The activity in the gustatory nerve from the anterior part of the tongue, the chorda tympani proper nerve, has been recorded during stimulation of the tongue of a New World monkey, Saguinus midas tamarin. A series of 0.3 M sugars and 0.07 M NaCl, 0.07 M sucrose, 0.004 M acetic acid and 0.00005 M quinine hydrochloride were used as taste stimuli. The concentrations of the last four stimuli were the same as those earlier found to be the lowest concentrations at which this monkey in behavioural experiments discriminates between them and water. The records showed that these threshold concentrations all elicited a neural response. Further, the neural activities during stimulation with the series of 0.3 M sugars were recorded. Using the amplitudes of the responses as a measure, the order between them was found to be fructose greater than glucose greater than lactose greater than arabinose greater than sucrose = galactose greater than raffinose. This order was discussed and related with the order found in behavioural experiments. With both methods, fructose seemed to be the strongest stimulus, but then the order among the sugars differed.  相似文献

The suitability of various nectar and honeydew sugars as a food source for the polyphagous ant species M. rubra (L.) was studied. The sugars used included monosaccharides (fructose, glucose, galactose, mannose, rhamnose), disaccharides (sucrose, maltose, trehalose, melibiose, lactose) and trisaccharides (melizitose, raffinose, erlose). Single-sugar solutions were tested on ant workers in a long-term laboratory bioassay in which acceptance of the solutions and ant survival were recorded. The acceptance of the sugars was confirmed in a second bioassay in which feeding time was established. Enzymatic hydrolysis of sucrose, maltose and melibiose was investigated through HPLC analyses of workers fed these disaccharides. Sugar acceptance and feeding time were related to ant survival. Considering the monosaccharide units of which the sugars are composed, fructose seems especially suitable as a short-term energy source, while glucose appears to be used both directly and for storage. The presence of a galactose unit appears to reduce sugar suitability. It is suggested that the workers possess invertase and maltase and to a lesser degree also galactosidase. The gustatory perception is correlated with the profitability of sugars in further metabolic processes.  相似文献

To clarify developmental changes in the gustatory system of the rat, integrated taste responses from the chorda tympani (CT) nerve were recorded and analyzed at different postnatal ages. The response magnitude was calculated relative to the response to the standard, 0.1 M NH4Cl. Even at 1 week of age, the CT responded well to all tested 0.1 M chloride salts (NH4Cl, NaCl, LiCl, KCl, RbCl and CsCl). The responses to 0.1 M NaCl and LiCl increased with increasing age of the rat while response magnitudes to KCl, RbCl and CsCl did not change up to 8 weeks. At 1 week, the integrated response pattern was quite similar to that in adult rats for NaCl, HCl and quinine hydrochloride (QHCl). The concentration-response functions for NaCl, HCl, QHCl and sucrose at 2 weeks were essentially the same as those at 8 weeks. These results suggest that taste buds in the 2-week-old rat are functionally mature for the detection of the four basic taste stimuli. The relative magnitude of the responses to the various sugars was smaller at 1 week compared to the adult rat and reached a maximum at weeks 3-4, then decreased gradually with age. Among the six sugars, sucrose was the most effective followed by lactose. From weeks 1-4, the magnitude of the integrated taste response to fructose was smaller than that to lactose except at 3 weeks of age. Maltose, galactose and glucose were less potent stimuli than the other sugars tested. The response magnitude to lactose at 4 weeks had decreased compared to that for the other sugars. Taste responses to the sugars in preweanling and adult rats were not cross-adapted by the individual sugars. These results suggest that after 1 week of age during postnatal development in the rat, taste information from the CT rapidly increases in its importance for feeding behavior.  相似文献

Summary The utilization of some mono- and oligosaccharides by the members of Choanephoraceae has been studied in detail. The filtrate was analysed by using circular paper chromatography. Amongst the seven monosaccharides tested, viz., glucose, galactose, fructose, mannose, xylose, sorbose and rhamnose, the first five were completely utilized within the specified period, while sorbose and rhamnose remained in the medium throughout the incubation period. A mixture of glucose, galactose and fructose was found to support better growth of all the present species, than that when these sugars were supplied singly. Out of the four oligosaccharides tested, only maltose could be hydrolysed, and it was completely consumed within the specified period. The other three oligosaccharides, viz., sucrose, lactose and raffinose were not hydrolysed and they remained in the medium throughout the incubation period.  相似文献

A synthetic culture medium which supports a high level of growth of a scrially propagated cell suspension culture of Acer pseudoplatanus is described. The sucrose of this medium can be effectively replaced by glucose or fructose or a mixture of glucose and fructose or galactose or maltose or soluble starch. When the carbohydrate is glucose or fructose no other sugars appear in the culture medium in significant amounts. Glucose is absorbed in greater quantity than fructose from an equimolar mixture of these sugars. When sucrose is supplied both glucose and fructose appear in the medium. Glucose appears in maltose medium, and maltose and glucose in soluble starch medium. Under the standard conditions of culture, media containing 2 % sucrose or 2 % glucose become depleted of sugar before the 25th day of incubation. Enhanced yield of the cultures can be obtained by raising the initial sucrose concentration to 6 %. – A supply of nitrate is essential for maximum yield and healthy growth. Growth, in the presence of nitrate, is significantly enhanced by a supply of urea. Addition of casein hydrolysate or of a mixture of amino acids enhances growth in the presence of nitrate and urea and particularly when nitrate is omitted. – When kinetin is omitted or incorporated at the standard level (0.25 mg/I), 2,4-dichlorophenoxyacetic acid (2,4-D) at 1.0 mg/l is essential for continuation of growth at a high level. It cannot be replaced by indol-3yl-acetic acid (IAA). 1-naphthaleneacetic acid (NAA) at 10 mg/l permits of a low level of growth with abnormal aggregation. When the level of kinetin is raised to 10 mg/l a high level of growth occurs in the absence of added auxin but the cultures become brown and tend to show increasing aggregation on subculture.  相似文献

1. Intracellular recordings of mouse taste cell responses were made using a glass micro-electrode filled with Procion yellow dye solution. 2. Six sugars (sucrose, maltose, lactose, glucose, galactose and fructose) produced the depolarization responses. 3. Gustatory cross adaptation between sugars was determined. When the taste cell was pre-adapted with one of the six sugars, the other five sugars, cross adapted, produced depolarization, hyperpolarization or null responses. 4. From these observations, it is suggested that there are multiple sugar receptor sites on the receptor membrane of the mouse taste cell.  相似文献

Adult parasites, Isoparorchis hypselobagri were maintained in vitro in four basic salt solutions, viz. Stephenson saline, AB saline, Tyrode's solution and Locke's solution. Survival was prolonged by adding sugars to Tyrode's solution. Of all the sugars, glucose was found most effective and parasites survived for 39 days in solution containing glucose, 6 days in galactose and sucrose, 10 days in fructose and lactose and 12 days in maltose. The parasites absorb carbohydrates through the cuticle. Ligated parasites survived for a longer period than unligated parasites. The pH 9 was considered as an optimum for maximum survival of the parasites Isoparorchis hypselobagri for 39 days.  相似文献

Glucose, maltose, sucrose, lactose, xylose, sorbose, galactose, fructose and gluconolactone were analyzed by means of immobilized pyranose oxidase as well as by the combination of immobilized glucose oxidase with immobilized glycoamylase, invertase, mutarotase, maltase (α-glucosidase) and glucose isomerase by flow injection analysis (FIA). For the simultaneous analysis of glucose and other sugars three different flow-injection configurations were applied and compared. The average error of prediction of the analyses were better than 3% in model media and better than 6% in yeast extract containing media.  相似文献

Fifth-stadium nymphs of the potato leafhopper, Empoasca fabae (Harris), were offered several concentrations of a number of carbohydrates contained in an agar-based medium. Imbibition (gustatory) response was based on how rapidly the nymphs became red from the rhodamine B dye incorporated into the media. Survival was expressed as the cumulative hours that nymphs remained alive on the sugar-containing media.Sucrose elicited a phagostimulatory response over the range of 0.0075 to 0.4 M. Glucose was phagostimulatory at 0.12 M, and lactose at 0.2 M. Concentrations of fructose, trehalose and raffinose equal to or greater than 0.4 M were phago-inhibitory while sucrose was inhibitory at 1 M.Raffinose, trehalose, maltose, sucrose, glucose, fructose and galactose all enhanced leafhopper survival, whereas lactose, rhamnose, and xylose were deleterious at concentrations of 0.12 M or greater. Four basic types of survival response are described.
Résumé Diverses concentrations de plusieurs hydrates de carbone contenus dans un substrat alimentaire à 2% d'agar, tamponné avec O, IM d'histidine (pH 6.1) ont été offertes à des larves au 5ème stade d'Empoasca fabae. La rapidité de la réaction d'absorption (gustative) est mesurée d'après la vitesse avec laquelle les larves sont colorées en rouge par la rhodamine B à 0,005% incorporée dans l'aliment. La survie est exprimée par le nombre d'heures pendant lequel les larves restent vivantes sur le substrat alimentaire contenant le sucre.Entre 0,0075 et 0,4 M, le sucrose provoque une phagostimulation; le glucose est phagostimulant à 0,12 M, et le lactose à 0,2 M. Les concentrations de fructose, de tréhalose, et de raffinose, égales ou supérieures à 0,4 M, sont phagoinhibitrices, tandis que le sucrose est inhibiteur à 1M.Le raffinose, le tréhalose, le maltose, le sucrose, le glucose, le fructose et le galactose prolongent tous la survie, tandis que le lactose, le rhamnose et le xylose sont nuisibles à partir de 0,12 M.Quatre types fondamentaux de survie ont été observés.Type I: La survie augmente rapidement avec la concentration en sucre, atteignant un maximum ou devenant proche du maximum à 0,03 M et restant élevée aux concentrations supérieures jusqu'à 0,2 M. Ultrasonic vocalizations were recorded from 10-day-old albinorats while they were isolated from their dam and siblings. Eachrat received a 1.0% BW intra-oral infusion of sucrose, fructose,glucose or lactose at a concentration range of 0.22–0.66M for 3 min and their vocalizations were determined during theinfusion and for an additional 7 min. Sucrose, fructose andglucose all significantly reduced vocalizations to about 50%of baseline levels, whereas lactose, the milk sugar, was ineffective.Moreover, the dose–response function was flat for thethree effective sugars. In a second experiment, the effectsof these sugars on heat escape latency were measured. Sucrose,fructose and glucose each elevated the latency with which infantrats removed a paw from a 48°C surface; lactose did not.These findings of lactose ineffectiveness and the flat dose-responsefunction for the other three sugars exactly parallel those obtainedfor human newborns. Summary An investigation was made of the effects of temperature and of mono- and disaccharides on lipids, biomass, odd-chain and unsaturated fatty acids production of Candida lipolytica. With different sugars as carbon source at 30°C, the order for biomass production was: fructose > glucose > sucrose > lactose > galactose, while lipids production/g biomass decreased as follows: lactose, sucrose, galactose, fructose and glucose. On the other hand, the odd-chain fatty acids contents decreased in the following order: fructose, lactose, glucose, sucrose and galactose. Lowering the temperature of cultivation to 15°C, biomass, lipids and unsaturated fatty acids decreased. However, a notable decrease in the content of odd-chain fatty acids was detected.  相似文献

Red beet hairy root cultures, obtained after genetic transformation with Agrobacterium rhizogenes, are completely heterotrophic and synthesize betalaines (BNs). Upon subjecting the hairy roots to treatments containing different sugars (3% w/v) it was found that sucrose was rapidly utilized, followed by maltose, and a very limited use of glucose, but the other hexoses – fructose, lactose, xylose and galactose or glycerol totally suppressed both growth and BN synthesis. No habituation or adaptability to maltose or glucose occurred, evidenced by the lack of growth upon re-culture in respective medium. Glycerol, was not taken up alone, but was utilized to a considerable extent in the presence of low levels of sucrose for growth only but not BN synthesis. Red beet hairy root culture did not exogenously hydrolyse sucrose to hexoses, as there were only traces of reducing sugar present in the medium soon after inoculation, without an increase later, confirmed by HPLC. There was an increase in medium osmolarity in the presence of fructose indicating the exudation of certain compounds from the roots. Red beet hairy roots appear useful as a model system to study sugar metabolism/signalling due to their sensitivity to different sugars that may directly link to morphological changes and BN synthesis.  相似文献

Responses of three groups of neural fibers from the chorda tympani of the hamster to binary mixtures of taste stimuli applied to the tongue were analyzed. The groups displayed different sensitivities to six chemicals at concentrations that had approximately equal effects on the whole nerve. Sucrose-best fibers responded strongly only to sucrose and D-phenylalanine. NaCl-best and HCl-best fibers, responded to four electrolytes: equally to CaCl2 and nearly equally to HCl, but the former responded more to NaCl, and the latter responded more to NH4Cl. The groups of fibers dealt differently with binary mixtures. Sucrose- best fibers responded to a mixture of sucrose and D-phenylalanine as if one of the chemicals had been appropriately increased in concentration, but they responded to a mixture of either one and an electrolyte as if the concentration of sucrose or D-phenylalanine had been reduced. NaCl- best fibers responded to a mixture as if it were a "mixture" of two appropriate concentrations of one chemical, or somewhat less. But, responses of HCl-best fibers to mixtures were greater than that, approaching a sum of responses to components. These results explain effects on the whole nerve, suggest that the sensitivity of a mammalian taste receptor to one chemical can be affected by a second, which may or may not be a stimulus for that receptor, and suggest that some effects of taste mixtures in humans may be the result of peripheral processes.  相似文献

Unimbibed Amaranthus caudatus seeds were found to contain stachyose, raffinose, verbascose, sucrose, galactinol, myo-inositol, glucose and fructose, while no galactose, maltose and maltotriose was detected. During imbibition, seed concentrations of verbascose, stachyose, raffinose, galactinol, myo-inositol (temporary) and fructose (transient) were observed to decrease; concentrations of galactose and maltose remained fairly constant, while those of sucrose, glucose and maltotriose increased, the increase in sucrose concentration was only temporary. Effects of gibberellin A3 (GA3) at 3 × 10−4 M and ethephon at 3 × 10−4 M alone or in the presence of methyl jasmonate (Me-JA) at 10−3 M on concentrations of soluble sugars during germination of A. caudatus seeds were examined. Me-JA was found to inhibit seed germination and fresh weight of the seeds, but did not affect sucrose, myo-inositol, galactose and maltose concentrations during imbibition for up to 20 h. The exogenously applied GA3 was observed to enhance germination, stachyose breakdown and glucose concentration after 20 h of incubation. Ethephon stimulated seed germination as well as utilisation of stachyose, galactinol (both after 14 and 20 h) and raffinose (after 14 h of incubation). Although the stimulatory effect of either GA3 or ethephon on seed germination was blocked by Me-JA; these stimulators increased mobilisation of raffinose and stachyose, but only ethephon enhanced both glucose and fructose after 14 and/or 20 h of incubation in the presence of Me-JA. The maltose concentration was increased by both GA3 and ethephon alone and in the presence of Me-JA. Of the growth regulators studied, ethephon alone and/or in combination with Me-JA significantly increased the concentrations of glucose, fructose, galactose, maltose and maltotriose. The differences in sugar metabolism appear to be linked to ethylene or GA3 applied simultaneously with Me-JA.  相似文献

The inductive effect of different sugars on beta-galactosidase synthesis in Phycomyces blakesleeanus has been studied. The enzyme was inducible by galactose and fructose. When grown on these sugars the enzyme level was 10-20 times greater than when grown on glucose. We have detected both intra- and extracellular beta-galactosidase activity when Phycomyces blakesleeanus was grown on galactose, but only extracellular beta-galactosidase activity when grown on fructose plus lactose.  相似文献

Fructose, galactose, glucose, maltose, melibiose, and trehalose were evaluated for their nutritional and survival values for Aedes aegypti. The development of the exogenous stages of Plasmodium gallinaceum was evaluated by counting and averaging the number of oocysts developing on the midguts of Aedes aegypti.The nutritional and survival value for A. aegypti was greatest on glucose, sucrose, and fructose and lowest on galactose and melibiose. Mosquitoes maintained on fructose produced the greatest numbers of oocysts. Only two other sugars, galactose and melibiose produced more oocysts than their respective controls. Glucose and maltose, both of which had high nutritive and survival value for A. aegypti were less efficient than the control (sucrose) for the development of P. gallinaceum.  相似文献

The effects of sugars (xylose, arabinose, fucose, fructose, galactose, glucose, sorbitol, maltose, sucrose, and lactose; 0-20% w/v) on the properties of the pressure-induced gel from a whey protein isolate (20%, 800 MPa, 30 degrees C, 10 min) were studied. All the sugars decreased the hardness, breaking stress and water-holding capacity of the gel at the same concentration of 55.5 mM. Increasing the sugar content changed the microstructure of the gel from a honeycomb-like structure to a stranded structure, while the strand thickness was progressively reduced. These results suggest that sugars decreased the degree of intermolecular S-S bonding of proteins and non-covalent interaction, and restrained the phase separation during gelation under high pressure.  相似文献

The effects of adding sugars to high- and low-tar cigarettes on the mutagenicity of their smoke condensates were studied using Salmonella typhimurium TA100 and TA98 with and without metabolic activation. The sugars tested were glucose, fructose, galactose, sorbitol, sucrose and lactose. The lowest mutagenicities observed with these sugars per mg of smoke condensate assayed on TA98 with metabolic activation were 37% (high-tar cigarettes) and 22% (low-tar cigaretts) of that of smoke condensate from untreated cigarettes. Addition of sugars increased the total amounts of smoke condensates, but the mutagenicities of the total condensates were also decreased by all the sugars, the lowest values being 35% (high-tar cigarettes) and 36% (low-tar cigarettes) of that of smoke condensates from cigarettes without added sugar. On assay with TA100 with metabolic activation, decreases in both specific and total mutangenicities of condensates of high-tar cigarettes were observed with all the sugars tested except galactose and sucrose. Treatment with glucose, fructose or sorbitol decreased the specific mutagenicity of condensates of low-tar cigarettes and glucose and fructose reduced also their total mutagenicity. The effects of added sugars were more marked when assayed on TA98 than on TA100 and of the sugars tested fructose and sorbitol had the greatest effects. Addition of sugars had no effect of the mutagenicity of cigarette-smoke condensate without metabolic activation.  相似文献
