In the annelid Enchytraeus albidus the ovary is composed of packets containing eight synchronously developing oocytes. Each oocyte in the packet is connected, via a bridge, to a common cytoplasmic mass. Developmental synchrony of oocytes within individual packets is probably related to the ooplasmic continuity. The young previtellogenic oocyte contains many polysomes, a few cisternae of smooth and rough endoplasmic reticulum, small Golgi complexes, and mitochondria. Many of the mitochondria are dumbbell-shaped and may thus represent division stages. Vitellogenesis is marked by the appearance of peripherally located lipid yolk and small, densely staining granules scattered throughout the ooplasm. There is an increase of smooth endoplasmic reticulum, mitochondria, and enlarged Golgi elements. Small multivesicular-like bodies, the early stages of developing yolk, are derived from the Golgi complex. The mature yolk sphere is bipartite and consists of (a) a variable number of dense spheres, the core bodies, which are produced in the ooplasm by the Golgi complex and which become embedded in (b) a dense matrix. The electron opaque tracer, horseradish peroxidase is incorporated into the oocyte and deposited in the matrix suggesting that this component of the yolk sphere is obtained by micropinocytosis. Enzyme digestions and various cytochemical techniques suggest that the core bodies are rich in carbohydrate, probably as glyco- or mucoproteins, and that the matrix is rich in lipid. 相似文献

Geographic variation in cold tolerance and associated physiological adaptations were investigated in the freeze tolerant enchytraeid Enchytraeus albidus (Oligochaeta). Specimens from Svalbard, Greenland (Nuuk), Iceland (Hólar and Mossfellsbær) and continental Europe [Norway (Bergen), Sweden (Kullen) and Germany] were reared in the laboratory in a common-garden experiment. The aim was to test for variations in minimum lethal temperature, freeze duration tolerance, carbohydrate reserves and metabolic rate among the populations. Cold tolerance was related to environmental temperature of the respective location. Populations from the coldest climatic regions were able to tolerate freezing down to at least ?15 °C and endured being frozen at ?5 °C for 27–48 days, respectively. Populations from milder climates had a lower freeze duration tolerance (about ?9 °C) and endured ?5 °C for a shorter period (between 9 and 16 days). Glucose accumulation and glycogen reserves varied significantly between populations, but was not related directly to cold tolerance. Metabolic rate varied significantly between populations, but was not significantly related to cold tolerance. The metabolic rates at ?2 °C of frozen and unfrozen worms from Germany and Svalbard were tested. The metabolic depression due to freezing of these populations was relatively small (<50 %), suggesting that the large carbohydrate accumulations may also be important as fuel during long-term freezing at moderately low temperatures. Differences in metabolic depression may partly explain the difference in cold tolerance of these two populations, however, the mechanisms behind local adaptation to low winter temperatures in these enchytraeid populations seem more complex than earlier studies have indicated. 相似文献

Two types of coelomocytes, the mucocyte and the phagocyte, occur in Enchytraeus fragmentosus. Other free cells observed within the coelomic cavity include chloragogen cells, peritoneal cells and some anucleate granular cells. Three forms of mucocytes occur and are believed to represent developmental stages. The first stage is one in which the mucous droplets are forming in the Golgi region. The second stage is a mature form, and the third stage is one in which the mucous droplets are being released. The phagocytes generally are quite large, and inclusions vary from recognizable portions of chloragogen cells to extremely small, electron-dense cytosomes. The origin of the coelomocytes could not be determined. Probable functions of coelomocytes are discussed. 相似文献

The clitellum of the annelid worm Enchytraeus fragmentosus was found to consist of two distinctive granular cell types in addition to the cells typically found within the epidermis. One cell type possessed granules containing microtubules. The other cell type had large secretory granules which fused and filled most of the cell. The contents of these granules were established to be responsible for the secretion of the cocoon wall and the nutritive substance surrounding the egg. 相似文献

Following amputation of the posterior half of Tubifex, all the nerve cells stainable by paraldehyde fuchsin (NF) immediately discharged. Fifteen minutes later, some stained NFs reappeared. Their number increased, reached a peak far above the normal level, then slowly decreased. This peak occurred 8 hours after amputation in the brain, after 3-4 days in the ventral ganglia. A similar cycle took place during the anterior regeneration of Tubifex whose four anterior segments were previously cut away. Following a double amputation (behind the first or fourth segment and at half length), the stimulation of the neurosecretory activity was even clearer, the number of NFs reaching a higher and earlier peak. Correlatively, the regenerative histogenesis was accelerated in the caudal blastema. 相似文献

The nephrostome of Enchytraeus albidus exhibits a longitudinal slit-like opening on the dorsal side of a bulbous organ which is mainly composed of three cells,
one flame cell situated centrally and endowed with a ciliary flame, and two cells lying superficially, called the mantle cell
and the accessory mantle cell. The mantle cell occupies the ventral side of the organ extending on both sides up to the opening
to constitute its immediate border on the frontal and lateral sides. Here it forms a kind of crest which is heavily subdivided
into many protrusions and infoldings endowed with long cilia which exclusively spread into the coelomic cavity. The accessory
mantle cell borders the narrow posterior slit of the opening, forming the roof of the initial canal which is devoid of cilia.
From its anterior region a projection arises extending above the opening. The flame cell forms a groove from which the ciliary
flame arises which extends into the canal. At its posterior region it replaces the accessory mantle cell displacing it onto
the dorsal surface of the organ. It is argued that the mantle cell and the accessory mantle cell have presumbly originated
from coelothelial cells. Thus the metanephridium may be a composite organ developing from a nephridioblastic and a coeloblastic
source. A discussion of results in other annelid species indicates that metanephridia in the Annelida may have evolved more
than once.

Enchytraeus albidus is able to absorb dissolved14C-labeled neutral amino acids (glycine, L-alanine, L-valine,-aminoisobutyric acid) and an amino-acid mixture from ambient water across the body surface against considerable concentration gradients. Saturation kinetics and susceptibility of glycine uptake to competitive inhibition by alanine suggest mediated transport. Absorption of neutral amino acids is an active process. Exchange diffusion of preloaded-aminoisobutyric acid against external glycine or-aminoisobutyric acid could not be detected. Results on inhibition of glycine uptake by a variety of low-molecular-weight substances indicate that glycine absorption is highly specific for neutral amino acids and somewhat less for basic amino acids; it is unspecific for non--amino acids, acidic amino acids, carbohydrates, and organic acids. Rates of transintegumentary net influx of glycine are nearly identical to14C-glycine influx, suggesting that only small amounts of amino acids are released, as compared with the capacity for uptake. Thus,14C-amino-acid influx data are used for characterization of the uptake system. Glycine uptake is positively correlated to external salinity. In fresh water, absorption is nearly zero; between 10 and 20 S, uptake increases markedly reaching maximum values at 30 S; these remain almost constant at 40 S. Transport constants and maximum uptake rates increase with rising salinities. Since absorption of glycine and L-valine is susceptible to sodium depletion, similar mechanisms presumably underly salinity-dependent uptake of amino acids and sodium-dependent solute transport. Oxygen consumption is not significantly modified by different external salinities. Estimates of nutritional profit gained from absorption of amino acids vary between 4 and 15 % of metabolic rate for glycine absorption and between 10 and 39 % for uptake of an amino-acid mixture, according to external concentrations (10 and 50 µM) and salinities (20 and 30 S). 相似文献

We have cloned and characterized the expression of a decapentaplegic homologue (designated Ttu-dpp) from the oligochaete annelid Tubifex tubifex. RT-PCR analysis and in situ hybridization revealed that Ttu-dpp begins to be expressed around the time of the onset of ectodermal germ band (GB) elongation (i.e., the onset of gastrulation). At this time, Ttu-dpp expression is detected in the anteriormost part of the GBs. As development proceeds and the GBs elongate, the domain of Ttu-dpp-expressing cells extends posteriorly. Then Ttu-dpp-expressing cells within the GB are divided into two groups: one group occurs along the ventral midline and coincides with the domain of ventral ganglia; the other is located more dorsally. The latter group of Ttu-dpp-expressing cells subsequently undergoes dorsalward expansion, which results in the formation of a lateral stripe of cells in every segment except the first (i.e., segment I). In embryos that undergo body elongation (that is one of the last morphogenetic movements occurring prior to hatchout), Ttu-dpp expression in the lateral region is confined to setal sacs, which are arranged in the same transverse plane around the periphery of each segment (except segment I). 相似文献

Enchytraeus japonensis is a highly regenerative oligochaete annelid that can regenerate a complete individual from a small body fragment in 4–5 days.
In our previous study, we performed complementary deoxyribonucleic acid subtraction cloning to isolate genes that are upregulated
during E. japonensis regeneration and identified glutamine synthetase (gs) as one of the most abundantly expressed genes during this process. In the present study, we show that the full-length sequence
of E. japonensis glutamine synthetase (EjGS), which is the first reported annelid glutamine synthetase, is highly similar to other known class
II glutamine synthetases. EjGS shows a 61–71% overall amino acid sequence identity with its counterparts in various other
animal species, including Drosophila and mouse. We performed detailed expression analysis by in situ hybridization and reveal that strong gs expression occurs in the blastemal regions of regenerating E. japonensis soon after amputation. gs expression was detectable at the cell layer covering the wound and was found to persist in the epidermal cells during the
formation and elongation of the blastema. Furthermore, in the elongated blastema, gs expression was detectable also in the presumptive regions of the brain, ventral nerve cord, and stomodeum. In the fully formed
intact head, gs expression was also evident in the prostomium, brain, the anterior end of the ventral nerve cord, the epithelium of buccal
and pharyngeal cavities, the pharyngeal pad, and in the esophageal appendages. In intact E. japonensis tails, gs expression was found in the growth zone in actively growing worms but not in full-grown individuals. In the nonblastemal
regions of regenerating fragments and in intact worms, gs expression was also detected in the nephridia, chloragocytes, gut epithelium, epidermis, spermatids, and oocytes. These results
suggest that EjGS may play roles in regeneration, nerve function, cell proliferation, nitrogenous waste excretion, macromolecule

The uptake of chemicals in soil organisms, especially earthworms, has been studied many times. However, in Europe no internationally accepted standardised test guideline for the assessment of bioaccumulation in the soil ecosystem exists. Therefore, the German Federal Environmental Agency recently funded a project in which a standardisable test method for measuring bioaccumulation of chemicals using earthworms and enchytraeids is being developed. In this contribution, initial results with the new method are presented, using two model chemicals (the insecticide lindane and the fungicide hexachlorobenzene). Two enchytraeid species (Enchytraeus luxuriosus and Enchytraeus albidus) were selected as test organisms due to their easy handling and their important ecological role in the soil compartment. Artificial soil and a natural standard soil were used as test substrates. Test concentrations were based on previous results of acute and reproduction toxicity tests performed with the same species. Uptake as well as the elimination of the test substances were examined under standardised conditions in a closed test system. The first results show that both chemicals were accumulated considerably by both enchytraeid species. The bioaccumulation factors (BAFs) of lindane and hexachlorobenzene found for enchytraeids are significantly higher compared to those for lumbricid earthworms. Evaluation of the preliminary data suggests that the smaller species E. luxuriosus accumulated the two chemicals to a greater extent than E. albidus. In most cases, both chemicals were eliminated completely. The use of this new test method appears suitable for the ecotoxicological risk assessment of bioaccumulative chemicals. 相似文献

Diastereoisomeric 4-substituted acidic amino acids occur in characteristic associations in the green parts of some species of the Filicinae. Subspecies of Phyllitis scolopendrium accumulate 2(S),4(R)-4-methylglutamic acid, 2(S)-4-methyleneglutamic acid and the two diastereoisomers of 2(S)-4-hydroxy-4-methylglutamic acid, the last two occurring at relative concentrations of 3: 1. All Asplenium species investigated were distinctive in accumulating 2(S),4(R)-4-methylglutamic acid, the two diastereoisomers of 2(S)-4-hydroxy-4-methylglutamic acid, and the two diastereoisomers of 2(S)-4-hydroxy-2-aminopimelic acid in a characteristic concentration ratio. Some Polystichum species do not accumulate 4-substituted acidic amino acids whereas others accumulate both diastereoisomers of 2(S)-4-hydroxy-4-methylglutamic acid and 'of 2(S)-4-hydroxy-2-aminopimelic acid, and thus resemble Asplenium species. The seasonal variation in the concentration of 4-substituted acidic amino acids in the green parts of Phyllitis, Asplenium and Polystichum species has also been determined. 相似文献

Abstract. Life-history attributes and the reproductive biology of Enchytraeus coronatus (Oligochaeta, Enchytraeidae) were studied in agar cultures, as a basis for using this species in toxicity tests and other experimental studies. The use of agar allows daily examination of the behavior, reproduction, and mortality of the worms. At 20°C, E. coronatus had a generation time of 21–22 days and an embryonic period of 7–10 days. Hatch rate was high (>80%) and worms became sexually mature 8–10 days after hatching. Cocoon production and number of eggs per cocoon were positively correlated. Mortality in the first stages of the life cycle (embryonic period and first week after hatching) was about 38%. Three different periods of cocoon production were distinguished: a period characterized by a regular increase in the number of cocoons produced (weeks 4–9), a period of production at a constant high rate of ∼5 cocoons per week (weeks 10–20), and a period of decline in production rate (weeks 21–32), down to a mean value of 1.6 cocoons per adult in the last week. This study showed that synchronized agar cultures of cohorts can provide adults for long-term experimental tests, with the age for optimum reproduction being 8–20 weeks. 相似文献

Although regeneration studies are useful for understanding how organs renew, little information is available about regeneration of reproductive organs and germ cells. We here describe the behavior of germ-cell precursors during regeneration of the oligochaete annelid worm Enchytraeus japonensis, which has the remarkable feature of undergoing asexual (by fission) and sexual reproduction . We first found that the gonad can regenerate from any body fragment yielded by fission during asexual reproduction. We then examined behavior of germ-cell lineage during this regenerative process, by using a homolog of the Piwi gene (Ej-piwi) as a marker. We found that in asexually growing animals, specialized cells expressing Ej-piwi are distributed widely in the body as single cells. These cells seem to serve as a reservoir of germ-cell precursors because during asexual propagation these cells migrate into the regenerating tissue, where they ultimately settle in the prospective gonads, and give rise to germ cells upon sexualization. These cells are distinct from the neoblasts, thought to be stem cells in other animals. This is the first report to directly show that the germ and somatic lineages are segregated in asexually growing animals and behave differently during regeneration. 相似文献

The terrestric oligochaete worm Enchytraeus buchholzi survives in cadmium (Cd)-polluted environments by aid of its Cd-inducible 25 kDa cysteine-rich protein (CRP). Here, we analyze promoter and structure of the crp gene and compare its relationship to MT genes. The crp gene, approximately 12 kbp long, consists of 10 exons with exons 2 to 9 encoding eight almost identical repeats of predominantly 31 amino acids of the CRP. The introns of the crp gene contain various repetitive elements including retrotransposon-like sequences. The 683-bp promoter of the non-constitutive crp gene exhibits a much higher basal activity than the mouse MT-II promoter in HepG2 cells. Essential for crp promoter activity is the distal region (-683/-521) with a GC box and the proximal region (-308/-8) with the four MREa, b, c, d and AP-1, -2, -3 elements, whereas the central portion (-521/-309) with CAAT box, CRE and a XRE causes promoter repression. The TATA box-, MREc- and the AP-2, -3-containing region are required for high crp promoter activity. Our data support the view that the crp gene is a unique MT-gene and has evolved by exon duplications from a MT-like ancestral gene. 相似文献
