A polymeric-resin-based microbiological assay is shown to yield results consistent with classical inhibition zone theory. This novel assay is applicable to cycloheximide, an antifungal antibiotic, at concentrations below 1.0 mug/mL and up to 800 mug/mL. Addition of ethanol to the cut-out wells of the assay plate helps to release the antibiotic and drastically increases the inhibition zone size. The antibiotic concentrations of both two component and multicomponent systems can be determined using this method. The use of polymeric-resin beads to monitor antibiotic concentrations is shown to be an effective technique for fermentation monitoring.  相似文献

Summary In order to study and control fermentation processes, indirect on-line measurements and mathematical models can be used. Here an on-line model for fermentation processes is presented. The model is based on atom and partial mass balances as well as on stability equations for the protolytes. The model is given an adaptive form by including transport equations for mass transfer and expressions for the fermentation kinetics. The state of the process can be estimated on-line using the balance component of the model completed with measurement equations for the input and the output flows of the process. Adaptivity is realized by means of on-line estimation of the parameters in the transport and kinetic expressions using recursive regression analysis. On-line estimation of the kinetic and mass transfer parameters makes model-based predictions possible and enables intelligent process control while facilitating testing of the validity of the measurement variables. A practical MS-Windows 3.1 model implementation called FMMS—Fermentation Monitoring and Modeling System is shown. The system makes it easy to configure the operating conditions for a run. It uses Windows dialogs for all set-ups, model configuration parameters, elemental compositions, on-line measurement devices and signal conditioning. Advanced on-line data analysis makes it possible to plot variables against each other for easy comparison. FMMS keeps track of over 100 variables per run. These variables are either measured or estimated by the model. Assay results can also be entered and plotted during fermentation. Thus the model can be verified almost instantly. Historical fermentation runs can be re-analyzed in simulation mode. This makes it possible to examine different signal conditining filters as well as the sensitivity of the model. Combined, the data analysis and the simulation mode make it easy to test and develop model theories and new ideas.  相似文献

Summary In an aerobic fermentor with a fermentation medium based on 4% corn steep liquor and salts, Bacillus sphaericus, strain NCTC 10338, produces an intracellular V-penicillin acylase, when pH is in the narrow interval 7.5–8.0. The strain grows well outside this pH range, but the growth is not associated with acylase activity in the cells. The enzyme was not induced by phenoxy acetic acid and was not repressed by glucose.  相似文献

Two substances besides rubiginol, of positive ferric chloride reaction were isolated from a shaking culture of Gluconoacetobacter liquefaciens, using glucose as the substrate. One of them was found to be a new γ-pyrone derivative and the composition was concluded to be 3,5-dihydroxy-l,4-pyrone-2-carboxylic acid. The other substance was found to be comenic acid. Assumptions were made with regard to the mechanism of the formation of these γ-pyrone derivatives from glucose.  相似文献

From an anaerobic digester a novel type of strictly anaerobic, Gram-negative, non-sporeforming mesophilic bacterium was isolated. The cells were curved rods, motile by means of lateral flagella and contained b- and c-type cytochromes. The G+C content of the DNA was 48.0±1.0 mol%. The isolate was able to ferment only glutamate, aspartate, lactate and pyruvate. Organic fermentation products were acetate, propionate and succinate. Propionate was probably formed via a reductive succinate pathway. Sheep fed a hay-concentrate diet were adapted to pectin administration and ruminal infusion of methanol. Both treatments resulted in a strong increase in the rate of methanogenesis from methanol. Quantitative data show that methanol was exclusively converted into methane. Treatments did not influence ruminal volatile fatty acid percentages.  相似文献

The parturograph is a composite record designed for the monitoring of fetal and maternal well-being and the progress of labour. It permits the early recognition of abnormalities and pinpoints the patients who would benefit most from intervention. Observations are made from the time of admission of the mother to the caseroom and recorded graphically. Factors assessed include fetal heart rate, maternal vital signs and urine, cervical dilatation, descent of the presenting fetal part, and frequency, duration and intensity of uterine contractions.  相似文献

A possible mechanism of fermentation conversion is described from polyalcohol fermentation to ethanol fermentation by Pichia miso. Little alcohol dehydrogenase activity was found in polyalcohol-producing cells, whereas higher enzyme activity was induced by ethanol-producing cells. The fermentation conversion may be caused by the different levels of alcohol dehydrogenase activity between polyalcohol- and ethanol-producing cells. It was also shown that yeast growth was inhibited and that yeast cells were lysed by ethanol (at 6g/100ml) that accumulated in 24 hr.  相似文献
