This study illustrates the benefits of Celite® supported lipase sol–gels for the transesterification of triolein to produce methyl oleate. A ping–pong bi–bi kinetic model was developed and validated taking into account the inhibition effects of methanol and glycerol as well as the effect of temperature. Although initial reaction rate models are useful for predicting the kinetics in the absence of products, a kinetic model beyond the initial conditions that considers glycerol inhibition is important. The model developed was consistent with the experimental data (R2 = 0.95) predicting an increase in methyl oleate production with increasing methanol concentration up to an optimal range of 1.3 M to 2.0 M depending on the temperature. In general, increasing the temperature increased the initial reaction rate for the immobilized lipase over the temperature range of 40–60 °C. Based on the kinetic constants, the maximum velocity of the reverse reaction is about 25% slower than that of the forward reaction and glycerol inhibition has a more significant effect on the reaction kinetics than methanol inhibition. The model developed would be useful for understanding the effects of methanol and glycerol inhibition as well as temperature on the production of methyl oleate using lipase-mediated enzymatic transesterification.  相似文献

A mild and efficient method for the conversion of fatty acid methyl esters from lard into ascorbyl esters via lipase-catalyzed transesterification in co-solvent mixture is described. A solvent engineering strategy was firstly applied to improve fatty acid ascorbyl esters production. The co-solvent mixture of 30% t-pentanol:70% isooctane (v/v) was optimal. Response surface methodology (RSM) and central composite design (CCD) were employed to estimate the effects of reaction parameters, such as reaction time (12–36 h), temperature (45–65 °C), enzyme amount (10–20%, w/w, of fat acid methyl esters), and substrate molar ratio of fatty acid methyl esters to ascorbic acid (8:1–12:1) for the synthesis of fatty acid ascorbyl esters in co-solvent mixture. Based on the RSM analysis, the optimal reaction conditions were determined as follows: reaction time 34.32 h, temperature 54.6 °C, enzyme amount 12.5%, substrate molar ratio 10.22:1 and the maximum conversion of fatty acid ascorbyl esters was 69.18%. The method proved to be applicable for the synthesis of ascorbyl esters using Novozym 435 in solvent.  相似文献

(−)-Epigallocatechin-3-O-gallate (EGCG) acetylated derivatives, which can be widely used as a natural antioxidant in both lipid containing food and cosmetic applications, were prepared by lipase catalyzed acylation of EGCG with vinyl acetate. Response surface methodology (RSM) and 5-level-4-factor central composite rotatable design (CCRD) were employed to evaluate the effects of synthesis parameters, such as reaction time (6–10 h), temperature (30–50 °C), enzyme amount (1.5–2.5% (w/w) of substrate), and substrate molar ratio of EGCG to vinyl acetate (0.5–1.5) on conversion of EGCG. By using multiple regression analysis, the experimental data were fitted to a second order polynomial model. The most suitable combination of variables was 40 °C, 2.12%, 10 h and 1.13 for the reaction temperature, the enzyme amount, the reaction time, and EGCG/vinyl acetate mole ratio, respectively. At these optimal conditions, the conversion yield reached 87.37%. The presence of mono-, di- and tri-acetylated derivatives in acetylated EGCG was confirmed by LC–MS-MS and identified as 5″-O-acetyl-EGCG, 3″, 5″-2-O-acetyl-EGCG and 5′, 3″, 5″-3-O-acetyl-EGCG by NMR.  相似文献

Enzymatic syntheses of fatty acid anilides are important owing to their wide range of industrial applications in detergents, shampoo, cosmetics, and surfactant formulations. The amidation reaction of Mucor miehei lipase Lipozyme IM20 was investigated for direct amidation of triacylglycerol in organic solvents. The process parameters (reaction temperature, substrate molar ratio, enzyme amount) were optimized to achieve the highest yield of anilide. The maximum yield of palmitanilide (88.9%) was achieved after 24 h of reaction at 40 °C at an enzyme concentration of 1.4% (70 mg). Kinetics of lipase-catalyzed amidation of aniline with tripalmitin has been investigated. The reaction rate could be described in terms of the Michaelis–Menten equation with a Ping–Pong Bi–Bi mechanism and competitive inhibition by both the substrates. The kinetic constants were estimated by using non-linear regression method using enzyme kinetic modules. The enzyme operational stability study showed that Lipozyme IM20 retained 38.1% of the initial activity for the synthesis of palmitanilide (even after repeated use for 48 h). Palmitanilide, a fatty acid amide, exhibited potent antimicrobial activity toward Bacillus cereus.  相似文献

l-Ascorbyl laurate is a fatty acid derivative of l-ascorbic acid which can be widely used as a natural antioxidant in both lipid containing food and cosmetic applications. To avoid any possible harmful effects from chemically synthesized product, the enzymatic synthesis appears to be the best way to satisfy the consumer demand for natural antioxidants. The ability of immobilized lipase from Candida antarctica (Novozym® 435) to catalyze the direct esterification between l-ascorbic acid and lauric acid was investigated. Response surface methodology (RSM) and 5-level-4-factor central composite rotatable design (CCRD) were employed to evaluate the effects of synthesis parameters, such as reaction time (2–10 h), temperature (25–65 °C), enzyme amount (10–50% w/w of l-ascorbic acid), and substrate molar ratio of l-ascorbic acid to lauric acid (1:1–1:5) on percentage molar conversion to l-ascorbyl laurate. Based on the analysis result of ridge max, the optimal enzymatic synthesis conditions were predicted as follows: reaction time 6.7 h, temperature 30.6 °C, enzyme amount 34.5%, substrate molar ratio 1:4.3; and the optimal actual yield was 93.2%.  相似文献

In this paper, a reaction scheme for fructose production by inulin enzymatic hydrolysis is proposed, taking into account the possibility of dealing with a mixture of enzymes acting on a mixture of polymers as substrate. The scheme is subsequently simplified to obtain a stoichiometric relationship between the fructose product and the reacted substrate. The former may be measured by HPLC, while the latter is the subject of kinetic investigations. Our proposed kinetic model is defined within temperature and substrate concentration ranges of industrial interest (40–60 °C and 3–60 g/L, respectively). Some assumptions were made in order to simplify the model, which is based on a minimum number of parameters. These hypotheses were always specified and assumed only on the basis of convenience and rational consideration. Eventually, the kinetic model was successfully validated by comparison with a vast set of experimental results.  相似文献

The enzyme reaction mechanism and kinetics for biosyntheses of deoxycytidine triphosphate (dCTP) and deoxythymidine triphosphate (dTTP) from the corresponding deoxycytidine diphosphate (dCDP) and deoxythymidine diphosphate (dTDP) catalyzed by pyruvate kinase were studied. The kinetic model for the two synthetic reactions was found to follow the Bi–Bi random rapid equilibrium mechanism similar to that of the biosynthesis of deoxyadenosine triphosphate (dATP) and deoxyguanosine triphosphate (dGTP) from the corresponding deoxyadenosine diphosphate (dADP) and deoxyguanosine diphosphate (dGDP). Kinetic constants involved in the reactions including the maximum reaction velocity, the Michaelis–Menten constants, and the inhibition constants for dCTP and dTTP biosyntheses were experimentally determined. This enzyme reaction requires Mg2+ ion and the optimal Mg2+ concentration was also determined. The experimental results showed a good agreement with the simulation results obtained from the kinetic model developed. The kinetics of the four biosynthetic reactions for deoxynucleoside triphosphates (dNTP) including dATP, dGTP, dCTP, and dTTP from the corresponding deoxynucleoside diphosphates (dNDP) including dADP, dGDP, dCDP, and dTDP were analyzed. The results suggest that the binding kinetics of phosphoenolpyruvate (PEP) and pyruvate are similar for all four biosynthetic reactions. The affinity of the dNDP substrates to enzyme is of the same order of magnitude as the corresponding dNTP as inhibitors. The order of reactivity and substrate specificity for dNDP is dADP > dGDP > dCDP > dTDP in the pyruvate kinase (PK) reactions. The results obtained from this study can be applied to bioreactor design and production of dCTP and dTTP for biosynthesis of DNA at a significantly lower cost compared to the currently available chemical method.  相似文献

Biodegradation kinetic behaviors of n-butyl alcohol and sec-butyl alcohol in a composite bead biofilter were investigated. The microbial growth rate of n-butyl alcohol was greater than that of sec-butyl alcohol in the inlet concentration range of 50–300 ppm. The microbial growth rate was inhibited at higher inlet concentration, and the inhibitive effect in the concentration range of 50–150 ppm was more pronounced than that in the concentration range of 150–300 ppm. The degree of inhibitive effect for n-butyl alcohol was more sensitive than that for sec-butyl alcohol in the concentration range of 50–150 ppm. The zero-order kinetic with the diffusion rate limitation could be regarded as the most adequate biochemical reaction model. For the biochemical reaction process, the biochemical reaction rate coefficient of n-butyl alcohol was greater than that of sec-butyl alcohol in the inlet concentration range of 50–300 ppm. The biochemical reaction rate coefficient was decreased with increasing inlet concentration. The inhibitive effect for sec-butyl alcohol was more pronounced than that for n-butyl alcohol. The factor of the chemical structure of compound was more predominant in the microbial growth and biochemical reaction processes. The maximum elimination capacity of n-butyl alcohol and sec-butyl alcohol were 55.7 and 20.9 g C h?1 m?3 bed volume, respectively. The primary alcohol was easily biodegraded by the microbial.  相似文献

Aerobic granular sludge was cultivated in a glass sequencing batch reactor (SBR) with glucose synthetic wastewater. The spherical shaped granules were observed on 4th day with the mean diameter of 0.1 mm. With the increase of chemical oxygen demand (COD) concentration of the influent, aerobic granules grew matured, the size of which ranged from 1.2 to 1.9 mm. The aerobic granular sludge could sustain high organic loading rate (about 4.0 g COD L−1 d−1), with good settling ability (settling velocity 36 m/h) and high biomass concentration (MLSS 6.7 ±0.2 g/L). Experimental data indicated that the substrate utilization and biomass growth kinetics followed Monod's kinetics model approximately. The corresponding kinetic coefficients of maximum specific substrate utilization rate (k), half velocity coefficient (Ks), growth yield coefficient (Y) and decay coefficient (Kd) were 13.2 d−1, 275.8 mg/L, 0.183–0.250 mg MLSS/mg COD and 0.023–0.075 d−1, respectively, which made aerobic granules have short setup period, high rate of substrate utilization and little surplus sludge.  相似文献

Short chain alkyl esters are well appreciated for fruity flavors they provide. These are mainly applied to the fruit-flavored products like jam, jelly, beverages, wine and dairy. Cutinase from Burkholderia cepacia NRRL B 2320 was found to be active in catalyzing the synthesis of alkyl esters in organic solvent. The optimal temperature range for the enzyme catalyzed synthesis was found to be from 35 °C to 40 °C. The maximum conversion (%) during synthesis of ester was obtained for butyric acid (C4) and valeric acid (C5) with butanol reflecting the specificity of the enzyme for short-chain length fatty acids. In case of alcohol specificity, butanol was found to be most preferred substrate by the enzyme and conversion (%) decreased with increasing carbon chain length of alcohol used in the esterification reaction. The kinetic analysis for the synthesis of butyl butyrate by varying concentration of one substrate at a time (butanol or butyric acid), showed that Ping–Pong Bi Bi model with acid inhibition and influence of initial water is most suitable model for the prediction of the reaction kinetics.  相似文献

Hexyl laurate, a medium-chain ester carried about fruity flavor, is primarily used in personal care formulations as an important emollient for cosmetic applications. On the basis of the hexyl laurate could be successfully synthesized by lipase within a batch system in our previous report. This study aimed to develop an optimal continuous procedure of lipase-catalyzed hexyl laurate synthesis in a packed-bed bioreactor to investigate the possibility of large-scale production further. The ability of lipase from Rhizomucor miehei (Lipozyme IM-77) to catalyze the direct-esterification of 1-hexanol and lauric acid in n-hexane was investigated. Response surface methodology (RSM) and 3-level-3-factor fractional factorial design were employed to evaluate the effects of synthesis parameters, such as reaction temperature (35–55 °C), mixture flow rate (1.5–4.5 mL/min) and substrate molar ratio 1-hexanol to lauric acid (1:1–1:3) on production rate (μmol/min) of hexyl laurate by direct-esterification. Based on the analysis of ridge max, the optimum synthesis conditions for hexyl laurate were as follows: 45 °C of reaction temperature, substrate molar ratio 1:2 and reaction flow rate 4.5 mL/min. The optimum predicted production rate was 435.6 ± 0.9 μmol/min and the actual value was 437.6 ± 0.4 μmol/min.  相似文献

The kinetic data obtained from the action of a cathepsin D-like enzyme from Biomphalaria glabrata hepatopancreas (digestive gland) on MOCAc-Gly-Lys-Pro-Ile-Leu-Phe-Phe-Arg-Leu-Lys(DNp)-D–Arg-NH2, was studied as a data prototype, generated by means of a fluorogenic substrate. An initial fluorescence, due to incomplete energy transfer, of about 8% of the values attained after complete substrate hydrolysis; a non-linear standard curve even at μM concentrations and an exponential decay of the steady state fluorescence of reaction product of the order of 10 4 × s 1 were the main analytical problems encountered. The standard curves for fluorescence of the substrate reaction product after 48 h of hydrolysis, and the reference compound MOCAc-Pro-Leu-Gly-NH2, were fitted by polynomial approximation and the point derivates used as calibration factors. Time dependence of the calibration factor for the reaction product was − 2.96 × 10 4 a.u μM 1 × s 1 that is, in the same order of observed enzymic reaction rates. A mathematical treatment was devised for obtaining rates corrected for errors derived from the three analytical problems indicated. The method is of general application in continuous fluorometric assays, irrespective of the particular enzyme used, but of special value for substrates that present significant initial fluorescence. The reaction rates were 11% higher; as calculated by means of the calibration factor [substrate] ÷ (final  initial fluorescence intensities), which is the prevalent procedure in the literature; leading to underestimation of Km and overestimation of Vmax.  相似文献

The combined effects of initial sucrose and initial Remazol Turquoise Blue-G (RTBG) reactive dye concentrations on the specific growth rate and dye bioaccumulation efficiency of Candida utilis was investigated and optimized using response surface methodology (RSM) in this study. A 22 full factorial central composite design was successfully used for experimental design and analyses of the results. Two numerical correlations fitted to a second-order quadratic equation were obtained to estimate the responses of specific growth rate and dye uptake yield. The statistical analysis indicated that both the microbial growth and removal yield of dye enhanced with raising sucrose concentration up to 15 g l?1 and diminished with the increase in initial RTBG dye concentration up to approximately 500 mg l?1 due to inhibition caused by high concentrations of RTBG dye. The optimum combination predicted via RSM confirmed that C. utilis was capable of bioaccumulating RTBG with the maximum uptake yield of 82.0% in 15 g l?1 sucrose and 50 mg l?1 dye containing growth medium.  相似文献

Enzymatic methanolysis of canola oil in the solvent-free system was studied in a packed-bed reactor (PBR) using small pieces of loofa plus Novozym 435. Response surface methodology (RSM) was applied to determine the effect of the transesterification conditions, namely flow rate of substrate (x1), temperature (x2) and methanol to canola oil molar ratio (x3) as the regressors, on the methyl ester production. A central composite design (CCD) was employed to optimize the reaction. A second-order polynomial multiple regression model was chosen and analysis of variance (ANOVA) showed a high coefficient of determination (R2) value of 0.996, thus adjustment of the model with experimental data was ensured. The methyl ester yield increased as the flow rate of the reaction mixture in the PBR increased from its low to the middle level thereafter, increasing the flow rate corresponded to decreasing the yield. The same trends of changes were observed for the other two factors. The optimum process conditions for biodiesel production in the PBR were found to be: x1 = 6.3 mL/min, x2 = 38 °C and x3 = 4.3. The same batch was successfully used repeatedly in the PBR for six enzymatic cycles (432 h), where the methyl ester yield was maintained above 97%.  相似文献

4-Methoxy cinnamoyl glycerol (4MCG) is a very promising UV filters material in personal care products. In order to effectively improve the yield of 4MCG, a systematic study on ultrasonic pretreatment enzymatic esterification for 4MCG products was carried out. An ultrasonic frequency of 35 kHz, ultrasonic power of 150 W and ultrasound irradiation time of 1.5 h was determined to guarantee satisfactory degree of esterification and lipase activity. The optimum production was achieved in organic solvent system at 65 °C with 4MCA to glycerol molar ratio of 1:5, enzyme amount of 15 mg/mL, resulting in a monoester yield of above 66% and 55% after 48 h and 24 h of reaction under ultrasonic pretreatment, respectively. The experimental kinetic data were studied. The reactions were modeled by a system of sequential first-order rate expressions, kinetic parameters were estimated from experimental data fit to the model equations. Results show that the monoester yield in the ultrasonic pretreatment process (24 h) were above 1.5-fold as that in mechanical stirring process without essential damaging to lipase activity. The enzymatic method using ultrasonic pretreatment was obviously superior to the mechanical stirring for enzymatic method and chemical method in terms of conversion rate and the monoester yield. These results are of great significance for applying ultrasonic pretreatment method to prepare 4MCG.  相似文献

Experimental determination of the separate effects of palm oil and methanol concentrations on the rate of their enzymatic transesterification was used to propose suitable mechanismic steps and to test the generated kinetic model. The reaction took place in n-hexane organic medium and the lipase used was from Mucor miehei. At a constant methanol concentration of 300 mol m−3, it was found that, initially as the palm oil concentration increased, the initial reaction rate increased. However, the initial rate dropped sharply at substrate concentrations larger than 1250 mol m−3. Similar behaviour was observed for methanol concentration effect, where at a constant substrate concentration of 1000 mol m−3, the initial rate of reaction dropped at methanol concentrations larger than 3000 mol m−3. Ping Pong Bi Bi mechanism with inhibition by both reactants was adopted as it best explains the experimental findings. A mathematical model was developed from a proposed kinetic mechanism and was used to identify the regions where the effect of inhibition by both substrates arised. The proposed model equation is essential for predicting the rate of methanolysis of palm oil in a batch or a continuous reactor and for determining the optimal conditions for biodiesel production.  相似文献

Phenylacetaldehyde (PA) can be produced by the oxidation of 2-phenylethanol (PE) through biotransformation. In order to prevent substrate and product inhibitions and the transformation of the PA to phenylacetic acid (PAA), utilization of a two-phase system is very attractive. Gluconobacter oxydans B-72 was used as the microorganism and iso-octane as the solvent. The effect of initial substrate concentration on the PA production was investigated in single- and two-phase systems. In the single-phase system, substrate inhibition occurred above 5 g/l, and in the two-phase system, above 7.5 g/l. Substrate inhibition kinetics were also studied in the two-phase system and kinetic constants were determined as rmax=0.64 g/l min, KM=8.15 g/l, KPA=2.5 g/l. Because it was observed that two-phase system is insufficient to remove the substrate inhibition effect, fed-batch operation was utilised in this study. For 7.5 g/l of PE, 1.65, 3.85, and 7.35 g/l of PA were obtained in the single-phase, two-phase, and two-phase three fed-batch systems, respectively. Effect of biotransformation time, initial substrate concentration, agitation speed, and fed-batch number on the PA production was investigated in a two-phase fed-batch system by the response surface methodology (RSM). The optimum values were found as 3 fed-batch number, 2.75 g/l initial substrate concentration, 150 rpm agitation speed, and 65 min of one batch biotransformation time. In order to verify these results, an experiment was performed at these optimum conditions and 7.10 g/l of PA concentration was obtained.  相似文献

(S)-(+)-2-Chlorophenylglycine 1 is an important intermediate in the synthesis of Clopidogrel. A recirculating packed bed reactor (RPBR) was constructed for efficient production of (S)-1 by kinetic resolution of racemic N-phenylacetyl-2- chlorophenylglycine 2 using immobilized penicillin G acylase (PGA). The immobilized PGA exhibited maximum activity at 50 °C and pH 8.0 with (R,S)-2 as substrate. The kinetic constants (Km and vmax) of immobilized PGA were calculated to be 20.61 mM and 83.2 mM/min/g, respectively. The substrate displayed inhibitory effect on immobilized PGA with inhibition constant of 221.23 mM. The immobilized PGA showed a strict enantiospecificity for substrate at different temperature, pH and substrate concentration examined. The performance and productivity of RPBR were evaluated by several critical parameters, including immobilized PGA load, substrate feeding rate, height to diameter ratio and so on. The kinetic resolution process shows higher initial reaction rate and conversion by recycling 100 mL of substrate solution (80 mM) through RPBRs packed with 6.0 g immobilized PGA with a feeding rate of 1.5 mL/min while the H/D ratio was 4.0. The immobilized PGA-catalyzed kinetic resolution of (R,S)-2 was successfully operated in the RPBR for 60 batches, with an average productivity of 1.2 g/L/h for (S)-1 in high optical purity (>97% enantiomeric excess) in semi-continuous operation. The residual (R)-2 can be easily racemized and then used as substrate.  相似文献

This study aimed to optimize the culture conditions (agitation speed, aeration rate and stirrer number) of hyaluronic acid production by Streptococcus zooepidemicus. Two optimization algorithms were used for comparison: response surface methodology (RSM) and radial basis function neural network coupling quantum-behaved particle swarm optimization algorithm (RBF-QPSO). In RBF-QPSO approach, RBF is employed to model the microbial HA production and QPSO algorithm is used to find the optimal culture conditions with the established RBF estimator as the objective function. The predicted maximum HA yield by RSM and RBF-QPSO was 5.27 and 5.62 g/l, respectively, while a maximum HA yield of 5.21 and 5.58 g/l was achieved in the validation experiments under the optimal culture conditions obtained by RSM and RBF-QPSO, respectively. It was indicated that both models provided similar quality predictions for the above three independent variables in terms of HA yield, but RBF model gives a slightly better fit to the measured data compared to RSM model. This work shows that the combination of RBF neural network with QPSO algorithm has good predictability and accuracy for bioprocess optimization and may be helpful to the other industrial bioprocesses optimization to improve productivity.  相似文献

A biocatalyst with high activity retention of lipase was fabricated by the covalent immobilization of Candida rugosa lipase on a cellulose nanofiber membrane. This nanofiber membrane was composed of nonwoven fibers with 200 nm nominal fiber diameter. It was prepared by electrospinning of cellulose acetate (CA) and then modified with alkaline hydrolysis to convert the nanofiber surface into regenerated cellulose (RC). The nanofiber membrane was further oxidized by NaIO4. Aldehyde groups were simultaneously generated on the nanofiber surface for coupling with lipase. Response surface methodology (RSM) was applied to model and optimize the modification conditions, namely NaIO4 content (2–10 mg/mL), reaction time (2–10 h), reaction temperature (25–35 °C) and reaction pH (5.5–6.5). Well-correlating models were established for the residual activity of the immobilized enzyme (R2 = 0.9228 and 0.8950). We found an enzymatic activity of 29.6 U/g of the biocatalyst was obtained with optimum operational conditions. The immobilized lipase exhibited significantly higher thermal stability and durability than equivalent free enzyme.  相似文献
