Late blight caused by Phytophthora infestans is one of the most limiting diseases in solanaceous crops in the world. This pathogen is a main constraint in the highland Andes, where these plants are grown under high humidity conditions and continuous cropping. The aim of this research was to increase the available information on the biology of P. infestans, specifically on its level of genetic variation in south-western Colombia, an area where various solanaceous crops susceptible to this pathogen converge. The study was carried out by using AFLP molecular markers with the restriction enzymes EcoRI and MseI and different primer combinations. Results indicated a low level of genetic variation among the 26 isolates evaluated, with only 18 polymorphic bands out of 135 amplicons obtained (13.43%), a Nei's genetic diversity index of 0.04, and a Shannon's information index of 0.06.  相似文献

致病疫霉(Phytophthora infestans)引起的晚疫病是马铃薯的一种毁灭性病害。有效控制马铃薯晚疫病需要明确致病疫霉的群体遗传结构特征。采用8对SSR引物对采自福建省福州、长乐、漳州2010年分离的95株马铃薯致病疫霉进行遗传多样性分析。结果共检测出21个等位基因和26个基因型。三个地点致病疫霉菌群体间的平均遗传分化系数FST为0.22,在8个位点中有5个位点的等位基因频率分布差异显著。三个群体的观测纯合度小于期望纯合度,观测杂合度大于期望杂合度,以无性生殖为主。结果表明福建群体的遗传多样性高,群体间的存在较高的遗传分化度。  相似文献

通过交配型和甲霜灵抗性以及线粒体DNA单倍型、SSR和AFLP基因型分析对40个超级生理小种菌株进行了遗传多样性分析。在被测菌株中发现了A1、A2和自育3种不同类型的交配型。其中,A1和自育型菌株数量多,分别为21株和14株,而A2交配型仅5株。甲霜灵抗性测定检测出高抗菌株26株,敏感菌株14株。线粒体DNA单倍型测定出Ia型和IIa型两种,比例接近1:1。基于5个基因座被测40个超级生理小种菌株共鉴定出了7种SSR基因型。利用6对荧光引物共检测到258条AFLP谱带,其中多态性谱带204条,多态性为79.1%。将供试的40个菌株划分为38个基因型,几乎每个菌株都为1个特有基因型。而且,我国南方和北方超级生理小种群体存在着明显的遗传差异。结果表明我国致病疫霉超级生理小种具有丰富的遗传多样性,可以推断致病疫霉中的任何小种都可在多个抗病基因的强大选择压力下,在短时间内通过与之对应的无毒基因快速突变而成为超级生理小种。当前对致病疫霉生理小种的鉴定及监测对生产上利用抗病品种防控晚疫病的指导意义不大。  相似文献

Ethiopian cattle are under threat from uncontrolled mating practices and are at high risk of becoming genetically homogeneous. Therefore, to evaluate genetic diversity, population structure and degree of admixture, 30 microsatellite markers were genotyped using 351 DNA samples from 10 Ethiopian cattle populations and the Holstein breed. The mean number of alleles per cattle population ranged from 6.93 ± 2.12 in Sheko to 7.50 ± 2.35 in Adwa. The mean observed and expected heterozygosities were 0.674 ± 0.015 and 0.726 ± 0.019 respectively. Ethiopian cattle populations have maintained a high level of within-population genetic differentiation (98.7%), the remainder being accounted for by differentiation among populations (1.3%). A highly significant deficiency in heterozygotes was detected within populations ( F IS = 0.071; P  <   0.001) and total inbreeding ( F IT = 0.083; P  <   0.001). The study populations were highly admixed but distinct from pure Bos taurus and Bos indicus breeds. The various levels of admixture and high genetic diversity make Ethiopian cattle populations suitable for future genetic improvement and utilization under a wide range of agro-ecologies in Ethiopia.  相似文献

Goodwin, S. B., and Fry, W. E. 1994. Genetic analyses of interspecific hybrids between Phytophthora infestans and Phytophthora mirabilis. Experimental Mycology 18, 20-32. Four crosses were made between isolates of two host-specific Phytophthora species. Phytophthora infestans and Phytophthora mirabilis. In the two most successful crosses involving a common P. infestans A2 parent, allozyme analysis confirmed that 79 of 86 progeny were interspecific hybrids, 3 were presumed selfs, and 4 were either selfs or nonrecombinant parental types. Mating type, alleles at the allozyme locus glucose-6-phosphate isomerase, and the + alleles at a number of DNA fingerprinting loci segregated independently according to Mendelian expectation. Three DNA fingerprinting loci were tightly linked in P. mirabilis, but no other linkages were detected among these markers. Mitochondrial DNA was uniparentally inherited, mostly from the P. infestans parent. Growth rate segregated as a quantitative character. None of the 68 progeny tested infected Mirabilis jalapa (the host of P. mirabilis), 3 infected potato, and 4 were weakly pathogenic to tomato. Because most of the F1 hybrids could not infect any of the hosts infected by the parents, host specialization could provide a postzygotic as well as a prezygotic reproductive isolating mechanism for P. infestans and P. mirabilis in central Mexico. These results indicate that P. mirabilis probably is capable of a regular outcrossing mating system.  相似文献

The genetic diversity of the populations of Phytophthora infestans from some major potato production regions in China were detected by amplified restriction fragment polymorphism (AFLP) analysis. Among 200 combinations of primer pair screened, 12 combinations could generate consistent polymorphic bands using six tested isolates. The twelve combinations were used to amplify the genomic DNA of 50 isolates collected in China from 1997 to 2002. A total of 922 AFLP bands were obtained, and 530 of them,covering 57.5%, showed polymorphism. Cluster analysis using the unweighted pair-group method with arithmetic averages (UPGMA) separated 50 isolates into five AFLP groups which were correlated to groups defined by geographical origin, however, they were not correlated to groups defined by mating type, or response to metalaxyl and virulence. Parameters of genetic diversity calculated by POPGENE software indicated that the genetic diversity level of Phytophtora infestans population in China was not high.  相似文献

中国马铃薯晚疫病菌AFLP遗传多样性分析   总被引:3,自引:0,他引:3  
应用AFLP分子标记检测了我国部分马铃薯主要产区马铃薯晚疫病菌的遗传多样性及不同地区菌株间的亲缘关系。在200对引物组合中,利用6个菌株筛选出12对多态性好、带型清晰的引物组合。利用这12对引物组合对1997-2002年间采自我国黑龙江、河北、四川和云南4省的50株菌株进行了PCR扩增,共扩增出922条谱带,其中多态性标记530条,占57.5%。利用NTSYSpc软件中UPGMA算法构建了我国马铃薯晚疫病菌的亲缘关系树状图,聚类分析结果表明我国马铃薯晚疫病菌的遗传多样性与病原菌的地理来源有一定的相关性,而与交配型、生理小种和对甲霜灵的抗性无明显的相关性。用POPGENE软件计算了各群体间的遗传多样性参数,结果表明我国马铃薯晚疫病菌的遗传多样性程度不高,不同地区种群间分化不明显。  相似文献

致病疫霉Phytophthora infestans为马铃薯晚疫病的重要病原菌。通过从昆明市寻甸县采集110P和H-6两株致病疫霉,明确其染色体倍性、交配型、线粒体单倍型、毒性和甲霜灵敏感性,经对峙培养,利用改良的卵孢子萌发方法获得有性生殖F1代群体POP1(60株),并对POP1进行表型和基因型测定。结果表明:冷冻处理24h为最佳条件,卵孢子萌发率达5.09%±0.15%;POP1的交配型、毒性和甲霜灵敏感性均发生了分离,其中交配型分离比为A1:A2:A1A2:自育型(SF)=16:5:17:22,毒性分离比为抗性(R):敏感性(S)=11:49,甲霜灵敏感性分离比为抗性(R):敏感性(S)=2:58;3个表型的分离均偏离孟德尔单基因显性遗传特点。基于8对SSR多态性引物对POP1基因型分析表明,遗传相似系数为0.98时,可将所有菌株分为14个基因型;遗传相似系数为0.95时,可将POP1分为6个分支,其中优势群体为S1,占分离群体的61.67%。关联分析进一步表明,8对SSR所代表的基因型和几个重要表型有显著相关性(R2=0.6667)。本研究建立了高效的致病疫霉卵孢子萌发体系,解析了有性生殖后代群体遗传结构特点,为深入探索致病疫霉的变异规律及病害流行趋势提供了理论基础。  相似文献

In the European bison, Bison bonasus (L), 15 proteins coded by 20 loci have been studied. Two of these loci (Es-3 and Ca) are polymorphic and the remaining 18 are monomorphic. The average heterozygosity of bisons in Polish herds is 3.5%. The value does not differ from those found in populations of other large mammals in spite of restoration of the European bison population from a mere 13 individuals.  相似文献

 The parasitic plant Orobanche cumana Wallr. has become a limiting factor for sunflower crops in infested countries. Over the past few years the progression of this parasitic plant, its introduction into new countries, and the development of new and more virulent races have all been observed. Consequently, the survey and understanding of broomrape population evolution is now crucial for the establishment of efficient breeding programmes. With this in prospect, the genetic variability of O. cumana populations from infested European countries, Bulgaria, Romania, Turkey and Spain, was studied using RAPD markers. Eight populations with a total of 180 plants were analysed. Twenty three primers were used to obtain 133 reproducible bands which led to a binary matrix. This matrix was subjected to various complementary analyses including pairwise distances computed with the Nei and Li coefficient, AMOVA, Nei’s genetic diversity statistics, and an estimation of gene flow among populations with the infinite-island formula. The results gave consistent conclusions whatever the method used for data treatment. We show that this parasitic plant is probably self-pollinated, that there is little intra-population variability, and very little gene exchange appears to occur between different geographic regions. Populations were well structured and organized into two distinct groups (one group corresponding to the East European countries, Bulgaria, Romania and Turkey, and the other group corresponding to Spanish populations) and could have a monophyletic origin. These results are discussed in relation to the applied uses of RAPD markers in the determination of true O. cumana races instead of populations. Numbers of light-footed clapper rails Rallus longirostris levipes, an endangered bird inhabiting southern California salt marshes, have substantially declined from historic levels. RAPD (randomly amplified polymorphic DNA) analysis was employed to assess the genetic variability within and among four of the largest remaining light-footed clapper rail populations. A single, larger population of the endangered Yuma clapper rail Rallus longirostris yumanensis was used for comparison. A total of 325 RAPD primers were tested on DNA from a subset of five clapper rails composed of a single representative for each of the four light-footed clapper rail populations and a representative for the single Yuma clapper rail population. Of the 1338 amplified bands (loci) surveyed in these five representative birds, approximately 1% were polymorphic, indicating the level of differentiation across all loci is quite low. Nine primers yielding these 16 polymorphic bands were used to analyse 48 individuals from five populations. Five of these bands were polymorphic in both subspecies, six were polymorphic only within the light-footed clapper rails, and five were polymorphic only within the Yuma clapper rail samples. Considering the few bands that were polymorphic among the light-footed clapper rail populations, a surprisingly high level of population differentiation (GST= 0.28) was found. This is in accord with the results of AMOVA analyses which show that a fairly high percentage of the limited variability among the rails is due to either differences between subspecies or differences between the light-footed rail populations. Because inbreeding depression is suspected and overall genetic distances between populations are low, movement of light-footed clapper rails from larger populations into smaller ones might be considered as a management strategy. Employing RAPDs as one of a series of assays is useful in revealing the population structure of genetically depauperate species.  相似文献

Sixty-seven isolates of Phytophthora infestans collected from Himalayan hill regions and subtropical planes of India were characterized by RAPD markers to assess diversity and differentiation based on location of origin. Ten random decamer primers generated 161 polymorphic fragments. Association of P. infestans isolates on the dendrogram and PCO plot revealed two clear grouping based on geographical location of origin-hill isolates and plane isolates. Quantification of diversity by Shannon index of diversity analysis demonstrated that most of the diversity was present with a particular population (hill or plane) of P. infestans isolates, with 85% variation being within and 15% being between hill and plane isolates. Subtropical plane isolates of P. infestans exhibited higher variability compared to hill isolates and they were more dispersed on the PCO plot. No clear differentiation of isolates based on mating type was reflected on the dendrogram and PCO plot.  相似文献

When grown in a medium containing heat-stable potato tuber proteins, the oomycete Phytophthora infestans (Mont.) de Bary produces a set of exoproteinases active at neutral and mildly basic pH values. These extracellular proteinases have been shown (by SDS-PAGE in the presence of gelatin) to include at least six components differing in molecular weight. Inhibitory analysis and studies of the effects of the enzymes on various synthetic substrates show that the culture liquid of P. infestans contains mainly serine proteinases (specific for trypsin and subtilisin) and metalloproteinases. Their activity is suppressed by proteinase-inhibiting proteins from potato tubers. It is suggested that exoproteinases of P. infestans may be the metabolic target for natural proteinase inhibitors from potato.  相似文献

While population declines can drive the loss of genetic diversity under some circumstances, it has been unclear whether this loss is a general consequence of overharvest in highly abundant marine fishes. We compiled data from 11 049 loci across 140 species and found that allelic richness was lower in overfished populations within 9 of 12 genera and families. A multiple linear regression showed that allelic richness was on average 12% lower (< 0.0001) in overharvested populations after accounting for the effects of body size, latitude and other factors. Heterozygosity was on average 2% lower (= 0.030). Simulations confirmed that these patterns are consistent with a recent bottleneck in abundant species and also showed that our analysis likely underestimates the loss of rare alleles by a factor of two or three. This evidence suggests that overharvest drives the decay of genetic diversity across a wide range of marine fishes. Such reductions of genetic diversity in some of the world's most abundant species may lead to a long‐term impact of fishing on their evolutionary potential, particularly if abundance remains low and diversity continues to decay.  相似文献

Phytophthora sojae causes root and stem rot, one of the most important diseases of soybean worldwide. Genetic diversity of 32 Phytophthora sojae isolates of different geographic origin from Argentina was evaluated with RAPD markers. The isolates were collected from diseased soybean plants and soil samples from Santa Fe, Buenos Aires, C6rdoba and Entre Rios provinces, in the Pampeana Region. DNA was amplified with 20 decanucleotides primers. Seven primers amplified 49 fragments, of which 35 were polymorphic, indicating high variability. RAPD analysis detected intraspecific variability even among isolates of the same geographic origin.  相似文献

Glacial and interglacial cycles of the Pleistocene have led to severe range fluctuations of many species. These range shifts of the past often are reflected by extant genetic signatures. Retractions of distribution areas often have fostered splits into several small and isolated retreats as remnants of the formerly interconnected range. These processes often go in line with losses of intraspecific diversity. By contrast, large and interconnected distribution ranges mostly sustain high levels of genetic variability. The genetic impact of both scenarios strongly depends on the temporal scale. In the present study, we tested the genetic effects of an assumed long‐lasting widespread distribution during glacial periods and more short‐term population retractions to mountain archipelagos during warm stages. We analyzed polymorphic allozymes for individuals of the Eastern Large Heath butterfly, Coenonympha rhodopensis, including major parts of its distribution, such as central Italy and the Balkan Peninsula. Our data show extraordinarily high genetic diversity. The only remarkable genetic split is detectable between the central Apennines (Italy) and the Balkan mountain systems. The populations sampled over seven Balkan mountain systems (Jakupica, Shar Planina, Ossogovo, Pirin, Rila, Rhodopes, and Stara Planina) show low genetic differentiation. This low genetic differentiation and high genetic diversity diverges from the genetic structures frequently found in species with disjunct distributions. We therefore hypothesize that the obtained molecular structure is the product of down‐slope shift during the last cold stage and subsequent expansion over the lowlands of the Balkan Peninsula. The current mountain restriction most probably occurred with the beginning of the postglacial warming, which is too short a time span to be of evolutionary relevance. Therefore, the recent high genetic diversities and low differentiation may still reflect long‐lasting glacial panmixia but not (yet) the recent disjunction. Glucose-6-phosphate isomerase (GPI) plays a key role in both glycolysis and gluconeogenesis. Isoforms of GPI are common, and therefore, its isozyme pattern is widely used to characterize isolates of Phytophthora infestans. Despite the importance of GPI in P. infestans studies, the gene encoding this enzyme has not yet been characterized. Furthermore, it has been suggested that P. infestans contains multiple copies of the gene but this hypothesis remains to be demonstrated. We have cloned and characterized GPI in various isolates of P. infestans as well as in several species of the genus Phytophthora. The gene contains 1671bp and encodes a protein with a predicted molecular weight of 60.8kDa. Multiple different alleles were identified and Southern analysis indicated certain P. infestans isolates carry several copies of the gene. Phylogenetic analysis revealed that P. infestans GPI is most closely related to sequences from Toxoplasma gondii, Arabidopsis thaliana, and Clarkia lewisii.  相似文献
