The present study was carried out as part of an ongoing general survey for myxosporean parasites infecting tilapias in the River Nile, Egypt. In the present study, 77 Nile tilapia (Oreochromis niloticus) were collected from boat landing sites at Beni-Suef governorate, Egypt and examined for the myxosporean infection. The infection was encountered as a huge number of free spores in the kidney and the spleen. The infection showed a prevalence of 51.9% (40/77) for Myxobolus brachysporus while it was 25.9% (20/77) for Myxobolus israelensis. Mature spores of M. brachysporus were ellipsoidal and measured 8.6 × 13.2 μm. The polar capsules were subcircular with 5–6 filament turns and measured 4.7 × 3.6 μm. Spores of M. israelensis were ellipsoidal in the frontal view and fusiform in the lateral view. Spore measurements were 13.4 μm long and 8.7 μm wide. The polar capsules were elongated with 6–7 filament coils and measured 8.6 × 3.1 μm. The findings presented here proved that tilapia fishes in the Nile River are still suffering from infections with Myxobolus species. Therefore, further studies should be carried out to survey the Myxobolus infection among tilapias under culture conditions to clarify the pathological impacts of this parasite in tilapias aquaculture.  相似文献

The microsporidium Unikaryon phyllotretae sp. n., a new pathogen of Phyllotreta undulata, is described based on light microscopic and ultrastructural characteristics. Microscopic examination of parasitized individuals revealed two types of spores. The majority of the spores were of the first type, which are oval and measured 2.74±0.17×1.93±0.17 μm when fresh. Fresh spores of the second type (very rare) are elongated and measured 4.39±0.18×1.61±0.20 μm. All life stages have single nuclei. Sporogony ends with uninucleate single sporoblasts and spores. The spores were only observed in Malpighian tubules. The isofilar polar filament of the parasite has six to eight coils, and a well-developed polaroplast was of the lamellated type, with closely packed anterior lamellae and loosely packed posterior lamellae.  相似文献

Unicapsula seriolae (Myxozoa; Multivalvulida) was found in the trunk muscle of Malabar grouper Epinephelus malabaricus caught off Wakayama Prefecture, Japan. Numerous filamentous or sesamoid brown to black lesions were observed in the skeletal muscle. Histopathological observation indicated that the lesions were myxosporean plasmodia encapsulated by a fibrous layer, accompanied by melanin deposition. Spores having one large and two rudimentary polar capsules were subspherical in shape and 6.6 × 6.9 μm in size. Scanning electron microscopy revealed that spores were composed of three spore valves. Morphological characteristics were consistent with U. seriolae, which is reported to cause myoliquefaction in yellowtail kingfish Seriola lalandi in Australia. Molecular analysis of the SSU and LSU rDNA supported identification of the species as U. seriolae. This is the first report of Unicapsula in Japan.  相似文献

Henneguya jocu n. sp. (Myxosporea, Myxobolidae) is described from the gill lamellae of the marine teleost fish Lutjanus jocu, with a focus on ultrastructural and molecular features. This myxosporean forms subspherical cysts up to ∼260 μm × 130 μm long, and develops asynchronously. Mature myxospores ellipsoidal with a bifurcated caudal process. Myxospore length 10.9 ± 0.4 μm (n = 50); width, 8.2 ± 0.3 μm (n = 50); and thickness, 2.9 ± 0.5 μm (n = 50). Two equal caudal processes, 34.1 ± 1.0 μm long (n = 50); and total myxospore length, 45.2 ± 1.0 μm (n = 50). Two symmetric valves surround two ellipsoidal polar capsules, 5.0 ± 0.3 × 1.4 ± 0.2 μm (n = 20), each containing an isofilar polar filament forming 4–5 coils along the inner wall of these structures, as well as a binucleated sporoplasm presenting a spherical vacuole and several globular sporoplasmosomes. Both the morphological data and molecular analysis of the SSU rDNA gene identify this parasite as a new species of the genus Henneguya. Maximum Likelihood and Maximum Parsimony analyses further indicate that the parasite clusters within others marine Myxobolidae species, forming a group alongside other Henneguya species described from marine hosts.  相似文献

Two new ectocarpic arbuscular mycorrhizal fungal species, Glomus drummondii and G. walkeri (Glomeromycota), found in maritime sand dunes of northern Poland and those adjacent to the Mediterranean Sea are described and illustrated. Mature spores of G. drummondii are pastel yellow to maize yellow, globose to subglobose, (58–)71(–85) μm diam, or ovoid, 50–80 × 63–98 μm. Their wall consists of three layers: an evanescent, hyaline, short-lived outermost layer, a laminate, smooth, pastel yellow to maize yellow middle layer, and a flexible, smooth, hyaline innermost layer. Spores of G. walkeri are white to pale yellow, globose to subglobose, (55–)81(–95) μm diam, or ovoid, 60–90 × 75–115 μm, and have a spore wall composed of three layers: a semi-permanent, hyaline outermost layer, a laminate, smooth, white to pale yellow middle layer, and a flexible, smooth, hyaline innermost layer. In Melzer's reagent, only the inner- and outermost layers stain reddish white to greyish rose in G. drummondii and G. walkeri, respectively. Both species form vesicular–arbuscular mycorrhizae in one-species cultures with Plantago lanceolata as the host plant. Henneguya cerebralis Pronin, 1972 (Myxozoa) was described from Kosogol graylings Thymallus arcticus nigrescens Dorogostaisky, 1923 in Lake Khovsgol (Mongolia) in 1972.H. cerebralis was redescribed using critical morphological features and 18S ribosomal DNA (rDNA) gene sequence. Parasite infects cranial cartilage of fish host. Plasmodia are white rounded or ovoid, by 0.1 to 2 mm in size, containing large quantities of spores. Spore body is ovoid or rounded, 11.18 ± 0.13 μm (range 9,71–12,56) in length and 9.06 ± 0.16 μm (range 7.22–10,06) in width with equal polar capsules (4.7 × 2.6 μm). The two caudal appendages have different lengths (one of them was shorter in 20%).Phylogenetic position inferred by 18S rDNA shows that H. cerebralis is closely related with H. zschokkei, H. nuesslini, H. salminicola and H. cartilaginis which are histozoic parasites of salmonid fish.  相似文献

Gray mold caused by Botrytis cinerea is an important disease of strawberry. Clonostachys rosea is a mycoparasite of B. cinerea that reduces fruit losses when used as a biocontrol agent. Since spore production by C. rosea has not been optimized, we investigated factors affecting sporulation under aseptic conditions on white rice grains. The greatest spore production in glass flasks, 3.4 × 109 spores/g-dry-matter (gDM), occurred with an initial moisture content of 46% (w/w wet basis), inoculated with 1 × 106 spores/gDM and hand shaken every 15 days. However, a lower inoculum density (9 × 103 spores/gDM) and no shaking also gave acceptable sporulation. In plastic bags 1.1 × 108 spores/gDM were produced in 15 days, suggesting that larger scale production may be feasible: with this spore content, 24 m2 of incubator space would produce sufficient spores for the continued treatment of 1 ha of strawberry plants.  相似文献

The veiled polypore, Cryptoporus volvatus, is distributed widely in North America and East Asia and is believed to have a mutualistic relationship with coleopteran species—the fungus providing food and shelter in basidiocarps and beetle helping disperse spores.Seventy fresh basidiocarps of C. volvatus were collected from the Japanese red pine (Pinus densiflora) in the spring season of 2013 from two sites in Korea. A total of 251 insects (101 adult and 150 larvae) were collected from the inside of basidiocarps and identified using morphology and mitochondrial cytochrome c oxidase subunit I (COI) sequences. Six species belonging to five coleopteran families were identified. The number of spores attached to the bodies of adult insects was counted and average spore counts for each of the six species ranged between 1.0 × 104 and 5.2 × 105 spores/individual. Across localities, three species were shared (Aethina suturalis, Trogossita japonica and Parabolitophagus felix) and carried spores at high densities on their bodies, making them more likely to aid in spore dispersal.  相似文献

A new microsporidian species, Glugea arabica n. sp., is reported infecting the intestinal wall of the marine teleost Epinephelus polyphekadion (=microdon) collected from the Red Sea coast off Saudi Arabia, and described on the basis of microscopic and molecular procedures. Spherical blackish xenomas formed parasitophorous vacuoles completely packed with several parasitic developmental stages, including spores. The nuclei were monokaryotic in all developmental stages. Spores were ellipsoidal to pyriform and measured 6.3 ± 0.3 (5.9–6.6) μm in length and 3.3 ± 0.4 (2.9–3.7) μm in width. A lamellar polaroplast surrounded the uncoiled portion of the polar filament, which extended into the spore's posterior pole and formed 27–29 coils organized in three or four rows. The posterior vacuole, located at the spore's posterior pole, appeared surrounded by the polar filament coils and displayed an irregular matrix composed of light material, in which was located the posterosome. Molecular analysis of the rRNA genes, including the ITS region, was performed using maximum parsimony, neighbor-joining and maximum likelihood methodologies. The ultrastructural features observed, in combination with the molecular data analysed, suggests the parasite to be a new species of the genus Glugea.  相似文献

Using standard methods, we describe two new Drepanomonas taxa: Drepanomonas hymenofera (Horváth 1956) nov. comb., which is composed of two (biogeographical?) subspecies, viz., D. hymenofera venezuelensis nov. subspec. and D. hymenofera hymenofera (Horváth 1956), was discovered in soil from Venezuela and Iceland, respectively. Both are comparatively large-sized (50 × 20 μm and 40 × 18 μm in vivo), differing in the cortex pattern and the structure of kineties 3 and 4. We agree with Corliss (1979) and Chardez (1990) that the genus Pseudocristigera, which was established by Horváth (1956) for Drepanomonas hymenofera, is a junior synonym of Drepanomonas. Drepanomonas vasta nov. spec., which was discovered in the mud of a tree hole in Austria, is a middle-sized species (35 × 18 μm) with thick body, wide left side ridges, a single anterior dikinetid in kinety 4, and an average of 99 basal bodies; it is unique in having the dorsal side much more flattened than the ventral side, thus being cuneate in transverse view. Ontogenetic data show that the ciliary pattern of Drepanomonas is homologous to that of Leptopharynx, specifically, the structure and origin of the postoral complex. Main features for distinguishing Drepanomonas species are discussed.  相似文献

A new myxosporean parasite, Ortholinea nupchi n. sp. (Myxozoa; Bivalvulida), was isolated from the urinary bladder of the olive flounder Paralichthys olivaceus cultured on Jeju Island, Korea. Mature spores were subspherical in the valvular and apical views and ellipsoidal in the sutural view. The spores measured 7.6 ± 0.5 μm in length, 6.7 ± 0.3 μm in thickness, and 7.3 ± 0.5 μm in width. Two pyriform polar capsules measured 3.2 ± 0.1 μm in length and 2.7 ± 0.1 μm in width and were located at the same level at the anterior half of the myxospores. The suture line was straight in the middle of the spores, and the surface ridges ranged between five and seven, forming an intricate pattern. The result of the 18S rDNA comparison showed ≤ 93.0% similarity with other Ortholinea species. The phylogenetic tree demonstrated that O. nupchi n. sp. was closest to O. auratae and clustered with oligochaete-infecting myxosporeans (OIM) having urinary system infection tropism. Based on the comparison of environmental and host factors in the phylogenetic groups of the OIM clade, we propose that the infection of O. nupchi n. sp. originated from marine oligochaetes.  相似文献

Anthracnose of lentil, caused by Colletotrichum truncatum is a serious threat to lentil (Lens culinaris) grown in western Canada. The teleomorph stage of this pathogen was induced to form under laboratory conditions. Random pairing of single conidium isolates enabled the identification of fertile isolates. The individual isolates of this fertile pair were crossed with 14 other isolates, and all isolates were also incubated alone. Self-sterility was observed for all 16 isolates tested. Three isolates did not produce perithecia with either tester isolate, and none of the isolates tested produced perithecia with both tester isolates. Perithecia were brown–black, superficial, solitary or in small groups, obpyriform to ovate or ampulliform, 200–520 × 110–320 μm (mean: 350 × 200 μm). Asci were cylindrical, narrowing slightly at the apex, unitunicate, evanescent, 53–142 × 5–14 μm (mean: 90 × 8 μm), and contained eight ascospores. Ascospores were hyaline, aseptate, oblong, 12–20 × 5–8 μm (mean: 15.7–6.7 μm). The characteristics agree with those described for the genus Glomerella, and the species was named G. truncata sp. nov. The morphology of the new species is compared with that of other species in the genus, and future research on G. truncata is described.  相似文献

The influence of infrared (IR) radiation on the viability and heat-activation of Bacillus subtilis spores, suspended in phosphate-buffered saline, was investigated. Two types of IR heaters with different spectral distributions were used. Near-infrared (NIR) and far-infrared (FIR) heaters with main wavelengths of approximately 1 μm and 3–6 μm, respectively, were utilized. Although both irradiation treatments decreased the number of B. subtilis colonies at a bulk temperature of approximately 75 °C, the mode of action was clearly different. In the case of the NIR heater, the number of colony-counts decreased gradually. In contrast, use of the FIR heater resulted in heat activation of the spores during the early stage of irradiation at a low bulk temperature (40–60 °C) over several minutes, followed by a decrease in the number of colonies. Consequently, FIR irradiation inactivated 90% of B. subtilis spores more effectively as compared to NIR irradiation for 20 min with a suspension volume of 20 ml and irradiation energy of 7.57 kW m?2. Spore exposure to FIR irradiation accelerated their germination rate in nutrient broth; however, this was not true for treatment with the NIR heater. The absorption IR spectrum of B. subtilis spores indicated that FIR radiation was absorbed easily by the spore cell components and might activate the bioactive substances involved in germination. Even at the same irradiation energy, the influence of infrared radiation on spore germination was dependent on the IR spectral distribution. Bacterial spores undergoing germination lose their resistance to stressors, such as heat, chemicals and ultraviolet rays. FIR heating promotes heat activation and germination, thereby producing vegetative cells that are more susceptible to other killing methods, enabling the killing of bacterial spores at lower stress without product damage.  相似文献

Thelohanellus macrovacuolaris n. sp. is described during a survey on myxozoan diversity of common carp Cyprinus carpio L. in China. It is characterized by the presence of round or ellipsoidal plasmodium in the palate in the mouth of host. Mature spores were pyriform in frontal view, lemon shaped in lateral view, measuring 21.6 ± 0.9 (19.3–23.8) long, 12.5 ± 0.7 (10.3–13.6) wide, and 10.2 ± 0.4 (9.8–11.8) thick. Most spores were surrounded by the membrane sheath. Single polar capsule was round with an apophysis at its top end presented close to apex of spore, measuring 9.1 ± 0.6 (8.0–10.0) in length, 8.6 ± 0.5 (7.8–9.6) in width. Polar filaments coiled, with 7 to 8 turns. A large, round iodinophilous vacuole was present, with 5.8–7.5 in diameter. The present species is morphologically distinct from all other Thelohanellus species. The BLAST search indicated that the newly obtained small subunit ribosomal RNA (ssrRNA) gene sequence of T. macrovacuolaris n. sp. did not match any available sequences in GenBank. Phylogenetically, T. macrovacuolaris n. sp. was placed sister to Thelohanellus wangi in the Thelohanellus clade. Both morphology and ssrRNA gene sequence data revealed that the present parasite is a new species of genus Thelohanellus.  相似文献

Exogenous and endogenous stages of Eimeria perforans naturally infected rabbits in Saudi Arabia were described. The prevalence of infection was 75%. Oocysts were ovoid to elliptical and measured 16 × 10 μm. The four dizoic sporocysts were ovoid and measured 7 × 5 μm. Endogenous stages were restricted to the duodenum. Light and scanning electron microscopy were used to study leaf and stem fine structure of the drought deciduous green-stemmed Mediterranean shrub Calicotome villosa (Poiret) Link. (Leguminosae). Each leaf consists of three small obovate leaflets with abundant but small (16 μm length) anomocytic stomata on both surfaces. Adaxial surface exhibits more than double stomatal density (440±8 mm−2) than the abaxial one (185±4 mm−2). T-shaped trichomes (36±3 mm−2) are present only on the abaxial leaf surface. Leaves are unifacial, furnished with palisade parenchyma on both sides. The stem is characterized by raised ridges and grooves. Beneath the one-cell-layered epidermis sclerenchyma is found on ridges, whereas stomata and palisade chlorenchyma are found in grooves. Hairs are abundant, especially in grooves. Stem and leaf palisade chlorenchymas are structurally similar. According to these data, photosynthesis could be efficiently supported by the stem.  相似文献

Marine toxic dinoflagellates of the genus Gambierdiscus are the causative agents of ciguatera fish poisoning (CFP), a form of seafood poisoning that is widespread in tropical, subtropical and temperate regions worldwide. The distributions of Gambierdiscus australes, Gambierdiscus scabrosus and two phylotypes of Gambierdiscus spp. type 2 and type 3 have been reported for the waters surrounding the main island of Japan. To explore the bloom dynamics and the vertical distribution of these Japanese species and phylotypes of Gambierdiscus, the effects of light intensity on their growth were tested, using a photoirradiation-culture system. The relationship between the observed growth rates and light intensity conditions for the four species/phylotypes were formulated at R > 0.92 (p < 0.01) using regression analysis and photosynthesis-light intensity (P-L) model. Based on this equation, the optimum light intensity (Lmax) and the semi-optimum light intensity range (Ls-opt) that resulted in the maximum growth rate (μmax) and ≥80% μ max values of the four species/phylotypes, respectively, were as follows: (1) the Lmax and Ls-opt of G. australes were 208 μmol photons m−2 s−1 and 91–422 μmol photons m−2 s−1, respectively; (2) those of G. scabrosus were 252 and 120–421 μmol photons m−2 s−1, respectively; (3) those of Gambierdiscus sp. type 2 were 192 and 75–430 μmol photons m−2 s−1, respectively; and (4) those of Gambierdiscus sp. type 3 were ≥427 and 73–427 μmol photons m−2 s−1, respectively. All four Gambierdiscus species/phylotypes required approximately 10 μmol photons m−2 s−1 to maintain growth. The light intensities in coastal waters at a site in Tosa Bay were measured vertically at 1 m intervals once per season. The relationships between the observed light intensity and depth were formulated using Beer’s Law. Based on these equations, the range of the attenuation coefficients at Tosa Bay site was determined to be 0.058–0.119 m−1. The values 1700 μmol photons m−2 s−1, 500 μmol photons m−2 s−1, and 200 μmol photons m−2 s−1 were substituted into the equations to estimate the vertical profiles of light intensity at sunny midday, cloudy midday and rainy midday, respectively. Based on the regression equations coupled with the empirically determined attenuation coefficients for each of the four seasons, the ranges of the projected depths of Lmax and Ls-opt for the four Gambierdiscus species/phylotypes under sunny midday conditions, cloudy midday conditions, and rainy midday conditions were 12–38 m and 12–54 m, 1–16 m and 1–33 m, and 0 m and 0–16 m, respectively. These results suggest that light intensity plays an important role in the bloom dynamics and vertical distribution of Gambierdiscus species/phylotypes in Japanese coastal waters.  相似文献

Community structure and species richness of arbuscular mycorrhizal fungi (Phylum Glomeromycota) were studied in sand dune sites at Itapiruba (southern), Joaquina (intermediate) and Praia Grande (northern) beaches along the coast of the state of Santa Catarina, Brazil. In each site, a 20 × 20 m plot was established and 20 soil samples collected in a regular grid pattern. Fungal spores were extracted from each sample, counted and identified to species level. A total of 25 species were recovered belonging to seven genera and five families in the Glomeromycota. Gigaspora albida and Acaulospora scrobiculata occurred in >50 % of samples at all three sites. Other common species whose sample frequency was >50 % in one or two sites were Scutellospora weresubiae, Scutellospora cerradensis and Racocetra gregaria, while the remaining majority of species were detected in <25 % of samples within a given site. Dune sites could be differentiated based on the higher frequency of occurrence of S. cerradensis and Acaulospora morrowiae in Itapiruba, S. weresubiae in Joaquina, and Scutellospora hawaiiensis in Praia Grande. No differences across sites were observed for species richness and total spore numbers, the latter averaging from 28.8 to 31.8 spores per 100 ml soil. Shannon diversity was significantly higher in Praia Grande compared to the other two sites. Differences in the relative spore abundance of genera among dunes were detected only for Scutellospora, which was significantly more abundant in the Joaquina beach. Community structure, as depicted by species rank/log abundance graphs, was not significantly different between areas according to the Kolmogorov–Smirnov two-sample test. Species accumulation curves demonstrated that 13 samples were enough to detect 90 % of all species. Overall, sand dune systems share similar arbuscular mycorrhizal fungal communities despite being geographically distant (150 km) from each other.  相似文献

Clostridium taeniosporum is a Gram-positive, anaerobic, rod-shaped non-toxigenic organism isolated from Crimean lake silt. It is unique in forming spores from which about twelve large, flat, ribbon-like appendages emanate. These ribbon-like structures, about 4.5 μm long and 0.45 μm wide, are assembled from smaller fibrils with 5 nm diameter spherical heads attached to thin tails about 1–2 nm in diameter and about 40 nm in length. The appendages have four major components, a glycoprotein with a collagen-like region, two proteins each of which contains two conserved domains of unknown function, and an ortholog of the Bacillus subtilis spore morphogenetic protein SpoVM. Genes for three of these and other, possibly related proteins, cluster on two chromosome fragments. Here we report that C. taeniosporum is saccharolytic, non-proteolytic, and produces both acetic and butyric acid fermentation products. It synthesizes α-d-glucosidase and N-acetyl-β,d-glucoseaminidase constitutively. These physiological properties are similar to those of the C. botulinum Group II. Genotypically, C. taeniosporum is also closely related to the same Group II, based on 16S rDNA sequences. C. taeniosporum differs from typical C. botulinum Group II strains because it is non-toxigenic and in forming the ribbon-like spore appendages. These major differences among otherwise closely related organisms suggest lateral transfer of genes for appendage synthesis and for toxigenicity.  相似文献

The capability of Bacillus subtilis spores to withstand extreme environmental conditions is thought to be conferred especially by their outermost proteinaceous protective layer, called the spore coat. Of the over 70 proteins that form the spore coat, only a small subset of them affect its morphogenesis, they are referred to as morphogenetic proteins. In this study we investigated the interaction between two spore coat morphogenetic proteins SpoVID and CotE. SpoVID is involved in the process of spore surface encirclement by individual coat proteins, these include CotE, which controls the assembly of the outer coat layer. Both proteins were proposed to be recruited to a common protein scaffold, but their direct association has not been previously shown. Here we studied the interactions between CotE and SpoVID in vitro for the first time by using molecule recognition force spectroscopy, which allows the detection of piconewton forces between conjugated biological pairs and also facilitates the investigation of dynamic processes. The most probable CotE–CotE unbinding force was 49.4 ± 0.1 pN at a loading rate of 3.16 × 103 pN/s while that of SpoVID–CotE was 26.5 ± 0.6 pN at a loading rate of 7.8 × 102 pN/s. We further analyzed the interactions with the bacterial two hybrid system and pull-down experiments, which also indicate that SpoVID interacts directly with CotE. In combination with the previously identified direct contacts among SpoIVA, SpoVID and SafA, our data imply that the physical association of key morphogenetic proteins forms a basic skeleton where other coat proteins could be attached.  相似文献
