To quantify both temperature (T) and water potential (ψ) effects on sesame (Sesamum indicum L.) seed germination (SG) and also to determine the cardinal T s for this plant, a laboratory experiment was carried out using hydrothermal time model (HTT). For this purpose, four sesame cultivars (‘Asbomahalleh’, ‘Darab’, ‘Dashtestan’ and ‘Yellowhite’) were germinated at seven constant T s (20, 25, 30, 35, 37, 39 and 43 °C) at each of the following ψ s (0, ? 0.12, ? 0.24 and ? 0.36 MPa; provided by PEG 8000). Germination rate (GR) and germination percentage (GP) significantly influenced by ψ, T and their interactions in all cultivars (P ≤ 0.01). There was no significant difference, based on the confidence intervals of the model coefficients, between cultivars, so an average of cardinal T s was 14.7, 35.4 and 47.2 °C for the minimum (T b), optimum (T o) and maximum (T c) T s, respectively, in the control condition (0 MPa). Hydrotime values in all cultivars decreased when T was increased to T o and then remained constant at T s > T o (15 MPa h?1). An average value of ψ b(50) was estimated to be ? 1.23 MPa at T s ≤ T o and then increased linearly (0.1041 MPa°Ch?1, the slope of the relationship between ψ b(50) and supra-optimal T s) with T when T s increased above T o and finally reached to zero at T c. The T b and T o values were not influenced by ψ, but T c value decreased (from 47.2 for zero to 43.5 °C for ? 0.36 MPa) at supra-optimal T s as a result of the effect of ψ on GR. Based on our findings, this model (as a predictive tool) and or the estimated parameter values in this study can easily be used in sesame SG simulation models to quantitatively characterize the physiological status of sesame seed populations at different T s and ψ s.  相似文献

This study evaluated the ability of a hydrothermal time model (HTT) to describe the kinetics of watermelon (Citrullus vulgaris cv. ‘Crimson sweet’) seed germination under different temperatures (T) and water potentials (ψ) and also to determine the cardinal temperatures of watermelon. Results indicated that ψ influenced germination rate and germination percentage. For this seed lot, cardinal temperatures were 10 °C for T b, 28.34 °C for T o and 40.8 °C for T c in the control (0 MPa) treatment. There was a decrease in hydrotime constant (θ H) when T was increased to T o and then remained constant at supra-optimal temperatures (30 MPah?1). Also, at temperatures above T o, ψ b(50) values increased linearly with T. The k T value (the slope of the relationship between ψ b(50) and T exceeds T o) of this seed lot was calculated as 0.076 MPa°Ch?1. Results this study show that when the HTT model is applied, it can accurately describe ψ b(g) and the course of germination around Ts (R 2 = 0.82). Moreover, the ψ b(50) was estimated to be ?0.96 MPa based on this model. Consequently, the germination response of watermelon for all Ts and ψs can be adequately described by the HTT model and enabling it to be used as a predictive tool in watermelon seed germination simulation models.  相似文献

Sublethal concentrations of chemical insecticides may cause changes in some behavioral characteristics of natural enemies such as functional responses. The residual effect of three synthetic insecticides including deltamethrin, fenvalerate and azadirachtin were studied on functional response of Habrobracon hebetor Say to Ephestia kuehniella Zeller larvae. Seven host densities (2, 4, 8, 16, 32, 64 and 96) were used during a 24 h period. The resulting data were appropriately fit to Type II functional response models in all treatments: (1) control (0.0916 h?1; and T h  = 0.2011 h); (2) deltamethrin (a = 0.0839 h?1; and T h  = 0.3560 h); (3) fenvalerate (a = 0.0808 h?1 and T h  = 0.3623 h); and (4) azadirachtin (a = 0.0900 h?1 and T h  = 0.2042 h). Maximum theoretical parasitism rate (T/T h ) was 119.34 estimated for control wasps. There was no significant difference between the values of attack rates (a and a + D a ) in all treatments while the handling time was statistically affected in female wasps treated with fenvalerate. Our findings will be useful in safe application of these insecticides in pest management programmes.  相似文献

A marine bacterial strain, F72T, was isolated from a solitary scleractinian coral, collected in Yap seamounts in the Pacific Ocean. Strain F72T is a Gram-negative, light-yellow-pigmented, motile, rod-shaped bacterium. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain F72T is related to the genus Novosphingobium and has high 16S rRNA gene sequence similarities with the type strains of Novosphingobium pentaromativorans US6-1T (97.7 %), Novosphingobium panipatense SM16T (97.6 %), Novosphingobium mathurense SM117T (97.2 %) and Novosphingobium barchaimii LL02T (97.1 %). Ubiquinone Q-10 was detected as the dominant quinone. The predominant cellular fatty acids were C18:1ω7c and C17:1ω6c. The genomic DNA G+C content of strain F72T was 63.4 mol %. The polar lipids profile contained phosphatidylglycerol, diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylmethylethanolamine, phosphatidylcholine, sphingoglycolipid and one uncharacterized lipid. Strain F72T shared DNA relatedness of 25 % with N. pentaromativorans JCM 12182T, 31 % with N. panipatense DSM 22890T, 21 % with N. mathurense DSM 23374T and 26 % with N. barchaimii DSM 25411T. Combined data from phenotypic, phylogenetic and DNA–DNA relatedness studies demonstrated that the strain F72T is a representative of a novel species of the genus Novosphingobium, for which we propose the name Novosphingobium profundi sp. nov. (type strain F72T = KACC 18566T = CGMCC 1.15390T).  相似文献

We examined changes in suspended-sediment yields (SSY) after a 50 % strip thinning in headwater streams draining a Japanese cedar (Cryptomeria japonica) and cypress (Chamaecyparis obtusa) plantation forest. We applied a paired-catchment analysis to treated (K T : 17.1 ha) and control (K C : 8.9 ha) catchments. Annual suspended-sediment yield (SSYan) in the prethinning period in the K T and K C catchments was 110.0 and 142.1 kg/ha per year. For the postthinning period, SSYan in the K T catchment became 5055.6 kg/ha per year, whereas that in the K C catchment increased 893.2 kg/ha per year. The paired-catchment analysis revealed that SSY in the K T catchment increased 17.0-fold compared with the prethinning period. However, the dominant hysteresis pattern remained clockwise in both pre- and postthinning periods. Sequences of large storm events in the postthinning period elevated SSY in both catchments. Increase in suspended sediment in the K T catchment was associated with the combined impacts of thinning and sequences of storm events during the period of thinning operation.  相似文献

The enhancement of the internal quantum efficiency (IQE) of deep-ultraviolet Al x Ga1-x N/Al y Ga1-y N (x < y) quantum wells (QWs) by fabricating one-dimensional Al nanogratings on a QW structure for inducing surface plasmon (SP) coupling is demonstrated. Through temperature-dependent photoluminescence (PL) measurement, the enhancements of IQE in different emission polarizations are illustrated. Due to the small difference in energy band level between the heavy/light hole and split-off valence bands, the IQEs of the transverse electric- (TE-) and transverse magnetic- (TM-) polarized emissions are about the same. When emission polarization is perpendicular to Al-grating ridges, the SP resonance mode for coupling with the QWs is dominated by localized surface plasmon (LSP). When emission polarization is parallel with Al-grating ridges, the coupled SP resonance mode may mix LSP and SP polariton. In this polarization, LSP can be excited because of the width fluctuation of a grating ridge. When the excitation laser polarization is perpendicular to Al-grating ridges, the strong LSP resonance at the excitation laser wavelength leads to stronger excitation and hence higher IQE levels.  相似文献

A moderately halophilic bacterium, designated strain 9-2T, was isolated from saline and alkaline soil collected in Lindian county, Heilongjiang province, China. The strain was observed to be strictly aerobic, Gram-negative, rod-shaped, oxidase-positive, catalase-positive and motile. It was found to require NaCl for growth and to grow at NaCl concentrations of 0.5–14 % (w/v) (optimum, 7–10 %, w/v), at temperatures of 10–45 °C (optimum 25–30 °C) and at pH 5.0–10.0 (optimum pH 8.0). Phylogenetic analysis based on 16S rRNA gene sequences revealed that strain 9-2T is a member of the genus Halomonas and is closely related to Halomonas desiderata DSM 9502T (96.68 %), Halomonas campaniensis DSM 1293T (96.46 %), Halomonas ventosae DSM 15911T (96.27 %) and Halomonas kenyensis DSM 17331T (96.27 %). The DNA–DNA hybridization value was 38.9 ± 0.66 % between the novel isolate 9-2T and H. desiderata DSM 9502T. The predominant ubiquinones were identified as Q9 (75.1 %) and Q8 (24.9 %). The major fatty acids were identified as C16:0 (22.0 %), Summed feature 8 (C18:1 ω6c/C18:1 ω7c, 19.6 %), Summed feature 3 (C16:1 ω6c/C16:1 ω7c, 12.6 %), C12:0 3-OH (12.0 %) and C10:0 (11.7 %). The DNA G+C content was determined to be 69.7 mol%. On the basis of the evidence presented in this study, strain 9-2T is considered to represent a novel species of the genus Halomonas, for which the name Halomonas heilongjiangensis sp. nov. is proposed. A novel actinomycete strain, designated TRM 49605T, was isolated from a desert soil sample from Lop Nur, Xinjiang, north-west China, and characterised using a polyphasic taxonomic approach. The strain exhibited antifungal activity against the following strains: Saccharomyces cerevisiae, Curvularia lunata, Aspergillus flavus, Aspergillus niger, Fusarium oxysporum, Penicillium citrinum, Candida albicans and Candida tropicalis; Antibacterial activity against Bacillus subtilis, Staphylococcus epidermidis and Micrococcus luteus; and no antibacterial activity against Escherichia coli. Phylogenetic analysis based on 16S rRNA gene sequences affiliated strain TRM 49605T to the genus Streptomyces. Strain TRM 49605T shows high sequence similarities to Streptomyces roseolilacinus NBRC 12815T (98.62 %), Streptomyces flavovariabilis NRRL B-16367T (98.45 %) and Streptomyces variegatus NRRL B-16380T (98.45 %). Whole cell hydrolysates of strain TRM 49605T were found to contain ll-diaminopimelic acid as the diagnostic diamino acid and galactose, glucose, xylose and mannose as the major whole cell sugars. The major fatty acids in strain TRM 49605T were identified as iso C16:0, anteiso C15:0, C16:0 and Summed Feature 5 as defined by MIDI. The main menaquinones were identified as MK-9(H4), MK-9(H6), MK-9(H8) and MK-10(H6). The polar lipids were diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, phosphatidylinositol and phosphatidylinositol mannoside. The G+C content of the genomic DNA was determined to be 71.2 %. The DNA–DNA relatedness between strain TRM 49605T and the phylogenetically related strain S. roseolilacinus NBRC 12815T was 60.12 ± 0.06 %, which is lower than the 70 % threshold value for delineation of genomic prokaryotic species. Based on the phenotypic, chemotaxonomic and phylogenetic data, strain TRM 49605T (=CCTCC AA2015026T = KCTC 39666T) should be designated as the type strain of a novel species of the genus Streptomyces, for which the name Streptomyces luozhongensis sp. nov. is proposed.  相似文献

A Gram staining negative, rod-shaped, aerobic bacterial strain J5-3T with a single polar flagellum was isolated from coking wastewater collected from Shaoguan, Guangdong, China. It was motile and capable of optimal growth at pH 6–8, 30 °C, and 0–2 % (w/v) NaCl. Its predominant fatty acids were 11-methyl C18:1 ω7c (29.2 %), C16:0 (20.6 %), C19:0 cyclo ω8c (18.2 %), C18:0 (11.0 %), and C18:1 ω7c/C18:1 ω6c (10.9 %) when grown on trypticase soy agar. The major polar lipids were diphosphatidylglycerol, phosphatidylglycerol, two unknown glycolipids (GL1, GL2), and two unknown phospholipid (PL1, PL2). The predominant ubiquinone was Q-10, and the genome DNA G+C content was 61.7 mol %. Phylogenetic analysis based on the 16S rRNA gene sequences indicated that strain J5-3T belonged to the family Hyphomicrobiaceae in Alphaproteobacteria. It shared the 16S rRNA gene sequence similarities of 93.8–96.1 % with the genus Devosia, 94.5–94.8 % with the genus Pelagibacterium, and <92.0 % with all the other type strains in family Hyphomicrobiaceae. It can be distinguished from the closest phylogenetic neighbors based on several phenotypic and genotypic features, including α-galactosidase activity, tetracycline susceptibility, major fatty acid composition, polar lipid profile, DNA gyrase B subunit (gyrB) gene sequence, and random-amplified polymorphic DNA profile. Therefore, we consider strain J5-3T to represent a novel species of a novel genus within the family Hyphomicrobiaceae, for which the name Paradevosia shaoguanensis gen. nov., sp. nov. is proposed. A strain 5-1-2T was isolated from a root nodule of Hedysarum multijugum collected from Zhangye city, Gansu province, north-west China. Phylogenetic analysis based on the 16S rRNA gene sequence and other housekeeping genes (recA and atpD) indicated that the strain represents a novel species in the genus Rhizobium close to the strain Rhizobium subbaraonis JC85T with similarities of 98.27, 88.92 and 89.62%, respectively. Strain 5-1-2T contained Q-10 as the predominant ubiquinone. Our results showed that the major fatty acids were feature 8 (C18:1 ω7c and/or C18:1 ω6c; 38.90%). In addition, the DNA–DNA hybridizations with the type strains R. subbaraonis JC85T and Rhizobium halophytocola YC6881T were 39.2 ± 2.1 and 44.3 ± 1.9, respectively. Therefore, a novel species Rhizobium hedysari sp. nov. is proposed, and 5-1-2T (=CGMCC1.15677T = NBRC112532T) is designated as the type strain.  相似文献

A novel pale pink-coloured, strictly aerobic, Gram-stain negative bacterial strain, designated strain KER25-12T, was isolated from a laboratory air-conditioning system in South Korea. Cells were observed to be non-motile cocci showing positive catalase and oxidase reactions. Strain KER25-12T was found to grow at 10–30 °C (optimum, 25–30 °C), at pH 4.0–9.0 (optimum, pH 6.0–7.0) and in the presence of 0–2% (w/v) NaCl (optimum, 0%). Ubiquinone-10 and spermidine were detected as the sole respiratory quinone and the predominant polyamine, respectively. The major fatty acids were identified as summed feature 8 (comprising C18:1 ω7c and/or C18:1 ω6c), summed feature 3 (comprising C16:1 ω7c and/or C16:1 ω6c), C16:0 and C18:0. The genomic DNA G+C content of strain KER25-12T was determined to be 70.0 mol%. The major polar lipids were identified as diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine and an unidentified aminolipid. Phylogenetic analysis based on 16S rRNA gene sequence comparison revealed that strain KER25-12T belongs to the genus Roseomonas and shows high sequence similarity to Roseomonas aerilata 5420S-30T (98.57%), Roseomonas pecuniae N75T (97.44%) and Roseomonas vinacea CPCC 100056T (97.40%). Based on the morphological, physiological, chemotaxonomic and phylogenetic features, strain KER25-12T is concluded to represent a novel species of the genus Roseomonas, for which the name Roseomonas aeriglobus sp. nov. is proposed. A Gram-stain-negative, strictly aerobic, non-motile and rod-shaped bacterial strain, designated T5T, was isolated from the Chishui River in Maotai town, Guizhou Province, Southwest of China. Strain T5T was found to grow optimally at pH 9.0 and 25 °C. The 16S rRNA gene sequence analysis indicated that strain T5T belongs to the family Sphingomonadaceae within the phylum Proteobacteria; the strain T5T clustered with the type strains of Sphingopyxis contaminans, Sphingorhabdus wooponensis and Sphingorhabdus rigui, with which it exhibits 16S rRNA gene sequence similarity values of 96.2–96.9%. The DNA G+C content was 58.5 mol%. The major respiratory quinone was Q-10 and the major polar lipid was phosphatidylethanolamine. The major polyamine was homospermidine and the major fatty acids were C18:1 ω7c (37.5%) and C16:1 ω7c (30.1%). On the basis of phylogenetic, phenotypic and genetic data, strain T5T represents a novel species of the genus Sphingorhabdus, for which the name Sphingorhabdus buctiana sp. nov. is proposed. The type strain is T5T (= CGMCC 1.12929T = JCM 30114T). It is also proposed that Sphingopyxis contaminans should be reclassified as a member of the genus Sphingorhabdus.  相似文献

A Gram-stain negative, aerobic, short rod-shaped, motile by flagella bacterial strain (THG-N2.35T), was isolated from Pu’er tea. Growth occurred at 10–40 °C (optimum 28 °C), at pH 4–7 (optimum 7) and at 0–5% NaCl (optimum 1%). Based on 16S rRNA gene sequence analysis, the near phylogenetic neighbours of strain THG-N2.35T were identified as Paracoccus hibisci KACC 18632T (99.0%), Paracoccus tibetensis CGMCC 1.8925T (98.7%), Paracoccus beibuensis CGMCC 1.7295T (98.2%), Paracoccus aestuarii KCTC 22049T (98.2%), Paracoccus rhizosphaerae LMG 26205T (98.1%), Paracoccus zeaxanthinifaciens ATCC 21588T (97.1%), Paracoccus marcusii DSM 11574T (97.0%). Levels of similarity between strain THG-N2.35T and other Paracoccus species were lower than 97.0%. DNA-DNA hybridization values between strain THG-N2.35T and P. hibisci KACC 18632T, P. tibetensis CGMCC 1.8925T, P. beibuensis CGMCC 1.7295T, P. aestuarii KCTC 22049T, P. rhizosphaerae LMG 26205T, P. zeaxanthinifaciens ATCC 21588T, P.marcusii DSM 11574T were 47.5% (42.3%, reciprocal analysis), 36.1% (32.3%), 24.7% (22.1%), 19.2% (16.3%), 11.3% (8.8%), 11.1% (10.8%), 6.1% (5.8%), respectively. The DNA G+C content of strain THG-N2.35T was 62.3 mol%. The polar lipids were diphosphatidylglycerol, phosphatidyl-N-methylethanolamine, phosphatidylethanolamine, phosphatidylglycerol and phosphatidylcholine. The quinone was ubiquinone-10 (Q-10). The major fatty acids were C10:0 3OH, C16:0, C18:0 and C18:1 ω7?. On the basis of the phylogenetic analysis, chemotaxonomic data, physiological characteristics and DNA-DNA hybridization data, strain THG-N2.35T represent a novel species of the genus Paracoccus, for which the name Paracoccus pueri sp. nov. is proposed. Experimental warming of forest ecosystems typically stimulates soil respiration (CO2 efflux), but most warming experiments have been conducted in northern latitudes (>?40°N) with relatively young soils. We quantified the influence of experimental warming on soil respiration (RT) in two adjacent forest habitats—a mature, closed canopy forest and a gap where trees were manually removed— on highly-weathered Ultisols of the southeastern U.S. (33°N). Using temperature variation, both natural and induced by experimental warming, we also quantified the temperature sensitivity of RT, defined as the activation energy, EA in the Arrhenius equation. Experimental warming (either + 3 °C or + 5 °C above ambient) did not significantly increase soil respiration rate or cumulative CO2 loss over the 3 years of the experiment, and did not influence the temperature sensitivity of soil respiration, once the influence of natural temperature variation was taken into consideration. Despite the absence of an experimental warming effect, we observed that EA varied on monthly time scales, and varied differently in each habitat. Soil moisture and habitat also influenced RT, but the effects were not consistent, and varied by month. Our results suggest that although RT does depend on temperature, the sensitivity of RT to temperature variation is influenced primarily by factors like microclimate and plant phenology that can change on relatively short (<?monthly) time scales. Thus, using the temperature sensitivity of RT to predict future CO2 losses due to warming is only reasonable if monthly variation in EA is incorporated into models for lower-latitude subtropical ecosystems with highly weathered soils, such as those in this study. Finally, our results suggest that higher temperatures may not enhance RT in highly-weathered, C-poor soils to the extent that has been reported in prior studies of high-latitude soils, which may constrain ecosystem-atmosphere carbon exchanges and feedbacks to the climate system.  相似文献

A Gram-stain negative, aerobic, motile by flagella, rod-shaped strain (THG-T16T) was isolated from rhizosphere of Hibiscus syriacus. Growth occurred at 10–40 °C (optimum 28–30 °C), at pH 6.0–8.0 (optimum 7.0) and at 0–1.0% NaCl (optimum 0%). Based on 16S rRNA gene sequence analysis, the near phylogenetic neighbours of strain THG-T16T were identified as Nibribacter koreensis KACC 16450T (98.6%), Rufibacter roseus KCTC 42217T (94.7%), Rufibacter immobilis CCTCC AB 2013351T (94.5%) and Rufibacter tibetensis CCTCC AB 208084T (94.4%). The DNA G+C content of strain THG-T16T was determined to be 46.7 mol%. DNA–DNA hybridization values between strain THG-T16T and N. koreensis KACC 16450T, R. roseus KCTC 42217T, R. immobilis CCTCC AB 2013351T, R.tibetensis CCTCC AB 208084T were 33.5?±?0.5% (31.7?±?0.7% reciprocal analysis), 28.1?±?0.2% (25.2?±?0.2%), 17.1?±?0.9% (10.2?±?0.6%) and 8.1?±?0.3% (5.2?±?0.1%). The polar lipids were identified as phosphatidylethanolamine, two unidentified aminophospholipids, an unidentified aminolipid and three unidentified lipids. The quinone was identified as MK-7 and the polyamine as sym-homospermidine. The major fatty acids were identified as C16:1 ω5c, C17:1 ω6c, iso-C15:0, summed feature 3 (C16:1 ω7c and/or C16:1 ω6c) and summed feature 4 (iso-C17:1 I and/or anteiso-C17:1 B). On the basis of the phylogenetic analysis, chemotaxonomic data, physiological characteristics, and DNA–DNA hybridization data, strain THG-T16T represents a novel species of the genus Nibribacter, for which the name Nibribacter flagellatus sp. nov. is proposed. A Gram-stain negative, rod-shaped, non-motile, strictly aerobic bacterium HK-28T was isolated from a mangrove sediment sample in Haikou city, Hainan Province, China. Strain HK-28T was able to grow at 10–45 °C (optimum 25–30 °C), pH 5.0–8.5 (optimum 6.0–7.0) and 0.5–12.0% (w/v) NaCl (optimum 1.0–3.0%, w/v). The major cellular fatty acids were C16:0, Summed Feature 8 (C18:1 ω7c and/or C18:1 ω6c), Summed Feature 3 (C16:1 ω7c and/or C16:1 ω6c), C17:0, C12:0 3-OH and C17:1ω8c. Ubiquinone-8 (Q-8) was the predominant respiratory quinone. The polar lipids consisted of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, two unidentified aminophospholipids, four unidentified phospholipids, two unidentified glycolipid, an unidentified glycophospholipid, an unidentified aminolipid and an unidentified lipid. The DNA G+C content was 50.2 mol%. Accoroding to 16S rRNA gene sequence similarities, strain HK-28T shared 97.1 and 96.7% sequence similarities to the validly named species Gallaecimonas xiamenensis MCCC 1A01354T and Gallaecimonas pentaromativorans MCCC 1A06435T, respectively, and shared lower sequence similarities (<?92.0%) to all other genera. Phylogenetic analysis showed strain HK-28T was clustered with G. pentaromativorans MCCC 1A06435T and G. xiamenensis MCCC 1A01354T. Strain HK-28T showed low DNA–DNA relatedness with G. xiamenensis MCCC 1A01354T (28.3?±?1.5%) and G. pentaromativorans MCCC 1A06435T (25.2?±?2.4%). On the basis of phenotypic, chemotaxonomic and genotypic characteristics, strain HK-28T is considered to represent a novel species in the genus Gallaecimonas, for which the name Gallaecimonas mangrovi sp. nov. is proposed. An endochitinase gene ‘ech42’ from the biocontrol fungus ‘Trichoderma virens’ was introduced to Brassica juncea (L). Czern and Coss via Agrobaterium tumefaciens mediated genetic transformation method. Integration and expression of the ‘ech42’ gene in transgenic lines were confirmed by PCR, RT-PCR and Southern hybridization. Transgenic lines (T1) showed expected 3:1 Mendelian segregation ratio when segregation analysis for inheritance of transgene ‘hpt’ was carried out. Fluorimetric analysis of transgenic lines (T0 and T1) showed 7 fold higher endochitinase activity than the non-transformed plant. Fluorimetric zymogram showed presence of endochitinase (42 kDa) in crude protein extract of transgenic lines. In detached leaf bioassay with fungi Alternaria brassicae and Alternaria brassicicola, transgenic lines (T0 and T1) showed delayed onset of lesions as well as 30–73 % reduction in infected leaf area compared to non-transformed plant.  相似文献

To investigate the symbiotic roles of the gut microbiota in the fungus-growing termite Macrotermes barneyi, a novel strain with chitinolytic and cellulolytic activity, designated strain an-chi-1T, was isolated from the hindgut of M. barneyi. Strain an-chi-1T grows optimally at 28–30 °C, pH 8.0 in PYG medium. On the basis of 16S rRNA gene sequence analysis, this isolate belongs to the genus Cellulomonas with high sequence similarity to Cellulomonas iranensis (99.4%), followed by Cellulomonas flavigena (98.4%), Cellulomonas phragmiteti (97.4%), Cellulomonas oligotrophica (97.2%) and Cellulomonas terrae (97.0%). The DNA–DNA relatedness between an-chi-1T and the type strains of C. iranensis and C. flavigena DSM20109T are 35.4% and 23.7%, respectively. The major cellular fatty acids are anteiso-C15:0 and C14:0. The polar lipid profile consists of diphosphatidylglycerol, phosphatidylinositol mannosides, phosphatidylinositol dimannosides and one unidentified phospholipid. The cell-wall sugar is ribose. The peptidoglycan contains glutamic acid, aspartic acid and alanine. The DNA G+C content is 67.3 mol%. Based on its distinctive phenotypic, phylogenetic, and chemotaxonomic characteristics, an-chi-1T represents a novel species of the genus Cellulomonas, for which the name Cellulomonas macrotermitis sp. nov. is proposed. Strain NHI-8T was isolated from a forest soil sample, collected in South Korea, by using a modified culture method. Comparative analysis of its nearly full-length 16S rRNA gene sequence showed that strain NHI-8T belongs to the genus Mesorhizobium and to be closely related to Mesorhizobium chacoense PR5T (97.32 %). The levels of DNA–DNA relatedness between strain NHI-8T and reference type strains of the genus Mesorhizobium were 32.28–53.65 %. SDS-PAGE of total soluble proteins and the sequences of the housekeeping genes recA, glnII, and atpD were also used to support the clade grouping in rhizobia. The new strain contained summed feature 8 (57.0 %), cyclo-C19:0ω8c (17.3 %), and C18:0 (11.0 %) as the major fatty acids, as in genus Mesorhizobium. The strain contained cardiolipin, phosphatidylglycerol, ornithine-containing lipid, phosphatidylethanolamine, phosphatidyl-N-dimethylethanolamine, and phosphatidylcholine. Morphological and physiological analyses were performed to compare the characteristics of our strain with those of the reference type strains. Based on the results, strain NHI-8T was determined to represent a novel member of the genus Mesorhizobium, and the name Mesorhizobium soli is proposed.
