Regional variations of cone density, cone types and cone mosaic were investigated by light microscopy in yolk sac fry, parr and adults of salmon (Salmo salar) and trout (Salmo trutta trutta) and related to the feeding habits of different stages. The retinal organization of the two species is similar, as are their feeding habits. In parr and adults of both species a region with high cone density was found in the ventro-temporal part of the retina. The lowest values of cone density were found centrally in the dorsal half of the retina. An increase in cone density towards the ora serrata was noted in all retinae investigated. There is a good agreement between the density distribution of the cones in parr of salmon and trout and their feeding habits. The parr feed to a great extent on the invertebrate drift. Facing the current, the parr search the water region in front and above themselves for food organisms, thereby probably making use of the ventro-temporal area of high cone density. The connection between the cone density distribution and the feeding habits is less clear in adult salmon and trout, but the high cone density region is probably used when they feed on surface organisms and schooling prey. The ecological significance of cone types and cone mosaics is discussed. During growth there was a change of the cone mosaic from a regular square pattern in the fry towards a less regular square or row pattern in the adults. The position of regions with high and low cone density did not change during development in each species and had a similar distribution in the two species. On the other hand, the range of cone density values becomes relatively greater as the animal grows older. The cone density distribution seems to be more homogeneous in parr than in adults. Differences in the relative density distribution of the cones are influenced by recruitment at the retinal periphery and by spreading out of existing cells. In addition to the ora serrata, mitoses were also observed in more central parts of the retina having well-differentiated receptors and a cone mosaic. Mitoses were noted in all three nuclear layers but were most frequent in the outer nuclear layer, in which the resulting cells probably differentiate into rods. Undifferentiated zones similar to the peripheral growth zone of the retina were found on both sides of the falciform process in fry. In parr and adults an undifferentiated zone persisted only on the temporal side. It probably contributes to the high cone density in the ventro-temporal part of the retina by a delivery of new cells.  相似文献

Root characteristics of soybean (Glycine max (L.) Merr.) improve drought avoidance by increasing water uptake from the soil profile. Screening genotypes for improved root architecture without breaking the taproots or losing lateral roots is a challenge. Due to difficulty in separating roots from field or potting soil, a rapid and effective screening method with a suitable growth medium to assess root characteristics under controlled conditions needs to be established. We describe two screening techniques “the cone system” and “the tube system” using turface:sand medium. In the cone system thirty four soybean lines including cultivars and exotic plant introduction (PI) lines were evaluated for tap root length and root biomass, 12 days after sowing. Eight replications per line were grown in a growth chamber. Significant differences among genotypes for tap root length were detected by the cone system. Validity of results from the cone system was tested by evaluating root growth 21 days after planting for eight lines in the tube system. A coefficient of determination of 0.72 indicated good agreement between the two screening systems for evaluating genotypes for rooting depth. The cone system will be a useful method to easily and rapidly assess soybean genotypes for root growth.  相似文献

Rod-specific photoreceptor dystrophies are complicated by the delayed death of genetically normal neighboring cones. In transgenic (Tg) swine with a rod-specific (rhodopsin) gene mutation, cone photoreceptor physiology was normal for months but later declined, consistent with delayed cone cell death. Surprisingly, cone postreceptoral function was markedly abnormal when cone photoreceptor physiology was still normal. The defect was localized to hyperpolarizing cells postsynaptic to the middle wavelength-sensitive cones. Recordings throughout postnatal development indicated a failure of cone circuitry maturation, a novel mechanism of secondary cone abnormality in rod dystrophy. The results have implications for therapy for human retinal dystrophies and raise the possibility that rod afferent activity plays a role in the postnatal maturation of cone retinal circuitry.  相似文献

Stable actin structures play important roles in the development and specialization of differentiated cells. How these structures form, are organized, and are used to mediate physiological processes is not well understood in most cases. In Drosophila testis, stable actin structures, called actin cones, mediate spermatid individualization, a large-scale cellular remodeling process. These actin cones are composed of two structural domains, a front meshwork and a rear region of parallel bundles. Myosin VI is an important player in proper actin cone organization and function. Myosin VI localizes to the cones' fronts and its specific localization is required for proper actin cone formation and function during individualization. To understand how these structures are organized and assembled, ultrastructural studies are important to reveal both organization of actin and the precise localization of actin regulators relative to regions with different filament organizations. In the present work, we have developed a novel pre-embedding immunogold-silver labeling method for high-resolution analysis of protein distribution in actin structures which allowed both satisfactory antibody labeling and good ultrastructural preservation. Electron microscopic studies revealed that myosin VI accumulated at the extreme leading edge of the actin cone and preferentially localized throughout the front meshwork of the cone where branched actin filaments were most concentrated. No myosin VI labeling was found adjacent to the membranes along the length of the cone or connecting neighboring cones. This method has potential to reveal important information about precise relationships between actin-binding proteins, membranes, and different types of actin structures.  相似文献

The geometrical surfaces of soil-burrowing animals were imitated and modeled on a cone component, the measuring tip part of a soil cone penetrometer. These biomimetic surfaces are concave dimples, convex domes and two wavy forms. The conventional cone surface and the biomimetic cone surfaces were analyzed in ANSYS 11.0 program to estimate cone equivalent stress and soil equivalent stress. Results show that biomimetic surfaces with the geometrical structures have lower cone equivalent stresses and soil equivalent stresses than that with conventional (smooth) surface. The least maximum cone equivalent stress and least maximum soil equivalent stress were recorded for biomimetic surfaces with concave dimples and wavy form-2 respectively. The two-body abrasive wear of biomimetic cone surfaces and conventional (smooth) cone surface were run on a rotary disk type of abrasive wear testing machine. The biomimetic cone surfaces were found to have lower abrasive wear than the conventional surface. It was found that and biomimetic cone surface with concave dimples has the lowest abrasive wear among the all tested surfaces.  相似文献

Retinitis pigmentosa (RP) is a major source of blindness caused by a large variety of mutations that lead to the death of rod photoreceptors. After rods die, cones gradually die from progressive oxidative damage. Several types of antioxidant formulations have been shown to reduce cone cell death over a relatively short-time frame, but in order for this strategy to be translated into a new treatment for patients with RP, prolonged effects will be needed. In this study, we determined that orally administered N-acetylcysteine (NAC) reduced cone cell death and preserved cone function by reducing oxidative damage in two models of RP, rd1(+/+) and rd10(+/+) mice. In rd10(+/+) mice, supplementation of drinking water with NAC promoted partial maintenance of cone structure and function for at least 6 months. Topical application of NAC to the cornea also reduced superoxide radicals in the retina and promoted survival and functioning of cones. Since oral and/or topical administration of NAC is feasible for long-term treatment in humans, and NAC has a good safety profile, it is reasonable to consider clinical trials to evaluate the effects of prolonged treatment with NAC in patients with RP.  相似文献

  • The study of intraspecific seed packaging (i.e. seed size/number strategy) variation across different populations may allow better understanding of the ecological forces that drive seed evolution in plants. Juniperus thurifera (Cupressaceae) provides a good model to study this due to the existence of two subspecies differentiated by phenotypic traits, such as seed size and cone seediness (number of seeds inside a cone), across its range.
  • The aim of this study was to analyse seed packaging (seed mass and cone seediness) variation at different scales (subspecies, populations and individuals) and the relationship between cone and seed traits in European and African J. thurifera populations.
  • After opening more than 5300 cones and measuring 3600 seeds, we found that seed packaging traits followed different patterns of variation. Large‐scale effects (region and population) significantly contributed to cone seediness variance, while most of the seed mass variance occurred within individuals. Seed packaging differed between the two sides of the Mediterranean Sea, with African cones bearing fewer but larger seeds than the European ones. However, no differences in seed mass were found between populations when taking into account cone seediness. Larger cones contained more pulp and seeds and displayed a larger variation in individual seed mass.
  • We validated previous reports on the intraspecific differences in J. thurifera seed packaging, although both subspecies followed the same seed size/number trade‐off. The higher seediness and variation in seed mass found in larger cones reveals that the positive relationship between seed and cone sizes may not be straightforward.We hypothesise that the large variation of seed size found within cones and individuals in J. thurifera, but also in other fleshy‐fruited species, could represent a bet‐hedging strategy for dispersal.

The biochemical bases for the differences in cone and rod photoreceptor physiology have not been thoroughly examined because of the difficulty in obtaining cone photoreceptor components. We report here the purification and preliminary characterization of a bovine cyclic GMP phosphodiesterase (PDE) which is enriched in cone photoreceptors. The cone PDE was purified at least 15,000-fold to apparent homogeneity from bovine retinas by DEAE-cellulose and cGMP-Sepharose affinity chromatography. The trypsin-activated cone PDE hydrolyzed cGMP with efficiency similar to that of the rod PDE. However, a number of characteristics distinguished the cone PDE from the rod isozyme including the subunit structure. As previously reported, the apparent molecular weight of the cone PDE large subunit (alpha') was slightly larger than either of the large subunits of the rod PDE (93,500 versus 88,000 and 84,000). Three other smaller polypeptides were associated with the alpha' subunit (Mr = 11,000, 13,000, and 15,000), one of which (11,000) may be identical to the rod PDE gamma subunit. Cone phosphodiesterase binds at least 10-fold more cyclic GMP/mol of PDE than the rod photoreceptor isozyme. Cyclic GMP binds to this noncatalytic site with high affinity (Kd = 11 nM) and dissociates very slowly (t1/2 = 10-20 min at 37 degrees C). Purified rod transducin activated the cone PDE in solution to at least 90% of the trypsin-activated level. The concentration of rod transducin required for half-maximal activation of cone PDE (15 nM) was 50-fold lower than that necessary for half-maximal activation of rod PDE. Thus several properties of the cone phosphodiesterase clearly distinguish it from the rod isozyme and could account for some differences in cone and rod physiology.  相似文献

 以系统揭示其表型变异程度和变异规律为目的,对祁连山青海云杉(Picea crassifolia )天然分布区的10个群体的8个种实性状进行了比较分析。采用方差分析、多重比较,相关分析、聚类分析等多种分析方法,对群体间和群体内的表型多样性进行了讨论。方差分析结果表明,各种实性状在群体间都存在极显著差异,除球果干质量和球果长/球果径外,其余性状在群体内都存在着极显著的差异;表型分化系数即群体间变异为27.18%,小于群体内的变异(72.82%);球果长、球果径、球果干质量、球果形状指数、种子长、种子宽、千粒重和种子形状指数的变异系数分别为:10.08%、5.80%、19.29%、9.66%、8.38%、15.34%、6.52%和13.94%;8个种实性状间多数呈极显著或显著的正相关,球果干质量、种子长、千粒重、球果长和球果径为青海云杉易测定和重要的种实性状;种实性状呈现出以经度变异为主的梯度规律性;通过表型性状的聚类分析可以将青海云杉10个群体划分为4类。  相似文献

Retinal cone cells exhibit distinctive photoresponse with a more restrained sensitivity to light and a more rapid shutoff kinetics than those of rods. To understand the molecular basis for these characteristics of cone responses, we focused on the opsin deactivation process initiated by G protein-coupled receptor kinase (GRK) 1 and GRK7 in the zebrafish, an animal model suitable for studies on retinal physiology and biochemistry. Screening of the ocular cDNAs identified two homologs for each of GRK1 (1A and 1B) and GRK7 (7-1 and 7-2), and they were classified into three GRK subfamilies, 1 A, 1B and 7 by phylogenetic analysis. In situ hybridization and immunohistochemical studies localized both GRK1B and GRK7-1 in the cone outer segments and GRK1A in the rod outer segments. The opsin/GRKs molar ratio was estimated to be 569 in the rod and 153 in the cone. The recombinant GRKs phosphorylated light-activated rhodopsin, and the Vmax value of the major cone subtype, GRK7-1, was 32-fold higher than that of the rod kinase, GRK1A. The reinforced activity of the cone kinase should provide a strengthened shutoff mechanism of the light-signaling in the cone and contribute to the characteristics of the cone responses by reducing signal amplification efficiency.  相似文献

Egg shells from 14 widely different species of wild birds were examined, using collodion casts of the inside surface of the shell. Organic cores, cones, cone junctions and pores were counted and in each case measurements of nearest neighbour distances were taken. Shell thickness was also measured.
It was found that, using mean values for each egg shell, there were highly significant, and in some cases almost perfect linear relationships between core numbers, cone numbers, cone junction numbers and pore numbers on the one hand and their respective nearest neighbour distances expressed as the reciprocal of the mean of the squares of these distances on the other. The results also showed that the distribution of all four structures was not random, but had a degree of uniformity increasing up the series pores < cone junctions < cores < cones.
The following relationships were also highly significant: core and cone numbers, core and cone junction numbers, and cone and cone junction numbers. The relationship of pore numbers to any of the other three structures was of no great significance.
Shell thickness was related curvilinearly to core numbers and to cone numbers and linearly to nearest neighbour values for cones. It was also related to pore numbers.
It is suggested that, apart from pores the other three structures and their distribution all depend upon the number of cores which are the original centres of crystallization from which the shell grows. To a lesser extent shell thickness also depends upon these structures. Tentative suggestions are also made concerning the origin of the pores.  相似文献

Fertilization Cone of Carp Eggs as Revealed by Scanning Electron Microscopy   总被引:3,自引:1,他引:2  
The process of formation of the fertilization cone in carp eggs was examined by scanning electron microscopy. The fertilized eggs responded to penetration of one sperm by primary and secondary steps of formation of a fertilization cone of unique morphology. In the primary step, the earliest fertilization cone was seen at the superior or anterosuperior part of a fused sperm head in inseminated eggs fixed 20 sec after immersion in fresh water. The cone reached a maximum of more than 10 μm in length and 3–4 μm in thickness by 40 sec, resulting in a transient plugging of the micropylar canal. In the secondary step, usually seen at 105–120 sec, a conformation reminiscent of a very small caldera volcano was formed, with the shortened earlier cone and part of the sperm tail at its top. By 2.5 min, the fertilization cone had become conical, and the sperm tail still extended from its top. At 3 min, the sperm tail was often not detectable, but a cytoplasmic eminence was still seen as a trace of the fertilization cone. The role of the earlier fertilization cone in blocking polyspermy is discussed.  相似文献

用扫描电镜(SEM)观察了北美香柏 Thuja occidentalis 雌球果的发育过程。在北京,北美香柏的雌球果是在八月初由营养芽转变而来,雌球果一般有4~6对苞片,中间2~3对可育,每一苞片腋部着生两枚胚珠,在可育苞片腋部最先观察到一扁平的隆起,并在其上分化出两个胚珠原基,接着分化出珠被和珠心,最后形成扁平而两侧对称的胚珠。在北美香柏雌球果发育过程中,约一半的雌球果在2~3对可育苞片中位于下面的1~2对的腋部产生3个胚珠原基,中间一个较小,并在以后的发育中逐渐退化。由此推测北美香柏的雌球果可能是由祖先类群中每一苞片具多于2个胚珠的雌球果演化而来。在光镜下对雌球果维管系统的观察发现,传粉前幼小雌球果的苞片内仅有一束维管束,传粉后随着苞片基部的居间生长,有4—8束维管束在苞片内形成,但是新发育的维管束木质部和韧皮部相对位置与正常叶性器官一致,这与在以往报道的柏科植物成熟雌球果的苞片中均有反向维管束的发育不同。北美香柏雌球果早期发育和维管束分析结果支持傅德志和杨亲二提出的解释裸子植物生殖器官形态演化的“苞鳞-种鳞复合体”理论。关键词北美香柏;雌球果的发育;胚珠分化;SEM  相似文献

Summary The ultrastructure of the rectal papillae of the parasitoid hymenopteran, Nasonia vitripennis (Walk), is described. These organs in this insect consist of four distinct cell types arranged as a closed, hollow cone. The majority of the cells are present in the raised cone, and are characterised by large numbers of mitochondria arranged in a membranous labyrinth. A series of cells form a collar around the base of the cone. Junction cells have been identified which are present at the point of insertion of the cone into the rectal epithelium. The base of the cone consists of cells with elaborately folded plasma membranes facing both the central cavity of the cone, and the haemolymph. The structure of this rectal papilla is compared with those found in other insects.We are indebted to Professor E. W. The two most common mechanisms of pupillary screening-pigment migration in arthropod superposition eyes are the cone and longitudinal pigment migration mechanisms. The dynamics of each were investigated by optical modelling and by determining experimentally the relationship between eye glow brightness and screening pigment position within the eyes of two representative insect species: the noctuid moth Agrotis infusa and the dung beetle Copris elphenor. During dark adaptation, in both mechanisms, the screening pigment is contracted distally to expose the proximal half of each crystalline cone. During light adaptation the pigment migrates proximally and reduces light flux in the retina. In the longitudinal mechanism, pigment migrates into the clear zone of the eye. In the cone mechanism, pigment never enters the clear zone and is instead restricted to the proximal half of each crystalline cone: a migrating sleeve of pigment creates a small aperture at the end of the crystalline cone, the area of which depends on the degree of light adaptation. According to the model, the cone mechanism provides a limited range of light attenuation (ca. 0.6 log units) for which both good spatial resolution and accuracy of control are maintained, and within this range attenuation is controlled very finely. Beyond this range, whilst attenuation is still possible, diffraction at the pigment aperture and increasing coarseness of control worsen visual performance significantly. In contrast, the longitudinal mechanism provides a much larger useful range of light attenuation (up to several log units) and maintains reasonable fineness of attenuation control over the entire range (although not as fine as the cone mechanism). The experimental results support the model. An extensive survey of arthropods with superposition eyes reveals that the cone mechanism is almost exclusively possessed by those animals experiencing a narrow range of light intensities, and the longitudinal mechanism by those experiencing a wide range.Dedicated to Professor Rolf Elofsson on the occasion of his retirement from the Chair of Zoology in Lund  相似文献

In mammals, each cone had been thought to contain only one single type of photopigment. It was not until the early 1990s that photopigment coexpression was reported. In the house mouse, the distribution of color cones shows a characteristic division. Whereas in the upper retinal field the ratio of short wave to middle-to-long wave cones falls in the usual range (1:10), in the ventral retinal field M/L-pigment expression is completely missing. In the transitional zone, numerous dual cones are detectable (spatial coexpression). In other species without retinal division, dual cones appear during development, suggesting that M/L-cones develop from S-cones. Dual elements represent a transitory stage in M/L-cone differentiation that disappear with maturation (transitory coexpression). These two phenomena seem to be mutually exclusive in the species studied so far. In the comparative part of this report the retinal cone distribution of eight rodent species is reported. In two species dual cones appear in adult specimens without retinal division, and dual elements either occupy the dorsal peripheral retina, or make up the entire cone population. This is the first observation proving that all cones of a retina are of dual nature. These species are good models for the study of molecular control of opsin expression and renders them suitable sources of dual cones for investigations on the role and neural connections of this peculiar cone type. In the developmental part, the retinal maturation of other species is examined to test the hypothesis of transitory coexpression. In these species S-pigment expression precedes that of the M/L-pigment, but dual cones are either identified in a small number or they are completely missing from the developing retina. These results exclude a common mechanism for M/L-cone maturation: they either transdifferentiate from S-cones or develop independently.  相似文献

This study describes the seasonal patterns of seed and cone abortion in natural Pinus halepensis trees and assesses the effects of pollen quantity, tree size and cone location on seed and cone abortion. The cone-set ratio was similar to the expected value, based on literature on woody perennials. The seed-ovule ratio, however, was almost twice as high as its expected value. The investment in protective cone elements was high and did not vary with the seed-ovule ratio of a cone. Pollination factors influenced cone abortion, as indicated by the high abortion rate at the end of the pollination period. Furthermore, abortive cones had lower pollination rates and lower pollen loads than well developed cones. Effects of resource availability were assessed as effects of tree size and cone position on twigs. Small trees aborted more cones than big trees and cone abortion was higher in apical cones than in basal cones. Abortion in P. halepensis is selective, mediated by both pollen quantity and resource availability. The relative importance of pollen and resources is suggested to be flexible, probably varying between trees and years. The high seed-ovule ratio of P. halepensis is the result of high pollination rate and selective abortion. The selective abortion might be due to the high allocation to protective cone elements relative to the allocation to seeds.  相似文献

以实际的北京萤火虫复眼结构与生理参数与依据,基于已知的GRIN透镜特征,把小眼的整个光学系统分成几个部分被模型化。模型中,在晶体柱部分使用了一个径向梯度折射率透镜柱具有的抛物线轮廓折射率公式;在晶锥末端所接的半椭球GRIN透镜部分,使用了一个径向梯度和纵向梯度分布都存在的折射率公式.模型的尺寸与折射率的大小根据实际晶锥的测量值.通过对模型小眼的光线追迹,证明模型小眼基本上是一个平行光入射,年行光出射的望远镜系统.小眼光学系统的前半部分把入射的平行光束聚焦到中间焦点,而这个点的位置恰好位于实际晶锥的腰的部位,而光线再次经过晶锥近端以后又以大致平行光束从晶锥末端出射。此模型小眼的最大入射角为30度,在这个入射角内其角放大率基本上是相同的。随着对平行入射光束与小眼光轴的角度的增加,所产生的中间像越来越远离晶锥光轴。  相似文献

Rod and cone photoreceptor neurons utilize discrete PDE6 enzymes that are crucial for phototransduction. Rod PDE6 is composed of heterodimeric catalytic subunits (αβ), while the catalytic core of cone PDE6 (α') is a homodimer. It is not known if variations between PDE6 subunits preclude rod PDE6 catalytic subunits from coupling to the cone phototransduction pathway. To study this issue, we generated a cone-dominated mouse model lacking cone PDE6 (Nrl(-/-) cpfl1). In this animal model, using several independent experimental approaches, we demonstrated the expression of rod PDE6 (αβ) and the absence of cone PDE6 (α') catalytic subunits. The rod PDE6 enzyme expressed in cone cells is active and contributes to the hydrolysis of cGMP in response to light. In addition, rod PDE6 expressed in cone cells couples to the light signaling pathway to produce S-cone responses. However, S-cone responses and light-dependent cGMP hydrolysis were eliminated when the β-subunit of rod PDE6 was removed (Nrl(-/-) cpfl1 rd). We conclude that either rod or cone PDE6 can effectively couple to the cone phototransduction pathway to mediate visual signaling. Interestingly, we also found that functional PDE6 is required for trafficking of M-opsin to cone outer segments.  相似文献
