Brain (median or lateral regions) or suboesophageal ganglion (SOG) homogenates of Day 1 fifth instar larvae of Trichoplusia ni induced the appearance of haemolymph juvenile hormone esterase (JHE) when injected into Day 1, Day 2 or early Day 4 fifth instar ligated hosts. Brain and SOG homogenates of late fourth instars also induced JHE when injected into Day 1 hosts, whole late fifth instar and pupal tissue did not. The pattern of JHE induction by early fourth through Day 3 fifth instar brain and SOG homogenates correlated with natural haemolymph JHE activity occurring at these times. Implantation of late fourth and Day 1 fifth instar brains and/or SOG into similar age hosts similarly induced JHE activity while prothoracic and abdominal ganglia did not. The relative levels of induction following implantation were SOG<brain<brain+SOG. JHE activity which appears in the haemolymph following injection of brain homogenates appears to be largely due to a single enzyme which has an isoelectric point indistinguishable from that of the natural haemolymph enzyme. Evidence is presented which suggests that inhibitory as well as stimulatory brain factors are involved in JHE regulation.  相似文献

The present in vitro studies using a suspension of Leydig cells from adult rat testis demonstrated that bromocriptine (BR, 2 × 10−5M) inhibits hCG-stimulated testosterone production (in the presence of submaximal and maximal doses of hCG), while basal production was unaffected. When the cells were exposed to 8-bromo-cAMP either in the presence or absence of hCG, the inhibitory effect of BR was not reversed. In intact cells, BR inhibited conversion of progesterone and 17-hydroxy-progesterone to testosterone while conversion of androstenedione was not affected. Incubation of homogenates of Leydig cells in the presence of limiting NADPH concentrations ( 0.1 mM) resulted in significant BR-induced inhibition of conversion of progesterone (10 μM) to testosterone, while in the presence of “high” concentrations of NADPH ( 0.5 mM) BR was without effect. Present results suggest that BR inhibits androgen production at the level of the microsomal enzymes 17-hydroxylase and/or 17,20-lyase. The inhibitory effect of BR using homogenates of Leydig cells was evident only in the presence of limiting NADPH concentrations that suggests a competitive-like pattern of inhibition, but mechanisms by which BR decreases activity of microsomal enzymes remain to be determined.  相似文献

Cachexia is a complex medical condition characterized by significant weight loss associated with decreased body fat and protein; the condition may present itself with or without anorexia. We have isolated and partially characterized a proteoglycan (azaftig) from the urine of cancer and AIDS patients experiencing weight loss. When given to mice, the purified azaftig resulted in a significant decrease in body weight and body fat without any effect on appetite. The results of these studies show that azaftig may be one of the many factors participating in the emergence of the cachectic state.  相似文献

1. 1.|The brain (BR), suboesophageal ganglion (SOG), metathoracic ganglion mass (TG3) or fourth free abdominal ganglion (AG4) were selectively warmed from room temperature (23°C) to 3°C and the effects of elevated ganglion temperature on abdominal ventilatory pumping rate were observed.

2. 2.|Warming the SOG or BR almost always induced a strong increase in ventilatory pumping rate while warming TG3 or AG4 had no consistent effect, inducing about equally often a slight increase, decrease or no change.

3. 3.|It is concluded that the most temperature sensitive elements associated with the ventilatory rhythm generating system are localized within the head ganglia.

Cadmium (Cd) exposure has been recognized to result in a wide variety of cellular responses, including oxidative stress and body weight loss. The aim of the present study was to examine the effect of lycopene supplementation on the antioxidant defense system, lipid peroxidation (LPO) level, nitric oxide (NO), tumor necrosis factor alpha (TNF-α) production, and body weight in Cd-exposed rats. Animals were divided into four groups (n = 7): control, Cd-treated, Cd plus lycopene-treated, and lycopene-treated. Cadmium (as CdCl2) was administrated orally for 20 days (6.6 mg kg−1 day−1), and lycopene (10 mg kg−1 day−1) was similarly administered. Lycopene administration significantly suppressed Cd-induced LPO in plasma and kidney homogenates. Lycopene also reversed Cd-decreased body weight compared to the control. Cadmium treatment had diverse effects on the antioxidant enzyme activities. Although antioxidant superoxide dismutase activity was unchanged, glutathione peroxidase activity was decreased, and catalase activity was elevated in kidney homogenates of Cd-administrated group. However, lycopene treatment reversed Cd-changed enzyme activities to the control level. Xanthine oxidase activity and TNF-α concentration were not altered by Cd administration, indicating that superoxide anion production and inflammation were not stimulated. Cadmium did not change NO levels in kidney homogenates but decreased those in plasma, and this effect was not prevented by lycopene supplementation. The result suggests that consumption of adequate levels of lycopene may be useful to prevent heavy-metal-induced LPO and body weight loss.  相似文献

Bacteriorhodopsin (BR) essentially free of native lipids has been prepared in a highly stable state. Purple membrane was solubilized in Triton X-100 and BR was purified by size exclusion chromatography using 3-[cholamidopropyl)dimethylammonio]-2-hydroxyl-1-propanesulfonic acid (CHAPSO) detergent at pH 5. Molar ratios of phospholipid/BR ranged from 0.4 to 0.05 corresponding to 94-98% phospholipid removal. Purified BR has an absorbance ratio (A280nm/A548nm) of 1.5-1.6 in the dark-adapted state which is the highest purified BR/protein ratio reported to date. The purified BR in CHAPSO shows maximum stability in the pH range 5.0-5.5. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis profiles of native purple membrane and solubilized BR from most Halobacterium halobium JW-3 cultures show 3 higher molecular weight bands in addition to BR. Immunological staining and amino acid sequencing indicates that these additional proteins are partially processed forms of the BR precursor protein. The BR preprotein contains 13 additional amino acids on the NH2 terminus which are removed by post-translational processing in at least four steps. Isoelectric focusing separated most delipidated and non-delipidated BR samples into 8 bands. Incomplete BR post-translational processing BR is thought to be largely responsible for the multiplicity of isoelectric BR species. The principal components have pI values of 5.20 and 5.24 and both have absorption maxima at 550 nm, characteristic of detergent-solubilized BR. BR in Triton X-100 or nonylglucoside, delipidated BR in CHAPSO, and BR in intact purple membrane all have a dark-adapted ratio of 13-cis to all-trans-retinal of 1.9:1.  相似文献

The physiological effects on submaximal and maximal exercise of three methods commonly used by athletes for achieving rapid weight loss were determined by measuring cardiorespiratory variables in 62 nonendurance athletes. A mean weight loss of 4.1% was achieved by those who followed either a sauna (SAU), diuretic (DIU), or exercise (ACT) protocol, compared with the average weight loss of 1.2% in the control group. At maximal exercise O2 consumption, O2 pulse, blood lactate concentration, and work load decreased in SAU and DIU groups relative to the ACT group, whereas only a few differences were observed at the aerobic threshold. Weight loss achieved over a 48-h period was less detrimental to an athlete than was a more rapid (24-h) weight reduction achieved through sauna bathing or the use of diuretics. We conclude that not only the quantity of weight loss but also the method itself may limit physical performance.  相似文献

Diuresis was studied in vivo by measuring the loss of tritiated water. The basal rate of water loss (5 nl/min) represents respiratory and cuticular losses, whereas higher rates reflect urine output, which reaches 20 nl/min after injection of 1 microl distilled water. This response to hypervolemia involves release of a diuretic hormone(s) into the hemolymph. However, housefly diuretic peptides increased urine output to a maximum of only 7 nl/min, and higher rates may require fluid reabsorption from the hindgut to be reduced. Diuresis is partially blocked by injected anti-muscakinin antibodies, providing evidence of a hormonal function for this insect myokinin.  相似文献

2001-2014年博斯腾湖流域植被物候时空变化及其驱动因子   总被引:1,自引:0,他引:1  
以博斯腾湖流域为研究对象,利用MODIS的MCD12Q2和LST产品、GHCN_CAMS气温观测/再分析资料与气象数据,采取趋势分析与相关性分析法探求了博斯腾湖流域2001-2014年植被物候的时空变化及其影响因素的相对作用,对博斯腾湖流域植被物候分区不同的驱动区域。结果表明:①在研究期内,整个研究区植被物候始期在第76-168天,末期在第172-295天;物候始期自南向北逐渐推迟、而末期逐渐提前,物候的空间分布特征与该区海拔高度的分布保持了较好的一致性;②2001-2014年植被始期和末期有明显提前趋势(提前3-6d),主要分布在流域的盆地和平原绿洲区,表示研究区植被物候受到人类活动的影响。③植被物候始期与末期变化受气候因子驱动影响的区域占比分别为57.10%和51.30%,主要分布在黄水沟流域,清水河流域,孔雀河流域,大尤路都斯盆地和小尤路都斯盆地周围地区;而非气候因子占42.90%和48.70%,主要位于博斯腾湖周围绿洲和库尔勒绿洲等地势较低的区域。④由植被生长季物候与降水、气温的偏相关性关系和复相关性关系可以得出,多年物候始期和末期与气温有关;而且随海拔升高,气温的敏感幅度越高。博斯腾湖流域植被物候的时空变化不仅是受气候变化的影响,还主要受人类活动和海拔高度差异等影响因素的共同作用。  相似文献

In order to study the role of membrane proteins in bilirubin (BR) binding phenomenon, selective removal of membrane proteins was carried out using various reagents, namely, ethylenediamine tetraacetic acid (EDTA), sodium hydroxide (NaOH), 3,5-diiodosalicylic acid, lithium salt (LIS), dimethylmaleic anhydride (DMMA), sodium iodide (NaI), o-phenanthroline-cupric sulfate (CuP) and phenanthroline-cupric sulfate containing 2-mercaptoethanol (CuP-mercaptoethanol). Effects of these treatments on the conformation and BR binding properties of the membrane were studied using circular dichroism (CD) spectroscopy as well as estimation of membrane-bound BR by diazotised-color reaction. Though a significant amount of protein (ranging from 23–69%) was lost from the membranes upon these treatments, only a small decrease (3–13%) was observed in BR binding, being maximum with NaOH-treated membranes. However, DMMA and NaI treatments produced a little increase in BR binding. Conformation of the membrane was retained to a significant extent as indicated by far-UV CD spectra upon these treatments except in DMMA and NaI treatments which resulted in the perturbation in CD spectra. Taken together, these results suggest that membrane proteins play little role in BR binding, rather act as barriers in BR binding phenomenon.  相似文献

Abstract A post-feeding diuretic response has been observed in adult Heliothis zea (Boddie) (Lepidoptera: Noctuidae). 2-day-old starved adults which were allowed to feed to repletion on a 10% (w/v) sucrose solution lost 49.1% (females) and 85.8% (males) of the weight of the ingested meal during the first hour following feeding. Ligation between head and thorax or frontal ganglionectomy, when performed immediately following feeding, each resulted in a significant and permanent reduction in this normal weight-loss. Injection of homogenates of the corpora cardiaca/corpora allata (CC/CA) complex into non-ligated insects immediately after feeding also reduced significantly the post-feeding weight-loss, but this inhibition was transient and disappeared after 1 h. Dissection and weighing of the crop from either ligated, frontal ganglionectomized, or CC/CA-injected insects confirmed the crop as the predominant site of fluid retention in each case. We suggest that a soluble antidiuretic factor from the CC/CA acts in conjunction with the frontal ganglion to control the rate of crop emptying and subsequent diuresis by regulating the volume of ingested fluid that is passed into the haemolymph from the crop/midgut.  相似文献

Apolipoproteins A-I, B, C-II and lipids were studied before and after rapid weight loss schedules. The compared groups were all athletic. Apolipoproteins were determined by electroimmunoassay methods using apoproteins purified by chromatofocusing column method. Dextran T10 was shown to increase rocket height in ApoB assay. Over 1% Dextran concentrations gave poor response. The linearity during calibration was from 0.3 to 3.0 g ApoB/l. Baseline values of ApoA-I in wrestlers, weightlifters, boxers and judokas were slightly higher as compared to "normal" population; ApoB was clearly reduced (mean value of 690 mg/l). Weight-loss was significant in each experimental group; mean value of 4.1% in active exercise, sauna and diuretic groups together. Compared as the whole sportsmen group in passive weight loss (or sauna) and diuretic groups the most pronounced changes were seen to be elevated apoprotein concentrations, whereas weight-loss by active rapid exercise resulted no apoprotein changes, but instead an increment in HDL cholesterol and decrement in triglycerides, respectively. The present study was the first to evaluate baseline values of apoproteins A-I, B and C-II in first class athletes and also the possible changes in these and lipid values in rapid weight-loss practices.  相似文献

Spinal cord injury (SCI) has serious long-term consequences on sympathetic cardiovascular regulation. Orthostatic intolerance results from insufficient baroreflex regulation (BR) of sympathetic outflow to maintain proper blood pressure upon postural changes. Autonomic dysreflexia occurs due to insufficient inhibition of spinal sources of sympathetic activity. Both of these conditions result from the inability to control sympathetic activity caudal to SCI. It is well established that limited motor ability recovers after incomplete SCI. Therefore, the goal of this study was to determine whether recovery of BR occurs after chronic, left thoracic spinal cord hemisection at either T(3) or T(8). Baroreflex tests were performed in rats by measuring the reflex response of left (ipsilateral) renal sympathetic nerve activity to decreases and increases in arterial pressure produced by ramped infusions of sodium nitroprusside and phenylephrine, respectively. One week after a T(3) left hemisection, BR function was modestly impaired. However, 8 wk after a T(3) left hemisection, BR function was normal. One week after a T(8) left hemisection, BR function was significantly impaired, and 8 wk after a T(8) left hemisection, BR function was significantly improved. These results indicate that BR of renal sympathetic nerve activity in rats may partially recover after spinal cord hemisections, becoming normal by 8 wk after a T(3) lesion, but not after a T(8) lesion. The nature of the spinal cord and/or brain stem reorganization that mediates this recovery remains to be determined.  相似文献

Bacteriorhodopsin (BR), from the purple membrane (PM) of Halobacterium halobium, was chemically modified with methoxypolyethylene glycol (m-PEG; molecular weight = 5,000 Da) succinimidyl carbonate. The polyethylene glycol-bacteriorhodopsin (m-PEG-SC-BR33) conjugate, containing one polyethylene glycol chain, was water soluble. The secondary structure of the conjugate in water appeared partially denatured, but was shown to contain alpha-helical segments by circular dichroism spectroscopy. The isolated bacteriorhodopsin conjugate, with added retinal, was refolded in a mixed detergent-lipid micelle and had an absorption maximum at 555 nm. The refolded conjugate was transferred into vesicles that pumped protons, upon illumination, as efficiently as did native BR. Modification of the PM with m-PEG did not alter the native structure or inhibit proton pumping, and therefore it is suggested that the glycol polymer is present as a moiety covalently linked to residues unnecessary for proton pumping and proper folding. The site of attachment of m-PEG was determined to be at either Lys 129 or Lys 159, with position Lys 129 the most probable site of attachment. The m-PEG-SC-BR33 could be stepwise refolded to the native conformation by the addition of trifluoroethanol to lower the dielectric constant, simulating the insertion of the BR into the phospholipid bilayer.  相似文献

The rabbit oviductal epithelium synthesizes and secretes a family of antigenically related, sulfated oviductal glycoproteins (SOG). Anti-SOG monoclonal antibodies (Mabs) were produced and two (Mab 1 and Mab 2) were selected for further characterization. Periodate oxidation of Western blots of oviductal fluid did not affect the binding of Mab 1 or Mab 2, thus suggesting that these antibodies recognized protein rather than carbohydrate epitopes on SOG. The specificity of Mab 1 was determined by Western blot analysis of tissues obtained from estrous rabbits and from the male rabbit reproductive tract. SOG was identified in tissue extracts of both the oviductal ampulla and isthmus. Cervix was the only non-oviductal tissue with which Mab 1 cross-reacted. Mab 1 was used to isolated SOG from whole oviductal fluid by immuno-affinity chromatography. Affinity-purified SOG and Mab 1 were used to develop a quantitative, SOG-specific, competitive enzyme-linked immunosorbent assay. This assay was used to quantify SOG in rabbit oviductal fluid collected during estrus and pseudopregnancy. SOG secretion during pseudopregnancy was resolved into two transient episodes of increased secretion. Maximum SOG secretion (X = 1039 +/- 199 micrograms/day) occurred within 48 h of the induction of pseudopregnancy. A second period of enhanced SOG secretion (X = 308 +/- 46 micrograms/day) occurred during the fifth and sixth days of pseudopregnancy. Baseline SOG secretion occurred during estrus at approximately 60% of maximum postovulatory secretion.  相似文献

High seed cost and low rates of establishment make tallgrass prairie restorations challenging and expensive endeavors. Typical seedling emergence rates in prairie restorations are approximately 10% and the causes of seed mortality are poorly understood. In this study, we examined the impact of small vertebrate granivores on prairie restoration by comparison of seedling emergence in open (sham) versus closed exclosures at three newly restored sites. To assess other causes of seed loss, we also tracked seed fates at one prairie restoration site. We coated seeds of four prairie species with fluorescent dye, placed them under closed exclosures, and monitored their fate (emerging seedling, partially germinated, nongerminated/viable, and nongerminated/nonviable) over a 5‐month period. On average, 9.6 more seedlings/m2 emerged in the closed than the opened exclosures, suggesting that small vertebrate granivores reduce seedling emergence in prairie restoration. Granivores influenced the composition of the emerging community but did not preferentially consume large‐seeded species. In the seed‐tracking experiment, we found that greater than 70% of seeds were lost within 30 days of sowing, that seed recovery and viability both decreased with time in soil, and that seed fates differed between species. Collectively, our results indicate that small vertebrate granivores are an important cause of seed loss in prairie restoration, but unidentified belowground (e.g. fungal decomposition, invertebrate predation) and environmental (wind, rain) factors account for a greater proportion of total seed loss. Until these causes of seed loss are better understood, high seed costs will persist and continue to impede prairie restoration.  相似文献

ABSTRACT. Homogenates of accessory glands from male Culex tarsalis Coquillet (Diptera; Culicidae) were fractionated using gel filtration column chromatography. A component of low molecular weight capable of suppressing female sexual receptivity was obtained and partially characterized. Proteins present in homogenates of isolated accessory glands formed aggregates with other proteins in extracts of whole mosquitoes. Consequently, previous reports of the molecular weight of biologically active components in homogenates from accessory glands from other mosquito species may be inaccurate.
When female Cx. tarsalis were mated with 3H-leucine labelled males. radioactivity could be found throughout the body of the female 24 h after mating. Autoradiography showed that the radioactivity was incorporated into the cells of various organs, and was not specifically localized in a particular target tissue. Approximately 30% of the total radioactivity transferred to the female was found in the head and thoracic regions, and proteins from the head had a specific activity twice that of the thoracic proteins. Bioassay of isolated body parts from mated and virgin females showed that injections of head tissues from mated females into virgin females suppressed sexual receptivity. These results suggest that the head is involved in the physiological mechanisms resulting in refractory mating behaviour.  相似文献

The rapid expulsion of Trichinella spiralis by mice of a variety of inbred and F1 mouse strains was examined. Mice were reinfected once with T. spiralis during and immediately after the natural termination of a primary infection and worm rejection was measured less than or equal to 24 hr after the challenge. The results showed that the challenge (super)infection was consistently rejected by all mouse strains before rejection of the adult worms from the primary infection commenced. Rejection of the challenge infection began at different times after the primary infection with NFS (2 days) less than C3H less than or equal to B10.Q approximately B10.BR (greater than 5 days). In all strains, rejection of the challenge infection preceded adult worm rejection from the primary infection by 5-8 days. At its peak, the loss of challenge worms related directly to the strength of the primary rejection process NFS greater than or equal to 98%, C3H 90-98%, and B10 mice 80-90%. Furthermore, loss of the capacity to reject the challenge followed approximately 7 days after the complete loss of the primary infection in each strain examined. Thus, the sooner worms from the primary infection were lost, the earlier the capacity to promptly reject the challenge infection disappeared. B10.Br mice still partially rejected a superinfection 35 days after the primary infection began, whereas NFS mice lost this capacity around 25 days. However, premature termination of the primary infection in B10.BR mice with methyridine at the same time that NFS mice naturally terminated their infection (15 days) abrogated the capacity of B10.BR mice to reject the superinfection at 24 days. Passive transfer of protective rat IgG monoclonal antibody to mice did not lead to rapid expulsion. Transfer of mouse immune serum to intestinally primed rats did result in rapid expulsion, suggesting that mouse antibody responses were adequate. The expression of superinfection rejection was susceptible to the administration in vivo of GK1.5, anti-mouse L3T4 antibody. The data indicate that the principal determinant of the strength, time of initiation, and longevity of rejection of a challenge infection was the response to the primary infection of that individual mouse strain. The genetic determinants of challenge infection rejection were seen to be identical to those that determined rejection of the primary infection. Since no evidence could be found to support the identity of this response with rapid expulsion, as defined in rats, a new term, "associative expulsion," is proposed.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Advances in our understanding of murine airway physiology have been hindered by the lack of suitable, ex vivo, small airway bioassay systems. In this study, we introduce a novel small murine airway bioassay system that permits the physiological and pharmacological study of intrapulmonary bronchial smooth muscle via a bronchial ring (BR) preparation utilizing BR segments as small as 200 microm in diameter. Using this ex vivo BR bioassay, we characterized small airway smooth muscle contraction and relaxation in the presence and absence of bronchial epithelium. In control BRs, the application of mechanical stretch is followed by spontaneous bronchial smooth muscle relaxation. BRs pretreated with methacholine (MCh) partially attenuate this stretch-induced relaxation by as much as 42% compared with control. MCh elicited a dose-dependent bronchial constriction with a maximal tension (E(max)) of 8.7 +/- 0.2 mN at an EC(50) of 0.33 +/- 0.02 microM. In the presence of nifedipine, ryanodine, 2-aminoethoxydiphenyl borate, and SKF-96365, E(max) to MCh was significantly reduced. In epithelium-denuded BRs, MCh-induced contraction was significantly enhanced to 11.4 +/- 1.0 mN with an EC(50) of 0.16 +/- 0.04 microM (P < 0.01). Substance P relaxed MCh-precontracted BR by 62.1%; however, this bronchial relaxation effect was completely lost in epithelium-denuded BRs. Papaverine virtually abolished MCh-induced constriction in both epithelium-intact and epithelium-denuded bronchial smooth muscle. In conclusion, this study introduces a novel murine small airway BR bioassay that allows for the physiological study of smooth muscle airway contractile responses that may aid in our understanding of the pathophysiology of asthma.  相似文献
