Degenerative alteration of the reproductive organs of female schistosomes in correlation with the change in egg-laying rate of schistosome pairs in vitro was studied by electron microscopy. The production of normal eggs by adult S. japonicum pairs decreased after 4 days in vitro followed by an increase of abnormal egg laying up to day 8. In S. mansoni, the yield of both normal and abnormal eggs decreased gradually from the start of maintenance in vitro in spite of a much higher pairing rate than in S. japonicum. The vitelline gland of 14-day in vitro-maintained S. japonicum stained with Fast red B, while that of S. mansoni did not. The ovary of both species exhibited regressive features after 14 days of maintenance in vitro. Ultrastructural examination showed that the vitelline cells and oocytes of S. japonicum and S. mansoni had already lost their structural integrity after 2 days in vitro and continued to exhibit signs of structural degeneration throughout the 14-day in vitro maintenance period. The regressive changes in reproductive potential of female S. mansoni maintained in vitro for 4 days could be reversed by surgically implanting the parasites into mouse mesenteric veins.  相似文献

International Journal for Parasitology14: 183–190. Sexually mature females of Schistosoma mansoni were separated from their male partners and surgically transferred to Nile rats (Arvicanthis niloticus). Over a period of 35 days there was a significant decrease in size of these worms and regression of the reproductive system took place. Electron microscope observations of the vitelline gland and ovary provided details of and a time scale for the regressive changes which took the form of a cessation of cell differentiation and turnover, together with extensive cell death. Survival of cells within these organs was restricted to the undifferentiated cells and by day 35 the worms resembled immature females. It is concluded that regression of the female reproductive system was the result of discontinued male stimulation.The nature and implications of the obligatory relationship between male and female S. mansoni are discussed.  相似文献

A comparative morphometric study was performed to identify host-induced morphological alterations in Schistosoma mansoni adult worms. A wild parasite population was obtained from a naturally infected rodent (Nectomys squamipes) and then recovered from laboratory infected C3H/He mice. Furthermore, allopatric worm populations maintained for long-term under laboratory conditions in Swiss Webster mice were passed on to N. squamipes. Suckers and genital system (testicular lobes, uterine egg, and egg spine) were analyzed by a digital system for image analysis. Confocal laser scanning microscopy (CLSM) showed details of the genital system (testicular lobes, vitelline glands, and ovary) and the tegument just below the ventral sucker. Significant morphological changes (p < 0.05) were detected in male worms in all experimental conditions, with no significant variability as assessed by CLSM. Significant changes (p < 0.05) were evident in females from the wild population related to their ovaries and vitelline glands, whereas allopatric females presented differences only in this last character. We conclude that S. mansoni worms present the phenotypic plasticity induced by modifications in the parasite's microenvironment, mainly during the first passage under laboratory conditions.  相似文献

The components of the female reproductive system of Diphyllobothrium latum, including ovary, ovicapt, oviduct, vitelline ducts, vitelline reservoir, vagina, seminal receptacle, ootype with unicellular Mehlis's gland, ootype-uterine duct and uterus were observed with the electron microscope. The epithelium of the female reproductive system ducts consists of a nucleate syncytial layer. Structural differences in apical surface of the ducts, the number of nuclei and organoids in syncytial layer as well as the number of underlaid muscles were revealed. The regional differentiations of the uterus wall were registered. The middle and distal region of uterus was covered with microtriches. The filamentous microtriches were observed in apical surface of vagina. The epithelium of seminal receptacle and distal region of uterus were underlaided by the powerful muscle layers. The fertilization canal was revealed. It was shown that the formation of egg shells implemented by the deposit of vitelline globules in their surface in the ootype-uterine duct. Structural and functional differences of different parts of female apparatus in various groups of cestodes are conditioned by species biology.  相似文献

Schistosoma mansoni adult worms with genital anomalies isolated from Nectomys squamipes (Muridae: Sigmodontinae) were studied by confocal laser scanning microscopy under the reflected mode. One male without testicular lobes (testicular agenesia/anorchism) and two females, one with an atrophied ovary and another with 17 uterine eggs, were identified. The absence of testicular lobes occurred in a worm presenting otherwise normal male adult characteristics: tegument, tubercles and a gynaecophoric canal with spines. In both female specimens the digestive tube showed a vacuolated appearance, and the specimen with supernumerary uterine eggs exhibited a developing miracidium and an egg with a formed shell. The area of the ventral sucker was similar in both specimens however the tegument thickness, ovary and vitelline glands of the specimen with the atrophied ovary were smaller than those of the one with supernumerary eggs. These reported anomalies in the reproductive system call attention to the need to improve our understanding of genetic regulation and the possible role of environmental influences upon trematode development.  相似文献

Successful reproduction is critical to pass genes to the next generation. Seminal proteins contribute to important reproductive processes that lead to fertilization in species ranging from insects to mammals. In Drosophila, the male's accessory gland is a source of seminal fluid proteins that affect the reproductive output of males and females by altering female post-mating behavior and physiology. Protein classes found in the seminal fluid of Drosophila are similar to those of other organisms, including mammals. By using RNA interference (RNAi) to knock down levels of individual accessory gland proteins (Acps), we investigated the role of 25 Acps in mediating three post-mating female responses: egg production, receptivity to remating and storage of sperm. We detected roles for five Acps in these post-mating responses. CG33943 is required for full stimulation of egg production on the first day after mating. Four other Acps (CG1652, CG1656, CG17575, and CG9997) appear to modulate the long-term response, which is the maintenance of post-mating behavior and physiological changes. The long-term post-mating response requires presence of sperm in storage and, until now, had been known to require only a single Acp. Here, we discovered several novel Acps together are required which together are required for sustained egg production, reduction in receptivity to remating of the mated female and for promotion of stored sperm release from the seminal receptacle. Our results also show that members of conserved protein classes found in seminal plasma from insects to mammals are essential for important reproductive processes.  相似文献

In Loligo forbesi Steenstrup, the female reproductive system consists of the ovary and accessory reproductive organs which include the oviducal gland, the nidamental gland, the accessory nidamental gland and seminal receptacle. Histological studies were made on the accessory reproductive organs of female L. forbesi. The various changes observed during maturation are described and the functional significance discussed. The secretions produced by the oviducal gland and nidamental gland apparently form the egg coats. The seminal receptacle serves to store spermatozoa after mating. Several components of the female reproductive system of Pharyngostomoides procyonis, including the vitellaria and vitelline duct, ovary and oviduct, Laurer's canal, and Mehlis' gland and associated ducts, were observed with the electron microscope. Vitelline follicles contain cells in various stages of development. Mature vitelline cells contain membrane-delimited clusters of vitelline globules near the plasma membrane. Cilia are present in the vitelline duct. The ovary contains germ cells in various stages of maturation. Oogonia are found in the peripheral region. Mature oocytes contain numerous dense bodies near the plasmalemma. Also included in the cytoplasm of mature oocytes are "nucleolus-like bodies," myelin-like bodies, and mitochondria containing dense granules and few cristae. The epithelium of the oviduct is ciliated. Sperm are present in the oviduct and in Laurer's canal. Two types of secretory cells found in Mehlis' gland are described.  相似文献

A tendency for increase both of numerical and of ecological factors of infestation in pseudophyllid cestodes is realized due to morphological peculiarities of the structure of their reproductive system. In the species E. rugosum with the closed bursiform uterus and intrauterine development of eggs, there are revealed the same degree of maturity of oocytes in ovary, the lack of sphincter or filtering cell at the base of the ovicapt infundibulum, and the presence of a large vitelline reservoir. By analogy with trematodes, an interconnection is traced between the degree of egg maturation in the uterus and morphology of uterus and vittelicles. The thickening of the egg membrane has been shown to occur not only by deposits of individual vitelline globules at the internal superficial egg layer, but also by binding of lipid droplets excreted from the surrounding parenchyma to the uterine tube along its entire length to the external egg surface. The male reproductive system of Eubothrium rugosum is characterized by the presence of a small cirrus pouch, by the cirrus supplied with microtrichia, which corresponds to the similar vagina structure, and by localization of prostate glands outside the cirrus pouch. Comparison is presented with other ultrastructural peculiarities of organization of the reproductive system in the earlier studied species of pseudophyllid cestodes, Diphyllobothrium latum.  相似文献

Male accessory gland proteins (Acps) in Drosophila are components of the seminal fluid and are transferred to females during copulation. In mated females, Acps enhance egg production, augment sperm storage, induce refractory mating behaviors, and affect the female's longevity. To address the functions of eight previously uncharacterized Acps and further analyze five others, we determined the tissues to which they target after transfer to females. Each Acp has multiple targets and is unique in its pattern of localization. Within the reproductive tract, Acps target to the uterus, oviduct, sperm storage organs, ovary and oocytes. Some Acps also leave the reproductive tract, to enter the hemolymph. Some Acps are detected on the surface of eggs laid by mated females but were not detectable within those eggs. Our results can help to identify the likely functions of these Acps as well as to create models for the mechanism of action of Acps.  相似文献

The objective of this work was to survey and compare the composition of the parts of the reproductive system of the female fowl in glycosaminoglycans and proteoglycans. Those parts analyzed were ovary, infundibulum, magnum, isthmus, shell gland and vagina. Methods of analysis included cellulose acetate electrophoresis, infrared spectroscopy, colorimetry, amino acid determination and scanning electron microscopy. Concentrations of glycosaminoglycans were higher in vagina, ovary, infundibulum and isthmus than in shell gland and magnum. Glycosaminoglycans may be important in those parts of the reproductive tract which contribute membraneous and mucosal material to the descending egg, and where fertilization of the egg occurs.  相似文献

Arginine vasotocin (AVT) is a neuropeptide involved in reproductive function in many nonmammalian vertebrates. We determined brain and plasma AVT concentrations during the estrous cycle and oviposition in the lizard Anolis carolinensis. There were no differences in AVT concentrations in the plasma or any brain region during the ovipositional sequence. However, we found that females with an egg in each oviduct and a large pre-ovulatory follicle (diameter > 4.5 mm) in one-ovary had significantly higher AVT concentrations in the supraoptic nucleus (SON) of the hypothalamus than did females with small pre-ovulatory follicles in both ovaries. In a second study, females with an egg in each oviduct and a large pre-ovulatory follicle had significantly greater AVT concentrations in the SON than females with only one oviductal egg and a large pre-ovulatory follicle or females with an egg in each oviduct and a small pre-ovulatory follicle in each ovary. Concentrations of AVT in other brain regions and in the plasma did not differ among these groups. Changes in steroid profiles during estrous and/or direct neural communication between the uterus, ovary, and brain may account for the changes in AVT concentrations seen in the supraoptic nucleus during the estrous cycle of Anolis carolinensis.  相似文献

Maturation and maintenance of normal reproductive function in female Schistosoma mansoni require a permanent association with the male, but the nature of this relationship is not well understood. The regional localization of a stimulatory factor in the male and its target in the female were investigated. Unisexual female and mature male worms were transected into segments of various lengths. Various combinations of transected male and female segments and intact worms were transferred to the mesenteric veins of recipient hamsters and were also maintained in vitro. In hamsters and in vitro, pairing took place between intact worms of each sex and segments of the other, and between segments of both sexes. The majority of female worms and segments so paired showed some reproductive development, as assessed by vitelline gland differentiation. In intact unisexual females paired with small male segments, vitelline gland development was limited to that portion of the worm that had been held by the male. Worm segments continued to display normal body contractions throughout 24 days of in vitro maintenance and morphological integrity was retained. It is concluded that 1) in the absence of a functioning gut, worm segments can survive for prolonged periods on nutrients absorbed through the tegument; 2) worm pairing, male stimulation, and the female developmental response are independent of central nervous control by the cerebral ganglia; 3) males have no centralized localization for the female-stimulating factor; 4) vitelline gland differentiation in the female requires local stimulation through male contact, and this is not propagated throughout the worm.  相似文献

瘤背石磺(Onchidium struma)是雌雄同体、异体交配的腹足纲贝类,其生殖系统较为复杂,通过解剖学和组织切片技术对成体瘤背石磺的生殖系统及产卵前后的组织学变化进行了系统的研究.结果表明:(1)雄性生殖系统主要南阴茎囊、阴茎、雄性附性腺、两性腺(早期主要产生精于)和储精囊等部分组成,而雌性生殖系统则由两性腺(后期主要产生卵子)、生殖细胞输送管、蛋白腺、黏液腺、受精囊和阴道等组成;(2)雄性生殖系统的组织学结构在产卵前后变化较小,但两性腺、卵蛋白腺和黏液腺的组织学在产卵前后变化显著;(3)产卵后的两性腺由于成熟卵子的排放,整体结构松散,部分腺泡中有少量未排出的成熟卵细胞和卵黄合成早期的卵母细胞;(4)产卵前的卵蛋白腺中含有许多强嗜碱性的小颗粒(组织学结构类似于卵鞘中的胚胎外周蛋白),产卵后腺体中的颗粒相对较大,且呈嗜酸性;(5)产卵前的黏液腺中存在嗜碱性区、嗜酸性区和混杂区三种区域,但是产卵前黏液腺以嗜酸性细胞为主,而产卵后的黏液腺中以嗜碱性细胞区域为主,且分泌管道中有一些嗜碱性物质.由此可见,卵蛋白腺的主要功能是分泌卵蛋白包裹受精卵形成卵外周蛋白层,而黏液腺则在产卵过程中分泌黏液物质形成卵鞘结构及链状的卵带.  相似文献

Recent investigations of sensory and behavioral cues that initiate sexual selection processes in the squid Loligo pealeii have determined that egg capsules deposited on the substrate provide a strong visual and chemotactile stimulus to males, even in the absence of females (1, 2, 3). The visual stimulus of egg capsules attracts males to the eggs, and when the males touch the eggs, they encounter a chemical stimulus that leads to highly aggressive fighting behavior. We have recently demonstrated that egg capsule extracts implanted in artificial egg capsules elicit this aggressive behavior (4). In this communication, we present evidence that the salient chemical factor originates in the ovary and perhaps the oviducal gland of the female reproductive tract.  相似文献

Observation of the sexual interactions of Sepioteuthis sepioidea squid during the short reproductive stage of their lives showed a scramble competition system, with both male and female polygyny. Mature females were faithful to a specific location in the daytime, whereas males moved from group to group and formed short-term consortships with females. Males defended females from other males, particularly with an agonistic Zebra display. Male–female pairs exchanged Saddle-Stripe displays, after which males might display an on–off Flicker. There was considerable female choice. Only if a female responded to this display with a parallel Rocking action would she pair and would the male deposit spermatophores at the base of her arms, and only 50% of the time did females move the spermatophores internally to where sperm might be released and stored in the oviducal gland for later fertilization of eggs. This long-term set of interactions and solitary deposition of hidden egg strings contrasts with the attraction of both sexes to a common ‘egg mop’ laid by many females which was a site of competition in other loliginid squid. Since Sepioteuthis is a primitive genus within the family Loliginidae, it may represent a generalist reproductive strategy that evolved into a specialized localization one.  相似文献

The reproductive development and maturation of female schistosomes are crucial since their released eggs are responsible for the host immunopathology and transmission of schistosomiasis. However, little is known about the nutrients required by female Schistosoma japonicum during its sexual maturation. We evaluated the promoting effect of several nutrients (calf serum, red blood cells (RBCs), ATP and hypoxanthine) on the reproductive development of pre-adult females at 18 days post infection (dpi) from mixed infections and at 50 dpi from unisexual infections of laboratory mice in basic medium RPMI-1640. We found RBCs, rather than other nutrients, promoted the female sexual maturation and egg production with significant morphological changes. In 27% of females (18 dpi) from mixed infections that paired with males in vitro on day 14, vitelline glands could be positively stained by Fast Blue B; and in 35% of females (50 dpi) from unisexual infections on day 21, mature vitelline cells were observed. Infertile eggs were detected among both groups. To analyze which component of mouse RBCs possesses the stimulating effect, RBCs were fractionated and included in media. However, the RBC fractions failed to stimulate development of the female reproductive organs. In addition, bovine hemoglobin hydrolysate, digested by neutral protease, was found to exhibit the promoting activity instead of untreated bovine hemoglobin. The other protein hydrolysate, lactalbumin hydrolysate, exhibited a similar effect with bovine hemoglobin hydrolysate. Using quantitative RT-PCR, we found the expression levels of four reproduction-related genes were significantly stimulated by RBCs. These data indicate that RBCs provide essential nutrients for the sexual maturation of female S. japonicum and that the protein component of RBCs appeared to constitute the key nutrient. These findings would improve laboratory culture of pre-adult schistosomes to adult worms in medium with well-defined components, which is important to investigate the function of genes related to female sexual maturation.  相似文献

Conflicts between females and males over reproductive decisions are common . In Drosophila, as in many other organisms, there is often a conflict over how often to mate. The mating frequency that maximizes male reproductive success is higher than that which maximizes female reproductive success . In addition, frequent mating reduces female lifespan and reproductive success , a cost that is mediated by male ejaculate accessory gland proteins (Acps) . We demonstrate here that a single Acp, the sex peptide (SP or Acp70A), which decreases female receptivity and stimulates egg production in the first matings of virgin females , is a major contributor to Acp-mediated mating costs in females. Females continuously exposed to SP-deficient males (which produce no detectable SP ) had significantly higher fitness and higher lifetime reproductive success than control females. Hence, rather than benefiting both sexes, receipt of SP decreases female fitness, making SP the first identified gene that is likely to play a central role in sexual conflict.  相似文献

Although the majority of onychophorans are viviparous or ovoviviparous, oviparity has been described in a number of species found exclusively in Australia and New Zealand. Light microscopy and scanning and transmission electron microscopy were used to examine developing eggs and the reproductive tract of the oviparous Planipapillus mundus. Deposited eggs and fully developed eggs dissected from the terminal end of the uteri have an outer thick, slightly opaque chorion, and an inner thin, transparent vitelline membrane. The chorion comprises an outermost extrachorion, sculptured with domes equally spaced over the surface; a middle exochorion, with pores occurring in a pattern of distribution equivalent to that of the domes of the extrachorion above; and an innermost, thick endochorion consisting of a spongelike reticulum of cavities comparable to the respiratory network found in insect eggs. The vitelline membrane lies beneath the chorion, from which it is separated by a fluid‐filled space. The vitelline membrane tightly invests the developing egg. Examination of oocytes in the ovary and developing eggs at various stages of passage through the uterus indicate that the majority of chorion deposition occurs in the midregion of the uterus, where vast networks of endoplasmic reticulum are present in the columnar epithelium. The vitelline membrane, however, is believed to begin its development as a primary egg membrane, surrounding the developing oocytes in the ovary. The vitelline membrane is transformed after fertilization, presumably by secretions from the anterior region of the uterus; hence, it should be more accurately referred to as a fertilization membrane.
