We characterized the physical/chemical conditions and the algal and bacterial assemblages in ballast water from 62 ballast tanks aboard 28 ships operated by the U.S. Military Sealift Command and the Maritime Administration, sampled at 9 ports on the U.S. West Coast and 4 ports on the U.S. East Coast. The ballast tank waters had been held for 2–176 days, and 90% of the tanks had undergone ballast exchange with open ocean waters. Phytoplankton abundance was highly variable (grand mean for all tanks, 3.21 × 104 viable cells m−3; median, 7.9 × 103 cells m−3) and was unrelated to physical/chemical parameters, except for a positive relationship between centric diatom abundance and nitrate concentration. A total of 100 phytoplankton species were identified from the ballast tanks, including 23 potentially harmful taxa (e.g. Chaetoceros concavicornis, Dinophysis acuminata, Gambierdiscus toxicus, Heterosigma akashiwo, Karlodinium veneficum, Prorocentrum minimum, Pseudo-nitzschia multiseries). Assemblages were dominated by chain-forming diatoms and dinoflagellates, and viable organisms comprised about half of the total cells. Species richness was higher in ballast tanks with coastal water, and in tanks containing Atlantic or Pacific Ocean source waters rather than Indian Ocean water. Total and viable phytoplankton numbers decreased with age of water in the tanks. Diversity also generally decreased with water age, and tanks with ballast water age >33 days did not produce culturable phytoplankton. Abundance was significantly higher in tanks with recently added coastal water than in tanks without coastal sources, but highly variable in waters held less than 30 days. Bacterial abundance was significantly lower in ballast tanks with Atlantic than Pacific Ocean source water, but otherwise was surprisingly consistent among ballast tanks (overall mean across all tanks, 3.13 ± 1.27 × 1011 cells m−3; median, 2.79 × 1011 cells m−3) and was unrelated to vessel type, exchange status, age of water, environmental conditions measured, or phytoplankton abundance. At least one of four pathogenic eubacteria (Listeria monocytogenes, Escherichia coli, Mycobacterium spp., Pseudomonas aeruginosa) was detected in 48% of the ballast tanks, but toxigenic strains of Vibrio cholerae were not detected. For ships with tanks of similar ballasting history, the largest source of variation in phytoplankton and bacteria abundance was among ships; for ships with tanks of differing ballasting histories, and for all ships/tanks considered collectively, the largest source of variation was within ships. Significant differences in phytoplankton abundance, but not bacterial abundance, sometimes occurred between paired tanks with similar ballasting history; hence, for regulatory purposes phytoplankton abundance cannot be estimated from single tanks only. Most tanks (94%) had adequate records to determine the source locations and age of the ballast water and, as mentioned, 90% had had ballast exchange with open-ocean waters. Although additional data are needed from sediments that can accumulate at the bottom of ballast tanks, the data from this water-column study indicate that in general, U.S. Department of Defense (DoD) ships are well managed to minimize the risk for introduction of harmful microbiota. Nevertheless, abundances of viable phytoplankton with maximum dimension >50 μm exceeded proposed International Maritime Organization standards in 47% of the ballast tanks sampled. The data suggest that further treatment technologies and/or alternative management strategies will be necessary to enable DoD vessels to comply with proposed standards.  相似文献

The role of ballast water in the introduction of exotic species has recently received extensive attention. The aim of this study is to assess the importance of ballast water discharge as a vector for the introduction of exotic species into Hong Kong waters. Twelve ballast water samples were collected from 5 container ships entering Hong Kong between June1994 and October 1995. The ballast water originated from ports on both sides of the Pacific Ocean. At least 81 species from 8 animal phyla and 5 protist phyla were found. Most of the major marine taxonomic groups were represented and many planktonic larval stages were included. Species richness in the ballast tanks decreased with the age of ballast water. Copepoda was the most diverse and abundant taxonomic group. The density of calanoid and cyclopoid copepods decreased with the age of ballast water, but that of harpacticoid copepods did not change significantly with time. Bivalve, crustacean, polychaete and ascidian larvae from ballast water samples were observed to settle in laboratory culture tanks. The mussel Mytilopsis sallei which was introduced to Hong Kong in 1980, was one of the bivalves that settled readily. Results of this study indicate that ballast water can be a major source for the introduction of exotic species to Hong Kong waters. Quantification of eukaryotes, such as diatom within a water community is extremely difficult due to the different copy number of the measured gene. ITS sequence was considered to be useful in defining intra-specific or population-level differences for their fast evolving. To assess the possibility of exploring rDNA-ITS region in quantification analysis of environment samples, the fragment of 5.8S + ITS-2 was amplified from extracted DNA and sedimental cell samples derived from the same natural-simulated water community, respectively. Two clone libraries were built from the above DNA-derived (DD) and cell-derived (CD) amplifications. The operational taxonomic units (OTUs) of the libraries were calculated. In most cases, number of OTUs for CD dataset was smaller than DD dataset. For the DD and CD datasets, 112 sequences yielded 6 OTUs-96 and 4 OTUs-97 and were assigned to six and four species, respectively. Intraspecific variations existed in these cultivations. The sequences percentage in datasets did not match with the cell portions in mixed sample, which revealed that 5.8S + ITS-2 copies varied between different diatom species, and it was not suitable for quantitative analysis of diatom community structures in mixed sample. More researches are necessary to seek for proper gene to quantify diatoms in the environmental samples.  相似文献

The species composition of surface sediment diatom assemblages in 33 Galloway lakes, pH range c. 4.5 to 7.4, is related in a statistically significant manner to water acidity (or to factors closely associated with pH). Predictive models of summer mean pH, using simple regression equations of Index B (Renberg & Hellberg, 1982) with both Scandinavian and Galloway data sets, and multiple regression equations using diatom pH preference groups and individual species, are described and applied to fossil diatoms in six sediment cores. Although multiple regression of individual species gives the highest correlation coefficient (r2=0.87) in the modern data set this method is least appropriate for reconstructing pH values from fossil material where predictor species are often absent. Of the four methods examined here it is suggested that multiple regression of diatom preference groups is probably the most suitable for pH reconstructions from sediment cores.  相似文献

Diatoms have been long collected from the Southern Ocean but almost no data exist for epiphytic communities, despite their high ecological significance as an important food source in Antarctic coastal food chains. Here, we present a first growth form analysis of diatoms associated with rhodophyte hosts from Terra Nova Bay, Ross Sea, Antarctica. We performed this study to gather baseline information on the species composition of epiphytic diatom communities, determine the influence of some environmental variables on the diatom distribution patterns, and assess the caveats that must be taken into account in terms of sampling design. Macroalgal material was collected during the Italian Antarctic expeditions between 1990 and 2004. Epiphytic diatoms were studied by means of scanning electron microscopy. In terms of growth forms, there were no significant differences between the diatom communities on the different macroalgal host species. Motile (mainly small-celled Navicula perminuta and other Navicula spp.) and adnate (Cocconeis spp.) diatoms dominated the community throughout the study period. Many of the macroalgal blades examined were also covered by epiphytic animals (calcareous bryozoans, hydroids) over most of their surface, with a significant effect on the associated diatom community structure. Our findings suggest that the bio-physicochemical characteristics of each sampling site affected the epiphytic diatom communities more than the substrate type provided by the macroalgal host or the sampling depth.  相似文献

We assessed the feasibility of using herbarium specimens to validate reference conditions in the UK by comparing diatom community composition of river sites with both recent and historic diatom samples. The question of substrate specificity was addressed by comparing epilithon (stone-derived) and epiphyton (plant-derived) samples from a number of rivers. No significant differences were found between the Trophic Diatom Index (TDI), species richness, species diversity, and percentage of motile valves between paired diatom samples (epilithic and epiphytic) from contemporary samples. Significant differences were recorded between a number of indices derived from analysis of the historic diatom samples on plant material sampled pre-1930 compared with diatoms from stones collected post-1990 from the same river location. The TDI, mean species richness, and species diversity and percentage of motile valves and nutrient tolerant valves were all significantly greater in the contemporary samples (p ≤ 0.05). The percentage of nutrient sensitive valves was significantly lower in the contemporary samples (p ≤ 0.05).The relative abundance of Achnanthidium minutissimum and Cocconeis placentula var. lineata was significantly greater on the herbarium material compared to matched contemporary samples. Calculated values for the TDI (43 ± 3) expected at reference conditions were similar to the observed TDI values derived from herbarium material (44 ± 12) showing no significant deviation in ecological status.  相似文献

Benthic diatoms are important indicators of human impacts on streams. Epilithic diatoms are collected most often for bioassessments, but potentially important ecological information from other habitats could be missed. Within our study region, substrata ranged from 100% rock to 100% sandy silt at 61 sites, leading us to question the appropriateness of sampling solely epilithic surfaces in riffles. We compared two protocols for collecting diatoms: (1) epilithic habitat samples (EHS) within riffles and (2) multi-habitat samples (MHS), which proportionately included different habitats (e.g., riffles and pools) and substrata (e.g., sand, silt, and rock). Three streams were not compared because rocks were absent. Nonmetric multidimensional scaling indicated that diatom communities from EHS and MHS responded similarly to alkalinity and agricultural gradients, and Procrustes analysis showed that ordinations were similar (P < 0.01). Percent motile diatoms and metrics indicating high or low P and N from MHS showed stronger relationships with land use variables than those metrics from EHS. Bray-Curtis (BC) similarity between the protocols increased as diversity and richness decreased in MHS, as agricultural impacts increased, and as motile diatoms became more abundant in EHS, which likely indicated greater habitat homogeneity or an overriding effect of water chemistry. The two protocols classified 78% of sites concordantly as minimally impacted or impacted. Multi-habitat sampling is preferred where stream habitats vary greatly because it assessed land use impacts more effectively than EHS, while indicating water chemistry impacts as effectively. Multiple habitats also contained greater species diversity, which is important to documenting species distributions, and were always available for sampling, whereas epilithic habitats could be sparse or absent.  相似文献

We investigated epilithic diatoms in rivers draining to the Nyanza Bay in Lake Victoria (Kenya) with the aim of determining environmental gradients in the assemblages and testing the usefulness of diatom metrics from temperate regions. Spatial and temporal variations of assemblages were studied in 12 sites of three rivers. Kibos, Nyando, and Kisat rivers contained 224 diatom taxa collected on seven sampling occasions over 4 years. Species richness showed a marginal decrease downstream and was the lowest at sites with high conductivity and ammonia–nitrogen levels. Two-Way Indicator Species Analysis and Canonical Correspondence Analysis revealed two major groups of river sites. Conductivity, alkalinity, turbidity, dissolved oxygen, and silicate were the most important variables influencing species distribution. Ecological diatom metrics of temperate regions and the Specific Pollution sensitivity Index showed good relationships with environmental variables. Both diatom assemblages and averaged indicator values were strong in predicting sites of ecological deterioration and in differentiating an upstream site of better quality (drinking water supply of Kisumu), thereby confirming epilithic diatoms as suitable water quality indicators in equatorial rivers. The use of metrics initially designed for temperate rivers, however, appears less valuable in good quality tropical rivers because potential indicators are not considered.  相似文献

Diatom and dinoflagellate species that are not endemic to aregion can be inadvertently introduced when their resistantresting stages are discharged with the ballast-tank waters andsediments of bulk cargo vessels. A survey of 343 cargo vesselsentering 18 Australian ports showed that 65% of ships were carryingsignificant amounts of sediment on the bottom of their ballasttanks. All of these samples contained diatoms, including speciesthat are not endemic to Australian waters. Diatom resting spores,especially of Chaetoceros, were also detected. Dinoflagellateresting spores (cysts) were present in 50% of the sediment samples.Of the 53 cyst species identified, 20 (including Diplopelta,Diplopsalopsis, Gonyaulax, Polykrikos, Protoperidinium, Scrippsiellaand Zygabikodinium spp.) were successfully germinated to produceviable cultures. Such diversity of diatom and dinoflagellatespecies in ships' ballast water suggests that the apparent cosmopolitanismof many coastal phytoplankton species may be due partly to theglobal transport of seawater ballast. Of considerable concernwas the detection in 16 ships of cysts of the toxic dinoflagellatesAlexandrium catenella, Alexandrium tamarense and Gymnodiniumcatenatum. One single ballast tank was estimated to contain>300 million viable A.tamarense cysts, some of which weresuccessfully germinated in the laboratory to produce toxic cultures.These toxic dinoflagellate species, which can contaminate shellfishwith paralytic shellfish poisons, pose a serious threat to humanhealth and the aquaculture industry. Ballast-water quarantinemeasures recently introduced in Australia are discussed. Mid-oceanexchange of ballast water is only partially effective in removingdinoflagellate cysts which have settled to the bottom of ballasttanks. The present work indicates that the most effective measureto prevent the spreading of toxic dinoflagellate cysts via ships'ballast water would be to avoid taking on ballast water duringdinoflagellate blooms in the water column of the world's ports.  相似文献

Large epiphytic and epilithic diatom species hosting other diatoms were observed in several fresh- and brackish water sites in Southern California. The most commonly encountered hosts were species forming long filaments attached to rocks or macroalgae, Hydrosera whampoensis (Schwarz) Hendey, Melosira varians Agardh, Pleurosira laevis (Ehrenberg) Compere and Terpsinoe musica Ehrenberg. These large diatoms often had smaller diatoms attached, usually to the girdle bands and occasionally to the mucilage pads connecting the cells. For example, cells of T. musica were observed supporting growth of a diverse diatom assemblage composed of species of the genera Achnanthes , Achnanthidium , Amphora , Cocconeis and Tabularia; Synedra sp. was attached to M. varians and B. paxillifer; and Cocconeis placentula was seen on H. whampoensis. Thus, large epiphytic and epilithic diatoms seem to provide suitable sites for attachment of small epiphytic diatom species, and it appears that this phenomenon is more common than previously thought.  相似文献

Dinoflagellates were identified and counted from preserved verticalnet haul samples collected from a station grid centred on SouthGeorgia in austral summer 1981 –1982. All species werescarce in comparison with diatoms, the dominant large-celledmicroplankton. Dinophysis antarctica Balech was the most abundantand widespread species. Maximum dinoflagellate numbers werefound at stations forming a belt to the south and west of theisland. Whilst previous analysis of diatom community compositionhad revealed distinct species associations, no differences inspecies composition between this region and areas of low dinoflagellateabundance could be resolved. Since both heterotrophic and photoautotrophictaxa were found in the samples, it is difficult to identifyenvironmental features which could act on most of the dinoflagellatetaxa. The large-scale pattern of dinoflagellate distributionmay reflect the pattern of water circulation along the edgeof the South Georgia continental shelf.  相似文献

Diatoms are present in all types of water bodies and their species diversity is influenced greatly by environmental conditions. This means that diatom occurrence and abundances are suitable indicators of water quality. Furthermore, continuous screening of algal biodiversity can provide information about diversity changes in ecosystems. Thus, diatoms represent a desirable group for which to develop an easy to use, quick, efficient, and standardised organism identification tool to serve routine water quality assessments. Because conventional morphological identification of diatoms demands specialised in-depth knowledge, we have established standard laboratory procedures for DNA barcoding in diatoms. We (1) identified a short segment (about 400 bp) of the SSU (18S) rRNA gene which is applicable for the identification of diatom taxa, and (2) elaborated a routine protocol including standard primers for this group of microalgae. To test the universality of the primer binding sites and the discriminatory power of the proposed barcode region, 123 taxa, representing limnic diatom diversity, were included in the study and identified at species level. The effectiveness of the barcode was also scrutinised within a closely related species group, namely the Sellaphora pupula taxon complex and relatives.  相似文献

为探讨福建敖江口表层沉积硅藻空间分布特征,2019年7月对敖江口进行表层沉积硅藻的采样调查,并研究了硅藻与环境因子的关系。结果表明,从13个站点共检出硅藻114种,隶属于39属。硅藻丰度具有从河口向外海先减少后增加的变化特征。聚类分析表明,主要硅藻属种可划分为淡水种组合带和半咸水-咸水种组合带。典型对应分析(CCA)表明,表层海水温度和盐度是影响硅藻分布的主要环境变量。河口区盐度相对较低,主要分布淡水种双面曲壳藻、优美曲壳藻和颗粒沟链藻;外海区北部盐度相对较高,主要分布咸水种流水双菱藻;外海区南部盐度相对较低,主要分布淡水种优美曲壳藻和颗粒沟链藻;滩涂的沉积物较粗,主要分布优美曲壳藻。此外,河口区和外海区南部可能存在一定的水体污染。  相似文献

The sampling skills of three common European fish species (Barbus barbus, Chondrostoma nasus and Squalius cephalus) were tested to assess their potential as biomonitors of past changes in river water quality through the analysis of diatoms in fish guts. The study was performed on three rivers with different chemical and physical conditions. Comparison of similarity indices revealed low diatom assemblage resemblance between epilithic samples and samples collected from fish guts at the same locality. In contrast, a mixed-effect linear model identified significant differences between locations through comparison of diatom-based water quality indices. Among fish species, diatom indices calculated from gut samples of B. barbus and C. nasus were in most cases not significantly different from those from epilithic samples while those from S. cephalus were often significantly different. The results of the study demonstrate that diatom analysis of fish guts provides a clear distinction between eutrophic and hypereutrophic rivers. Finer nutrient variations within hypereutrophic conditions were also found to be significant for some diatom indices. In this context, choice of appropriate diatom water quality index proved to be crucial in maximizing the sensitivity of the method.  相似文献

The freshwater ultraoligotrophic Lake Labynkyr is located near the Pole of Cold in the northern hemisphere (Yakutia, Russia). The lake is covered by ice during 240 days a year. We undertook several expeditions to the lake during the ice and open water periods for sampling ice fouling, plankton and periphyton that were then analyzed by means of scanning electron microscopy. As a result, we identified a high biodiversity of diatoms—123 species and intraspecific taxa from 53 genera, among them 3 species were new for Russia and 26 taxa were new for the algal flora of Yakutia. The oligo- and xenosaprobionts and their variations dominate—71 taxa. 18 Species were evaluated as tolerant to cold oligotrophic waters, 12 occurred on the ice bottom, and 62 in the water column under ice (0–25 m). 104 taxa were found during the open water period, 70 taxa were identified in the periphyton. We showed the diatom flora of Lake Labynkyr to be unique compared with other lakes of Yakutia and to share taxa with the diatom flora of Lake Baikal. The diatoms being indicators of the global climate changes and ecological status of lakes, our data can be used as an evidence of such changes as well as to be useful studies of biogeography and history of formation of flora in Arctic and Subarctic waters.

The spatial and temporal distribution of planktonic, sediment-associated and epiphytic diatoms among 58 sites in Biscayne Bay, Florida was examined in order to identify diatom taxa indicative of different salinity and water quality conditions, geographic locations and habitat types. Assessments were made in contrasting wet and dry seasons in order to develop robust assessment models for salinity and water quality for this region. We found that diatom assemblages differed between nearshore and offshore locations, especially during the wet season when salinity and nutrient gradients were steepest. In the dry season, habitat structure was primary determinant of diatom assemblage composition. Among a suite of physicochemical variables, water depth and sediment total phosphorus (STP) were most strongly associated with diatom assemblage composition in the dry season, while salinity and water total phosphorus (TP) were more important in the wet season. We used indicator species analysis (ISA) to identify taxa that were most abundant and frequent at nearshore and offshore locations, in planktonic, epiphytic and benthic habitats and in contrasting salinity and water quality regimes. Because surface water concentrations of salts, total phosphorus, nitrogen (TN) and organic carbon (TOC) are partly controlled by water management in this region, diatom-based models were produced to infer these variables in modern and retrospective assessments of management-driven changes. Weighted averaging (WA) and weighted averaging partial least squares (WA-PLS) regressions produced reliable estimates of salinity, TP, TN and TOC from diatoms (r2 = 0.92, 0.77, 0.77 and 0.71, respectively). Because of their sensitivity to salinity, nutrient and TOC concentrations diatom assemblages should be useful in developing protective nutrient criteria for estuaries and coastal waters of Florida.  相似文献

We measured the presence, viability and potential toxicity of cyanobacteria in ships’ ballast tanks during three domestic voyages through the North American Great Lakes. Using molecular methods, the toxin-producing forms of Microcystis and Anabaena were monitored in ballast water after ships’ ballast tanks were filled at their first port of call, and at subsequent ports as ships transited the Great Lakes. Microcystis was detected in ballast water at intermediate and final ports of call in all three experiments, but the presence of Anabaena was more variable, suggesting low abundance or patchy distribution in ballast tanks. Both species were detected in ballast water up to 11 days old. Detection of the microcystin synthetase gene, mcyE, in ballast tanks indicated entrained cells were capable of producing microcystin, and further analyses of RNA indicated the toxin was being expressed by Microcystis, even after 11 days in dark transit. These data demonstrate within-basin transport and delivery of planktonic harmful algal bloom (HAB) species to distant ports in the world's largest freshwater reservoir, with potential implications for drinking water quality. These implications are discussed with respect to management of microbial invasions and the fate of introduced phytoplankton in their receiving environment.  相似文献

Following the European Water Framework Directive, this study aims to be the first step to (i) identify diatom type assemblages in unpolluted streams in NW Italy, and (ii) find which ecological factors explain most of the variation. To achieve this, we collected physical, chemical and benthic community data from four streams in NW Italy from 2001 to 2004, for a total of 72 samples. All sampling sites were between 200 m a.s.l. and 800 m a.s.l., but differed in the dominant lithological substrate, i.e. alluvial or siliceous. Relationships between diatom communities and environmental factors were examined using canonical correspondence analysis (CCA), while Indicator Species Analysis was used to define characterizing species and accompanying species of three environmental groups identified by CCA: (1) high water quality and medium saline content, (2) high water quality and low saline content, (3) poor water quality. The diatom assemblages of the three groups of sites differed significantly, as shown by the Multi-Response Permutation Procedure. There were strong correlations between diatoms and environmental factors, especially chlorides (also highly correlated with sulphates and carbonate hardness), nitrate concentration and conductivity. The group 1 assemblage was typical of the alluvial Alpine streams with medium saline content and was characterized by mostly oligosaprobic/β-mesosaprobic taxa such as Cymbella affinis, Diatoma ehrenbergii and Staurosira pinnata. The species assemblage found in the siliceous Alpine rivers with good water quality make them suitable reference sites for a benthic diatom community. It is well established that cyst-forming phytoplankton species are transported in ships' ballast tanks. However, there is increasing evidence that other phytoplankton species which do not encyst are also capable of surviving ballast transit. These species have alternative modes of nutrition (hetero- or mixotrophy) and/or are able to survive long-term darkness. In our studies of no-ballast-on-board vessels arriving in the Great Lakes, we tested for the presence of the harmful algal bloom species Aureococcus anophagefferens (brown tide) in residual (i.e., unpumpable) ballast water using methods based on the PCR. During 2001, the brown tide organism was detected in 7 of 18 ballast water tanks in commercial ships following transit from foreign ports. Furthermore, it was detected after 10 days of ballast tank confinement during a vessel transit in the Great Lakes, a significant result given the large disparity between the salinity tolerance for active growth of Aureococcus (>22 ppt) and the low salinity of the residual ballast water (~2 ppt). We also investigated the potential for smaller, recreational vessels to transport and distribute Aureococcus. During the summer of 2002, 11 trailered boats from the inland bays of Delaware and coastal bays of Maryland were sampled. Brown tide was detected in the bilge water in the bottoms of eight boats, as well as in one live-well sample. Commercial ships and small recreational boats are therefore implicated as potential vectors for long-distance transport and local-scale dispersal of Aureococcus.  相似文献
