The model of Wilson and co-workers (2., 3., Arch. Biochem. Biophys. 182, 749–762) for the regulation of mitochondrial oxidative phosphorylation has been extended to include the dependence on oxygen tension. The derived rate expression correctly describes the observed dependence of cellular energy metabolism on oxygen tension, including the oxygen dependence at “normoxic” physiological values. Experimental evidence is presented that oxidative phosphorylation by suspensions of isolated rat liver mitochondria is also dependent on oxygen concentration up to values of at least 100 μM.  相似文献

A theory of 1f and conductance noise is given for ionic channels in nerve membrane. The theory is based on the assumption that the channels are in constant, stochastically independent, rotational motion within a fluid bilayer membrane. The resulting expression for the current noise power density S contains a conduction noise term consistent withStevens (1972) and Hill & Chen (1972) and a 1f noise term consistent with Lundstrom & McQueen (1974) and Clay & Shlesinger (1976). The expression for S also contains a third term which is the spectrum of the product of the single channel conduction noise and 1f noise correlation functions. This term is independent of the number of channels in the membrane, R. Consequently, the expression for S effectively reduces to a sum of 1f and conduction noise for R 10–100 which is in agreement with noise measurements on squid axon. The theory is applied in detail to potassium squid noise measurements of Conti, DeFelice & Wanke (1975) using the stochastic analysis of single file ion motion developed in our previous paper (Clay & Shlesinger (1976)).  相似文献

This note considers sampling theory for a selectively neutral locus where it is supposed that the data provide nucleotide sequences for the genes sampled. It thus anticipates that technical advances will soon provide data of this form in volume approaching that currently obtained from electrophoresis. The assumption made on the nature of the data will require us to use, in the terminology ofKimura (Theor. Pop. Biol.2, 174–208 (1971)), the “infinite sites” model of Karlin and McGregor (Proc. Fifth Berkeley Symp. Math. Statist. Prob.4, 415–438 (1967)) rather that the “infinite alleles” model of Kimura and Crow (Genetics49, 174–738 (1964)). We emphasize that these two models refer not to two different real-world circumstances, but rather to two different assumptions concerning our capacity to investigate the real world. We compare our results where appropriate with corresponding sampling theory of Ewens (Theor. Pop. Biol.3, 87–112 (1972)) for the “infinite alleles” model. Note finally that some of our results depend on an assumption of independence of behavior at individual sites; a parallel paper byWatterson (submitted for publication (1974)) assumes no recombination between sites. Real-world behavior will lie between these two assumptions, closer to the situation assumed by Watterson than in this note. Our analysis provides upper bounds for increased efficiency in using complete nucleotide sequences.  相似文献

During recent years, numerous attempts have been made to correlate both quantitative (Davies &; Taylor, 1959; Engen, 1962; Beck, 1964; Engen, Cain &; Rovee, 1968; Cain, 1969; Dravnieks &; Laffoit, 1970; Laffort, 1969a,b) and qualitative (Davies, 1965; Amoore &; Venstrom, 1965; Döving, 1966a,b; Wright &; Michels, 1964; Leveteau &; MacLeod, 1969) odorous properties of single compounds to their molecular properties. These attempts have been only partially successful.In the present paper we will try to explain the several odorous properties of single compounds on the basis of the non-specific properties of odorants involved in solubility.This model is a first approach, and although it gives statistically highly significant relations, it is not as accurate as those advanced with respect to the physical and sensory dimensions of stimuli in the fields of vision and audition.We will first give the present definitions of the most suitable physicochemical parameters, and then advance quantitative and qualitative models for single compounds. Quantitative odorous properties are: odour threshold, rate of change of odour intensity with odorant concentration in the suprathreshold region, and the somewhat controversial upper odour intensity. Kidney cortex from alloxan-diabetic rats has an elevated activity of hexokinase, enzymes of the pentose phosphate pathway and of certain enzymes of the glucuronate-xylulose route relative to age-matched control rats. These changes are highly significant when expressed as total kidney units/100 g body weight, a parameter relating biochemical activity to the functional requirement of the whole animal. These changes are discussed in relation to the hypothesis of ‘glucose over-utilization’ in diabetes in tissues not requiring insulin for glucose uptake [R.G. In recent papers, it has been shown experimentally that the force-velocity relationship in single muscle fibres presents deviations from hyperbolicity at high values of the load (Edman, Mulieri & Scubon-Mulieri, 1976; Edman & Hwang,1977). It has been shown independently and on theoretical bases, that the parameter “b” in Hill's characteristic equation also presents deviations from its normal value at low values of the speed of shortening, i.e. at high values of the load (Morel, Pinset-Härström & Gingold, 1976). In the present paper, it is shown that both the experimental and the theoretical results are in excellent agreement and a theoretical force-velocity relationship is proposed.  相似文献

Migrating cells possess surface glycosyltransferase activity toward extracellular substrates, and the appearance of enzyme activity coincides with the onset of cellular migration (Shur, 1977a, Shur, 1977b, Develop. Biol.58, 23–39, 40–55; E. A. Turley and S. Roth, 1979, Cell17, 109–115). In this paper, surface glycosyltransferases were examined during normal and TT mutant mesenchyme migration. Of six glycosyltransferases that were assayed, only galactosyltransferase was present at significant levels on the cell surface, despite the presence of a variety of intracellular glycosyltransferases. All controls have been performed to show clearly the enzyme activity was cell surface localized. In both normal and TT embryos, surface galactosyltransferase activity was localized, by autoradiography, primarily to migrating mesenchymal cells, and to a lesser degree, to presumptive neural epithelium. During primitive streak formation, putative TT embryos were devoid of surface galactosyltransferase activity. However, as development progressed, the TT level of activity eventually exceeded wild-type levels by two- to sixfold and was evident in TT tissues prior to the onset of microscopic pathology. Other surface enzymes assayed did not show any TT-dependent increase in activity. The extracellular galactosyl acceptors were not chloroform:methanol soluble, and glycopeptides prepared by exhaustive Pronase digestion were excluded from Sephadex G-50. This large galactosylated glycoconjugate was readily digestable with endo-β-galactosidase, and, therefore, is similar to the poly-N-acetyllactosamine chains previously identified on early embryonic tissues (A. Kapadia, T. Feizi, and M. J. Evans, 1981, Exp. Cell. Res.131, 185–195; T. Muramatsu, G. Gachelin, M. Damonneville, C. Delarbre, and F. Jacob, 1979, Cell18, 183–191; A. Heifetz, W. J. Lennarz, B. Libbus, and Y. -C. Hsu, 1980, Develop. Biol.80, 398–408). These results support an involvement of surface galactosyltransferases in mesenchyme formation and during migration on poly-N-acetyllactosamine substrates.  相似文献

The Island Model introduced by Sewall Wright (1951) has proven to be a useful construction for studying the interaction of genetic drift, population subdivision, and mutation. Interest in the model has recently increased because of its relevance to certain questions involving the rate of differentiation of sub-populations under the neutral allele hypothesis (e.g., Smith, 1970; Latter, 1973). It is perhaps the only realistic population structure in which the test for neutrality proposed by Lewontin and Krakauer (1973) is valid (Lewontin and Krakauer, 1975). If data from natural populations is to be compared to the predictions of the Island Model, it is desirable to have an alternative model with the same migration pattern but with natural selection operating. In this paper one such model will be introduced where the stochastic element comes from random fluctuations in the environment rather than from genetic drift. The model is a direct extension of the one in the previous paper in this series (Gillespie, 1975) which dealt with a population which is subdivided into two patches with restricted migration between them.  相似文献

Hepatotoxic studies of xylidines differ with regard to the animal species and type of isomer. The aim of the present paper is to correlate electronic structure data and physical-chemical properties, studied by the authors during a few previous papers (Sahini & Weinberg, 1973; Sahini, Weinberg & Vasilescu 1976, Weinberg & Sahini, to be published; Weinberg & Sahini, 1980), with the hepatotoxic activity of xylidines. A modified Hodgkin & Huxley (1952) model for axons was used to simulate smooth muscle action potentials. The modifications were such as to match our own experimental results and published data on the passive and active behavior of smooth muscle.A brief account of the modifications introduced to the HH model is as follows. The resting ionic conductances were obtained from the data of Casteels (1969). Chloride conductance was replaced by an ad hoc leakage conductance (g?L) in order to obtain a resting membrane resistance of about 11 kΩcm2. The ionic equilibrium potentials were according to Kao & Nishiyama (1969). The rate constants m, n and h have similar form to those in axons, but their different numerical values produce action potentials that match the duration of the smooth muscle action potential (about 16 ms) at half its maximum amplitude. The effective membrane capacitance was taken as 2.5 μF/cm2.The results obtained by implementing those smooth muscle parameters in the HH formulation include: (a) a membrane potential that matches the main characteristics of experimentally recorded action potentials in uterine smooth muscle and guinea-pig taenia-coli, and (b) a propagated action potential which, on a cable diameter of 5 μm (similar to the diameter of a single smooth muscle cell), has a speed of propagation within the range of the values experimentally recorded in smooth muscle. This observed velocity of propagation is not compatible with a large cable and it is concluded that “functional units” are not required to sustain propagation of action potentials in smooth muscle.  相似文献

The time of the last DNA replication of the Mauthner's neuron precursor cell has been investigated using radioautography. Embryos of Xenopus laevis were labeled at different stages of early development by single microinjections of tritiated thymidine. Labeling times were designed to cover the entire period of development between gastrula and hatching stages. The embryos were fixed at later stages (41 to 44, according to Nieuwkoop and Faber, 1967), when the Mauthner neuron can be readily distinguished by its characteristically large size and large nucleolus.Mauthner neurons of embryos which received tritiated thymidine from stage 10 (beginning of gastrulation) to stage 12 (advanced gastrula, medium yolk plug) were always labeled. Those embryos which received the isotope at or after stage 1212 (advanced gastrula, small yolk plug) were never found labeled. These results imply that the last DNA replication of the cell destined to give rise to the Mauthner neuron occurs during the last gastrula stages. This last DNA replication immediately proceeds the time of the so-called “histogenetic determination” of the Mauthner neuron proposed to correspond to stage 13 (slit blastopore) by Stefanelli (1951).Therefore it appears that the developmental program of the Mauthner neuron involves a remarkably early cessation of DNA replication closely followed by histogenetic determination. This is the earliest known event of this type for a specific, well characterized neuron in the amphibian embryo.  相似文献

Electron microscopic examination of the histone H1-depleted, folded genomes of Drosophila melanogaster reveals that they are composed of long cylindrical cables of about 100 Å diameter. Limited single-strand nicking with DNAase I relaxes the 100 Å fibers to a “beads-on-a-string” structure, showing the nucleosomes and internucleosome DNA.Based on these results and other available data, we have constructed a detailed space-filling model for the higher order DNA coiling in chromatin, starting with the symmetrical nucleosome core previously described (Weintraub, Worcel and Alberts, 1976). The model defines the path of the DNA helix and the nucleosome arrangement along the DNA coil for both the 100 Å and the 200–300 Å fibers.Following Sobell et al. (1976), we believe that the DNA is coiled in the 100 Å nucleofilament in a uniform left-handed supercoil of about 90 base pairs (bp) per turn and 47 Å pitch; the 140 bp symmetrical nucleosome cores align themselves along this uniform DNA superhelix so that the isologous outer surfaces of adjacent nucleosomes touch and the internucleosome spacer DNA coils between them. A few single-strand discontinuities [about one nick per 85 kilobases (kb); Benyajati and Worcel, 1976] in the H1-depleted 100 Å fiber can thus relax the negatively supercoiled internucleosome DNA generating the “beads-on -a-string” appearance.We propose that histone H1 binds to the 100 Å diameter superhelix and coils it into tightly packed, 110 Å pitch super-superhelices (“solenoids;” Finch and Klug, 1976) of variable diameter (between 200–300 Å). In our model, the “thick” 200–300 Å fiber is stabilized at metaphase by histone H1-H1 heterologous interactions between adjacent helical turns of the nucleofilament, and the internucleosome spacer DNA is located on the outside. Symmetry considerations demand that changes in the length of the repeat should lead to variations in the number of nucleosomes per helical turn and in the handedness of these turns in the 200–300 Å metaphase fiber.  相似文献

Price's (1970) covariance theorem can be used to derive an expression for gene frequency change in kin selection models in which the fitness effect of an act is independent of the genotype of the recipient. This expression defines a coefficient of relatedness which subsumes r(Wright, 1922), b(Hamilton, 1972), ρ (Orlove &; Wood, 1978), and R(Michod &; Hamilton, 1980). The new coefficient extends the domain of Hamilton's rule to models in which the average gene frequency of actors differs from that of recipients.  相似文献

An analysis has been made of the composition and structure of the two types of sheets assembled from material from dissociated bacteriophage T2 (Poglazov &; Mesyhanzhinov, 1967) and T4 capsids. Serological techniques have been used to show that both types of sheet are assembled from proteolytic fragment of P231, the major capsid constituent. The two types of sheets have been found to interconvert depending on the concentration of Mg2+ ions in the buffer. Computer modelling experiments show that the “hexagonal” and “rectangular” morphologies observed in the negative stain are due to in-register and staggered associations, respectively, of a single basic hexagonal lattice. Analysis by polyacrylamide gel electrophoresis of samples of sheets and dissociated capsids, together with previous results from immune electron microscopy (Kistler et al., 1978), suggest that hexamers of the proteolytic fragment are derived conservatively from capsomers of the phage head.The value of this proteolytic P23 fragment has been twofold: (1) it has proved to be a useful peptide in the ongoing primary sequence determination of P23 and (2) antibodies raised against it have been employed to follow the fate of P23 antigenic sites during various steps of phage capsid maturation (Kistler et al., 1978).  相似文献

The Rhesus monkey (Macaca mulatta) has 21 pairs of chromosomes, many of which can easily be confused with one another by the traditional staining methods. By using the method of banding with trypsin (Seabright, 1971) we have been able to characterize the various pairs of homologous chromosomes and we have classified them by following the criteria adopted by Rothfels & Siminovich (1958). The evolutionary meaning of the results are discussed.  相似文献

The pH dependence of proton uptake upon binding of NADH to porcine heart mitochondrial malate dehydrogenase (l-malate: NAD+ oxidoreductase, EC 1.1.1.37) has been investigated. The enzyme has been shown to exhibit a pH-dependent uptake of protons upon binding NADH at pH values from 6.0 to 8.5. Enzyme in which one histidine residue has been modified per subunit by the reagent iodoacetamide (E. M. Gregory, M. S. Rohrbach, and J. H. Harrison, 1971, Biochim. Biophys. Acta253, 489–497) was used to establish that this specific histidine residue was responsible for the uptake of a proton upon binding of NADH to the native enzyme. It has also been established that while there is no enhancement of the nucleotide fluorescence upon addition of NADH to the iodoacetamide-modified enzyme, NADH is nevertheless binding to the modified enzyme with the same stoichiometry as with native enzyme. The data are discussed in relation to the involvement of the essential histidine residue in the catalytic mechanism of “histidine dehydrogenases” recently proposed by Lodola et al. (A. Lodola, D. M. Parker, R. Jeck, and J. J. Holbrook, 1978, Biochem. J.173, 597–605) and the catalytic mechanism of “malate dehydrogenases” recently proposed by L. H. Bernstein and J. Everse (1978, J. Biol. The plasticity of growth of animals in time, due to the resilience of their response to the ways they can be fed, suggests the difficulty of describing growth by a stochastic model in the time domain. A model is presented which avoids this difficulty by describing growth as a Markov process in the food-consumed domain, assuming that, at conception, (1) the maximum mature weight as a number α of biomass units of mass μ, and (2) the probability B of production of a biomass unit per unit of food consumed, are specified. Constancy of α, μ and B, as the animal feeds, is the basis of the proposed Markov process. The mean growth from infancy to maturity in the food-consumed domain is then the old law of diminishing returns empirically formulated first by Spillman (1924) for cattle and swine, and confirmed by Titus, Jull &; Hendricks (1934) for fowl, and by Parks (1972) across species from mice to steers. The solution also leads to the possibility that the distribution of weights in a population of growing animals of the same species, is related to the distribution of mature weights among the individuals. An experiment by Lister &; McCance (1967) with well-fed and severely undernourished pigs, shows the stability of growth in the foodconsumed domain compared to the plasticity in the time domain. Other implications of the model are discussed.  相似文献

The “intruder” hypothesis for the presence of more arginine codons in the genetic code than are needed leads to the expectation for selection against arginine in protein synthesis.
