A rapid solvent evaporation method based on the triple point of a processing solvent is presented to prepare carbon nanotube (CNT) foam with a porous structure for thermoelectric (TE) power generators. The rapid solvent evaporation process allows the preparation of CNT foam with various sizes and shapes. The obtained highly porous CNT foam with porosity exceeding 90% exhibits a low thermal conductivity of 0.17 W m?1 K?1 with increased phonon scattering, which is 100 times lower than that of a CNT film with a densely packed network. The aforementioned structural and thermal properties of the CNT foam are advantageous to develop a sufficient temperature gradient between the hot and cold parts to enhance TE output characteristics. To improve the electrical conductivity and Seebeck coefficient further, p‐ and n‐molecular dopants are easily introduced into the CNT foam, and the optimized condition is investigated based on the TE properties. Finally, optimized p‐ and n‐doped CNT foams are used to fabricate a vertical and flexible TE power generator with a combination of series and parallel mixed circuits. The maximum output power and output power per weight of the TE generator reach 1.5 µW and 82 µW g?1, respectively, at a temperature difference of 13.9 K.  相似文献

The introduction of new materials and processes to microfabrication has, in large part, enabled many important advances in microsystems, lab-on-a-chip devices, and their applications. In particular, capabilities for cost-effective fabrication of polymer microstructures were transformed by the advent of soft lithography and other micromolding techniques 1, 2, and this led a revolution in applications of microfabrication to biomedical engineering and biology. Nevertheless, it remains challenging to fabricate microstructures with well-defined nanoscale surface textures, and to fabricate arbitrary 3D shapes at the micro-scale. Robustness of master molds and maintenance of shape integrity is especially important to achieve high fidelity replication of complex structures and preserving their nanoscale surface texture. The combination of hierarchical textures, and heterogeneous shapes, is a profound challenge to existing microfabrication methods that largely rely upon top-down etching using fixed mask templates. On the other hand, the bottom-up synthesis of nanostructures such as nanotubes and nanowires can offer new capabilities to microfabrication, in particular by taking advantage of the collective self-organization of nanostructures, and local control of their growth behavior with respect to microfabricated patterns. Our goal is to introduce vertically aligned carbon nanotubes (CNTs), which we refer to as CNT "forests", as a new microfabrication material. We present details of a suite of related methods recently developed by our group: fabrication of CNT forest microstructures by thermal CVD from lithographically patterned catalyst thin films; self-directed elastocapillary densification of CNT microstructures; and replica molding of polymer microstructures using CNT composite master molds. In particular, our work shows that self-directed capillary densification ("capillary forming"), which is performed by condensation of a solvent onto the substrate with CNT microstructures, significantly increases the packing density of CNTs. This process enables directed transformation of vertical CNT microstructures into straight, inclined, and twisted shapes, which have robust mechanical properties exceeding those of typical microfabrication polymers. This in turn enables formation of nanocomposite CNT master molds by capillary-driven infiltration of polymers. The replica structures exhibit the anisotropic nanoscale texture of the aligned CNTs, and can have walls with sub-micron thickness and aspect ratios exceeding 50:1. Integration of CNT microstructures in fabrication offers further opportunity to exploit the electrical and thermal properties of CNTs, and diverse capabilities for chemical and biochemical functionalization 3.  相似文献

Amiloride is a known blocker of the mechano-electrical transduction current in sensory hair cells. Measurements of cupular motion in the lateral line organ of fish now show that amiloride concurrently changes the micromechanical properties of the hair cell bundles. The effects of amiloride on the mechanics and receptor potentials of the hair cells resemble those previously observed for the aminoglycoside drug dihydrostreptomycin (DHSM) and are similarly antagonized by Ca2+. We hypothesize that amiloride and DHSM act on hair cells in two correlated ways which manifest themselves in both the electrical and mechanical properties of the transduction process. One action is the reduction of the transduction current with a concurrent increase of the hair bundle stiffness. The other action is a shift of the hair cell''s operating point on a current–displacement curve, with a concomitant shift along the associated hair bundle stiffness–displacement curve. The latter action has the opposite effect to that of the first and thus may lead, at relatively low blocker concentrations, to both an increase of transduction current and a decrease in hair bundle stiffness.  相似文献

In this study, molecular dynamics simulation is used to investigate the adsorption of an anticancer drug, doxorubicin, on bundles of functionalized single-walled carbon nanotubes (SWNTs) in an aqueous solution. Carboxylic group has been selected as the functional group. Molecular dynamics (MD) simulations are performed for both separated systems containing a SWNT bundle and a functionalized carbon nanotube bundle, and results are compared with existing experimental data. MD results show that doxorubicin can be adsorbed on CNTs using different methods such as entrapment within CNT bundle, attachment to the side wall of the CNT, and adsorption on the CNT inner cavity. For functionalized CNT, the adsorption of drugs on the functional groups is essential for predicting the enhancement of drug loading on the functionalized nanotubes. Furthermore, the adsorption behavior of doxorubicin on CNTs is fitted with Langmuir and Freundlich isotherm models. The results show that Langmuir model can predict the adsorption behavior of doxorubicin on CNTs more accurately than Freundlich model does. As predicted by this isotherm model, the adsorption process of doxorubicin on CNTs is relatively difficult, but it can be improved by increasing the functional groups on the CNTs surface.  相似文献

The method of Triton X-100 extraction and critical point drying of whole mounts of cultured chick and human myoblasts was used to study the presence of intracellular bundles of filaments within these cells. Observation by means of transmission and scanning electron microscopy demonstrated a complex system of filament bundles which appeared morphologically and spatially heterogeneous. Most obvious were long dense bundles or cables traversing along the ventral surface of developing myoblasts, presumably the ‘stress fibers’ seen in light microscopy. Other bundle types occurred which were composed of loose aggregates of filaments coursing through the remnant cell body. A prominent accumulation of filaments was also seen at the lateral edges of these myoblasts. These lateral edge cables were thicker and denser than any other type of filament bundle observed in the myoblasts. Reaction of unextracted myoblasts directly to human antiplatelet myosin conjugated to rhodamine demonstrated that the most intense reaction also occurred along the lateral edges of both human and chick myoblasts. During development of chick myoblasts the filament bundles became oriented parallel to the cell axis giving the cell a fusiform morphology. It is possible that the various filament bundle structures and their differing structural and spatial dispositions could be related to functional differences among the diverse population of intracellular bundles of filaments.  相似文献

The vascular system of the grapevine (Vitis vinifera L.) flower is a channel for transporting water and nutrients to the ovary. It plays an important role in the development of the ovary and fertilization through pollination. However, the vascular bundles in the flower are so tiny that they are difficult to sample and observe by traditional slicing techniques. In this study, ‘Summer Black’ grape flowers were selected as the test materials, and the tissue samples were treated by the optical clearing technique. After simple compaction, the structure and development of the vasculature were observed by common microscopy, fluorescence microscopy and laser confocal microscopy. The results showed that the transparency effects of 3% NaOH and a saturated trichloroacetaldehyde composite agreed well with the observations of the vascular structure and the developmental process of the flower in different periods. Moreover, the samples after optical clearing could be reconstructed in 3D, which helped us know more about its development and function. According to these observations, the vasculature of the ‘Summer Black’ flower can be divided into ovule vascular bundles, peripheral vascular bundles and central vascular bundles. The peripheral vascular bundles were composed of the first-order vascular bundles and the inferior vascular bundles which branched from the superior vascular bundles. These bundles branched in different directions with no discernible pattern. The two different branching methods were as follows. First, the inferior vascular bundle was directly connected to a superior vascular bundle. Secondly, some of the superior vascular bundles bent in different ways, forming the inferior vascular bundle connecting the superior vascular bundles by a metamorphosed vessel with a triangular shape. In a comparison of the developmental changes in various periods, the growth of vascular bundles at each period was directly proportional to the growth of the flower. Laser confocal scanning was used to explore the three-dimensional morphology of the peripheral vascular bundle and showed that the peripheral vascular bundle of grapes was not completely parallel to the flower’s epidermal cells. As a result, the optical clearing technique was convenient and authentic compared with the traditional slicing operation for tiny flower organs. With these advantages according to the observations, this study provides a feasible technique and useful information for the study of vascular bundle development in grape flower organs.  相似文献

Spontaneous oscillations exhibited by free-standing hair bundles from the Bullfrog sacculus suggest the existence of an active process that might underlie the exquisite sensitivity of the sacculus to mechanical stimulation. However, this spontaneous activity is suppressed by coupling to an overlying membrane, which applies a large mechanical load on the bundle. How a quiescent hair bundle utilizes its active process is still unknown. We studied the dynamics of motion of individual hair bundles under different offsets in the bundle position, and observed the occurrence of spikes in hair-bundle motion, associated with the generation of active work. These mechanical spikes can be evoked by a sinusoidal stimulus, leading to an amplified movement of the bundle with respect to the passive response. Amplitude gain reached as high as 100-fold at small stimulus amplitudes. Amplification of motion decreased with increasing amplitude of stimulation, ceasing at ∼6–12 pN stimuli. Results from numerical simulations suggest that the adaptation process, mediated by myosin 1c, is not required for the production of mechanical spikes.  相似文献

Earlier studies have shown that the helical content of α‐helical peptide decreases upon its interaction with carbon nanotube (CNT). Further, the length of the α‐helix varies from few residues in the small globular protein to several number of residues in structural and membrane proteins. In structural and membrane proteins, helices are widely present as the supercoil i.e., helical bundles. Thus, in this study, the length‐dependent interaction pattern of α‐helical peptides with CNT and the stability of isolated α‐helical fragment versus supercoiled helical bundle upon interaction with CNT have been investigated using classical molecular dynamics (MD) simulation. Results reveal that the disruption in the helical motif on interaction with CNT is directly proportional to the length of the helix. Also it is found that the shorter helix does not undergo noticeable changes in the helicity upon adsorption with CNT. On the other hand, helicity of longer peptides is considerably affected by its interaction with CNT. In contrast to the known fact that the stability of the helix increases with its length, the disruption in the helical peptide increases with its length upon its interaction with CNT. Comparison of results shows that structural changes in the isolated helical fragment are higher than that in supercoiled helix. In fact, helical chain in supercoiled bundle does not undergo significant changes in the helicity upon interaction with CNT. Both the length of the helical peptide and the inherent stability of the helical unit in the supercoiled helix influence the interaction pattern with the CNT. © 2012 Wiley Periodicals, Inc. Summary Bundles of fibrils and tubular structures were found to be associated with growing trabeculae ofCaulerpa simpliciuscula. In the rhizome tips, the bundles had an average diameter of 0.1 to 0.3 m, and a length greater than 10 m. The fibrils in the bundles were oriented in a strictly parallel fashion, with an individual thickness of 3–8 nm. The development of trabeculae started with the apposition of material of low electron density onto the bundles, which in this way became the inner skeleton of the trabeculae.Although fibre bundles with the same internal structure also occurred in the frond tip, these rarely contributed to trabecula formation. In the frond tips a different type of bundle with paracrystalline structure was found associated with the trabecular surface, forming a temporary connection between adjacent trabeculae. Permanent connection was achieved by deposition of further layers of trabecular material. These bundles in the frond tip consisted of two layers of tubular elements with a wall thickness of 80 Å and an inner diameter of 20–25 nm.Both fibre bundles and tubular bundles appear to contribute to trabecula formation. The similarity of these structures to the vacuolar inclusions observed in other siphonous algae is discussed.  相似文献

Summary The maculae utriculi and sacculi of the inner ear from the European roach (Rutilus rutilus) were investigated by transmission electron microscopy. The stereovilli of peripherally and centrally located sensory cells differ in several features that suggest a developmental gradient. The stereovilli of the peripheral sensory cells, shown to be differentiating cells by other research groups, are short and less steeply graded in height than in central hair cells. All stereovilli in both kinds of hair bundles are interconnected. In the central bundles of stereovilli basal, tip, and vertical connectors are separated by unconnected regions. In contrast, filaments and sometimes other additional structures connect the stereovilli of peripheral bundles over their entire length, but vertical connectors are usually absent. Osmiophilic material occurring inside peripheral stereovilli is interpreted to be monomeric actin. Central and peripheral hair bundles also differ in their reaction to ruthenium red and cationized ferritin. Only the stereovilli of the central cells can be fused by these polycations. Ruthenium red also discriminates between supporting and sensory cells indicating differences in amount or distribution of extracellular material. Hair bundles, intermediate in properties and position between central and peripheral sensory cells, were also found, so that it became possible to propose a scheme of developmental steps leading from microvilli or microvillus-like stereovilli to the fully differentiated hair bundle.  相似文献

Nuclear pore complexes (NPCs) form gateways for material transfer across the nuclear envelope of eukaryotic cells. Disordered proteins, rich in phenylalanine-glycine repeat motifs (FG-nups), form the central transport channel. Understanding how nups are arranged in the interior of the NPC may explain how NPC functions as a selectivity filter for transport of large molecules and a sieve-like filter for diffusion of small molecules (< or ). We employed molecular dynamics to model the structures formed by various assemblies of one kind of nup, namely the 609-aa-long FG domain of Nsp1 (Nsp1-FG). The simulations started from different initial conformations and geometrical arrangements of Nsp1-FGs. In all cases Nsp1-FGs collectively formed brush-like structures with bristles made of bundles of 2–27 nups, however, the bundles being cross-linked through single nups leaving one bundle and joining a nearby one. The degree of cross-linking varies with different initial nup conformations and arrangements. Structural analysis reveals that FG-repeats of the nups not only involve formation of bundle structures, but are abundantly present in cross-linking regions where the epitopes of FG-repeats are highly accessible. Large molecules that are assisted by transport factors (TFs) are selectively transported through NPC apparently by binding to FG-nups through populated FG-binding pockets on the TF surface. Therefore, our finding suggests that TFs bind concertedly to multiple FGs in cross-linking regions and break-up the bundles to create wide pores for themselves and their cargoes to pass. In addition, the cross-linking between Nsp1-FG bundles, arising from simulations, is found to set a molecular size limit of < for passive diffusion of molecules. Our simulations suggest that the NPC central channel, near the periphery where tethering of nups is dominant, features brush-like moderately cross-linked bundles, but in the central region, where tethering loses its effect, features a sieve-like structure of bundles and frequent cross-links.  相似文献

We present a two-step approach to modeling the transmembrane spanning helical bundles of integral membrane proteins using only sparse distance constraints, such as those derived from chemical cross-linking, dipolar EPR and FRET experiments. In Step 1, using an algorithm, we developed, the conformational space of membrane protein folds matching a set of distance constraints is explored to provide initial structures for local conformational searches. In Step 2, these structures refined against a custom penalty function that incorporates both measures derived from statistical analysis of solved membrane protein structures and distance constraints obtained from experiments. We begin by describing the statistical analysis of the solved membrane protein structures from which the theoretical portion of the penalty function was derived. We then describe the penalty function, and, using a set of six test cases, demonstrate that it is capable of distinguishing helical bundles that are close to the native bundle from those that are far from the native bundle. Finally, using a set of only 27 distance constraints extracted from the literature, we show that our method successfully recovers the structure of dark-adapted rhodopsin to within 3.2 A of the crystal structure.  相似文献

The bonding rules for actin filament bundles do not lead to a particular packing symmetry, but allow for either regular or disordered filament packing. Indeed, both hexagonal and disordered types of packing are observed in vivo. To investigate factors which control bundle order, as well as size, we examined the effect of protein concentration on the growth of actin-fascin bundles in vitro. We found that bundles require 4-8 d to achieve both maximum size and order. The largest and best ordered bundles were grown at low fascin and high actin concentrations (an initial fascin/actin ratio of 1:200). In contrast, a much larger number of poorly ordered bundles were formed at ratios of 1:25 and 1:50, and most surprisingly, no bundles were formed at 1:300 or 1:400. Based on these observations we propose a two-stage mechanism for bundle growth. The first stage is dominated by nucleation, which requires relatively high concentrations of fascin and which is therefore accompanied by rapid growth. Below some concentration threshold, nucleation ceases and bundles enter the second stage of slow growth, which continues until the supply of fascin is exhausted. By analogy with crystallization, we hypothesize that slower growth produces better order. We are able to use this mechanism to explain our observations as well as previous observations of bundle growth both in vitro and in vivo.  相似文献

This work presents the fabrication and investigation of thermoelectric cells based on composite of carbon nanotubes (CNT) and silicone adhesive. The composite contains CNT and silicon adhesive 1∶1 by weight. The current-voltage characteristics and dependences of voltage, current and Seebeck coefficient on the temperature gradient of cell were studied. It was observed that with increase in temperature gradient the open circuit voltage, short circuit current and the Seebeck coefficient of the cells increase. Approximately 7 times increase in temperature gradient increases the open circuit voltage and short circuit current up to 40 and 5 times, respectively. The simulation of experimental results is also carried out; the simulated results are well matched with experimental results.  相似文献

Fusion proteins of many viruses, including HIV-1 envelope protein (Env), fold into six-helix bundle structures. Fusion between individual Env-expressing cells and target cells was studied by fluorescence microscopy, and a temperature jump technique, to determine whether folding of Env into a bundle is complete by the time fusion pores have formed. Lowering temperature to 4 degrees C immediately after a pore opened halted pore growth, which quickly resumed when temperature was raised again. HIV gp41-derived peptides that inhibit bundle formation (C34 or N36) caused the cold-arrested pore to quickly and irreversibly close, demonstrating that bundle formation is not complete by the time a pore has formed. In contrast, lowering the temperature to an intermediate value also halted pore growth, but the pore was not closed by the bundle-inhibiting peptides, and it enlarged when temperature was again elevated. This latter result shows that bundle formation is definitely required for the fusion process, but surprisingly, some (if not all) bundle formation occurs after a pore has formed. It is concluded that an essential function of the bundle is to stabilize the pore against collapse and ensure its growth.  相似文献

The aerial stem of Prionium has been studied by motion-picture analysis which permits the reliable tracing of one among hundreds of vascular strands throughout long series of transverse sections. By plotting the path of many bundles in the mature stem, a quantitative, 3-dimensional analysis of their distribution has been made, and by repeating this in the apical region an understanding of vascular development has been achieved. In the mature stem axial continuity is maintained by a vertical bundle which branches from each leaf trace just before this enters the leaf base. Lateral continuity results from bridges which link leaf traces with nearby vertical bundles. Development of the provascular system involves a meristematic cap into which the blind ends of vertical bundles can be followed. Leaf traces are produced continuously in association with developing leaf primordia for a period of over 30 plastochrones; they connect with the vertical bundles in the meristematic cap and so establish the essential vascular configuration which is later reorientated through about 90° by overall growth of the crown. The last bundles to differentiate from the leaf do so outside the meristematic cap and thus fail to make contact with the axial system; they appear in the mature axis as blind-ending cortical bundles. Prionium is only distantly related to palms and its vascular histology is quite different. Nevertheless, the course of vascular bundles and the origin of this pattern in the stem resembles that of a palm. It is suggested that we are examining the fundamental pattern of vascular development in large monocotyledons.  相似文献

Here we demonstrate that a free-standing carbon nanotube (CNT) array can be used as a large surface area and high porosity 3D platform for molecular imprinted polymer (MIP), especially for surface imprinting. The thickness of polymer grafted around each CNT can be fine-tuned to imprint different sizes of target molecules, and yet it can be thin enough to expose every imprint site to the target molecules in solution without sacrificing the capacity of binding sites. The performance of this new CNT–MIP architecture was first assessed with a caffeine-imprinted polypyrrole (PPy) coating on two types of CNT arrays: sparse and dense CNTs. Real-time pulsed amperometric detection was used to study the rebinding of the caffeine molecules onto these CNT-MIPPy sensors. The dense CNT-MIPPy sensor presented the highest sensitivity, about 15 times better when compared to the conventional thin film, whereas an improvement of 3.6 times was recorded on the sparse CNT. However, due to the small tube-to-tube spacing in the dense CNT array, electrode fouling was observed during the detection of concentrated caffeine in phosphate buffer solution. A new I–V characterization method using pulsed amperometry was introduced to investigate the electrical characterization of these new devices. The resistance value derived from the I–V plot provides insight into the electrical conductivity of the CNT transducer and also the effective surface area for caffeine imprinting.  相似文献

The de novo design of proteins is a rigorous test of our understanding of the key determinants of protein structure. The helix bundle is an interesting de novo design model system due to the diverse topologies that can be generated from a few simple α-helices. Previously, noncomputational studies demonstrated that connecting amphipathic helices together with short loops can sometimes generate helix bundle proteins, regardless of the bundle''s exact sequence. However, using such methods, the precise positions of helices and side chains cannot be predetermined. Since protein function depends on exact positioning of residues, we examined if sequence design tools in the program Rosetta could be used to design a four-helix bundle with a predetermined structure. Helix position was specified using a folding procedure that constrained the design model to a defined topology, and iterative rounds of rotamer-based sequence design and backbone refinement were used to identify a low energy sequence for characterization. The designed protein, DND_4HB, unfolds cooperatively (Tm >90°C) and a NMR solution structure shows that it adopts the target helical bundle topology. Helices 2, 3, and 4 agree very closely with the design model (backbone RMSD = 1.11 Å) and >90% of the core side chain χ1 and χ2 angles are correctly predicted. Helix 1 lies in the target groove against the other helices, but is displaced 3 Å along the bundle axis. This result highlights the potential of computational design to create bundles with atomic-level precision, but also points at remaining challenges for achieving specific positioning between amphipathic helices.  相似文献
