The adherence of Escherichia coli B cells to cell wall associated-agglutinin of the soil borne plant pathogen Rhizoctonia solani, was inhibited by l-fucose, l-galactose, trypsin, SDS, cycloheximide and Na2-EDTA. The coiling of the biocontrol agent Trichoderma harzianum around Rhizoctonia hyphae was prevented by SDS, cycloheximide, Na2-EDTA and methyl--l-fucoside — an inhibitor of Rhizoctonia agglutinin not metabolized by both fungi. The possible role of the agglutinin in Trichoderma-Rhizoctonia interaction is discussed.  相似文献

Evidence is presented that the high levels of internal l-glutamic and l-aspartic acid in frog Rana esculenta red blood cells are due to the existence of a specific carrier for acidic amino acids of high affinity K m = 3 m and low capacity (Vmax) 0.4 mol l-Glu · Kg–1 dry cell mass · 10 min–1. It is Na+ dependent and the incorporation of l-glutamic acid can be inhibited by l and d-aspartate and l-cysteic acid, while d-glutamic does not inhibit. Moreover, this glutamic uptake shows a bell-shaped dependence on the external pH. All these properties show that this carrier belongs to the system X AG family. Besides the incorporation through this system, l-glutamic acid is also taken up through the ASC system, although, under physiological conditions, this transport is far less important, since it has relatively low affinity K m 39 m but high capacity (V max) 1.8 mol l-Glu · Kg–1 dry cell mass · 10 min–1.  相似文献

Summary A detailed study on the reductive amination of -ketoisovalerate to l-valine by l-valine dehydrogenase using glucose dehydrogenase as an NADH regeneration enzyme was performed. The presence of both enzyme activities in Bacillus megaterium ATCC 39 118 permitted a direct and systematic comparison of the performances (initial l-valine production rate, productivity, molar conversion yield) of different types of conversion systems: purified enzymes or crude extract and whole cells, intact or permeabilized. A maximal l-valine productivity of 8 mmol·l–1 · h–1 was obtained using purified enzymes which constituted the most efficient system with a maximal rate of 0.87 mol · ml–1 · min–1 and a molar conversion yield of 0.91. The ability to convert d-galactose into d-tagatose was compared among a number of bacterial l-arabinose isomerases (araA). One of the most efficient enzymes, from the anaerobic thermophilic bacterium Thermoanaerobacter mathranii, was produced heterologously in Escherichia coli and characterised. Amino acid sequence comparisons indicated that this enzyme is only distantly related to the group of previously known araA sequences in which the sequence similarity is evident. The substrate specificity and the Michaelis–Menten constants of the enzyme determined with l-arabinose, d-galactose and d-fucose also indicated that this enzyme is an unusual, versatile l-arabinose isomerase which is able to isomerise structurally related sugars. The enzyme was immobilised and used for production of d-tagatose at 65 °C. Starting from a 30% solution of d-galactose, the yield of d-tagatose was 42% and no sugars other than d-tagatose and d-galactose were detected. Direct conversion of lactose to d-tagatose in a single reactor was demonstrated using a thermostable -galactosidase together with the thermostable l-arabinose isomerase. The two enzymes were also successfully combined with a commercially available glucose isomerase for conversion of lactose into a sweetening mixture comprising lactose, glucose, galactose, fructose and tagatose.  相似文献

Exopolysaccharides of a number of slime-producingAchromobacter strains, isolated from activated sludge, were prepared and analysed. They consist ofD-glucose,D-galactose, pyruvic acid and O-acetyl in the approximate molar ratio of 1:1:1:1/2. The polysaccharides were shown by methylation and partial hydrolysis to consist of unbranched chains of alternately arrangedD-glucose andD-galactose residues, exclusively linked by -1,3-linkages, and with pyruvate substituents linked by acetal bonds at positions 0–4 and 0–6 of theD-galactose residues. The significance of the exopolysaccharides in relation to some relevant properties of activated sludge organisms is discussed.  相似文献

The xylogalactan sulfate from Chondria macrocarpa (Ceramiales,Rhodophyta)   总被引:1,自引:1,他引:0  
A structure is proposed for the complex xylogalactan sulfate from Chondria macrocarpa. The hot-water extract of C. macrocarpa was desulfated or alkali-treated and Smith degraded. Constituent sugars and their substitution patterns were identified using a modified Hakamori methylation procedure suited to sulfated polysaccharides and a double hydrolysis-reduction protocol that yielded derivatives from all of the sugar residues, including the labile 3,6-anhydrogalactosyl residues. The polymer has an agar-type backbone of alternating 3-linked \-d- and 4-linked -L-galactopyranosyl units. The d-residues are partially sulfated on O-2 (50%) and O-6 (20–30%). About 40% of the l-residues are present as the 3,6-anhydride and 25% as its precursor l-galactose 6-sulfate. A significant proportion of the remaining l-galactosyl residues have both a d-xylopyranosyl substituent on O-3 and a sulfate ester on O-6 and are stable to alkali.  相似文献

Summary In the present work the chemical cell wall composition and some other biochemical characteristics were studied in staphylococci with the intention of utilizing the data obtained in their classification.According to the cell wall peptidoglycans and teichoic acids, the 130 strains of staphylococci studied are divided into 10 major groups. This division of staphylococci into groups is in good agreement with their present classification only in some cases. All of the 47Staphylococcus aureus strains contain a cell wall peptidoglycan of thel-Lys-Gly5–6 type and ribitol teichoic acid. Coagulase-negative staphylococci are more heterogeneous and are divided according to their cell wall composition into 9 major groups. 21 strains of them are classified asS. epidermidis sensu stricto. They form a natural group and are distinguished by the occurrence of thel-Lys-Gly4–5,l-Ser0.5–1.8 peptidoglycan type, glycerol teichoic acid and anl-lactate dehydrogenase which is activated by fructose-1,6-diphosphate. 8 strains with peptidoglycan of thel-Lys-Gly4–5,l-Ser0.5–1.8 type and ribitol teichoic acid are labeled asS. saprophyticus. The remaining groups have not been given species names and require further extensive comparative study.  相似文献

Zusammenfassung Von 18 Stämmen von Bifidobacterium longum Reuter und einem Stamm von B. lactentis Reuter wurden die Zellwände in üblicher Weise hergestellt. Sie enthielten Mur, GlcNH2, d-Glu, Ala, l-Orn (l-Lys), Thr, Ser in einem Molverhältnis von rund 1:1:1:4:1:1:1. Die Molverhältnisse änderten sich nicht, wenn die Zellwände mit Trichloressigsäure oder heißem Formamid extrahiert wurden. In einigen Stämmen trat mehr als 1 Mol Glutaminsäure pro Mol Diaminosäure auf. Die zusätzliche Glutaminsäure hatte die l-Konfiguration. Sie war kein Bestandteil des Mureins, sondern einer lysozymunempfindlichen, unbekannten Zellwandkomponente, vermutlich einer Polyglutaminsäure. l-Ornithin war in den meisten Stämmen die dominierende Diaminosäure, während l-Lysin nur mit einem Anteil von 10–20% vertreten war. In 2 Stämmen war l-Lysin dominierend (90%). Die Aminosäuresequenz wurde durch die Analyse der Oligopeptide aus Partialhydrolysaten bestimmt. Die an Mureinsäure gebundenen Peptiduntereinheiten hatten die auch von anderen Mureinen bekannte Sequenz: l-Ala-d-Glu-l-Orn (oder l-Lys)-d-Ala. Glutaminsäure ist wahrscheinlich amidiert, wie aus dem Auftreten von rund 1 Mol NH3 im Hydrolysat der Zellwände zu schließen ist. Die Interpeptidbrücke besteht aus dem Peptid l-Ala-Thr-l-Ala-l-Ser. Sie ist mit dem C-terminalen Serin an die -Aminogruppe der Diaminosäure der Peptiduntereinheit gebunden. Die Quervernetzung erfolgt zwischen dem N-terminalen Alanin der Interpeptidbrücke zum C-terminalen d-Alanin einer Peptiduntereinheit. Da 4% des gesamten Alanins und 3% der -Aminogruppe des Ornithins dinitrophenylierbar sind, ist anzunehmen, daß die Quervernetzung nur zu etwa 80% verwirklicht ist.

Summary Cell walls of 18 strains of Bifidobacterium longum Reuter and one strain of B. lactentis Reuter were prepared in the usual way. They contained Mur, GlcNH2, d-Glu, Ala, l-Orn (l-Lys), Thr, Ser in a molar ratio of about 1:1:1:4:1:1:1. The ratio was not changed when the cell walls were extracted by trichloroacetic acid or hot formamide. In some strains more than 1 mole glutamic acid per mole of diamino acid was present. The additional glutamic acid was of the l-rather than the d-form. It was not a constituent of the murein, but of an unknown lysozyme insensitive cell wall component, probably a polyglutamic acid. l-Ornithine was the dominating diamino acid in most strains but l-lysine was also present in a portion of 10 to 20%. In 2 strains l-lysine was dominating (90%).The amino acid sequence was determined by analysing the oligopeptides arising during partial acid hydrolysis. It was shown that the peptide subunits attached to the muramic acid are the same as those of other mureins: l-Ala-d-Glu-l-Orn (or l-Lys)-d-Ala. glutamic acid is probably amidated, since about 1 mole of NH3 is released by acid hydrolysis of the cell walls. The interpeptide bridge consists of the peptide l-Ala-Thr-l-Ala-l-Ser which is bound by its C-terminal serine to the -amino group of the diamino acid of one peptide subunit and by its N-terminal l-alanine to the C-terminal d-alanine of another peptide subunit. About 4% of the total alanine and 3% of the -amino groups of ornithine of the cell wall can be dinitrophenylated. This indicates that about 20% of the peptide subunits are not crosslinked.

l-Lysine is known to stimulate production of -lactam antibiotics byStreptomyces clavuligerus via provision of the lysine breakdown product,l--aminoadipic acid, which is a limiting precursor. Previous investigations utilized levels of 10–20 mMl-lysine as an addition to chemically-defined media resulting in 50–100% improvement in antibiotic production. We were surprised to note that as the concentration was further increased, the organism responded by producing even higher titers of antibiotics. Summary A new process (Living Cell Reaction Process) forl-isoleucine production using viable, non-growing cells ofBrevibacterium flavum AB-07 was optimised using ethanol as the energy source and -ketobutyric acid (-KB) as precursor.l-valine also could be produced from glucose at high yield by this process. This process differs from the usual fermentation method in that non-growing cells are used, and the production ofl-isoleucine andl-valine were carried out under conditions of repressed cell division and growth. Minimal medium missing the essential growth factor, biotin was employed as the reaction mixture for the production ofl-isoleucine andl-valine. The productivity ofl-isoleucine andl-valine were 200 mmol·l–1 · day–1 (molecular yield to -KB: 95%) and 300 mmol · l–1 · day–1 (molecular yield to glucose: 80%) respectively. The content ofl-isoleucine andl-valine in total amino acids produced in the each mixture were 97% and 96% respectively.  相似文献

Sternbergia fischeriana is an endangered geophyte and therefore in vitro micropropagation of this plant will have great importance for germplasm conservation and commercial production. Bulb scale and immature embryo explants of S. fischeriana were cultured on different nutrient media supplemented with various concentrations of plant growth regulators. Immature embryos produced higher number of bulblets than bulb scales. Large numbers of bulblets were regenerated (over 80 bulblets/explants) from immature embryos on Murashige and Skoog (MS) medium supplemented with 4 mg l–1 6-benzylaminopurine (BA) and 0.25 mg l–1 -naphthaleneacetic (NAA) or 2 mg l–12,4-dichlorophenoxyacetic acid (2,4-D) after 14 months of culture initiation. Regenerated bulblets were kept at 5 °C for 5 weeks and then transplanted to a potting mixture.  相似文献

Summary We examined the direct effects of isoproterenol (ISO) andl-norepinephrine (NE) on electrolyte transport in isolated rabbit cortical collecting tubules (CCT) perfusedin vitro. The addition of either ISO (10–6 m) or NE (10–6 m) to the bath decreased transepithelial potential difference (PD), on average by 51 and 25%, respectively. These effects of ISO and NE were abolished by prior addition of the -adrenergic blocker,l-propranolol. ISO (10–5 m) had no effect from lumen. Also, osmotic water permeability was not influenced by ISO. Ouabain and ISO had additive effects on PD. Elimination of chloride from both perfusate and bath, or addition of acetazolamide, abolished the effect of ISO on PD. Although isotopic sodium flux from lumen to bath was not influenced by ISO, chemical net chloride absorption increased from 1.1±0.4 to 2.7±0.6 peq·cm–1·sec–1 (n=8,p<0.005). In conclusion, both ISO and NE are capable of decreasing PD in rabbit CCT perfusedin vitro. This effect is mediated by -adrenergic receptors and is accompanied by the increase in net chloride absorption. Although the mechanism responsible for this decrease in PD with ISO is unclear, active chloride absorption, active hydrogen secretion, or membrane chloride permeability changes may account for the effects of ISO.  相似文献

Polysaccharide extracts from four New Zealand members of the Gracilariaceae have been characterized by 13C-NMR spectroscopy and GLC analysis of alditol acetate derivatives prepared using a new double hydrolysis-reduction procedure. All were based on variously substituted repeating disaccharide units of agarobiose and 20% of its precursor containing l-galactose-6-sulfate. Gracilaria truncata yielded a firm gelling agar with 67% methylation on the 6-position of the d-galactose residues. The other extracts belong to a new class of agar molecules having methylation on both the 6-position of the d-galactose units and the 2-position of the l-sugar units. The Curdiea coriacea polysaccharide displayed this double methylation almost completely ( 96 %); the alkali-modified polymer thus had only two free hydroxy-groups per disaccharide repeat unit, yet still gave a firm gel. The Curdiea flabellata and Melanthalia abscissa extracts had this double methylation pattern but to a lesser extent, and additional xylosyl branch units on up to 18% of the repeating disaccharide units.  相似文献

Arabidopsis thaliana mur1 is a dwarf mutant with altered cell-wall properties, in which l-fucose is partially replaced by l-galactose in the xyloglucan and glycoproteins. We found that the mur1 mutation also affects the primary structure of the pectic polysaccharide rhamnogalacturonan II (RG-II). In mur1 RG-II a non-reducing terminal 2-O-methyl l-galactosyl residue and a 3,4-linked l-galactosyl residue replace the non-reducing terminal 2-O-methyl l-fucosyl residue and the 3,4-linked l-fucosyl residue, respectively, that are present in wild-type RG-II. Furthermore, we found that a terminal non-reducing l-galactosyl residue, rather than the previously reported d-galactosyl residue, is present on the 2-O-methyl xylose-containing side chain of RG-II in both wild type and mur1 plants. Approximately 95% of the RG-II from wild type and mur1 plants is solubilized as a high-molecular-weight (>100 kDa) complex, by treating walls with aqueous potassium phosphate. The molecular mass of RG-II in this complex was reduced to 5–10 kDa by treatment with endopolygalacturonase, providing additional evidence that RG-II is covalently linked to homogalacturonan. Zusammenfassung Es wird die Aktivität von intaktenL. plantarum-Zellen verschiedener Arten gegenüberl-Äpfelsäure, Oxalessigsäure und Brenztraubensäure getestet. Bei der Dissimilation vonl-Äpfelsäure lassen sich zwei pH-Optima unterscheiden, 2,6–3,0 für eine MDH-Aktivität und 3,6–4,0 für eine Malic-Enzym-Aktivität. Stoffwechselprodukte der Brenztraubensäure-Decarboxylierung sind außer Kohlendioxid Äthylalkohol und Acetoin bzw. Diacetyl.L. plantarum ist außerdem zur Oxydation der Brenztraubensäure befähigt. Ausl-Äpfelsäure entsteht kein Acetoin (Stamm L).

Summary The decomposition ofl-malic, oxaloacetic and pyruvic acids by intact cells of three strains ofL. plantarum is investigated. The dissimilation ofl-malic acid shows two pH-optima, at pH2.6–3.0 for a malatedehy drogenase activity and at pH 3.6–4.0 for a malic enzyme activity. The decarboxylation of pyruvic acid yields CO2, ethyl alcohol, acetoin and diacetyl.L. plantarum is also able to oxidize pyruvic acid. The acetoin produced byL. plantarum Strain L does not originate froml-malic acid.

Summary Escherichia coli 044 K74 grown under anaerobic conditions in the presence of l(–)-carnitine is able to convert d(+)-carnitine into the l(–)-enantiomer. This activity is repressed by electron acceptors such as oxygen and nitrate as well as by glucose. d(+)-Carnitine shows no effect on the induction or repression of the corresponding enzyme or enzyme system. Resting cells of E. coli 044 K74 were used for the formation of l(–)-carnitine from d(+)-carnitine. The maximum obtained yield was 50%. -Butyrobetaine was formed as a by-product. Fluorescence and stopped-flow spectrophotometric studies on three plant lectins fromPsophocarpus tetragonolobus (winged bean),Glycine max (soybean) andArtocarpus integrifolia (jack fruit) have been studied usingN-dansylgalactosamine as a fluorescent ligand. The best monosaccharide for the winged bean agglutinin I (WBA I) and soybean (SBA) is Me-GalNAc and for jack fruit agglutinin (JFA) is Me-Gal. Examination of the percentage enhancement and association constants (1.51×106, 6.56×106 and 4.17×105 M–1 for SBA, WBA I and JFA, respectively) suggests that the combining regions of the lectins SBA and WBA I are apolar whereas that of JFA is polar. Thermodynamic parameters obtained for the binding of several monosaccharides to these lectins are enthalpically favourable. The binding of monosaccharides to these lectins suggests that the-OH groups at C-1, C-2, C-4 and C-6 in thed-galactose configuration are important loci for interaction with these lectins. An important finding is that the JFA binds specifically to Galß1-3GaINAc with much higher affinity than the other disaccharides which are structurally and topographically similar.The results of stopped-flow spectrometry on the binding ofN-dansylgalactosamine to these lectins are consistent with a bimolecular single step mechanism. The association rate constants (2.4×105, 1.3×104, and 11.7×105 M–1 sec–1 for SBA, WBA I and JFA, respectively) obtained are several orders of magnitude slower than the ones expected for diffusion controlled reactions. Summary The specific growth rate () during cultivation of Bacteroides polypragmatus in 2.51 batch cultures in 4–5% (w/v) l-arabinose medium was 0.23 h-1 while that in either d-xylose or d-ribose medium was lower (=0.19 h-1). Whereas growth on arabinose or xylose occurred after about 6–8 h lag period, growth on ribose commenced after a 30 h lag phase. The maximum substrate utilization rate for arabinose, ribose and xylose in media with an initial substrate concentration of 4–5% (w/v) was 0.77, 0.76, and 0.60 g/l/h respectively. In medium containing a mixture of glucose, arabinose, and xylose, the utilization of all three substrates occurred concurrently. The maximum amount of ethanol produced after 72 h growth in 4–5% (w/v) of arabinose, xylose, and ribose was 9.4, 6.5, and 5.3 g/l, respectively. Summary An ethionine resistant mutant of Candida utilis was found to maintain an expanded intracellular pool of free l-methionine in batch and continuous cultures. During glucose-limited growth in mineral salts medium in a continuous fermenter, the free l-methionine pool of the mutant was 40–80% higher than in batch cultures, and varied in the range of 25–30 moles/g dry cells (3.7–4.5 mg/g dry cells).  相似文献

A new method was established for somatic embryogenesis and plant regeneration from callus cultures of Dioscorea zingiberensis C.H. Wright. Primary callus was induced by culturing stems, leaves and petioles on Murashige and Skoog (MS) medium supplemented with 0.5–2.0 mg l–1 N6-benzyladenine (BA) and 0–2.0 mg l–1 -naphthaleneacetic acid (NAA) or 2,4-dichlorophenoxyacetic acid (2,4-D) for 1 month. The highest frequency (87%) of callus formation was achieved from stem explants treated with 0.5 mg l–1 BA and 2.0 mg l–1 2,4-D. Somatic embryos were obtained by subculturing embryogenic calli derived from stem explants on MS medium supplemented with 2.0–4.0 mg l–1 BA and 0–0.4 mg l–1 NAA or 2,4-D for 3 weeks. The optimum combination of 4.0 mg l–1 BA and 0.2 mg l–1 NAA promoted embryo formation on one-third of the calli. After a further month of subculture on the same medium, mature embryos were transferred to MS medium supplemented with 0–4.0 mg l–1 BA, NAA or indole-3-butyric acid (IBA) for further development of plantlets and tuber formation. Plant growth regulators had a negative effect on the development of mature embryos.  相似文献
