Personal protection measures against biting arthropods include topical insect repellents, area repellents, insecticide‐treated bednets and treated clothing. The literature on the effectiveness of personal protection products against arthropods is mainly limited to studies of prevention of bites, rather than prevention of disease. Tungiasis was successfully controlled by application of topical repellents and scrub typhus was reduced through the use of treated clothing. Successful reduction of leishmaniasis was achieved through the use of topical repellents, treated bednets and treated clothing in individual studies. Malaria has been reduced by the use of insecticide‐treated bednets (ITN), certain campaigns involving topical repellents, and the combination of treated bednets and topical repellents. Although area repellents such as mosquito coils are used extensively, their ability to protect humans from vector‐transmitted pathogens has not been proven. Taken together, the literature indicates that personal protection measures must be used correctly to be effective. A study that showed successful control of malaria by combining treated bednets and topical repellents suggests that combinations of personal protection measures are likely to be more effective than single methods. Implementation of successful programmes based on personal protection will require a level of cooperation commonly associated with other basic societal functions, such as education and food safety.  相似文献

For many decades effective insect repellents have relied on synthetic actives such as N,N -diethyl- meta -toluamide. Increasingly, consumers are seeking natural-based alternatives to many everyday products including insect repellents. While many studies have been published detailing the potential of essential oils to act as insect repellents, few oils have been identified as viable alternatives to synthetic actives. This study details the process involved in the selection of Australian essential oils effective as repellents and the subsequent testing of natural-based insect repellents using the selected oils. Using a combination of laboratory-based and field-based testing, oil from Melaleuca ericifolia was identified as being an effective insect repellent. When formulated into three different bases: an alcohol-based spray, an emulsion and a gel, these Melaleuca -based repellents were shown to be as effective at repelling mosquitoes Aedes vigilax (Skuse) (Diptera: Culicidae) and Verrallina carmenti (Edwards) (Diptera: Culicidae), the bush fly Musca vetustissima (Walker) (Diptera: Muscidae), and biting midges Culicoides ornatus (Taylor) (Diptera: Ceratopogonidae) and Culicoides immaculatus (Lee & Reye) (Diptera: Ceratopogonidae) as a synthetic-based commercial repellent. This study has shown that effective insect repellents based on natural active ingredients can deliver repellency on par with synthetic actives in the field. Three Melaleuca -based formulations have been registered as repellents and are now commercially available.  相似文献

The identification of molecular targets of insect repellents has been a challenging task, with their effects on odorant receptors (ORs) remaining a debatable issue. Here, we describe a study on the effects of selected mosquito repellents, including the widely used repellent N,N-diethyl-meta-toluamide (DEET), on the function of specific ORs of the African malaria vector Anopheles gambiae. This study, which has been based on quantitative measurements of a Ca2+-activated photoprotein biosensor of recombinant OR function in an insect cell-based expression platform and a sequential compound addition protocol, revealed that heteromeric OR (ORx/Orco) function was susceptible to strong inhibition by all tested mosquito repellents except DEET. Moreover, our results demonstrated that the observed inhibition was due to efficient blocking of Orco (olfactory receptor coreceptor) function. This mechanism of repellent action, which is reported for the first time, is distinct from the mode of action of other characterized insect repellents including DEET.  相似文献

Recently, the bed bug, Cimex lectularius L. has re-emerged as a serious and growing problem in many parts of the world. Presence of resistant bed bugs and the difficulty to eliminate them has renewed interest in alternative control tactics. Similar to other haematophagous arthropods, bed bugs rely on their olfactory system to detect semiochemicals in the environment. Previous studies have morphologically characterized olfactory organs of bed bugs’ antenna and have physiologically evaluated the responses of olfactory receptor neurons (ORNs) to host-derived chemicals. To date, odorant binding proteins (OBPs) and odorant receptors (ORs) associated with these olfaction processes have not been studied in bed bugs. Chemoreception in insects requires formation of heteromeric complexes of ORs and a universal OR coreceptor (Orco). Orco is the constant chain of every odorant receptor in insects and is critical for insect olfaction but does not directly bind to odorants. Orco agonists and antagonists have been suggested as high-value targets for the development of novel insect repellents. In this study, we have performed RNAseq of bed bug sensory organs and identified several odorant receptors as well as Orco. We characterized Orco expression and investigated the effect of chemicals targeting Orco on bed bug behavior and reproduction. We have identified partial cDNAs of six C. lectularius OBPs and 16 ORs. Full length bed bug Orco was cloned and sequenced. Orco is widely expressed in different parts of the bed bug including OR neurons and spermatozoa. Treatment of bed bugs with the agonist VUAA1 changed bed bug pheromone-induced aggregation behavior and inactivated spermatozoa. We have described and characterized for the first time OBPs, ORs and Orco in bed bugs. Given the importance of these molecules in chemoreception of this insect they are interesting targets for the development of novel insect behavior modifiers.  相似文献

Scaling up of insecticide treated nets has contributed to a substantial malaria decline. However, some malaria vectors, and most arbovirus vectors, bite outdoors and in the early evening. Therefore, topically applied insect repellents may provide crucial additional protection against mosquito-borne pathogens. Among topical repellents, DEET is the most commonly used, followed by others such as picaridin. The protective efficacy of two formulated picaridin repellents against mosquito bites, including arbovirus and malaria vectors, was evaluated in a field study in Cambodia. Over a period of two years, human landing collections were performed on repellent treated persons, with rotation to account for the effect of collection place, time and individual collector. Based on a total of 4996 mosquitoes collected on negative control persons, the overall five hour protection rate was 97.4% [95%CI: 97.1–97.8%], not decreasing over time. Picaridin 20% performed equally well as DEET 20% and better than picaridin 10%. Repellents performed better against Mansonia and Culex spp. as compared to aedines and anophelines. A lower performance was observed against Aedes albopictus as compared to Aedes aegypti, and against Anopheles barbirostris as compared to several vector species. Parity rates were higher in vectors collected on repellent treated person as compared to control persons. As such, field evaluation shows that repellents can provide additional personal protection against early and outdoor biting malaria and arbovirus vectors, with excellent protection up to five hours after application. The heterogeneity in repellent sensitivity between mosquito genera and vector species could however impact the efficacy of repellents in public health programs. Considering its excellent performance and potential to protect against early and outdoor biting vectors, as well as its higher acceptability as compared to DEET, picaridin is an appropriate product to evaluate the epidemiological impact of large scale use of topical repellents on arthropod borne diseases.  相似文献

The effects of the action of extracts from 82 plant species included in 39 families of indigenous flora against the larvae and adults of house fly (Musca domestica), the mosquitoes (Aedes aegypti and Anopheles atroparvus) and the German cockroach (Blattella germanica) are presented. Some of the extracts were prepared from air-dried and ground plant material which was exhaustively extracted with successive solvents of different polarity: ethyl ether, ethyl alcohol and water. Other extracts were prepared either by distillation of the whole water extracts of 24 hours macerated plants or by water extraction from residues of plants remained after distillation. These plant extracts act as toxicants, growth and development and reproduction inhibitors and repellents. The differential responses induced by these plant extracts on assessed insects were influenced by several factors such as the plant species, the solvents used for extractions, the species and the stages of insect life and also the methods employed for evaluation. Some of the tested plants appear to have a great potential for providing safer insect control agents.  相似文献

Background

Chemicals are used on bed nets in order to prevent infected bites and to kill aggressive malaria vectors. Because pyrethroid resistance has become widespread in the main malaria vectors, research for alternative active ingredients becomes urgent. Mixing a repellent and a non-pyrethroid insecticide seemed to be a promising tool as mixtures in the laboratory showed the same features as pyrethroids.

Methodology/Principal Findings

We present here the results of two trials run against free-flying Anopheles gambiae populations comparing the effects of two insect repellents (either DEET or KBR 3023, also known as icaridin) and an organophosphate insecticide at low-doses (pirimiphos-methyl, PM) used alone and in combination on bed nets. We showed that mixtures of PM and the repellents induced higher exophily, blood feeding inhibition and mortality among wild susceptible and resistant malaria vectors than compounds used alone. Nevertheless the synergistic interactions are only involved in the high mortality induced by the two mixtures.

Conclusion

These field trials argue in favour of the strategy of mixing repellent and organophosphate on bed nets to better control resistant malaria vectors.  相似文献

Mosquito repellents are used around the globe to protect against nuisance biting and disease‐transmitting mosquitoes. Recently, there has been renewed interest in the development of repellents as tools to control the transmission of mosquito‐borne diseases. We present a new bioassay for the accurate assessment of candidate repellent compounds, using a synthetic odour that mimics the odour blend released by human skin. Using DEET (N,N‐diethyl‐meta‐toluamide) and PMD (p‐menthane‐3,8‐diol) as reference compounds, nine candidate repellents were tested, of which five showed significant repellency to the malaria mosquito Anopheles gambiae sensu stricto (Diptera: Culicidae). These included: 2‐nonanone; 6‐methyl‐5‐hepten‐2‐one; linalool; δ‐decalactone, and δ‐undecalactone. The lactones were also tested on the yellow fever mosquito Aedes aegypti (Stegomyia aegypti) (Diptera: Culicidae), against which they showed similar degrees of repellency. We conclude that the lactones are highly promising repellents, particularly because these compounds are pleasant‐smelling, natural products that are also present in human food sources.  相似文献

The incidence of malaria may vary substantially between adjacent communities and within an individual community, even in areas of high malaria transmission. Analysis of the factors responsible for these local variations in the incidence of malaria may identify potential control measures. Factors shown to be associated with local protection against malaria in some situations include house position, house design, the use of insect repellents and mechanical barriers such as bednets and curtains. The efficacy of insecticide treated nets and curtains in preventing mortality and morbidity from malaria, at least in the short-term, has been demonstrated convincingly. However, other measures of personal protection have not been evaluated in large trials which have clinical malaria as their endpoint. Such trials are needed to see if new malaria control tools can be identified that will assist current international efforts to improve malaria control, especially in Africa. The millions of non-immune travellers who visit malaria endemic areas each year need to protect themselves against malaria and the ways in which they can do this most effectively have been studied extensively. However, less attention has been paid to the local population of malaria endemic areas. What steps can they adopt to provide personal protection against malaria and how effective are these measures? Clues to which measures might be effective can come from study of the reasons for local variations in the incidence of malaria.  相似文献

嗅觉在蚊虫的吸血寄主搜寻、产卵和糖源搜寻行为中起决定作用,而在交配行为中的作用并不清楚。本文系统全面地综述了近20年来蚊虫化学生态学和嗅觉识别的分子机理的研究。蚊虫的触角、下颚须和口喙上的嗅觉感器感觉环境中释放的各种挥发性化合物。气味分子与嗅觉气味结合蛋白和气味受体的结合所启动的一系列生化反应产生神经动作电位。蚊虫嗅觉神经元编码气味中化合物的组成、浓度及其暂时瞬间的浓度变化和空间分布。吸血前后神经元的活性在数量和质量上有变化,反映了蚊虫在搜寻吸血寄主和产卵行为上的调节。在吸血寄主搜寻中,人体和动物释放的二氧化碳、乳酸以及其他气味协同引诱蚊虫向目标气味源定向飞行,最后找到吸血寄主。而成熟产卵雌蚊是利用产卵场所释放的腐烂气味寻找适宜的产卵场所,一些蚊虫卵、幼虫或蛹分泌的产卵信息素引诱和刺激雌蚊产卵,并与产卵生境气味起协同作用。植物气味尤其是花香味引诱蚊虫找到蜜源。驱避剂也是直接或间接通过嗅觉起作用,一些驱蚊剂由于阻断嗅觉反应而抑制蚊虫的定向飞行。从植物、动物或人体以及产卵场所释放的气味中有望找到有效的引诱和驱避化合物。对蚊虫嗅觉识别机理的认识将使我们开发出有效的蚊虫诱捕技术,进而应用于种群监测和控制。  相似文献

Several lines of evidence suggest that insect repellent molecules reduce mosquito-host contacts by interacting with odorants and odorant receptors (ORs), thereby ultimately affecting olfactory-driven behaviours. We describe the molecular effects of 10 insect repellents and a pyrethroid insecticide with known repellent activity on two highly specific Aedes aegypti (Diptera: Culicidae) ORs, AaOR2 + AaOR7 and AaOR8 + AaOR7, exquisitely sensitive to key mosquito attractants indole and (R)-(-)-1-octen-3-ol, expressed in oocytes of Xenopus (Anura: Pipidae). Our study demonstrates that insect repellents can both inhibit odorant-evoked currents mediated by ORs and independently elicit currents in the absence of odorants. All of the repellents had effects on one or both ORs; most of these compounds were selective inhibitors and showed a high degree of specificity in their capacity to activate the two ORs. These results show that a range of insect repellents belonging to structurally diverse chemical classes modulate the function of mosquito ORs through multiple molecular mechanisms.  相似文献

Abstract

Insects have extremely sensitive systems of olfaction. These systems have been explored as potential sensors for odourants associated with forensics, medicine, security, and agriculture application. Most sensors based on insect olfaction utilize associative learning to “program” the insects to exhibit some form of behavioural response to a target odourant. To move to the next stage of development with whole-insect programmable sensors, an examination of how odourants are captured, processed and used to create behaviour is necessary. This review article examines how the neurophysiological, molecular, genetic and behavioural system of olfaction works and how an understanding of these systems should lead the way to future developments in whole-insect programmable sensors.  相似文献

嗅觉受体是参与昆虫嗅觉识别过程的一类重要蛋白。在昆虫的众多嗅觉受体中, 有一类受体明显不同于其他受体, 被称为Orco。该受体基因在不同昆虫种间高度保守, 且表达广泛。Orco在昆虫嗅觉识别过程中发挥关键作用。采用基因突变或RNAi等技术使Orco基因沉默后, 昆虫会出现严重的嗅觉缺陷, 但Orco本身不与气味配体结合, 它与传统嗅觉受体形成复合体Or-Orco, 促进传统嗅觉受体在神经元树突膜上的定位并维持其稳定性, 提高传统嗅觉受体对气味反应的效率。昆虫嗅觉受体的结构与脊椎动物的G蛋白偶联受体相似, 均有7个跨膜区, 但二者的膜拓扑结构相反, 昆虫嗅觉受体的N末端位于细胞质膜内, C末端在细胞质膜外, Orco与传统嗅觉受体通过保守的C末端区域相互作用形成一种新型的配体门控离子通道--Or-Orco复合体。阐明Orco在昆虫嗅觉识别中的功能机制, 可为开创基于昆虫嗅觉行为干扰的新的害虫防治措施提供基础。  相似文献

Plant-based repellents have been used for generations in traditional practice as a personal protection measure against host-seeking mosquitoes. Knowledge on traditional repellent plants obtained through ethnobotanical studies is a valuable resource for the development of new natural products. Recently, commercial repellent products containing plant-based ingredients have gained increasing popularity among consumers, as these are commonly perceived as "safe" in comparison to long-established synthetic repellents although this is sometimes a misconception. To date insufficient studies have followed standard WHO Pesticide Evaluation Scheme guidelines for repellent testing. There is a need for further standardized studies in order to better evaluate repellent compounds and develop new products that offer high repellency as well as good consumer safety. This paper presents a summary of recent information on testing, efficacy and safety of plant-based repellents as well as promising new developments in the field.  相似文献

Chemical repellents are promoted as a method to reduce ungulate–vehicle collisions and ungulate browsing damages to agricultural and forestry resources. We tested the effectiveness of two odour repellents (Mota FL and Wolf Urine) on the foraging behaviour and area avoidance of free-ranging roe deer (Capreolus capreolus) and red deer (Cervus elaphus). The effects of the repellents were assessed by comparing deer visitation rates to sand arenas before and after application of repellents and visitation rates to control arenas. Neither of the tested products reduced deer visitation rates. Rapid habituation to olfactory stimuli and lack of sensitivity to predator odours may explain the ineffectiveness of the repellents to alter the behaviour of the deer. The results indicate that the tested products have no effects on roe deer and red deer behaviour and suggest that the effectiveness of the chemical area repellents as a measure to reduce deer–vehicle collision risk and browsing damages is questionable.  相似文献

Background

Insect repellents are prophylactic tools against a number of vector-borne diseases. There is growing demand for repellents outperforming DEET in cost and safety, but with the current technologies R&D of a new product takes almost 10 years, with a prohibitive cost of $30 million dollar in part due to the demand for large-scale synthesis of thousands of test compounds of which only 1 may reach the market. R&D could be expedited and cost dramatically reduced with a molecular/physiological target to streamline putative repellents for final efficacy and toxicological tests.

Methodology

Using olfactory-based choice assay we show here that the fruit fly is repelled by not only DEET, but also IR3535 and picaridin thus suggesting they might have “generic repellent detector(s),” which may be of practical applications in new repellent screenings. We performed single unit recordings from all olfactory sensilla in the antennae and maxillary palps. Although the ab3A neuron in the wild type flies responded to picaridin, it was unresponsive to DEET and IR3535. By contrast, a neuron housed in the palp basiconic sensilla pb1 responded to DEET, IR3535, and picaridin, with apparent sensitivity higher than that of the DEET detectors in the mosquitoes Culex quinquefasciatus and Aedes aegypti. DmOr42a was transplanted from pb1 to the “empty neuron” and showed to be sensitive to the three insect repellents.

Conclusions

For the first time we have demonstrated that the fruit fly avoids not only DEET but also IR3535 and picaridin, and identified an olfactory receptor neuron (ORN), which is sensitive to these three major insect repellents. We have also identified the insect repellent-sensitive receptor, DmOr42a. This generic detector fulfils the requirements for a simplified bioassay for early screening of test insect repellents.  相似文献

In this Overview , we explore the linkages between basic research and the commercial development of novel pest management products in Australia. Despite the large volume of research in fundamental and applied aspects of entomology, very few new pest management products are developed and commercialised in Australia. Reasons for this include demanding and expensive regulatory requirements which (as in many other countries) mean that commercial development is the province of large multinational agrochemical companies. We describe the Australian regulatory system and the opportunities and difficulties it can present, using examples from recently registered Australian products, Magnet® moth attractant and the MOOV® range of insect repellents. The science behind these products is described in a series of papers in this issue of Australian Journal of Entomology . We also explore some of the commercial imperatives in novel product development, and aspects of the interactions between researchers and commercial partners. Finally, we discuss potential advantages of Australia as a locale for commercial development of novel products.  相似文献

Drosophila olfactory sensory neurons (OSNs) each express two odorant receptors (ORs): a divergent member of the OR family and the highly conserved, broadly expressed receptor OR83b. OR83b is essential for olfaction in vivo and enhances OR function in vitro, but the molecular mechanism by which it acts is unknown. Here we demonstrate that OR83b heterodimerizes with conventional ORs early in the endomembrane system in OSNs, couples these complexes to the conserved ciliary trafficking pathway, and is essential to maintain the OR/OR83b complex within the sensory cilia, where odor signal transduction occurs. The OR/OR83b complex is necessary and sufficient to promote functional reconstitution of odor-evoked signaling in sensory neurons that normally respond only to carbon dioxide. Unexpectedly, unlike all known vertebrate and nematode chemosensory receptors, we find that Drosophila ORs and OR83b adopt a novel membrane topology with their N-termini and the most conserved loops in the cytoplasm. These loops mediate direct association of ORs with OR83b. Our results reveal that OR83b is a universal and integral part of the functional OR in Drosophila. This atypical heteromeric and topological design appears to be an insect-specific solution for odor recognition, making the OR/OR83b complex an attractive target for the development of highly selective insect repellents to disrupt olfactory-mediated host-seeking behaviors of insect disease vectors.  相似文献

The behaviour of two of the most anthropophilic malaria vectors in the world, Anopheles gambiae Giles and An. arabiensis Patton, is revisited with respect to recent studies on their host preferences and the chemical ecology of host-seeking.
