In general, copper is primarily an osmoregulatory toxicant to fish and Cu toxicity is thought to be related to the rate of sodium loss. Looking at a previous research it is striking that gibel carp, Carassius auratus gibelio, do not seem so susceptible to the first ionoregulatory shock phase of Cu exposure, but rather build up physiological disturbances slowly until mortality occurs. Since it was noted that gibel carp experience severe hypoxia under Cu exposure, we hypothesised that, besides the Na loss, the slow depletion of liver glycogen stores contributed equally to the collapse of physiological integrity. It is clear from our results that glycogen stores are being depleted in Cu exposed fish and that dead fish suffered more extensive glycogen losses compared to surviving fish, with liver glycogen levels of 125 ± 8 mg g1 in dead fish compared to 230 ± 13 mg g1 in surviving fish. However, changes in liver glycogen did not contribute significantly to mortality, while changes in whole body sodium and the rate of sodium loss did. Whole body Na levels dropped from 1111 ± 48 μg g1 dry weight in control fish to 850 ± 54 μg g1 in surviving fish and to 607 ± 24 μg g− 1 in fish that had died resulting in Na loss rates of 1.25 ± 0.22 μg g− 1 h− 1 and 3.39 ± 0.19 μg g− 1 h− 1 in surviving and dead fish respectively. Our results support the finding that the rate of Na loss largely determines Cu toxicity in fish, even in resistant species.  相似文献

Three Algerian populations of female Pistacia atlantica shrubs were investigated in order to check whether their terpenoid contents and morpho-anatomical parameters may characterize the infraspecific variability. The populations were sampled along a gradient of increasing aridity from the Atlas mountains into the northwestern Central Sahara.As evidenced by Scanning Electron Microscopy, tufted hairs could be found only on seedling leaves from the low aridity site as a population-specific trait preserved also in culture. Under common garden cultivation seedlings of the high aridity site showed a three times higher density of glandular trichomes compared to the low aridity site. Increased aridity resulted also in reduction of leaf sizes while their thickness increased. Palisade parenchyma thickness also increases with aridity, being the best variable that discriminates the three populations of P. atlantica.Analysis of terpenoids from the leaves carried out by GC-MS reveals the presence of 65 compounds. The major compounds identified were spathulenol (23 μg g−1 dw), α-pinene (10 μg g−1 dw), verbenone (7 μg g−1 dw) and β-pinene (6 μg g−1 dw) in leaves from the low aridity site; spathulenol (73 μg g−1 dw), α-pinene (25 μg g−1 dw), β-pinene (18 μg g−1 dw) and γ-amorphene (16 μg g−1 dw) in those from medium aridity and spathulenol (114 μg g−1 dw), α-pinene (49 μg g−1 dw), germacrene D (29 μg g−1 dw) and camphene (23 μg g−1 dw) in leaves from the high aridity site. Terpene concentrations increased with the degree of aridity: the highest mean concentration of monoterpenes (136 μg g−1 dw), sesquiterpenes (290 μg g−1 dw) and total terpenes (427 μg g−1 dw) were observed in the highest arid site and are, respectively, 3-, 5- and 4-fold higher compared to the lower arid site. Spathulenol and α-pinene can be taken as chemical markers of aridity. Drought discriminating compounds in low, but detectable concentrations are δ-cadinene and β-copaene. The functional roles of the terpenoids found in P. atlantica leaves and principles of their biosynthesis are discussed with emphasis on the mechanisms of plant resistance to drought conditions.  相似文献

Amphibious fishes often emerse (leave water) when faced with unfavourable water conditions. How amphibious fishes cope with the risks of rising water temperatures may depend, in part, on the plasticity of behavioural mechanisms such as emersion thresholds. We hypothesized that the emersion threshold is reversibly plastic and thus dependent on recent acclimation history rather than on conditions during early development. Kryptolebias marmoratus were reared for 1 year at 25 or 30°C and acclimated as adults (one week) to either 25 or 30°C before exposure to an acute increase in water temperature. The emersion threshold temperature and acute thermal tolerance were significantly increased in adult fish acclimated to 30°C, but rearing temperature had no significant effect. Using a thermal imaging camera, we also showed that emersed fish in a low humidity aerial environment (30°C) lost significantly more heat (3.3°C min−1) than those in a high humidity environment (1.6°C min−1). In the field, mean relative humidity was 84%. These results provide evidence of behavioural avoidance of high temperatures and the first quantification of evaporative cooling in an amphibious fish. Furthermore, the avoidance response was reversibly plastic, flexibility that may be important for tropical amphibious fishes under increasing pressures from climatic change.  相似文献

The existence of a hydrodynamically relevant endothelial glycocalyx of ∼0.5 μm in thickness is well established in capillaries and venules in vivo. Since the glycocalyx is likely to have implications for broad areas of vascular physiology and pathophysiology, including endothelial-cell mechanotransduction, vascular permeability, and atherosclerosis, it is necessary to determine the extent to which the glycocalyx is present on arteriolar endothelium. We applied microviscometric analysis to data obtained using microparticle image velocimetry in cremaster-muscle arterioles of wild-type mice. Due to the pulsatile nature of the flow regimes in arterioles, data acquisition was triggered with the electrocardiogram at specific time points in the cardiac cycle. Results show the existence of a hydrodynamically relevant glycocalyx having a mean thickness of 0.38 μm in arterioles ∼20-70 μm in diameter (n = 20), which is ∼0.13 μm thinner (p = 0.03) than that found previously in venules having a similar diameter range and under similar hemodynamic conditions. Results from data obtained at multiple time points in the cardiac cycle show that the glycocalyx remains hydrodynamically relevant in arterioles with statistically insignificant changes in mean thickness throughout the cardiac cycle, despite the inherent unsteadiness of the flow regimes in these microvessels. These results provide direct in vivo confirmation of the existence of a hydrodynamically relevant surface glycocalyx that essentially eliminates fluid shear stress on arteriolar endothelium throughout the entire cardiac cycle.  相似文献

Aphanius dispar, a freshwater fish (mean total length ± sd, 3.42 ± 0.33 cm) was exposed to different concentrations of deltamethrin (2.25, 2.50 and 3.00 μg L−1), an insecticide used to control Dubas bug, Ommatissus lybicus in Oman. The histopathological changes in the structure of the gills in fish exposed to deltamethrin was studied using light, transmission and scanning electron microscopy. In light microscopy, the fish exposed to 2.25 μg L−1 deltamethrin, showed hypertrophy and hyperplasia of chloride cells, while in 2.50 μg L−1, additional changes like vacuolization, lifting of the lamellar epithelium and fusion of secondary lamellae were observed. The most severe alteration including vacuolization, desquamation of cells and dilation of intercellular space was recorded at 3.00 μg L−1. Transmission and scanning electron microscopy, supports the findings of light microscopy and further document the ultrastructural damage caused, especially in exposure to 2.50 and 3.00 μg L−1. Al l changes observed are described in detail. The deltamethrin exposure concentration of 2.25 μg L−1 seems to be a threshold above which any small increase in concentration results in severe damage. All present results have been compared with those of previous studies on gill damage caused by various chemical substances. The impacts of the damage on the functioning of gills as a respiratory organ are discussed.  相似文献

This study was carried out to determine upper (CTMax) and lower (CTMin) thermal tolerance, acclimation response ratio (ARR) and thermal tolerance polygon of the European sea bass inhabiting the Iskenderun Bay, the most southeasterly part of the Mediterranean Sea, at three acclimation temperatures (15, 20, 25 °C). Acclimation temperature significantly affected the CTMin and CTMax values of the fish. At 0.3 °C min−1 cooling or heating rate, CTMin ranged from 4.10 to 6.77 °C and CTMax ranged from 33.23 to 35.95 °C in three acclimation temperatures from 15 to 25 °C. Thermal tolerance polygon for the juveniles at the tested acclimation temperatures was calculated to be 296.14 °C2. In general, the current data show that our sea bass population possesses acclimation response ratio (ARR) values (0.25-0.27) similar to some tropical species. The cold tolerance values attained for this species ranged from 4.10 to 6.77 °C, suggesting that cold winter temperatures may not pose danger during the culture of European sea bass in deep ponds or high water exchange rate systems. Upper thermal tolerance is more of a problem in the southern part of the Mediterranean as maximum water temperature in ponds may sometimes exceed 33-34 °C, during which underground cool-water should be used to lower ambient water temperature in the mid-summer. For successful culture of sea bass in ponds, temperature should be maintained around 25 °C throughout the year and this can be managed under greenhousing systems using underground well-waters, commonly available in the region.  相似文献

The Lesser Mulberry Pyralid, Glyphodes pyloalis, is an important pest of mulberry. This pest feeds on mulberry leaves, and causes some problems for the silk industries in the north of Iran. The study of digestive enzymes is highly imperative to identify and apply new pest management technologies. Glucosidases have an important role in the final stages of carbohydrate digestion. Some enzymatic properties of α- and β-glucosidases from midgut and salivary glands of G. pyloalis larvae were determined. The activities of α- and β-glucosidase in the midgut and salivary glands of 5th instar larvae were obtained as 0.195, 1.07, 0.194 and 0.072 μmol−1 min−1 mg protein−1, respectively. Activity of α- and β-glucosidase from whole body of larval stages was also determined. Data showed that the highest activity of α- and β-glucosidase was observed in the 5th larval stage, 0.168 and 0.645 μmol−1 min−1 mg protein−1, respectively and the lowest activity in the 2nd larval stage, 0.042 and 0.164 μmol−1 min−1 mg protein−1, respectively. Results showed that the optimal pH for α- and β-glucosidase activity in midgut and salivary glands were 7.5, 5.5, 8-9 and 8-9 respectively. Also, the optimal temperature for α- and β-glucosidase activity in the midgut was obtained as 45 °C. The addition of CaCl2 (40 mM) decreased midgut β-glucosidase activity whereas α-glucosidase activity was significantly increased at this concentration. The α-glucosidase activity, in contrast to β-glucosidase, was enhanced with increasing in concentration of EDTA. Urea (4 mM) and SDS (8 mM) significantly decreased digestive β-glucosidase activity. Characterization studies of insect glucosidases are not only of interest for comparative investigations, but also understanding of their function is essential when developing methods of insect control such as the use of enzyme inhibitors and transgenic plants to control insect pest.  相似文献

The discovery of natural and natural-based compounds has resulted in its application as an alternative to synthetic algicides to control harmful algae in aquatic systems. Of the many natural-product-based algicides, sorgoleone, a natural plant product from Sorghum bicolor root exudates has been investigated for its controlling effect on different algal species and its acute fish toxicity. Growth of the blue green algal species Microcystis aeruginosa Kützing was completely inhibited by the crude methanol extract of sorghum root at 20 μg mL−1. The most noticeable inhibition was observed in the bioassay of n-hexane soluble extract, where 98% growth inhibition occurred in M. aeruginosa at the concentration of 1.25 μg mL−1. Sorgoleone very effectively controlled blue green algae inhibiting 97% of M. aeruginosa at 0.5 μg mL−1 and 99% of Anabaena affinis Lemmermann at 4 μg mL−1. In contrast, inhibition of the green algae species Chlorella vulgaris Beijerinck and Scenedensmus spp. at 16 μg mL−1 sorgoleone was 87 and 68%, respectively. There were no mortalities or adverse effects observed in any of the fish exposed to water control, solvent control, and a nominal concentration of 1 μg mL−1 during the test period. The no observed effect concentration (NOEC) value was 1.5 μg mL−1 for the tested fish (O. latipes). Sorgoleone can be considered as an effective and an ecologically and environmentally sustainable approach to controlling harmful algae.  相似文献

Community respiration and primary production were measured in a dense intertidal Zostera noltii bed on the Banc d’Arguin, Mauritania (West Africa) under aerial and submerged conditions. Metabolism was studied in situ in dark and transparent benthic chambers. CO2 fluxes in the air were measured over a series of short-term incubations (3 min) using an infrared gas analyzer. Dissolved inorganic carbon fluxes were calculated from concentration changes during one-hour underwater incubations. Air and underwater irradiance levels were measured every minute throughout the experiments. Carbon respiration was lower in the air (2.2 mmol m−2 h−1) than underwater (5.0 mmol m−2 h−1); similarly, a production-irradiance model fitted to the data indicated that gross maximal photosynthetic rate was markedly lower during emergence (6.0 mmol C m−2 h−1) than under water (42.7 mmol C m−2 h−1). The δ13C values observed in shoots indicated a decrease in atmospheric CO2 contribution, compared to dissolved inorganic carbon, in Z. noltii metabolism along a depth gradient within a single location. As the seagrass bed remains under a thin layer of water at low tide at the studied site, the large difference in primary production can be mainly attributed to photosynthesis inhibition by high pH and oxygen concentration, as well as to the negative feedback of self-shading by seagrass leaves during emersion. The observed differences in respiration can be explained by the oxygen deficit at night during low tide near the sediment surface, a deficit that is consistent with the abundance of anoxia-tolerant species.  相似文献

Ghrelin (GHR) is a recently discovered endocrine regulatory peptide of gastrointestinal origin with multiple functions including cardiovascular effects. However, contradictory data are available on the vascular actions of GHR in different organs and species. The aim of this study was to characterize the direct effect of the peptide on the canine coronary bed and to evaluate the role of the growth hormone secretagogue receptor (GHS-R) in the effect of GHR on coronary arterioles. The presence of GHS-R1a and 1b subtypes in canine coronary arterioles was investigated using Western blotting and immunohistochemistry. Responses of coronary arterioles with spontaneous and elevated vascular tone (the latter evoked by the thromboxane mimetic agent U46619, 10−7-10−6 mol/l) to GHR (10−9-3 × 10−7 nmol/l) were recorded by video-microscopy as changes of vessel diameter. Positive immunostaining for both GHS-R subtypes was found in the wall of intramural arterioles. The microarteriographic study results showed that GHR alone could not elicit any significant effect on vessel diameter of arterioles with spontaneous tone. However, when vascular smooth muscle was preconstricted by the thromboxane mimetic agent U46619, administration of GHR induced further constriction (+31 ± 9% increase in contraction p < 0.01). This was not abolished by the specific blockade of GHS-R1a by d-Lys3-GHRP-6 (5 × 10−6 mol/l). The results suggest that GHR induces tone-dependent constriction of canine coronary arterioles which is mediated by a receptor other than GHS-R1a.  相似文献

ADP-ribosyl cyclase and NAD+ glycohydrolase (CD38, E.C.3.2.2.5) efficiently catalyze the exchange of the nicotinamidyl moiety of NAD+, nicotinamide adenine dinucleotide phosphate (NADP+) or nicotinamide mononucleotide (NMN+) with an alternative base. 4′-Pyridinyl drugs (amrinone, milrinone, dismerinone and pinacidil) were efficient alternative substrates (kcat/KM = 0.9-10 μM−1 s−1) in the exchange reaction with ADP-ribosyl cyclase. When CD38 was used as a catalyst the kcat/KM values for the exchange reaction were reduced two or more orders of magnitude (0.015-0.15 μM−1 s−1). The products of this reaction were novel dinucleotides. The values of the equilibrium constants for dinucleotide formation were determined for several drugs. These enzymes also efficiently catalyze the formation of novel mononucleotides in an exchange reaction with NMN+, kcat/KM = 0.05-0.4 μM−1 s−1. The kcat/KM values for the exchange reaction with NMN+ were generally similar (0.04-0.12 μM−1 s−1) with CD38 and ADP-ribosyl cyclase as catalysts. Several novel heterocyclic alternative substrates were identified as 2-isoquinolines, 1,6-naphthyridines and tricyclic bases. The kcat/KM values for the exchange reaction with these substrates varied over five orders of magnitude and approached the limit of diffusion with 1,6-naphthyridines. The exchange reaction could be used to synthesize novel mononucleotides or to identify novel reversible inhibitors of CD38.  相似文献

Water dispersions of TEMPO-oxidized α-chitin nanowhisker (TOChN), partially deacetylated α-chitin nanowhisker/nanofiber mixture (DEChN), HCl-hydrolyzed chitin nanowhisker (HHChN) and squid-pen β-chitin nanofiber (SQChN) were prepared, and the properties of nano-dispersions and their cast films were characterized between the four chitin nano-samples. Because SQChN has the highest aspect ratio, its 0.1% dispersion had the highest shear stress and viscosity at the same shear rate in the four chitin nano-samples, and showed gel-like behavior in the whole shear rate range from 10−3 to 103 s−1. AFM images of the self-standing films showed that film surfaces consisted of characteristic chitin nano-elements with different morphologies and degrees of orientation between the four chitin samples, whereas all chitin nanowhisker/nanofiber films had similar thermal degradation points at ∼200 °C. The DEChN film had the highest tensile strength of ∼140 MPa, elongation at break of ∼10% and light-transmittance of 87% at 400 nm. In contrast, the SQChN film had the lowest tensile strength, Young's modulus and light-transmittance. All chitin nanowhisker/nanofiber films had similar oxygen permeabilities of ∼1 mL μm m−2 day−1 kPa−1, which was clearly lower than that (184 mL μm m−2 day−1 kPa−1) of a poly(lactic acid) film.  相似文献

The uptake kinetics of phosphate (Pi) by Myriophyllum spicatum was determined from adsorption and absorption under light and dark conditions. Pi uptake was light dependent and showed saturation following the Michaelis-Menten relation (in light: V = 16.91 × [Pi](1.335 + [Pi]), R2 = 0.90, p < 0.001; in the dark: V = 5.13 × [Pi](0.351 + [Pi]), R2 = 0.77, p < 0.001). Around 77% of the loss of Pi in the water column was absorbed into the tissue of M. spicatum, and only 23% was adsorbed on the surface of the plant shoots. Our study shows that M. spicatum shoots have a much higher affinity (in light: 3.9 μmol g−1 dw h−1 μM−1; in the dark: 3.7 μmol g−1 dw h−1 μM−1) and Vmax (maximum uptake rate, shoot light) for Pi uptake than many other aquatic macrophytes (in light: 0.002-0.23 μmol g−1 dw h−1 μM−1; in the dark: 0.002-0.19 μmol g−1 dw h−1 μM−1), which may provide a competitive advantage over other macrophytes across a wide range of Pi concentrations.  相似文献

The effect of salinity on growth, photosynthetic performance and osmotic acclimation was investigated in the eulittoral red algal species Bangiopsis subsimplex (Stylonematophyceae). The strain grew in a broad salinity range between 1 and 70 psu showing optimum growth between 10 and 50 psu. The saturation point Ik of the photosynthesis irradiance curves ranged between 153 and 83 μmol photons m− 2 s− 1 at all salinities and indicates an adaptation of B. subsimplex to moderate radiation conditions. Adjustments on the photosynthetic level (non-photochemical quenching) were sufficient to prevent damage to the photosynthetic apparatus as Fv/Fm values were constantly high (> 0.7) even when grown at the most hypo- and hypersaline conditions. As main low molecular weight carbohydrates, B. subsimplex contains the heteroside digeneaside and the polyol sorbitol. Digeneaside concentration was low and almost unchanged after hypersaline treatment (< 20 μmol g− 1 DW), i.e. it did not play a role in osmotic acclimation. By contrast, sorbitol levels increased linearly from 150 to 380 μmol g− 1 DW with increasing salinities between 5 and 60 psu, indicating its important function as an osmolyte and compatible solute under hypersaline conditions. The data presented are consistent with the natural habitat of B. subsimplex, i.e. the upper eulittoral zone.  相似文献

ATP prevents G-actin cysteine oxidation and vanadyl formation specifically induced by decavanadate, suggesting that the oxometalate-protein interaction is affected by the nucleotide. The ATP exchange rate is increased by 2-fold due to the presence of decavanadate when compared with control actin (3.1 × 10− 3 s− 1), and an apparent dissociation constant (kdapp) of 227.4 ± 25.7 μM and 112.3 ± 8.7 μM was obtained in absence or presence of 20 μM V10, respectively. Moreover, concentrations as low as 50 μM of decameric vanadate species (V10) increases the relative G-actin intrinsic fluorescence intensity by approximately 80% whereas for a 10-fold concentration of monomeric vanadate (V1) no effects were observed. Upon decavanadate titration, it was observed a linear increase in G-actin hydrophobic surface (2.6-fold), while no changes were detected for V1 (0-200 μM). Taken together, three major ideas arise: i) ATP prevents decavanadate-induced G-actin cysteine oxidation and vanadate reduction; ii) decavanadate promotes actin conformational changes resulting on its inactivation, iii) decavanadate has an effect on actin ATP binding site. Once it is demonstrated that actin is a new potential target for decavanadate, being the ATP binding site a suitable site for decavanadate binding, it is proposed that some of the biological effects of vanadate can be, at least in part, explained by decavanadate interactions with actin.  相似文献

Northern pike (Esox lucius L.) spermatozoa are uniflagellated cells differentiated into a head without acrosome, a midpiece and a flagellar tail region flanked by a fin structure. Total, flagellar, head and midpiece lengths of spermatozoa were measured and show mean values of 34.5, 32.0, 1.32, 1.17 μm, respectively, with anterior and posterior widths of the midpiece measuring 0.8 and 0.6 μm, respectively. The osmolality of seminal plasma ranged from 228 to 350 mOsmol kg−1 (average: 283.88 ± 33.05). After triggering of sperm motility in very low osmolality medium (distilled water), blebs appeared along the flagellum. At later periods in the motility phase, the tip of the flagellum became curled into a loop shape which resulted in a shortening of the flagellum and a restriction of wave development to the proximal part (close to head). Spermatozoa velocity and percentage of motile spermatozoa decreased rapidly as a function of time postactivation and depended on the osmolality of activation media (P < 0.05). In general, the greatest percentage of motile spermatozoa and highest spermatozoa velocity were observed between 125 and 235 mOsmol kg−1. Osmolality above 375 mOsmol kg−1 inhibited the motility of spermatozoa. After triggering of sperm motility in activation media, beating waves propagated along the full length of flagella, while waves appeared dampened during later periods in the motility phase, and were absent at the end of the motility phase. By increasing osmolality, the velocity of spermatozoa reached the highest value while wave length, amplitude, number of waves and curvatures also were at their highest values. This study showed that sperm morphology can be used for fish classification. Sperm morphology, in particular, the flagellar part showed several changes during activation in distilled water. Sperm motility of pike is inhibited due to high osmolality in the seminal plasma. Osmolality of activation medium affects the percentage of motile sperm and spermatozoa velocity due to changes in flagellar wave parameters.  相似文献

The two-spotted spider mite, Tetranychus urticae, is a worldwide pest species that overwinters as diapausing females. Cold hardening is presumed to start during diapause development to ensure the successful overwintering of this species. To address this hypothesis, we compared cold tolerance between non-diapausing and diapausing females. We measured supercooling point (SCP) and survival to acute cold stress by exposing the mites at a range of sub-zero temperatures (from −4 to −28 °C for 2 h). The mean SCPs of non-diapausing and diapausing females were −19.6±0.5 and −24.7±0.3 °C respectively, and freezing killed the mites. Diapausing females were significantly more cold tolerant than non-diapausing ones, with LT50 of −19.7 and −13.3 °C, respectively. Further, we also examined the effects of cold acclimation (10 d at 0 or 5 °C) in non-diapausing and diapausing females. Our findings indicated that diapause decreased SCP significantly, while cold acclimation had no effect on the SCP except for non-diapausing females that were acclimated at 5 °C. Acclimation at 5 °C enhanced survival to acute cold stress in diapausing and non-diapausing females, with LT50 of −22.0 and −17.1 °C, respectively. Altogether, our results indicate that T. urticae is a chill tolerant species, and that diapause and cold acclimation elevate cold hardiness in this species.  相似文献

The sydnone SYD-1 (3-[4-chloro-3-nitrophenyl]-1,2,3-oxadiazolium-5-olate] possesses important antitumor activity against Sarcoma 180 and Ehrlich tumors. We previously showed that SYD-1 depresses mitochondrial phosphorylation efficiency, which could be involved in its antitumoral activity. Considering the important role of mitochondria in the generation of reactive oxygen species (ROS) and the involvement of ROS in cell death mechanisms, we evaluated the effects of SYD-1 on oxidative stress parameters in rat liver mitochondria. SYD-1 (0.5 and 0.75 μmol mg−1 protein) inhibited the lipoperoxidation induced by Fe3+/ADP-oxoglutarate by approximately 75% and promoted total inhibition at the highest concentration tested (1.0 μmol mg−1 protein). However, SYD-1 did not affect lipoperoxidation started by peroxyl radicals generated by α-α′-azodiisobutyramidine dihydrochloride. The mesoionic compound (0.25–1.0 μmol mg−1 protein) demonstrated an ability to scavenge superoxide radicals, decreasing their levels by 9–19%. The activities of catalase and superoxide dismutase did not change in the presence of SYD-1 (0.25–1.0 μmol mg−1 protein). SYD-1 inhibited mitochondrial swelling dependent on the formation/opening of the permeability transition pore induced by Ca2+/phosphate by approximately 30% (1.0 μmol mg−1 protein). When Ca2+/H2O2 were used as inducers, SYD-1 inhibited swelling only by approximately 12% at the same concentration. NADPH oxidation was also inhibited by SYD-1 (1.0 μmol mg−1 of protein) by approximately 48%. These results show that SYD-1 is able to prevent oxidative stress in isolated mitochondria and suggest that the antitumoral activity of SYD-1 is not mediated by the increasing generation of ROS.  相似文献

Binding of the utmost N-terminus of essential myosin light chains (ELC) to actin slows down myosin motor function. In this study, we investigated the binding constants of two different human cardiac ELC isoforms with actin. We employed circular dichroism (CD) and surface plasmon resonance (SPR) spectroscopy to determine structural properties and protein–protein interaction of recombinant human atrial and ventricular ELC (hALC-1 and hVLC-1, respectively) with α-actin as well as α-actin with alanin-mutated ELC binding site (α-actinala3) as control. CD spectroscopy showed similar secondary structure of both hALC-1 and hVLC-1 with high degree of α-helicity. SPR spectroscopy revealed that the affinity of hALC-1 to α-actin (KD = 575 nM) was significantly (p < 0.01) lower compared with the affinity of hVLC-1 to α-actin (KD = 186 nM). The reduced affinity of hALC-1 to α-actin was mainly due to a significantly (p < 0.01) lower association rate (kon: 1018 M−1 s−1) compared with kon of the hVLC-1/α-actin complex interaction (2908 M−1 s−1). Hence, differential expression of ELC isoforms could modulate muscle contractile activity via distinct α-actin interactions.  相似文献
