The effects of stress produced by noxious cutaneous stimulation on the concentration of 18 amino acids in the hemolymph of Aplysia californica were examined. The concentration of alanine and glutamate increased in response to stress and remained elevated for at least 6 h. The total amino acid concentration in the hemolymph did not change significantly in response to stress. It is concluded that stress evokes an increase in circulating alanine and glutamate levels. Molting in crayfish, essential for growth is preceded by a decreasein total organic content during premolt. Mineral, as calcium,is conserved in gastroliths after mobilization from the exoskeleton.Crustecdysone treatment results in gastrolith formation by intactcrayfish in intermolt, state C, but does not influence the rateof gastrolith formation in eyestalkless animals. Moreover, therate of gastrolith formation is not increased at higher hormonedoses; conversely, less dense, abnormal gastrohths result, whileapolysis is evident by 48 to 60 hours after treatment. It appearsthat crustecdysone promotes reabsorption of the organic matrixsince treatment with this hormone does not lead to a measureabledecrease in calcium content of shell based on dry weight. Premolt crayfish normally have a higher amino acid pool in alltissues than do those in intermolt, and eyestalk removal fromintermolt crayfish results in an increase in the free-aminoacid level of all tissues by 24 hours; crustecdysone treatmentresults in a significant increase in muscle amino acids by 12hours after a hormone dose of 5 µg/g body weight. In thecontext of integumentary growth and protein synthesis low dosesof crustecdysone (1 µg/g) increase the in vivo incorporationof amino acid into hypodermis, but not hepatopancreas, by 24hours, and the increase is significant by 36 hours; a higherdose (5 µg) does not change this rate of incorporation.Selective increase in hypodermis protein is not evident in acidphosphatase levels from 12 to 48 hours, but respiration of thistissue is significantly elevated within two hours after hormoneinjection.  相似文献

The effects of contaminated river water on the filtration rate of zebra mussels from a clean reference site were studied. After a 48-h exposure period to filtered water from the rivers Rhine, Meuse and Amstel (The Netherlands), the filtration rate was measured. It was demonstrated that water from contaminated locations inhibited the filtration rate. Inhibition was higher during low water levels in the rivers Rhine and Meuse than during high water levels, suggesting that contaminants are diluted during high water levels. It is concluded that the shortterm filtration assay with D. polymorpha can be used for assessing water quality.  相似文献

Changes in the free amino acid levels of extracts from body whole homogenates of 3rd instar boll weevilAnthonomus grandis Boheman (Curculionidae) larvae which had been stung once by females of the ectoparasitoidBracon mellitor Say (Braconidae) were determined. Stung larvae exhibited an initial rise in free amino acid levels within 6 min after the venom was injected followed by a decline within 2h. A second free amino acid peak occurred 4 days after initial sting. Observed increases in free amino acid levels were always followed by synchronized decreases in total soluble proteins. The possible nutritional regulatory effect of ectoparasitoid venom on the physiology of the boll weevil is discussed.  相似文献

l-Dopa therapy has been suggested as effective in the reversal of hepatic coma both in humans and in animals. Beneficial effects have been reported also in chronic hepatic encephalopathy. There are many possible mechanisms through whichl-dopa could ameliorate this pathological state. The present study was carried out to clarify whether thel-dopa effect could be mediated through an improvement of the brain neutral amino acid patterns, since it competes for the same transport carrier at the blood-brain barrier. A first group of rats was orally administeredl-dopa (10 mg/100 g body weight daily) for 1 month following portocaval anastomosis. A second group was intraperitoneally injected (1.5 mg/100 g body weight daily) for 1 week, a month after portocaval shunt. Amino acid levels were determined in plasma and in four cerebral regions. No beneficial effects were observed clinically (in general condition, body weight, or hypertonic posture) in rats receivingl-dopa compared to controls. The large increase of tyrosine, phenylalanine, tryptophan, histidine, and glutamine that occurs in the cerebral tissue after portocaval shunt was also not affected byl-dopa administrations. In conclusion, in this experimental condition we had no clinical improvement in shunted animals receivingl-dopa. Moreover, this compound did not seem to influence the pathological increase of aromatic amino acids in the brain, which is considered to play an important role in hepatic encephalopathy.  相似文献

The symbiotic bacteria Buchnera contribute to the nutrition of pea aphids, Acyrthosiphon pisum, through the provision of essential amino acids which are lacking in the diet. However, chemically defined diets, containing nutritionally adequate amounts of essential amino acids, fail to rescue aposymbiotic aphids, in which the bacteria have been disrupted with antibiotics. In this study the injection of a mixture of essential amino acids into the haemocoel of aposymbiotic aphids was shown to alleviate, at least partially, the impact of symbiont loss. Specifically, the total amino acid content in the tissues of aposymbiotic aphids was reduced by approximately 40% to levels comparable with symbiotic insects, and there was a 1.7-fold increase in the number of embryos, suggesting that the availability of essential amino acids promotes aphid protein synthesis by rejuvenating the free amino acid pool of aposymbiotic aphids. In addition, a similar effect on the total amino acid content was observed when phenylalanine alone, but not glutamine, lysine or tryptophan, was injected into the haemocoel of aposymbiotic aphids, and there was also a significant increase in the number of embryos following injection of phenylalanine or tryptophan alone. The impact of amino acid injection on the embryo complement of aposymbiotic aphids was limited to an increase in the number of embryos, with no increase in basal embryo size. It is proposed that older embryos may rely on their own complement of symbiotic bacteria for essential amino acid provisioning. Taken together, the data highlight the importance of bacterial provisioning of essential amino acids, particularly the aromatic amino acids, in the intact symbiosis.  相似文献

利用外源镉添加的袋栽试验,系统研究了镉对姬松茸J37和J1两菌株农艺性状、子实体产量、氨基酸和镉含量的影响,以求为低富集镉的姬松茸品种选育和安全栽培提供科学依据。结果表明外源镉添加对姬松茸J37和J1品种子实体的农艺性状的影响各异,两菌株单个子实体重量、子实体高度、菌盖直径、菌盖厚度和菌柄直径等均与外源镉浓度具有显著的负相关性,其中子实体重量与外源镉浓度间的相关性最大;15 mg/kg镉水平是两菌株农艺性状发生显著变化的敏感点。不同镉水平下J37菌株单个子实体重量比J1菌株高10.9%—36.6%,表现为较强镉耐受性。姬松茸J37与J1菌株子实体产量、氨基酸和镉含量均随着潮次的增加而降低,子实体产量和氨基酸含量随着外源镉水平增加而降低,而镉含量随外源镉水平增加而增加。与外源镉0mg/kg处理相比,当外源镉水平达15 mg/kg时,J1和J37子实体总产量分别下降23.4%和13.4%以上,J1和J37子实体镉含量分别提高51.5%、13.7%以上,且J1子实体镉含量已达到食品卫生标准的临界值。当外源镉水平达到35 mg/kg时,J1和J37子实体产量分别下降45%、32%以上,J1和J...  相似文献

Bivalves are filter-feeders that can accumulate large numbers of bacteria, in particular Vibrio species; these can persist within bivalve tissues largely depending on their sensitivity to the hemolymph bactericidal activity. In this work, functional parameters of the hemolymph of Mytilus galloprovincialis were evaluated in response to in vivo challenge with different bacteria (Gram(−) Vibrio anguillarum and V. splendidus, Gram(+) Micrococcus lysodeikticus). Mussels were injected with heat-killed bacteria or PBS-NaCl (controls) and hemolymph sampled from 3 to 48 h post-injection (p.i.). In hemocytes, all bacteria induced significant lysosomal membrane destabilisation (LMS) from 3 h p.i. with V. splendidus > V. anguillarum > M. lysodeikticus. LMS showed recovery for both M. lysodeikticus and V. anguillarum, whereas a further time-dependent decrease was observed for V. splendidus. Bacterial challenge also induced a rapid (from 3 h p.i.) and significant increase in serum lysozyme activity; the effect was persistent with M. lysodeikticus and transient for the two Vibrio species. In order to evaluate whether in vivo challenge may affect the subsequent capacity of hemolymph to kill bacteria, the bactericidal activity was tested in an in vitro assay towards E. coli. At 48 h. p.i. hemolymph samples from V. anguillarum-injected mussels showed a significant increase in E. coli killing (+ 35% with respect to controls); a smaller effect was observed with V. splendidus-injected mussels (+ 16%), whereas M. lysodeikticus was ineffective. Moreover, hemolymph from V. anguillarum-injected mussels showed an in vitro bactericidal activity towards V. anguillarum 2-folds higher than that of controls. Changes in total hemocyte counts (THC) and in hemocyte populations were evaluated by Flow cytometry at 6 and 48 h p.i., indicating a decrease in THC followed by recovery with all bacteria. Moreover, at 6 h p.i. a general decrease in the percentage of granulocytes was observed (V. splendidus > V. anguillarum > M. lysodeikticus), followed by complete and partial recovery with M. lysodeikticus and V. anguillarum, respectively, but not with V. splendidus. The results demonstrate the existence of differential functional immune responses in M. galloprovincialis to different bacteria.  相似文献

We investigated the taste responses to fructose and tannic acid compounds among 6 western gorillas (Gorilla gorilla gorilla) at the San Francisco Zoo. We presented the subjects with 12 reference concentrations of 3–300 mM fructose, paired with tap water in paired-choice experimental trials. We subsequently presented them with tap water paired with 8 dilute concentrations of tannic acid (0.25–6 mM) dissolved in 100 mM fructose solutions and 8 concentrations (0.25–6 mM) dissolved in 300 mM fructose solutions. The gorillas exhibited a broadly similar preference threshold for fructose (50 mM) to those of other nonhuman primate and human samples. The gorillas tolerated moderate levels of tanninc acid, especially when presented in a very sweet package. The depressing effect of tannic acid on the ingestion of fructose solutions appeared to increase progressively with tannin concentration and was lower as fructose concentration increased. The inhibition threshold for tannic acid solutions was reached at 4 mM tannic acid in 100 mM fructose solution, and at 6 mM tannic acid in 300 mM fructose solutions. These results suggest that gorillas use sweetness as a criterion for food selection and regard those with moderate levels of tannins as palatable. Our findings corroberate food preference studies and nutritional analyses of wild gorilla foods indicating that they prefer sugary foods and readily consume ones containing moderate levels of tannins. Taste responses may facilitate a flexible frugivorous/folivorous diet among gorillas.  相似文献

Summary The aims of the present study were to assess the changes of individual plasma amino acid levels in relation (1) to the severity of liver damage and (2) to the process of liver recovery. Acute liver injury was induced by an intragastric administration of CCl4 diluted in olive oil in doses of 2, 4 and /or 6 g of CCl4 per kg b.w. The control rats received olive oil only. Animals were sacrificed at 16, 24, 48 and 96 hours after treatment. The severity of liver injury was assessed by histological examination, by changes in ALT and AST in the blood plasma and by changes in liver weight. Statistical analysis was carried by ANOVA, p < 0.05 was considered significant. The Spearman rank correlation coefficient was used as a measure of the degree of linear relationship between variable and dose. In the period of the development of acute liver damage, i.e. at 16 and 24 hours after treatment, an increase in blood plasma amino acid levels and positive correlations with the dose of CCl4 were observed for most individual amino acids. The only exception was arginine which decreased in a dose dependent manner. At a phase of liver recovery, i.e. at 48 and 96 hours after CCl4 treatment, the concentrations of some individual amino acids decreased below the control values. The negative correlation with the dose of CCl4 occurred for taurine and isoleucine (at 48 hours) and taurine, threonine, valine, methionine, isoleucine and leucine (at 96 hours).  相似文献

Batch cultures of the toxigenic diatomNitzschia pungens Grunow f.multiseries Hasle were fed to blue mussels (Mytilus edulis) and deep sea Atlantic scallops (Placopecten magellanicus) to elucidate conditions under which domoic acid (DA) was accumulated and excreted (depurated). Mussels accumulated the toxin to a maximum level of 13 g g-1, at rates of 0.21 to 3.7 g h-1 g-1, dry weight. Accumulation efficiency (the proportion of accumulated DA to estimated net uptake) ranged from 1–5%. The highest filtration rate of 1.71 h-1 occurred at concentrations of 4–8 × 106 Nitzschia cells 1-1 with no formation of pseudofeces. Depuration rates between fed and starved mussels over a 2 h test period were the same. The depuration rate of domoic acid was about 17% d-1 and did not account for the low uptake efficiencies, so it is suggested that most of the DA is lost from mussels in the solution during the feeding process. Domoic acid accumulation in mussels was dependent on the amount of toxin available, which in turn was a function of the density and growth phase of theNitzschia population. Changes in filtration rate withNitzschia concentration and depuration rate with time can account for the DA levels of mussels collected during toxic episodes in Cardigan Bay, Prince Edward Island, Canada in 1988 and 1989.Scallops accumulated DA (0.39–1.3 g h-1 g-1, more slowly than mussels, however, accumulation efficiencies ranged from 5–100%. Filtration rates remained relatively low and constant at 0.081 h-1. Scallops retained domoic acid longer than mussels, a fact which must be considered in the marketing of whole scallops for human consumption.  相似文献

An experiment was conducted to follow the fate of the cyanobacterial toxin, nodularin, produced by Nodularia spumigena through ingestion by Mytilus edulis and re-ingestion of faecal material (coprophagy). Mussels were fed with cultures of N. spumigena, and the faeces that were produced were fed to other mussels not previously exposed to N. spumigena. Concentrations of nodularin were measured in the food (N. spumigena), the mussels and in the faeces in order to make a toxin budget. High concentrations of nodularin were found in the mussels and their faeces after 48 h incubation with N. spumigena. When the toxic faeces were fed to new mussels, the toxin content of faeces was reduced from 95 μg nod g−1 dry weight (DW) to 1 μg nod g−1 DW through the process of coprophagy. Hence, when toxic faeces were fed to mussels, the nodularin concentration of the resulting faecal material was reduced by 99%. Pseudofaeces were produced when the mussels were grazing on N. spumigena, but not when grazing on faeces. The pseudofaeces contained high concentrations of nodularin and apparently intact N. spumigena cells. However, these cells were growth-inhibited and their potential contribution to seeding a bloom is probably limited. Our data indicate that a large fraction of ingested nodularin in M. edulis is egested with the faeces, and that the concentration of nodularin in the faeces is reduced when faeces are re-ingested.  相似文献

An 8‐week feeding trial was conducted to determine the effects of dietary arginine levels on growth performance and body composition of juvenile grouper Epinephelus coioides (initial weight: 16.10 ± 0.26 g per fish) in 500‐L indoor flow‐through circular fiberglass tanks. Diets contained six levels of l ‐arginine ranging from 1.78% to 3.31% of dry diet in 0.30% increments. The diets, in which 25% crude protein came from fishmeal and soybean protein concentrate, and 23% from crystalline amino acids, were formulated to simulate the amino acid profile of 48% whole chicken egg protein except for arginine. Each diet was assigned to three tanks (20 fish per tank) in a completely randomized design. Grouper were fed to apparent satiation (about 5% body weight per day at the initial 1–4 weeks and 3% body weight per day in the following 5–8 weeks) twice daily on weekdays and once daily on weekends. Weight gain (WG) and specific growth rate (SGR) showed an incremental trend with increasing dietary arginine levels from 1.78% to 2.72% and remained thereafter relatively constant. Productive protein value (PPV) was poorest in fish fed the lowest arginine diet (P < 0.05) and showed no significant differences among other treatments (P > 0.05). Dietary arginine contents significantly influenced body composition. With the increase in dietary arginine, contents of total essential amino acids (EAAs), total non‐EAAs and total amino acids of whole body significantly increased (P < 0.05). The retention of dietary EAA in whole body of juvenile grouper was significantly influenced by dietary arginine. Arginine retention increased with increasing dietary arginine levels from 1.78% to 2.72%, then declined above the level of 2.7% (P < 0.05). Based on broken‐line regression analysis of WG against dietary arginine levels, the dietary arginine requirement of juvenile grouper E. coioides was determined as 2.7% of the diet (corresponding to 5.5% of dietary protein).  相似文献

Hospital tap water is a recognised source of Pseudomonas aeruginosa. UK guidance documents recommend measures to control/minimise the risk of P. aeruginosa in augmented care units but these are based on limited scientific evidence. An experimental water distribution system was designed to investigate colonisation of hospital tap components. P. aeruginosa was injected into 27 individual tap ‘assemblies’. Taps were subsequently flushed twice daily and contamination levels monitored over two years. Tap assemblies were systematically dismantled and assessed microbiologically and the effect of removing potentially contaminated components was determined. P. aeruginosa was repeatedly recovered from the tap water at levels above the augmented care alert level. The organism was recovered from all dismantled solenoid valves with colonisation of the ethylene propylene diene monomer (EPDM) diaphragm confirmed by microscopy. Removing the solenoid valves reduced P. aeruginosa counts in the water to below detectable levels. This effect was immediate and sustained, implicating the solenoid diaphragm as the primary contamination source.  相似文献

1. Portacaval shunting in rats results in several metabolic alterations similar to those seen in patients with hepatic encephalopathy. The characteristic changes include: (a) diminution of cerebral function; (b) raised plasma ammonia and brain glutamine levels; (c) increased neutral amino acid transport across the blood-brain barrier; (d) altered brain and plasma amino acid levels; and (e) changes in brain neurotransmitter content. The aetiology of these abnormalities remains unknown. 2. To study the degree to which ammonia could be responsible, rats were made hyperammonaemic by administering 40 units of urease/kg body weight every 12 h and killing the rats 48 h after the first injection. 3. The changes observed in the urease-treated rats were: (a) whole-brain glucose use was significantly depressed, whereas the levels of high-energy phosphates remained unchanged; (b) the permeability of the blood-brain to barrier to two large neutral amino acids, tryptophan and leucine, was increased; (c) blood-brain barrier integrity was maintained, as indicated by the unchanged permeability-to-surface-area product for acetate; (d) plasma and brain amino acid concentrations were altered; and (e) dopamine, 5-hydroxytryptamine (serotonin) and noradrenaline levels in brain were unchanged, but 5-hydroxyindoleacetic acid (5-HIAA), a metabolite of 5-hydroxytryptamine, was elevated. 4. The depressed brain glucose use, increased tryptophan permeability-to-surface-area product, elevated brain tryptophan content and rise in the level of cerebral 5-HIAA were closely correlated with the observed rise in brain glutamine content. 5. These results suggest that many of the metabolic alterations seen in rats with portacaval shunts could be due to elevated ammonia levels. Furthermore, the synthesis or accumulation of glutamine may be closely linked to cerebral dysfunction in hyperammonaemia.  相似文献

The protein content in the dry body weight of the starfish Odontaster validus is 30%. The composition of amino acids is characterized by a very high level of glycine (27%). Glutamic acid (Glu) in a concentration of 10 mM/liter in seawater causes an increase in the metabolic rate of starved O. validus. As the period of starvation increases, this reaction is enhanced. A two-hour exposure to kynurenic acid (Kyn) in a concentration of 0.1 mM/liter blocks the reaction to glutamic acid.  相似文献

Small scale spatial patterns of abundance, growth and condition of the mussel Mytilus edulis on sub-littoral boulders (approx. 1–2 m high) were investigated at the island of Askö in the northern Baltic proper. The effect of side (exposed/sheltered with respect to wave action and sunlight) of boulder and position (up/down) on boulder was investigated. A large spatial variability in abundance of M. edulis between boulders and between various sites within boulders were found. The highest numbers of mussels were found on the wave exposed side, near the top of boulders. Shell growth was favoured by a sheltered side and a down position. The body condition (meat weight/shell weight) of mussels was, on the other hand, affected only by position, the condition of mussels being better at the up position. Consequently, there seems to be temporal differences in the condition for growth within a spatial position. The body condition of the mussels was best near the top of boulders in the spring, but long-term shell growth was favoured by a sheltered side and a down position. This may reflect changes in the composition and availability of food during the year with phytoplankton as the major food source during the spring bloom and resuspension of benthic production and detritus as relatively more important during the rest of the year.  相似文献

Water stress induced an increase in endogenous concentrations of ABA in Lavandula stoechas L. plants to 13100 pmol ABA g–1 FW, which may contribute to the maintenance of water relations between the second and the third day of water stress treatment. After the third day, a sharp decrease in ABA levels was observed to 2630 pmol ABA g–1 FW, together with a decrease in water content and water potential and a loss of plant response to water stress. Water deficit did not induce an increase in endogenous ABA concentration, which remained at 514 pmol ABA g–1 FW in Rosmarinus officinalis L., which is more sclerophyllous than L. stoechas. Nevertheless, the relative water content of Rosmarinus officinalis L. after seven days of water stress decreased more than 40% and reached values of –3.2 MPa. R. officinalis showed lower levels of ABA, but significantly higher levels of IAA and ZR than L. stoechas (4 times and 6 times respectively in well watered-plants).
