Signal I, the EPR signal of P-700, induced by long flashes as well as the rate of linear electron transport are investigated at partial inhibition of electron transport in chloroplasts. Inhibition of plastoquinol oxidation by dibromothymoquinone and bathophenanthroline, inhibition of plastocyanin by KCN and HgCl2, and inhibition by 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide are used to study a possible electron exchange between electron-transport chains after plastoquinone. (1) At partial inhibition of plastocyanin the reduction kinetics of P-700+ show a fast component comparable to that in control chloroplasts and a new slow component. The slow component indicates P-700+ which is not accessible to residual active plastocyanin under these conditions. We conclude that P-700 is reduced via complexed plastocyanin. (2) The rate of linear electron transport at continuous illumination decreases immediately when increasing amounts of plastocyanin are inhibited by KCN incubation. This is not consistent with an oxidation of cytochrome f by a mobile pool of plastocyanin with respect to the reaction rates of plastocyanin being more than an order of magnitude faster than the rate-limiting step of linear electron transport. It is evidence for a complex between the cytochrome b6 - f complex and plastocyanin. The number of these complexes with active plastocyanin is concluded to control the rate-limiting plastoquinol oxidation. (3) Partial inhibition of the electron transfer between plastoquinone and cytochrome f by dibromothymoquinone and bathophenanthroline causes decelerated monophasic reduction of total P-700+. The P-700 kinetics indicate an electron transfer from the cytochrome b6 - f complex to more than ten Photosystem I reaction center complexes. This cooperation is concluded to occur by lateral diffusion of both complexes in the membrane. (4) The proposed functional organization of electron transport from plastoquinone to P-700 in situ is supported by further kinetic details and is discussed in terms of the spatial distribution of the electron carriers in the thylakoid membrane.  相似文献

The pgr5 mutant of Arabidopsis thaliana has been described as being deficient in cyclic electron flow around photosystem I, however, the precise role of the PGR5 protein remains unknown. To address this issue, photosynthetic electron transport was examined in intact leaves of pgr5 and wild type A. thaliana. Based on measurements of the kinetics of P700 oxidation in far red light and re-reduction following oxidation in the presence of DCMU, we conclude that this mutant is able to perform cyclic electron flow at a rate similar to the wild type. The PGR5 protein is therefore not essential for cyclic flow. However, cyclic flow is affected by the pgr5 mutation under conditions where this process is normally enhanced in wild type leaves, i.e. high light or low CO2 concentrations resulted in enhancement of cyclic electron flow. This suggests a different capacity to regulate cyclic flow in response to environmental stimuli in the mutant. We also show that the pgr5 mutant is affected in the redox poising of the chloroplast, with the electron transport chain being substantially reduced under most conditions. This may result in defective feedback regulation of photosynthetic electron transport under some conditions, thus providing a rationale for the reduced efficiency of cyclic electron flow.  相似文献

The oxidation and reduction of cytochrome f and P-700 is measured spectrophotometrically in leaves of low-light and high-light plants. After illumination with red light, an induction phenomenon for cytochrome f oxidation is observed which indicates a regulation of photosystem I activity through energy distribution between the pigment systems by the energy state of the membrane. After far-red excitation the reduction of cytochrome f in the dark is much slower in low-light leaves. This shows that cyclic electron transport is not improved in low-light plants under these conditions. P-700 is oxidized on excitation with far-red light. However, with high intensities of far-red light, P-700 is partially reduced again which is due to a low extent of photosystem II excitation with the far-red used in the experiments. The low-light leaves show greater sensitivity of photosystem II to this excitation. The initial rate of the cytochrome f oxidation-rate is the same in low-light and high-light leaves. This shows that several P-700 are connected with only one electron transport chain. The consequences of these results concerning the tripartite concept and the photosynthetic unit are discussed. In the high-light plants the experimental data can be well explained by the tripartite organization of the photosynthetic unit. In low-light plants, however, a multipartite organization has to be postulated. The reduction rate of oxidized reaction center chlorophyll of Photosystem I after laser-flash excitation at 25 K has been determined for D-144 subchloroplast fragments and chloroplasts. A maximum of 40% of Photosystem I reaction centers undergo irreversible charge separation (P-700, Cluster A: P-700+, Cluster A?) at 25 K, a percentage which is independent of laser-flash intensity. The remaining reaction centers in chloroplasts and D-144 fragments undergo reversible charge separation with biphasic recombination. Similar amplitudes and time constants (chloroplasts, 49 μs (61%); D-144 fragments, 90 μs (67%)) were obtained for the fast component, while the slower component differed considerably in time (chloroplasts, 2.9 ms; D-144 fragments, 170 ms). It is known that Fe-S Cluster A is photoreduced in less than 1 ms at 25 K. Data obtained support a model for Photosystem I involving a single intermediate in the decay path between the reduced primary electron acceptor (A?1) and P-700+ and a second intermediate in the decay path between a reduced secondary electron acceptor and P-700+. Dual laser-flash experiments to determine rate constants for these processes are included.  相似文献

Geum montanum L. is an alpine plant usually found at altitudes between 1700 and 2600 m. Its wintergreen leaves can be subjected to very low temperatures and at the same time receive high photon flux densities at the beginning of the growth season when the snow melts. We report results of a study, performed with classical methods of biophysics, showing that leaves of G. montanum were remarkably tolerant to sunlight even at low temperatures. This tolerance results from the interplay of photorespiration and CO2 photosassimilation. When temperatures approach 0°C, responses include stomatal opening and CO2 uptake even under desiccation stress. This permits linear electron transport that is sufficient to avoid the excessive reduction of the electron transport chain which is known to lead to photodamage. In addition, excitation energy was shifted from photosystem (PS)II to PSI which is a very efficient energy quencher. Sensitivity of P700 in PSI to oxidation by far-red light was decreased and rates of dark reduction of photooxidized P700 were increased by actinic illumination, suggesting activation of cyclic electron transport. Consistent with this, far-red light was able to decrease the quantum yield of PSII (measured by the F v/F m ratio of chlorophyll fluorescence). We suggest that cyclic electron transport decreases the lumenal pH under strong light. In the presence of zeaxanthin, this increases energy dissipation at the PSII level. At low temperatures, P700 remained strongly oxidized under high irradiation while the primary electron acceptor of PSII, QA, was largely reduced. This shows efficient control of electron transport presumably at the level of the cytochrome b/f complex and suggests formation of a protective transthylakoid proton gradient even when linear electron transport is much reduced in the cold. Thus, several mechanisms cooperate to effectively protect the photosynthetic apparatus of G. montanum from photodamage. We see no indication of destructive “photostress” in this species during the growth season under alpine low-temperature and drought conditions. The yield of the triplet state of the primary electron donor of Photosystem I of photosynthesis (PT-700) and the characteristic parameters (g value, line shape, saturation behavior) of the ESR signal of the photoaccumulated intermediary acceptor A have been measured for two types of Photosystem I subchloroplast particles: Triton particles (TSF 1, about 100 chlorophyll molecules per P-700) that contain the iron-sulfur acceptors FX, FB and FA, and lithium dodecyl sulfate (LDS) particles (about 40 chlorophyll molecules per P-700) that lack these iron-sulfur acceptors. The results are: (i) In Triton particles the yield of PT-700 upon illumination is independent of the redox state of A and of FX,B,A and is maximally about 5% of the active reaction centers at 5 K. The molecular sublevel decay rates are kx = 1100 s?1 ± 10%, ky = 1300 s?1 ± 10% and kz = 83 s?1 ± 20%. In LDS particles the triplet yield decreases linearly with concentration of reduced intermediary acceptors, the maximal yield being about 4% at 5 K assuming full P-700 activity. (ii) In Triton particles the acceptor complex A consists of two acceptors A0 and A1, with A0 preceding A1. In LDS particles at temperatures below ?30°C only A0 is photoactive. (iii) The spin-polarized ESR signal found in the time-resolved ESR experiments with Triton particles is attributed to a polarized P-700-A?1 spectrum. The decay kinetics are complex and are influenced by transient nutation effects, even at low microwave power. It is concluded that the lifetime at 5 K of P-700A0A?1 must exceed 5 ms. We conclude that PT-700 originates from charge recombination of P-700A?0, and that in Triton particles A0 and A1 are both photoaccumulated upon cooling at low redox potential in the light. Since the state P-700AF?X does not give rise to triplet formation the 5% triplet yield in Triton particles is probably due to centers with damaged electron transport.  相似文献

After transferring the dark-acclimated cyanobacteria to light, flavodiiron proteins Flv1/Flv3 serve as a main electron acceptor for PSI within the first seconds because Calvin cycle enzymes are inactive in the dark. Synechocystis PCC 6803 mutant Δflv1flv3 devoid of Flv1 and Flv3 retained the PSI chlorophyll P700 in the reduced state over 10?s (Helman et al., 2003; Allahverdiyeva et al., 2013). Study of P700 oxidoreduction transients in dark-acclimated Δflv1flv3 mutant under the action of successive white light pulses separated by dark intervals of various durations indicated that the delayed oxidation of P700 was determined by light activation of electron transport on the acceptor side of PSI. We show that the light-induced redox transients of chlorophyll P700 in dark-acclimated Δflv1flv3 proceed within 2?min, as opposed to 1–3?s in the wild type, and comprise a series of kinetic stages. The release of rate-limiting steps was eliminated by iodoacetamide, an inhibitor of Calvin cycle enzymes. Conversely, the creation with methyl viologen of a bypass electron flow to O2 accelerated P700 oxidation and made its extent comparable to that in the wild-type cells. The lack of major sinks for linear electron flow in iodoacetamide-treated Δflv1flv3 mutant, in which O2- and CO2-dependent electron flows were impaired, facilitated cyclic electron flow, which was evident from the decreased steady-state oxidation of P700 and from rapid dark reduction of P700 during and after illumination with far-red light. The results show that the photosynthetic induction in wild-type Synechocystis PCC 6803 is largely hidden due to the flavodiiron proteins whose operation circumvents the rate-limiting electron transport steps controlled by Calvin cycle reactions.  相似文献

Light quality was shown to exert well-coordinated regulatory effects on the composition and function of the thylakoid membranes as well as on the photosynthetic rates of intact leaves from Atriplex triangularis grown in continuous blue, white and red lights (50 μE · m?2 · s?1). The higher photosynthetic rates in plants grown in blue light, as compared to those in white and red lights, resulted from marked changes in both light-harvesting complexes and electron carriers. The concentrations of electron carriers such as atrazine binding sites, plastoquinone, cytochromes b and f and P-700 on a chlorophyll basis were markedly increased in Atriplex grown in blue light; and the apparent light-harvesting antenna unit sizes of Photosystems I and II were greatly reduced. Consequently, the electron transport capacities of Photosystems I and II were also increased as was the coupling factor CF1 activity. Atriplex grown in red light had lower photosynthetic rates than those grown in blue or white light by incorporating changes in the composition and function of the thylakoids in a direction opposite to those caused by growth in blue light. When these regulatory effects of light quality were compared with those of light quantity [6,7], it is clear that ChlaChl b ratios, electron transport capacities of Photosystems I and II, concentrations of plastoquinone, atrazine binding sites, coupling factor CF1 activity and the apparent antenna unit size of Photosystem II are more affected by light quantity, whereas light quality has a greater influence on the concentration of P-700, the apparent antenna unit size of Photosystem I and the overall photosynthetic rates of intact leaves.  相似文献

Photosynthetic electron flux from water via photosystem II (PSII) and PSI to oxygen (water-water cycle) may act as an alternative electron sink under fluctuating light in angiosperms. We measured the P700 redox kinetics and electrochromic shift signal under fluctuating light in 11 Camellia species and tobacco leaves. Upon dark-to-light transition, these Camellia species showed rapid re-oxidation of P700. However, this rapid re-oxidation of P700 was not observed when measured under anaerobic conditions, as was in experiment with tobacco performed under aerobic conditions. Therefore, photo-reduction of O2 mediated by water-water cycle was functional in these Camellia species but not in tobacco. Within the first 10 s after transition from low to high light, PSI was highly oxidized in these Camellia species but was over-reduced in tobacco leaves. Furthermore, such rapid oxidation of PSI in these Camellia species was independent of the formation of trans-thylakoid proton gradient (ΔpH). These results indicated that in addition to ΔpH-dependent photosynthetic control, the water-water cycle can protect PSI against photoinhibition under fluctuating light in these Camellia species. We here propose that the water-water cycle is an overlooked strategy for photosynthetic regulation under fluctuating light in angiosperms.  相似文献

The flash-induced absorption transient at 698 nm in a Photosystem I subchloroplast particle showed the following characteristics after addition of 0.25–2.0% lithium dodecyl sulfate (LDS). (i) The 30-ms transient corresponding to the P-700+ P-430 backreaction was replaced by a 1.2-ms transient. (ii) The amplitude of the transient did not change immediately after LDS addition, but decayed with a half-life of 10 min at pH 8.5. (iii) Methyl viologen had no effect on the magnitude or kinetics of the transient, indicating that it cannot accept an electron from this component. (iv) The difference spectrum of the transient from 400 nm to 500 nm was characteristic of an iron-sulfur protein. (v) The transient followed first-order Arrhenius behavior between 298 K and 225 K with an activation energy of 13.3 kJ/mol; between 225 K and 77 K, the 85-ms half-time remained temperature-invariant. These properties suggest that the LDS-induced absorption transient corresponds to the P-700+ A2 change recombination seen in the absence of a reduced electron-acceptor system. In the presence of LDS, the reaction-center complex was dissociated, allowing removal of the smaller peptides from the 64-kDa P-700-containing protein. With prolonged incubation, the iron-sulfur clusters were destroyed through conversion of the labile sulfide to zero-valence sulfur. About 35% of the zero-valence sulfur was found associated with the 64-kDa protein under conditions that allowed separation of the small peptides. We interpret the long lifetime of the P-700+ A2 transient after LDS addition and the association of zero-valence sulfur with a 64-kDa protein to indicate that A2 is closely associated with, and perhaps integral with, the P-700-containing protein.  相似文献

The plastid terminal oxidase PTOX is a plastohydroquinone:oxygen oxidoreductase that is important for carotenoid biosynthesis and plastid development. Its role in photosynthesis is controversially discussed. Under a number of abiotic stress conditions, the protein level of PTOX increases. PTOX is thought to act as a safety valve under high light protecting the photosynthetic apparatus against photodamage. However, transformants with high PTOX level were reported to suffer from photoinhibition. To analyze the effect of PTOX on the photosynthetic electron transport, tobacco expressing PTOX‐1 from Chlamydomonas reinhardtii (Cr‐PTOX1) was studied by chlorophyll fluorescence, thermoluminescence, P700 absorption kinetics and CO2 assimilation. Cr‐PTOX1 was shown to compete very efficiently with the photosynthetic electron transport for PQH2. High pressure liquid chromatography (HPLC) analysis confirmed that the PQ pool was highly oxidized in the transformant. Immunoblots showed that, in the wild‐type, PTOX was associated with the thylakoid membrane only at a relatively alkaline pH value while it was detached from the membrane at neutral pH. We present a model proposing that PTOX associates with the membrane and oxidizes PQH2 only when the oxidation of PQH2 by the cytochrome b6f complex is limiting forward electron transport due to a high proton gradient across the thylakoid membrane.  相似文献

Absorbance changes ΔA 810 were measured in pea (Pisum sativum L., cv. Premium) leaves to track redox transients of chlorophyll P700 during and after irradiation with far red (FR) light under various preillumination conditions in the absence and presence of inhibitors and protonophorous uncoupler of photosynthetic electron transport. It was shown that cyclic electron transport (CET) in chloroplasts of pea leaves operates at its highest rate after preillumination of leaves with white light and is strongly suppressed after preillumination with FR light. The FR light-induced suppression was partly released during prolonged dark adaptation. Upon FR illumination of dark-adapted leaves, the induction of CET was observed, during which CET activity increased to the peak from the low level and then decreased gradually. The kinetics of P700 oxidation induced by FR light of various intensities in leaves preilluminated with white light were fit to empirical sigmoid curves containing two variables. In leaves treated with a protonophore FCCP, the amplitude of FR light-induced changes ΔA 810 was strongly suppressed, indicating that the rate of CET is controlled by the pH gradient across the thylakoid membrane.  相似文献

A model of primary photosynthetic reactions in the thylakoid membrane was developed and its validity was tested by simulating three types of experimental kinetic curves: (1) the light-induced chlorophyll a fluorescence rise (OJIP transients) reflecting the stepwise transition of the photosynthetic electron transport chain from the oxidized to the fully reduced state; (2) the dark relaxation of the flash-induced fluorescence yield attributed to the QA? oxidation kinetics in PSII; and (3) the light-induced absorbance changes near 820 or 705 nm assigned to the redox transitions of P700 in PSI. A model was implemented by using a rule-based kinetic Monte-Carlo method and verified by simulating experimental curves under different treatments including photosynthetic inhibitors, heat stress, anaerobic conditions, and very high light intensity.  相似文献

In addition to compounds which inhibit the function of calmodulin (a ubiquitous calcium regularity protein found in plants and mammalian tissues), Ca2+ or Cl channel blockers in mammalian tissues were also found to inhibit electron transport in Photosystem II submembrane preparations. Their inhibition was overcome by electron donation to P-680 by diphenylcarbazide (for all of the compounds used) and by H2O2 (except with trifluoperazine). Addition of Ca2+ and/or Cl also partially prevented the inhibitory action. We postulate that the inhibitory action occurs at the level of the water-splitting system at the site of Ca2+ modulation of the Cl cofactor requirement for O2 evolution (as hypothesized in Nakatani, H.Y. It is still a controversial issue how the electron transport reaction is carried out around photosystem I (PSI) in the photosynthetic electron transport chain. The measurable component in PSI is the oxidized P700, the reaction center chlorophyll in PSI, as the absorbance changes at 820–830 nm. Previously, the quantum yield at PSI [Y(I)] has been estimated as the existence probability of the photo-oxidizable P700 by applying the saturated-pulse illumination (SP; 10,000–20,000 µmol photons m?2 s?1). The electron transport rate (ETR) at PSI has been estimated from the Y(I) value, which was larger than the reaction rate at PSII, evaluated as the quantum yield of PSII, especially under stress-conditions such as CO2-limited and high light intensity conditions. Therefore, it has been considered that the extra electron flow at PSI was enhanced at the stress condition and played an important role in dealing with the excessive light energy. However, some pieces of evidence were reported that the excessive electron flow at PSI would be ignorable from other aspects. In the present research, we confirmed that the Y(I) value estimated by the SP method could be easily misestimated by the limitation of the electron donation to PSI. Moreover, we estimated the quantitative turnover rate of P700+ by the light-to-dark transition. However, the turnover rate of P700 was much slower than the ETR at PSII. It is still hard to quantitatively estimate the ETR at PSI by the current techniques.

Thiocapsa. roseopersicina BBS has four active [NiFe] hydrogenases, providing an excellent opportunity to examine their metabolic linkages to the cellular redox processes. Hyn is a periplasmic membrane-associated hydrogenase harboring two additional electron transfer subunits: Isp1 is a transmembrane protein, while Isp2 is located on the cytoplasmic side of the membrane. In this work, the connection of HynSL to various electron transport pathways is studied. During photoautotrophic growth, electrons, generated from the oxidation of thiosulfate and sulfur, are donated to the photosynthetic electron transport chain via cytochromes. Electrons formed from thiosulfate and sulfur oxidation might also be also used for Hyn-dependent hydrogen evolution which was shown to be light and proton motive force driven. Hyn-linked hydrogen uptake can be promoted by both sulfur and nitrate. The electron flow from/to HynSL requires the presence of Isp2 in both directions. Hydrogenase-linked sulfur reduction could be inhibited by a QB site competitive inhibitor, terbutryne, suggesting a redox coupling between the Hyn hydrogenase and the photosynthetic electron transport chain. Based on these findings, redox linkages of Hyn hydrogenase are modeled.  相似文献

Summary The light curve of Cl-uptake (uptake vs. light intensity) byElodea densa in pure N2 shows that saturation is reached at a very low light intensity. In N2+3% CO2, on the other hand, there is considerably less Cl- uptake. Under these conditions, the saturation attained at low light intensity is only temporary, and the Cl--uptake increases steadily with a further rise in light intensity. It is suggested that the reason for the low intensity of light saturation may be the necessity for an intracellular transport of ATP from the site of its formation to the site of Cl--uptake.CO2 exerts a strong inhibitory influence on the Cl--uptake, especially at low light intensities. At higher intensities the inhibition diminishes and it is nearly absent at high intensities of white light.The inhibition by CO2 is also a function of the wavelength of the light; it is greatest in the region below 683 nm, where photosynthesis occurs with high efficiency, but it is still present at wavelengths beyond 700 nm.CO2 also inhibits the Cl--uptake at high light intensities of white light when small concentration of DCMU (5×10-7 M) is present at the same time.The inhibitory action of CO2 is partly interpreted as a consequence of a competition for ATP between CO2-assimilation (espectially below 683 nm) and the light-dependent Cl--uptake. In addition, however, it is suggested that at low light intensities the presence of CO2 effects a regulation between noncyclic and cyclic electron transports and photophosphorylation which is supposed to be a consequence of a change in the redox potential of ferredoxin or another cofactor acting in noncyclic and cyclic electron transports. Especially the inhibition of the Cl--uptake by CO2 in far-red light (>700 nm) and in the presence of DCMU is taken to be an indication of this intracellular regulation.
