SUMMARY. Colonizing aquatic macroinvertebrates were collected from two kinds of artificial substrate placed on wing dams in Pool 13 of the Upper Mississippi River in September 1978. Thirty-one taxa were collected from basket samplers containing cement spheres and twenty-one taxa from multiplate samplers constructed from tempered hardboard. Hydro-psychidae (Trichoptera), especially Cheumatopsyche sp., Potamyla flava and Hydropsyche sp., were the dominant macroinvertebrates which colonized both samplers. Basket samplers had a significantly greater macroinvertebrate density, biomass and number of taxa compared with multiplate samplers. Precision of the arithmetic mean for density, biomass and number of taxa was 19.9, 18.3 and 8.1% for basket samplers and 18.8, 18.7 and 8.5% for multiplate samplers. The number of sampling units required for a precision of 20% for macroinvertebrate density, biomass and number of taxa was 13, 11 and 2 for basket samplers and 11, 11 and 2 for multiplate samplers. Basket samplers with 7.5-cm cement spheres are recommended for use instead of multiplate samplers.  相似文献

In the summers of 1974 and 1975 pollen deposition was studied in two bays of the Baltic Sea near Stockholm with roofed and unroofed samplers mounted on rafts and with samplers submerged to levels of 30 and 60 cm over the bottom. The unroofed traps show larger totals of pollen grains than the roofed samplers. Sufficient data could not be obtained to establish that the totals of pollen grains in the submerged samplers at level 30 cm were larger than the totals at level 60 cm. The differences in pollen rain in the two seasons can be attributed mainly to the meteorological conditions, viz, to the particularly dry and warm summer of 1975. In the raft samplers the percentages of redeposited arboreal pollen are lower than in the submerged ones. Transport by currents and redeposition influence the pollen spectra in the submerged samplers. The pollen spectra in samplers over and under the water surface depend mainly on the specific composition of the surrounding vegetation. Pollen transport over short distances is therefore of great importance. No evidence was found of long-distance pollen transport.  相似文献

Reconstructing biological networks using high-throughput technologies has the potential to produce condition-specific interactomes. But are these reconstructed networks a reliable source of biological interactions? Do some network inference methods offer dramatically improved performance on certain types of networks? To facilitate the use of network inference methods in systems biology, we report a large-scale simulation study comparing the ability of Markov chain Monte Carlo (MCMC) samplers to reverse engineer Bayesian networks. The MCMC samplers we investigated included foundational and state-of-the-art Metropolis–Hastings and Gibbs sampling approaches, as well as novel samplers we have designed. To enable a comprehensive comparison, we simulated gene expression and genetics data from known network structures under a range of biologically plausible scenarios. We examine the overall quality of network inference via different methods, as well as how their performance is affected by network characteristics. Our simulations reveal that network size, edge density, and strength of gene-to-gene signaling are major parameters that differentiate the performance of various samplers. Specifically, more recent samplers including our novel methods outperform traditional samplers for highly interconnected large networks with strong gene-to-gene signaling. Our newly developed samplers show comparable or superior performance to the top existing methods. Moreover, this performance gain is strongest in networks with biologically oriented topology, which indicates that our novel samplers are suitable for inferring biological networks. The performance of MCMC samplers in this simulation framework can guide the choice of methods for network reconstruction using systems genetics data.  相似文献

OBJECTIVE: To compare three types of esophageal exfoliative cytology samplers in terms of patient acceptability, ease of use, final destination of the sampler tip in the gastrointestinal tract and cellular yield. STUDY DESIGN: A controlled, single-blind, cross-over study was undertaken to compare the balloon, sponge and sponge-mesh samplers in healthy volunteers. After completing the three procedures in random order, participants were asked about their preferred method. Ease of use was defined as the technician's ability to perform the intubation successfully. Final destination of the samplers was assessed fluoroscopically. Cytopathologists determined the cellular yield of each sampler using the Bethesda System. RESULTS: Sixty-two volunteers participated. The two encapsulated samplers were significantly preferred over the balloon (P < .0001). There was no significant difference in ease of use, final destination or cellular yield of the three techniques. All three samplers were successfully intubated on the first attempt and retrieved adequate numbers of squamous and glandular cells in > 78% of cases. CONCLUSION: All three samplers obtained satisfactory yields of squamous and glandular cells, but the encapsulated samplers were more patient acceptable. The sponge-mesh sampler may be the least complicated sampler for field screening use. Larger-scale studies will be required to test the accuracy of these three samplers for detecting esophageal dysplasia and carcinoma.  相似文献

Airborne fungi were monitored at five sample sites with the Burkard portable, the RCS Plus, and the SAS Super 90 air samplers; the Andersen 2-stage impactor was used for comparison. All samplers were calibrated before being used simultaneously to collect 100-liter samples at each site. The Andersen and Burkard samplers retrieved equivalent volumes of airborne fungi; the SAS Super 90 and RCS Plus measurements did not differ from each other but were significantly lower than those obtained with the Andersen or Burkard samplers. Total fungal counts correlated linearly with Cladosporium and Penicillium counts. Alternaria species, although present at all sites, did not correlate with total count or with amounts of any other fungal genera. Sampler and location significantly influenced fungal counts, but no interactions between samplers and locations were found.  相似文献

Summary Distance sampling is a widely used methodology for assessing animal abundance. A key requirement of distance sampling is that samplers (lines or points) are placed according to a randomized design, which ensures that samplers are positioned independently of animals. Often samplers are placed along linear features such as roads, so that bias is expected if animals are not uniformly distributed with respect to distance from the linear feature. We present an approach for analyzing distance data from a survey when the samplers are points placed along a linear feature. Based on results from a simulation study and from a survey of Irish hares in Northern Ireland conducted from roads, we conclude that large bias may result if the position of samplers is not randomized, and analysis methods fail to account for nonuniformity.  相似文献

Two high-speed plankton samplers (Gulf VII/PRO-NET and MAFF/Guildline)currently in use to sample pelagic eggs and larvae in watersaround the UK are described. Both samplers are modificationsof Gulf III type samplers and are capable of being towed atspeeds of up to 7 knots (3.6 m s–1). The performance ofthe samplers (flow rates, degree of clogging of the nets andsample depth profiles) can be monitored during deployment. Thereal-time monitoring packages also provide vertical profilesof temperature, salinity and chlorophyll a fluorescence. Bothsamplers can be constructed in different sizes to suit the vesselfrom which the sampler is deployed and the expected concentrationof plankton in the sampling area.  相似文献

The Andersen six-stage impactor, the SAS (Surface Air System) impactor, the AGI-30 impinger, and gravity plates were evaluated for the retrieval of aerosol-released Pseudomonas syringae. The upper limits of the impactor samplers were exceeded at a spray concentration of 107 CFU/ml, indicating that these samplers are not appropriate for monitoring high airborne concentrations. Decreased cell concentrations were retrieved with increased sampling time for the Andersen and AGI samplers, indicating that a minimum sampling time is preferable for monitoring aerosolized vegetative cells.  相似文献

Experiments comparing spore concentrations obtained by a single versus multiple Rotorod samplers in a 2 m×2 m×2 m grid were conducted within similar ecological areas and environmental conditions. Mean spore concentrations obtained by samplers at the same height above ground level were not significantly different at the p=0.05 level in all tests. However, significant differences in spore concentrations were measured among samplers operating at different heights above ground level when wind velocities were greater than 5 m/sec and no free moisture was on the spore bearing tissue. Samplers operating near a spore source measured significant differences in spore concentrations at various heights above ground but not among samplers at the same height. A single volumetric sampler will adequately measure spore concentrations for powering simulation models, but care must be taken to determine the location of the sampling unit in relationship to spore source and ecological or environmental conditions.  相似文献

Two biological aerosol samplers, the Andersen two-stage microbial impactor and the May three-stage glass impinger, were examined to determine the benefits and effectiveness of the May sampler compared with the Andersen sampler, one of the most widely accepted samplers. Side-by-side samples were collected during simulated wastewater spray irrigation dispersion studies. Escherichia coli colony counts and air concentrations were statistically treated to determine the dependability of the May results with respect to the Andersen results. After data pairs containing potentially overloaded Andersen counts were eliminated, a linear regression of the remaining data was performed. It indicates that although the May sampler reports 82% of the Andersen sampler value, the correlation between the two samplers is good with an r2 value of 0.84. This comparison indicates that although there are differences between the two samplers, they do give comparable results and that when both are used in a sampling program, they tend to complement each other.  相似文献

SUMMARY. From 16 air-lift samplers described in the literature, three were selected for operation from a small boat: Mackey, Pearson et al. and Verollet & Tachet samplers. Random samples (number of sampling units n= 10) were taken in a large tank with a known number of dried peas, representing invertebrates, amongst stones of uniform size. Separate experiments were performed with three sizes of stones (modal sizes 2–4, 16–20 and 32–36 mm). Stratified-random samples (usually n= 10) were taken in rivers at lour sites with modal particle sizes of < 0.1 2–4. 32–128 and 64–128 mm. The samplers were compared with a Ponar grab in the tank experiments and the field, and with a Naturalist's dredge in the field. Rates of pumping at different air-flows were established for each sampler. Water-How increased with increasing air-How and increasing submergence of the riser-pipe. The Mackey sampler achieved the highest water-flow and lifted more substratum and larger stones (64–128 mm) than the other samplers. The Verollet & Tachet sampler had the lowest pumping rate and failed to lift stones > 8 mm long, whilst the Pearson et al. sampler, with an intermediate performance, lifted mud and stones < 32 mm long. In the tank experiments, the area from which the Mackey and Pearson et al. samplers collected peas varied with air-flow, operating time and type of substratum. The Maekey sampler over-sampled and the Pearson et al. sampler usually under-sampled their respective sampling areas, except the latter sampler at high air-flows gave approximately quantitative results comparable to those of the Ponar grab for peas on the surface of stones 16–20 mm. No sampler collected much at a depth of 3 cm on larger stones (32–36 mm), and the Verollet & Tachet sampler failed completely on all substrata. In field trials, the Mackey and Pearson et al. samplers provided good qualitative samples except on large stones (32–128mm). The Verollet & Tachet sampler lifted little material. The relative abundance of taxa often differed between samplers. Estimates of the number of invertebrates per m2 differed widely between samplers except for the Pearson et al. and Ponar grab which gave similar quantitative results at three sites. Values were very low for the Verollet & Tachet sampler and Naturalist's dredge and always very high for the Mackey sampler which often grossly over-sampled its sampling area. Therefore, these three samplers cannot be considered as quantitative. The relationship between the variances and the means of samples of peas in the tank and most invertebrates in the field followed a power law with values of the exponent b in the range 1.13–2.51. The present study completes the evaluation of 14 samplers used to sample benthic macro-invertebrates in deep rivers, and the more important conclusions from the comparative studies of seven grabs, four dredges and three air-lift samplers are summarized in a table.  相似文献

Millipore Coli-Count Samplers were used to enumerate colonies of laboratory cultures of coliform bacteria. The samplers gave significantly lower counts than standard membrane-filter procedures for both total and fecal coliforms. Although the samplers are useful for semiquantitative analysis as indicated by the manufacturer, they are not suitable for standard examinations of recreational waters.  相似文献

The validity of obtaining soil water for fecal coliform analyses by porcelain cup soil water samplers was examined. Numbers from samples of manure slurry drawn through porcelain cups were reduced 100- to 10,000,000-fold compared to numbers obtained from the external manure slurry, and 65% of the cups yielded coliform-free samples. Fecal coliforms adsorbed to cups apparently were released, thus influencing the counts of subsequent samples. Fecal coliforms persisted in soil water samplers buried in soil and thus could significantly influence the coliform counts of water samples obtained a month later. These studies indicate that porcelain cup soil water samplers do not yield valid water samples for fecal coliform analyses.  相似文献

Olive and grass pollen grains are the major causes of hay fever in the Mediterranean region. A number of samplers and methods have been developed in recent years in order to obtain reliable data regarding airborne allergen concentrations. This paper reports on a detailed comparison of two samplers—Cyclone and ChemVol—and on the parameters that could influence their efficiency. Airborne concentrations of two key olive and grass allergens, Ole e 1 and Phl p 5, respectively, were monitored over two years with different weather patterns, 2012 and 2014. Allergenic particles were quantified by ELISA assay, and results were compared with pollen concentrations monitored using a Hirst-type volumetric spore trap over the same study periods. The influence of weather-related parameters on local airborne pollen and allergen concentrations was also analysed. Although a positive correlation was detected between results obtained using the two samplers during the pollen season, results for the cumulative annual Allergen Index varied considerably. The two samplers revealed a positive correlation between pollen concentrations and both minimum temperature during the warmer year (2012) and maximum temperature during the cooler year (2014); a negative significant correlation was observed in both cases with rainfall and relative humidity. In summary, although some differences were observed between the two samplers studied, both may be regarded as suitable for allergen detection.  相似文献

Millipore HPC samplers are simple, self-contained test devices that can be used by personnel in dental offices who do not have microbiologic training to easily and economically monitor dental unit water quality without laboratory support. This study evaluated the correlation of HPC samplers to R2A agar for enumerating planktonic bacteria in dental unit treatment water. Eight different dental units were sampled. Five replicates were performed for each media at each dilution. The Pearson correlation coefficient between the R2A agar and HPC sampler is 0.89. These data suggest HPC samplers correlate with conventional laboratory-based R2A culture techniques for determining dental unit water line contamination. Summary The relative sampling efficiency of the Samplair (Allergenco Inc.) and Rotorod samplers (Sampling Technologies Inc.) were compared for different conifer pollen sizes and at different wind speeds. The wind tunnel experiments conducted with the two samplers are valid for the range of wind speeds and conifer pollen types which might be expected in seed orchards in Canada. A Quadratic relationship was established between the two samplers which revealed a decrease in relative efficiency on the part of the Samplair as compared to the Rotorod at wind speeds less than 5 m/s. An apparent increase in the efficiency of the samplair at higher wind speeds resulted from a decrease in the sampling ability of the rotorod.  相似文献

Endotoxins are a component of Gram-negative bacteria cell walls and are known to be present in biosolids. Endotoxins have been shown to be potent stimulators of the innate immune response causing airway irritation and shortness of breath. Class B biosolids are routinely applied to agricultural lands to enhance soil properties and can be used as an alternative to chemical fertilizers. This study investigated the aerosolized endotoxin dispersed during the land application of Class B biosolids on agricultural land and a concrete surface at two sites in Colorado, USA. Aerosolized endotoxin was captured using HiVol samplers fitted with glass fiber filters, polycarbonate filter cassettes (both open and closed) and BioSampler impinger air samplers. Endotoxins were also measured in the biosolids to allow for correlating bulk biosolids concentrations with aerosol emission rates. Endotoxin concentrations in biosolids, impinger solutions and filter extracts were determined using the kinetic Limulus amebocyte lysate assay. Aerosolized endotoxin concentration was detected from all sites with levels ranging from 0.5 to 642 EU/m3. The four types of sampling apparatus were compared, and the HiVol and open-faced cassette samplers produced higher time-weighted average (TWA) measurements (EU/m3) than the impinger and closed cassette samplers. Ambient wind speed was found to be the variable best describing the observed results with optimal wind speed for highest deposition estimated at 5 m s?1. It is argued that HiVol air samplers are a particularly reliable approach and subsequent analyses relating TWA measurements to wind speed and biosolids characteristics were based on the measurements collected with those samplers.  相似文献

Two commercially available soil water samplers and a ceramic sampler constructed in our laboratories were evaluated for their ability to recover viruses from both tap water and secondary sewage effluent. The ceramic sampler consistently gave the best recoveries of viruses from water samples. Soil columns containing ceramic samplers at various depths provide a simple method for studying virus transport through sewage-contaminated soils.  相似文献
