Griffonia simplicifolia II, an N-acetylglucosamine-specific legume lectin, has insecticidal activity when fed to the cowpea weevil, Callosobruchus maculatus (F.). A cDNA clone encoding G. simplicifolia II was isolated from a leaf cDNA library, sequenced, and expressed in a bacterial expression system. The recombinant protein exhibited N-acetylglucosamine-binding and insecticidal activity against cowpea weevil, indicating that glycosylation and multimeric structure are not required for these properties. These results support the hypothesis that genes of the legume lectin gene family encode proteins that function in plant defense against herbivores.  相似文献

Phytohemagglutinin (PHA), the major seed lectin of the common bean (Phaseolus vulgaris), is found largely in the cotyledons, but is also present in the embryonic axis. At mid-maturation, the percentage of total protein synthesis which is directed towards making PHA is 5 to 10 times greater in the cotyledons than in the axes. This lower rate of synthesis in the axes is correlated with a lower abundance of mRNA for PHA, as determined by dot blot hybridization using a cDNA clone for PHA. Manen and Pusztai (Planta 1982 155: 328-334) have claimed on the basis of immunocytochemical evidence that, in the axis, PHA is found in the cytosol although it is present in protein bodies in the cotyledons. In the cotyledons, PHA is synthesized on rough endoplasmic reticulum, and its transport to the protein bodies via the Golgi complex is associated with specific posttranslational processing steps (Vitale and Chrispeels, J Cell Biol 1984 In press). A cytosolic localization of axis PHA would be an indication of a different site of synthesis and transport pathway. The results presented here indicate that the site of synthesis of PHA and the posttranslational modifications of PHA are the same in the axes as in the cotyledons. Since in the cotyledons these modifications take place in the endoplasmic reticulum, the Golgi, and the protein bodies, it appears that the transport pathway and the site of accumulation of PHA in the axes is similar to that in the cotyledons. On the basis of our evidence, we suggest that the subcellular localization of PHA in the axes should be reexamined.  相似文献

Artificial feeding assays were used to study the effect of purified galactose-specific lectins from African yam beans (Sphenostylis stenocarpa) on development of larvae of the cowpea weevil, Callosobruchus maculatus (Coleoptera : Bruchidae) and the legume pod-borer, Maruca vitrata (Lepidoptera : Pyrialidae). Inhibition of development of C. maculatus was observed when larvae were fed on artificial cowpea seeds containing 0.2%, 2.0% and 5.0% (wt/wt) of dietary lectin. Larval mortality was between 30% and 88%, while delays in total developmental time ranged between 7 and 13 days. The lectin had no effect on development of larvae of M. vitrala, when tested through topical artificial diet incorporation assays, except at the extremely high dose of 35% dietary level.  相似文献

One California processor of organic garbanzo beans (Cicer arietinum L.), unable to use chemical fumigants, relies on 30-d storage at -18 degrees C to disinfest product of the cowpea weevil, Callosobruchus maculatus (F). To determine whether the storage period may be shortened, the most cold-tolerant life stage of the cowpea weevil was identified. Laboratory studies showed that the egg stage was most tolerant to -18 degrees C and that adults were most susceptible. To examine the efficacy of cold storage disinfestation, bags of black-eyed peas, Vigna unguiculata (L.) Walp., infested with cowpea weevil eggs were buried within garbanzo bean bins placed in a commercial cold storage facility kept at approximately -18 degrees C and removed after 7, 14, and 21 d. Survival was highest in eggs located at the center of the bins and coincided with the slowest cooling rate. Although temperatures within the bins did not reach -18 degrees C until after 14-19 d, egg mortality was estimated to be >98% after just 7 d of exposure. Complete mortality of eggs occurred after 14 d of cold storage. A 2-wk treatment regimen may be sufficient for control of cowpea weevil in organic legumes.  相似文献

Waterlettuce,Pistia stratiotes L., has been successfully controlled in Australia with the weevil,Neohydronomus affinis Hustache. In this study, the weevil was tested in quarantine against 34 plant species in 27 families. In non-replicated no-choice tests, it fed and oviposited on 3 species of duckweeds [(Lemna minor L.,Spirodela punctata (Meyer) Thomps.,S. polyrhiza (L.) Schleid.], frogbit [(Limnobium spongia (Bosc.) Steud.], mosquitofern (Azolla caroliniana Willd) and waterfern (Salvinia mimima Baker). Feeding also occurred on golden club (Orontium aquaticum L.) and one egg was laid on the terrestrial panda plant (Kalanchoe tomentosa Baker). Plants oviposited and fed upon were retested in choice tests. The weevils then fed and oviposited exclusively on waterlettuce. In the current work, the toxicity and mechanism of phytohaemagglutinin (PHA), lectin isolated from the kidney bean (Phaseolus vulgaris) to the grain aphid (Sitobion avenae F.) were studied. When S. avenae was fed an artificial diet containing the lectin PHA, toxicity assays indicated that fecundity decreased, the pre-reproductive period and generation time were prolonged, and mortality increased. To elucidate the mode of action of PHA, the interaction between the lectin and the insect gut was investigated. Interestingly, DNA fragmentation was observed in extract of gut of treated grain aphid, and this was accompanied by an increase in caspase-3 activity. Results indicate that the detrimental activity of PHA on S. avenae may involve effects on death of the gut epithelial cells.  相似文献

Three digestion experiments involving cryptorchid weaned lambs were conducted for 14-day periods in the spring, summer and autumn to determine the in vivo digestibility of dry matter (DMD), gross energy (GE), organic matter (OMD), digestible organic matter in the dry matter (DOMD) and estimated metabolisable energy (ME) concentration of Lotus corniculatus L. (birdsfoot trefoil; cv. Grasslands Goldie) at different stages of maturity. The experiments were carried out indoors at Massey University's Riverside farm, in the Wairarapa, North Island, New Zealand. In vivo digestibility samples were then used as standards to investigate if the enzymic in vitro system of [Roughan, P.G., Holland, R., 1977. Predicting in vitro digestiblities of herbages by exhaustive enzymic hydrolysis of cell walls. J. Sci. Food Agric. 28, 1057–1064] could predict OMD and DOMD of condensed tannin (CT)-containing L. corniculatus.Digestibility of L. corniculatus declined as it matured, but the rate of decline was much less than that which occurs for perennial ryegrass (Lolium perenne)/white clover (Trifolium repens) pasture. It was concluded that the in vitro enzymic system of [Roughan, P.G., Holland, R., 1977. Predicting in vitro digestiblities of herbages by exhaustive enzymic hydrolysis of cell walls. J. Sci. Food Agric. 28, 1057–1064] can be used to predict OMD and DOMD of L. corniculatus with a high degree of accuracy, provided a standard curve involving in vivo data generated with L. corniculatus is used. Using a standard curve with in vivo data from perennial ryegrass/white clover pasture led to bias which increased at lower OMD values. Reasons for the differences between L. corniculatus and pasture standard curves are discussed, including possible effects of bound CT.  相似文献

Laboratory studies were carried out to determine the functional response of the parasitoid, Pteromalus cerealellae (Boucek) to cowpea weevil, Callosobruchus maculatus (F.) hosts, and the relationship between parasitism and cowpea variety susceptibility to the weevil. The attack response of P. cerealella to weevil larvae was best described by a type III functional response using an egg limitation model. The mean attack rate (a) of the parasitoid was estimated as 1.7 and the upper limit of the response was 24 weevil larvae per individual parasitoid within a 48-h period. The results also showed that cowpea varieties resistant to weevil infestation supported fewer weevil progeny and facilitated a greater suppression of the weevil by the parasitoid. The parasitoid progeny index, a measure that compares the number of the parasitoid progeny to the number of the beetle progeny was higher in cowpea weevil resistant varieties than in susceptible varieties. Additionally, the presence of P. cerealellae resulted in reduced weight losses of cowpea caused by the weevil infestation in both susceptible and resistant varieties.  相似文献

Four isolectin forms of a seed lectin from mature seed of tepary bean (Phaseolus acutifolius) were isolated using solubility fractionation, affinity chromatography, and high performance liquid chromatography. The subunits are polypeptides with an apparent molecular mass of 30,000 daltons. The 30 kilodalton subunits are produced starting approximately 13 days after flowering and subsequently comprise a major fraction of the proteins found in the mature seed. The amino terminus of each isolectin fraction was determined to be highly homologous with that of the subunits of common bean (Phaseolus vulgaris L.) phytohemagglutinin (PHA). The tepary isolectin cross-reacts with both erythroagglutinating and leucoagglutinating subunits of PHA antibodies, although differential cross-reactivity was noted. A seed protein fraction enriched in tepary bean lectin was found to be toxic to bean bruchid beetles (Acanthoscelides obtectus), when incorporated into their diets at incremental concentrations from (1-5% w/w) above that of PHA concentrations in mature seeds of the susceptible common bean variety “Red Kidney.”  相似文献

Insecticidal lectins were isolated from 20 resistant Vigna and non-Vigna legumes and tested againstn 3 pests of cowpea namely: Maruca vitrata, Callosobruchus maculatus and Clavigralla tomentosicollis. Crude lectins were separated from seeds using sodium chloride extraction, ammonium sulfate fractionation, and dialysis. SDS-PAGE indicated the molecular size of ca. 30 kDa for the most intense (and presumably active) band. Haemagglutination assays using trypsin-treated rabbit erythrocytes suggested that lectins were among the extracted proteins. Extracts from Lablab purpureus and Sphenostylis stenocarpa both non-Vigna spp., caused greater agglutination than those from the wild Vigna species. Bioassays on all three insect species using the lectin extracts incorporated in either artificial cowpea seeds (5% w/w) or in modified Vanderzant legume pod borer diet (1% w/v) indicated that the non-Vigna extracts were highly toxic to the insects. Mortality after 10 days was >80% in the most toxic extracts. The extract from one of the accessions of Sphenostylis stenocarpa, an edible legume, was singled out for lectin purification and future gene cloning with the view of using it for engineering resistance to cowpea pests.  相似文献

Bruchid beetle larvae cause major losses in grain legume crops throughout the world. Some bruchid species, such as the cowpea weevil (Callosobruchus maculatus) and the Mexican bean weevil (Zabrotes subfasciatus), are pests that damage stored seeds. The Mediterranean flour moth (Anagasta kuehniella) is of major economic importance as a flour and grain feeder; it is often a severe pest in flour mills. Plant lectins have been implicated as antibiosis factors against insects. Bauhinia monandra leaf lectin (BmoLL) was tested for anti-insect activity against C. maculatus, Z. subfasciatus and A. kuehniella larvae. BmoLL produced ca. 50% mortality to Z. subfaciatus and C. maculatus when incorporated into an artificial diet at a level of 0.5% and 0.3% (w/w), respectively. BmoLL up to 1% did not significantly decrease the survival of A. kuehniella larvae, but produced a decrease of 40% in weight. Affinity chromatography showed that BmoLL bound to midgut proteins of the insect C. maculatus. 33 kDa subunit BmoLL was not digested by midgut preparations of these bruchids. BmoLL-fed C. maculatus larvae increased the digestion of potato starch by 25% compared with the control. The transformation of the genes coding for this lectin could be useful in the development of insect resistance in important agricultural crops.  相似文献

The maize weevil Sitophilus zeamais Motsch. is an important pest of maize that attacks the grain both in the field and during storage. The damage caused by the maize weevil S. zeamais on maize landraces, Amarelo Antigo, Asteca, Caiano, Carioca, and Ferrinho, was evaluated by no-choice tests under laboratory conditions. The commercial varieties Sol da Manh?, BR 106, BR 451, and the synthetics PC 0203 and PC 9903 were evaluated for comparisons with the maize landraces. The parameters evaluated were susceptibility index, number of weevil progeny, development time, weevil progeny dry weight, and grain dry weight loss. The landraces were more susceptible to the maize weevil as compared to the commercial varieties. Based on the cluster analysis, two groups of susceptibility to the maize weevil were observed: one of more susceptible populations formed by local landraces and BR 451, and another less susceptible, with commercial varieties, synthetics, and the landrace Amarelo.  相似文献

A mannose-binding lectin found in vegetative tissues of peanut, Arachis hypogaea, was compared with mannose-binding lectin from pea, Pisum sativum, for toxic effects on larvae of the stem borer Chilo partellus (Swinhoe). After 10 days, the mortality of larvae fed on artificial diet containing 0.5% (m/m) peanut lectin was 46.2%. The mortality of larvae fed on 1.0% peanut lectin was similar (48.1%) but insects were significantly smaller than those of the 0.5% treatment. Larvae of both lectin treatments stopped feeding within three days. Larval size and mortality was not significantly reduced by 0.1% peanut lectin and 1% heat-treated lectin did not show toxic effects. The mannose-binding lectin from pea was not toxic to C. partellus at concentrations up to 1%. Peanut lectin bound to the apical membranes of columnar epithelial cells in the mid-gut of C. partellus. This suggests that peanut lectin has an antinutritive action and that it may protect vegetative tissues of peanut against insect pests.  相似文献

A set of 14 plant lectins was screened in a binary choice bioassay for inhibitory activity on cowpea weevil Callosobruchus maculatus (F.) oviposition. Coating of chickpea seeds (Cicer arietinum L.) with a 0.05% (w/v) solution of plant lectins caused a significant reduction in egg laying. Control experiments with heat inactivated lectin and BSA indicated that the observed deterrent effects are specific and require carbohydrate-binding activity. However, no clear correlation could be established between deterrent activity and sugar-binding specificity/molecular structure of the lectins. Increasing the insect density reduced the inhibitory effect of the lectins confirming that female insects are capable of adjusting their oviposition rates as a function of host availability.  相似文献

The phenomena of heat-induced G1 delay and thermal resistance were compared in synchronous populations of CHO cells. Mildly toxic induction doses of 5 min (Single cell survival, (SCS) = 0.90 +/- 0.06) and 10 min (SCS = 0.69 +/- 0.12) at 45 degrees C resulted in G1 delays of 4.3 and 11.3 h, respectively. Thermal resistance was tested (30 min, 45 degrees C) for up to 32-92 h following the induction dose. Thermal resistance did not start to decay prior to 26 h following the induction dose. These data confirm reports by R. R. Read, M. H. Fox, and J. S. Bedford [Radiat. Res. 98, 491-505 (1984)] and G. L. Rice, J. W. Gray, P. N. Dean, and W. C. Dewey [Cancer Res. 44, 2368-2376 (1984)] that acutely heated G1 populations of CHO cells progress into S phase without a concurrent loss of thermal resistance, using 45 degrees C induction doses even less toxic than used by other workers.  相似文献

Pseudomonas aeruginosa lectin PA-II agglutinates human peripheral lymphocytes and stimulates mitogenesis (predominantly in T cells), like the plant lectins PHA and Con A. Murine splenocytes are also agglutinated and stimulated by PA-II as by Con A. Sialidase treatment of the human and murine cells enhances their agglutination and augments the stimulation of human lymphocytes at low PA-II concentrations. The PA-II agglutinating and mitogenic effects are specifically inhibited by L-fucose. The bacterial source and the specificity of PA-II for L-fucose are both rare features among the hitherto described mitogenic lectins. However, since this lectin also binds mannose, a mannose-bearing receptor might be involved in its mitogenicity.  相似文献

Naturally occurring cyclic tetrapeptides (CTPs) such as tentoxin (Halloin et al., Plant Physiol 1970, 45, 310-314; Saad, Phytopathology 1970, 60, 415-418), ampicidin (Darkin-Rattray, Proc Natl Acad Sci USA 1996, 93, 13143-13147), HC-toxin (Walton, Proc Natl Acad Sci USA 1987, 84, 8444-8447), and trapoxin (Yoshida and Sugita, Jpn J Cancer Res 1992, 83, 324-328; Itazaki et al., J Antibiot (Tokyo) 1990, 43, 1524-1532) have a wide range of biological activity and potential use ranging from herbicides (Walton, Proc Natl Acad Sci USA 1987, 84, 8444-8447; Judson, J Agric Food Chem 1987, 35, 451-456) to therapeutics (Loiseau, Biopolymers 2003, 69, 363-385) for malaria (Darkin-Rattray, Proc Natl Acad Sci USA 1996, 93, 13143-13147) and cancer (Yoshida and Sugita, Jpn J Cancer Res 1992, 83, 324-328). To elucidate scaffolds that have few low-energy conformations and could serve as semirigid reverse-turn mimetics, the flexibility of CTPs was determined computationally. Four analogs of cyclic tetraproline c[Pro-pro-Pro-pro] with alternating L- and D-prolines, namely c[pro-Pro-pro-NMe-Ala], c[pip-Pro-pip-Pro], c[pro-Pip-pro-Pro], and c[Ala-Pro-pip-Pro] were synthesized and characterized by NOESY NMR. Both molecular mechanics and Density Functional Theory quantum calculations found these head-to-tail CTPs to be constrained to one or two relatively stable conformations. NMR structures, while not always yielding the same lowest energy conformation as expected by in silico predictions, confirmed only one or two highly populated solution conformations for all four peptides examined. c[pro-Pro-pro-NMe-Ala] was shown to have a single all trans-amide bond conformation from both in silico predictions and NMR characterization, and to be a reverse-turn mimetic by overlapping four Calpha-Cbeta bonds with those for approximately 6.5% (Tran, J Comput Aided Mol Des 2005, 19, 551-566) of reverse-turns in the Protein Data Bank PDB with a RMSD of 0.57 A.  相似文献

在害虫治理中, 在消费或贮藏粮食加工产品的建筑设施或场所进行害虫防治需要将杀虫剂施用在各种基质表面上。为了测定不同基质表面上杀虫剂的药效, 将四纹豆象Callosobruchus maculatus (F.)成虫接触田间推荐剂量的阿维菌素、 溴氰菊酯和毒死蜱。结果表明: 施用在玻璃、 瓷砖、 塑料和纸盘表面上, 阿维菌素对四纹豆象成虫的致死率分别为63.33%, 22.41%, 12.9%和11.9%, 而溴氰菊酯在这4种基质表面上对四纹豆象成虫的致死率分别为 55%, 44.2%, 41.3%和 37.4%。在所有基质表面上接触毒死蜱, 四纹豆象成虫的死亡率均为100%。对数据进行的Probit 分析表明, 毒死蜱制剂在玻璃、 瓷砖、 塑料和纸盘上对四纹豆象成虫 的LC50 值分别为 8.66, 13.6, 29.16和 56.5 μg/mL, 阿维菌素制剂的相应数值分别为119.4, 446.2, 774.2 和 836.4 μg/mL, 溴氰菊酯制剂的相应数值分别为 1 008, 1 131, 1 210和 1 336 μg/mL。据此推断, 毒死蜱对四纹豆象的毒性最强, 且在玻璃、 瓷砖、 塑料和纸盘表面上的毒性依次降低。  相似文献

Survival of bacteria (Rhizobium, Agrobacterium, and Arthrobacter spp.), fungal spores (Penicillium sp.), and yeasts (Saccharomyces sp.) was studied in relation to water activity (aw) and the presence of nutritive solutes. The cells were entrapped in polysaccharide gels, as is done to immobilize cells or enzymes, and then dehydrated. The number of living cells (1010 g of dry polymer−1) remained constant for periods of storage of >3 years at 28°C when the inocula were kept at an aw of <0.069. At aw values between 0.069 and 0.83 the number of survivors diminished more and more rapidly as the aw was raised. For a given aw and organism, there were large differences in survival rate as a function of the nutritive solutes used to culture the microorganisms. Low-molecular-weight compounds (with three or five carbon atoms) had a deleterious effect on survival, whereas compounds of higher molecular weight (C6 to C12) had a protecting effect. Thus, the aw alone was not a sufficient explanation for the deterioration of the inocula. Survival seemed to be more directly related to some properties of the water in the biopolymer. New concepts such as the discontinuity of properties of water and the point of mobilization of solutes, already proposed by Duckworth and Kelly (J. Food Technol. 8:105-113, 1973) and Seow (J. Sci. Food Agric., 26:535-536, 1975), have been taken into consideration to explain the interactions of water with the biopolymer and their specific effects on the microorganisms.  相似文献
