Abstract.
  • 1 Like other checkerspots, Euphydryas gillettii butterflies may contain the defensive chemicals, iridoid glycosides, which are sequestered from their hostplants during larval feeding.
  • 2 We analysed the iridoid glycoside content of E.gillettii adults from two different populations, Warm Lake, Idaho, and Granite Creek, Wyoming, that have different patterns of hostplant use.
  • 3 Gas chromatographic analysis of thirty butterflies from the Wyoming population showed that they contained a mean of 1.27 (±0.19 SE) % dry weight iridoid glycosides. Notably, 20% of these butterflies contained no detectable iridoid glycosides.
  • 4 In contrast, nineteen butterflies from the Idaho population contained a mean of 3.89 (±0.38 SE) % dry weight iridoid glycosides, and all butterflies contained iridoid glycosides.
  • 5 These results illustrate how the chemical defence of herbivorous insects varies according to differential use of potential hostplants.

Burenella dimorpha, a microsporidian parasite of the tropical fire ant, Solenopsis geminata, produces two morphologically distinct types of spores. The binucleate free spores (spores not bound by a pansporoblast membrane) develop normally at temperatures at least as low as 20°C and as high as 32°C. The uninucleate octospores (spores bound in octets by a pansporoblast membrane), however, develop in a restricted range of temperature. Octospores constituted 35.9%± 2.6 of the spores in 25 pupae held at 28°C. Raising the temperature to 30°C reduced octospores to < 1% of the total spore population. Lowering the temperature to 25° or 22°C reduced the octospore population to 8.5%± 6.5 or 0.4 ± 0.5, respectively. Inhibition of octospore development was complete at 20°C. In contrast, the octospores of Vairimorpha necatrix and Vairimorpha plodiae are reported to be abundant at 16°C and 21°C, respectively. The critical event blocked in octospore development may be meiosis, as evidenced by an abundance of binucleate sporonts in the octospore sequence of development, and absence of more advanced sporogonic stages in hosts held at inhibitory temperatures. Free spore size is not affected by temperature although yield may be slightly reduced at elevated temperature.  相似文献

Aims

To develop test methods and evaluate the survival of Bacillus anthracis ?Sterne and Bacillus thuringiensis Al Hakam spores after exposure to hot, humid air.

Methods and Results

Spores (>7 logs) of both strains were dried on six different test materials. Response surface methodology was employed to identify the limits of spore survival at optimal test combinations of temperature (60, 68, 77°C), relative humidity (60, 75, 90%) and time (1, 4, 7 days). No spores survived the harshest test run (77°C, 90% r.h., 7 days), while > 6·5 logs of spores survived the mildest test run (60°C, 60% r.h., 1 day). Spores of both strains inoculated on nylon webbing and polypropylene had greater survival rates at 68°C, 75% r.h., 4 days than spores on other materials. Electron microscopy showed no obvious physical damage to spores using hot, humid air, which contrasted with pH‐adjusted bleach decontamination.

Conclusions

Test methods were developed to show that hot, humid air effectively inactivates B. anthracis ?Sterne and B. thuringiensis Al Hakam spores with similar kinetics.

Significance and Impact of the Study

Peristenus spretus Chen et van Achterberg (Hymenoptera: Braconidae), a parasitoid of the plant bug Apolygus lucorum (Hemiptera: Miridae), has been studied for use in augmentative biological control in China. Under laboratory conditions, we explored the development, survival, age-specific and potential lifetime fecundity, oviposition period and progeny sex ratio of P. spretus reared at six constant temperatures (15°C, 19°C, 23°C, 27°C, 31°C, 35°C) on the second instar nymphs of A. lucorum. At 15°C, male and female P. spretus took 48.7 ± 0.3 and 52.5 ± 0.3 days to complete their immature development, while developmental time was reduced by more than half at 23°C and 27°C. The parasitoid can only develop to the larval stage at 31°C and neither larva nor pupa survived at 35°C. The estimated lower developmental threshold of the immature stage was 7.3°C. When parasitoid adults were exposed at 15°C, females laid 90% of their eggs at first 19 days of oviposition and had an extended reproductive life. In contrast, females held at 27°C laid most of their eggs (90%) in their first of 10 days of oviposition and had shorter longevity. The highest potential lifetime fecundity of P. spretus was 671.2 ± 34.7 SE eggs produced over 23.4 ± 1.4 SE days at 23°C. At 15°C, 19°C and 23°C, sex ratios of reared parasitoids were male-biased, but at 27°C there was no male bias.  相似文献

The optimum conditions for Peronospora farinosa betae to produce spores were temperature 8–10 °C and relative humidity 90 % or more, but many spores were produced between 5 and 20 °C and between 80 and 90 % R.H. Most spores were formed in darkness after leaves were exposed to light for 6–8 h. Spores survived exposure to 60 % R.H. for up to 5 days, but were soon killed by temperatures above 20 °C. The germination capacity of spores collected from the field was often very small, but this could not be related to the weather. Most seedlings were infected when inoculated at the growing point and incubated in a saturated atmosphere between 3 and 15 °C for at least 8 h.  相似文献

In this study, we developed an oviposition model of Neoseiulus californicus (McGregor) with Tetranychus urticae Koch as prey. To obtain data for the model, we investigated the longevity, fecundity and survivorship of adult female N. californicus at six constant temperatures (16, 20, 24, 28, 32 and 36°C), 60–70% RH and a photoperiod of 16 : 8 (L : D) h. Longevity (average ± SE) decreased as temperature increased and was longest at 16°C (46.7 ± 5.25 days) and shortest at 36°C (12.8 ± 0.75 days). Adult developmental rate (1/average longevity) was described by the Lactin 1 model (r2 = 0.95). The oviposition period (average±SE) was also longest at 16°C (29.8 ± 2.93 days) and shortest at 36°C (6.7 ± 0.54 days). Fecundity (average±SE) was greatest at 24°C (43.8 ± 3.23 eggs) and lowest at 36°C (15.9 ± 1.50 eggs). The oviposition model comprised temperature‐dependent fecundity, age‐specific cumulative oviposition rate and age‐specific survival rate functions. The temperature‐dependent fecundity was best described by an exponential equation (r2 = 0.81). The age‐specific cumulative oviposition rate was best described by the three‐parameter Weibull function (r2 = 0.96). Heleomyza borealis Boh. (Diptera, Heleomyzidae) overwinters as larvae in Arctic habitats, where they may experience winter temperatures below ? 15°C. The larvae freeze at c.? 7°C but in acclimation experiments 80% survived when exposed to ? 60°C. Of the larvae exposed to between ? 4 and ? 15°C, only 3% pupated. However, when cooled to ? 20°C this increased to 44%, with 4% emerging as adults. Larvae maintained at 5°C contained low levels of glycerol, sorbitol and trehalose, which did not increase with acclimation to low temperatures. However, levels of fructose increased from 6.1 μg mg?1 fw in control animals to 17 μg mg?1 fw when exposed to ? 2°C for 1 week. Larval body water (2.2 ± 0.1 g/g dw, mean ± SD, n = 100) and lipid content (0.22 ± 0.002 g/g fw, mean ± SE) showed no significant change during acclimation to low temperatures. Larvae maintained at a constant 5°C survived for over 18 months with little loss of body mass (from 7.5 ± 1.2 to 7.0 ± 1.2 mg fw, mean ± SD, n = 20), but none pupated. Heleomyza borealis larvae appear to feed and grow until they reach a body mass of about 7.5 mg and then become dormant. They remain in this state until they experience a low temperature stimulus (< ? 15°C) followed by a warm period (≈ 5°C). This ensures that the larvae pupate and adults emerge in early summer, allowing the maximum growing period before the following winter. Heleomyza borealis are adapted to survive long winters in a dormant larval state. They have a low metabolic rate, can conserve body water even at subzero temperatures but do not synthesize large quantities of cryoprotectants.  相似文献

A technique for simultaneous inoculation of cucumber cotyledons with Colletotrichum orbiculare race 1 and Cladosporium cucumerinum has been developed. The procedure permitted both resistant and susceptible plants to be recovered. Seedlings were grown at 20°C and inoculated 24 h after emergence with Colletotrichum orbiculare (200 spores in 2 μ1 of water) and Cladosporium cucumerinum (1000 spores in 5 μ1 of water) followed by 48 h of incubation in the dark at 20°C and 100% r.h., and 48 h in a 20°C lighted growth chamber. Seedlings were then moved to a growth chamber at 21°C at night and at 26°C during the day for 4 days and plants were rated as resistant or susceptible 8 days after inoculation. No interference in the expression of resistance or susceptibility of cultivars to either pathogen was detected in simultaneous inoculations.  相似文献

  • 1 Absorption efficiencies of nitrogen and carbon in two Chironomus species found dominating in the profundal zone of mesotrophic Lake Erken were determined gravimetrically.
  • 2 Absorption efficiencies for C. plumosus showed greater seasonal variation than those of C. anthracinus, with low efficiencies coinciding with the summer dominance of flagellate phytoplankton and with high C:N ratios.
  • 3 Overall mean absorption efficiencies (±SE) for carbon and nitrogen, respectively, were 26.8% (±1.9) and 29.3% (±1.9) for C. plumosus, 24.6% (±1.7) and 28.1% (±1.8) for C. anthracinus.
  • 4 Significant differences were found to exist between the C: N ratios of the superficial 2 cm sediment layers and those of Chironomus anterior midgut contents.
  • 5 C. anthracinus appears to be a deposit feeder ingesting particulate matter scraped from the recently deposited surface sediments. The greater seasonal variation found in the absorption efficiencies of C. plumosus, together with the lower C:N ratios, support the contention that this species is a filter feeder with the nutritional quality of ingested matter depending primarily on pelagic inputs.

SUMMARY.
  • 1 We evaluated survival, growth and time to maturation of the fairy shrimp, Streptocephalus seali Ryder, in the laboratory at various combinations of temperature and water hardness.
  • 2 Both independent factors affected survival and growth of S. seali. Multiple regression analysis and response surface modelling predict that after 4 days, over 80% survival is obtained at temperatures from 14 to 28°C and water hardnesses from 60 to 130 mg CaCO3 1-?1.
  • 3 Growth rates of larvae were often maximum at physicochemical conditions other than those which had promoted maximum rates of survival. For example, after 12 days mean total body length was almost 12 mm in larvae which had been maintained at 34°C (80 mg CaCO3 1-?1): the maximum survival rate had been obtained at 19°C. Total length was directly correlated with temperature at the lowest hardness tested, but not at the other two hardnesses (100 and 120 mg CaCO3 1-?1). At the latter water hardnesses, total length was significantly less at 34°C than at 32°C on all three sampling occasions (4, 8 and 12 days post-hatch).
  • 4 Similarly, developmental stage of larvae correlated well with temperature but larvae reared at 34°C did not develop more quickly than those reared at 32°C. After 12 days, most larvae at the two highest temperature treatments had developed at least to Stage 14 and many were nearly mature; at 17°C most larvae were still at Stage 10.
  • 5 During our study of maturation rate of females we noted that egg production was initiated after completion of fourteen or fifteen moults. Mean time to maturation at 27°C (17.3±2.8 days) exceeded that at 32°C (12.3±2.6days). The minimum time to maturation of a shrimp was 9 days at 32°C.

Two larval instars of Carcinops pumilio (Erichson) were identified from a frequency distribution of the head capsule measurements. The total developmental time from egg to adult emergence averaged 20.5 ± 0.1 days at 30 °C. The 2nd instar was the longest, accounting for 39% of the total development time. The 1st instar larvae sustained the highest mortality of 26% whilst there was no mortality in the pupal stage. The immature stages sustained about 50% mortality before adult emergence. The adult females of C. pumilio can live for 140 days and the rate of survival was high in the young adults but decreased as the beetles aged. Newly emerged adults survived for 25.5 ± 0.4 days without feeding. The sex ratio was 0.48 females: 0.52 males. The intrinsic rate of natural increase (rm) was 0.065, the generation time (T) was 50.019 days and the net reproduction rate (Ro) was 20.191.  相似文献

The temperature‐dependent predation by Anthocoris nemorum L. (Het.: Anthocoridae) on second instar Brevicoryne brassicae L. (Hom.: Aphididae) at constant temperatures of 12, 15 and 20°C increased linearly with temperature with a mean (±SE) consumption of 6.9 (±0.8), 9.28 (±1) and 15.9 (±0.8) respectively.   1. The invasive sawfly Aproceros leucopoda causes severe defoliation of various elm species and thus can be a major pest in forest stands and urban environments.
  2. The overwintering biology of A. leucopoda has not been investigated so far; therefore, the aim of this study was to determine the cold tolerance strategy and cold hardiness of hibernating A. leucopoda eonymphs.
  3. The supercooling points (SCPs) of overwintering individuals varied geographically, monthly and interannually and ranged between ?12.14 °C and ?24.22 °C.
  4. As none of the eonymphs survived once the SCP had been reached, A. leucopoda is classified as a freeze‐avoidant species.
  5. Survival rates of overwintering eonymphs exposed to different sub‐zero temperatures above the SCP (?1.6 °C and ?4.0 °C for 10, 20 and 30 days and ?10.5 °C for 9 days) ranged between 89.2% and 100%, suggesting that A. leucopoda is not a chill‐susceptible species.
  6. Our results suggest that low winter temperatures may not be expected to be an important limiting factor for the overwintering success of A. leucopoda.

Secretory vesicles isolated from a variety of mammalian tissues are known to lyse and thereby release their secretory products when exposed to ATP. This process, which will be termed ATP-induced lysis, has been studied most extensively using adrenal chromaffin-granule preparations. We report here that ATP causes the lysis of a highly purified preparation of rat parotid secretory granules. The rate of granule lysis was measured spectrophotometrically, and ATP-induced lysis was expressed as the increase in the rate of lysis (r = % lysis per min) when ATP was added. This lytic process was characterized with respect to pH, temperature, osmolarity, and the ionic composition of the media ATP-induced lysis of parotid granules was found to have the following properties in common with the extensively characterized chromaffin-granule process:
  • 1 It is a saturable function of ATP with half-maximal rates observed at 0.5 ± 0.1 mM ATP.
  • 2 It is temperature dependent, eg, r = 6.1 ± 2.1%/min at 30°C vs 12.2 ± 2.5%/min at 37°C.
  • 3 It is inhibited in hyperosmotic media, eg, r = 5.3 ± 0.3%/min at 0.3 OsM vs 0.8 ± 0.2%/min at 0.4 OsM.
  • 4 It shows a nucleotide preference of ATP = GTP > ADP > AMP > CTP = ITP.
  • 5 It has an anion requirement.
The above findings, combined with reports of ATP-induced lysis of cholinergeric, insulin, and posterior-pituitary vesicles, imply that ATP-induced lysis may reflect an ATP-dependent property of all secretory vesicles, and as such, this vesicle property could play a similar role in each exocytotic release process. Using a model system, Miller and Racker [22] made a surprising finding that the extent to which liposomes fuse with a black lipid membrane depends on the osmotic gradient across the vesicle membrane. In view of the osmotic dependence of ATP-induced lysis in this and other secretory-vesicle preparations, we postulate that ATP may prime secretory vesicles for fusion with the plasma membrane by inducing and/or maintaining an osmotic gradient across the vesicle membrane.  相似文献

The viability of Mattesia trogodermae spores stored at different temperatures was assessed by the percentage infection induced in 30-day-old Trogoderma glabrum larvae. Exposure to 73°C and higher temperatures for 30 min was lethal to the spores. Spores stored at ?19°C survived better than those stored at 26.7°, 3.5°, or ?30°C.  相似文献

We have developed an experimental system of cohort monospores from clonal culture of leafy gametophytes in Porphyra yezoensis Ueda (strain TU-1). This system is quite different from traditional systems for algal protoplast experimentation, which require expensive enzymatic treatment and utilize an ineffective method of preservation. Cohort monospores were obtained by utilizing a mode of asexual reproduction in the culture strain (monospores) and artificial regulation (thallus length, temperature, light, etc.) of monospore release. When the leafy gametophytes that formed monospores were frozen at - 20°C in a cryoprotective solution composed of 5% DMSO and 5% dextran in 100% seawater, about 98% survived for 3 months. When stored at 5°C without cryoprotectants, these leafy gametophytes could be kept without monospore release for 1 week. Maximum monospore yield was about 3000 spores per 100 gametophytes, and germination rate was about 70%, This system will accelerate developmental biology studies in Porphyra.  相似文献
