This study focuses on evaluating the degradation of n-hexane/methanol mixture in trickle-bed-air-biofilters (TBABs). Two different concentration ratios of methanol:n-hexane were evaluated (3:1) for TBAB “A” and (5:1) for TBAB “B”. Both TBABs were run and fed with nutrients buffered at pH 4 for encouraging the growth of fungi. The TBABs were loaded with pelletized diatomaceous earth support media and were run at an empty bed residence time of 120 s. n-Hexane loading rates (LRs) ranged from 0.9 to 13.2 g/m3 h for both TBABs. The corresponding methanol LRs varied from 2.3 to 37.7 g/m3 h and from 4.6 to 64.5 g/m3 h for TBABs “A” and “B”, respectively. Experimental results have shown that the degradation of n-hexane in presence of methanol is enhanced for n-hexane LRs less than 10.6 g/m3 h as compared to previous study for sole-fed n-hexane, but for n-hexane LRs of 13.2 g/m3 h, the performance of TBABs in eliminating n-hexane depended on the methanol to n-hexane ratios. The impact was less severe for TBAB “A” (RE 85%) as compared to TBAB “B” (RE 72%). This is attributed to the high LRs of methanol in TBAB “B”. n-Hexane performance stability was another advantage attained.  相似文献

Comparing analytical results for Sulfite Pretreatment to Overcome the Recalcitrance of Lignocelluloses (SPORL) of Douglas-fir (Pseudotsuga menziesii) at two different temperatures shows that the apparent activation energy of sugar degradation is higher than that of hemicellulose hydrolysis, approximately 161 kJ/mole versus 100 kJ/mole. Thus, one can balance the production of degradation products against hemicellulose hydrolysis and therefore the enzymatic saccharification efficiency of the resultant substrate by changing pretreatment temperature and duration. Specifically, pretreatment at 165 °C for 75 min significantly reduced furan formation compared with the pretreatment at 180 °C for 30 min while maintaining the same pretreatment severity and therefore the same substrate enzymatic digestibility (SED). Obtaining high SED with Douglas-fir is also limited by lignin content. Fortunately, the bisulfite in SPORL provides delignification activity. By combining kinetic models for hemicelluloses hydrolysis, sugar degradation, and delignification, the performance of pretreatment can be optimized with respect to temperature, duration, acid, and bisulfite loading. The kinetic approach taken in this study is effective to design viable low temperature pretreatment processes for effective bioconversion of lignocelluloses.  相似文献

Oil palm empty fruit bunch (OPEFB) was pretreated with 2% (v/v) HNO3 and degraded by Aspergillus niger EFB1 crude cellulase. Through 2 Level Factorial Design (2LFD), it was found that OPEFB concentration, temperature, incubation time, concentration of Tween 80 and agitation speed have significant effect in reducing sugar production. A standard Response Surface Methodology (RSM) design known as Central Composite Design (CCD) was used to optimize the enzymatic degradation condition of OPEFB in rotary drum bioreactor. Reducing sugar level of 1.183 g/L was obtained with the following optimized degradation conditions: 1.95% (w/v) OPEFB, 0.5% (v/v) Tween 80, 55 °C, 87.5 rpm in the incubation period of 3 days and 16 h. The optimal degradation condition improved reducing sugar production by 1.07 fold compared to that before optimization in shake flasks culture. The optimization strategy of enzymatic degradation of OPEFB inside rotary drum bioreactor led to increase in glucose, xylose, arabinose, galactose and mannose production by 3, 2.5, 1.64, 19.37 and 22.52 fold, respectively. The improvement in reducing sugar and polyoses production were comparable with the reduction in OPEFB cellulose and hemicellulose content by 89.32% and 48.17% respectively after enzymatic degradation in optimized condition.  相似文献

This study analyzed the effects of biliprotein C-phycocyanin (C-PC) on the enzymatic antioxidant defence system in lymphocytes of nuclear power-plant workers and non-exposed controls. Changes in the protein levels of manganese super oxide dismutase (MnSOD), catalase and glutathione-S-transferase (GST) were used as markers for early biological effects of a single in vitro exposure of cells to: (i) 2 Gy gamma rays; (ii) 5 μM C-PC; and (iii) a combination of C-PC plus irradiation with 2 Gy. The results showed that C-PC selectively stimulated the lymphocyte antioxidant defence system of occupationally exposed subjects. The activation of the antioxidant protective mechanisms as part of the early radiation response was probably related to the chronic low-dose occupational exposure. The modulating capacity of C-PC at the molecular level may be of interest for the protection of occupationally exposed persons.  相似文献

Biomonitoring by means of the supersensitive cultivar Nicotiana tabacum Bel-W3 is frequently used to obtain information on ozone effects on plants and estimates of ozone exposure. However, most of biomonitoring surveys do not account for other environmental variables (predictors in a statistical model) and their inherent multicollinearity with ozone. We tested the relative role of different predictors (fixed: time and site; random: ozone, temperature and humidity) on height growth and on the development of visible foliar symptoms of N. tabacum Bel-W3 plants. To do this, we investigated a relatively small area (256 km2), used a random design at every stage of the survey, controlled watering and protected plants from direct solar radiation and wind. QA/QC procedures were adopted at every stage of the investigation. Linear correlation shows that Leaf Injury Index (LII) and height increment (H.I.) positively related to ozone concentration, elevation and temperature, and negatively to relative humidity. All the predictors correlate to each other. However, relationships between response and ozone vary with the site and the monitoring week. The effect of the random factor “ozone” in combination with fixed factors “site” and “time” on the response variables was therefore formally investigated using the ANCOVA model. Besides ozone, the interactions “ozone × site” and “ozone × time” resulted always significant (0.001 < P < 0.05). While the factor “time” emphasize the inherent development of injury and growth through time, the interaction “ozone × site” pointed out the importance of local conditions. When watering, solar radiation, wind and plant characteristics were controlled, the remaining site-specific covariates of interest were temperature (T) and humidity (RH). When T and RH were accounted for by means of partial correlation analysis, no significant relationship was found between ozone and LII. On the other side, O3 and RH resulted significant for both absolute and relative height increment. In short, LII seemed to be not solely dependent on ozone, T and RH, but showed to integrate their combined effect. On the other side, H.I. seemed to be favoured by high RH and T, and depressed by high ozone. Based on the above results, we recommend caution when handling bioindicator data: if the purpose is to infer ozone concentrations by leaf injury data, results may be affected by a serious bias, as the frequently reported correlations may be partly an artefact due to co-variation between predictors.  相似文献

Although long-lived trees grow in the Northern Caucasus, no single tree-ring chronology has been reported thus far from this area in the International Tree-Ring Data Base (ITRDB), neither has one been published in international journals. Extensive tree-ring studies were conducted over the last decade, and a tree-ring network was developed for the investigated area. The data on the minimum blue intensity based on 33 series of pine (Pinus sylvestris L.) and fir (Abies nordmanniana (Steven) Spach) is presented in this study. The minimum blue intensity (BI) chronology covers the period 1596–2011 with EPS value ≥0.85. The BI chronology strongly correlates with the mean June-September temperature (R = 0.74; p < 0.05) from the weather station “Kluhorskij Pereval” (1951–2011). Mean June-September temperature anomalies were reconstructed using the rescaling method. Based on the reconstruction provided in this study the twentieth century is characterized by highly increased June-September temperature. According to this study, the minimum blue intensity approach demonstrates a great potential for paleoclimatic research in the Caucasus. Vast spatial coverage of the new BI-based reconstruction based on data from only two locations in the Northern Caucasus provides prospects for reconstruction of temperature variations for a great region in the Middle East and Northern Africa.  相似文献

Temperature and pH play an important role in the stability of phycocyanin, a natural blue colorant. Systematic investigations showed the maximum stability of phycocyanin was in the pH range 5.5–6.0. Incubation at temperatures between 47 and 64 °C caused the concentration (CR) and half-life of phycocyanin in solution to decrease rapidly. The CR value remained at approximately 50% after incubating for 30 min at 59 °C. After heating at 60 °C for 15 min, the CR value of phycocyanin at pH 7.0 was maintained at around 62–70% when 20–40% glucose or sucrose was added, and the half-life increased from 19 min to 30–44 min. 2.5% sodium chloride was found to be an effective preservative for phycocyanin at pH 7.0 as a CR value of 76% was maintained and the half-life of 67 min was increased.  相似文献

The abdominal muscle activity has been shown to be variable in subjects with chronic obstructive pulmonary disease (COPD) when respiratory demand increases and their recruitment pattern may change the mechanics, as well as the work and cost of breathing. The scientific evidence in subjects “at risk” for the development of COPD may be important to understand the natural history of this disease. This study aims to evaluate the effect of inspiratory and expiratory loads on the abdominal muscle activity during breathing in subjects “at risk” for the development of COPD and healthy. Thirty-one volunteers, divided in “At Risk” for COPD (n = 17; 47.71 ± 5.11 years) and Healthy (n = 14; 48.21 ± 6.87 years) groups, breathed at the same rhythm without load and with 10% of the maximal inspiratory or expiratory pressures, in standing. Surface electromyography was performed to assess the activation intensity of rectus abdominis (RA), external oblique and transversus abdominis/internal oblique (TrA/IO) muscles, during inspiration and expiration. During inspiration, in “At Risk” for COPD group, RA muscle activation was higher with loaded expiration (p = 0.016); however, in Healthy group it was observed a higher activation of external oblique and TrA/IO muscles (p < 0.050). During expiration, while in “At Risk” for COPD group, RA muscle activation was higher with loaded inspiration (p = 0.009), in Healthy group TrA/IO muscle showed a higher activation (p = 0.025). Subjects “at risk” for the development of COPD seemed to have a specific recruitment of the superficial layer of ventrolateral abdominal wall for the mechanics of breathing.  相似文献

This article presents a microbiological system composed of a “BT” bioassay (Beta-lactams and Tetracyclines) and a “QS” bioassay (Quinolones and Sulfonamides). The “BT” bioassay contains spores of Geobacillus stearothermophilus, bromocresol purple and cloramphenicol in a culture medium (incubation time: 2.45 h), while the “QS” bioassay uses spores of Bacillus subtilis, trifenyltetrazolium – toluidine blue and trimethoprim in a suitable culture medium (incubation time: 5.5 h). The detection capability (CCβ) of 27 antimicrobial agents in ovine milk were determined by logistic regression models. Thus, the “BT” bioassay detects amoxycillin, ampicillin, penicillin “G”, cloxacillin, oxacillin, cephalexin, cefoperazone, ceftiofur, chlortetracycline, oxytetracycline, tetracycline, neomycin, gentamycin and tylosin, while “QS” bioassay detects: ciprofloxacin, enrofloxacin, marbofloxacin, sulfadiazine, sulfadimethoxine, sulfamerazine, sulfamethazine, sulfamethoxazole, sulfathiazole, erythromycin, lincomycin and spiramycin at levels close to their respective Maximum Residue Limits. The simultaneous use of both bioassays detects a large number of antibiotics in milk given each method's adequate complementary sensitivity.  相似文献

Various supplements (abscisic acid (ABA) or sucrose) were added to the initial embryo culture medium (M3) with the aim of improving the vigour of vitroplants deriving from date palm somatic embryogenesis. ABA (20 and 40 μM) and sucrose (90 g/l) applied for 4 and 2 weeks respectively increased embryo thickness, with no apparent difference in length. ABA (5–40 μM) increased embryo proliferation rate. Somatic embryos maintained in modified M3 (M3 supplemented with ABA and an increased sucrose concentration) contained a higher amount of protein than those maintained in initial M3 (no ABA, 30 g/l of sucrose), with a 1.5–1.7-fold increase depending on the compound and concentration assayed. The 1-D and 2-DE protein profiles showed qualitative and quantitative differences between the somatic embryos cultured in initial M3 (control) and in modified M3. Statistical analysis of spot intensity was performed by principal component analysis, yielding two accurate groups of samples and determining the most discriminating spots. Samples were also clustered using Euclidean distance with an average linkage algorithm. Thirty-four variable spots were identified using mass spectrometry analysis. Identified proteins were classified into the following functional categories: energy metabolism (five proteins); protein translation, folding and degradation (9); redox maintenance (5); cytoskeleton (3); storage protein (2); and with no assigned function as (10). While “up-regulation” of stress-related proteins and “down-regulation” of energy metabolism proteins were observed in somatic embryos matured in M3 supplemented with ABA, storage proteins (legumin) were “up-regulated” in somatic embryos matured in M3 supplemented with increased sucrose.  相似文献

Cheese whey powder (CWP) solution with different CWP or sugar concentrations was fermented to ethanol in a continuous fermenter using pure culture of Kluyveromyces marxianus (DSMZ 7239). Sugar concentration of the feed CWP solution varied between 55 and 200 g l−1 while the hydraulic residence time (HRT) was kept constant at 54 h. Ethanol formation, sugar utilization and biomass formation were investigated as functions of the feed sugar concentration. Percent sugar utilization and biomass concentrations decreased and the effluent sugar concentration increased with increasing feed sugar concentrations especially for the feed sugar contents above 100 g l−1. Ethanol concentration and productivity (DP) increased with increasing feed sugar up to 100 g l−1 and then decreased with further increases in the feed sugar content. The highest ethanol concentration (3.7%, v v−1) and productivity (0.54 gE l−1 h−1) were obtained with the feed sugar content of 100 g l−1 or 125 g l−1. The ethanol yield coefficient (YP/S) was also maximum (0.49 gE gS−1) when the feed sugar was between 100 and 125 g l−1. The growth yield coefficient (YX/S) decreased steadily from 0.123 to 0.063 gX gS−1 when the feed sugar increased from 55 to 200 g l−1 due to adverse effects of high sugar contents on yeast growth. The optimal feed sugar concentration maximizing the ethanol productivity and sugar utilization was between 100 and 125 g l−1 under the specified experimental conditions.  相似文献

A novel aqueous two phase system (ATPS) using trimethylamine-polyethylene glycols (TMA-PEG) to promote the extraction of C-phycocyanin (C-PC) from S.platensis was introduced. The purity of C-PC (EP) obtained in the ATPS of PEG1000/Na3PO4 was increased 2.1 times by the addition of TMA-PEG1000. The purification factor was enhanced from 2.9 to 10.1 when 65% TMA-PEG1000 was added in the system. The ATPS operation must be carried out in the pH range of 6.0-7.0 and at temperatures less than 35 °C for maintaining the stability of C-PC. The partition coefficient and recovery ratio of C-PC increased with the increasing concentration of TMA-PEG. The system parameters like TMA-PEG1000 content, tie line length (TLL), pH, temperature and phase volume ratio (Vr) were screened and optimized using the fractional factorial design and Box-Behnken experiment design. The optimized system is composed of 11.8% PEG1000/TMA-PEG1000 (w/w), 64.42% TMA-PEG1000 (w/w PEG1000) and 9.5% Na3PO4 (w/w) with 38.19% TLL (w/w) and 0.89 Vr at pH 6.5 and 25 °C. The obtained value of EP was 5.21 in one-stage ATPS and 6.7 in two-stage ATPS. The recovery ratio of C-PC in the new ATPS extraction system was more than 97%.  相似文献

The photosynthetic thermophiles have advantage in sequestering CO2 emitted from the energy sector due to their adaptation to high temperatures, growth at high concentrations of CO2, and economically important metabolites. The characterization of such a microorganism, a cyanobacterium from Taian hot springs in Taiwan is described here. This thermophilic cyanobacterium is rod-shaped with a size of 1.2–2.5 μm × 6.0–9.0 μm. A comparison of the 16S RNA and cpcBA-IGS sequences revealed that it is closely related to Thermosynechococcus elongatus BP-1 and so named as Thermosynechococcus elongatus TA-1. This cyanobacterium has better growth at 10% and 20% CO2, at 50 °C with 6000 lx light intensity, at a starting pH of 7–9 and in a medium with 20 mM NaCl. The preferred nitrogen source is NaNO3 of which the minimal requirement is 10 mM. The purified phycocyanin (C-PC) from TA-1 is still kept native and active at a wide range of temperatures (4–60 °C) with a 65.65% activity even at 60 °C, as well as pH values from 4 to 9 and thus exhibiting a good thermal and acid–base stability. This thermophilic cyanobacterium could make integration of CO2 mitigation from industrial flue gas and production of economically important product, like C-PC, more feasible.  相似文献

For the first time, the investigation of Indigo carmine decolorization was done using an atypical Scytalidium thermophilum laccase. Crude and purified laccases required high temperatures and slight acidic pH to achieve maximum Indigo decolorization. Kinetic parameters (Km and kcat) of the homotrimeric laccase toward Indigo carmine were determined and laccase efficacy toward repeated dye decolorization process was studied. For the first time, 5 g l−1 as initial Indigo carmine concentration were efficiently transformed up to 50% within 6 h of incubation using 0.1 U ml−1 of laccase and without presence of any mediators. In this study, we showed that the atypical laccase transformed the indigoid dye structure, confirmed by the color changing from blue to red. This intermediate (red) was a subject to an efficient microbial consortium treatment monitored by measuring the decrease in optical density and the total organic carbon removal efficiencies. Toxicological studies via micro-toxicity test showed that the released enzymatic and adapted consortium degradation products were both non-toxic while the initial product was toxic.  相似文献

Nectar-feeding bats play an important role in natural communities acting as pollinators; however, the characteristics that affect their food selection are unclear. Here we explore the role that sugar gustatory thresholds and sugar concentration play on sugar selection of Glossophaga soricina and Leptonycteris yerbabuenae. We offered bats paired feeders containing sugar solutions of sucrose (S), glucose (G) or fructose (F) vs. pure water, and sucrose vs. 1:1 equicaloric solutions of glucose–fructose at 5, 15 and 35% (wt./vol.). To see the effect of sweetness on sugar selection, we habituated the bats with a diet containing either sucrose or hexoses and subsequently evaluated sugar preferences. Sugar thresholds were S < G,F for G. soricina and G < S < F for L. yerbabuenae. These thresholds did not match with sugar preferences when the bats fed on dilute nectars. L. yerbabuenae changed its sugar preferences with concentration while G. soricina did not. Finally, the bats consistently preferred the sugar they were habituated to. Our results suggest that bats become accustomed to the sugar found in the most abundant plants they use, and thus prefer the most common sugars included in their diet. This could confer an advantage by allowing them shifting sugar preferences on the most common food present in their environment.  相似文献

Many studies have found elevated cortisol linked to negative events (“stress”) and subsequent negative outcomes, such as reduced immunity and stunted growth, leading to the conclusion that high cortisol is “bad.” However, a growing number of studies have found more advantaged groups showing relatively elevated cortisol. For example, higher morning cortisol followed by a steeper diurnal decline among Caucasians compared to ethnic minorities has been interpreted as a context-specific “adaptive boost” to meet daily demands. We tested the adaptive boost hypothesis using data on socioeconomic status, depressive affect and salivary cortisol among adult men (n = 32) in Botswana. Three findings emerged: (i) depressive affect was associated with lower morning cortisol (r = ? 0.43, p = 0.014); (ii) depressive affect was associated with a diurnal increase in cortisol when comparing morning and evening samples (r = 0.49, p = 0.004); and (iii) depressive affect was associated with lower income (r = ? 0.55, p = 0.001). Findings are consistent with the adaptive boost hypothesis and add to a growing body of evidence that elevated cortisol is not universally bad. Hypothalamic–pituitary adrenal axis (HPAA) activity, such as cortisol, may be adaptive depending on a person's contextual circumstances. Based on our findings and those of previous studies, we develop a “person-in-context” model of the threat appraisal process. Integrated with life history theory, our model facilitates testable hypotheses about intra- and inter-individual variability in HPAA and adaptive consequences.  相似文献

We investigated the effects of environmental light colors (blue, yellow and white) on the stress responses (measured by changes in ventilatory frequency – VF) of Nile tilapia to confinement. After 7 days of light treatment, the VF was similar for fish in each color. On the 8th day, fish were confined for 15 min. After release, the post-confinement VF was measured six times (first period: 0, 2 and 4 min; second period: 6, 8 and 10 min). Irrespective of the light color treatment, confinement increased the VF to higher levels during the first post-confinement period than during the second one. When color was analyzed, irrespective of time, fish under white light increased their VF post-confinement, and blue light prevented this effect. We conclude that blue light is the preferred color for Nile tilapia in terms of reducing stress. This finding is in contrast to previous choice test studies that indicated that yellow is their preferred color.  相似文献

AimsProtein O-GlcNAcylation is both a nutrient sensing and cellular stress response that mediates signal transduction in the heart. Chronically elevated O-GlcNAc has been associated with the development of cardiac dysfunction at both the cellular and organ levels in obesity, insulin resistance and diabetes. Development of these pathologies is often attributed to diets high in saturated fat and sugar (a “Western” diet; WES) but a role for O-GlcNAc in diet-induced cardiac dysfunction has not been established. The aims of this study were to examine the effect of chronic consumption of WES on cardiac O-GlcNAcylation and investigate associations of O-GlcNAc with cardiac function and markers of cellular stress.Main methodsYoung male rats received either a control diet (CON; n = 9) or WES (n = 8) diet for 52 weeks.Key findingsThere was no evidence of cardiac dysfunction, advanced glycation endproduct (AGE) accumulation, pathological cardiac hypertrophy, calcium handling impairment, fibrosis or endoplasmic reticulum stress in WES hearts. However, cardiac O-GlcNAc protein, particularly in the higher molecular weight range, was significantly higher in WES hearts compared to CON (P < 0.05). Protein levels of the enzymes that regulate O-GlcNAc attachment were not different between groups; thus, the increased O-GlcNAcylation in WES hearts appears to be due to increased nutrient availability rather than enzymatic regulation of cellular stress.SignificanceThese data suggest that diets high in saturated fat and sugar may contribute to the adverse effects of metabolic syndrome and diabetes by an O-GlcNAc-mediated process and that this may occur in the absence of overt cellular stress.  相似文献

An experimental method for producing ethanol continuously was designed and tested with a cell-recycling two-tank system, which was composed of two fermentors, each of which was individually equipped with a settler for recycling flocculent yeast. This system was effective for the continuous fermentation of ethanol from sucrose at high cell-recycling (r = 0.8–0.9) and dilution (up to 0.48 h?1) rates. The system has several advantages; the high cell concentration in the fermentors and relief of substrate and product inhibition. Thus, the enhanced productivity using this continuous fermentation with the two-tank cell-recycling system was significantly higher compared with that of the batch fermentation. The results indicate that increased recycling ratios caused an increase in biomass concentration and subsequently, product concentration in the tank. The ethanol productivity increased with the dilution rate, but higher dilution rates could render increasing amounts of sugar unconverted. Continuous fermentation with the sugar feed concentration of 160 g/l at r = 0.9 and dilution rate of 0.2 h?1 achieved the highest productivity with less than 2% of the unconverted sugar in the product steam. Under the same cell recycling ratios a productivity range of 6.9–7.5 g/l h?1 could be achieved with feeding concentrations of 80–200 g/l, while batch fermentation at these sugar concentrations led to productivities of 3.85–4.48 g/l h?1.  相似文献

Boric acid and sodium borates have been considered as being “toxic to reproduction and development”, following results of animal studies with high doses. However unfavorable effects of boron exposure on reproduction and development have not been proved in epidemiological studies so far. The aim of the present study was to investigate the reproductive toxicity indicators in highly exposed workers employed in a boric acid production plant in Band?rma, Turkey. Two hundred and four workers participated in this study. The mean blood boron concentration of the high exposure group of workers was 223.89 ± 69.49 (152.82–454.02) ng/g. Unfavorable effects of boron exposure on the reproductive toxicity indicators were not observed.  相似文献
