We performed a short-term in vitro cultivation of interna fragments of the rhizocephalan barnacleSacculina polygenea, a parasite of the shore crabHemigrapsus sanguineus, and a histological study of an interna isolated from the host organism. It is shown that the colonial interna ofS. polygenea includes an extensive trophic system (a “root” system), which performs the functions of absorption, accumulation, and transportation of nutrients, and a reproductive system represented by numerous externa primordia at different stages of development. Vitellogenic oocytes were found in the ovary of the late primordia of externae. Thus, we observed the colonial structure of interna inS. polygenea with numerous elements of the reproductive and trophic systems and characteristic signs of fractal organization.  相似文献

We performed histological studies on trophic and reproductive systems of colonial interna in Sacculina polygenea, a parasite of the coastal crab Hemigrapsus sanguineus. The trophic system that performs functions of absorption, accumulation, and transportation of nutrients from the hemolymph of the host comprises the trophic epithelium of distal canals and transporting trunks. The reproductive system of interna consist of nuclei (early stages of development of the primordia of externae) and the primordia of externae in later stages of development. It has been shown that during morphogenesis of the nucleus two primordia arise, a primordium of the externa itself and a primordium of its trophic system. In the primordium of the ovary, we found oogonia; early oocytes and vitellogenic oocytes were found in the ovaries of the late primordia of the externae. The damaging effects of the interna on the ovaries and testes of the crab host are discussed. Thus, we have found numerous elements of reproductive and trophic systems in the colonial interna of S. polygenea. The term individual is proposed to be used for the externa in rhizocephalan barnacles with its trophic system.  相似文献

Over the past several decades, global warming has been linked to shifts in the distributions and abundances of species. In the southern North Sea, temperatures have increased in the last three decades and this will likely have consequences on the seasonality of marine organisms living in the area. Ctenophores such as Beroe gracilis and Pleurobrachia pileus could be particularly affected by changes in their own phenology and that of their prey, thus causing shifts in ecosystem function. Despite their global relevance and their potentially deleterious effect on the fishing industry, only a few long‐term records of ctenophore abundance exist, and most of these records are semiquantitative in nature. Therefore, our knowledge of the influence of environmental factors on their population development is presently very limited. In this study, the long‐term abundance dynamics of B. gracilis, P. pileus and their food calanoid copepods were analysed for a highly temporally resolved time series in the German Bight at Helgoland Roads. Special attention was focused on the response of these organisms to climate warming. Bayesian statistics showed that the phenology of the two ctenophores shifted in a step‐like mode in the year 1987/1988 to permanent earlier appearances. The seasonal change in the population blooms of P. pileus and B. gracilis correlated remarkably well with a step‐like increase in winter and spring sea surface temperatures of the southern North Sea. Possible explanations for the changes observed in these organisms include higher reproductive rates, increased winter survival rates or both. Interannual variations in ctenophore abundances correlated best with the interannual changes in spring temperatures, although the impact of temperature on B. gracilis appeared less pronounced. The changes in copepods abundance were not consistent with changes in P. pileus and B. gracilis. P. pileus showed longer periods of high abundance after the permanent seasonal advancement. These longer periods were correlated with a decline in the average autumn abundance of copepods. Changes in the phenology of these organisms raise the concerns on the declining state of fish stocks, which could potentially be exacerbated by gelatinous zooplankton outbreaks. These conditions may ultimately lead to trophic dead ends by channelling the flow of energy away from higher trophic levels.  相似文献

Marine planktonic algae are frequently exposed to metallic contaminants. Because heavy metals can be assimilated and accumulated by algal cells, they can then be transferred to higher trophic levels of food chains. We studied the effects of cadmium on protein production and the growth of the marine prasinophyte Tetraselmis gracilis (Kylin) Butcher. By means of toxicological assays, we estimated the LC50 of cadmium as 3.2 ppm and 1.8 ppm after 48 h and 96 h of exposure to this heavy metal, respectively. The growth curves and survival percentages of cell cultures in the presence of cadmium were determined, and a proportional reduction of both parameters with increasing metal concentrations was found. When chronically exposed to sublethal concentrations of cadmium, T. gracilis contained high levels of superoxide dismutase (SOD) activity, one of the main enzymes of the cell's antioxidant defense mechanism. Under these growth conditions, total SOD activity in crude extracts was increased by 41% (at 1.5 ppm) and 107% (at 3.0 ppm). Assays of SOD activity in nondenaturing polyacrylamide gels also showed a similar induction by cadmium. These results show that cadmium has potentially toxic properties since it significantly inhibited the growth of T. gracilis at low concentrations and promoted the induction of SOD activity, suggestive of an oxidative stress state. Besides being the first report of SOD in T. gracilis, this work describes experimental evidence of SOD induction by cadmium in this species.  相似文献

The capability of granulosa and theca interna cells, from preovulatory follicles of the domestic hen, to metabolize steroid precursors was evaluated. Granulosa and theca interna cells were isolated from ovarian preovulatory follicles at three different developmental stages: F1, F3 and F5. Tritiated pregnenolone (P5), progesterone (P4), dehydroepiandrosterone (DHEA), androstenedione (A4) and testosterone (T) were employed as precursors and their metabolic products were evaluated. The major metabolite of P5 by granulosa cells was P4, but we also observed low amounts of 5β-pregnandione. DHEA metabolism by granulosa cells yielded mainly A4, and minute quantities of 5β-androstan-3,17-dione (5β-dione) were detected. The only significant metabolite obtained in granulosa cells from A4 was 5β-dione, whereas T was only transformed into A4. On the other hand, P5 metabolism by theca interna cells yielded A4 as the main product, also P4, 17α-OHP4, 17α-OHP5, 5β-pregnandione, and DHEA, were found. When DHEA was the precursor A4 was produced in higher amounts than 5β-dione. A4 was mainly transformed into 5β-dione. In similar conditions, T was transformed into A4. These results show that granulosa cells have enzymatic activities of 3β-hydroxysteroid dehydrogenase/5-4 isomerase (3β-HSD from P5 and DHEA), 17β-hydroxysteroid dehydrogenase (17β-HSD from T) and 5β-reductase (from P5, DHEA and A4). Whereas theca interna cells have enzymatic activities of cytochrome P450c17 (from P5 and P4), 3β-HSD (from P5 and DHEA), 17β-HSD (from T) and 5β-reductase (from P4, DHEA and A4). These data support the concept that theca interna cells have the ability to synthesize androgens from progestins produced in granulosa cells. In addition, since theca interna cells did not show the capacity to aromatize androgens suggests that interaction between theca interna and theca externa cells occurs in vivo, thus confirming the three cell model for estrogen production. Furthermore, the fact that other metabolites were produced both in granulosa and theca interna cells, but in a different extent, suggests that complex mechanisms are participating in the regulation of steroid synthesis in avian ovary follicles.  相似文献

Diatoms form large spring blooms in lakes and oceans, providing fuel for higher trophic levels at the start of the growing season. Some of the diatom blooms, however, are not grazed by filter-feeding zooplankton like Daphnia due to their large size. Several of these large diatoms are susceptible to chytrid infections. Zoospores of chytrids appeared to be excellent food for Daphnia, both in terms of size, shape, and quality (PUFAs and cholesterol). Thus, zoospores of chytrids can bridge the gap between inedible diatoms and Daphnia. In order to examine the effects of diatoms and chytrids on the survival of copepods, we performed one grazing and one survival experiment. The grazing experiment revealed that the diatom, Asterionella formosa, was not grazed by the copepod, Eudiaptomus gracilis, even after being infected by the chytrid Zygorhizidium planktonicum. However, carbon and nitrogen concentrations were significantly reduced by E. gracilis only when A. formosa was infected by Z. planktonicum, indicating that the chytrids might facilitate material transfer from inedible diatoms to the copepods. The survival experiment revealed that E. gracilis lived shorter with A. formosa than with the cryptophyta Cryptomonas pyrenoidifera. However, the survival of E. gracilis increased significantly in the treatment where A. formosa cells were infected by Z. planktonicum. Since E. gracilis could not graze A. formosa cells due to their large colonial forms, E. gracilis may acquire nutrients by grazing on the zoospores, and were so able to survive in the presence of the A. formosa. This provides new insights into the role of parasitic fungi in aquatic food webs, where chytrids may improve copepod survival during diatom blooms.  相似文献

The eastern Colorado shortgrass steppe is dominated by the C4 grass, Bouteloua gracilis, but contains a mixture of C3 grasses as well, including Pascopyrum smithii. Although the ecology of this region has been extensively studied, there is little information on how increasing atmospheric CO2 will affect it. This growth chamber study investigated gas exchange, water relations, growth, and biomass and carbohydrate partitioning in B. gracilis and P. smithii grown under present ambient and elevated CO2 concentrations of 350 μl l−1and 700 μl l−1, respectively, and two deficit irrigation regimes. The experiment was conducted in soil-packed columns planted to either species over a 2-month period under summer-like conditions and with no fertilizer additions. Our objective was to better understand how these species and the functional groups they represent will respond in future CO2-enriched environments. Leaf CO2 assimilation (A n), transpiration use efficiency (TUE, or A n/transpiration), plant growth, and whole-plant water use efficiency (WUE, or plant biomass production/water evapotranspired) of both species were greater at elevated CO2, although responses were more pronounced for P. smithii. Elevated CO2 enhanced photosynthesis, TUE, and growth in both species through higher soil water content (SWC) and leaf water potentials (Ψ) and stimulation of photosynthesis. Consumptive water use was greater and TUE less for P. smithii than B. gracilis during early growth when soil water was more available. Declining SWC with time was associated with a steadily increased sequestering of total non-structural carbohydrates (TNCs), storage carbohydrates (primarily fructans for P. smithii) and biomass in belowground organs of P. smithii, but not B. gracilis. The root:shoot ratio of P. smithii also increased at elevated CO2, while the root:shoot ratio of B. gracilis was unresponsive to CO2. These partitioning responses may be the consequence of different ontogenetic strategies of a cool-season and warm-season grass entering a warm, dry summer period; the cool-season P. smithii responds by sequestering TNCs belowground in preparation for summer dormancy, while resource partitioning of the warm-season B. gracilis remains unaltered. One consequence of greater partitioning of resources into P. smithii belowground organs in the present study was maintenance of higher Ψ and A n rates. This, along with differences in photosynthetic pathway, may have accounted for the greater responsiveness of P. smithii to CO2 enrichment compared to B. gracilis. Summary

Responses of larvae of two rhizocephalan species to changes in seawater temperature and salinity were studied under laboratory conditions. Peltogasterella gracilis parasitizes the hermit crab Pagurus pectinatus, which occurs at stable salinity and gradually changing temperature in summer. Sacculina polygenea is a parasite of the crab Hemigrapsus sanguineus, which lives in the intertidal zone in summer where salinity and temperature can fluctuate during the day. The development of both species is comprised of five naupliar stages and the cyprid stage, and it was considered successful if more than 50% of the nauplii attained the cyprid stage. P. gracilis nauplii successfully developed at 12–20°C and 30–34‰, but at 22°C successful development occurred in a narrower salinity range (32–34‰). All nauplii died both at 25°C and in 26‰. S. polygenea nauplii successfully reached the cyprid stage at higher temperatures (18–25°C) and a wider salinity range (18–34‰) than P. gracilis nauplii, but at 12°C and 16‰ larval development of S. polygenea was suppressed. Under favorable conditions, naupliar development lasted 3.5 days in P. gracilis and 2–3 days in S. polygenea. The cyprids of both rhizocephalan species demonstrated a greater resistance to temperature and salinity changes than nauplii. However, P. gracilis cyprids were active in a narrower salinity range (16–34‰), as compared to S. polygenea cyprids (8–34‰). Under favorable conditions the cyprids of both species survived for 6 to 10 days.  相似文献

This study reports some of the steroidogenic characteristics of the interna and externa theca cells taken from young and eCG primed calves. These cells were isolated from large healthy follicles. They were separately cultured for 3 days in absence or in presence of steroid substrates. Androstenedione (A4) and progesterone (P4) were measured by radioimmunoassay (RIA). In control conditions, A4 levels, higher in interna than in externa cells (**P<0.01), decreased during cultures (**P<0.01). In both cell types, A4 increased in presence of 17α-hydroxypregnenolone (17OHP5), pregnenolone (P5) and 22R-hydroxycholesterol (22R-chol) (*P<0.05) but not with P4 or 17α-hydroxyprogesterone (17OHP4) (P>0.05). The most efficient substrate was dehydroepiandrosterone (DHEA) (*P<0.05). In control conditions, P4 levels increased in both cell types. They were higher in externa than in interna cells on day 1, the reverse was observed on day 3. P4 levels increased after addition of 22R-chol and P5 (*P<0.05) but not with 17OHP5, 17OHP4 and DHEA (P>0.05) from day 1 in externa cells and only on day 3 in interna cells. P4 levels measured on day 1 were lower than the quantity of P4 added as a substrate. These results, obtained with theca cells from young calf follicles, indicate: 1/A4 is synthesized by the Δ5 pathway and 17α-hydroxylase activity decreases in vitro, 2/externa and interna cells differ by the quantities of A4 and P4 produced, 3/both lack precursors to produce A4 and P4 but their 3β-hydroxysteroid dehydrogenase activity subsists, 4/P4 could be metabolized during the first 2 days in both cell cultures.  相似文献

Ovaries of four flatfish species—Liopsetta pinnifasciata, Pleuronectes obscurus, Pleuronectes yokohamae, and Glyptocephalus stelleri, as well as of Far Eastern navaga Eleginus gracilis and rudd Tribolodon brandtii caught in Peter the Great Bay of the Sea of Japan in prespawning and/or spawning periods were studied. Under conditions of anthropogenic pollution of the bay, the state of fish ovaries is determined by several factors, such as migration activity, mode of life (bottom or pelagic), and the degree of sites pollution where females stayed in the period of the vitellogenous growth of oocytes. The pollution of the bay exerts the most damaging effect on the reproductive system of shallow species of flatfish—L. pinnifasciata and P. obscurus—that do not perform far migrations and throughout their life stay in the zone exposed to toxicants. In navaga Eleginus gracilis and flatfish P. yokohamae, and G. stelleri that perform seasonal migrations, no damages of ovaries were found.  相似文献

The studies described indicate that the UV bleached mutant, Euglena gracilis W3BUL does not serve as a suitable cytoplasmic control for the phenylalanyl-tRNA synthetase system. Chromatography of wild-type E. gracilis on Sephadex G100 revealed three peaks of activity identified as the chloroplastic, cytoplasmic and mitochondrial enzymes. The chloroplastic activity was greater in log than in stationary phase cells and was the only activity recovered from purified chloroplasts. Cell-free extracts of the achloroplastic mutant, E. gracilis W3BUL, contained wild-type levels of the cytoplasmic and mitochondrial phenylalanyl-tRNA synthetases. However, no chloroplastic synthetase was detected in the mutant extracts. Anomalies in the aminoacylation behavior of the W3BUL system were observed which suggest the possibility of a mutation affecting non-chloroplastic tRNAs in this UV-induced mutant. These anomalies significantly reduce the ability of the E. gracilis W3BUL mutant to serve as a cytoplasmic control in the phenylalanyl-tRNA synthetase system.  相似文献

Vitamins B6 and B12, biotin, folates, riboflavin, nicotinate, pantothenate, biopterin, and vitamin C (l-ascorbate) were assayed in Poteriochromonas stipitata, Euglena gracilis, and Tetrahymena thermophila cells grown in defined media and in spent culture supernatants. P. stipitata and E. gracilis synthesized, stored and excreted folates (mainly as 5-methyltetrahydrofolate), B6, riboflavin, pantothenate, nicotinate, biopterin, and ascorbate. E. gracilis synthesized and stored biotin. T. thermophila did not synthesize the above vitamins except for B12, biopterin, and ascorbate; it excreted biopterin and stored B12 and ascorbate. Thiamin was left of consideration because all 3 organisms are thiamin auxotrophs. Possible ecological implications of these findings are considered.  相似文献

Summary Long term continuous plankton measurements at Helgoland (North Sea) have provided a set of data which could be used for ecological functional analysis with respect to prey predator theory. Local dynamics display trophic feedback between selected populations. Phytoplankton, small copepods, Noctiluca miliaris, Pleurobrachia pileus and Beroe gracilis conform with theoretical assumptions. The functional relationships are less significant in averaged population dynamics compared with single ear processes. The local prey-predator cycles are to be understood as population waves travelling through German Bight. Such population waves over an area of 18 000 km2 have been investigated and are displayed for the above given zooplankton populations as computer graphics and analysed in their progression for the population of P. pileus.  相似文献

The organization of the body cavities is an important morphological trait that can be used for establishing the phylogenetic relationships between different groups of animals. In the present study, the hemocoel and coelomic systems of 10‐hr‐old juveniles and adults of the hermaphroditic oikopleurid Oikopleura gracilis were examined using light and transmission electron microscopy. The trunk hemocoel in 10‐hr‐old juveniles was represented by small clefts containing layers of extracellular matrix of adjacent tissues or interstices with migrating primordial germ syncytium. The wide hemocoel in the tail contained extracellular strands, subdividing the hemocoel into hemal sinuses. In adults, a large hemocoel appeared in the trunk and tail, and also contained extracellular strands. The hermaphroditic gonad was surrounded by its own lining, separating it from the hemocoel. The gamete‐filled cavity in the ovary and testis appeared only at late‐stage gonadogenesis, when the pre‐spawning reduction of syncytium occurred in the gonads. The true coelom in 10‐hr‐old juveniles and adults was represented by the pericardium. The lining of the pericardium consisted of myoepithelial and peritoneal cells. In the myoepithelial cells of 10‐hr‐old juveniles, myofibrils had been formed. The myoepithelial cells of adults had several parallel rows of completely differentiated myofibrils. The substantial reduction of the coelomic and circulatory systems in O. gracilis evidently results from the extreme shortening of ontogeny in appendicularians. Development in O. gracilis from early juvenile to adult involves the following steps, which also suggest how the tunicate heart may have evolved: a single‐layered coelomic sac gives rise to a grooved pericardium with an open hemal sinus (simple heart). In ascidians, this simple heart in turn gives rise to a closed tubular, double‐layered heart–pericardial complex, with a separate pericardial cavity and a closed heart, whose wall is formed by specialized myocardium.  相似文献

Pseudochorda gracilis sp. Nov. (Pseudochordacease, Laminariales) is described from the Japan Sea coast of Hokkaido/ the species is subtidal, epilithic and annual, appearing in spring and maturing in winter. Erect thalli grow solitary or in tufts on a small discoid holdfast. They are simple, cord-shaped and hollow, with inner hyphal filaments, cylindrical medullary cells and paraphyses consisting of 3–6 cells. Hair tufts are observed only in young thalli. Unilocular sporangia are sessile and narrowly ovate. In culture, P. gracilis shows a heteromorphic life history with oogamy, characteristic of the order Laminariales. Gametophytes are dioecious and dimorphic. Gametophytes mature under lower temperature conditions (usually below 10°C), and sporophytes mature under low temperature and short-day conditions (5°C, SD). The seasonal growth pattern of the species results from the photoperiod-temperature conditions controlling saprophyte maturation.  相似文献

When far red light preincubated cells of Euglena gracilis are transferred to dark or light, chlorophyll fluorescence (F0 and Fm) decreases. Non-photochemical quenching in the dark is suggested to be induced partly by chlororespiration and partly by changes in the distribution of excitation energy between the photosystems. Depending on the light intensities it was possible to resolve the non-photochemical quenching into at least three different components. The slowest relaxation phase of non-photochemical quenching occurred only after exposure to high light and was assigned to photoinhibition. The other two components were an energy-dependent quenching (qE), and the one which we attribute to a spill over mechanism. We suggest that both photosystems use a common antenna system consisting of LHC I and LHC II proteins. In contrast to higher plants, qE in Euglena gracilis is independent of the xanthophyll cycle and an aggregation of LHC II. 毛竹是我国重要的经济竹种,在长期栽培适应过程中产生了丰富的变异。为揭示毛竹竹秆变异变型的全基因组突变类型,以黄皮毛竹、金丝毛竹、绿皮花毛竹和花毛竹4个毛竹变型为实验材料,采用高通量重测序技术获得全基因组序列,进行单核苷多态性(SNP)、小片段插入缺失(InDel)和结构变异(SV)检测和注释,并将变异基因进行功能注释。结果表明:花毛竹基因组检测得到的基因变异数最多,为12 555个; 金丝毛竹样品变异位点数最少,为11 923个; 4个样品都有7 000多个变异基因得到功能注释。GO注释分类包括细胞组件、分子功能和生物过程三个基因功能分类体系的56个功能组。在细胞组件方面,叶绿素合成相关基因有2 431个; 在生物过程方面,参与类胡萝卜素合成过程的基因有75个,参与花青素合成过程中的调控以及紫外光下组织中花青素积累的相关基因有80个。COG分类表明参与复制、重组和修复的基因数为369个,信号转导机制的基因数为291个,转录的相关基因为222个。通过KEGG数据库系统地分析变异基因参与的黄酮类、类胡萝卜素等物质代谢合成途径。深入研究这些差异基因的调控途径,从DNA水平上解释竹秆的变异机制,可为深入研究毛竹种内丰富的多态性和遗传变异提供数据支持,阐析不同变异类型的基因家族、功能基因等遗传基础。  相似文献

Summary The organ of Bellonci of Anaspides tasmaniae (Thomson) (Crustacea, Syncarida) is described light and electron microscopically, and a few histochemical tests are reported. Located ventrally in the eyestalk below the medulla interna, the organ is composed of a number of cavities. These are similar in structure in their contents and associated cellular components, which include two types of glia cells delimiting each cavity and the terminal parts of a few dendrites. Each dendrite usually bears two cilia, which project into the cavity where they split up into numerous branches. The organ is supplied by three nerve tracts: two from the medulla terminalis and one from the medulla interna. The sensory pore, which is innervated from the medulla interna, is not closely associated with the organ of Bellonci in Anaspides. No marked secretory activity is detectable by histochemical or ultrastructural observations. It is thought that the organ has a sensory function.This investigation was supported by a grant (to T.K.) from Helge Ax:son Johnsons Stiftelse. The effects of activated charcoal on growth and morphogenesis in plate cultures of different plant cells have been studied. It was shown that medium containing charcoal induced embryogenesis in cultures of Daucus carota in which embryo formation could not be brought about by omitting auxin from the medium. Charcoal-medium also induced abundant root formation in older cultures of Allium cepa, which normally did not produce roots. The growth of cultures of Glycine max and Haplopappus gracilis was totally inhibited by charcoal. It is thought that activated charcoal removes substances from the medium, one of which might be auxin.  相似文献

Between 1981–83 the gut contents ofDaphnia galeata, D. cucullata, Eudiaptomus gracilis, andCyclops vicinus were examined with light and scanning electron microscope to obtain information on the feeding of these species in Lake Balaton. The twoDaphnia species feed mainly on abioseston, and it is assumed that their primary nutrient source was organic matter adsorbed onto the surfaces of the abioseston granules plus bacteria and detritus.E. gracilis feeds on algae, showing a preference for green algae and diatoms.C. vicinus is also a prodigious consumer of algae in Lake Balaton, utilizing the whole size spectrum of phytoplankton. Concerning the trophic relationships between phytoplankton and zooplankton in Lake Balaton, that between diatoms and bothE. gracilis andC. vicinus is the most conspicouos. Convincing evidence for an extensive utilization of blue-green algae was not found. Though there is no firm evidence yet, it is likely that theDaphnia are dependent on organic matter adsorbed on the abioseston.  相似文献
