垂体腺苷酸环化酶激活肽(PACAP)是近年新发现的神经多肽,属于促胰液素/胰高血糖素/血管活性肽(VIP)家族中的新成员,广泛分布于脑和外周组织器官,尤其在内分泌胰腺、性腺、呼吸和生殖系统,在能量代谢、神经保护、免疫系统等发挥重要生理学功能。糖尿病是一种常见的主要以高血糖为特征的慢性代谢性疾病,糖尿病并发症日益严重威胁着人们的身体健康,已成为导致糖尿病患者致死、致残的主要原因。主要对PACAP治疗糖尿病及其并发症国内外研究的最新进展进行论述。  相似文献

Overexpression of Regulator of Calcineurin 1 (RCAN1/DSCR1/Adapt78) is known to inhibit the calcineurin-NFAT dependent signaling pathway. In this report, we find that activation of adenylate cyclase by forskolin increases the expression of RCAN1 through the increase of the protein’s half-life. The ability of forskolin to increase the accumulation of RCAN1 protein is significantly inhibited with protein kinase A inhibitors such as KT5720 and H-89. Furthermore, forskolin targets the central and C-terminal region of RCAN1 and enhances the inhibitory effect of RCAN1 on the calcineurin-mediated activation of NFAT. Our findings provide the first evidence that the accumulation of the RCAN1 protein by cAMP acts as an important regulatory mechanism in the control of the calcineurin-dependent cellular pathway.

采用环腺苷酸 (cAMP)放射免疫测定法和活细胞内Ca2 荧光探针Indo 1,研究绵羊垂体腺苷酸环化酶激活多肽 (oPACAP)对原代培养的鲤鱼脑垂体细胞内cAMP和游离Ca2 ([Ca2 ]i)的影响 ,以期探讨PACAP调节脑垂体生长激素 (GH)分泌的机制受体后。oPACAP 38和oPACAP 2 7以剂量依存方式促进脑垂体细胞内cAMP释放和合成。oPACAP 38和oPACAP 2 7也能升高脑垂体细胞内 [Ca2 ]i 水平 ,该作用会因用EGTA消竭细胞外Ca2 ([Ca2 ]e)而迅速消失 ;L型电位敏感性Ca2 通道 (VSCC)阻断剂硝苯吡啶可抑制oPACAP 38诱导的 [Ca2 ]i 水平的升高 ,而当用硝苯吡啶预处理脑垂体细胞 ,oPACAP 38诱导 [Ca2 ]i 水平升高作用完全被抑制。可见 ,PACAP刺激鲤鱼脑垂体GH分泌机制包括依赖于cAMP和依赖于通过L型VSCC内流的 [Ca2 ]e 的机制。  相似文献

Pituitary adenylate cyclase-activating polypeptide (PACAP) functions as a neuroprotective factor through the PACAP type 1 receptor, PAC1. In a previous work, we demonstrated that nerve growth factor augmented PAC1 gene expression through the activation of Sp1 via the Ras/MAPK pathway. We also observed that PAC1 expression in Neuro2a cells was transiently suppressed during in vitro ischemic conditions, oxygen-glucose deprivation (OGD). Because endoplasmic reticulum (ER) stress is induced by ischemia, we attempted to clarify how ER stress affects the expression of PAC1. Tunicamycin, which induces ER stress, significantly suppressed PAC1 gene expression, and salubrinal, a selective inhibitor of the protein kinase RNA-like endoplasmic reticulum kinase signaling pathway of ER stress, blocked the suppression. In luciferase reporter assay, we found that two Sp1 sites were involved in suppression of PAC1 gene expression due to tunicamycin or OGD. Immunocytochemical staining demonstrated that OGD-induced transglutaminase 2 (TG2) expression was suppressed by salubrinal or cystamine, a TG activity inhibitor. Further, the OGD-induced accumulation of cross-linked Sp1 in nuclei was suppressed by cystamine or salubrinal. Together with cystamine, R283, TG2-specific inhibitor, and siRNA specific for TG2 also ameliorated OGD-induced attenuation of PAC1 gene expression. These results suggest that Sp1 cross-linking might be crucial in negative regulation of PAC1 gene expression due to TG2 in OGD-induced ER stress.  相似文献

C3H10T1/2多潜能干细胞成脂过程分为定向和分化两个阶段,骨形成蛋白4(BMP4)可以诱导其定向成前脂肪细胞.已有的研究表明,脂肪组织特异性敲除低密度脂蛋白受体相关蛋白1(Lrp1)的小鼠体重减轻,脂肪组织含量减少,揭示此基因对成脂具有重要作用.然而,目前尚不清楚Lrp1是否在成脂定向过程中发挥作用.采用小干扰RNA技术(RNAi),在体外水平研究低密度脂蛋白Lrp1对C3H10T1/2多潜能干细胞成脂定向的作用.分别在C3H10T1/2成脂的定向期和脂滴成熟期敲低Lrp1,通过显微镜下观察、油红O染色、Western blotting等实验证实,定向期而非脂滴成熟期敲低Lrp1显著抑制C3H10T1/2多潜能干细胞成脂.BMP4通过激活下游Smad1/5/8信号通路发挥作用,而敲低Lrp1显著抑制BMP4诱导的Smad1/5/8磷酸化.这些结果说明:敲低Lrp1通过下调Smad信号通路,抑制BMP4诱导的C3H10T1/2多潜能干细胞成脂定向.  相似文献

The melanocortin-1 receptor (MC1R) is a key regulator of mammalian pigmentation. Melanism in the grey squirrel is associated with an eight amino acid deletion in the mutant melanocortin-1 receptor with 24 base pair deletion (MC1RΔ24) variant. We demonstrate that the MC1RΔ24 exhibits a higher basal activity than the wildtype MC1R (MC1R-wt). We demonstrate that agouti signalling protein (ASIP) is an inverse agonist to the MC1R-wt but is an agonist to the MC1RΔ24. We conclude that the deletion in the MC1RΔ24 leads to a receptor with a high basal activity which is further activated by ASIP. This is the first report of ASIP acting as an agonist to MC1R.  相似文献

Homer proteins are commonly known as scaffold proteins at postsynaptic density. Homer 1 is a widely studied member of the Homer protein family, comprising both synaptic structure and mediating postsynaptic signaling transduction. Both an immediate-early gene encoding a Homer 1 variant and a constitutively expressed Homer 1 variant regulate receptor clustering and trafficking, intracellular calcium homeostasis, and intracellular molecule complex formation. Substantial preclinical investigations have implicated that each of these Homer 1 variants are associated with the etiology of many neurological diseases, such as pain, mental retardation syndromes, Alzheimer's disease, schizophrenia, drug-induced addiction, and traumatic brain injury.  相似文献

Cardiovascular diseases are a major cause of human death worldwide. Excessive proliferation of vascular smooth muscle cells contributes to the etiology of such diseases, including atherosclerosis, restenosis, and pulmonary hypertension. The control of vascular cell proliferation is complex and encompasses interactions of many regulatory molecules and signaling pathways. Herein, we recapitulated the importance of signaling cascades relevant for the regulation of vascular cell proliferation. Detailed understanding of the mechanism underlying this process is essential for the identification of new lead compounds (e.g., natural products) for vascular therapies.  相似文献
