Mammals live in a homeostatic symbiosis with their gastrointestinal microbiota. The mammalian host provides the microbiota with nutrients and a stable environment; whereas the microbiota helps shaping the host's gut mucosa and provides nutritional contributions. Microorganisms start colonizing the gut immediately after birth followed by a succession of populations until a stable, adult microbiota has been established. However, physiological conditions differ substantially among locations in the gut and determine bacterial density and diversity. While Firmicutes and Bacteroidetes dominate the gut microbiota in all mammals, the bacterial genera and species diversity is huge and reflects mammalian phylogeny. The main function of the gastrointestinal epithelium is to absorb nutrients and to retain water and electrolytes, yet at the same time it is an efficient barrier against harmful compounds and microorganisms, and is able to neutralize antagonists coincidentally breaching the barrier. These processes are influenced by the microbiota, which modify epithelial expression of genes involved in nutrient uptake and metabolism, mucosal barrier function, xenobiotic metabolism, enteric nervous system and motility, hormonal and maturational responses, angiogenesis, cytoskeleton and extracellular matrix, signal transduction, and general cellular functions. Whereas such effects are local at the gut epithelium they may eventually have systemic consequences, e.g. on body weight and composition.  相似文献

Ambiguous odor compounds, partly citrus-like and partly woodyin odor character, were seen to change in odor quality whenevaluated in the same session as more prototypical odors. Whentested with characteristically citrus odors, the ambiguous compoundsseemed more woody, and when tested with characteristically woodyodors, the ambiguous odorants were higher in citrus character,an example of perceptual contrast. Response frequency biaseswere ruled out as an explanation for this shift by an experimentin which responses other than citrus and woody ratings wereasked of the subjects during the contextual exposure. Simplesensory adaptation was found to be a potential contributor tothe effect, and a sufficient condition to produce similar shiftsin odor quality. However, adaptation was not a necessary conditionto produce the effect. This was seen in reversed pair experimentsin which the contextual odors were presented after the ambiguousstimuli. The contextual shift was robust—it was obtainedwith different ambiguous odors, contextual (conditioning) odors,numbers of contextual odors, orders of presentation of contextualodors relative to ambiguous odors, scale types, and rating tasksduring the presentation of contextual odors.  相似文献

Mitochondrial DNA (mtDNA) is essential for the ability of mammalian cells to generate a functional oxidative phosphorylation system. Mutations in mtDNA occur in human disease and also during ageing. Here, we address three questions concerning the occurrence and accumulation of mtDNA mutations during the lifespan of the mammalian cell. What sort of mutations accumulate with age in humans and other mammals? How is the female germ line spared from the accumulation of such mutations as occurs in many somatic tissues, so that neonates normally start life with a ‘clean sheet'? Is the occurrence of mtDNA mutations associated with the functional decline of cells and tissues during ageing? We argue that mtDNA mutations in somatic cells do not just reflect a passive imprint of ageing, but they are causally associated with the loss of bioenergetic function during the ageing process.  相似文献

The history of somatic cell genetics from the late 1950s to the present day is considered. Studies in this field provided for the elucidation of numerous fundamental and applied problems, including spontaneous mutagenesis, gene mapping with somatic cell hybrids, and genetic mechanisms of carcinogenesis (e.g., cell protooncogenes, oncogenes, and tumor suppressor genes were revealed). Mice have become the mammalian model of choice for the application of genetics in biomedical research due to the evolutionary conservation of physiological systems and their attendant pathologies among all mammals as well as the exceptional power of genetic research technologies in the species. Beginning from aberrant phenotypes, a large number of mouse mutants and natural polymorphisms have been cloned, providing much information about the molecular basis of physiological processes. Additionally, the variable expression of these mutations in different inbred strain backgrounds has demonstrated the importance of modifier genes, which are also susceptible to cloning. Research efforts are keeping pace with these developments. In the area of gene discovery, large, government-funded mutagenesis programs now exist, and as a matter of great practical importance, recent evidence suggests that the same genes may be involved in the natural polymorphisms affecting disease in mice and humans. In parallel, dramatic advances are also being made in our ability to measure physiological processes in mice, and the advent of expression profiling promises revolutionary advances in understanding phenotype at the molecular level. Gene-driven approaches have relied on engineering the mouse genome, including adding, subtracting, and replacing genes and, most recently, the ability to control gene activity reversibly. Together, these multiple advances in our technical abilities have created extraordinary opportunities for future discovery.  相似文献

Defended prey frequently advertise to potential predators usingmultimodal warning displays. Signaling through more than onesensory pathway may enhance the rate of avoidance learning andthe memorability of these learned avoidances. If this is so,then mimetic insects would gain more protection from mimickinga multimodal rather than a monomodal model. Day-old domesticchicks (Gallus gallus domesticus) were used to examine whethera common insect warning odor (pyrazine) enhanced learning andmemorability of yellow prey, a common warning color. Pyrazineincreased the rate at which the chicks learned to avoid unpalatableyellow prey, and how well this learned avoidance was rememberedafter a 96-h interval. After 96 h, mimics of the multimodalprey were avoided, whereas mimics of the monomodal prey werenot. In the absence of pyrazine, chicks generalized their learnedavoidance of the unpalatable yellow prey to palatable greenprey; however, the presence of pyrazine reduced this color generalization.These results suggest that much is to be gained from signalingmultimodally, for both models and mimetic prey species. Thepresence of multimodal prey in the habitat may also advantagethe predators as it allows it them to distinguish more easilybetween palatable and unpalatable prey.  相似文献

Compared with visual and auditory imagery, little is known aboutolfactory imagery. There is evidence that respiration may bealtered by both olfactory perception and olfactory imagery.In order to investigate this relationship, breathing parameters(respiratory minute volume, respiratory amplitude, and breathingrate) in human subjects during olfactory perception and olfactoryimagery were investigated. Fifty-six subjects having normalolfactory function were tested. Nasal respiration was measuredusing a respiratory pressure sensor. Using an experimental blockdesign, we alternately presented odors or asked the subjectsto imagine a given smell. Four different pleasant odors wereused: banana, rose, coffee, and lemon odor. We detected a significantincrease in respiratory minute volume between olfactory perceptionand the baseline condition as well as between olfactory imageryand baseline condition. Additionally we found significant differencesin the respiratory amplitude between imagery and baseline conditionand between odor and imagery condition. Differences in the breathingrate between olfactory perception, olfactory imagery, and baselinewere not statistically significant. We conclude from our resultsthat olfactory perception and olfactory imagery both have effectson the human respiratory profile and that these effects arebased on a common underlying mechanism.  相似文献
