To investigate the variability within species of the genus Naegleria, the ITS1,5.8S and ITS2 rDNA were sequenced of several strains of N. lovaniensis and its Western Australian variants, N. australiensis, N. fowleri, N. andersoni, N. jamiesoni, N. tihangensis, N. pringsheimi, N. pagei, N. gruberi sensu lato and a Naegleria lineage that lost a group I intron from the SSUrDNA twintron. As a result, it is possible to define a molecular species within the Naegleria genus. In addition, one strain of each different allozyme cluster was sequenced to investigate whether they belong to described species or should be treated as distinct new species. This leads to the proposal of eleven new species. The sequencing results from those Naegleria spp. of which several strains are available indicate that these species are ubiquitous. The only exception might be the species represented by the WA variants. However, there are still many Naegleria spp. for which only one strain has been isolated, hence, it is important that the search for more isolates should be continued worldwide.  相似文献

The loculoascomycete Moristroma quercinum sp. nov. and Moristroma japonicum sp. nov. are described from Northern Europe (Denmark, Lithuania, Sweden) and Japan, respectively. M. quercinum is reported from wood of Quercus robur and Q. petraea, and M. japonicum is reported from wood of Quercus mongolica var. grossoserrata. Ascostromata of both species were found on hard heartwood of attached or shed branches. The two new species differ from the type species of the genus, M. polysporum, by the presence of pycnidia, and by the size of ascostromata, asci and ascospores. Drawings illustrate ascostromata, pycnidia, asci, hamathecium and ascospores of the two new species. A phylogenetic analysis suggests that Moristroma belongs to the Chaetothyriomycetes, rather than to the Dothideomycetes as previously suggested.  相似文献

Eostrobilopshumicolus Páll-Gergely & Hunyadi, sp. n. is described from Guangxi Province, China. It is characterized by the combination of a small shell (diameter: 2.3–2.4 mm), strongly ribbed dorsal surface, an infraparietal lamella not reaching the callus, and long basal folds. The new species is found approximately 500 and 800 km from the two nearest species Eostrobilopsinfrequens (northern Vietnam), and Eostrobilopsdiodontina (Hunan, China), respectively. A checklist of extant Eostrobilops Pilsbry, 1927 and Enteroplax Gude, 1899 species is provided. Enteroplaxyaeyamensis Habe & Chinen, 1974, Enteroplaxkanjiokuboi Minato & Tada, 1992 and Enteroplaxtaiwanica Minato & Tada, 1992 are moved to the genus Eostrobilops because of the lack of an elevated parietal callus and a peripheral thread. Summary. Internal transcribed spacer 1 (ITS-1) sequences of the nuclear rDNA of eight bee species of the genus Melipona were studied. Complete ITS-1 sequence and flanking regions from three Melipona species were PCR-amplified, cloned, sequenced, and their variability compared. These sequences show length variation (1391 to 1417 bp), several repeated elements of one, two, three, and four nucleotides, and a repeated tandem sequence of approximately 80 bp. The low variation level between M. quadrifasciata and M. mandacaia sequences supports the hypothesis that they diverged recently. PCR-amplification, cloning, and sequencing of a partial ITS-1 sequence (394 to 496 bp) of eight Melipona species and two outgroups were performed and the obtained sequences used for phylogenetic analysis. The single tree estimated from parsimony analysis recovered four well-defined clades and monophyly of the genus Melipona. The phylogenetic relationships derived from sequences of ITS-1 fragments corroborate the taxonomic classification of Melipona based on morphological characters.Received 17 July 2003; revised 10 May 2004; accepted 1 June 2004.  相似文献

Complete nuclear ribosomal DNA (nrDNA) internal transcribed spacer (ITS) sequences were obtained for 18 Chondrus populations collected at 15 sites from eight countries worldwide. Pairwise comparisons with the multiple alignment revealed that intraspecific divergences of ITS sequences ranged from 0.3 to 1.8% in C. crispus Stackhouse (except for the entity SVLH from France) and from 0.0 to 0.6% in C. ocellatus Holmes, whereas interspecific divergences in Chondrus varied from 1.4 to 5.0%. Three phylogenetic methods (neighbour joining, maximum parsimony and maximum likelihood) confirmed three main lineages: the North Atlantic lineage containing entities of C. crispus from Canada, France, Germany, England, Portugal, Ireland and Wales; a second lineage comprising three species: C. sp. 1, C. armatus (Harvey) Yamada et Mikami, and C. pinnulatus (Harvey) Okamura from the Northern Pacific; and a third lineage containing just one species: C. ocellatus from the Northern Pacific. Chondrus yendoi Yamada et Mikami separated from other Chondrus species singly. nrDNA ITS data indicate that a previous assignment of C. sp. 2 to Mazzaella japonica (Mikami) Hommersand may be incorrect, and additional evidence is needed to resolve the generic placement of this entity. It is inferred from the nrDNA ITS data that three Chondrus species are presently known in China with two, C. ocellatus and C. nipponicus, in Qingdao and two, C. armatus and C. nipponicus, in Dalian. We hypothesize that the ancestor of North Atlantic C. crispus had a Pacific origin, and that the present distribution of C. crispus in the Atlantic Ocean correlates with a trans-Arctic dispersal and vicariance events associated with Pleistocene glaciation maxima.  相似文献

Thirteen new microsatellite loci were isolated and tested on two land snail species, Trochulus villosus and T. sericeus (Pulmonata: Hygromiidae), resulting in a set of eight polymorphic markers for each species. The expected heterozygosity was high for all loci and species (between 0.616 and 0.944). Such levels of variability will allow detailed insights into the population genetic structure of some Trochulus species.  相似文献

The genus Rhagada is the second most diverse camaenid genus in Australia. We examined anatomical and mitochondrial characters of previously unidentified material from the Kimberley that was earmarked to potentially represent new species in recently published molecular phylogenetic studies. Our comparisons revealed that specimens from Gibbings Island (‘R. sp. Gibbings’) were morphologically and genetically most similar to Rhagada cygna from the Dampier Peninsula. Hence, ‘R. sp. Gibbings’ is considered to be identical to R. cygna. In addition, we found that R. cygna as so delimited is not clearly distinguished from the second species on the Dampier Peninsula, Rhagada bulgana. Both species differ rather subtly in anatomical and mitochondrial characters, indicating their close relationships and potentially incomplete evolutionary differentiation. Furthermore, we describe two new species based on comparative morphology and mitochondrial sequences: Rhagada worora n. sp. from the Prince Regent Reserve in the Kimberley and Rhagada karajarri n. sp. from Dampierland. The present study confirms that species in Rhagada are best identified by means of both morphological and molecular data.

The phylogenetic origin of Beckmannia remains unknown. The genus has been placed within the Chlorideae, Aveneae (Agrostideae), Poeae, or treated as an isolate lineage, Beckmanniinae. In the present study, we used nuclear internal transcribed spacer (ITS) and chloroplast trnL-F sequences to examine the phylogenetic relationship between Beckmannia and those genera that have assumed to be related. On the basis of the results of our studies, the following conclusions could be drawn: (i) Beckmannia and Alopecurus are sister groups with high support; and (ii) Beckmannia and Alopecurus are nested in the Poeae clade with high support. The results of our analysis suggest that Beckmannia should be placed in Poeae.  相似文献

The phylogeny of Ptychostomum was first spacer (ITS) region of the nuclear ribosomal (nr) DNA DNA rps4 sequences. Maximum parsimony, maximum undertaken based on analysis of the internal transcribed and by combining data from nrDNA ITS and chloroplast likelihood, and Bayesian analyses all support the conclusion that the reinstated genus Ptychostomum is not monophyletic. Ptychostomum funkii (Schwagr.) J. R. Spence (≡ Bryum funkii Schwaigr.) is placed within a clade containing the type species of Bryum, B. argenteum Hedw. The remaining members of Ptychostomum investigated in the present study constitute another well-supported clade. The results are congruent with previous molecular analyses. On the basis of phylogenetic evidence, we agree with transferring B. amblyodon Mull. Hal. (≡ B. inclinatum (Brid.) Turton≡ Bryum archangelicum Bruch & Schimp.), Bryum lonchocaulon Mull. Hal., Bryum pallescens Schleich. ex Schwaigr., and Bryum pallens Sw. to Ptychostomum.  相似文献

Parapatric, morphologically characterized forms of the Mediterranean snail genus Albinaria have traditionally been regarded as biological species. Recently, this view has been challenged on the basis of small interspecific genetic distances and cross-breeding experiments. In order to test whether traditionally recognized species are reproductively isolated lineages, two cases of syntopy were analysed by means of isozyme electrophoresis. The data suggest that complete reproductive isolation is present. In one of the two cases, ecological differentiation on a small spatial scale was observed.  相似文献

Variation within the internal transcribed spacer (ITS) of the ribosomal RNA gene of 15 isolates representing seven European Armillaria species, was examined by sequencing of the PCR-amplified products. The analysis of an 744-bp region showed that the 5.8S gene appeared to be highly conserved in the 15 isolates and in other Basidiomycetes and Ascomycetes, whereas ITS1 and especially ITS2 spacers exhibited polymorphisms due to base substitutions, insertions or deletions of up to eight nucleotides. An initial dendrogram for the full sequence was drawn using cluster analysis (UPGMA), and a tree was constructed using the maximum parsimony method. Both methods indicated that the isolates could be divided into four major groups. One group, consisting of A. ectypa , was distinct from all the other species. Examination of the other groups indicated that A. tabescens and A. mellea were in a separated cluster, with a significant variation between the two isolates of the latter species. A. gallica and A. cepistipes constituted another closely related group distinguishable from A. ostoyae and A. borealis , these latter two species exhibiting the highest similarity. The results are consistent with, and discussed in regard to, the relationships estimated previously by pairing tests, morphological and physiological comparisons, as well as by restriction fragment length polymorphism of the rDNA.  相似文献

The radiation of the endemic Jamaican land snail genus Sagda is examined through analysis of the comparative shell morphology, anatomy, ecology and biogeography of the four species inhabiting north-central Jamaica: S. centralis sp. nov. , S. spei, S. bondi and S. montegoensis. The species are all rather similar in their morphology and ecology. The shells show some differences in the presence or absence of internal lamellae, the position of the lamellae, the form of the depression in the centre of the base, the regularity of sculpture (low ribs) and the pattern of microsculpture. Slight differences in the form of the radular teeth are found between some species. The structure of the secondary ureter is peculiar: in all species it is open shortly behind the mantle collar, then branches into two closed tubes which open separately on the outside of the mantle collar. Despite its great complexity, the reproductive system is rather uniform among species; differences are seen in the number of pockets that form spines on the spermatophore. The complex, three-branched spermatophore is formed within the entire epiphallar branch of the penis, whereas the appendicular branch apparently assumes the copulatory function. When inactive, all species are found associated with rocks or, less commonly, among leaf litter. Activity occurs primarily at night and mostly on leaf litter rather than rocks. The snails eat dead plant material, including a variety of plant species. All species (with the possible exception of S. montegoensis) are ovoviviparous. They overlap broadly in the range of climates occupied. The geographic ranges of the four species are overlapping, although Sagda bondi has a parapatric distribution in relation to the western species S. grandis. Fossil and subfossil evidence shows that shifts in the ranges of the species have occurred in the late Quaternary; Pleistocene-Holocene environmental changes, habitat destruction by man, and possibly other environmental changes during historical times are probably involved. Radiations of other land snail groups on tropical islands have generally produced greater morphological and ecological differentiation than in Sagda. Allopatric or parapatric species distributions appear to be more common in other land snail radiations.  相似文献

Albinaria (Gastropoda, Clausiliidae) is a pulmonate genus distributed around the north-eastern coasts of the Mediterranean. It is the most 'speciose' genus within the family of Clausiliidae, exhibiting a high degree of morphological and genetic differentiation, and serving as a model for several ecological, systematics and evolutionary studies. Nevertheless, many aspects remain uncertain mainly due to the large number of taxa whose classification has not yet been evaluated with solid synapomorphic characters. Thirty-one morphological species are currently recognized on the island of Crete and its satellite islets. Four of them (A. retusa, A. torticollis, A. jaeckeli, and A. teres) are distributed on the island of Dia (north of Crete); the first three are island endemics. Here, we combined mitochondrial and nuclear DNA information and Bayesian and Maximum Likelihood approaches to evaluate the phylogenetic relationships, and assess the genetic distinctiveness and cohesiveness of all described species of Dia Island. The produced phylogeny was not congruent with the morphological species, demonstrating a more complex pattern of speciation and diversification. Although each island endemic constitutes a monophyletic lineage, the number of island endemic species could be greater than the three currently recognized species so far (A. retusa, A. torticollis, and A. jaeckeli), since a newly discovered lineage (north-western part of the island), that morphologically differs from the populations of A. torticollis in the eastern part, and genetically is more closely related to A. jaeckeli, could be elevated to the species level. Considering the fourth species found on the island of Dia, A. teres is genetically highly variable, showing low geographic structuring due to either long-distance gene flow or retained ancestral polymorphisms, or a combination of both. Further work (analysis of more specimens and DNA data) both from Dia and Crete is indispensable in order to shed light on aspects of the evolutionary history of the genus in Dia.  相似文献

Two species of the genus Boysidia Ancey, 1881 were reported as new to science, Boysidia(Boysidia) xianfengensis sp. nov. and B.(B.) xiaoguanensis sp. nov., while the distribution of the 23 species of Boysidia from China was discussed. The materials of the new species were collected from Hubei Province, China by authors. The shell of B.(B.) xianfengensis sp. nov. looks small(height 4.90 mm, diameter 3.10 mm), and has five apertural teeth: one large parietal tooth, one long columellar tooth, one small nipple-shaped upper palatal tooth, one large flake-shaped lower palatal tooth, one small infrapalatal tooth. The shell of B.(B.) xiaoguanensis sp. nov. looks small(height 3.55 mm, diameter 2.20 mm), and has four apertural teeth: one large parietal tooth, one long columellar tooth, one large flake-shaped upper palatal tooth, one slightly small flake-shaped lower palatal tooth. The type specimens of the new species are deposited in the Institute of Zoology, Chinese Academy of Sciences, China.  相似文献

Individuals of five nominal species of Grania (Annelida: Clitellata: Enchytraeidae) were collected from locations in Sweden, Norway and France, for studies on the intraspecific variation at the Cytochrome Oxidase I (COI) locus of mitochondrial DNA and internal transcribed spacer (ITS) region of nuclear DNA. It was found that the previously described morphospecies in general contain low variation compared to the between‐species variation in both loci. In one instance, however, an individual morphologically indistinguishable from G. ovitheca was found to be deviant and instead cluster with G. postclitellochaeta both by COI and ITS. We describe this individual as a new species: G. occulta sp.n. Furthermore, phylogenetic analyses were conducted, showing a close relationship between G. variochaeta, G. occulta, G. ovitheca and G. postclitellochaeta, as well as between G. pusilla and G. maricola. Using the results from the phylogenetic analyses, we discuss the evolution of morphological characters in Scandinavian species of Grania.  相似文献

The nucleotide sequences of the internal transcribed spacer (ITS) regions of the ribosomal DNA including the 5.8S rRNA gene and the 5′ end of the 28S rRNA gene have been determined for 19 species in 10 genera of the powdery mildew fungi in order to analyze their phylogenetic relationship. These fungi were divided into two large groups based on the nucleotide length of the ITS regions, and this grouping was in line with that based on the morphological characters of the anamorphic stage rather than the teleomorphic stage. Although the variable ITS sequences were often ambiguously aligned, conserved sites were also found. Thus, a neighbor-joining tree was constructed using the nucleotide sequence data of the conserved sites of the ITS regions, the 5.8S rRNA gene, and the 5′ end of the 28S rRNA gene. The phylogenetic tree displayed the presence of four groups in the powdery mildews, which were distinguished by their morphology and/or host ranges. In the ITS2 region, the presence of a common secondary structure having four hairpin domains was suggested, in spite of the highly variable nucleotide sequences of this region. The predicted secondary structure was supported by the compensatory mutations as well as compensatory conserved sequences and high G+C content in the predicted stem regions. The following eleven Empria species are reported from Japan: Empria candidata (Fallén, 1808), Empria japonica Heidemaa & Prous, 2011, Empria liturata (Gmelin, 1790), Empria loktini Ermolenko, 1971, Empria plana (Jakowlew, 1891), Empria quadrimaculata Takeuchi, 1952, Empria rubicola Ermolenko, 1971, Empria tridens (Konow, 1896), Empria tridentis Lee & Ryu, 1996, Empria honshuana Prous & Heidemaa, sp. n., and Empria takeuchii Prous & Heidemaa, sp. n. The lectotypes of Poecilosoma pallipes Matsumura, 1912, Empria itelmena Malaise, 1931, Tenthredo candidata Fallén, 1808, and Tenthredo (Poecilostoma) hybrida Erichson, 1851 are designated. Empria itelmena Malaise, 1931, syn. n. is synonymized with Empria plana (Jakowlew, 1891). Poecilosoma pallipes Matsumura, 1912, previously assigned to Empria, is transferred to Monsoma, creating Monsoma pallipes (Matsumura, 1912), comb. n. Results of phylogenetic analyses using mitochondrial (COI) and nuclear (ITS1 and ITS2) sequences are also provided.  相似文献

Taxonomy within the sponge Order Haplosclerida remains somewhat contentious. Both morphology and biochemistry have been employed to help unravel the relationships of the Order with limited results perhaps because of the small number of reliable characters. We employed phylogenetic analysis of 750 base pairs of the 5′ end of the 28S rRNA gene to study relationships between 20 taxa within the Order. While preliminary (low number of taxa, one gene region) results suggested strong discrepancy to former phylogenies, there was no support for the monophyly of the Order and almost all morphologically defined genera appeared to be polyphyletic.  相似文献

Based on the sequence analysis of 5.8S subunit and internal transcribed spacers (ITS ) of ribosomal RNA gene (rDNA), the molecular phylogenetic tree of representative species of Pipizini and three groups of Syrphidae with different feeding habits (seven species belong to six genera) was constructed. Meanwhile, the phylogenetic tree of tribes (including Pipizini and other 17 tribes of Syrphidae) was constructed using morphological characteristics of adults and larvae and the number of chromosomes. Both the results show that the relationship between Pipizini and predatory groups is closer than that between Pipizini and saprophagous groups. So it is suggested that Pipizini be transferred from Milesiinae to Syrphinae.  相似文献
