A comparison of the effects of foliar and soil N application was made in field-grown mature fruiting Gala/M9 apple trees (Malus domestica Borkh) in 2001 and 2002 growing seasons under Pacific Northwest growing conditions in southern British Columbia, Canada. The trees, six years old at the start of the experiment, were treated: (1) with 5 g/l urea sprays supplied every two weeks (7 times) from mid May to mid August (total about 50 g N/tree/year), (2) with the same amount of N applied to the soil with the same timing and quantity as for the foliar treatment, and (3) with no N (control). Leaf color (as SPAD readings) and N concentrations (mg/g), and soil NH4+-N and NO3-N were measured periodically throughout the two seasons. Leached NO3-N was monitored monthly via an anion exchange probe from June to October in 2001 and from May to November in 2002. Shoot length was measured in October and N concentration of one-year-old wood and roots was determined in December of each growing s eason. Soil N application significantly increased shoot length relative to control or foliar N application. Leaf color, leaf N, and N concentration of one-year-old wood and roots were similarly increased relative to control by both soil and foliar N application. These treatments also increased fruit yield relative to control. There was no significant difference in yield and fruit quality between soil and foliar N applications. Soil N application increased soil NH4+-N and NO3-N content in the root zone, and also increased the NO3 leaching loss below the root zone especially late in the growing season. Our results suggested that tree N status and yield and fruit quality could be maintained by multiple urea sprays during the growing season in apple orchards, and foliar N application will reduce the risk of soil NO3-N leaching.  相似文献

Summary A study of the effects of malathion and parathion applied at 10 and 50 g/g of soil on transformations of urea and (NH4)2SO4–N in a sandy loam showed that the insecticides retarded urea hydrolysis as well as nitrification of urea and (NH4)2SO4–N. At 50 parts/106 rate of the insecticides, inhibition of urea hydrolysis ranged from 44 to 61% after 0.5 week and from 7 to 21% after 3 weeks of application. The insecticides inhibited the conversion of NH4 + to NO2 without appreciably affecting the subsequent oxidation of NO2 to NO3 –N. This resulted in accumulation of higher amounts of NH4 +–N in soil samples treated with ammonium sulfate or urea N.The results suggest that transformations of urea and NH4 + fertilizers in soils may be influenced by the amount of organophosphorus insecticide present and this may affect plant nutrition and fertilizer use.  相似文献

Nitrification and denitrification rates were estimated simultaneously in soil-floodwater columns of a Crowley silt loam (Typic Albaqualfs) rice soil by an15N isotopic dilution technique. Labeled NO 3 was added to the floodwater of soil-water columns, half were treated with urea fertilizer. The (NO 3 +NO 2 )–N and (NO 3 +NO 2 )–N concentrations in the floodwater were measured over time and production and reduction rates for NO 3 calculated. Nitrate reduction in the urea amended columns averaged 515 mol N m–2h–1 and nitrification averaged 395 mol N m–2h–1 over the 35–153 d incubation. The nitrification rate for 4–19 d sampling period (1,560 mol N m–2h–1) in the urea amended columns was almost 9 times greater than the reduction rate (175 mol N m–2h–1) over the same period. Without the addition of urea the NO 3 production rate averaged 32 mol N m–2h–1 and reduction 101 mol N m–2h–1.  相似文献

Copper deficiency can delay flowering and plant maturity. However, the effect of copper deficiency on the rate of leaf emergence has not been quantified. We tested the hypothesis that low copper supply decreases the rate of leaf emergence of wheat (Triticum aestivum L. cv Gamenya). Copper foliar sprays are commonly applied to wheat. We examined the response of the rate of leaf emergence to a foliar application of copper sulphate.Wheat was grown in root cooling tanks (20°C) in the glasshouse. Soil copper treatments were applied as solutions of CuSO4.5H2O at three rates: Cu0=no added Cu, Cu400=400 g Cu per 3 kg pot, and Cu1600=1600 g Cu per pot. An additional treatment of a foliar spray of CuSO4.5H2O (0.4 mg Cu per plant) was applied to Cu0 and Cu400 plants 45 days after sowing (5.5 leaves on the main stem). Leaves on the main stem were counted and the rate of leaf emergence was estimated from the regression of number of emerged leaves against thermal time (base 0°C). The phyllochron was calculated as 1/rate of emergence.Leaves on Cu0 and Cu400 plants took longer to emerge than on Cu1600 plants, with the phyllochron of Cu1600 plants being 130 compared to 137 for the Cu400 plants and 158 for the Cu0 plants. The foliar application of CuSO4 at the 5–6 leaf stage resulted in a decrease in the phyllochron of the Cu0 plants to 127, but no change in that of the Cu400 plants.  相似文献

Transformed Nicotiana plumbaginifolia plants with constitutive expression of nitrate reductase (NR) activity were grown at different levels of nitrogen nutrition. The gradients in foliar NO 3 content and maximum extractable NR activity observed with leaf order on the shoot, from base to apex, were much decreased as a result of N-deficiency in both the transformed plants and wild type controls grown under identical conditions. Constitutive expression of NR did not influence the foliar protein and chlorophyll contents under any circumstances. A reciprocal relationship between the observed maximal extractable NR activity of the leaves and their NO 3 content was observed in plants grown in nitrogen replete conditions at low irradiance (170 mol photons·m–2 ·s–1). This relationship disappeared at higher irradiance (450 mol photons·m–2·S–1) because the maximal extractable NR activity in the leaves of the wild type plants in these conditions increased to a level that was similar to, or greater than that found in constitutive NR-expressors. Much more NO 3 accumulated in the leaves of plants grown at 450 mol photons·m–2·s–1 than in those grown at 170 mol photons·m–2·s–1 in N-replete conditions. The foliar NO 3 level and maximal NR activity decreased with the imposition of N-deficiency in all plant types such that after prolonged exposure to nitrogen depletion very little NO 3 was found in the leaves and NR activity had decreased to almost zero. The activity of NR decreased under conditions of nitrogen deficiency. This regulation is multifactoral since there is no regulation of NR gene expression by NO 3 in the constitutive NR-expressors. We conclude that the NR protein is specifically targetted for destruction under nitrogen deficiency. Consequently, constitutive expression of NR activity does not benefit the plant in terms of increased biomass production in conditions of limiting nitrogen.Abbreviations Chl chlorophyll - N nitrogen - NR NADH-nitrate reductase - WT wild type  相似文献

Measurement of nitrogen isotopic composition (15N) of plants and soil nitrogen might allow the characteristics of N transformation in an ecosystem to be detected. We tested the measurement of 15N for its ability to provide a picture of N dynamics at the ecosystem level by doing a simple comparison of 15N between soil N pools and plants, and by using an existing model. 15N of plants and soil N was measured together with foliar nitrate reductase activity (NRA) and the foliar NO3 pool at two sites with different nitrification rates in a temperature forest in Japan. 15N of plants was similar to that of soil NO3 in the high-nitrification site. Because of high foliar NRA and the large foliar NO3 pool at this site, we concluded that plant 15N indicated a great reliance of plants on soil NO3 there. However, many 15N of soil N overlapped each other at the other site, and 15N could not provide definitive evidence of the N source. The existing model was verified by measured 15N of soil inorganic N and it explained the variations of plant 15N between the two sites in the context of relative importance of nitrification, but more information about isotopic fractionations during plant N uptake is required for quantitative discussions about the plant N source. The model applied here can provide a basis to compare 15N signatures from different ecosystems and to understand N dynamics.  相似文献

To investigate the effect of inorganic fertilizer and composted manure amendments on the N isotope composition (delta 15N) of crop and soil, maize (Zea mays L.) was cultivated under greenhouse conditions for 30, 40, 50, 60, and 70 days. Composted pig manure (delta 15N= +13.9) and urea (-2.3) were applied at 0 and 0 kg N ha–1 (C0U0), 0 and 150 kg N ha–1 (C0U2), 150 and 0 kg N ha–1 (C2U0), and 75 and 75 kg N ha–1 (C1U1), respectively. The delta 15N of total soil-N was not affected by both amendments, but delta 15N of NH+ 4 and NO 3 provided some information on the N isotope fractionation in soil. During the early growth stage, significant differences (P < 0.05) in delta 15N among maize subjected to different treatments were observed. After 30 days of growth, the delta 15N values of maize were +6.6 for C0U0, +1.1 for C0U2, +7.7 for C2U0, and +4.5 for C1U1. However, effects of urea and composted manure application on maize delta 15N progressively decreased with increasing growth period, probably due to isotope fractionation accompanying N losses and increased uptake of soil-derived N by maize. After 70 days of growth, delta 15N of leaves and grains of maize amended with composted pig manure were significantly (P < 0.05) higher than those with urea. The temporal variations in delta 15N of maize amended with urea and composted manure indicate that plant delta 15N is generally not a good tracer for N sources applied to field. Our data can be used in validation of delta 15N fractionation models in relation to N source inputs.  相似文献

The Central Asian Taklamakan desert is characterized by a hyperarid climate with less than 50 mm annual precipitation but a permanent shallow groundwater table. The perched groundwater (2–16 m) could present a reliable and constant source of nitrogen throughout the growing season and help overcome temporal nitrogen limitations that are common in arid environments. We investigated the importance of groundwater and nitrogen fixation in the nitrogen metabolism of desert plants by assessing the possible forms and availability of soil N and atmospheric N and the seasonal variation in concentration as well as isotopic composition of plant N. Water availability was experimentally modified in the desert foreland through simulated flooding to estimate the contribution of surface water and temporally increased soil moisture for nutrient uptake and plant–water relations. The natural vegetation of the Taklamakan desert is dominated by plants with high foliar nitrogen concentrations (2–3% DM) and leaf nitrate reductase activity (NRA) (0.2–1 mol NO2 g–1 FW h–1). There is little evidence that nitrogen is a limiting resource as all perennial plants exhibited fast rates of growth. The extremely dry soil conditions preclude all but minor contributions of soil N to total plant N so that groundwater is suggested as the dominant source of N with concentrations of 100 M NO3. Flood irrigation had little beneficial effect on nitrogen metabolism and growth, further confirming the dependence on groundwater. Nitrogen fixation was determined by the 15N natural abundance method and was a significant component of the N-requirement of the legume Alhagi, the average contribution of biologically fixed nitrogen in Alhagi was 54.8%. But nitrogen fixing plants had little ecological advantage owing to the more or less constant supply of N available from groundwater. From our data we conclude that the perennial species investigated have adapted to the environmental conditions through development of root systems that access groundwater to satisfy demands for both water and nutrients. This is an ecologically favourable strategy since only groundwater is a predictable and stable resource.  相似文献

The measurement of natural 15N abundance is a well-established technique for the identification and quantification of biological N2 fixation in plants. Associative N2 fixing bacteria have been isolated from sugarcane and reported to contribute potentially significant amounts of N to plant growth and development. It has not been established whether Australian commercial sugarcane receives significant input from biological N2 fixation, even though high populations of N2 fixing bacteria have been isolated from Australian commercial sugarcane fields and plants. In this study, 15N measurements were used as a primary measure to identify whether Australian commercial sugarcane was obtaining significant inputs of N via biological N2 fixation. Quantification of N input, via biological N2 fixation, was not possible since suitable non-N2 fixing reference plants were not present in commercial cane fields. The survey of Australian commercially grown sugarcane crops showed the majority had positive leaf 15N values (73% >3.00, 63% of which were >5.00), which was not indicative of biological N2 fixation being the major source of N for these crops. However, a small number of sites had low or negative leaf 15N values. These crops had received high N fertiliser applications in the weeks prior to sampling. Two possible pathways that could result in low 15N values for sugarcane leaves (other than N2 fixation) are proposed; high external N concentrations and foliar uptake of volatilised NH3. The leaf 15N value of sugarcane grown in aerated solution culture was shown to decrease by approximately 5 with increasing external N concentration (0.5–8.0 mM), with both NO3 and NH4 + nitrogen forms. Foliar uptake of atmospheric NH3 has been shown to result in depleted leaf 15N values in many plant species. Acid traps collected atmospheric N with negative 15N value (–24.45±0.90) from above a field recently surface fertilised with urea. The 15N of leaves of sugarcane plants either growing directly in the soil or isolated from soil in pots dropped by 3.00 in the same field after the fertiliser application. Both the high concentration of external N in the root zone (following the application of N-fertilisers) and/or subsequent foliar uptake of volatilised NH3 could have caused the depleted leaf 15N values measured in the sugarcane crops at these sites.  相似文献

Container-grownEuphorbia lathyris plants were treated with foliar sprays of various combinations of BA and GA4+7 or 0–3600 mg L–1 Promalin (11 BA + GA4+7) in separate experiments. GA4+7 and Promalin stimulated plants to grow taller. BA and Promalin promoted axillary shoot growth. Multiple applications of Promalin stimulated branching more than single treatments. Dry weight accumulation was stimulated only if the growth regulators were applied to 28–33-cm and not to 56-cm tall plants. Chemical names used: (1, 2, 4a, 4b, 10)-2,4a,7-trihydroxy-1-methyl-8-methylenegibb-3-ene-1,10-dicarboxylic acid 1,4a-lactone (GA4+7),N-(phenylmethyl)-H-purin-6-amine (BA), and Promalin [11 (wt/wt) GA4+7 and BA].The use of the name Promalin or other trade names does not imply endorsement to the exclusion of other products or vendors that may also be suitable.  相似文献

The short-term acclimation (10-d) of Norway spruce [Picea abies (L.) Karst] to elevated CO2 concentration (EC) in combination with low irradiance (100 mol m–2 s–1) resulted in stimulation of CO2 assimilation (by 61 %), increased total chlorophyll (Chl) content (by 17 %), significantly higher photosystem 2 (PS2) photochemical efficiency (Fv/Fm; by 4 %), and reduced demand on non-radiative dissipation of absorbed excitation energy corresponding with enhanced capacity of photon utilisation within PS2. On the other hand, at high cultivation irradiance (1 200 mol m–2 s–1) both Norway spruce and spring barley (Hordeum vulgare L. cv. Akcent) responded to EC by reduced photosynthetic capacity and prolonged inhibition of Fv/Fm accompanied with enhanced non-radiative dissipation of absorbed photon energy. Norway spruce needles revealed the expressive retention of zeaxanthin and antheraxanthin (Z+A) in darkness and higher violaxanthin (V) convertibility (yielding even 95 %) under all cultivation regimes in comparison with barley plants. In addition, the non-photochemical quenching of minimum Chl a fluorescence (SV0), expressing the extent of non-radiative dissipation of absorbed photon energy within light-harvesting complexes (LHCs), linearly correlated with V conversion to Z+A very well in spruce, but not in barley plants. Finally, a key role of the Z+A-mediated non-radiative dissipation within LHCs in acclimation of spruce photosynthetic apparatus to high irradiance alone and in combination with EC was documented by extremely high SV0 values, fast induction of non-radiative dissipation of absorbed photon energy, and its stability in darkness.  相似文献

We investigated the natural abundance of 15N in sun leaves and other components of tropical rain forests on altitudinal sequences of eight sites that form a gradient of soil N availability with varying ectomycorrhizal abundances on Mt. Kinabalu, Borneo. We investigated how soil N availability and ectomycorrhizal abundance related to the 15N abundance of ecosystem components. 15N values (15N abundance relative to 14N) increased consistently in the following order at each site: sun leaves, leaf litter, fine roots and from shallower organic to deeper mineral soil horizons. Enrichment (3–6 15N) of 15N occurred at the litter–topsoil interface at all sites, and the magnitude of the enrichment correlated negatively with 15N depletion in the foliage, irrespective of ectomycorrhizal abundance. Foliar 15N values significantly positively correlated with their N concentrations. Foliar (and litter and root) 15N values correlated positively with NO3 availability, and negatively with NH4 availability. The two positive correlations of foliar 15N with foliar N and NO3 availability were inconsistent with the assumption that stronger nitrification (hence a greater nitrate availability) produced a more 15N-depleted active inorganic N pool. The isotopic fractionation during the passage of N through ectomycorrhizas to plants might explain the positive correlation of foliar 15N and N concentration; however, this mechanism could not fully explain the correlation in our case because strong foliar 15N depletions occurred at the sites that lacked ectomycorrhizas. Alternatively, the positive correlation across sites reflected the tightness of N cycling. Strong nitrification and associated isotopic fractionation might have occurred at N-richer sites and the subsequent removal of NO3 from the system could decrease isotopically `lighter' N at these sites.  相似文献

The impact of sustained low external concentrations of NO 3 (0, 10, 100 and 1000 mmol m–3) on plant growth and the relative acquisition of N through N2 fixation and NO 3 uptake by established, nodulated white clover (Trifolium repens L. cv. Blanca) was studied over 28 days in flowing solution culture. Nitrogen fixation was measured by N difference and 15N dilution methods. Plants supplied with NO 3 achieved higher relative growth rates (% MathType!MTEF!2!1!+-% feaafiart1ev1aaatCvAUfeBSjuyZL2yd9gzLbvyNv2CaerbuLwBLn% hiov2DGi1BTfMBaeXatLxBI9gBaerbd9wDYLwzYbItLDharqqtubsr% 4rNCHbGeaGqiVu0Je9sqqrpepC0xbbL8F4rqqrFfpeea0xe9Lq-Jc9% vqaqpepm0xbba9pwe9Q8fs0-yqaqpepae9pg0FirpepeKkFr0xfr-x% fr-xb9adbaqaaeGaciGaaiaabeqaamaabaabaaGcbaGabmiEayaara% aaaa!3702!\[\bar x\]=0.091 d–1) compared with control plants dependent on N2 fixation (0.073 d–1). Nitrate plants showed progressive increases in shoot: root d.w. ratios from 4 to 6.5–7.6 between days 0–28, compared with 5.1 on day 28 for control plants. Increases in both nodule d.w. and numbers per plant were inhibited after day seven at all concentrations of NO 3 . The severity of inhibition of N2 fixation increased with increasing NO 3 concentration and with time. The total amounts of N2 fixed per plant between days 0–7 after supplying 10, 100 and 1000 mmol m–3 NO 3 , respectively, were 37–39, 28–30 and 0–13%, of the total N acquired. Between days 7–28 the proportional contributions of N2 fixation to total N acquisition declined to 3, 0.5 and 0%, respectively, in these treatments. The corresponding mean specific rates of N2 fixation between days 0–7 were, respectively, 5.4, 3.2, and 2.0 mmol N d–1 g–1 nodule d.w., compared with 7.9 mmol N d–1 g–1 nodule d.w. for zero NO 3 plants. There was no evidence of a transitory increase in N2 fixation following the addition of NO 3 , even at the lowest supply concentration.  相似文献

The responses of photosynthesis, Rubisco activity, Rubisco protein, leaf carbohydrates and total soluble protein to three carbon dioxide treatments were studied in winter wheat [Triticum aestivum (L.)] and barley [Hordeum vulgare (L.)]. Barley and wheat plants were grown in small field plots during 1995 and 1996 in clear, acrylic chambers (1.2–2.4 m2) and were provided with continuous carbon dioxide fertilization at concentrations of 350, 525 and 700 mol mol–1. Photosynthetic rates of barley penultimate leaves and wheat flag leaves measured at growth carbon dioxide concentrations decreased with leaf age in all three CO2 treatments during 1995 and 1996. Photosynthetic acclimation to elevated CO2 was observed on seven of eight measurement dates for barley and ten of eleven measurement dates for wheat over both years. Initial Rubisco activity, total soluble protein and Rubisco protein in barley penultimate leaves and wheat flag leaves also decreased with leaf age. Total Rubisco activity was not used because of enzyme degradation. There was a significant CO2 treatment effect on initial Rubisco activity, total soluble protein and Rubisco protein for wheat in 1995 and 1996 and for barley in 1995. Responses of barley penultimate leaf Rubisco activity and leaf protein concentrations to elevated carbon dioxide were nonsignificant in 1996. A significant CO2 treatment effect also was detected when means of Rubisco activity, soluble protein and Rubisco protein for wheat flag leaves were combined over harvests and years. These three flag leaf parameters were not significantly different in the 350 and 525 mol mol–1 CO2 treatments but were decreased during growth in 700 mol mol–1 CO2 relative to the other two CO2 treatments. Ratios of photosynthesis at 700 and 350 mol mol–1 were compared to ratios of Rubisco activity at 700 and 350 mol mol–1 using wheat flag leaf data from 1995 and 1996. Regression analysis of these data were linear [y = 0.586 + 1.103t x (r2 = 0.432)] and were significant at P 0.05. This result indicated that photosynthetic acclimation was positively correlated with changes of initial Rubisco activity in wheat flag leaves in response to CO2 enrichment. Effects of elevated CO2 on wheat leaf proteins during 1995 and 1996 and on barley during 1995 were consistent with an acceleration of senescence.  相似文献

In order to investigate the effect of N form on dry matter (DM) formation and water uptake rate, French bean (Phaseolus vulgaris L. `Sotaxa') plants were grown with a split-root system. Three treatments were compared: sole nitrate (NO 3) supply (NN), sole ammonium (NH+ 4) supply (AA) and spatially separated supply of NO 3 and NH+ 4 (NA). The pH of the nutrient solutions was kept constant at 6.3 using a pH-stat system. 9 days after onset of the treatments, NN plants had higher root (36%) and shoot dry matter (11%) than AA plants. N form drastically influenced partitioning of assimilates: in the NA treatment, the root half exposed to NO 3 revealed a 170% higher DM than the root half exposed to NH+ 4. N form affected stable carbon-isotope discrimination () of leaf tissue. In leaves of plants which were supplied with NH+ 4 (AA; NA) was significantly more negative (–29.4, –29.6) than in NN treatment (–28.2). We explain this effect by differences in stomatal conductance. We suppose that the significantly less negative of root tissue under NH+ 4 supply is most probably related to higher PEP-case activity. The water uptake rate was higher in NN than in AA grown plants. This effect was found in both, short- and long-term experiments. In case of NA plants, the water uptake in the root part being exposed to NO 3 was 104% higher than in those receiving NH+ 4. At least in the case of the NA treatment we can exclude shoot growth effects as being responsible for differences in water uptake. We therefore assume that differences in root hydraulic conductivity are responsible for the observed effects.  相似文献

Pyracantha (Pyracantha coccinea M. J. Roem. Lalandei) plants were treated with uniconazole at 0.5 mg ai container–1 as a medium drench, 150 mg ai L–1 as a foliar spray, or left untreated. Plants from all treatments were placed under three water regimes: drought acclimated, nonacclimated and later exposed to drought, or nonstressed. Acclimated plants were conditioned by seven 4-day stress cycles (water withheld), while nonacclimated were well watered prior to a single 4-day stress cycle at the same time as the seventh drought cycle of acclimated plants. Nonstressed plants were well watered throughout the study. Nonstressed plants had higher leaf water potentials and leaf conductances than acclimated and nonacclimated plants, and transpiration rates were higher in nonacclimated than acclimated plants. Uniconazole did not affect leaf water potential, leaf conductance, or transpiration rate. Acclimated plants had smaller leaf areas and leaf, stem, and root dry weights than nonacclimated or nonstressed plants. Plants drenched with uniconazole had the lowest stem and root dry weights. Acclimated plants also contained higher N concentrations than nonacclimated or nonstressed plants, and higher P concentrations than nonacclimated plants. Uniconazole medium drench treatments increased levels of Mn and P. Calcium concentration was increased in plants receiving either medium drench or foliar applications.  相似文献

A field lysimeter experiment was conducted over a 406 day period to determine the effect of different soil types on the fate of synthetic urinary nitrogen (N). Soil types included a sandy loam, silty loam, clay and peat. Synthetic urine was applied at 1000 kg N ha-1, during a winter season, to intact soil cores in lysimeters. Leaching losses, nitrous oxide (N2O) emissions, and plant uptake of N were monitored, with soil 15N content determined upon destructive sampling of the lysimeters. Plant uptake of urine-N ranged from 21.6 to 31.4%. Soil type influenced timing and form of inorganic-N leaching. Macropore flow occurred in the structured silt and clay soils resulting in the leaching of urea. Ammonium (NH 4 + –N), nitrite (NO 2 - –N) and nitrate (NO3 -–N) all occurred in the leachates with maximum concentrations, varying with soil type and ranging from 2.3–31.4 g NH 4 + –N mL-1, 2.4–35.6 g NO 2 - –N mL-1, and 62–102 g NO 3 - –N mL-1, respectively. Leachates from the peat and clay soils contained high concentrations of NO 2 - –N. Gaseous losses of N2O were low (<2% of N applied) over a 112 day measurement period. An associated experiment showed the ratio of N2–N:N2O–N ranged from 6.2 to 33.2. Unrecovered 15N was presumed to have been lost predominantly as gaseous N2. It is postulated that the high levels of NO 2 - –N could have contributed to chemodenitrification mechanisms in the peat soil.  相似文献

In this study, we examined the use of the 15N natural abundance method to quantify the percentage N derived from fixation of atmospheric N2 in honeybush (Cyclopia spp.) shrubs and trees in the fynbos, South Africa. Non-fixing shrubs and trees of similar phenology to the Cyclopia species were chosen as reference plants. These reference plants were selected to cover a range of mycorrhizal associations (ericoid mycorrhizal, arbuscular mycorrhizal and non-mycorrhizal). Isotopic analysis revealed a wide range of foliar 15N values for the reference plants, including many very negative values. The marked differences in 15N values were defined by the mycorrhizal status of the reference plant species, with the ericoid and arbuscular mycorrhizal plants showing lower foliar 15N values relative to their non-mycorrhizal counterparts. In contrast, the 15N values of the N2-fixing Cyclopia species were uniformly clustered around zero, from –0.11 to –1.43. These findings are consistent with the observation that mycorrhizal fungi discriminate against the heavier 15N isotope during transfer of N from the fungus to the host plant, leaving the latter depleted in 15N (i.e. with a more negative 15N value). However, a major assumption of the 15N natural abundance method for estimating N2 fixation is that both legume and reference plant should have the same level of fractionation associated with N uptake. But, because mycorrhizal associations may strongly affect the level of fractionation during N uptake and transfer, the test legume should belong to the same mycorrhizal group as the chosen reference plant species. As shown in this study, if the mycorrhizal status of the legume and the reference plant differs, or cannot be assessed, then the 15N natural abundance technique cannot be used to quantitatively estimate N2 fixation.  相似文献

Summary Subterranean clover plants were grown as swards (about 2000 plants/m2) under controlled conditions with N provided either by N2-fixation (NO 3 withheld) or by assimilation of NO 3 (NO 3 supplied). Crop growth rates were measured by dry matter sampling over periods of up to 70 days at PPFD values of 400–1000 mole quanta/m2/s. When NO 3 was supplied from sowing the swards grew more rapidly than when the swards were not supplied with NO 3 and plants had to establish an N2-fixing apparatus. When inter-plant competition was reduced within the sward, a difference in growth rate in favour of NO 3 -fed plants continued for at least 50 days. When however, a closed canopy was allowed to form, the NO 3 -fed swards had more dry weight than the N2-fed swards at the time of canopy closure but thereafter the two swards grew at similar rates at light flux densities of above about 800 mole quanta/m2/s. At light flux densities of about 400 mole quanta/m2/s N2-fed swards had a growth rate 70–80% of that of NO 3 -fed plants. NO 3 -fed plants had a higher organic N content than did N2-fed plants under all conditions.  相似文献

Wild-type (WT) rye (Secale cereale) and barley (Hordeurn vulgare) as well as the barley chlorophyllb-less chlorina F2 mutant were grown under control and intermittent light (IML) conditions. (PSII) in control barley F2 was similar to IML grown WT rye and barley, which, in turn was 2.5 to 3.5 times smaller than for control WT plants. In contrast, PSI was similar for all control plants. This was 2.5 to 4 times larger than for IML-grown WT plants. IML-grown barley mutant plants had the smallest absorption cross-sections. Photosynthetic light response curves revealed that the barley chlorina F2-mutant had rates of oxygen evolution on a per leaf area basis that were only slightly lower than control WT rye and barley while IML-grown plants had strongly reduced photosynthetic performance. Convexity () for control barley chlorina F2-mutants was equal to the WT controls (0.6–0.7), while all IML-grown plants had a of 0. This indicates that, in contrast to control barley mutants, IML-plants were limited by PS II turn-over rates at all irradiances. However, on a per leaf Chl-basis the IML-grown plants exhibited the highest photosynthetic rates. Thus, the comparatively poor photosynthetic rates for IML-grown plants on a per leaf area basis were not due to less efficient photosynthetic reaction centers, but may rather be due to an increased limitation from PS II turn-over and a reduction in the number of reaction centers per leaf area.  相似文献
