分析基因型、环境及其互作对小麦品质性状的影响。对两年度3个试点13个小麦品种的16个主要品质性状进行分析。结果表明:品质性状在不同小麦品种及不同试点间皆存在显著差异,其中硬度、PPO活性、AWRC、稀懈值的变异系数较大;膨胀势、最终粘度、峰值粘度的变异系数较小;糊化温度、碱性水保持力、峰值时间、蛋白质含量、湿面筋含量的环境效应大于基因型效应;SDS沉降值、PPO活性、黄色素含量、膨胀势、峰值粘度、保持粘度、稀懈值、最终粘度、回升值、Zeleng沉降值、硬度的基因型效应大于环境效应,保持粘度这一品质性状基本上不受环境影响,比较稳定。  相似文献

本试验以普通大麦(Hordeum vulgare L.)品种单核晚期的花药为材料,研究了他们在液体培养过程中海藻酸钠的作用。结果表明:在供试的4个海藻酸钠浓度(1、2、3、6%)中,以浓度为2-3%的效果最佳。当他们代替20-30%的Ficoll时,无论是用BAC3培养基,还是用MS培养基,培养基中加NAA,还是家2,4-D,都能诱导出愈伤组织,但以每升培养基加2mg NAA和1 mg 6-BA的效果最佳,在含这种激素的培养基上,一些基因型的出愈率相当高,供试基因型都在100%以上,而且有些花粉粒能直接发育成能正常发芽出苗的胚状体。Barley anthers at the late uninucleate stage were cultured in order to study the use of sodium alginate in liquidmedium.The results showed that, the effect of 2-3% (W/V) sodium alginate was best in 4 concentration studied (1%,2%,3%and 6%). When the 2-3% sodium alginate was substituted for 20-30% ficoll, the calli could be induced from anthers of all genotypes whether on BAC3 medium or MS medium and whether adding NAA or 2,4-Din medium.2mg/L NAA+1mg/L 6-BA added to medium could give better results and on the medium with these hormones the callus rate of all genotypes were above 100%. Plant hormones play many important roles in plant growth and development. We previously tried to control barley (Hordeum vulgare L.) flowering in ear cultures treated with plant hormones and related compounds, and found that anther extrusion from florets was reduced by treatment with 100 ppm methyl jasmonate (MJA). In the present study, we show that 10 ppm MJA also inhibits anther extrusion in barley ear cultures. Spraying field-grown barley ears with MJA clarified the efficacy of this chemical under field conditions, but higher concentrations (100 to 1000 ppm MJA) were required. In addition, we investigated the activity of jasmonic acid (JA) analogues and cucurbic acid analogues in barley ear cultures. Some esters of JA showed similar effects to that of MJA, but effects of 9,10-dihydrojasmonic acid (DJA) and cucurbic acid derivatives were lower than that of MJA. In light of these data, we discuss the structural requirements for increased inhibition of barley anther extrusion.  相似文献

The effects of mannitol pretreatment on androgenesis of barley were systematically studied in comparison with that of cold pretreatment and control. The results showed that mannitol pretreatment could significantly increase the frequency of pollen survival reaching 19.0% on the eighth day, while in cold pretreatment and control they were 8.4% and 6.6 %, respectively. Mannitol pretreatment could also improve the quality of pollen and inhibit starch production from microspore, which were quite advantageous to microspore division and development. The developing period was shortened 2--3 days as compared with cold pretreatment and control. The major developmental pathways of androgenesis after mannitol pretreatment were the equal division (B pathway). In addition, the majority of microspore nuclei were diploids. On the contrary, the major microspores pretreated with low temperature had fewer chromosomes than with mannitol pretreatment, the microspore nuclei were haploids.  相似文献

采用SSR技术对沪95-2639和91冬27携带的两个新的矮秆基因进行了分子标记.在大麦4H染色体的长臂上,发现SSR标记位点HVM67同时与这两个新的矮秆基因连锁,距91冬27的较近,约10.0cM,离沪95-2639的较远,为23.3cM.初步绘制出大麦4H染色体上矮秆基因与SSR标记位点的遗传连锁图谱.  相似文献

本文就大麦黄花叶病(BYMV)的抗性进行了遗传分析。研究表明,在本研究中,大麦黄花叶病抗性表现为多基因控制的数量性状,符合“加性-显性”遗传模型,但主要受加性效应控制。回归分析与平均显性度((H_1/D)~(1/2))测定均表明为部分显性。且控制BYMV的严重度的显性基因数约为3—6组。遗传力估算较高。最后就实验结果对BYMV抗性育种进行了初步分析讨论。  相似文献

 Genetic transformation using shoot meristematic cultures (SMCs) derived from germinated seedlings is established in commercial varieties of oat cv 'Garry' and barley cv 'Harrington'. Six-month-old SMCs of oat were induced on MPM and bombarded with bar and uidA; 9-month-old SMCs of barley were induced on an improved medium (MPM-MC) containing maltose and high levels of copper and bombarded with bar/nptII and uidA. After 3–4 months on selection, seven independent transgenic lines of oat were obtained, two lines of barley. All transgenic lines produced T0 plants; five lines of oat and one line of barley were self-fertile, and the other barley line produced T1 seed when out-crossed. Both Mendelian and non-Mendelian segregation ratios of transgene expression were observed in T1 and T2 progeny of transgenic oat. Normal as well as low physical transmission of the transgenes was also seen in T1 and T2 progeny of oat. The bar-containing line of barley showed stable transgene expression in all of the T1 and T2 progeny tested. Received: 4 January 1999 / Accepted: 14 January 1999  相似文献

导入TrxS基因后,转基因大麦籽粒的硫氧还蛋白h活性明显提高;淀粉酶活性也明显提高,其中α-淀粉酶活性在开花后30d提高了3倍以上,随着籽粒的发育,转基因对α-淀粉酶活性影响作用减少,对β-淀粉酶活性的影响有同样的趋势;转基因大麦种子发芽势明显提高。说明TrxS基因有望改善啤酒大麦的制麦特性和品质特性。  相似文献

Summary The inheritance of the ability of barley anthers to produce microspore-derived callus in vitro was investigated. The genotypes selected were the two spring cultivars Dissa (D) and Sabarlis (S), the two F1 hybrids (DxS, SxD), the two backcross generations [Dx(DxS), Sx(DxS)], and an F2 generation derived from DxS. From a number of individuals of each generation, the first five spikes were harvested sequentially and after pre-treatment the anthers were removed and placed in culture. Cultures were scored for microspore callus production and plantlet differentiation. Although Dissa gave a significantly higher level of callus production than Sabarlis, the overall frequencies of green and albino plant production were higher from Sabarlis. There was no significant difference between reciprocal F1 hybrids. Analysis of variance revealed significant differences in response between the spikes sampled from the plants. This was the major source of variation in the experiment. Spike to spike variation also appeared to be a heritable character.  相似文献

One hundred and forty six barley doubled-haploid lines (DH lines) were tested for variation in grain yield, yield components, plant height, and heading date after artificial infection with a German isolate of barley yellow dwarf virus (BYDV-PAV-Braunschweig). Of these 146 lines 76 were derived from the cross of the barley yellow dwarf virus (BYDV) tolerant cultivar ’Post’ to cv ’Vixen’ (Ryd2) and 70 from the cross of Post to cv ’Nixe’. Phenotypic measurements were gathered on both non-infected plants and plants artificially inoculated with BYDV-PAV by viruliferous aphids in pot and field experiments for three years at two locations. For all traits a continuous variation was observed suggesting a quantitative mode of inheritance for tolerance against BYDV-PAV. Using skeleton maps constructed using SSRs, AFLPs and RAPDs, two QTLs for relative grain yield per plant after BYDV infection, explaining about 47% of the phenotypic variance, were identified in Post × Vixen at the telomeric region of chromosome 2HL and at a region containing the Ryd2 gene on chromosome 3HL. In Post × Nixe, a QTL was found in exactly the same chromosome 2HL marker interval. In this cross, additional QTL were mapped on chromosomes 7H and 4H and together these explained about 40% of the phenotypic variance. QTL for effects of BYDV infection on yield components, plant height, and heading date generally mapped to the same marker intervals, or in the vicinity of the QTL for relative grain yield, on chromosomes 2HL and 3HL, suggesting that these regions are of special importance for tolerance to the Braunschweig isolate of BYDV-PAV.
