A lectin (LEL) was isolated from the fresh fruiting bodies of the shiitake mushroom Lentinula edodes by a combination of gel filtration chromatography on Sephadex G-150 and affinity chromatography on an N-acetyl-Dgalactosamine-Sepharose 4B column. Its molecular mass, as determined by gel filtration, was estimated to be 71, 000 Daltons and its structure is homotetrameric with subunit molecular weight of approximately 18,000 Daltons. LEL agglutinated non-specifically red blood cells from the human ABO system as well as rabbit erythrocytes and in haemagglutination inhibition assays, exhibited sugar-binding specificity toward N-acetyl-D-galactosamine. EDTA had no inhibitory effect on its haemagglutinating activity, which was stable up to 70°C and was not affected by changes in pH. The lectin had no covalently-linked carbohydrate and amino acid composition analysis revealed that it contained 124 amino acid residues and was rich in tyrosine, proline, phenylalanine, arginine, glutamic acid and cysteine. LEL did not cause mortality neither was it observed to alter the morphology of key organs when administered intraperitoneally at concentrations up to 10,000 mg kg-1 body weight of mice.  相似文献

Mushrooms have been highly regarded as possessing enormous nutritive and medicinal values. In the present study, we evaluated the anti-oxidative and anti-atherosclerotic potential of shiitake mushroom (Lentinula edodes) using its solvent–solvent partitioned fractions that consisted of methanol:dichloromethane (M:DCM), hexane (HEX), dichloromethane (DCM), ethyl acetate (EA) and aqueous residue (AQ). The hexane fraction (1 mg/mL) mostly scavenged (67.38%, IC50 0.55 mg/mL) the 2,2-diphenyl-1-picryl hydrazyl (DPPH) free radical, contained the highest reducing capacity (60.16 mg gallic acid equivalents/g fraction), and most potently inhibited lipid peroxidation (67.07%), low density lipo-protein oxidation and the activity of 3-hydroxy 3-methyl glutaryl co-enzyme A reductase (HMGR). GC–MS analyses of the hexane fraction identified α-tocopherol (vitamin E), oleic acid, linoleic acid, ergosterol and butyric acid as the bio-functional components present in L. edodes. Our findings suggest that L. edodes possesses anti-atherosclerotic bio-functionality that can be applied as functional food-based therapeutics against cardiovascular diseases.  相似文献

Antimicrobials are used as feed additives to improve growth performance and to prevent subclinical disease challenge in industrial animals. However, these drugs can lead to the development of resistant strains of bacteria. Shiitake mushrooms (SM) (Lentinula edodes) have long been popular as a health food in East Asia. Moreover, SM-derived polysaccharides are well-known as immunostimulants that possess antimicrobial properties. The aim of the present study was to evaluate the immunoprophylactic effects of SM against Bordetella bronchiseptica infection in mice as an initial step towards the development of eco-friendly feed additives to reduce the use of antimicrobials. Although SM had no effect on body weight gain under the un-infected conditions, SM alleviated progressive weight loss and helped in the recovery of body weight in B. bronchiseptica infected mice. Dietary supplementation with SM reinforced bacterial clearance in the infected mice. Of note, SM markedly increased the percentage of various T lymphocytes and the relative mRNA expression levels of tumor necrosis factor-α and interferon-γ in the bronchial lymph node early in the infection. Taken together, these findings suggest that SM could help in the improvement of body weight gain during B. bronchiseptica infection and may enhance the protective immune activity against a subclinical disease challenge, such as B. bronchiseptica infection in mice, probably by a strong stimulation of non-specific immune responses. Hence, SM may provide an alternative to reduce use of antimicrobials. Confirmation of the beneficial effects of SM as a feed additive is now required in industrial animals.  相似文献

The possibility of using hazelnut husk (HH) as a new basal ingredient for substrate preparation in Lentinula edodes cultivation was investigated. Some chemical properties of the substrates prepared by HH alone and its mixtures with wheat straw (WS), beech wood-chip (BWC) and wheat bran (WB) in different ratios were compared, and their effects on spawn run time, days to first harvest (earliness), yield and biological efficiency (BE) were determined. The N content of the substrate prepared from HH alone was very high (0.82%), and thus the C:N ratio of substrates decreased with an increase in the rate of HH in the mixtures. Yield and BE in the HH alone substrate was considerably low compared with the controls (80BWC:10WS:10M and 60BWC:20WS:20WB), and decreased with an increase in the rate of HH in the mixtures. However, when the HH content in the mixtures was kept below 50%, the yield was relatively high (50HH:50WS and 50HH:50BWC). Even when the HH content increased to 75% in the mixture, the comparable yield and BE to the controls could be obtained by adding 10% of WB as nutrients (75HH:15WS:10WB and 75HH:15BWC:10WB). The results revealed that HH could be used as a new basal ingredient for substrate preparation in L. edodes cultivation.  相似文献

A lectin, with a specificity for N-acetylgalactosamine and N-acetylglucosamine and a molecular weight of 43 kDa, was isolated from fruiting bodies of the edible shiitake mushroom Lentinus edodes. The purification procedure entailed extraction with aqueous buffer, ammonium sulfate precipitation, gel filtration on Sephadex G-100 and affinity chromatography on N-acetylgalactosamine-agarose. The lectin was unique in that it was tenaciously bound on anion exchangers including DEAE-cellulose, DEAE-Sepharose, DEAE-Sephadex, Q-Sepharose, Dowex and PEI-cellulose and also on hydroxyapatite and phenyl Sepharose. It was largely unadsorbed on cation exchangers including CM-cellulose, CM-Sepharose, SP-Sepharose and Amberlite, and also on protein G-Sepharose, Red Sepharose and Affi-gel Blue gel, wheat germ lectin-Sepharose and p-aminophenyl-d-glucopyranoside agarose.  相似文献

The white-rot shiitake mushroom, Lentinula edodes, was used to degrade an environmentally hazardous compound, 2,4-dichlorophenol (DCP), using vanillin as an activator. Vanillin increased the mycelial growth from 74 to 118 mg/150 ml culture and accelerated laccase and Mn-peroxidase production from the maximum on days 24–28 without vanillin to days 10–14. It eliminated 92 % of 100 mM DCP with 50 mg vanillin/l compared with only 15 % without vanillin. GC–MS revealed that a diaryl ether dimer of DCP was formed in the culture without vanillin, whereas dimer formation was diminished with vanillin addition. This indicates that vanillin enhances the degradation of DCP and disrupts the formation of the toxic dimer. Therefore, lignin-derived phenol such as vanillin can be used as natural and eco-friendly activators to control white-rot mushrooms, thereby facilitating the effective degradation of environmentally hazardous compounds.  相似文献

Synthetic substrate consisting of oak sawdust (50%), white millet (28%), winter rye (11%) and soft red wheat bran (11%) was non-supplemented or supplemented with 0.2%, 0.4% or 0.6% (dry weight basis) precipitated calcium carbonate (CaCO(3)). Shiitake (Lentinula edodes) was grown in two crops to determine the effect of three CaCO(3) levels on mushroom yield and size. Yields and biological efficiencies (averages for two crops) from substrates non-supplemented with CaCO(3) were lower by 14.1%, 18.4% and 24.9% compared to treatments supplemented with 0.2%, 0.4% and 0.6% CaCO(3), respectively. Mushroom size (weight) was larger with non-supplemented substrate (16.8 g) compared to substrate supplemented with 0.6% CaCO(3) (15.1 g). However, mushroom production was more consistent from crop to crop when 0.6% CaCO(3) was added to substrate.  相似文献

The presence of plasmids was surveyed in 90 wild isolates ofLentinula edodes collected from geographically different world regions. DNA plasmids of different sizes were found in about 80% of the isolates. The plasmids detected were of six kinds, designated as pLE1 (9.0 kb), pLE2 (11.1 kb,=pLLE1 described by other authors), pLE3A (9.8 kb), pLE3B (10.8 kb), pLE3C (12.1 kb), and pLE3D (12.3 kb). Hybridization analysis suggested that pLE1 and pLE2 were distinct plasmid types of different homology groups to each other, and the four other plasmids were variant types belonging to a third homology group. These plasmids had no homology with their host's and non-host's nuclear and mitochondrial genome DNAs. Restriction analysis and electron microscopy indicated that the plasmids are linear in form. Since all six plasmids were transmitted uniparentally in sexual crosses and were consistently associated with the DNA preparations from mitochondria fractionated from mycelia of representative isolates, they were suggested to be located in mitochondria, similar to many other known fungal DNA plasmids. Geographically, pLE1 and pLE2 were widely distributed in natural populations ofL. edodes, while the remaining four plasmids were uniquely present in delimited natural populations. The Japanese wood mushroom, Lentinula edodes, can be cultivated under UK climatic conditions using extensive cultivation systems adapted from traditional growing methods developed in Japan and Taiwan. As well as producing a desirable and high value food product, cultivation offers a viable alternative enterprise for farmers. The growing methods described could provide incentives for the management of broad-leaved woodland in Britain with consequent conservation benefits. Four different strains of Lentinula edodes were grown on oak logs under natural environmental conditions below the woodland canopy and fruited in polyethylene tunnels. Yields ranged from 0.5–0.9 kg wet weight shiitake per cordwood log per year over a three year fruiting period.  相似文献

比较分析香菇球状菌株与正常菌株的形态特征、氨基酸特征和蛋白质品质,并基于现行国际氨基酸模式谱,采用蛋白质的氨基酸评分(amino acid score,AAS)、氨基酸比值系数分(ratio coefficient of amino acid,SRC)、IOM(Institute of Medicine)模式评分、化学评分(chemical score,CS)、必需氨基酸指数(essential amino acid index,EAAI)以及蛋白质校正氨基酸计分(protein digestibility corrected amino acids score,PDCAAS)多种指标进行评价。结果表明:与正常菌株相比,球状菌株形态上没有菌褶和菌柄的分化,同时营养成分也发生了变化,就粗蛋白含量而言,球状菌株为正常菌株的1.37倍(分别为32.32%和23.60%);就平均总氨基酸含量而言,球状菌株(209.58mg/g)是正常菌株(163.10mg/g)的1.28倍。这些球状菌株的特征可作为新品种的培育材料进一步被利用。  相似文献

Two selected Lentinula edodes strains (S4080 and SIEF0231) were cultivated on oak-wood sawdust (OS), wheat straw (WS) and corn-cobs (CC) substrates in order to examine the influence of those residues on mycelium growth and on basidiomata production. For both strains, mycelial growth measurements conducted in race tubes demonstrated faster colonization of OS and WS media. Lag-phase and complete colonization periods were correlated to mycelium extension rates in the three substrates tested. Similar patterns of pH and electrical conductivity (Ec) changes were detected in all media and for all strains tested; the pH decreased steadily throughout the colonization process to reach values of 4.49–5.06; Ec increased by the end of mycelium colonization, and it presented the highest and lowest values in the WS and OS media respectively. In addition, a negative correlation was established between final salt content of the substrates and mycelium extension rates. Subjecting fully colonized substrates to a cold-shock treatment resulted in fruiting 58–65 days after inoculation in tubes; WS and CC promoted earlier sporophore initiation than OS. Monitoring CO2 emissions by strain SIEF0231 in pilot-scale cultivation on synthetic blocks, revealed higher respiration rates from OS and CC than from WS, which were further correlated with substrate colonization rates. Among residues colonized by the same strain, WS appeared to promote earliness and crop productivity (BE 54.17%) by presenting shorter cropping periods and equal yield distribution among flushes, while on OS and CC maximum yields were obtained within the first two flushes. Moreover, heavier basidiomata were produced by WS and OS substrates.  相似文献

香菇多糖是从香菇菌丝体或子实体中提取纯化得到的高分子葡聚糖,作为香菇主要的生物活性物质,具有提高免疫力、抗病毒、抗氧化和抗真菌等生物活性。本研究对幼菇期、菌褶期、采收期、成熟期和开伞期5个不同发育阶段香菇粗多糖含量、理化性质和体外免疫活性进行比较分析。结果表明,其粗多糖含量、理化性质和体外免疫活性具有明显差异。粗多糖得率呈现先增加后降低的趋势,菌褶期得率最高;粗多糖含量也呈现先增加后降低的趋势,成熟期含量最高;多糖的分子量因发育期不同而有较大差异,发育后期所得粗多糖分子量千万级以上组分的比例增加,分子量百万级和十万级的多糖组分比例降低;不同发育阶段香菇子实体多糖的单糖组成均包含果糖、半乳糖、葡萄糖和甘露糖4种单糖,且都以葡萄糖为主;5个阶段的多糖组分均有较好的体外免疫活性,其中幼菇期粗多糖在浓度500μg/mL时表现出最高的体外免疫活性。本研究探讨了香菇不同发育阶段子实体多糖的理化性质及体外免疫活性,为香菇采收期的确定及香菇多糖的制备与利用提供理论依据。  相似文献

Extracts fromshiitake (Lentinula edodes) mycelial culture broth, by an organic solvent ethyl acetate, inhibited the proliferation of cultured cells. At lower concentrations (1.25–15 μg/ml), this inhibition, measured by the MTT assay, was dose- and cell line-dependent. Inhibition of tumor cells, such as Caski, SiHa, HeLa, HP-1 and A375, byL. edodes-436 extracts was stronger than inhibition of normal cells (3T3). At 20 μg/ml, the extracts induced changes in cell shape, DNA-fragmentation and the activation of caspase-3. The extracts also inhibited the binding of E2F protein to its promoter. The results suggest that extracts ofL. edodes culture broth contain substances that have the ability to induce apoptosis in the cultured cells.  相似文献

Vitamin B12 is an essential biomolecule that assists in the catalysis of methyl transfer and radical-based reactions in cellular metabolism. The structure of B12 is characterized by a tetrapyrrolic corrin ring with a central cobalt ion coordinated with an upper ligand, and a lower ligand anchored via a nucleotide loop. Multiple methyl groups decorate B12, and their presence (or absence) have structural and functional consequences. In this minireview, we focus on the methyl groups that distinguish vitamin B12 from other tetrapyrrolic biomolecules and from its own naturally occurring analogues called cobamides. We draw information from recent advances in the field to understand the origins of these methyl groups and the enzymes that incorporate them, and discuss their biological significance.  相似文献

Production costs for shiitake (Lentinula edodes) are on the rise in the United States due to increasing expenses including materials, labor and energy. Increased yield and improved bioconversion of raw materials may improve grower profit margins and may help reduce the cost of shiitake to the consumer. Two crops (Crop 1 and 2) of shiitake were grown to evaluate effects of three substrate moisture contents (50%, 55% and 60%), two log weights (2.7 and 3.2kg) and three porosities of bag filter (low, medium and high) on mushroom yield (g/log) and biological efficiency (BE). Yield data were collected under controlled environmental conditions for two breaks. The formulation with 55% substrate moisture gave the highest yield and BE. Higher mushroom yields were produced from heavier logs (3.2kg), but BE was not significantly affected. Filter porosity significantly affected yield and BE in Crop 1 but not in Crop 2. Significant interactions were observed for log moisture contentxfilter porosity for both crops. There were no significant two-way interactions observed for filter porosityxlog weight or three-way interactions observed for moisture contentxfilter porosityxlog weight. Maximum yields were obtained from 3.2kg logs with a substrate moisture content of ca. 55% using medium or low porosity-filtered bags. This study may provide growers with additional information to better optimize production practices and become more efficient and competitive.  相似文献

Mycelium of shiitake (Lentinula edodes) was cultured continuously in liquid medium. The liquid culture was carried out for the production of liquid spawn in the cultivation of this mushroom on synthetic sawdust substrate, and its performance was compared with that of the solid spawn. The initial colonization in culture bags was faster with the solid spawn than with the liquid spawn, but after this stage CO2 production was higher with the liquid spawn than with the solid spawn. For harvesting sufficient amount of good quality mushrooms, 120 d of incubation in bags was needed with the solid spawn, but this was reduced to 90 d for the sawdust blocks using liquid spawn of less than 50 d old. If continuous culture of the liquid spawn was prolonged over 50 d, immature fruit-bodies or their initials formed during the period of bag incubation. The solid subcultures of the liquid spawns retained the fruiting characteristics acquired in the liquid culture. Liquid culture could be a useful tool for breeding of mushrooms.  相似文献
