Summary The improvement of rate of seedling emergence and early seedling growth of maize (Zea mays L.) under cool conditions has been an objective of breeding programs in cool regions for many years. To study inheritance of emergence time, and to determine if differences in emergence time were due to differences in seedling growth, F1, F2 and backcross generations of a diallel cross of two rapidly emerging lines from CIMMYT Pool 5, 5-113 and 5-154, and two elite Corn Belt Dent lines, A619 and A632, were grown in controlled environment rooms at low temperatures.The lines from Pool 5 emerged significantly faster than A619 and A632 over a range of low temperature conditions. This difference occurred both when the lines themselves were tested and when the lines were tested as male and female parents in crosses. The Pool 5 lines converted a higher proportion of their original seed to new root and shoot tissue than did A619 and A632, indicating that they had a faster seedling growth rate. Primarily this was due to a faster loss of seed reserve, rather than a more efficient conversion process.A significant difference occurred between A619 and A632 for emergence time, but this was not due to a difference in seedling growth rate.Reciprocal differences occurred only in the F1 generation in crosses involving A619, and then marked effects could be attributed to the male parent. Reciprocal differences tended to disappear in the F2. This suggested that the genotype of the embryo and endosperm was of much greater importance than the genotype of the maternal parent in determining differences of time to emergence and seedling growth.Mid-parent heterosis occurred for time to emergence and seed loss, a measure of mean rate of utilization of seed reserve, in all crosses. High parent heterosis occurred in several crosses for these traits. High parent heterosis occurred in all crosses for efficiency of utilization of seed reserve.A generation means analysis indicated that both additive and dominance effects were present for rate of seedling growth in crosses between A632 and the Pool 5 lines.  相似文献

Somaclonal-variation-induced multiple mutations were observed in a progeny of the S1587 plant, regenerated from type I calli of the aluminum-tolerant inbred maize line Cat-100-6. After five generations of self-pollination, 14 progeny families of the S1587 somaclone were found to show aluminum toxicity symptoms with altered root tip morphology and reduced primary root growth. The most sensitive progeny, S1587-17, was crossed to the Cat-100-6 inbred line. The parental lines and the F1 were tested in nutrient solutions containing an aluminum activity gradient of 0–93 ⋅ 10–6. The heterozygote behaves like the tolerant parent at aluminum activities up to 40 ⋅ 10–6 and showed an intermediate phenotype at higher aluminum concentrations. Histological sections of aluminum-treated roots from tolerant and sensitive plants stained with hematoxylin, an aluminum marker, showed a progressive destruction of the root tip of the aluminum-sensitive genotype over time and indicated that tolerance in Cat-100-6 could be due to an aluminum exclusion mechanism. Segregation analysis of the F2 and backcross to the sensitive parent based on root morphology of plants subjected to an aluminum activity of 30 ⋅ 10–6 showed the typical 3:1 and 1:1 tolerant:sensitive segregation ratios, respectively, indicating that tolerance in the Cat-100-6 inbred maize line is controlled by a single nuclear, semidominant gene, named Alm1. Hematoxylin staining is an early indicator of Aluminum (Al) toxicity effects on the apices of young, developing roots grown in nutrient solution. In this work, the potential of this technique as a reliable and reproducible phenotypic index for Al tolerance in tropical maize genotypes was assessed, with its performance systematically compared to two other parameters widely used in breeding programs – relative seminal-root length (RSRL) and net seminal-root length (NSRL). Seeding roots from contrasting genotypes for Al sensitivity stained remarkably different after 24- and 48-h and 7-day exposures to 222 μM Al in nutrient solution, with the Al-dye complex being detected in both the outer (epidermis) and inner (cortex) portions of the roots from the sensitive cultivar. Hematoxylin staining was compared to the RSRL and NSRL parameters using 20 families from the third generation of selfing (S3) following the cross between two contrasting inbred lines that had been previously classified by the RSRL index in an independent procedure. The coloration technique showed the highest capacity to discriminate among tolerant and sensitive genotypes and displayed significant correlation coefficients to the other two indexes. Evaluation of the results from diallel crosses involving nine inbred lines proved that hematoxylin staining was also particularly adequate for identifying expressive hybrid vigor, as demonstrated by the general (GCA) and specific (SCA) combining ability estimates obtained by using the three indexes simultaneously. Hence, hematoxylin staining of Al-stressed root apices appears to be a powerful tool to assist in Al-tolerance selection in tropical maize breeding programs. In crosses between hexaploid wheat and inbred lines of rye, a small number of rye genotypes produce seeds carrying undifferentiated, non-viable embryos. Hybrids between such lines and those not showing this phenotype were used as pollen donors in crosses with bread wheat in order to determine the genetic basis of disturbed embryo development. A single gene, designated Eml-R1b, is causing this character. Molecular markers associated with F2 genotypes derived from a contrasting rye inbred progeny were used for a linkage study. Recombinant inbred lines of an F5 population served as testers. Eml-R1b maps to chromosome arm 6RL, along with two co-segregating microsatellite loci, Xgwm1103 and Xgwm732. Complementary interactions of deleterious genes in wheat and rye are discussed.  相似文献

Summary Performance of a random array of recombinant inbred lines derived by single seed descent from five different source populations of Brussels sprouts (Brassica oleracea var. gemmifera) is presented. A total of 2,356 lines were tested in trials during 1985 and 1986. Three of the source populations were derived from double crosses between F1 hybrids. These hybrids show a considerable heterotic advantage over their inbred parents for the most important agronomic traits. The recombinant inbred lines performed, on average, less well than the parental inbred material, indicating that additive x additive genie interactions may make a significant contribution to the performance of current inbred material. Nevertheless, the very large variation among the recombinant inbred lines permitted many lines to be identified which outperformed the best parental inbred for all traits. Two lines outperformed the reference F1 hybrid, Gower, for an index that included marketable yield and quality. Consideration was also given to the dangers of misinterpreting phenotypically based proportions. Accordingly, response equations were used to ascertain the real genetic progress that was made. Advance seemed small when compared with the large heterotic effect, which is consistent with the segregation of a large number of loci. The distribution of the recombinant inbred lines was compared to predictions made from early generation trials. There was broad agreement but significant discrepancies existed which, it is suggested, may arise from the effects of genotype-environment interactions.  相似文献

Summary The agronomic performance of 9 doubled haploid (DH) lines of Chinese Spring, 6 DH lines of Hope, 14 DH lines of the single chromosome substitution line Chinese Spring (Hope 5 A) and their respective parents was analyzed under field conditions. Seventeen Chinese Spring DH lines derived from wheat x Hordeum bulbosum crosses were also included for comparison. No significant variation was detected in either population of Chinese Spring DH lines and neither DH population differed from its parent. The Hope DH lines differed significantly for tiller biomass, spikelet number per ear, ear grain weight and 50-grain weight. However, all the variation could be attributed to the poor performance of only one line. Chinese Spring (Hope 5 A) DH lines showed significant variation for ear emergence time, but this was probably due to genetic heterogeneity in the parental stock. Overall, the results suggest that most DH lines produced by the wheat x maize method resemble their wheat parent, and that the variation induced in DH production is likely to be similar to that found in DHs from wheat x Hordeum bulbosum crosses.  相似文献

In this paper, the causes of early embryo abortion in the reciprocal crosses between Phaseolus vulgaris L. (a cultivar) and Phaseolus coccineus L. (a wild form) were studied. Methacrylate resin sections, 3–5 μm thick, of 3 to 14 day-old seeds were used to examine the embryo developmental stages and the state of seed tissue. It was observed that, embryos aborted at different developmental stages (globular to early cotyledon) depending on the maternal parent. The use of P. coccineus cytoplasm resulted in a higher number of abortion than in reciprocal crosses. Many of them took place between 5 and 6 days after pollination (DAP). Histological analyses permitted to observe that the embryo development was slower in the cross between P. coccineus and P. vulgaris, compared to parental seeds. It would be related to a deficient endosperm development in reciprocal crosses and, in some extent, hypertrophy of the suspensor might be the main cause of early embryo abortion. Then, it would be practical to overcome this incompatibility by rescuing the embryo at the globular stage of development.  相似文献

Summary Cold acclimation responses of latitudinal ecotypes of Cornus sericea L. (C. stolonifera Michx.) and F1, F2 and BC1 hybrid progenies were measured under natural photoperiod conditions in St. Paul, MN and artificially shortened photoperiods in the glass-house. The 65 °N and 62 °N ecotypes (Alaska and Northwest Territories, respectively) were characterized by a short night length for hardiness induction, the 42 °N ecotype (Utah A and B) by a long night length for hardiness induction, while the F1 was intermediate to the parents. Results from reciprocal crosses indicated there was no significant unilateral maternal influence on cold acclimation. Acclimation responses of the F2 were highly variable but generally ranged between the parental extremes. However, three individuals from the 42 ° × 62 °N crosses exhibited greater cold resistance than the northern parent on two successive freezing test dates. F2 plants were also found with less freezing resistance than the southern parent. We used bulked segregant analysis (BSA) to identify microsatellite markers associated with water-stress tolerance in wheat. Two DNA pools (tolerant and sensitive) were established from the selected F2 individuals of crosses between water-stress-tolerant and -sensitive wheat parental genotypes on the basis of the paraquat (PQ) tolerance, leaf size, and relative water content. All three traits were previously shown to be associated with water-stress tolerance on segregating F2 progeny of the wheat crosses used in this study. Microsatellite analysis was then performed on the established DNA pools, using 35 primer pairs that included all of the chromosome group 5 (5A, 5B, 5D) markers, to detect microsatellite fragments that were present, absent, or both in the DNA pools and their parental lines. We identified one microsatellite fragment that was present in tolerant parent wheat and the tolerant bulk but absent in the sensitive parent wheat and sensitive bulk. We then followed the segregation of this marker in the tolerant F2 individuals. Use of this marker may significantly enhance the success of selection for PQ- and water-stress-tolerant genotypes in wheat breeding programs.  相似文献

The phenotypic manifestation and genetic control of embryo lethality observed in crosses between common wheat and rye were studied. It was found that crosses between common wheat and inbred self-fertile rye lines L2 and 535 gave rise to ungerminating grains, in which the development and differentiation of the hybrid embryo are arrested. Study of the degree of embryo development in the hybrid grains obtained by crossing common wheat varieties with inbred rye lines L2 and 535 showed that genotypes of the parents affected the ratio between undifferentiated embryos of various sizes. Analysis of this trait was performed by test crosses according to a novel pedigree program with the use of interlinear hybrids and a set of fourth-generation hybrid recombinant inbred lines. Rye line L2 was shown to bear the Eml (Embryo lethality) gene, which terminates the development of the hybrid embryo in amphihaploids. The suggestion of complementary interaction between wheat and rye genes during formation of a “new” character in wheat-rye F1 hybrids is discussed. A method of detecting an allele not complementary to the rye Eml allele in wheat is proposed. Early generations of hybrids can express both genetic incompatibilities and phenotypic novelty. Insights into whether these conflicting interactions between intrinsic and extrinsic selection persist after a few generations of recombination require experimental studies. To address this question, we use interpopulation crosses and recombinant inbred lines (RILs) of the copepod Tigriopus californicus, and focus on two traits that are relevant for the diversification of this species: survivorship during development and tolerance to thermal stress. Experimental crosses between two population pairs show that most RILs between two heat‐tolerant populations show enhanced tolerance to temperatures that are lethal to the respective parentals, whereas RILs between a heat‐tolerant and a heat‐sensitive population are intermediate. Although interpopulation crosses are affected by intrinsic selection at early generational hybrids, most of the sampled F9 RILs have recovered fitness to the level of their parentals. Together, these results suggest that a few generations of recombination allows for an independent segregation of the genes underlying thermal tolerance and cytonuclear incompatibilities, permitting certain recombinant lineages to survive in niches previously unused by parental taxa (i.e., warmer thermal environments) without incurring intrinsic selection.  相似文献

Summary Interspecific crosses between Zinnia angustifolia clones (maternal parents) and Z. elegans lines (paternal parents) were performed to investigate postzygotic barriers among Z. angustifolia X Z. elegans hybrids and to determine influence of parental genotype on embryonic and vegetative development of interspecific hybrids. Variation in percentage of emerged seedlings (PES) and percentage of morphologically normal hybrids (PNH) was attributable to Z. angustifolia clones with minor or no effect attributable to Z. elegans lines. Heterogeneity in PES values among Z. angustifolia clones was due to differences in amount of hybrid embryo breakdown and ungerminable seed. Cytological observations of normal and abnormal interspecific hybrids revealed similar chromosome numbers (2n=23) but indicated a low mitotic index for abnormal hybrids. Genetic analysis of PES and PNH suggested control by multiple genes inherited from the Z. angustifolia genome. Zymograms of single individuals of Aedes aegypti were obtained by means of starch gel electrophoresis, using alpha-naphthyl acetate as substrate. Inbred lines gave consistently homogeneous patterns; earlier results from random-breeding laboratory strains had shown considerable variability. Six distinct bands were observed. The furthest moving band, designated Esterase 6, showed differential migration in two inbred lines. Reciprocal crosses between these lines gave F1 progeny showing both bands. Backcrosses of F1 to either parental line gave a 1:1 segregation. These results are consistent with the hypothesis that the two forms of Esterase 6 are controlled by a single pair of codominant alleles at a single gene locus (Est 6 a and Est 6 b). Linkage tests with marker genes have demonstrated that Est 6 is on linkage group 2, with the following alignment: spot-abdomen (9.0±1.0) yellow-larva (17.4±1.3) Est 6. Crosses with another inbred line demonstrated a third band with intermediate mobility, designated Est 6 c. Distortion of expected Mendelian segregation ratios, commonly observed in many plant taxa, has been detected in an experimental three-generation inbred pedigree of Populus founded by interspecific hybridization between P. trichocarpa and P. deltoides. An RFLP linkage map was constructed around a single locus showing severe skewing of segregation ratio against F2 trees carrying the P. trichocarpa allele in homozygous form. Several hypotheses for the mechanism of segregation distortion at this locus were tested, including directional chromosome loss, segregation of a pollen lethal allele, conflicts between genetic factors that isolate the parental species, and inbreeding depression as a result of genetic load. Breeding experiments to produce inbred and outcrossed progenies were combined with PCR-based detection of RFLPs to follow the fate of the deficient allele throughout embryo and seedling development. A recessive lethal allele, lth, inherited from the P. trichocarpa parent, was found to be tightly linked to the RFLP marker locus POP1054 and to cause embryo and seedling mortality.  Inbreds selfed from the same F2 or backcross population are referred to as sister inbreds. In some situations, maize (Zea mays L.) sister inbreds may not have testcross data available for best linear unbiased prediction (BLUP) of single-cross performance. This study evaluated the usefulness of BLUP and restriction fragment length polymorphism (RFLP)-based coefficients of coancestry ( f ) in predicting the testcross performance of sister inbreds. Parental contributions (p) were estimated from 70 RFLP loci for 15 inbreds that comprised three sister inbreds selfed from each of five F2 populations. Estimates of p were subsequently used to calculate RFLP-based f. Grain yield, moisture, and stalk lodging data were obtained for 2265 single crosses tested by Limagrain Genetics in multilocation trials from 1990 to 1995. Performance of the sister inbreds when crossed to several inbred testers was predicted from the performance of the tested single crosses and RFLP-based f. Correlations between predicted and observed performance, obtained with a delete-one cross-validation procedure, were erratic and mostly low for all three traits. Correspondence was poor between ranks for predicted and observed general combining ability of the sister inbreds. The results suggested that the proportion of the genome derived by a sister inbred from a given parental inbred does not solely determine its testcross performance. The failure of BLUP and RFLP-based f to consistently predict testcross performance indicated that actual field testing will continue to be necessary for preliminary evaluation of sister inbreds. Summary Diallel crosses among Triticum boeoticum (4 lines from different geographical areas), T.urartu, Aegilops squarrosa and Ae. speltoides exhibited reciprocal differences in hybrid seed morphology, endosperm development, and embryo viability. T. urartu and Ae. squarrosa as females with T. boeotiaum and Ae. speltoides lead to shrivelled inviable seed. T.boeoticum accessions as female with Ae.speltoides also lead to shrivelled seeds. The reciprocal crosses produced plump seeds which either resembled the maternal parent or showed size differences. By altering the endospermic genome ratios, hybrid seeds with 1 (PF)/1 (PM) showed extreme shrivelling whereas those with 4 (PF)/1 (PM) were medium shrivelled to plump. Genetic experiments involving hybrids of T. boeoticum, T. urartu and T. monococcum showed that a factor is present in pollen or male gametes, which shows dosage effect and which, by interacting with the maternal genome, leads to endosperm abortion.  相似文献

Zusammenfassung Dreizehn homozygote Inzuchtlinien aus europäischen Roggensorten wurden auf ihre Fähigkeit getestet, in Einfachkreuzungen mit der cytoplasmatisch männlich sterilen Inzuchtlinie L1-P die Pollen-fertilität wiederherzustellen. Zwei von den Bestäuberlinien erwiesen sich als voll wirksame Restorerformen. Acht Linien wurden als Nicht-Restorergenotypen eingestuft. Für die übrigen drei Linien konnten keine schlüssigen Ergebnisse erzielt werden.
Restoration of pollen fertility to cytoplasmic male sterile rye
Summary Thirteen homozygous inbred lines derived from European rye (Secale cereale L.) varieties were evaluated for their ability to restore pollen fertility to single crosses with the cytoplasmic male-sterile inbred line L1 — P as female parent. Two of the pollinator lines proved to be fully effective in restoring fertility. Eight lines were classified as nonrestorer genotypes. No conclusive results were obtained for the three remaining pollinator lines.

Results are reported of crossing a dwarf seed stalk line (mean height 44.8 cm) from Israel with five inbred lines of the Rijnsburger type each having normal tall seed stalks (mean heights 83.1–105.9 cm). The seed stalk height of F1 generations from these crosses was slightly greater than that of the Rijnsburger parents but segregation occurred in the F2 giving plants which could be considered ‘dwarf’ and ‘normal’ with respect to seed stalk height. Selfing individual F2‘dwarf’ plants produced F3 progenies whose mean heights were in the dwarf range, and were significantly correlated with those of their F2 parent. The results obtained in the F2 and F3 families suggest that although a major recessive gene for dwarfness may be present, substantial minor gene and environmental variation is also involved.  相似文献
