Winter wheat (Triticum aestivum L. cv. Hereward) plants were grown for 35 d either at 350 μ mol mol–1 CO2 or at 650 μ mol mol–1 CO2. Lipid synthesis was studied in these plants by incubating the 5th leaf on the main stem with [1–14C]acetate. Increased CO2 concentrations did not significantly affect the total incorporation of radiolabel into lipids of whole leaf tissue, but altered the distribution for individual lipid classes. Most noticeable amongst acyl lipids was the reduction in labelling of diacylglycerol and a corresponding increase in the proportion of phosphatidylcholine labelling. In the basal regions, there were similar changes and, in addition, phosphatidylglycerol labelling was particularly increased following growth in an enriched CO2 atmosphere. The stimulation of labelling of the mitochondrial-specific lipid, diphosphatidylglycerol, prompted an examination of the mitochondrial population in wheat plants. Mitochondria were localized in intact wheat sections by immunolabelling for the mitochondrial-specific chaperonin probe. Growth in elevated CO2 doubled the number of mitochondria compared to growth in ambient CO2. Fatty acid labelling was also significantly influenced following growth at elevated CO2 concentrations. Most noticeable were the changes in 16C:18C ratios for the membrane lipids, phosphatidylcholine, phosphatidylglycerol and monogalactosyldiacylglycerol. These data imply a change in the apportioning of newly synthesized fatty acids between the 'eukaryotic' and 'prokaryotic' pathways of metabolism under elevated CO2.  相似文献

Plants of garden pea ( Pisum sativum L.) were exposed to charcoal-filtered air with or without addition of 65 ± 5 l−1 ozone. Plants were harvested daily for 9 days and lipids were extracted from the second-oldest leaf. Visible injury of this leaf was evident from day 5 on, while the differences in lipids between ozone and control treatments were observed earlier. Ozone caused large decreases in the contents of monogalactosyldiacylglycerol (MGDG), digalactosyldiacylglycerol (DGDG) and sulfoquinovosyldiacylglycerol (SQDG), a slower decrease in the content of phosphatidylcholine (PC), but an increase in the content of phosphatidylethanolamine (PE) per leaf area, compared with exposure to charcoal-filtered air. The content of phosphatidylglycerol (PG) was unaffected by ozone. Compared with charcoal-filtered air, fumigation with ozone resulted in a decrease in the proportion of linolenic acid (18:3) of the total lipid extract, with a concomitant increase in the proportion of linoleic acid (18:2). For individual lipids, ozone caused a similar pattern of decreased 18:3 and increased 18:2 in MGDG, SQDG, PC and PE, while the fatty acid composition of DGDG was unaffected. In PG, ozone decreased the proportions of 18:3 and trans -Δ3-decenoic acid (16:1trans), balanced by increased proportions of palmitic and oleic acids. The contents of chlorophylls and carotenoids were unaffected by ozone. Our results show that moderately elevated levels of ozone cause significant changes in the polar lipid composition of garden pea leaves and in the level of unsaturation of the lipid acyl groups and, furthermore, that ozone has different effects, which could be direct or indirect, on chloroplast lipids (MGDG, DGDG, SQDG, PG acylated with 16:1trans) and cytosolic membrane lipids.  相似文献

In crude extracts of the unicellular green alga Chlorella kessleri Fott et Novákóva grown in red light the activity of the glycolytic enzyme phosphofructokinase (PFK, EC 2.7.1.11) is about 40% higher compared to white light conditions giving the same dry matter production. Application of cycloheximide and density labelling with D2O indicate that this increase depends on the de novo synthesis of the enzyme: Twelve h of illumination at a fluence rate of 7 × 1018 quanta m−2 s−1 (11.6 μmol m−2 s−1) suffice to saturate the effect. In autotrophically grown algae maximal increase in enzyme saturate the effect. In autotrophically grown algae maximal increase in enzyme activity is reached in light of 680 nm, while in 3-(3,4-dichlorophenyl)-1,1-dimethyl urea (DCMU)-poisoned, glucose-fed cells, light of wavelengths around 727 nm is most effective. Involvement of a phytochrome-like photoreceptor is discussed.  相似文献

The abundance and cellular location of Fe-containing superoxide dismutase (Fe-SOD) in trichomes of Nodularia , Aphanizomenon and Anabaena collected from various depths in the Baltic Sea, and in trichomes of a cultured Nodularia strain, BC Nod-9427, isolated from the Baltic Sea, was examined by immunogold labelling. For trichomes collected from natural populations the areal concentration of Fe-SOD labelling decreased with depth: trichomes collected from surface accumulations had between 8 and 11 gold particles μm−2 whereas trichomes collected from a depth of 18 m were unlabelled. When trichomes collected from a depth of 10 m (mean areal labelling density 0·5 gold particles μm−2) were exposed to the higher irradiances present at 1 m, the areal concentration of Fe-SOD increased to 3·5–4 gold particles μm−2 within 4 h. When cultures of Nodularia strain BC Nod-9427, adapted to low light (10 μmol m−2 s−1), were transferred to an incident irradiance of 1350 μmol m−2 s−1, a doubling of the areal concentration of Fe-SOD gold label was observed within 1 h. Addition of 3-(3,4-dichlorophenyl)-1,1'-dimethylurea (DCMU) to cultures immediately before their transfer to increased illumination resulted in a decrease in areal Fe-SOD concentrations whereas addition of CdCl2 caused an increase over and above that induced by the elevated irradiance. These results suggest that Baltic Sea cyanobacteria are able to modulate their Fe-SOD content but that this might be in response to oxidative stress rather than to light per se .  相似文献

Effects of canopy shade on the lipid composition of soybean leaves   总被引:1,自引:0,他引:1  
The effect of canopy shade on leaf lipid composition was examined in soybeans ( Glycine max cv. Young) grown under field conditions. Expanding leaves were tagged at 50, 58 and 65 days after planting (DAP) in plots with either a high (10 plants m−1 row) or low (1 plant m−1 row) plant density. At 92 DAP, light conditions ranged from a pho-tosynthetic photon flux density (PPFD) of 87% of full sun with a far-red/red (735 nm/645 nm) ratio of 0.9 at upper canopy leaves to extreme shade where the PPFD was 10% of full sun with a far-red/red ratio greater than 6. Highly shaded leaves in the high plant density treatment accumulated triacylglycerol (TG) up to 25% of total leaf lipid, a 2.4-fold increase in TG on a chlorophyll basis compared to leaves in the upper canopy. Although total polar lipid content was reduced up to 50% in shaded leaves, shade had little affect on the lipid content or composition of thylakoid membranes. Shade did not affect leaf chlorophyll content. Therefore, the changes in leaf lipid composition were not related to senescence. These findings suggest that conditions of low irradiance and/or a high FR/R ratio cause a shift in carbon metabolism toward the accumulation of TG, a storage lipid. Eighteen-carbon fatty acid desaturation was also affected in highly shaded leaves where a reduction in linolenic acid (18:3) content was accompanied by a proportional increase in oleic (18:1) and linoleic (18:2) acids.  相似文献

Three 9-day-old cultivars of Hordeum vulgare L. (Barberousse, Gerbel and Panda) were exposed to low levels of SO2 fumigation (40 ± 5 and 117 ± 20 ppb). After 48 days of treatment the seedlings were harvested for lipid analysis. In comparison to the control (plants exposed to charcoal-filtered air), the total lipid content of fumigated seedlings declined at 40 ppb SO2 and even more so at 117 ppb in all three cultivars. A large reduction in diacylglycerols. polar lipids and free sterols was also observed after fumigations at both SO2 concentrations, whereas the treatments resulted in an increase in triacylglycerols and free fatty acids. The percentage composition of total fatty acids and that of each lipid class were changed by the fumigations. resulting in an increase in the degree of unsaturation. No changes in the percentage composition of sterols occurred in the fumigated leaves. These results suggest that even if SO2 may not directly oxidize unsaturated fatty acids at the low concentrations used here (which do not cause visible injury). it may alter lipid metabolism. This alteration. which was particularly evident in the polar lipids and sterols, could affect the functions associated with membrane stabilization, in which lipids plus sterols play a key part.  相似文献

以模式藻株莱茵衣藻(Chlamydomonas reinhardtii)为材料, 基于液质联用技术对其极性甘油酯组进行定性定量分析。通过综合利用UPLC-ESI-Q-Trap/MS的一级质谱扫描(中性丢失或母离子扫描)及UPLC-ESI-Orbitrap/MS2的二级碎片信息扫描, 共鉴定出109种极性甘油酯分子; 再通过外标法利用UPLC-ESI-Q-Trap/MS在多级反应监测模式下对各分子进行靶向定量分析。结果表明, 莱茵衣藻的极性脂以糖脂MGDG、DGDG及甜菜碱脂DGTS为主, 所有极性脂的分子组成表明, DGDG、SQDG、DGTS及PI是C18脂肪酸的去饱和载体。该研究利用液质联用技术建立了莱茵衣藻极性甘油酯组的结构图谱及定量分析技术平台, 为微藻极性脂生物学功能及脂质代谢研究奠定了基础。  相似文献

Abstract— Subsynaptosomal localization of stimulation of phospholipid labelling by cholinergic agents was investigated. Synaptosomes prepared from guinea-pig cortex were incubated with [32P]orthophosphate in the presence or absence of 10−3 m carbamylcholine. Following incubation and osmotic shock, lysed synaptosomes were subjected to density gradient fractionation. Subsynaptosomal fractions were examined by electron microscopy and analysed for enzyme activities and 32P-labelled lipids.
In the absence of carbamylcholine, labelled phosphatidate and phosphatidylinositol were recovered in layers and interfaces A, B, C and D formed over 0.9, 1.1, 1.2 and 1.3 m sucrose, with highest amounts of label in fractions C and D for both lipids. Carbamylcholine induced the greatest increment in these two labelled lipids in fractions A and B. This distribution correlated with the presence of acetylcholinesterase activity and membrane ghosts. No correlation was found among the four fractions between the induced increase in labelling and succinic dehydrogenase activity or with the abundance of mitochondria, synaptic vesicles, or cytoplasmic fragments identified by electron microscopy. In contrast with the increases seen in phosphatidylinositol and phosphatidate labelling, carbamylcholine caused a decrease in 32P-labelling of the polyphosphoinositides, and this effect was seen primarily in the heavier subsynaptosomal fractions, C and D.  相似文献

Seeds of Salicornia europaea L. were analyzed for their nutrient reserves. The content of potassium and sodium was 216 and 39 mmol (kg dry seeds)-1, respectively. Calcium and magnesium accounted for 30 and 138 mmol (kg dry seeds)-1, respectively. Whereas most of the alkali metals were water soluble, the alkaline earth metals were mostly acid soluble. The acid-soluble calcium plus magnesium corresponded well with the acid-soluble phosphate. Chloride was accumulated to a level equivalent to that of sodium. Carbonate was present at a concentration of 9 mmol (kg dry seeds)-1. Carbohydrates accounted for 93 g (kg dry seeds)-1, nearly half of which was derived from sucrose. Fructose and glucose were present only in traces. Total nitrogen was determined to be 55 g (kg dry seeds)-1, 16% of which was diethylether soluble. The remaining nitrogen was separated into 39 g (kg dry seeds)-1 ethanol-insoluble and 8 g (kg dry seeds)-1 ethanol-soluble nitrogen. About 10% of the ethanol-soluble nitrogen were derived from amino acids. Total lipid content was about 280 g (kg dry seeds)-1. The alcoholic component of the storage lipids was glycerol and the glycerides were calculated from gas chromatography to be 66% of the total lipids. About 90% of the fatty acids consisted of unsaturated acids, linoleic and oleic acid, the majority (77%) of which was linoleic acid.  相似文献

Plasma total lipids were significantly higher in 17β-oestradiol(E2)-treated immature rainbow trout Oncorhynchus mykiss at week 4 after implantation, due to increases in polar and neutral lipids. The lipid classes responding were phosphatidylcholine, phosphatidylinositol, phosphatidylethanolamine, sterols and sterol esters, in a proportion that approximately reflected the increase in plasma vitellogenin (VtG) levels as measured by a non-competitive enzyme-linked immunosorbent assay (ELISA). Plasma non-esterified fatty acids and triacylglycerol were not affected by E2 treatment. Plasma growth hormone GH levels were increased, and plasma somatostatin-14 (SRIF) levels decreased in E2-treated fish, responses which could be secondary to elevated plasma lipid (VtG) content, although a direct E2 action on somatotroph function is possible. Plasma T4 concentrations were not affected by E2 treatment, but plasma T3 concentrations were significantly lower than in controls 1 week after implantation when plasma E2 concentrations were the highest; this is in support of the hypothesis that E2 has a suppressive action on T3 production.  相似文献

Abstract— A novel phospholipase A1, present in human cerebral cortex and active at pH 9.25 upon ultrasonicated phosphatidylethanolamine is described. It has been purified 39-fold from acetone-dried powders of cortex by ammonium sulphate fractionation at 25% saturation, followed by Sephadex G150 gel filtration. A molecular weight of approx 500,000 has been found by Sepharose 6B gel filtration. The enzyme was slightly stimulated by 2 mM-Ca2+ and was inhibited by 1 mM-Hg2+ and by all detergents tested. The enzyme hydrolysed phosphatidylcholine at 18% of the rate for phosphatidylethanolamine, lysophospholipids to a lesser degree and neutral lipids not at all. There appeared to be a preference for fluid phosphatidylethanolamine substrates. The similarities of the enzyme to other phospholipases A1 are discussed.  相似文献

Cyanidium caldarium (Tilden) Geitler, a non-vacuolate unicellular alga, resuspended in medium flushed with air enriched with 5% CO2, assimilated NH4+ at high rates both in the light and in the dark. The assimilation of NO3, by contrast, was inhibited by 63% in the dark. In cell suspensions flushed with CO2-free air, NH4+ assimilation decreased with time both in the light and in the dark and ceased almost completely after 90 min. The addition of CO2 completely restored the capacity of the alga to assimilate NH4+. NO3 assimilation, by contrast, was 33% higher in the absence of CO2 and was linear with time. It is suggested that NO3 and NH4+ metabolism in C. caldarium are differently controlled in response to the light and carbon conditions of the cell.  相似文献

SUMMARY. The oxygen consumption of shrimps ranging from 1 to 30 mg dry mass was determined at 18, 24 and 30°C using a continuous flow recording respirometer based upon a Clark-type oxygen electrode. Respiration (ascribed to routine metabolism) is described by the power curve: R = a Mb , ( R =μg O2 h−1, M = mg dry mass), which gives values of a = 1.632, 2.564 and 4.181, and b = 0.800, 0.898, and 0.793, at 18, 24 and 30°C respectively. The single expression, R = 0.008 T 1.829 M 0.830 provides a reasonable prediction of respiration as a combined function of shrimp size ( M ) and temperature (T, °C). Using an energy equivalent of 14.14 J mg O2−1 estimates of the energy requirements ( E , J h−1 10−3) of routine metabolism are given by the expression: E = 0.115 T 1.829 M 0.830.
Variability in oxygen consumption values between individuals is discussed and the observations on C. nilotica are compared with other crustacean studies.  相似文献

Abstract: The conjugative transposon Tn 919 was introduced at high frequency to L. lactis subsp. lactis biovar. diacetylactis 18-16 and transconjugants were screened for mutations in two chromosomally located genotypes; citrate metabolism and maltose utilization. A citrate negative mutant, lacking citritase activity, was isolated at a frequency of 1.18 × 10−4. The mutant, 18-16C5, contained a single copy of Tn 919 in a chromosomal location. A junction fragment of Tn 919 ::18-16C5 chromosomal DNA was cloned in Escherichia coli . Mutations in maltose metabolism were detected at a frequency of 4.0 × 10−4. No mutants were detected when Tn 919 was not introduced. Reversion to a Mal+ phenotype occurred at high frequency, but was not due to Tn 919 transposition.  相似文献

Pyoverdine (PvdI) is the major siderophore secreted by Pseudomonas aeruginosa PAOI in order to get access to iron. After being loaded with iron in the extracellular medium, PvdI is transported across the bacterial outer membrane by the transporter, FpvAI. We used the spectral properties of PvdI to show that in addition to Fe3+, this siderophore also chelates, but with lower efficiencies, all the 16 metals used in our screening. Afterwards, FpvAI at the cell surface binds Ag+, Al3+, Cd2+, Co2+, Cu2+, Fe3+, Ga3+, Hg2+, Mn2+, Ni2+ or Zn2+ in complex with PvdI. We used Inductively Coupled Plasma-Atomic Emission Spectrometry to monitor metal uptake in P. aeruginosa : TonB-dependent uptake, in the presence of PvdI, was only efficient for Fe3+. Cu2+, Ga3+, Mn2+ and Ni2+ were also transported into the cell but with lower uptake rates. The presence of Al3+, Cu2+, Ga3+, Mn2+, Ni2+ and Zn2+ in the extracellular medium induced PvdI production in P. aeruginosa . All these data allow a better understanding of the behaviour of the PvdI uptake pathway in the presence of metals other than iron: FpvAI at the cell surface has broad metal specificity at the binding stage and it is highly selective for Fe3+ only during the uptake process.  相似文献

The amount and distribution of proteins of the light-harvesting complex associated with photosystem II (PS II) were investigated using immunogold labelling of chloroplasts of wheat ( Triticum aestivum L. cv. Walde). The seedlings were grown in weak red light (16 mW m−2) after imbibition of grains with SAN-9789 (Norflurazon, 0.028 to 28 mg I−1). Chloroplasts of these plants exhibited thylakoids with different degrees of stacking. Thylakoids of untreated plants grown in a greenhouse had most gold particles per unit membrane length in both appressed and non-appressed regions compared to red light grown plants. The ratios of labelling between appressed and non-appressed membranes were fairly constant in red light- and greenhouse-grown plants. The labelling densities were 2.5–3 times higher in the appressed thylakoids compared to the non-appressed thylakoids. However, at a SAN concentration of 2.8 mg I−1 there was a sharp decrease in thylakoid appressions and in labelling density of both appressed and non-appressed membranes. The total amount of particles per chloroplast was also much lower as compared to that at lower SAN concentrations. Plants treated with the highest concentration of SAN (28 mg I−1) contained chloroplasts devoid of normal grana structures. In these plastids, the thylakoids were elongated and single. The labelling density in these membranes was ca 50% of that observed at 2.8 mg I−1. This paper thus supports earlier observations that proteins of the light-harvesting complex of PS II (LHC II) are mainly localized in the appressed regions of the grana membranes, and may be involved in the formation of grana.  相似文献

The fatty acid composition of the total lipids from two Conidiobolus species was studied by gas—liquid chromatography. The major fatty acids of C. lamprauges were palmitic acid (C16:0), oleic acid (C18:1), linolenic acid (C18:3), and arachidonic acid (C20:4). For C. eurymitus , myristic acid (C14:0), C16:0, and linoleic acid (C18:2) were the most abundant acids. The fatty acid composition of C. eurymitus was quite different from that of the Conidiobolus species as mentioned in other reports. The lipid composition of the total lipids of C. lamprauges and C. eurymitus was also studied by thinchrography on quartz rods. Triglycerides and phospholipids were the major components in the two Conidiobolus species.  相似文献

Abstract Pseudomonas fluorescens appeared to circumvent the challenge imposed by millimolar amounts of metals (5 mM Al3+, 5 mM Fe3+, 2 mM Ca2+, 1 mM Ga3+ and 3 mM Zn2+) by the formation of phosphatidylethanolamine. This lipid moiety constituted an important organic component of an insoluble gelatinous residue in which most of the test metals were immobilized at stationary phase of growth. Ultracentrifugation and dialysis experiments showed that the metals were associated with phosphatidylethanolamine from early stages of growth. Transmission electron microscopy revealed metal rich bodies in the cytoplasm prior to their secretion in the spent fluid. These results demonstrate a role of phosphatidylethanolamine in multiple-metal homeostasis.  相似文献

Lipid metabolism and the effect of Pb(2+) and Cu(2+) on this process was studied in the moss Rhytidiadelphus squarrosus collected from both a lead-contaminated and a non-contaminated site. Total radiolabelling of lipids from [1-(14)C]acetate was similar in both populations and Cu or Pb (1 microM, 10 microM) did not cause much alteration in acute exposure experiments. However, there were significant qualitative changes. Of the major labelled neutral lipid classes, samples from the lead-polluted site showed a decrease in labelling of triacylglycerols and an increase in wax esters. Acute lead exposure caused similar effects. Cu caused a decrease in the labelling of wax esters and an increase in diacylglycerols. These data suggest that heavy metals cause a change in carbon flux through the acylation reactions associated with the Kennedy pathway. R. squarrosus obtained from the Pb-contaminated site also showed changes in polar lipid labelling compared to the uncontaminated site. The labelling of phosphatidylcholine was more than halved and replaced by increased labelling of other zwitterionic lipids. The chloroplast glycerolipids were also increasingly labelled. Acute exposure to Pb, however, caused little alteration of labelling patterns within 24 h. R. squarrosus contains high levels of polyunsaturated fatty acids (PUFAs), but moss obtained from the Pb-polluted site had significantly less PUFAs containing three or more double bonds. Such samples, when incubated with [1-(14)C]acetate also showed decreased PUFA labelling. By contrast, acute exposure to Pb produced different results. These data provide a foundation for examining lipid metabolism in bryophytes and the effects of pollution in this important class of organism. The results also emphasize that acute and chronic exposure to heavy metals may produce different effects and that caution must be exercised in extrapolating data from one system to another.  相似文献
