将L-谷氨酰胺与苯磺酰氯缩合,合成了与抗瘤酮A_(10)(3-苯乙酰氨基-2,6-哌啶二酮)的水解产物相类似的标题化合物。采用X-射线单晶衍射分析法研究了它的晶体结构。标题化合物分子式C_(11)H_(14)N_2O_5S,Mr=286.30,晶胞参数:a=5.373(2)A,b=15.073(1)A,c=16.277(3)A;α=89.99(1)~0,β=90.00(2)~0,γ=89.96(2)~0;正交晶系,空间群P2_12_12_1,V=1318.3A~3,Z=4,D_c=1.442g/cm~3,最终晶体学偏差因子R=0.031。  相似文献

从猪屎豆种子中提取分离,得到单猪屎豆碱(Monocrotaline)。经X射线衍射分析测定了晶体结构。该化合物属正交品系,空间群为P2_12_12_1,晶体学参数:a=10.072(7),b=11.431(1),c=13.560(2)A,V=1561.3(7)A,Z=4,D=1.384g·cm~(-1) ,μ(MoKa)=0.699mm~(-1),F(000)=696,M=325.36,分子式为C_(16)H_(23)NO_6。在1≤3a(1)收集1627个衍射点,最终偏差因子R和R_0分别为0.058和0.05232。  相似文献

目的:探讨圣草酚对Aβ_(25-35)诱导的PC12细胞损伤的保护作用及其机制。方法:采用MTT法筛选圣草酚对Aβ_(25-35)诱导的PC12细胞损伤的有效保护浓度,进一步采用流式细胞术检测圣草酚对Aβ_(25-35)诱导的PC12细胞的凋亡率的影响,并通过试剂盒检测圣草酚对Aβ_(25-35)诱导的PC12细胞胆碱能系统受体活性的影响。结果:MTT结果显示2×10~(-3)μmol/L圣草酚可显著促进Aβ2_(25-35)诱导的PC12细胞增殖(P0.05)。流式细胞术结果显示2×10~(-3)μmol/L圣草酚和雌二醇均可以显著抑制Aβ_(25-35)诱导的PC12细胞凋亡(P0.05),提高其乙酰胆碱及乙酰胆碱转移酶活性并且降低乙酰胆碱酯酶的活性(P0.05),且二者的作用相当,差异无统计学意义(P0.05)。结论:圣草酚可能通过提高Ach及ChAT的含量,降低AchE的含量,调节胆碱能系统相关酶的活性,发挥对Aβ_(25-35)诱导的PC12细胞的保护作用。  相似文献

用靶向探针追踪淀粉样蛋白(Aβ_(25-35))的亚细胞定位情况,同时基于Nrf2信号通路探讨阿里红多糖组分(FOAPs-a)和(FOAPs-b)对Aβ_(25-35)诱导的PC12细胞线粒体损伤通路的保护作用机制。采用40μmol/L Aβ_(25-35)诱导PC12细胞建立阿尔茨海默病(AD)细胞模型,将PC12细胞分为空白组、模型组(加40μmol/L Aβ_(25-35))、阳性组(加50μmol/L盐酸多奈哌齐)、不同浓度的FOAPs-a和FOAPs-b干预组(各50、100、200μg/mL)。以靶向探针追踪Aβ_(25-35)在各组PC12细胞中的亚细胞定位情况;通过试剂盒检测PC12细胞中活性氧自由基ROS的变化情况;Western blotting法测定细胞凋亡相关蛋白Bax及Bcl-2的表达量以及和Nrf2通路相关的Nrf2、APK1和磷酸化的APK1蛋白的表达情况。结果发现,Aβ_(25-35)处理PC12细胞会影响线粒体的完整性;在200μg/mL FOAPs-a/b预处理PC12细胞后,能够显著缓解Aβ_(25-35)对线粒体的损伤,同时使Aβ_(25-35)的亚细胞共定位减弱;与空白组比较,模型组细胞中ROS含量增加,与模型组比较,FOAPs-a及FOAPs-b干预组均能降低ROS的沉积,差异有统计学意义;Western blotting结果显示:与模型组比较FOAPs-a及FOAPs-b均能减少APK1的磷酸化水平,上调Nrf2的蛋白表达水平。总之Aβ_(25-35)可进入到PC12细胞的线粒体中引起其损伤,阿里红多糖组分能够通过激活Nrf2信号通路显著缓解Aβ_(25-35)对PC12细胞线粒体的损伤。  相似文献

采用大鼠整体灌流模型,同时测定灌流大鼠后肢血管床灌流压和全身平均动脉压,通过动态观察,比较多种α_1-肾上腺素受体(α_1-AR)亚型选择性拮抗剂对两者影响的异同,初步探讨α_1-AR亚型在大鼠整体血压调节中的作用。结果表明:α_1-AR选择性拮抗剂(prazosin组13.5±3.6vs 15.1±4.3,n=11)和α_1-AR亚型选择性拮抗剂(5-mithyl-urapidil组2.4±0.9vs 3.7±2.3,n=12;RS-17053组3.2±1.6vs 4.4±3.3,n=12)均对正常大鼠苯肾上腺素引起后肢血管床升压反应曲线和平均动脉压升压反应曲线的右移程度(dose ratio,Dr)无明显影响,α_1-AR亚型选择性拮抗剂(BMY 7378组1.9±0.9vs 2.2±0.8,n=8)对正常大鼠两者苯肾上腺素加压反应也无差别;自发性高血压大鼠(RS-17053组3.4±0.6vs 4.3±0.9,n=5;BMY 7378组1.7±0.5vs 1.7±0.5,n=8)的反应同正常大鼠相似。提示介导苯肾上腺素引起麻醉大鼠全身动脉加压效应的α_1-AR与引起大鼠后肢血管床收缩的α_1-AR可能是同一种亚型,即α_1-AR。  相似文献

Crystals of benzamidine-inhibited trypsin from the North Atlantic salmon (Salmo salar) have been grown from ammonium sulphate solution at pH 5.0. Two crystal forms suitable for X-ray structure analysis, obtained from a hanging-drop experiment, have been characterized. Both belong to space-group P22(1)2(1) with cell dimensions a = 39.2 A, b = 62.4 A, c = 84.6 A and a = 31.4 A, b = 74.8 A, c = 83.5 A, for forms I and II, respectively. Intensity data to 1.82 A have been collected for crystal form I on a CAD4 diffractometer, and initial phases have been obtained by molecular replacement methods. The conventional R-factor after two rounds of model building and subsequent refinement is 0.25 for data between 6.0 and 2.0 A. So far no water molecules have been included in the model.  相似文献

Nucleoside diphosphate (NDP) kinase catalyzes the transfer of the gamma-phosphate from a nucleoside triphosphate to a nucleoside diphosphate. Human and rodent forms of this enzyme have been shown to be suppressors of metastasis. Crystals that diffract X-rays to high resolution have been obtained for the recombinant Myxococcus xanthus NDP kinase expressed in and purified from Escherichia coli. Two crystal forms have been obtained. Both forms are orthorhombic, space group I222 (or I2(1)2(1)2(1)) with a = 267.1 A, b = 74.0 A and c = 75.1 A for form I and a = 53.5 A, b = 74.0 A and c = 75.1 A for form II. Form I appears to have five molecules in the asymmetric unit approximately related to each other by a translation of 0.2 along the a axis. Diffraction data have been recorded to 1.9 A for form I and to 2.2 A for form II.  相似文献

The type II restriction endonuclease EcoRV purified from a genetically engineered, overproducing strain has been crystallized. Four crystal forms all obtained by precipitation with polyethylene glycol 4000 have been characterized. Two of these are suitable for high resolution structure analysis. Both are orthorhombic, have space group P2(1)2(1)2(1) and have similar unit cell dimensions of a = 58.2 A, b = 71.7 A, c = 130.6 A (form A) and a = 59.9 A, b = 74.5 A, c = 121.8 A (form B). They diffract to about 2A resolution and appear to have one dimer of 2 X 29,000 daltons in the asymmetric unit.  相似文献

Human apo-lactoferrin in both native and deglycosylated forms has been purified, and crystals obtained by dialysis against low ionic strength buffer solutions. The crystals of native apo-lactoferrin are orthorhombic, space group P2(1)2(1)2(1) with cell dimensions a = 222.0 A, b = 115.6 A, c = 77.8 A and have two protein molecules per asymmetric unit. Two crystal forms of deglycosylated apo-lactoferrin have been obtained. One is orthorhombic, space group P2(1)2(1)2(1), with cell dimensions a = 152.1 A, b = 94.6 A, c = 55.8 A. The second is tetragonal, space group I4, with cell dimensions a = b = 189.4 A, c = 55.1 A. Both of the latter have only one molecule per asymmetric unit, and are suitable for high-resolution X-ray structure analysis.  相似文献

Crystals of recombinant wild-type antichymotrypsin have been prepared by the method of vapor diffusion with polyethylene glycol 4000 as a precipitant at pH 5.7. Two crystal forms are observed. One form belongs to tetragonal space group P4(3)2(1)2 (or P4(1)2(1)2) and has unit cell dimensions a = b = 126 A, c = 243 A, with two molecules in the asymmetric unit. The other crystal form belongs to orthorhombic space group P2(1)2(1)2(1) and has unit cell parameters of a = 73 A, b = 78 A and c = 80 A, with one molecular in the asymmetric unit. Diffraction intensity measurements have been made on the tetragonal crystal form to a limiting resolution of 4.1 A, and reflections have been observed on X-ray still photographs to a limiting resolution of 2.5 A for the orthorhombic form. An activity assay of redissolved tetragonal form crystals indicates that the uncleaved, functional serpin has been crystallized.  相似文献

A monoclonal antibody of the subclass IgG2a specific for canine lymphoma cells has been crystallized by vapor diffusion from polyethylene glycol 8000. the crystals, which occasionally measure nearly a millimeter on edge, have been examined by X-ray diffraction. The crystals are of triclinic space group P1 with unit cell parameters of a = 66.39 A, b = 77.34 A, c = 101.42 A, alpha = 87.60 degrees, beta = 92.55 degrees, gamma = 97.54 degrees and cell volume of V = 4.84 x 10(5) A3. There is one entire antibody molecule as the asymmetric unit of the crystals. Three-dimensional X-ray diffraction data have been collected to 2.8 A resolution and a self rotation function calculation shows a pronounced peak indicating at least an approximate non-crystallographic dyad axis.  相似文献

Thaumatin is a plant protein that in the mature form contains 8 disulfide bonds and 207 amino acids. Several forms of this protein occur naturally and each elicits an intense sweetness sensation when tasted in microgram quantities. The two major forms of thaumatin are easily separable by ion exchange chromatography. Crystals of the two proteins (designated here A and B) have been grown by vapor equilibration from solutions containing polyethylene glycol and examined by X-ray diffraction. The thaumatin A crystals are of space group P2(1)2(1)2(1) with a = 44.3 A, b = 63.7 A and c = 72.7 A. The crystals of thaumatin B are of space group C2 with a = 117.7 A, b = 44.9 A, and c = 38.0 A and beta = 94.0 degrees. Both crystals diffract to well beyond 2.3 A and appear suitable for high resolution structure analysis. Four heavy atom derivatives of thaumatin B have been generated and diffraction data to 4 A resolution have been collected. This work is designed to provide a basis for studying the 3-dimensional structure of more than 100 genetically generated thaumatin derivatives, several of which show enhanced stability and improved taste characteristics.  相似文献

Ace is a collagen-binding bacterial cell surface adhesin from Enterococcus faecalis. The collagen-binding domain of Ace (termed Ace40) and its truncated form Ace19 have been crystallized by the vapor-diffusion hanging-drop method. Ace19 was crystallized in two different crystal forms. A complete 1.65 A data set has been collected on the orthorhombic crystal form with unit cell parameters a=38.43 b=48.91 and c=83.73 A. Ace40 was crystallized in the trigonal space group P3(1)21 or P3(2)21 with unit cell parameters a=b=80.24, c=105.91 A; alpha=beta=90 and gamma=120 degrees. A full set of X-ray diffraction data was collected to 2.5 A. Three heavy atom derivative data sets have been successfully obtained for Ace19 crystals and structural analysis is in progress.  相似文献

Crystals of the acyl carrier protein of Escherichia coli have been grown and analyzed by X-ray diffraction. The crystals grow in space group C2 with unit cell dimensions a = 46.8 A, b = 52.1 A, c = 47.3 A and beta = 93.2 degrees. The Type C staphylococcal enterotoxin produced by Staphylococcus aureus strain FRI-909 has been crystallized using a combination of two precipitants, ammonium sulfate and polyethylene glycol 400, with the addition of small amounts of detergent. Two related crystal forms have been obtained, one triclinic, and one tetragonal, both with one toxin molecule per asymmetric unit. These crystals are stable for at least 75 hr in the X-ray beam and diffract to at least 2.2 and 2.6 A, respectively. The triclinic crystals have unit cell parameters a = 38.5 A, b = 43.7 A, c = 36.9 A, and interaxial angles alpha = 99.9 degrees, beta = 95.8 degrees, and gamma = 98.5 degrees. The tetragonal crystals are of space group P4(1)22 with unit cell parameters a = 43.4 A and c = 278.0 A.  相似文献
