Between January, 1, 1986 and December, 31, 2000, dermatological specimens from 10.678 animals (7.650 cats and 3.028 dogs) were examined for dermatophytes. All the animals presented clinical signs of ringworm. Two thousand-four hundred fifty-six of the 10.678 (23%) examined animals scored positive for dermatophytes, 566 out of 3.028 canine (18.7%) and 1890 out of 7.650 feline specimens (24.7%). Microsporum canis constituted 83% and 97% of the isolated dermatophytes respectively in dogs and cats, M. gypseum represented 13% and 2.6% and T. mentagrophytes 5.5% and 0.2%. A sexual predisposition for mycotic infections was not observed. The animals with less than 1 year of age were more frequently infected. Canine toy breeds showed a significantly higher (P < 0.001) prevalence of infections by M. canis. Microsporum gypseum was mostly recorded from sporting (hunting) breeds [such as T. mentagrophytes (6.7%)]. Microsporum canis was isolated from long-haired cats with a ratio of 2:1 versus short-haired cats, while M. gypseum and T. mentagrophytes were never recovered from Persian cats. The annual distribution of the infections in dogs showed a significantly higher incidence for M. gypseum in summer versus winter and spring, while the recovery rate of M. canis from cats was very significantly higher in fall and winter than in summer and spring. Macroconidia ofMicrosporum canis, when placed in a nutrient medium produce germ tubes within 4–6 h. Precursor incorporation studies showed that protein synthesis occurred prior to RNA synthesis. Sucrose density gradient analysis of wet and dry spore extracts revealed the presence of 16 % and 11 % polysomes respectively. The polysomal content increased to about 50% within 15 min of germination. Synthesis of RNA occurred only after 2 h of germination. Pool equilibration of the radioactive precursors was not limiting to these measurements. Polyadenylated RNA was isolated from macroconidia and was found to comprise 2–2.5 % of the total RNA. The poly(A)+ RNAs were heterodisperse and translatable in a wheat germ cell free translating system. Water-soluble antigens liberated from the disrupted mycelium of nine dermatophytes (seven isolates ofMicrosporum canis, one each ofMicrosporum gypseum andTrichophyton mentagrophytes) were compared by analytical slab SDS-PAGE. No substantial differences were observed between the protein bands of theM. Canis isolates, but certain distinctive bands were apparent in the other two species examined. Western immunoblotting usingM. canis-derived antigens separated by SDS-PAGE was used to investigate the humoral immune response in 79 cats with naturally-occurring dermatophytosis (72 withM. canis, six withM. gypseum and one withT. mentagrophytes) and this information was compared to results of immunoblots from 46 control (non-dermatophyte exposed) cats. Seven dominant bands (bands which occurred frequently and stained heavily) were identified in immunoblots from the dermatophyte-infected cats with apparent molecular weights varying between 39 and 120 kD. None of these bands were totally specific markers for dermatophytosis as a variable proportion of the control cats showed reactivity to all these proteins. However, most (73%) of the dermatophyte-infected cats showed reactivity to six or seven of the identified bands whereas most (80%) of the control cats showed reactivity to between zero and three of these bands (p<0.005). Western immunoblotting could be used to select individual immunodominant antigens for further evaluation of protective (cell-mediated) immunity.  相似文献

The enzymatic activity of 70 feline and canineMicrosporum canis isolates was determined by the Api-Zym® test. The liquid phase of cultures, inoculated into Tryptic Soy Broth, was used to examine 19 enzymes. Considerable differences were observed among the extracellular enzymatic patterns. All the isolates produced alkaline phosphatase and beta-glucosidase, while lipase (C14), trypsin, chymotrypsin, beta-glucuronidase, and alpha-fucosidase activity was never revealed. Esterase (C4) activity was present in 57 samples (81%), esterase lipase (C8) in 31 (44%), leucine arylamidase in 35 (50%), valine arylamidase and cystine arylamidase in 7 (10%), acid phosphatase in 64 (91%), naphthol-AS-BI-phosphohydrolase in 60 (86%), alpha-galactosidase in 5 (7%), beta-galactosidase in 6 (8%), alpha-glucosidase in 25 (36%), N-acetyl-beta-glucosaminidase in 41 (58%), and alpha-mannosidase in 51 (73%). The beta-galactosidase activity ofM. canis has not been reported previously. Remarkable variations of intensity for each enzymatic activity were also detected. It is believed that these results could provide basic data for further investigations on the pathogenic role of enzymes secreted byM. canis.  相似文献

Retrospective studies have shown the occurrence of episodes of deep or superficial fungal infections in 58 to 81% of HIV/AIDS patients as a result of impairment of cell immunity. We describe a case of disseminated cutaneous dermatophytoses caused by Trichophyton mentagrophytes and Microsporum canis in a patients with AIDS. Diagnostic and therapeutic problems in relation to this unsual presentation are emphasized as well as the importance of an early mycologic diagnosis to prescribe antifungal therapy.  相似文献

Between 1985 and 1990 we observed 2085 cases of dermatophytoses in the Florence area of Italy.Trichophyton rubrum andMicrosporum canis were the major etiological agents. 126 cases of dermatophytoses observed during this period were caused byTrichophyton mentagrophytes. Most of the patients came from rural areas or they kept pets.Epidermophyton floccosum ranked fourth in frequency (95 cases). The patients were adults and mainly males, most of whom engaged in sports or lived in group environments.Microsporum gypseum was isolated in 31 cases. Infections occurred directly from contact with soil or domestic animals. We report the principal clinical data concerning these last three dermatophyte infections.  相似文献

The isolation of a dysgonic variety ofMicrosporum canis from a large number of cats and kittens in a cattery is described. The normal variety of this fungus was isolated at the same time from the same animals. Dysgonic varieties are thought to be mutants of normal strains, but this isolation of both forms together suggests that the relationship may be more complex.  相似文献

The mycoflora of 220 skin scrapings, hair, nail samples and pens’ materials of apparently healthy animals including cows, sheep, goats, rabbits, pigs and dogs were determined. Twenty eight species of fungi belonging to ten genera were recovered. Chrysosporium spp. were the most common and C. keratinophilum was recovered from all animals. Dermatophytes which are known causal agents of dermatophytosis were also isolated indifferent frequencies (Microsporum gypseum, M. canis, Trichophytonmentagrophytes, T. rubrum). The causative agents of tinea capitis in Libyan nationals attending the out patient Dermatology Clinic of the Tripoli Medical Centre over the period December 1997 to December 1999 were investigated. Samples (hair and scalp scrapings) were taken from 940 patients who presented with suspected tinea capitis. The etiological agents were identified in 584 cases. Trichophyton violaceum was found to be the most prevalent organism isolated being responsible for 64.4% (376/584) of culture positive cases, followed by Microsporum canis at 24.7% (144/584) and T. mentagrophytes at 5.5% (32/584). The majority of infections (380/584) occurred in females and in children with ages less than 12 years (554/584). Since 1957, Microsporum (M.) canis has been one of the common causative agents of dermatophytosis in Korea. We analyzed 944 patients infected with M. canis who attended outpatient clinic over a 17-year period (1993–2009). M. canis infections were diagnosed by mycological examinations, including direct microscopic examinations with 15% KOH and cultures on potato dextrose agar complemented with 0.5% chloramphenicol. Mycological examinations confirmed 164,903 cases of dermatophytosis, 944 (0.6%) of which were M. canis infections. The annual prevalence of patients with M. canis infection was the highest in 2002 (91 cases). Then, the prevalence of patients with M. canis infection markedly decreased until 2008 (15 cases). The ratio of male to female patients was 0.65:1, but the ratio in children under the age of 15 was 1.14:1 and was 0.20:1 in adults. Seasonally, 274 cases occurred in winter, showing higher incidence than in other seasons. There was a difference in the clinical forms of M. canis infections between children and adults; tinea (T.) capitis was the most common form in children, but T. corporis was the most common in adults. We demonstrate that the decreasing prevalence of M. canis infections over the study period has been noted in Korea.  相似文献

This study was undertaken to assess the in vitro antifungal potential of the essential oil and n-hexane, chloroform, ethyl acetate, and methanol extracts of Nandina domestica Thunb. against dermatophytes, the casual agents of superficial infections in animals and human beings. The oil (1,000 μg/disc) and extracts (1,500 μg/disc) revealed 31.1–68.6% and 19.2–55.1% antidermatophytic effect against Trichophyton rubrum KCTC 6345, T. rubrum KCTC 6375, T. rubrum KCTC 6352, Trichophyton mentagrophytes KCTC 6085, T. mentagrophytes KCTC 6077, T. mentagrophytes KCTC 6316, Microsporum canis KCTC 6591, M. canis KCTC 6348, and M. canis KCTC 6349, respectively, along with their respective minimum inhibitory concentration values ranging from 62.5 to 500 and 125 to 2,000 μg/ml. Also, the oil had strong detrimental effect on spore germination of all the tested dermatophytic fungi as well as concentration and time-dependent kinetic inhibition of T. rubrum KCTC 6375. The present results demonstrated that N. domestica mediated oil and extracts could be potential sources of natural fungicides to control certain important dermatophytic fungi.  相似文献

Abstract The role of Musca domestica Linnaeus as a vector of the dermatophyte Microsporum canis was investigated under experimental laboratory conditions. About 400 4‐day‐old M. domestica flies were divided into two groups. Group A consisted of about 200 infected flies and group B comprised about 200 uninfected flies that were used as controls. Each trial was run three times. Flies from group A were fed for 24 h with a solution of ultra‐high temperature‐treated (UHT) milk containing about 106 colony‐forming units (CFU) per mL of M. canis (infected milk inoculum [IMI]). The control group (group B) was fed with only UHT milk spiked with a teaspoon of honey. Microsporum canis was detected from faeces, vomitus, external surfaces and internal organs of 20 adult flies, eggs, first‐, second‐ and third‐stage (L1, L2, L3) larvae and pupae of each group, as well from 20 adult newly emerged flies (NEFs; from infected generations only). Samples were collected at 2, 4, 6 and 24 h post‐infection (p.i.) (i.e. the times at which IMI was available) and on 2, 5, 7 and 8 days p.i. from adult flies, faeces and vomitus. Eggs, L1, L2, L3 and pupae were processed as soon as they appeared. Equivalent samples were taken from group B. All the samples were individually cultured. Microsporum canis was not isolated from the control group, from eggs, larvae, pupae or NEFs, or from faeces and vomitus, although it was detected on the body surface (26.2%) and internal organs (26.9%) of adult flies. The highest positivity for M. canis was detected on flies within the first 6 h p.i. (i.e. 57.2% on the body surface and 71.6% in the internal organs). No M. canis was isolated at 24 h p.i., but it was isolated from the body surface only at 2 and 5 days p.i. The results presented provide evidence that M. domestica transmits M. canis mechanically with its outer body surface for up to 5 days p.i., but does not do so through its vomitus and faeces or transovarially. The role played by M. domestica in the epidemiology of human and animal dermatophytoses is discussed.  相似文献

Among 67 endophytic fungi isolated from Quercus variabilis, 53.7% of endophytic fungal fermentation broths displayed growth inhibition on at least one test microorganism, such as pathogenic fungi (Trichophyton rubrum, Candida albicans, Aspergillus niger, Epidermophyton floccosum, Microsporum canis) and bacteria (Escherichia coli, Bacillus subtilis, Pseudomonas fluorescens). Moreover, 19.4% of strains showed a broader antimicrobial spectrum, such as Aspergillus sp., Penicillium sp., Alternaria sp., 20.9% of strains showed strong inhibition (+++) to pathogenic bacteria, while only 7.5% displayed that to test fungi. The most active antifungal strain I(R)9-2, Cladosporium sp. was selected and fermented. From the broth, a secondary metabolite, brefeldin A was obtained. This is the first report on the antimicrobial potentials of endophytic fungi residing in Q. variabilis and isolation of brefeldin A produced by Cladosporium sp.  相似文献

A crude ribosomal fraction was extracted from Microsporum canis. The ratio of spectrophotometric readings at 260 and 280 nm was 1.56 and the nucleic acid to protein ratio of the fraction was 1.5. A delayed intradermal hypersensitivity test in guinea pigs was positive and dose related.  相似文献

This is a comparative study to isolate the dermatophytes of tinea capitis using the cytobrush and comparing it versus the standard method. A prospective, observational, comparative trial of 178 probable cases of tinea capitis was conducted in two dermatological centers. Each patient underwent mycological tests that included direct exam with KOH and cultures with either of two methods: scraping the scalp to remove hair and cell debris, and the cytobrush. A total of 135 clinically and mycologically proven cases of tinea capitis were included; 119 were non-inflammatory and 16 inflammatory tinea. A total of 131 had a positive direct exam and subsequent primary isolation cultures were obtained in 135 cases. The main dermatophytes isolated were Microsporum canis (68%) and Trichophyton tonsurans (20%). A total of 115/135 (85.1%), were detected with the traditional method, with an average of 11.2 days until positive, while the number detected with the cytobrush was 132/135 (97.7%) with an average of 8.5 days until positive. The chi-square statistical method showed that the cytobrush culture was superior to the standard one with a chi-square of 5.078 (P = 0.025), with a statistically significant difference versus the standard method.  相似文献

In this study, we examined dermatophyte infections in patients referred to the Department of Dermatology, EL-Houd El-Marsoud Hospital, Cairo, during March 2004 to June 2005. Of 506 patients enrolled in this investigation, 403 (79.6%) were clinically diagnosed as having dermatophytoses (age range 6–70 years; males 240; females 163). Species identification determined by observation of their macroscopic and microscopic characteristics was complemented with sequencing of the internal transcribed spacer ITS1-5.8S-ITS2 rDNA region. The most common dermatophyte infection diagnosed was tinea capitis (76.4%), followed by tinea corporis (22.3%) and tinea unguium (1.2%). The most frequently isolated dermatophyte species was Trichophyton violaceum, which accounted for most (71.1%) of all the recovered dermatophytes, followed by Microsporum canis (21.09%), Trichophyton rubrum (6.2%), and Microsporum boullardii (0.49%); both Epidermophyton floccosum and Trichophyton tonsurans were each only rarely isolated (0.24%).  相似文献

A total of 4,287 primary school children, comprising 1,740 males and 2,547 females in Arochukwu local government area of Abia state Nigeria were examined for clinical signs of dermatophytoses. About 873 (20.4%), consisting of 505 males and 368 females had lesions consistent with dermatophytoses. The disease was more prevalent in males (29%) than females (14.4%) in a ratio of approximately 2:1 (P < 0.05). The infection rate increased from 16.8% in the 4–6 year age group to a peak of 28.1% in the 10–12 year age bracket and dropped sharply to 5.6% in the 16–18 year group. The highest prevalence (39%) was observed among males aged 10–12 years while females 16–18 years had the lowest prevalence (2.5%). Tinea capitis was the predominant clinical type of dermatophytoses, and occurred in 13.7% of the total population studied and 67% of lesion positive cases. Trichophyton soudanense and Trichophyton tonsurans the predominant aetiological agents of dermatophytoses with a prevalence of 26.2% and 21.6%, respectively. Others include Trichophyton mentagrophytes (18.8%), Epidermophyton floccosum (8.3%), Microsporum audouinii (6.4%), Microsporum gypseum (6.0%), Trichophyton rubrum (5.5%) and Microsporum ferrugineum (7.3%), which was isolated for the first time in Nigeria.  相似文献

Cutaneous lesions which can lead to false positive results have been observed in several rabbits used for the determination of the cutaneous irritation capacity of a product (ITA, PII). The responsible agent was Microsporum canis. A preventive treatment by an antifungal agent did not modify toxicological experimental results.  相似文献
