  • 1.1. The growth characteristics of Burkitt's lymphoma cells in suspension culture have been studied, and a mean population doubling time of 20–21 hr established for this cell line under a range of nutritional and physical conditions; data which have provided a basis for the assessment of the reproducibility of the culture techniques and conditions which were employed in the subsequent studies.
  • 2.2. Activities of lactate dehydrogenase (LDH), aldolase and esterase, as well as the cellular content of total soluble protein, and the isoenzyme pattern of LDH, were monitored in randomly growing Raji cells for the duration of a complete growth cycle.
  • 3.3. In this period, the temporal pattern of variation in the levels of total soluble protein were seen to reflect alterations in LDH activity during a single growth cycle.
  • 4.4. The fluctuations observed in LDH activity were greater than those observed for either aldolase or esterase activity, and, from the data considered, the maximum degree of variation appeared to be confined to the initial stages of growth.
  • 5.5. Extracellular levels of LDH activity remained relatively constant throughout the growth cycle. so that the large fluctuations in intracellular LDH activity could not be attributed to either secretion or leakage of the enzyme into the culture medium.
  • 6.6. No gross changes in the pattern of LDH isoenzymes in these Raji cells were detected during the course of a single growth cycle.

  • 1.1. Groups of mice were subjected to different degrees of thiamine deprivation in their diet. In particular, the effects of complete thiamine deficiency and a continuation of minimal nutritional levels of thiamine were compared.
  • 2.2. The effects of these treatments on the turnover characteristics of lactate dehydrogenase and total soluble protein have been studied by means of double labelling experiments, and determinations of the relative emphases of synthesis and degradation of these tissue components.
  • 3.3. Marked divergences from normal were apparent with each of these nutritional regimens-complete thiamine deficiency causing a considerably increased rate of degradation for both total protein and lactate dehydrogenase in all tissues; whereas maintenance of minimal levels of thiamine led to increased degradation of total protein in liver, but reduced rates of degradation for lactate dehydrogenase in brain, heart and liver.
  • 4.4. The significance of these results has been discussed in relation to the relative influence of vitamin and calorie deficiencies on turnover parameters, the individuality of specific tissue behaviour, differences in protein redistribution in response to separate physiological perturbations, and the role of thiamine in specific proteolysis.

  • 3.3. The A4 homopolymer appears simultaneously with initial muscle contractility and is correlated with the physiological function of muscular contraction.
  • 4.4. The activation of the Ldh-C locus is correlated with the first functioning of liver. It is suggested that the state of differentiation of liver cells may be the stimulus required for C locus expression.

  • 1.1. The effects of niacin deficiency on the relative turnover rates of proteins in various tissues of Japanese quail were investigated.
  • 2.2. The level of liver NAD was not affected by niacin deficiency whereas the level of pectoral muscle NAD was markedly reduced.
  • 3.3. In all dietary treatments the liver had the highest turnover rates of proteins, heart and brain had intermediate rates, and pectoral muscle had the lowest rates.
  • 4.4. Relative turnover rates of proteins in all tissues (particularly pectoral muscle) of the niacin deficient group were significantly higher than those of pair-fed control group, although there were no significant differences in turnover rate between pair-fed control and control groups.
  • 5.5. The high turnover rate of proteins in niacin deficiency was primarily attributed to enhanced degradation rate of proteins rather than enhanced synthesis rate of proteins.
  • 6.6. Optical density scanning (or densitometric) of water-soluble pectoral muscle proteins separated by isoelectric focusing revealed several additional minor protein bands between major protein bands in the niacin deficient group which were more pronounced in the acidic region of the gel.
  • 7.7. These results suggest that proteins with a low pI value in pectoral muscle of the niacin deficient animal are highly sensitive to protein degradation.

  • 1.1. The carcinoma showed higher enzyme activities than the normal mammary tissue.
  • 2.2. The ratios of glutamate dehydrogenase, glutathione reductase and catalase to lactate dehydrogenase were lower in carcinomas than in normal tissues. Similarly, the ratios of glutamate dehydrogenase, glutathione reductase and catalase to glucose-6-phosphate dehydrogenase were also significantly lower in carcinomas.
  • 3.3. There were no significant differences in enzyme activities between stages I and II of disease, however in the metastatic tissues, there were significant differences between stages I and II.
  • 4.4. SH groups were higher in the tissues of cancer patients than in normal tissues. The levels of thiols groups were higher in carcinomas at stage III of disease.

  • 2.2. During pre-pupal growth, LDH is the most active enzyme, followed by MDH and α-GPDH; while during post-pupal growth, MDH is most active followed by α-GPDH and LDH.
  • 3.3. Increased enzyme activity is in response to changing physiological and physical environments, and is due to temporal activation of new gene loci indicated by the production of new α-GPDH and MDH isozymes. LDH activity is probably controlled by the temporal action of regulatory gene(s).

  • 1.1. Opine dehydrogenases (OpDHs) and lactate dehydrogenase (LDH) activities were determined in various marine animals. OpDHs were detected in six marine invertebrate phyla; Porifera, Coelenterata, Annelida, Mollusca, Arthropoda and Echinodermata in phylogenic sequence.
  • 2.2. Among several OpDHs, tauropine dehydrogenase (TaDH) occurred widely in marine invertebrates, from Porifera to Echinodermata.
  • 3.3. With a few exceptions, total OpDHs activities exceeded that of LDH activity in the marine invertebrates investigated.
  • 4.4. With respect to anaerobic glycolysis, OpDHs are indicated to play an important role in phylogenically lower invertebrates, whereas LDH is more important in higher animals.

  • 1.1. Activities of several enzymes and protein constituents of magnum, isthmus, shell gland and breast muscle (pectoralis major) of Japanese quail, Coturnix coturnix japonica were compared.
  • 2.2. The respective activity per g wet weight of lactate dehydrogenase, malate dehydrogenase and l-glycerol 3-phosphate dehydrogenase in pectoralis major was approx 20 times that of these enzymes in isthmus which showed the highest activity among oviducal tissues. On the other hand, the respective activity of lactate dehydrogenase was similar to that of malate dehydrogenase and was approx 10–20 times that of l-glycerol 3-phosphate dehydrogenase in each tissue.
  • 3.3. Among NADPH-producing enzymes, NADP+-isocitrate dehydrogenase showed the highest activity in all tissues. The activity of malic enzyme was lowest in oviducal tissues, but was next to NADP+-isocitrate dehydrogenase in pectoralis major.
  • 4.4. Sodium dodecyl sulfate polyacrylamide gel electrophoretic patterns of the whole homogenate, the supernatant and the precipitate fraction of magnum, isthmus, shell gland and pectoralis major showed tissue specific protein compositions. Proteins with molecular weight of 55,000, 70,000, 110,000 and 130,000 were observed in the respective precipitate (myofibrilar) fraction of magnum, isthmus and shell gland, but not in that of pectoralis major. It was noteworthy that the amount of myosin heavy chain of magnum was markedly less than that of other three tissues.

  • 1.1. Role of NADP-glutamate dehydrogenase in the depletion of citrate was analyzed using permeabilized yeast cells.
  • 2.2. Citrate was converted to 2-oxoglutarate, which was then metabolized to glutamate by NADP-glutamate dehydrogenase in the presence of ammonium ion.
  • 3.3. Formation of 2-oxoglutarate plus glutamate was in good agreement with the concentration of citrate decreased. Glutamate formation can be a good indicator of the depletion of citrate, because 70% of the citrate decreased was converted to glutamate.
  • 4.4. Glycolytic activity was closely correlated with the decrease in citrate under the in situ conditions.
  • 5.5. NADP-glutamate dehydrogenase increased in anaerobically grown yeast cells.
  • 6.6. An effective depletion of citrate by increased synthesis of NADP-glutamate dehydrogenase can explain the lowered mechanism of citrate causing glycolytic stimulation under the anaerobic growth conditions of yeast.

  • 4.4. During the larval development, both L1 and L2 remain at a low level.
  • 5.5. After the larvae hatch L1 and L2 gradually rise although L1 is predominant.
  • 6.6. Measurement of kinetic parameters shows that the general behaviour of the enzymes of the embryo resembles that of the adult enzymes.
  • 7.7. However, one can observe during the development a constant increase in the affinity of the enzyme towards its substrate, lactate.

  • 1.1. Lactate dehydrogenase in ovine tissues was separated by electrophoresis, one dimensional isoelectric focusing (IEF) and a two dimensional technique.
  • 2.2. Tissues showed five zones of enzyme activity consisting of multiple bands after IEF.
  • 3.3. The IEF zymograms were unique for each tissue and differed from those of other species.
  • 4.4. Two dimensional separation revealed that the five zones of activity observed on IEF corresponded to the five isoenzymes separable by conventional electrophoresis.

  • 3.3. Considerable variation was evident between the individual tissues, with synthesis being indicated as most rapid in intestine while degradation was highest in liver.

  • 1.1. The extent of fatty acid synthesis from [1-14C]acetate in liver slices was reduced 6-fold when eels were fasted for 1–7 weeks and 20-fold when fasted for 39 weeks; thereafter hepatic lipogenesis seemed to remain constant for up to 95 weeks of fasting.

  • 1.1. Various tissues of the porcupine Hystrix hodgesoni including liver, intestine, stomach, spleen, kidney, brain and lung were examined for the presence of growth hormone binding sites.
  • 2.2. Membranes were prepared from the aforementioned tissues and tested for binding to 125I-bovine growth hormone (125I-bGH).
  • 3.3. Porcupine kidney membranes yielded 1.3 and 2.7% specific binding when tested at 1000 and 2500 μg protein, respectively. Porcupine liver membranes demonstrated approximately 1% specific binding at 3000 μg protein. The other tissues gave low specific binding. The results indicate that porcupine kidney contained binding sites for growth hormone.
  • 4.4. Various tissues of two teleosts, the snakehead Channa maculata and the winter founder Pleuronectes americanus, were similarly processed and tested for binding to 125I-bGH. It was found that among the different tissues studied, the liver membranes of Channa maculata and the gonad membranes of Pleuronectes americanus gave the highest specific binding of 125I-bGH.
  • 5.5. Liver and intestine membranes of the lamprey Petromyzon marinus did not bind 125I-bGH.

  • 1.1. The double isotope ratios of individual lipid fractions in major tissues of mice were determined before and after a period of total dietary deprivation.
  • 2.2. Several significant alterations in these ratios were caused by this treatment, with a marked individuality evident in the response of separate tissues.
  • 3.3. The increased metabolic flux overall was contributed to by increased rates of lipid degradation in liver, and active mobilization from the peripheral depots.
  • 4.4. Significant utilization of phospholipid fractions for energy purposes was also noticeable, however. along with tissue specific variation in the patterns of both double isotope ratios and specific radioactivities.
  • 5.5. These data are discussed in relation to the comparative priorities of synthesis and degradation in the different tissue and lipid sources in response to this physiological perturbation.

  • 1.1. The activities of all the eight enzymes of conversion of fructose to glucose, of all the three key enzymes of glycolysis and of the two dehydrogenases of pentose shunt were determined in proximal and distal mucosa of small intestine.
  • 2.2. With the exception of hexokinase, all of these enzymes have an activity significantly higher in the proximal than distal mucosa.
  • 3.3. The gradient along the intestine is particularly important for the three enzymes which are typical for fructose metabolism (ketohexokinase, triokinase and fructose-1-phosphate aldolase), for glucose-6-phosphatase and for phosphofructokinase.
  • 4.4. The effects of fructose diet on the enzyme activities are compatible with the results, described in other papers, concerning the final products of metabolism.
  • 5.5. The increase of fructose metabolism appears to result mainly from the stimulation of the activities of ketohexokinase and fructose-1-phosphate aldolase which control all the pathways of ketohexose utilization.
  • 6.6. The activation of glucose-6-phosphatase, in comparison with the other enzymes which are involved in glucose-6-phosphate metabolism, explains the appearance of the ability to synthesize glucose with fructose as substrate. This enzyme is the only key enzyme of fructose to glucose conversion which responds to fructose feeding in distal mucosa.
  • 7.7. The activities of hexokinase and phosphofructokinase are not increased by fructose feeding.
  • 8.8. The activity of pyruvate kinase. the only key glycolytic enzyme which is necessarily implicated when fructose is the substrate, is stimulated but less than the typical enzymes of fructose metabolism.
  • 9.9. But, because of its quantitative importance, the glycolytic pathway is responsible for the most part of the observed increase of fructose utilization.
  • 10.10. The responses of pyruvate kinase, glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase activities to fructose feeding are similar in the two parts of small intestine.
  • 11.11. The activities of ketohexokinase, triokinase and glucose-6-phosphate isomerase are stimulated only in the proximal small intestine mucosa.
  • 12.12. The other enzyme activities which are stimulated in proximal segment are also increased in distal segment.
  • 13.13. All segments of small bowel show adaptive changes to dietary manipulation but not necessarily for all their functions.
  • 14.14. The gradient of enzyme activities from the proximal to the distal small intestine persists despite dietary modification, but the data do not determine that this gradient is intrinsic or that it is not intrinsic.

  • 1.1. Activities of the three ammonia-forming enzymes, glutamate dehydrogenase, AMP deaminase and serine dehydrase (SerDH), were measured in tissues of gill, digestive diverticula, mantle and foot muscle of the brackish-water bivalve Corbicula japonica.
  • 2.2. High levels of SerDH activity were detected in gill and digestive diverticula, while the activity levels of the other two enzymes were low.
  • 3.3. The result suggests the significance of SerDH in amino acid degradation of this species.
