Sicklepod (Cassia obtusifolia L.) leaflet epicuticular fatty acid, fatty alcohol, and hydrocarbon contents were measured by gas-liquid chromatography from plants grown under 10-, 12-, 14-, or 16-hour photoperiods and treated with S-(2,3-dichloroallyl)diisopropylthiocarbamate (diallate) (0, 0.14, 0.28, 0.56, 1.12 kilograms per hectare). As diallate concentration increased, epicuticular fatty acid content decreased. Fatty alcohol content was maximal in plants treated with 0.28 kilograms per hectare diallate and was reduced from that level at herbicide concentrations above or below this rate. Hydrocarbon content patterns were similar to those of the fatty alcohols except that the hydrocarbons at 0.28 kilograms per hectare were 61% of that present in the control, whereas the concentration of fatty alcohols increased to 200% of the control in treatments of 0.28 kilograms per hectare diallate.  相似文献

Proserpinaca palustris L. produced juvenile leaves on 8-hour photoperiods, adult leaves on 12-hour photoperiods, and adult leaves and flowers on 14-hour photoperiods. Treatment of plants growing on 8- and 14-hour photoperiods with gibberellic acid caused stem elongation and inhibited flowering. Saltcedar (Tamarix pentandra Pall.) stem cuttings responded vegetatively to various photoperiods of 11-, 14-, 17-, and 8 + 1-hour duration. Total stem length, average stem length, cuticle thickness, and xylem vessel member outside diameter were significantly maximal under a 14-hour photoperiod and less under photoperiods above and below this level. Numbers of stems and roots produced were significantly less in an 11-hour than under a 14-hour photoperiod; but neither was significantly different from responses under 17- and 8 + 1-hour photoperiods. Vegetative photoperiodic responses, except for cuticle thickness, were affected by a photoperiodic after-effect produced by cuttings collected on various dates. Average stem length, total stem length, and number of stems produced were significantly correlated with field photoperiods from November to May.  相似文献

Fatty acids were extracted from saponified vegetative cells and myxospores of Myxococcus xanthus and examined as the methyl esters by gas-liquid chromatography. The acids consisted mainly of C14 to C17 species. Branched acids predominated, and iso-pentadecanoic acid constituted half or more of the mixture. The other leading component (11–28%) was found to be 11-n-hexadecenoic acid. Among the unsaturated acids were two diunsaturated ones, an n-hexadecadienoic acid and an iso-heptadecadienoic acid. No significant differences between the fatty acid compositions of the vegetative cells and myxospores could be detected. The fatty acid composition of M. xanthus was found to be markedly similar to that of Stigmatella aurantiaca. It is suggested that a fatty acid pattern consisting of a large proportion of iso-branched C15 and C17 acids and a substantial amount of an n-16:1 acid is characteristic of myxobacteria.  相似文献

Analysis of fatty acids from the cyanobacterium Cyanothece sp. PCC 8801 revealed that this species contained high levels of myristic acid (14:0) and linoleic acid in its glycerolipids, with minor contributions from palmitic acid (16:0), stearic acid, and oleic acid. The level of 14:0 relative to total fatty acids reached nearly 50%. This 14:0 fatty acid was esterified primarily to the sn-2 position of the glycerol moiety of glycerolipids. This characteristic is unique because, in most of the cyanobacterial strains, the sn-2 position is esterified exclusively with C16 fatty acids, generally 16:0. Transformation of Synechocystis sp. PCC 6803 with the PCC8801_1274 gene for lysophosphatidic acid acyltransferase (1-acyl-sn-glycerol-3-phosphate acyltransferase) from Cyanothece sp. PCC 8801 increased the level of 14:0 from 2% to 17% in total lipids and the increase in the 14:0 content was observed in all lipid classes. These findings suggest that the high content of 14:0 in Cyanothece sp. PCC 8801 might be a result of the high specificity of this acyltransferase toward the 14:0-acyl-carrier protein.  相似文献

In a winter variety of rapeseed, low temperatures enhance fatty acid desaturation as evidenced by 14C-acetate incorporation into fatty acids or 14C-oleate desaturation in vivo; similarly, low temperatures favour polyunsaturated fatty acid accumulation during the maturation of the seeds. Oleate desaturation was slightly higher under 16 hr daylight exposure than under 9 hr treatment.  相似文献

The cyanobacteriumSynechocystis PCC 6803 was grown photoautotrophically in an inorganic medium at constant growth temperatures of 20, 38 (control) or 43°C for 9 h. The up and down-shift of cultivation temperature decreased the growth as measured by culture absorbance and chlorophylla content. However, high temperature slightly increased the oxygen evolution while temperature lower than control inhibited oxygen evolution during the whole incubation period. The protein synthesis studied by14C-labeled protein declined under low temperature by about 50%. The fatty acid pattern is characterized as lacking in C20/C22 acids but containing large amounts of C16 and C18 polyunsaturated fatty acids, 16:2 and 18:3 in particular. The lower temperature increased the percentage of monounsaturated fatty acids while higher temperature increased the saturated fatty acid content in total lipids and lipid classes studied.  相似文献

Etiolated Cucumis sativus L. cotyledons preferentially catabolized exogenous [1-14C]oleic acid and [1-14C]linoleic acid with relatively little incorporation into complex lipids or desaturation of the 14C-labeled fatty acids. Following a 16-hour exposure to light, the greening cotyledons efficiently desaturated the exogenous 14C-labeled fatty acids. A small amount of oleate desaturation to linoleate was observed in etiolated tissue, but hardly any linoleate desaturation to α-linolenate was detected. Both oleate and linoleate desaturation showed diurnal variations with maxima at the end of light periods and minima at the end of dark periods. Illumination of etiolated tissue by flashing light, as opposed to continuous light, failed to stimulate either chlorophyll or α-linolenic acid biosynthesis, and both processes could be halted or reversed by 10 micrograms per milliliter cycloheximide. Production of polyunsaturated fatty acids from [1-14C]acetate, [1-14C]oleic acid, and [1-14C]linoleic acid, by greening cucumber cotyledons, was markedly affected by tissue integrity with finely chopped cotyledons having very little capacity for their synthesis and intact seedlings showing the highest rates.  相似文献

The effect of recalcitrant hydrocarbons on the fatty acid profile from leaf, basal corm, and roots of Cyperus laxus plants cultivated in greenhouse phytoremediation systems of soils from aged oil spill-impacted sites containing from 16 to 340 g/Kg total hydrocarbons (THC) was assessed to investigate if this is a C18:3 species and if the hydrocarbon removal during the phytoremediation process has a relationship with the fatty acid profile of this plant. The fatty acid profile was specific to each vegetative organ and was strongly affected by the hydrocarbons level in the impacted sites. Leaf extracts of plants from uncontaminated soil produced palmitic acid (C16), octadecanoic acid (C18:0), unsaturated oleic acids (C18:1-C18:3), and unsaturated eichosanoic (C20:2-C20:3) acids with a noticeable absence of the unsaturated hexadecatrienoic acid (C16:3); this finding demonstrates, for the first time, that C. laxus is a C18:3 plant. In plants from the phytoremediation systems, the total fatty acid contents in the leaf and the corm were negatively affected by the hydrocarbons presence; however, the effect was positive in root. Interestingly, under contaminated conditions, unusual fatty acids such as odd numbered carbons (C15, C17, C21, and C23) and uncommon unsaturated chains (C20:3n6 and C20:4) were produced together with a remarkable quantity of C22:2 and C24:0 chains in the corm and the leaf. These results demonstrate that weathered hydrocarbons may drastically affect the lipidic composition of C. laxus at the fatty acid level, suggesting that this species adjusts the cover lipid composition in its vegetative organs, mainly in roots, in response to the weathered hydrocarbon presence and uptake during the phytoremediation process.  相似文献

We investigated how dietary fats and oils of different fatty acid composition influence the seasonal change of body mass, fur colour, testes size and torpor in Djungarian hamsters, Phodopus sungorus, maintained from autumn to winter under different photoperiods and temperature regimes. Dietary fatty acids influenced the occurrence of spontaneous torpor (food and water ad libitum) in P. sungorus maintained at 18°C under natural and artificial short photoperiods. Torpor was most pronounced in individuals on a diet containing 10% safflower oil (rich in polyunsaturated fatty acids), intermediate in individuals on a diet containing 10% olive oil (rich in monounsaturated fatty acids) and least pronounced in individuals on a diet containing 10% coconut fat (rich in saturated fatty acids). Torpor in P. sungorus on chow containing no added fat or oil was intermediate between those on coconut fat and olive oil. Dietary fatty acids had little effect on torpor in animals maintained at 23°C. Body mass, fur colour and testes size were also little affected by dietary fatty acids. The fatty acid composition of brown fat from hamsters maintained at 18°C and under natural photoperiod strongly reflected that of the dietary fatty acids. Our study suggests that the seasonal change of body mass, fur colour and testes size are not significantly affected by dietary fatty acids. In cells of Rhodococcus opacus GM-14, GM-29, and 1CP, the contents of branched (10-methyl) fatty acids increased from 3% to 15 to 34% of the total fatty acids when the cells were grown on benzene, phenol, 4-chlorophenol, chlorobenzene, or toluene as the sole source of carbon and energy, in comparison with cells grown on fructose. In addition, the content of trans-hexadecenoic acid increased from 5% to 8 to 18% with phenol or chlorophenol as the carbon source. The 10-methyl branched fatty acid content of R. opacus GM-14 cells increased in a dose-related manner following exposure to phenol or toluene when toluene was not utilized as the growth substrate. The results suggest that 10-methyl branched fatty acids may participate in the adaptation of R. opacus to lipophilic aromatic compounds.  相似文献

The degree of unsaturation of fatty acids was higher in Chlorella vulgaris 11h cells grown with air (low-CO2 cells) than in the cells grown with air enriched with 2% CO2 (high-CO2 cells). The change in the ratio of linoleic acid to α-linolenic acid was particularly significant. This change of the ratio was observed in four major lipids (monogalactosyldiacylglycerol, digalactosyldiacylglycerol, phosphatidylcholine, and phosphatidylethanolamine). The relative contents of lipid classes were essentially the same both in high-CO2 and low-CO2 cells. After high-CO2 cells were transferred to low CO2 condition, total amount of fatty acids remained constant but the relative content of α-linolenic acid increased during a 6-hour lag phase in growth with concomitant decreases in linoleic and oleic acids. When low-CO2 cells were transferred to high CO2 condition, total amount of fatty acids and relative content of oleic acid increased significantly. The amount of α-linolenic acid remained almost constant, while the amounts of palmitic, oleic, and linoleic acids increased. Similar, but smaller, changes in fatty acid compositions were observed in two species of green algae Chlamydomonas reinhardtii and Dunaliella tertiolecta. However, no difference was found in Euglena gracilis, Porphyridium cruentum, Anabaena variabilis, and Anacystis nidulans.  相似文献

The metabolism of lipids, like that of other components, was adversely and strongly affected when rice (Oryza sativa L.) coleoptiles were grown anaerobically. In aerobic coleoptiles, the amounts of total fatty acid, phospholipid, and total lipid per coleoptile increased by 2.5- to 3-fold between days three and seven, whereas under anoxia, the increases were all less than 60%. The total amount of lipid at day seven in anoxia was less than 30% of that in air. In air, the total fatty acid content at day three was 25 nanomoles per coleoptile and this increased to over 71 nanomoles per coleoptile at day seven. All acids except 18:0 showed substantial increases. In anoxia, the corresponding values for total fatty acids were 24 nanomoles and 27 nanomoles. The small increases were confined to the saturated fatty acids; no significant increase occurred in unsaturated fatty acids. A minor fatty acid constituent (16:1) increased from 0.09 to 1.99 nanomoles per coleoptile between days three and seven in air. This component was never observed in any fatty acid preparation from anaerobic coleoptiles. The major phospholipids under all conditions were phosphatidylcholine, phosphatidylethanolamine, phosphatidylinositol, and phosphatidic acid. A small amount of unidentified phosphoester, not present on thin layer chromatography plates from aerobic coleoptiles, was seen in extracts of anaerobic coleoptiles. The fatty acyl substituents of each of the phospholipids were analyzed at days three and seven in coleoptiles grown aerobically and in anoxia. Each phospholipid had its own distinctive fatty acid composition which remained fairly constant under all treatments; 16:0 and 18:2 were the most abundant fatty acids in every phospholipid class. In air, the percentages of total fatty acids that were in the phospholipids were 86% on day three and 87% on day seven. In anoxia, the values at the corresponding ages were 47 and 57%. Since no net synthesis of unsaturated fatty acids occurred in anaerobic conditions, the small increase in total unsaturated acids in the phospholipids between days three and seven must have occurred at the expense of fatty acids preexisting in the neutral lipid. No unusual pathways of biosynthesis or unusual precursors are required to explain the presence of unsaturated fatty acids in the rice coleoptile. The present study and results of experiments where coleoptiles were fed [14C]acetate (BB Vartapetian et al. 1978 Plant Sci Lett 13:321-328) clearly show that unsaturated fatty acid synthesis in rice coleoptiles requires O2, as it does in other plants.  相似文献

Crassulacean acid metabolism (CAM) was investigated in leaves and stems of the succulent C4 dicot Portulaca oleracea L. Diurnal acid fluctuations, CO2 gas exchange, and leaf resistance were monitored under various photoperiod and watering regimes. No CAM activity was seen in well watered plants grown under 16-hour days. Under 8-hour days, however, well watered plants showed a CAM-like pattern of acid fluctuation with amplitudes of 102 and 90 microequivalents per gram fresh weight for leaves and stems, respectively. Similar patterns were also observed in detached leaves and defoliated stems. Leaf resistance values indicated that stomata were open during part of the dark period, but night acidification most likely resulted from refixation of respiratory CO2. In water-stressed plants maximum acid accumulations were reduced under both long and short photoperiods. At night, these plants showed short periods of net CO2 uptake and stomatal opening which continued all night long during preliminary studies under natural environmental conditions. Greatest acid fluctuations, in P. oleracea, with amplitudes of 128 microequivalents per gram fresh weight, were observed in water-stressed plants which had been rewatered, especially when grown under short days. No net CO2 uptake took place, but stomata remained open throughout the night under these conditions. These results indicate that under certain conditions, such as water stress or short photoperiods, P. oleracea is capable of developing an acid metabolism with many similarities to CAM.  相似文献

Recently, microalgae have gained a lot of attention because of their ability to produce fatty acids in their surrounding environments. The present paper describes the influence of organic carbon on the different fatty acid pools including esterified fatty acids, intracellular free fatty acids and extracellular free fatty acids in Ochromonas danica. It also throws light on the ability of O. danica to secrete free fatty acids in the growth medium under photoautotrophic and mixotrophic conditions. Biomass production of photoautotrophically grown O. danica was higher than that of mixotrophically grown, where a cellular biomass formation of 1.8 g L?1 was observed under photoautotrophic condition which was about five folds higher than that under mixotrophic conditions. Contrary, the esterified fatty acid content reached up to 99 mg g?1 CDW under photoautotrophic conditions at the late exponential phase, while during mixotrophic conditions a maximum of 212 mg g?1 CDW was observed at the stationary phase. Furthermore, O. danica cells grown under mixotrophic conditions showed higher intracellular free fatty acid and extracellular free fatty acid contents (up to 51 and 20 mg g?1 CDW, respectively) than cells grown under photoautotrophic conditions (up to 26 and 4 mg g?1 CDW, respectively). The intra- and extracellular free fatty acids consisted of a high proportion of polyunsaturated fatty acids, mainly C18:2n?6, C18:3n?3 and C20:4n?6.  相似文献

Fatty acid compositions in growing and resting cells of several strains of Pseudomonas putida (P8, NCTC 10936, and KT 2440) were studied, with a focus on alterations of the saturation degree, cis-trans isomerization, and cyclopropane formation. The fatty acid compositions of the strains were very similar under comparable growth conditions, but surprisingly, and contrary to earlier reports, trans fatty acids were not found in either exponentially growing cells or stationary-phase cells. During the transition from growth to the starvation state, cyclopropane fatty acids were preferentially formed, an increase in the saturation degree of fatty acids was observed, and larger amounts of hydroxy fatty acids were detected. A lowered saturation degree and concomitant higher membrane fluidity seemed to be optimal for substrate uptake and growth. The incubation of cells under nongrowth conditions rapidly led to the formation of trans fatty acids. We show that harvesting and sample preparation for analysis could provoke the enzyme-catalyzed formation of trans fatty acids. Freeze-thawing of resting cells and increased temperatures accelerated the formation of trans fatty acids. We demonstrate that cis-trans isomerization only occurred in cells that were subjected to an abrupt disturbance without having the possibility of adapting to the changed conditions by the de novo synthesis of fatty acids. The cis-trans isomerization reaction was in competition with the cis-to-cyclopropane fatty acid conversion. The potential for the formation of trans fatty acids depended on the cyclopropane content that was already present.  相似文献

The rate of fatty acid synthesis withStreptomyces aureofaciens was measured by means of incorporation of 1-14C-acetic acid. The maximum specific rate of incorporation was achieved in the 12th hour. The incorporation rate of the labelled acetate into chlortetracycline was roughly twice that into fatty acids during the production phase. The rate of fatty acid synthesis in the high producing strain was five times that of the low producing one, although the dry weight fatty acid content was almost the same in both the strains.  相似文献

Starch accumulation rate in leaves of vegetative soybeans is inversely related to the length of the daily photosynthetic period. However, it is not known whether a similar response would be observed during reproductive growth. Soybeans (Glycine max L. Merr. cv Amsoy 71) were grown to three stages of reproductive growth (beginning seed, mid seed-fill, and late seed-fill) under 12-hour daylengths, and then shifted to 6-hour photosynthetic periods (12-hour photoperiods) for 4 days. One and 4 days after treatment, a mid-canopy leaf was pulsed with 14CO2, and sampled for radiolabeled starch and water-soluble compounds at 0.5, 1, 3, 9, and 21 hours after labeling.

Plants exposed to the 6-hour photosynthetic periods at the beginning seed stage retained and incorporated significantly more label as starch than did those given 12-hour photosynthetic periods. However, plants exposed to the shortened photosynthetic periods at the late seed-fill stage partitioned less label into starch. Plants exposed at mid seed-fill gave a variable response.

Shortened photosynthetic periods resulted in preferential partitioning of recently fixed carbon to the seed at the expense of the pod wall. The results of these experiments suggest that the increased sink demand present during late reproductive growth may be of greater importance in control of leaf starch accumulation than is the length of the daily photosynthetic period.

Photoperiods of 10 or 12 h enhanced vegetative propagule production by Potamogeton nodosus Poir. and P. pectinatus L. The proportion of plants producing vegetative propagules and the number and weight of propagules per plant increased over time, and were greatest after 8 weeks. Maximum vegetative propagule production was 12±6 tubers per plant (mean±S.D.; n=7) for P. pectinatus and 5±3 for P. nodosus. Tuber or winter-bud dry weight accounted for 38 or 27% of total plant dry weight, respectively, for plants grown for 56 days under a 10-h photoperiod. Vegetative propagules appeared to be produced instead of new shoot tissue under short-day conditions. Chlorophyll a and carotenoid content were lower for P. nodosus at short photoperiods, but not for P. pectinatus. The changes in chlorophyll a:b and carotenoid:ratio chlorophyll a suggest that synthesis/degradation of these pigments is tightly coupled and that short photoperiods may initiate senescence in P. nodosus.  相似文献
