Brahman (Bos indicus) cows, were selected at 28+/-10 days after calving and analyzed by real time rectal ultrasonography three times a week, in order to evaluate and compare follicular and corpus luteum development during postpartum (PP) anestrus and the first PP estrous cycle under sylvopastoril conditions. Suckling (S, n=11) or non-suckling (NS, n=5) cows were evaluated in a zone of tropical dry forest (450m of altitude, mean temperature=27 degrees C, annual rainfall=1000mm). Estrous detection was performed twice daily by direct observation. Progesterone was quantified using RIA. From 28+/-10 days postcalving to resumption of estrous cycles, there were no differences (P>0.05) between NS and S cows for diameter of the dominant or first subordinate follicle, follicular growth rate, or interdominance interval. Silent ovulation, corpus luteum formation and subsequent progesterone concentrations ranging from 0.3 to 9. 7ng/ml, were found in both groups. The first calving to ovulation and calving to standing estrus intervals were shorter (P<0.01) in NS (34.8+/-5.81 and 41.2+/-9.03 days) than in S (65+/-4.82 and 81+/-6. 21 days) cows. Follicular development and progesterone concentrations during the first PP estrous cycle did not differ (P>0. 05) between NS and S cows. These results suggest that Brahman cows could have an early PP resumption of follicular recruitment if fed under sylvopastoril system conditions. However, non-suckled cows did have an earlier standing estrus and ovulation than did suckled cows.  相似文献

The effects of dietary energy and suckling on adrenal function and luteinizing hormone (LH) concentrations were investigated in primiparous postpartum cows. Ten heifers were assigned at calving to either high (22.8 Mcal/day) or low (15.2 Mcal/day) energy diets. Blood samples were collected every 15 minutes for 8 hours on 28, 42, and 56 days post partum. Calves were allowed to suckle ad libitum during sampling periods. Serum samples were analyzed by radioimmunoassay for LH and cortisol. Concentrations of catecholamines were quantified by reverse-phase HPLC. Body weights were decreased (P<0.01) by low energy intake. In addition, low energy diet cows had lower mean LH concentrations (0.97 +/- 0.09 vs 1.57 +/- 0.07 ng/ml), P<0.05) than high energy diet cows. Luteinizing hormone concentrations in high energy diet cows increased with days post partum, resulting in a treatment-by-time interaction (P<0.005). Treatment did not affect mean cortisol concentrations. However, within 15 minutes of suckling cortisol release was significantly above baseline in 77% of the observed suckling events. Dihydroxyphenylalanine (DOPA) increased in high energy diet cows compared with that of low energy diet cows (2,833 +/- 243 vs 1,294 +/- 243 pg/ml, P<0.01). Norepinephrine (NE) and 3,4-dihydroxyphenylacetic acid (DOPAC) were not influenced by treatment. Plasma NE decreased during the postpartum interval (P<0.005). These data suggest that reduced energy intake may prevent the increase in LH associated with increasing days post partum and alter adrenal function. In addition, spontaneous suckling events elicit a release of cortisol.  相似文献

This study was conducted to determine relationships between the uterine microflora and reproductive characteristics of dairy cows. Uterine lumina were swabbed during estrus immediately prior to artificial insemination (A I). Swabbings were cultured for bacteria and accessed for cytologic evidence of inflammation. The time of sampling averaged 104 d post partum. Bacteria in the uterus were cultured from 65% of the cows (n = 85). The number of cows with any given microbial genus ranged from 1 to 35 and the number of different genera per cow ranged from 0 to 8. There were on significant correlations between amount and type of uterine microflora, days post partum, numbers of observed estrus, conception rate and uterine inflammation. The number of observed estrus periods was not correlated with the presence of aerobic bacteria. No significant relationships were found between microbes and uterine inflammation or conception to the A.I. service at the time of sampling. A negative correlation between uterine inflammation and conception approached significance (r=-0.24, P > 0.10). Acquisition of uterine lumen samples by the technique utilized had no effect on conception to A I service.  相似文献

Characteristics of the intrafollicular environment to which the preovulatory oocyte is exposed may be one of the major factors determining subsequent fertility. The aim of our study was to examine to what extent metabolic changes that occur in early post partum high-yielding dairy cows are reflected in the follicular fluid (FF) of the dominant follicle (>8 mm). Nine blood samples were taken per cow from nine high-yielding dairy cows between 7 days before and 46 days after parturition. From Day 14 post partum on and together with blood sampling, FF samples of the largest follicle were collected from the same cows by means of transvaginal follicle aspiration. Serum and FF samples were analyzed using commercial clinical and photometric chemistry assays for glucose, beta-hydroxybutyrate (beta-OHB), urea, total protein (TP), triglycerides (TG), non-esterified fatty acids (NEFA) and total cholesterol (TC). All cows lost body condition during the experimental period (0.94+/-0.09 points) illustrating a negative energy balance during the experimental period. In FF, glucose concentrations were significantly higher and the TP, TG, NEFA and TC concentrations were significantly lower than in serum (P<0.05). The concentrations of glucose, beta-OHB, urea and TC in serum and in FF changed significantly over time (P<0.05). Throughout the study, changes of all metabolites in serum were reflected by similar changes in FF. Especially for glucose, beta-OHB and urea the correlations were remarkably high. The results from the present study confirm that the typical metabolic adaptations which can be found in serum of high-yielding dairy cows shortly post partum, are reflected in follicular fluid and, therefore, may affect the quality of both the oocyte and the granulosa cells.  相似文献

The pattern of follicular growth was studied in 17 suckled zebu cows with average body condition and under extensive management in a tropical environment (23 degrees C, 78% humidity; 2200 mm annual rainfall; 1000 m altitude). The study covered the period from parturition to weaning at 12 months postpartum (PP). Data were collected by transrectal ultrasonography (7.5 MHz) at 48 h intervals, and progesterone (P4) measurements were performed by RIA. The sequential development of ovarian follicles greater than 4 mm was followed until regression or ovulation. Ovarian activity as characterized by growth and regression of follicles of 4 to 6 mm, with sporadic dominance, and a long interdominance interval was observed in every cow and from as early as 26 +/- 2 days PP. This follicular pattern was highly variable during the first 6 months: cows presented 2 to 20 follicular waves (FW) in which a dominant follicle (DF) grew to 8 +/- 1 mm with daily growth rates of 1.1 +/- 0.5 mm/day. The duration of dominance varied from 2 to 8 days and the interdominance time interval was 0 (overlapped waves) to 60 days. Neither behavioural oestrus nor ovulation was observed during this period. From 6 to 12 months PP, cows presented 7 to 20 FW, some with ovulation and/or corpora lutea (CL) formation. The ovulation was preceded by oestrus in some cases (43%). The mean (+/- sem) diameter of DF was 9 +/- 2.7 mm, their mean growth rate 1.4 +/- 0.2 mm/day, their duration of dominance was 2 to 8 days and the interdominance interval was 0 to 14 days. Progesterone concentrations (P4) from 1.0 to 13 ng/ml were found when a CL was present. Once cyclicity re-commenced at 217 to 278 days PP, the cows presented either normal (21 +/- 3 days), short (10 +/- 2 days), or long (50 +/- 4 days) cycles. The resumption of cyclicity was characterized by an increased frequency of emerging follicular waves. Under the conditions of this study, the suckled Bos indicus cows re-commenced ovarian follicular activity as early as described in B. taurus breeds, but the establishment of cyclicity was substantially later. These data add further to the panorama of postpartum reproductive physiology in tropical cattle.  相似文献

Seasonal effects on luteal activity during post partum were evaluated in two consecutive studies in 253 dairy cows in Northern Italy. In study 1, plasma progesterone concentrations were determined on days 14, 21, 28, 35, 42, 49 and 56 post partum and in study 2 cows were synchronized and inseminated at a fixed time using two regimes based on the 'Ovsynch' protocol. Study 1: Animals were classified as luteal (progesterone >1.5 ng/ml in at least two consecutive samples) or non-luteal (progesterone <1.5 ng/ml in all samples). The proportion of cows without luteal activity from calving to day 56 post partum was 47/253 (18.5%). Of the 47 cows without luteal activity, 42 (89%) were detected during the warm months of the year and five were detected during the cold months of the year, and the effect of season was highly significant (P < 0.001). Study 2: Three study groups were established; control (CONT, untreated cows, n = 92), GPG (cows receiving gonadotropin-releasing hormone (GnRH) on day 0, PGF2α on day 7 followed by a second dose of GnRH 24 h later, n = 80); and HPH (the same as the GPG group, but with human chorionic gonadotropin (hCG) substituted for GnRH, n = 81). In the GPG and HPH groups, cows were inseminated 16 to 22 h after the second GnRH or hCG injection. Untreated cows were inseminated at first estrus after a voluntary weaning period. Because the effects of the GPG and HPH regimes on pregnancy rate were not significantly different, data were pooled into a single treatment group (TREAT). Pregnancy rates during the warm months of the year were 16% and 15% at first service and 65% and 66% at day 135 post partum for CONT and TREAT groups, respectively. Pregnancy rates during the cold months of the year were 36% and 38% at first service and 72% and 76% at day 135 post partum for CONT and TREAT groups, respectively. There was an effect of season (P < 0.05) but not of treatment on pregnancy rate. Treatment reduced the number of days from calving to conception during both the cold (101 ± 3.2 v. 121 ± 3.1 days; P < 0.001) and warm seasons (122 ± 3.2 v. 145 ± 3.1 days; P < 0.001). In conclusion, the present study shows that (i) heat stress during the warm season can compromise luteal activity and (ii) that regimes based on the Ovsynch protocol did not improve pregnancy rate at first service or by 135 post partum, but they had a positive effect on the calving-to-conception interval.  相似文献

The primary objective was to determine the effect of supplemental progesterone, administered via an intravaginal device (CIDR), on conception rates to timed-artificial insemination (timed-AI) in postpartum suckled Japanese Black beef cows treated with the Ovsynch protocol. A secondary objective was to compare the effects of treatments on plasma concentrations of progesterone and estradiol. Cows in the control group (Ovsynch, n=38) received a standard Ovsynch protocol (100 microg GnRH analogue on Day 0, 500 microg PGF2alpha analogue on Day 7, and 100 microg GnRH analogue on Day 9), with AI on Day 10, approximately 20 h after the second GnRH treatment. Cows in the treatment group (Ovsynch+CIDR; n=40) received a standard Ovsynch protocol plus a CIDR for 7 days (starting on Day 0). Plasma progesterone concentrations were determined on Days 0, 1, 7, 9, 10, and 17 and plasma estradiol-17beta concentrations were determined on Days 7, 9, 10, and 17. The odds ratio for likelihood of conception was 3.29 times greater (P=0.02) in the Ovsynch+CIDR group compared to Ovsynch group. The conception rate was greater (P=0.03) in the Ovsynch+CIDR group than in the Ovsynch group (72.5% versus 47.7%). Insertion of a CIDR device significantly increased plasma progesterone concentrations only on Days 1 and 7 (P<0.001 and P=0.05, respectively), but had no significant effect on plasma estradiol-17beta concentrations. Including a CIDR with the Ovsynch protocol significantly improved conception rates in postpartum suckled Japanese Black beef cows.  相似文献

32 of 35 sexually mature CFHB rats paired with proven males and thereafter allowed to mate post partum and nurse young concurrent with pregnancy, produced a total of 227 litters. The proportion of females producing litters decreased both with increasing age and parity, and with the number of young nursed. Litter size declined both as age and parity increased, and when gestation was extremely prolonged. The incidence and extent of delayed implantation (gestations greater than 25 days) were affected by the number of young nursed but not by increasing age and parity. There was no evidence for the existence of implantation periods coinciding with multiples of oestrous cycles.  相似文献

An epidemiological study of risk factors for postpartal ovarian disturbances was carried out on 334 high-yielding dairy cows in 6 well-managed Belgian herds. Ovarian activity was closely monitored using progesterone profiles, based on twice weekly RIA-analysis for progesterone in milk fat, starting at 10 d after calving and continuing until the confirmation of a new pregnancy. Attention was focused on abnormal cyclicity during the preservice, postpartum period; cows were divided into 6 different categories. Three of these categories (normal profile, delayed cyclicity, and prolonged luteal phase) were of major importance and were analyzed using a multiple variable logistic regression model. Season of calving (stable vs pasture, odds ratio (OR)=5.7), an extended length of the previous dry period (> 77 vs < or = 63 d, OR=2.9), problem calvings (OR=3.6), abnormal vaginal discharge (OR=4.5), health problems during the first month of lactation (clinical disease, OR=5.4; ketosis, OR=11.3), and clinical parameters illustrating the appearance of a severe negative energy balance significantly increased the risk for delayed cyclicity before service. Parity (> or = 4 vs 1, OR=2.5), problem calvings (OR=2.9), occurrence of puerperal disturbances (OR ranged from 3.5 to 11.0), health problems during the first month of lactation (OR=3.1), and an early resumption of ovarian cyclicity after calving (< 19 d vs > 32 d, OR=2.8) increased the risk for prolonged luteal cycles before service.  相似文献

It was the aim of this investigation to study the combined effect of superovulation and non-surgical recovery on the fertility of the donor animals. Injection of a prostaglandin analogue at the day of collection (days 6–8 after standing heat), significantly shortened the super-ovulatory estrus cycle (21.3 ± 10.0 days), when compared with untreated donor animals (38.7 ± 15.1 days). The prostaglandin treatment, however, also led to more irregular estrous cycles, resulting in a lower pregnancy rate after first insemination (47%) and a need for more inseminations per conception (2.00 ± 1.00), than in animals not treated with prostaglandin analogue (92.3% and 1.06 ± 0.29 AI/conception). For all animals these parameters were 66.6% and 1.60 ± 0.99 AI/conception). The average time from calving to pregnancy was 143.1 ± 34.3 days, slightly longer for prostaglandin treated (148.2 ± 39.4 days) than for untreated animals (137.3 ± 22.8 days). One animal developed endometritis and one had adhesions in the bursa ovarii. It was found that most donors attained normal fertility, and that superovulation was more likely to affect the fertility (abnormal cyclicity, early embryonic mortality e.g.) than the flushing of the uterus. Treatment with a prostaglandin analogue at the day of collection did not improve the subsequent fertility.  相似文献

Doses of 125, 250 or 500 micrograms LH-RH were injected i.m. into suckled beef cows on approximately Day 11 of an oestrous cycle synchronized by prostaglandin treatment. There was a positive linear relationship between dose of LH-RH and the area under the measured LH peak. Administration of 500 micrograms LH-RH as a single injection to suckled cows 13-32 days post partum resulted in LH release but failed to induce normal ovarian activity. A small transient rise in plasma progesterone for 6--9 days occurred at the expected time after injection in 50% of animals. Administration of 500 micrograms LH-RH to suckled beef cows approximately 20--30 days post partum and a second injection approximately 10 days later at the time when the resulting transient rise in plasma progesterone had returned to basal values induced normal cyclic activity (as shown by progesterone concentrations and observed oestrus) at 35 days compared with 70 days for untreated controls. Pituitary responsiveness to LH-RH, as assessed by LH levels, was found to increase up to 20 days post partum.  相似文献

A total of 402 two suckled postpartum beef cows at 2 locations (CSU and ECRC) were utilized in 3 trials to evaluate the effectiveness of a combination melengestrol acetate (MGA)-PGF2alpha estrus synchronization system in spring-calving cows. The cows were allocated by days post partum, body condition score (1 = emaciated; 9 = obese), sire breed, and dam age at the beginning of treatment to 1 of 2 treatments within location and trial: MGA-PGF2alpha (0.5 mg MGA/head/d for 14 d with 25 mg of PGF2alpha injected 17 d after MGA withdrawal), and unsynchronized controls. All cows were observed for estrus at 12-h intervals for at least 5 d post injection. Cows observed in standing estrus were inseminated 12 to 18 h later. There was a location effect on response to treatment that was attributed to differences in body condition score between locations so data were analyzed within a location. Body condition score at the CSU location was 5.7 compared with 4.0 at the ECRC location. The CSU MGA-PGF2alpha treated cows had higher (P < 0.05) 5-day estrus and 5-d pregnancy rates (78.6 and 61.0%, respectively) than the CSU controls (11.1 and 6.9%, respectively). Similar results for 5-d estrus and pregnancy rates but of decreased magnitude were also observed for the ECRC MGA-PGF2alpha treated (31.6 and 21.4%, respectively) cows compared with that of the ECRC controls (11.9 and 8.5%, respectively). The CSU MGA-PGF2alpha treated cows had higher (P < 0.05) 25- and 60-d pregnancy rates (82.5 and 94.8%) than the CSU controls (65.3 and 87.5%). The 25- and 60-d pregnancy rates were similar between the ECRC MGA-PGF2alpha treated cows and ECRC control cows. The MGA-PGF2alpha estrus synchronization system appears to contribute to pregnancy early in the breeding season in postpartum beef cows, although its effectiveness is limited by cow body condition.  相似文献

Calcium content and distribution of the 25-day pregnant (PR) and post partum (PP) rabbit uterus was studied by atomic absorption spectrophotometry and 45Ca determination. Total Ca content [2.28 +/- 0.28 (PR) and 2.19 +/- 0.12 (PP) mM/kg wet wt] extracellular [1.21 +/- 0.09 (PR) and 1.25 +/- 0.11 (PP) mM/kg wet wt] cellular [1.07 +/- 0.08 (PR) and 0.94 +/- 0.09 (PP) mM/kg wet et], total exchangeable [1.86 +/- 0.11 (PR) and 1.84 +/- 0.09 (PR) mM/kg wet wt] and inexchangeable [0.43 +/- 0.05 (PR) and 0.35 +/- 0.04 (PP) mM/kg wet wt] Ca fractions were identical in the two extreme endocrinological conditions. In contrast compartment size and rate constant of different exchangeable Ca fractions determined by kinetic analysis of 45Ca desaturation "urves (curve-peeling tecnique and computer method), revealed significant differences between PR and PP uteri. Two exchangeable phases could be identified in both endocrinological states. The rate constants of both phases of efflux were significantly higher in the PP (alpha 1 = 0.173 +/- 0.02 min-1; alpha 2 = 0.023 +/- 0.001 min-1) than in the PR uterus (alpha 1 = 0.099 +/- 0.01 min-1; alpha 2 = 0.018 +/- 0.01 min-1). Compartment size of phase 1 (fast component) was significantly higher in the PR (1.13 +/- 0.1 mM/kg wet wt) than in the PP uterus (0.77 +/- 0.06 mM/kg wet wt). In contrast, compartment size of phase 2 (slow component) was significantly smaller in PR than in PP uterine strips (0.74 +/- 0.06 and 1.08 +/- 0.11 mM/kg wet wt). The last portion of desaturation curves represents efflux from one homogenous compartment. The present results suggest that endocrinological control of the rabbit myometrium is linked to the regulation of the binding of a superficial exchangeable Ca fraction.  相似文献

Ovarian activity and uterine involution were monitored by rectal palpation, oestrus detection and plasma progesterone analysis from 3 to 4 days to approximately 150 days post partum in 38 suckled swamp buffaloes (Bubalus bubalis). The intervals from parturition to regression of the corpus luteum (CL) of pregnancy and involution of the uterus were 10 and 28 ± 6 (S.D.) days respectively. First detected oestrus and first elevation of plasma progesterone (> 0.7 ng/ml) occurred at 88 ± 26 and 96 ± 22 days in 8 and 12 buffaloes respectively. During the first 150 days post partum, 26 of 38 suckling buffaloes (68%) were acyclic (anoestrus) and of 12 animals (32%) exhibiting ovarian cycles, 4 were not detected in oestrus. The tentative diagnosis, based on rectal palpation, that CL were present between days 30 and 90 after parturition (without concurrent luteal levels of progesterone in plasma) suggests that confirmation should be by laparoscopy. It is concluded that a delay in the resumption of ovarian cyclicity post partum represents an important factor contributing to the prolonged calving to conception interval in the suckled swamp buffalo.  相似文献

The objective was to determine the effect of sequence of insemination after simultaneous thawing of multiple 0.5 mL semen straws on conception rate in suckled multiparous Nelore cows. The effect of this thawing procedure on in vitro sperm characteristics was also evaluated. All cows (N = 944) received the same timed AI protocol. Ten straws (0.5 mL) of frozen semen from the same batch were simultaneously thawed at 36 °C, for a minimum of 30 sec. One straw per cow was used for timed AI. Frozen semen from three Angus bulls was used. Timed AI records included sequence of insemination (first to tenth) and time of semen removal from thawing bath. For laboratory analyses, the same semen batches used in the field experiment were evaluated. Ten frozen straws from the same batch were thawed simultaneously in a thawing unit identical to that used in the field experiment. The following sperm characteristics were analyzed: sperm motility parameters, sperm thermal resistance, plasma and acrosomal membrane integrity, lipid peroxidation, chromatin structure, and sperm morphometry. Based on logistic regression, there were no significant effects of breeding group, body condition score, AI technician, and sire on conception rate, but there was an interaction between sire and straw group (P = 0.002). Semen from only one bull had decreased (P < 0.05) field fertility for the group of straws associated with the longest interval from thawing to AI. However, the results of the laboratory experiment were unable to explain the findings of the field experiment. Sperm width:length ratio of morphometric analysis was the single sperm characteristic with a significant interaction between sire and straw group (P = 0.02). It was concluded that sequence of insemination after simultaneous thawing of 10 semen straws can differently affect conception rates at timed AI, depending on the sire used. Nevertheless, the effects of this thawing environment on in vitro sperm characteristics, remain to be further investigated.  相似文献
