The genetic population structure inBdallophyton bambusarum, an endoparasite, was studied in ten subpopulations from a subdeciduous tropical forest in Veracruz Mexico. The sample was analyzed using seven polymorphic loci in cellulose acetate electrophoresis. Isozyme data indicated that the subpopulations ofB. bambusarum contained high genetic variability (Hep = 0.452 ± 0.045, S.E.). Our analysis suggests that almost each inflorescence ofB. bambusarum is an individual. The subpopulations studied were genetically similar (average Nei's genetic identity 0.941 ± 0.051 and F st values 0.097 ± 0.026), suggesting that genetic differentiation among subpopulations was small. Direct estimates of effective population size was derived from observations of three fluorescent dyes, and from the genetic neighborhood area derived from these data. The neighborhood area, multiplied by the total density of individuals, gave an Ne = 124.84 plants, and when corrected to consider the proportion of males and females gave an Ne = 118.59 individuals. An indirect estimate of Nm was obtained from the F st values (mean Nm=2.037), giving an indirect estimate of the effective population size Nb = 12.8 individuals. Both values are relatively high when compared to other plant studies. The gene flow and/or effective populations size of the studied subpopulations ofB. bambusarum are believed to be large enough to prevent differentiation among subpopulations due to genetic drift.  相似文献

In breeding programs, robustness of animals and uniformity of end product can be improved by exploiting genetic variation in residual variance. Residual variance can be defined as environmental variance after accounting for all identifiable effects. The aims of this study were to estimate genetic variance in residual variance of body weight, and to estimate genetic correlations between body weight itself and its residual variance and between female and male residual variance for broilers. The data sets comprised 26 972 female and 24 407 male body weight records. Variance components were estimated with ASREML. Estimates of the heritability of residual variance were in the range 0.029 (s.e. = 0.003) to 0.047 (s.e. = 0.004). The genetic coefficients of variation were high, between 0.35 and 0.57. Heritabilities were higher in females than in males. Accounting for heterogeneous residual variance increased the heritabilities for body weight as well. Genetic correlations between body weight and its residual variance were -0.41 (s.e. = 0.032) and -0.45 (s.e. = 0.040), respectively, in females and males. The genetic correlation between female and male residual variance was 0.11 (s.e. = 0.089), indicating that female and male residual variance are different traits. Results indicate good opportunities to simultaneously increase the mean and improve uniformity of body weight of broilers by selection.  相似文献

Drosophila engyochracea, an endemic Hawaiian fly found only in two, finite populations in Volcano National Park, has extensive electrophoretic heterozygosity on a par with that found in species with much wider distributions. A study of six polymorphic loci in both populations over an 18-month period revealed that the population in the more xeric environment is more dynamic genetically as well as more variable. In addition, genetic changes at one locus, Pgm, are correlated to changes in an environmental moisture parameter. These findings confirm that migration is not necessary to maintain genetic variation in isolated populations and demonstrate that D. engyochracea gene pools are susceptible to errors in Hardy-Weinberg equilibria during specific seasonal periods.Portions of this article were submitted in partial fulfillment for a Doctor of Philosophy degree in Genetics awarded to W. Many ecologically important traits exhibit latitudinal variation. Body size clines have been described repeatedly in insects across multiple continents, suggesting that similar selective forces are shaping these geographical gradients. It is unknown whether these parallel clinal patterns are controlled by the same or different genetic mechanism(s). We present here, quantitative trait loci (QTL) analysis of wing size variation in Drosophila simulans. Our results show that much of the wing size variation is controlled by a QTL on Chr 3L with relatively minor contribution from other chromosome arms. Comparative analysis of the genomic positions of the QTL indicates that the major QTL on Chr 3 are distinct in D. simulans and D. melanogaster, whereas the QTL on Chr 2R might overlap between species. Our results suggest that parallel evolution of wing size clines could be driven by non-identical genetic mechanisms but in both cases involve a major QTL as well as smaller effects of other genomic regions.  相似文献

The flight ability ofDrosophila aldrichi (Patterson & Crow) andD. buzzatii (Patterson & Wheeler) using tethered flights, was measured with respect to age-related changes, genetic variation and adult body size variation induced by rearing at different larval densities.Drosophila buzzatii flew for much longer thanD. aldrichi, especially females, but age-related changes in flight duration were significant only forD. aldrichi. Effects of body size on flight ability were significant inD. buzzatii, but not inD. aldrichi. InD. buzzatii, there was a significant genotype-environment interaction (larval density × line) for flight duration, with short and average flight duration isofemale lines showing longer flights, but a long flight duration line shorter flights as body size decreased (i.e., as larval density increased). Heritability estimates for flight duration were similar in the two species, but flight duration showed no significant genetic correlations with developmental time, body size or wing dimensions (except for one wing dimension inD. buzzatii). Although not significantly different between the species, heritabilities for life-history traits (adult size and developmental time) showed contrasting patterns — with higher heritability for body size (body weight and thorax length) inD. buzzatii, and higher for developmental time inD. aldrichi. In agreement with limited previous field evidence,D. buzzatii is better adapted for colonization than isD. aldrichi.  相似文献

Patterns of restriction fragment length polymorphisms (RFLPs) have been proposed as estimators of genetic diversity among breeding lines and as predictors of heterosis and genetic variance. We evaluated these proposals by using a set of nine elite oat lines crossed in a diallel mating design without reciprocals. RFLP analysis was conducted using HindIII-digested DNA and a total of 107 probes from three different sources: 14 heterologous wheat cDNA clones, 17 oat genomic clones, and 76 oat cDNA clones. Of the 77 probes that produced high-quality autoradiographs, 26 detected polymorphisms among this set of lines, with an average of 2.6 variants per probe. RFLP-based genetic distance (FD) was calculated from these data by using Nei and Li's measure of genetic similarity, and was compared with two other measures of genetic divergence. Genealogical distance (GD *) was obtained from the coefficients of parentage based on known parental pedigrees, and multivariate distance (DI) was calculated by using the first five principal components of the parental correlation matrix for 12 agronomic traits. FD was significantly correlated with GD * (r=0.63, P<0.01), but not with DI (r=-0.05). Cluster analysis based on these three distance estimates did not produce equivalent groupings, but the FD and GD * clusters were more similar to each other than to the DI clusters. These results indicate that: (1) sufficient variation exists for further application of RFLP technologyto oats, (2) RFLPs could provide accurate estimates of genetic divergence among elite oat lines, and (3) it is unlikely that dispersed markers can predict heterosis or population genetic variance in oats. Three dipeptidases in Drosophila melanogaster are under independent genetic control and their structural genes have been localized, Dip-A to 2R and Dip-B and Dip-C to 3R (Voelker and Langley, 1978; Ohnishi and Voelker, 1981). These enzymes were characterized with respect to their substrate specificities, genetic variability (electrophoretic mobility and quantitative activity level), ontogeny (activity and isozyme pattern), and tissue localization. The dipeptide substrate specificities of DIP-A and DIP-B overlap each other considerably, but do not overlap with DIP-C. In natural populations, DIP-B and DIP-C are essentially monomorphic electrophoretically whereas DIP-A is polymorphic for three allozymes. Both DIP-A and DIP-B show quantitative genetic variation of activity level within an allozyme class. All three enzymes are expressed at all stages in the life cycle, but DIP-A and DIP-B activities vary considerably according to developmental stage and sex of adult. The tissue localizations of DIP-A and DIP-B activities show similar patterns and a nearly ubiquitous occurrence of both enzymes, but with particularly high values in larval and adult midguts and in the adult female reproductive system. We examined the geographical pattern in growth and adult body size among 14 populations of Swedish moose (Alces alces) using data from 4,294 moose (1.5 years old) killed during the hunting season in 1989–1992. In both sexes, adult body mass was significantly positively correlated with latitude. Moose in northern populations had a 15–20% larger adult body mass than moose in the south. Juvenile body mass was correlated with neither latitude nor adult body mass. Thus, variation in time (years) and rate of body growth after the juvenile stage were responsible for most of the variation in adult body mass among populations. Moose in northern populations grew for approximately 2 more years of life than southern moose. In contrast to adult body mass, skeletal size (measured as jawbone length) was not correlated with latitude, suggesting that variation in adult body mass was primarily due to differences in fat reserves. Discrimination between population characteristics, such as moose density, climate, and the amount of browse available to moose, showed climatic harshness to be the most important variable explaining geographical variation in body mass among populations. The results support the notion that in mammals body size increases with latitude in accordance with Bergmann's rule. We conclude that (1) variation in patterns of growth after the juvenile stage is the main cause of the latitudinal trend in adult body size in moose, and (2) climatic conditions are a more important factor than population density and availability of food in explaining geographical variation in growth patterns and adult body mass between populations of Swedish moose.  相似文献

Genetic monitoring of reintroduced plantpopulations can allow assessment of the successin establishing new populations thatgenetically resemble native populations. Weused a PCR-based method (Intersimple SequenceRepeats) to quantify genetic variation in fourreintroduced populations of Abroniaumbellata ssp. breviflora, an annualforb native to the Pacific Coast that isstate-listed endangered in Oregon. Thereintroduced populations ranged in size from 18to 4,111 individuals in the year they weresampled. Genetic variation was also quantifiedin the natural population that served as theseed source for the reintroduction efforts. Atotal of 77 loci (bands) was observed using twoISSR primers, providing 65 polymorphic loci. Asignificant, positive regression was observedbetween the log of population size for the fivepopulations and genetic variation when measuredas percent polymorphic loci (P), expectedheterozygosity (He> ), and with adissimilarity index (1 – Sxy) based on bandsharing. Two of the reintroduced populationsmaintained approximately 90% of the geneticvariation we observed in the source population. Based on these results, we predict thatreintroduced populations of A. u. ssp.breviflora that have at least 1,000individuals should maintain 90% of the geneticvariation of the source population.  相似文献

Reaction norms across seven constant and one fluctuating temperature of development were measured for thorax length and several wing size traits for up to 10 isofemale lines of each of the cactophilic Drosophila species, D. aldrichi and D. buzzatii, originating from the same locality. Maximum thorax length was reached at different low to intermediate temperatures for the two species, whereas wing length was highest at the lowest temperature in both species. Various ratio parameters showed pronounced species differences. The reaction norm for the wing loading index (wing length/thorax length) decreased monotonically with temperature in both species, but was much steeper and spanned a wider range in D. aldrichi than in D. buzzatii, suggesting either that wing loading is not a good characterization of flight capacity or, more likely, that flight optimization does not occur in the same manner in both species. The vein ratio (distal length/proximal length of the third vein) increased with temperature in D. buzzatii but decreased in D. aldrichi. Wing development in the two species thus is very different, with the proximal part of the wing in D. buzzatii more closely allied to the thorax than to the distal part. Among line variation was significant for all traits in both species, and most pronounced for thorax length and the ratio parameters. Coefficients of variation were significantly different between the species for all traits, with those in D. aldrichi higher than in D. buzzatii. Genetic variance in plasticity was significant for all traits in D. buzzatii, but only for seven out of 12 in D. aldrichi. Additive genetic variances for all traits in both species were significantly larger than zero. Genetic correlations between thorax length and several wing length parameters, and between these and wing area, were positive and generally significant in both species. The genetic correlation between the distal and the proximal length of the third vein was not significantly different from zero in D. aldrichi, but negative and significant in D. buzzatii. Heritabilites varied significantly among temperatures for almost all traits in both species. Phenotypic variances were generally higher in D. aldrichi than in D. buzzatii, and commonly highest at the extreme temperatures in the former species. At the high temperature the genetic variances also were usually highest in D. aldrichi. The data clearly suggest that the process of thermal adaptation is species specific and caution against generalizations based on the study of single species.  相似文献

Restricted maximum likelihood was used to estimate genetic parameters of male and female wing and thorax length in isofemale lines ofDrosophila melanogaster, and results compared to estimates obtained earlier with the classical analysis of variance approach. As parents within an isofemale line were unknown, a total of 500 parental pedigrees were simulated and mean estimates computed. Full and half sibs were distinguished, in contrast to usual isofemale studies in which animals were all treated as half sibs and hence heritability was overestimated. Heritability was thus estimated at 0.33, 0.38, 0.30 and 0.33 for male and female wing length and male and female thorax length, respectively, whereas corresponding estimates obtained using analysis of variance were 0.46, 0.54, 0.35 and 0.38. Genetic correlations between male and female traits were 0.85 and 0.62 for wing and thorax length, respectively. Sexual dimorphism and the ratio of female to male traits were moderately heritable (0.30 and 0.23 for wing length, 0.38 and 0.23 for thorax length). Both were moderately and positively correlated with female traits, and weakly and negatively correlated with male traits. The genetic differentiation ofOrchis papilionacea from Italy, Corsica, Sardinia, Sicily, and Greece was analyzed by isozyme studies. No significant differences at any of the 28 loci tested were observed between samples of the putative subspeciespapilionacea from Italy,grandiflora from Sicily andheroica from Greece, or by comparing sympatricpapilionacea-like andgrandiflora-like subsamples from Sardinia and Corsica. The genetic relationships found among the populations studied reflect more their geographic origin than their taxonomic assignment;papilionacea, grandiflora, andheroica are, therefore, at least in the study area, not to be considered as subspecies, but as simple morphs, controlled by few genes (possibly two) with Mendelian inheritance. The genetic divergence observed among the samples studied is very low and consistent with that of geographic populations of a single cohesive gene pool, linked by a high gene flow (Nm = 5.9).  相似文献

Electrophoretic variation within and between North Atlantic minke whale samples(Balaenoptera acutorostrata) from West Greenland, Iceland, and Norway was investigated. In the West Greenland samples, 28 enzyme systems were examined, representing 36 loci, of which 6 were found to be polymorphic. In Icelandic and Norwegian samples, 22 enzyme systems were examined, representing 29 loci, of which 6 and 5 were found to be polymorphic, respectively. The average heterozygosity was 0.058 (SE=0.024) in samples from West Greenland, 0.074 (SE=0.028) in samples from Iceland, and 0.054 (SE=0.023) in samples from Norway. No significant deviations from the expected Hardy—Weinberg genotypic frequencies, within samples taken from the same area, were found. Significant differences in allele frequencies were observed, however, between samples from the three different areas. The average Nei's genetic distance was 0.014 and the averageF st value was 0.126. The genetic differences between the samples from the different areas indicate that those from West Greenland, Iceland, and Norway represented different breeding populations.  相似文献

Allozyme polymorphisms at seven loci have been studied in nine natural populations of Drosophila melanogaster from the Saône and Rhône valleys sampled in 1973 and 1974. A great deal of polymorphism was observed; an individual was on the average heterozygous at 20.2% of its loci. The populations were genetically very homogeneous throughout the region sampled. The number of ovariolae per female varied from one group of populations to another depending on their geographical separation. Yet the number of ovariolae remained constant from one year to the next. The results show that migration alone cannot explain the homogeneity of the allozyme frequencies. It seems reasonable to conclude that selection plays a major role in maintaining the homogeneity of populations living in proximal biotopes.E.R.A. This work investigated the effects of increasing temperature from 30°C to 47°C on the physiological and genetic characteristics of Saccharomyces cerevisiae strain 63M after continuous fermentation with cell recycling in a system of five reactors in series. Steady state was attained at 30°C, and then the temperature of the system was raised so it ranged from 35°C in the last reactor to 43°C in the first reactor or feeding reactor with a 2°C difference between reactors. After 15 days at steady state, the temperature was raised from 37°C to 45°C for 25 days at steady state, then from 39°C to 47°C for 20 days at steady state. Starter strain 63M was a hybrid strain constructed to have a MAT a/α, LYS/lys, URA/ura genotype. This hybrid yeast showed vigorous growth on plates at 40°C, weak growth at 41°C, positive assimilation of melibiose, positive fermentation of galactose, raffinose and sucrose. Of 156 isolates obtained from this system at the end of the fermentation process, only 17.3% showed the same characteristics as starter strain 63M. Alterations in mating type reaction and in utilization of raffinose, melibiose, and sucrose were identified. Only 1.9% of the isolates lost the ability to grow at 40°C. Isolates showing requirements for lysine and uracil were also obtained. In addition, cell survival was observed at 39–47°C, but no isolates showing growth above 41°C were obtained.  相似文献

Drosophila melanogaster larvae were subjected to 10 generations of selection on 6% ethanol at 17, 25, and 30°C. For each temperature there was a significant (P<0.01) increase in the frequency of the Adh isoallele. Controls with no ethanol showed no change in the frequency of the Adh F isoallele. Larvae subjected to stronger selection on 8% ethanol confirmed the results. When adults of various ages were subjected to 16 and 32°C, the ADHF isoenzyme retained its twofold advantage in activity over ADHS regardless of the temperature. The same result was obtained with larvae at 16 and 35°C. Although some effect of temperature was demonstrated, it was concluded that the effect was not strong enough for temperature to be a selective factor under the conditions studied. However, ethanol is a strong selective factor for laboratory populations.  相似文献

Wing and body lengths of polyphagous parasitoids of leafminers are analyzed in order to understand the relationship between host and parasitoid sizes. A distinct positive relationship was observed: within each parasitoid species, smaller individuals were reared from smaller hosts. There was no difference between idio-and koinobionts in sexual dimorphism, with females being significantly larger than males in both groups.  相似文献

Genetic variation of body size along latitudinal clines is found globally in Drosophila melanogaster, with larger individuals encountered at higher latitudes. Temperature has been implicated as a selective agent for these clines, because the body size of laboratory populations allowed to evolve in culture at lower temperatures is larger. In this study, we investigated the hypothesis that larger size is favoured at lower temperature through natural selection on adult males. We measured life‐span and age‐specific fertility of males from lines of flies artificially selected for body size at two different experimental temperatures. There was an interaction between experimental temperature and body size selection for male fitness; large‐line males were fitter than controls at both temperatures, but the difference in fitness was greater at the lower experimental temperature. Smaller males did not perform significantly differently from control males at either experimental temperature. The results imply that thermal selection for larger adult males is at least in part responsible for the evolution of larger body size at lower temperatures in this species. The responsible mechanisms require further investigation.  相似文献
