A new white truffle species, Tuber panzhihuanense from China   总被引:1,自引:0,他引:1  
A new white Tuber species, T. panzhihuanense, is described based on collections from Sichuan and Yunnan, China. This is one of 10 new white truffle species, which were discovered in 2010 and 2011. Morphological and molecular analysis shows that T. panzhihuanense is clearly different from any other taxon in the genus of Tuber. It grows in mycorrhizal association with trees of Pinus yunnanensis in limestone soils. It has a pleasant aroma and is good edible. This is the first white truffle species discovered in China that has commercial potential.  相似文献

Thambus planifrons Muona & Meng sp. nov. and Otho rutriclypeus Muona & Meng sp. nov. are described from China. Both genera are recorded for the first time from China. Except for the European T. friwaldszkyi Bonvouloir, 1871, and the Japanese T. curvicaria Hisamatsu, 1956, all other previously known species belonging to Otho were from the Americas. A new species of the genus Trematosphaeria was isolated and identified from the soil in Jiuzhaigou Nature Reserve, southwestern China. The new taxon was studied based on morphological characters and phylogenetic analyses of three nuclear loci, i.e., the internal transcribed spacer rDNA operon (ITS), and large and small subunit nuclear ribosomal DNA (nuc28S rDNA, nuc18S rDNA). The analyzed Trematosphaeria species were clustered in several clades within the Pleosporales and the genus was shown to be polyphyletic. The new species is introduced as Trematosphaeria terricola, and it is characterized by short-papillate ascomata with suborbicular ostiolum, 3-septate, cinnamon-brown, fusiform ascospores with narrowly rounded ends, verruculose at maturity. Trematosphaeria terricola is compared with morphologically similar species T. confusa, T. mangrovis, and T. pertusa, which differ from the former species in the number of septa, and the shape and size of the ascospores with or without a sheath. This is the first report of a Trematosphaeria isolate from alpine soil in China.  相似文献

Theileria sergenti and Theileria sinensis are closely related members of benign Theileria species found in cattle and yaks in China. They are morphologically indistinguishable. A polymerase chain reaction (PCR) targeting major piroplasm surface protein of T. sergenti and T. sinensis was developed in this study. The newly developed oligonucleotide primer set was able to specifically amplify the DNA of T. sinensis and in conjunction with primers for T. sergenti and these two species could be detected and distinguished. Specificity testing also revealed that there was no cross-reaction with the other tick-borne diseases Theileria annulata, Babesia ovata, Anaplasma marginale as well as bovine white blood cells. Phylogenetic analysis based on the MPSP gene sequences confirmed the specificity of PCR assays. The sensitivity of the methods was 0.1 pg DNA for the T. sergenti PCR and 1 pg DNA for T. sinensis PCR. Two hundred and thirty-six field blood samples from of cattle and yaks were collected from five different geographical regions in China where benign Theileria species have been found. T. sergenti was found in all five provinces but was absent from one county in Gansu Province. T. sinensis was only found in Gansu Province. In both counties in Gansu where the parasites co-existed, mixed infections were detected. Our results indicate that the PCR methods developed in this study are suitable for the detection and differentiation of T. sergenti and T. sinensis.  相似文献

Theileria sinensis was recently isolated and named as an independent Theileria species that infects cattle in China. To date, this parasite has been described based on its morphology, transmission and molecular studies, indicating that it should be classified as a distinct species. To test the validity of this taxon, the two internal transcribed spacers (ITS1 and ITS2) and the 5.8S rRNA gene were cloned and sequenced from three T. sinensis isolates. The complete ITS sequences were compared with those of other Theileria sp. available in GenBank. Phylogenetic analyses based on sequence data for the complete ITS sequences indicate that T. sinensis lies in a distinct clade that is separate from that of T. buffeli/orientalis and T. annulata. Sequence comparisons indicate that different T. sinensis isolates possess unique sizes of ITS1 and ITS2 as well as species-specific nucleotide sequences. This analysis provides new molecular data to support the classification of T. sinensis as a distinct species from other known Theileria spp. based on ITS sequences.  相似文献

In this paper, one new species of the leafhopper genus Thaioneura Song, Li & Dietrich, 2016 from Guizhou Province, China are described and illustrated: Thaioneura huajiangensis sp. nov. And the female ovipositors are photographed and described. A key to distinguish all known species of the genus is provided. The hind wing venation of the new species are the same as the T. suphanburia and T. sinuata, and not common in the tribe Erythroneurini.  相似文献

BackgroundToxoplasma gondii is widely distributed and can infect many species of warm-blooded animals, including swine. This study aimed to evaluate the prevalence of T. gondii in swines from the central of China. A total of 2798 samples, including 305 hearts, 2086 diaphragms, and 407 sera were collected from different swine in Henan Province, China. The modified agglutination test was used to detect antibodies against T. gondii in sera from jugular vein blood and heart blood (cut-off: 1:25), diaphragm juice (cut-off: 1:10). T. gondii DNA was screened from the digestive fluids of all diaphragm tissue samples and seropositive hearts, and attempt to isolate viable T. gondii strain by bioassay in mice.ResultsA total of 9.94% (278/2798) swine tested positive for T. gondii antibodies. Region, but not gender, was associated with T. gondi seropositivity in swine. T. gondii nucleic acid was not found in the tissue digestive fluids (2090 swines). Three groups of mice showed T. gondii antibodies after having been bioassayed with diaphragm samples (n = 81, which came from 2090 swine). No viable T. gondii strain was isolated from muscle of swine.ConclusionsThis is the first large-scale survey T. gondi infection in swine from the central of China. Overall, the prevalence of viable T. gondii in swine was low. Nevertheless, T. gondii infection is present in swine from the central of China. Consumers may acquire T. gondii infection from ingestion of raw or undercooked pork.  相似文献

Water chestnut, Trapa natans, has become a major invasive plant in shallow water bodies in the northeastern United States. The failure of chemical and mechanical means to provide long-term and economically sustainable suppression of the species resulted in interest in the development of biological control. Field surveys in Asia and Europe identified a number of potential biological control agents in the native range of T. natans. The most promising species appeared to be the leaf beetle Galerucella birmanica, which is considered a pest of farmed T. natans in China. However, initial attempts to develop biological control faltered when field observations in China suggested that G. birmanica may not be host specific. Of particular concern was attack on water shield, Brasenia schreberi, a species native to China and North America. We conducted a number of laboratory and field investigations in China to assess preference and performance of G. birmanica on T. natans and B. schreberi. Initial no-choice experiments using 19 different plant species in 13 different families demonstrated that G. birmanica oviposited and was able to complete development only on Trapa spp. and B. schreberi. In larval no-choice tests G. birmanica was able to complete development on B. schreberi, however, larvae showed a 20% increase in mortality and longer development time compared to larvae developing on T. natans. In laboratory and field choice tests adults strongly preferred T. natans and in the field only occasionally laid eggs on B. schreberi. In addition, adults emerging from larvae reared on B. schreberi were less fit with reduced feeding and a declining oviposition rate. Their strong preference for T. natans was maintained in the field, even when T. natans was completely defoliated and adults were forced to migrate. We found only occasional “spill-over” of beetles onto B. schreberi and our data indicate that G. birmanica is a more promising biological control agent of T. natans than previously thought, although additional host specificity tests with many more North American plant species need to be completed.  相似文献

Taenia asiatica and T. saginata are frequently confused tapeworms due to their morphological similarities and sympatric distribution in Asian regions. To resolve this problem, a high-resolution multiplex PCR assay was developed to distinguish T. asiatica infections from infection with other human Taenia tapeworms. For molecular characterization, the species specificity of all materials used was confirmed by sequencing of the cox1 gene. Fifty-two samples were analyzed in this study, comprising 20 samples of T. asiatica genomic DNA from China, Korea, and the Philippines; 24 samples of T. saginata from Belgium, Chile, China, Ethiopia, France, Indonesia, Korea, Laos, the Philippines, Poland, Taiwan, Thailand, and Switzerland; and 10 samples of T. solium from Cape Verde, China, Honduras, and Korea. The diagnostic quality of the results obtained using PCR and species-specific primers designed from valine tRNA and NADH genes was equal to that based on the nucleotide sequencing of the cox1 gene. Using oligonucleotide primers Ta4978F, Ts5058F, Tso7421F, and Rev7915, the multiplex PCR assay was useful for the differentially diagnosing T. asiatica, T. saginata, and T. solium based on 706-, 629-, and 474-bp bands.  相似文献

A total of 16 Taenia multiceps isolates collected from naturally infected sheep or goats in Gansu Province, China were characterized by sequences of mitochondrial cytochrome c oxidase subunit 1 (cox1) gene. The complete cox1 gene was amplified for individual T. multiceps isolates by PCR, ligated to pMD18T vector, and sequenced. Sequence analysis indicated that out of 16 T. multiceps isolates 10 unique cox1 gene sequences of 1,623 bp were obtained with sequence variation of 0.12-0.68%. The results showed that the cox1 gene sequences were highly conserved among the examined T. multiceps isolates. However, they were quite different from those of the other Taenia species. Phylogenetic analysis based on complete cox1 gene sequences revealed that T. multiceps isolates were composed of 3 genotypes and distinguished from the other Taenia species.  相似文献

Tetranychus truncatus Ehara is a phytophagous spider mite that is now one of the most important pests of agricultural and economic crops in East and Southeast Asia. However, population genetics and other studies of T. truncatus have been impeded by the lack of microsatellite markers, which are expensive and time-consuming to identify. Previous studies indicated a high potential of cross-amplification of microsatellites in Tetranychus species, meaning that the microsatellite flanking sequences are sufficiently homologous among Tetranychus species that the primers for one species may work in another species. Here, we tested 205 primer pairs designed from the whole genome sequence of Tetranychus urticae Koch, a sister species of T. truncatus, for microsatellite markers in three populations of T. truncatus in China (N = 94). About half (102) of these primer pairs yielded the desired PCR products, 36 of which revealed polymorphism in T. truncatus. Each of the 36 markers harbored between 2 and 23 alleles, with a mean polymorphic information content of 0.589 (0.119–0.922 range). The mean observed and expected heterozygosity across loci and the three populations were 0.468 and 0.628, respectively. Of the 36 primer pairs, 22 also worked in Tetranychus piercei, but only a few of them worked in T. ludeni and T. phaselus. Cross-amplification is thus a cost-effective way to develop microsatellite markers, which can be of great value in population genetics studies.  相似文献

During an investigation of parasitic ciliates in northern China, two Trichodina species, T. acuta Lom, 1970 and T. nigra Lom, 1960, were isolated from the freshwater fish Cyprinus carpio Linnaeus, 1758. The morphology of each species was investigated based on dry silver nitrate-stained specimens. In addition, the molecular phylogeny of each was analyzed based on small subunit ribosomal DNA (SSU rDNA) sequence data. Trichodina acuta can be distinguished from its congeners by the undefined periphery of the central circle, the distinct gap between the rays and the central circle, and the distinctly sickle-shaped blades. Trichodina nigra is a cosmopolitan ciliate and is characterized by its densely linked denticles, broad, rounded spatula-shaped blades, robust central parts, and well developed rays. Phylogenetic analyses revealed that T. acuta and T. nigra nest within different clades, supporting the assertion that the GC content of SSU rDNA sequences could reflect evolutionary relationships among Trichodina species.  相似文献

The distribution of rhcRST and rhcJ-C1 fragments located in different loci of the type III secretion system (T3SS) gene cluster in the peanut-nodulating bradyrhizobia isolated from Guangdong Province, China was investigated by PCR-based sequencing. T3SS was detected in approximately one-third of the peanut bradyrhizobial strains and the T3SS-harboring strains belonging to different Bradyrhizobium genomic species. Diverse T3SS groups corresponding to different symbiotic gene types were defined among the 23 T3SS-harboring strains. The same or similar T3SS genes were detected in different genospecies, indicating that interspecies horizontal transfer of symbiotic genes had occurred in the Bradyrhizobium genus.  相似文献

Additional material of Todites (Osmundaceae) from the Lower Jurassic Xiangshan Group in Jiangsu and Anhui Provinces, Eastern China is investigated. Five species of Todites are described based on collections (including sterile and fertile pinnae) from eight localities along the Yangtze River, including Todites denticulatus (Brongniart) Krasser, Todites cf. goeppertianus (Muenster) Krasser, Todites nanjingensis sp. nov., Todites princeps (Presl) Gothan and Todites sp. Material of T. denticulatus and T. nanjingensis sp. nov. contains further data of the fertile pinnae. The stratigraphical ranges and the geographical distribution of the genus Todites in China are discussed and reviewed. Available data show that Todites mostly occurs in the Late Triassic to Middle Jurassic of China. The genus was widely distributed in both the Northern and Southern Floristic Provinces of China, and especially abundant and diverse in the Southern Floristic Province.  相似文献

The study deals with similarity among 25 species of the genus Trifolium represented by 134 accessions. Clustering of the species based on isozyme banding pattern of five enzymes revealed that T. repens was distinctly different from other species. T. repens and T. retusum formed independent clusters. The group of species comprising of T. pratense, T. cherleri, T. spumosum, T. subterraneum, T. resupinatum, T. alexandrinum, T. echinatum, T. constantinopolitanum and T. tembense exhibited considerable similarity to the second cluster. This group joined another group of five species, i.e. T. nigrescens, T. glomeratum, T. apertum, T. alpestre and T. hybridum with nearly 50% similarity. T. purpureum, T. hirtum, T. campestre, T. incarnatum, and T. argutum grouped separately. There was no marked difference for banding pattern among T. alexandrinum genotypes. T. alexandrinum showed close affinity with T. subterraneum and T. resupinatum. T. lappaceum, T. diffusum, T. campestre, T. incarnatum and T. argutum showed only 44.8% similarity with other Trifolium species. Grouping together of accessions belonging to individual species indicated that incompatibility among species under study had restricted interspecific hybridization. Species belonging to subgenus Lotoidea clustered with species of subgenus Trifolium. Chonosemium species T. campestre formed one cluster with two Trifolium species T. hirtum and T. incarnatum. T. nigrescens was placed quite apart from the T. repens.  相似文献

Borrelia sp. prevalence in ticks on migratory birds was surveyed in central Japan. In autumn, a total of 1,733 birds representing 40 species were examined for ticks. A total of 361 ticks were obtained from 173 birds of 15 species, and these ticks were immature Haemaphysalis flava (94.4%), Haemaphysalis longicornis, Ixodes columnae, Ixodes persulcatus, Ixodes turdus, and an unidentified Ixodes species. Of these, 27 juveniles of H. flava on Turdus pallidus, Turdus cardis, or Emberiza spodocephala, 2 juveniles of I. persulcatus on T. pallidus, and 1 female H. flava molted from a T. pallidus-derived nymph were positive for the presence of Borrelia by Barbour-Stoenner-Kelly culture passages. In spring, a total of 16 ticks obtained from 102 birds of 21 species were negative for the spirochete. Isolates from 15 ticks were characterized by 5S-23S rRNA intergenic spacer restriction fragment length polymorphism analysis; all isolates were identified as Borrelia garinii with pattern B/B′ based on the previous patterning. According to the intergenic spacer sequences, 2 of 15 isolates, strains Fi14f and Fi24f, were highly similar to B. garinii strains 935T of Korea and ChY13p of Inner Mongolia, China, respectively. These findings indicate that Lyme disease-causing B. garinii may have been introduced to Japan by migratory birds from northeastern China via Korea. Additionally, a case of transstadial transmission of B. garinii from nymph to adult H. flava suggests that the infected H. flava may transmit Borrelia to large animals.  相似文献

Results of comparative analysis of specific features of morphology of larval lanternfishes of the Indo-Pacific genus Triphoturus based on materials from the southeastern part of the Pacific Ocean, northwestern Pacific, South China Sea, and the western part of the Indian Ocean, as well as on published data, are provided. Noticeable differences are shown in specific features of pigmentation of larval T. nigrescens (sensu Hulley, 1986) from different parts of the vast range of this species, that may be accounted for by its individual and/or geographic variation and by the collective pattern of this taxon. It is stated that it is necessary to use genetic data for the verification of the taxonomic status of the representatives of the genus. Morphological evidence in favor of the species isolation of T. oculeum and T. microchir based on larval materials is presented. The presence of three morphologically different forms of larval Triphoturus from the eastern Pacific differing in the pattern of body pigmentation that are identified with three nominal species of the genus Triphoturus—T. nigrescens, T. mexicanus, and T. oculeum—is confirmed. The study of specific features of distribution of larvae of these species in southeastern waters of the Pacific Ocean based on our own and published materials revealed the presence of larval T. mexicanus and T. oculeum off Peru, which is supported by data on the presence of adult individuals of both species here.  相似文献
