A蛋白定向固定抗体用于椭偏光学生物传感器免疫检测   总被引:9,自引:0,他引:9  
椭偏光学生物传感器是在椭偏光学显微成像技术的基础上发展的一项生物传感技术。它能够直接观测固体表面上的生物分子面密度,毋需任何标记辅助,适合发展成为一种无标记免疫检测技术。研究了在硅片表面上通过A蛋白定向固定抗体分子用于椭偏光学生物传感器免疫检测的可能性。实验结果表明,通过A蛋白固定抗体得到的抗体膜层的均一性和固定量的重复性能够保证椭偏光学生物传感器免疫检测结果的质量。通过A蛋白定向固定的抗体的抗原结合位点趋向一致,显著提高了抗体与抗原结合的能力。此外,通过蛋白A固定的免疫球蛋白G分子能够结合更多的多克隆抗体分子说明通过A蛋白固定的蛋白质分子能够较好地保持其空间构象。  相似文献

利用热带次生林林窗边缘树表温和林窗区域地表温的观测资料,探讨了昼间林窗各热力作用面的热力效应及其变化规律,通过分析林窗边缘树表温和林窗地表温的变化,指出在林窗区域林窗边缘墙面是林冠面,林窗地面,林内地面之外的亲的第4热力作用面;各个热力作用面的热力效应随季节,位置和时刻的不同有着明显差异,在受浓雾影响的上午,林窗地面热力作用较强,在中午和下午林窗东侧林缘壁面,林窗东侧-东北侧地面的热力作用显著,中乖以林窗地面作用较强;而下午则以林窄边缘墙面作用较强,各个较力作用面的热力特征和相互作用将制约林窗的热力变化,影响植物的生长。  相似文献

The surface properties of nine Streptococcus mitis and four Peptostreptococcus micros strains from the oral cavity were examined and compared with a large group of oral streptococci. Zeta potential and contact angle measurements were employed to determine physico-chemical cell surface properties. In addition, elemental surface concentration ratios were obtained via X-ray photoelectron spectroscopy, and surface structures were examined with transmission electron microscopy. The S. mitis and P. micros strains were found to have higher isoelectric points, higher hydrophobicities and higher N/C surface concentration ratios than some other oral streptococci. The combined data suggest that both species possess large amounts of surface protein. All the S. mitis strains displayed abundant surface fibrils in negative staining, but the P. micros strains were devoid of surface appendages indicating that surface protein is present in different forms in the two species. The surfaces of S. mitis and P. micros type strains differed significantly from the other strains examined.  相似文献

A novel biotinylated gradient surface was designed to bind a streptavidin-FITC. The gradient polyethylene (PE) surface was prepared by a corona treatment with a power increase along the PE length, and successively, biotinylated gradient PE surface was prepared by the reaction of polyethylenimine (PEI) and biotin. Surface properties of the gradient PE surface changed according to PE length and chemical modification. The biotinylated gradient PE surface provided a continuous gradient of streptavidin-FITC through binding of avidin-biotin. The luminosity of fluorescence that corresponds to a streptavidin-bound FITC on the biotinylated gradient PE surface increased gradually on the surface along the PE length.  相似文献

The aim of this work was to study the influence of refining on various pulp properties such as freeness, specific surface area, specific volume, surface charge, total charge and elastic modulus. The results indicated that specific surface area of the pulps increased with increased refining, and at the same freeness level the pine pulp exhibited higher surface charge, surface area, and specific volume than the eucalyptus pulps. Also, the eucalypt pulps were much easier to beat than the pine pulps. The total fibre charge, as determined by conductometric titrations, was not affected by refining. However, the surface charge, as determined by titrations with poly-DADMAC, increased with refining. Increasing the specific surface area by refining resulted in a higher fibre surface charge and better fibre-fibre bonding. The change of the fibre surface charge during refining could be monitored using the FTIR characteristic bands within 1700-1300cm(-1).  相似文献

The role that bacterial surface hydrophobicity (surface tension) plays in determining the extent of adhesion of polymer substrates and phagocytic ingestion is reviewed. The early attachment phase in bacterial adhesion is shown to depend critically on the relative surface tensions of the three interacting phases; i.e., bacteria, substrate, and suspending liquid surface tension. When suspended in a liquid with a high surface tension such as Hanks balanced salt solution, the most hydrophobic bacteria adhere to all surfaces to the greatest extent. When the liquid surface tension (gamma LV) is larger than the bacterial surface tension (gamma BV), then for any single bacterial species the extent of adhesion decreases with increasing substrate surface tension (gamma SV). When gamma LV less than gamma BV then adhesion increases with increasing gamma SV. Bacterial surface tension also determines in part the extent of phagocytic ingestion and the degree to which antibodies specifically adsorb onto the bacterium resulting in opsonization. The nonspecific adsorption of antibodies results in a considerable modification in the surface properties of the bacteria. Bacterial surface hydrophobicity can be altered significantly through exposure to subinhibitory concentrations of antibiotics, surfactants, lectins, etc. The effect of these changes on subsequent phagocytic ingestion is discussed.  相似文献

This article presents an analogy to illustrate the effect of surface roughness on surface wettability. I used a water-filled balloon to represent water droplet, a toothpick to represent surface roughness and Styrofoam as the surface. The coordinated control of extracellular matrix degradation on the cell surface involves three crucial elements: secreted proteases and their inhibitors, surface protease receptors and integral membrane proteases. The roles that each of these elements play in cell surface proteolysis are described. The localization of proteases to the cell surface, protease activation, and regulation of cell surface proteolysis by protease inhibitors are key issues for elucidating the role of membrane proteases in tissue remodeling and tumour invasion.  相似文献

在扫描电镜下观察了龙胆科双蝴蝶属8个种和蔓龙胆属6个种的种子表面纹饰,观察结果表明,2属的种子表面特征均为网状纹饰类型。其中Tripterospermun cordatum,T.volubile,T.chinense,T.discoideum,T.japonicum,T.filicaule,Crawfurdia delavayi和C.pri-cei的种子表面为细网状;T.hirticalyx,T.pingbianense,C.puberula,C.tibetica,C.campanu-lacea和C.crawfordioides的种子表面为粗网状,在龙胆属Gentiana的不同组中均存在网状纹饰,因此,从种子表面特征可看出,龙胆属,双蝴蝶属和蔓龙胆属3属间具较近的亲缘关系。同时,在双蝴蝶属中,种之间在种子表面特征上有分化。在表面纹饰的具体特征上存在较大差别,而在蔓龙胆属中种间差别很小,仅在一些种子表面有无附属物上存在差异。  相似文献

The thermal behavior of luciferase on nanofabricated hydrophilic Si surface was investigated using molecular dynamics simulations. At high temperature, LUC_CHANGES (structural changes in the active site of luciferase) were smaller on the nanofabricated hydrophilic Si surface than on a non-nanofabricated hydrophilic Si surface, although LUC_CHANGES were slightly larger on the nanofabricated hydrophilic Si surface than on a non-nanofabricated hydrophobic Si surface. At room temperature, LUC_CHANGES were smaller on the nanofabricated hydrophilic Si surface than on a non-nanofabricated and wet-treated Si surface. Thus, nanofabricated hydrophilic Si surface would be preferable for practical applications.  相似文献

Three types of rough surface were processed by laser irradiation on the 3Cr2W8V material hot-work die steel surface.The wear experiments with smooth surface and rough surface samples were repeated on the pin-tray wear machine. According to the wear results, we studied the regularity of wear resistance of different rough surface samples. The results indicated that bionic rough surface can improve the wear resistance of the material and the wear resistance can be increased 1 - 2 times, compared with the smooth surface. Also, the wear resistance of the rough surface was affected by laser current and duration of impulse. The bigger the laser current or the impulse duration, the better is the wear resistance. When the distance between the same kind of units which are distributed on the surfaces is changed, the wear resistance changes. The wear resistance of a bionic rough surface on which the grid units were distributed at spacing of 1 mm was the best. And we designed the wear models.  相似文献

The distribution of charged groups on the surface of Dictyostelium cells and their change during development were examined by electronmicroscopy using cationic and anionic ferritins. The number of anionic sites on the cell surface decreased greatly during the course of development. The whole surface of vegetative cells stained strongly with cationic ferritin (CF). On the other hand, the surface of aggregation-competent cells had fewer negative charges and these were unequally distributed, the surface of the advancing area (lobopodial region) being devoid of anionic sites. The number of anionic sites on the cell surface decreased progressively during further development, and the suface of slug cells did not stain at all with CF. The cell surface did not stain with anionic ferritin at any developmental stage, indicating the absence of detectable cationic sites. The biological significance of these findings is discussed in connection with cell adhesiveness and movement.  相似文献

为探索岩溶地区城市遗存山体(urban remnant mountains, URMs)表面空气负离子浓度(negative air ion concentration, NAIC)特征及其影响因素,以贵阳市建成区内22座城市遗存山体为研究对象,通过样地调查法分析了城市遗存山体表面空气负离子浓度空间格局特征,运用单因素方差分析LSD法、pearson相关性及线性回归分析等方法分析了山体规模大小、山体斑块特征、山体平均植被覆盖度、植物物种多样性、植物群落体积生物量、地表辐射、空气温湿度、人为干扰程度、近红外辐射、短波红外辐射及城市遗存山体周边城市景观格局等因素对城市遗存山体表面空气负离子浓度的影响。结果表明:(1)不同规模城市遗存山体的表面空气负离子浓度表现为中型山体>小型山体>大型山体;空气负离子浓度在城市遗存山体坡向上呈东坡面和北坡面大于西坡面和南坡面的两级格局,同一坡向的不同坡位间无明显差异。(2)空气负离子浓度与山体表面温度极显著负相关,与山体表面空气湿度极显著正相关;植物群落体积生物量对城市遗存山体表面空气负离子浓度影响较大,且小型山体表现最明显;植物物种多样性对山...  相似文献

This work studied the influence of refining on various pulp properties such as freeness, specific surface area, specific volume, water retention value, surface charge, total charge and tensile index of five different pulps. At a freeness level the wheat straw pulp exhibited higher surface charge and total charge compared to other pulps. The specific surface area, specific volume and water retention value of the pulps increased with increased refining. The total fibre charge as determined by conductometric titrations was not affected by refining. However, the surface charge as determined by titrations with poly-DADMAC increased with refining. The increases in specific surface area of pulps by refining resulted in a higher fibre surface charge and also better fibre-fibre bonding. The linear regression models developed using experimental data were found to be accurate. The comparison of model predicted data and the experimental data showed an excellent agreement between them.  相似文献

The behavior of phosphatidylcholine monolayers at the air/water interface was studied by measuring their surface isotherm, surface potential, surface viscosity, and rate of hydrolysis by the dimeric phospholipase A2 from the venom of Crotalus atrox. The monolayers showed typical liquid-expanded behavior. In this phase, the surface potential was linearly dependent on surface concentration and extrapolated at zero concentration to a value characteristic of a liquid hydrocarbon/water interface. The rate of the reaction was measured by monitoring changes in area at constant surface pressure for 1,2-dioctanoyl- and 1,2-didecanoyl-3-sn-phosphatidylcholines, and by monitoring changes in surface potential for 1,2-dimyristoyl-, 1,2-dipalmitoyl-, 1-palmitoyl-2-oleoyl-, and 1-oleoyl-2-palmitoyl-sn-glycero-3-phosphocholines. The enzymatic hydrolysis is first order with respect to the enzyme-calcium complex which forms with a Kd = 1.5 mM. A mechanism is proposed to account for the dependency of the reaction rates on the surface concentration of the substrate. We postulate that the rate-limiting step is the decomposition of a quaternary complex formed from two phospholipid molecules, one calcium ion and one dimeric enzyme. The rate is independent of the surface pressure per se; addition of inert lipids to a monolayer at constant area, and hence constant surface concentration of the substrate, increases the surface pressure without changing the surface density of the substrate yielding maximal enzymatic rate. The enzyme is specific for loosely packed substrate molecules in the liquid-expanded state: transition into the liquid-condensed state or compression of the liquid-expanded layer beyond 80 A2/phospholipid strongly inhibits the enzymatic reaction. Our results show that surface recognition is a direct consequence of a bifunctional active site since it is only at a phospholipid surface that the distance between two substrate molecules is optimal for forming a catalytically competent enzyme-Ca2+-(substrate)2 complex.  相似文献

Following the theory of surface recombination in semiconductors, we have derived an expression for the rate of ion recombination at the membrane surface. The surface recombination rate is used in the boundary conditions of current flows at the interfaces. Expressions for the ion fluxes are then derived as functions of environmental variables and membrane parameters. Our analysis strongly suggests that the ion flow through a thin lipid membrane consists of two major components: the surface barrier jumping current and the surface recombination current that are controlled decisively by surface barrier height, surface trap density and surface recombination rates. These general formulations are useful not only for the calculation but also for the understanding of ion transport in thin lipid membranes under a variety of experimental conditions. The implications of this theory to biological membranes and its possible extensions are discussed.  相似文献

To study multi-fractal behavior of corroded steel surface, a range of fractal surfaces of corroded surfaces of Q235 steel were constructed by using the Weierstrass-Mandelbrot method under a high total accuracy. The multi-fractal spectrum of fractal surface of corroded steel was calculated to study the multi-fractal characteristics of the W-M corroded surface. Based on the shape feature of the multi-fractal spectrum of corroded steel surface, the least squares method was applied to the quadratic fitting of the multi-fractal spectrum of corroded surface. The fitting function was quantitatively analyzed to simplify the calculation of multi-fractal characteristics of corroded surface. The results showed that the multi-fractal spectrum of corroded surface was fitted well with the method using quadratic curve fitting, and the evolution rules and trends were forecasted accurately. The findings can be applied to research on the mechanisms of corroded surface formation of steel and provide a new approach for the establishment of corrosion damage constitutive models of steel.  相似文献
