A novel actinomycete strain (PK1) was isolated from soil in Khon Kaen Province, Thailand, and was capable of inhibiting the cyanobacterium Microcystis aeruginosa. The isolate PK1 was identified as Streptomyces aurantiogriseus based on an analysis of biochemical and morphological characteristics and 16S rDNA sequence. The algicidal activity of PK1 against M. aeruginosa depended on the growth phase of PK1, but not on the cyanobacterial growth phase. Stationary growth phase cultures of the strain PK1 exhibited the highest anti-Microcystis activity when co-cultivated with M. aeruginosa. Complete growth inhibition was observed after 8 days of co-cultivation in liquid culture medium. The algicidal compounds were extracted from PK1 with ethyl acetate and then purified by silica gel column chromatography. These partially purified compounds demonstrated algicidal activity against M. aeruginosa, suggesting that the strain PK1 provides a potential source of extracellular compounds for the control of M. aeruginosa bloom. This is the first report of anti-cyanobacterial activity from the soil actinomycete S. aurantiogriseus.  相似文献

As part of efforts to enhance the strategies explored to eliminate the adverse impacts of cyanobacterial blooms, we isolated an algicidal bacterium, J4, from Lake Taihu. Analysis of 16S rDNA sequence revealed that strain J4 belonged to the genus Brevundimonas. Bacterium J4 exhibited algicidal activity mainly through excretion of extracellular algicidal compounds that were further extracted with methanol and purified by silica gel chromatography and high performance liquid chromatography (HPLC). The compounds showed thermal stability, strong polarity and water solubility in J4 cultures. Study on the algicidal activity of J4 against two dominant cyanobacterial bloom-forming species in Lake Taihu showed that J4 exhibited lower algicidal rate against Synechococcus sp. BN60 (48.6%, t = 6 days) than against Microcystis aeruginosa 9110 (91.8%, t = 6 days). Additionally, rapid reduction in cell density of J4 was observed in co-cultures of Synechococcus sp. BN60 and bacterium J4 but not observed in co-cultures of M. aeruginosa 9110 and bacterium J4 during algicidal process, which was the main reason why the algicidal rate of J4 against BN60 was lower than against 9110. The reduction in cell density of J4 resulted from inducible production of antimicrobial-like compound secreted by Synechococcus sp. BN60 in co-cultures of Synechococcus sp. BN60 and bacterium J4, which reflected a kind of chemical defense from cyanobacteria (BN60) against algicidal bacteria (J4). However, M. aeruginosa 9110 had no chemical defense against J4, suggesting that whether cyanobacterial chemical defense occurs or not between cyanobacteria and algicidal bacteria depends on specific cyanobacteria–algicidal bacteria pairs. These results show that not only one-sided algicidal effect but also two-sided reciprocal inhibition interactions exist between algicidal bacteria and cyanobacteria, indicating the complexity of cyanobacteria–algicidal bacteria interactions in Lake Taihu and the need to take the cyanobacterial defensive responses into consideration when assessing potential use of algicidal bacteria.  相似文献

As part of efforts to enhance the strategies employed to manage and mitigate algal blooms and their adverse effects, algicidal bacteria have shown promise as potential suppressors of these events. Nine strains of bacteria algicidal against the toxic dinoflagellate, Alexandrium tamarense, were isolated from the East Sea area, China. Sequence analysis of 16S rDNA showed that all the algicidal bacteria belonged to the γ-proteobacteria subclass and the genera Pseudoalteromonas (strain SP31 and SP44), Alteromonas (strain DH12 and DH46), Idiomarina (strain SP96), Vibrio (strain DH47 and DH51) and Halomonas (strain DH74 and DH77). To assess the algicidal mode of these algicidal bacteria, bacterial cells and the filtrate from bacterial cultures were inoculated into A. tamarense cultures, and fluorescein diacetate vital stain was applied to monitor the growth of the algal cells. The results showed that all the algicidal bacteria exhibited algicidal activity through an indirect attack since algicidal activity was only detected in cell free supernatants but not the bacterial cells. This is the first report of bacteria from the genus Idiomarina showing algicidal activity to the toxic dinoflagellate A. tamarense and these findings would increase our knowledge of bacterial–algal interactions and the role of bacteria during the population dynamics of HABs.  相似文献

The dynamics of cyanophage-like particles and algicidal bacteria that infect the bloom-forming cyanobacterium Microcystis aeruginosa was followed in a hyper-eutrophic pond from September 1998 to August 1999. The densities of M. aeruginosa ranged between 4.0 × 105 and 1.9 × 107 cells ml−1, whereas those of algicidal bacteria were between 4.0 and 5.1 × 102 plaque-forming units (PFU) ml−1 and those of cyanophage-like particles were between <5.0 × 102 and 7.1 × 103 PFU ml−1. A significant relationship was found between the densities of algicidal bacteria and M. aeruginosa (r = 0.81, n = 69, P < 0.001), suggesting that the dynamics of the algicidal bacteria may regulate the abundance of M. aeruginosa. Occasional peaks of density of cyanophage-like particles were detected in October, June, and August, when sharp declines in M. aeruginosa cell densities were also observed. The densities of cyanophage-like particles became undetectable when the abundance of M. aeruginosa was low, suggesting the density-dependent infection of M. aeruginosa by cyanophage-like particles. Thus, we suggest that infections of both algicidal bacteria and cyanophage-like particles are important biological agents that decompose blooms of M. aeruginosa in freshwater environments. Cyanobacterial blooms have become a serious problem in Lake Taihu during the last 20 years, and Microcystis aeruginosa and Synechococcus sp. are the two dominant species in cyanobacterial blooms of Lake Taihu. A freshwater bacterial strain, Shewanella sp. Lzh-2, with strong algicidal properties against harmful cyanobacteria was isolated from Lake Taihu. Two substances with algicidal activity secreted extracellularly by Shewanella sp. Lzh-2, S-2A and S-2B, were purified from the bacterial culture of strain Lzh-2 using ethyl acetate extraction, column chromatography, and high performance liquid chromatography (HPLC) in turn. The substances S-2A and S-2B were identified as hexahydropyrrolo[1,2-a]pyrazine-1,4-dione and 2, 3-indolinedione (isatin), respectively, based on liquid chromatography-mass spectrometry (LC-MS), gas chromatography-mass spectrometry (GC-MS), and hydrogen-nuclear magnetic resonance (H-NMR) analyses, making this the first report of their algicidal activity toward cyanobacteria. S-2A (hexahydropyrrolo[1,2-a]pyrazine-1,4-dione) had no algicidal effects against Synechococcus sp. BN60, but had a high level of algicidal activity against M. aeruginosa 9110. The LD50 value of S-2A against M. aeruginosa 9110 was 5.7 μg/ml. S-2B (2, 3-indolinedione) showed a potent algicidal effect against both M. aeruginosa 9110 and Synechococcus sp. BN60, and the LD50 value of S-2B against M. aeruginosa 9110 and Synechococcus sp. BN60 was 12.5 and 34.2 μg/ml, respectively. Obvious morphological changes in M. aeruginosa 9110 and Synechococcus sp. BN60 were observed after they were exposed to S-2A (or S-2B) for 24 h. Approximately, the algicidal activity, the concentration of S-2A and S-2B, and the cell density of Lzh-2 were positively related to each other during the cocultivation process. Overall, these findings increase our knowledge about algicidal substances secreted by algicidal bacteria and indicate that strain Lzh-2 and its two algicidal substances have the potential for use as a bio-agent in controlling cyanobacterial blooms in Lake Taihu.  相似文献

A brownish yellow pigmented bacterial strain, designated antisso-27, was recently isolated from a water area of saltpan in Southern Taiwan. Phylogenetic analyses based on 16S rRNA gene sequences indicate that strain antisso-27 belongs the genus Aquimarina in the family Flavobacteriacea and its only closest neighbor is Aquimarina spongiae (96.6%). Based on screening for algicidal activity, strain antisso-27 exhibits potent activity against the toxic cyanobacterium Microcystis aeruginosa. Both the strain antisso-27 bacterial culture and its culture filtrate show algicidal activity against the toxic cyanobacterium, indicating that an algicidal substance is released from strain antisso-27. The algicidal activity of strain antisso-27 occurs during the late stationary phase of bacterial growth. Strain antisso-27 can synthesize an algicidal protein with a molecular mass of 190 kDa, and its isoelectric point is approximately 9.4. This study explores the nature of this algicidal protein such as l-amino acid oxidase with broad substrate specificity. The enzyme is most active with l-leucine, l-isoleucine, l-methionine and l-valine and the hydrogen peroxide generated by its catalysis mediates algicidal activity. This is the first report on an Aquimarina strain algicidal to the toxic M. aeruginosa and the algicidal activity is generated through its enzymatic activity of l-amino acid oxidase.  相似文献

The aim of the study was to determine the activity of an immobilized algicidal bacterium, Pseudomonas fluorescens HYK0210-SK09 (SK09), against blooms of the diatom Stephanodiscus hantzschii and to characterize its effect in an ecosystem. To this end, SK09 cells that had been immobilized using an activated carbon polyvinyl alcohol sponge (APVAS) were tested in a field mesocosm in the Nakdong River, South Korea. Immobilized SK09 cells showed species-specific activity toward S. hantzschii throughout the study, whereby up to 72 % of the population of this species was killed. The APVAS carrier absorbed nutrients in the mesocosms effectively, which reduced the free concentrations of nutrients. Other phytoplankton species, such as Cryptomonas ovata, did not show any fluctuation in abundance. The abundance of heterotrophic protists, such as heterotrophic flagellates and ciliates, increased significantly owing to the utilization of SK09 as a food source. The high level of algicidal activity of SK09 against S. hantzschii was sustained by the release of these algicidal cells from the carrier. Thus, the immobilization of algicidal bacteria has various advantages, which include high packing ability, enhancement of bacterial growth, protection from bacterial predators, effective nutrient removal, and improved algicidal activity. The present study demonstrates that immobilized SK09 is an effective biocontrol agent for natural S. hantzschii blooms. Taken together, the findings of this study clearly demonstrate that SK09 cells immobilized in APVAS can improve the water quality in mesocosm ecosystems without producing any ecological disturbance.  相似文献

The present study aims to maximize proteases production by Bacillus mojavensis SA strain and their use to produce bioactive protein hydrolysates from a meat by-product. The production of SA bacteria proteases was maximized using a culture medium based on wheat bran, which offer an advantage in minimizing the production cost and enhancing the enzyme activity by using agro-industrial wastes. The composition of media and cultural conditions for optimal proteases production by B. mojavensis SA strain were investigated. A successful and significant improvement of the alkaline proteases production (four folds) by the SA strain was achieved using the medium composed of (g/l): wheat bran, 50.0; KH2PO4, 0.5; K2HPO4, 0.5; CaCl2, 2.0; pH 6.0, where the growth conditions were monitored at 37 °C with an agitation speed of 200 rpm. Interestingly, the enzyme preparation of B. mojavensis was applied for the preparation of protein hydrolysates from a meat by-product. Hydrolysis was carried out for 180 min at pH 12.0. The resulting hydrolysate displayed an important antioxidant activity as evaluated by the radical scavenging capacity, the reducing power, and the β-carotene bleaching inhibition. The present study showed the high proteases’ producing level by B. mojavensis SA strain in a low-cost fermentation medium (wheat bran) and their potential use in the production of bioactive protein hydrolysate from meat by-products.  相似文献

A new sensing system based on the immobilization of luminescent bacteria,Photobacterium phosphoreum, was proposed for continuous real-time monitoring of pollutants. The response curves demonstrate thatPhotobacterium phosphoreum immobilized on the strontium alginate was very sensitive to seven reference chemicals used. The significant inhibitory concentrations for bioluminescence emission were 5 ppm for Pb(NO3)2, NiCl2, CdCl2, 50 ppm for NaAsO2, 0.1 ppm for HgCl2, 0.5 ppm for pentachlorophenol and less than 5 ppm for SDS, respectively. The alginate mixed-cells (AMC) retained their luminescence during experimental period (29 days) under storage condition of ? 80°C. The variables affecting performance of continuous flow through monitoring (CFTM) were optimized in order to ensure stability and efficiency. The flow through cell with strontium-alginate immobilized luminescent bacteria was tested with salicylate and 4-nitrophenol and a rapid response of luminescence was recorded by time drivemode in bioluminescence spectrometer after exposure to both toxicants.  相似文献

A bacterial strain designated antisso-27T, previously isolated from saltpan in Taiwan while screening for bacteria for algicidal activity, was characterized using the polyphasic taxonomic approach. Strain antisso-27T was Gram-negative, aerobic, brownish yellow colored, rod-shaped, non-flagellated and non-gliding. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain antisso-27T belonged to the genus Aquimarina within the family Flavobacteriaceae with relatively low sequence similarities of 94.0–96.6% to other valid Aquimarina spp. It contained iso-C17:0 3-OH, iso-C15:0, iso-C16:0, iso-C15:1 and iso-C15:0 3-OH as the main fatty acids and contained a menaquinone with six isoprene units (MK-6) as the major isoprenoid quinone. Major polar lipids were phosphatidylethanolamine, diphosphatidylglycerol, an uncharacterized aminolipid and five uncharacterized phospholipids. Strain antisso-27T employed direct mode of algicidal lysis to Chlorella vulgaris strain 211-31; nevertheless, it released an algicidal substance against M. aeruginosa strain MTY01. This is the first study that the Aquimarina species possesses both direct and indirect algicidal activities. On the basis of the phylogenetic and phenotypic data, strain antisso-27T should be classified as representing a novel species, for which the name A. salinaria sp. nov. is proposed. Interactions between bacteria and harmful algal bloom (HAB) species have been acknowledged as an important factor of regulating the population of these algae. In the study, two strains of algicidal bacteria, DHQ25 and DHY3, were screened out because of their probably secreting algicidal proteins against axenic Alexandrium tamarense. Molecular characterization classified them to the γ-proteobacteria subclass and to the genus Vibrio and Pseudoalteromonas, respectively. After centrifugation and ultrafiltration, chromatography of the cultural supernatants of DHQ25 revealed 8 peaks by HPLC. SDS-PAGE and Native PAGE determination showed that peak 7 to be a monoband peak. Both xenic and axenic culture of A. tamarense were susceptible to the purified protein (short for P7 below) indicated by algicidal activity assay. Observation of algicidal process demonstrated that algal cells were lysed and cellular substances were released under visual fields of microscope. P7 proved to be a challenge controller of A. tamarense by the above characterizations of algicidal activity assaying and algicidal process. This is the first report of a protein algicidal to the toxic dinoflagellate A. tamarense. The findings increase our knowledge of bacterial–algal interactions and the role of bacteria during controlling HABs.  相似文献

The aim of this study was to isolate and identify algicidal bacteria against the dinoflagellate Cochlodinium polykrikoides, and to determine the algicidal activity and algicidal range. During the declining period of C. polykrikoides blooms, seven algicidal bacteria were isolated. The algicidal bacteria against C. polykrikoides were enumerated using the most probable number (MPN) method. The number of algicidal bacteria was high (3.7 × 103 mL−1). Algicidal bacteria were identified on the basis of biochemical and chemotaxonomic characteristics, and analysis of 16S rDNA sequences. Seven algicidal bacteria isolated in this study belonged to the genera Bacillus, Dietzia, Janibacter, and Micrococcus. The most algicidal bacterium, designated Micrococcus luteus SY-13, is assumed to produce secondary metabolites. When 5% culture filtrate of this strain was applied to C. polykrikoides cultures, over 90% of C. polykrikoides cells were destroyed within 6 h. M. luteus SY-13 showed significant algicidal activities against C. polykrikoides and a wide algicidal range against various harmful algal bloom (HAB) species. Taken together, our results suggest that M. luteus SY-13 could be a candidate for controlling HABs.  相似文献

Summary A fungal strain isolated from soil and identified asAspergillus athecius, when grown on moistened wheat bran produced large amounts of extracellular invertase. Most of the invertase from the moldy bran was easily extracted by low ionic strength buffer (0.005 M, pH 5.7). The crude invertase immobilized on DEAE cellulose showed not only increased activity (45%) but also greater thermal and storage stability than the free enzyme. The free and the bound enzymes showed a temperature optimum of 50–55°C and a pH optimum of 5.7 and 4.8 respectively. The Km app. of the bound enzyme was lower than that of the free enzyme.  相似文献

A lytic phage (øZCW1) was isolated from an algicidal bacterium Pseudoalteromonas sp. strain SP48 that specifically kills the toxic dinoflagellate Alexandrium tamarense. We demonstrated that øZCW1 could trigger the growth of A. tamarense by inhibiting the growth of algicidal bacterium SP48. In contrast, the growth of A. tamarense was suppressed when cocultured with either SP48 or the øZCW1-resistant mutant of SP48. This study provides the first evidence of the indirect impact of bacteriophage on bloom-forming microalgae via phage lysis of alga-killing bacteria.  相似文献

Nursing piglets are entirely dependent, for their micronutrient provisions, upon in utero, colostrum and milk transfers from the dam. An adequate maternal transfer of micronutrients is all the more important during these periods which, in fact, lasts for approximately half the life cycle (conception to slaughter) of modern pigs. The present study aimed to set up a simple approach to assess the maternal perinatal transfer of vitamins and trace elements in sows. Prenatal transfer (R-u) was estimated as limited, passive or active using the ratio between pre-colostral serum concentrations of a given micronutrient in newborn piglets and corresponding pre-farrowing values in sows. Efficiency of the postnatal transfer (R-c) was estimated from the ratio between serum concentrations of post- and pre-colostral micronutrients in piglets. Data from literature (12 studies) were used for vitamins A, D, E, C, folic acid and B12, whereas vitamins B2, B3, B6 and B8 as well as Zn, Fe, Cu and Se were generated from a trial where blood sera from 20 sows, and their litter were collected during the perinatal period. In sow trial, statistical t tests were used to determine if ratios differed from 1. Prenatal transfer was active and in favour of piglets (R-u > 1, P < 0.03) for Zn and vitamins B6 and B8 (sow trial) as well as for vitamins C and B12 (literature data). This transfer was limited (R-u < 1, P < 0.01) for vitamin B2, Fe, Cu and Se (sow trial) and for vitamins A, E, D and folic acid (literature data) whereas it was passive for vitamin B3 (R-u = 1, P > 0.37). After birth, the early postnatal transfer through colostrum was active towards piglets for most micronutrients but vitamins B6 and B8 (R-c < 1, P < 0.01). Globally, the perinatal transfer (combination of R-u and R-c) was favourable to the neonatal piglets for most micronutrients except for vitamins A and D as well as Fe, Cu and Se whereas there is apparently a barrier for prenatal transfer which is not compensated by the colostrum provision to neonatal piglets. Then, post-colostral concentrations of these micronutrients in piglets remain below prenatal levels of their dam. Neonatal strategies of micronutrient provision are known for Fe (intramuscular injection) and Se (sow milk enrichment). Further studies are needed to assess the importance of the unfavourable perinatal transfer for Cu and vitamins A and D for piglet robustness later in life.  相似文献

In yeast crops which were grown in the presence of various inhibitors, there was considerable variation in content of the various B vitamins. A higher degree of parallelism in variation in content was found to exist between thiamine and niacin than between any other pair of vitamins; this has been interpreted as indicating that the predominant functions of these two vitamins are their established rôles in fermentation. Values for inositol indicate that it may be involved in fermentation processes, but this is not the case for other members of the B complex. Biotin appears to be unique since in no case did the biotin content of yeast grown in the presence of an inhibitor fall below that of the control yeast. There was some evidence of synthesis of biotin, or a material with biotin activity, in the presence of certain inhibitors, the most striking instance being with sulfaguanidine. An exogenous supply of biotin was essential for extensive proliferation of F. B. yeast, and yeast grown in a medium to which biotin was the only added vitamin contained the B vitamins in amounts very similar to those found in the control yeast, the most marked differences being in increased vitamin B6 and p-aminobenzoic acid contents. In the absence of biotin, significant amounts of all of the B vitamins except biotin were synthesized, both in the presence and absence of certain other members of the B complex. The addition of thiamine, pyridoxine, inositol, and β-alanine to the culture medium caused a reduction in the amounts of vitamin B6 and p-aminobenzoic acid synthesized. F. B. yeast was able to grow in a xylose medium only when certain of the B vitamins were present, and even then growth was limited. Evidence was obtained for some synthesis of all of the vitamins investigated except biotin and vitamin B6. The most significant differences in vitamin content between galac yeast and the parent F. B. strain were in folic acid and vitamin B6, the former being considerably reduced in amount, the latter being increased.  相似文献

One strain of algicidal bacterium which can inhibit Harmful algal blooms (HABs), FDT5, was isolated from activated sludge and found to have good algicidal effects on Microcystis aeruginosa. It was revealed that: The FDT5 was a Gram-negative bacterium and identified as Ochrobactrum sp.; The greater the initial bacterial cell density, the faster the degradation of chlorophyll a.; The algicidal efficiency was evaluated at the most favorable conditions which were a temperature of 30–35°C, a pH of 7.6 and complete darkness; The FDT5 strain lysed Microcystis aeruginosa not directly but by secreting metabolites which could withstand high temperatures and pressure.  相似文献
