Summary Substantial numbers of predominantly O+ fish were impinged on the cooling water screens of Harculo, Gelderland, Merwedehaven and Amer power stations. Low numbers were impinged at the two stations located on the river Meuse (Maas and Claus). Two possible reasons for this difference are suggested,viz. a low fish density in the river Meuse and the deep position of the cooling water intakes. In comparison with Bergum power station situated on a lake the impingement rate at the river stations was more than ten times as low.Most fish were impinged during the summer months when large numbers of juvenile fish are present in the rivers. Dominating species impinged were roach, pikeperch, bream and perch. The abundance of pikeperch in the screensamples, points to a recent increase in the population of pikeperch in the rivers. The growth pattern of O+ pikeperch and O+ roach during summer seemed very similar for power stations located at different rivers. Thirty fish species and four crustacean species were collected, among whichOrconectus limosus was quite abundant at Maas and Claus power stations.  相似文献

The harpacticoid fauna of a sandy sediment in the Sluice Dock of Ostend, Belgium, was dominated by a single species, Canuella perplexa, that attained an average annual density of over 400 ind. 10 cm–2 in 1976 and 1977, whereas other species had a total mean density of less than 10 ind. 10 cm–2. In the summer of 1979 a dystrophic crisis with abundant growth of Ulva occurred and in March 1980 the lagoon was drained. After refilling of the lagoon, the sediment was immediately colonized by good swimming harpacticoids, that attained densities of 200 ind. 10 cm–2. Three months after refilling Canuella perplexa reappeared and the system returned to its original state within one month.  相似文献

Decreased serum concentrations that substantially alter the growth of normal 3T3 cells alter neither the active and non-active components of unidirectional 86Rb+ influx nor the intracellular K+ content when compared with cells in exponential growth. Thus the changes in K+ transport (measured with 86Rb+ as an analogue for K+ movements) that occur on density-dependent growth inhibition of the mouse 3T3 cell are not mimicked by serum deprivation of the cells before density inhibition.  相似文献

SUMMARY 1. Following fish removal, the water quality in biomanipulated lakes often improves concomitant with decreased phosphorus (P) levels. Because the decrease in P concentrations derives most probably either directly or indirectly from fish, which are the main target of biomanipulation, this study examined the P release of 0+, 1+ and 2+ roach [Rutilus rutilus (L.)] and changes in the P release during summer in a shallow eutrophic lake in Finland. 2. The P release was separated into P derived from benthic and littoral food items and into recycled P derived from feeding on zooplankton, to estimate the contribution of net P additions to the water column by the fish to the increase in P concentrations of the lake water (75–110 mg P m?3) in summer 1991–96. 3. Individual P release of roach by both egestion and excretion was estimated with a bioenergetics model. The size of the roach population was estimated with a depletion method and the proportions of different age groups from catch samples, using a programme separating mixtures of normal distributions. The sensitivity of the release estimates to variation in the growth data was estimated with the jackknife technique. 4. The biomass‐specific P release by 0+ roach (0.36–0.54 mg P g?1 day?1) was higher than that by older roach (0.07–0.16 mg P g?1 day?1) throughout the summer. The P release by the whole roach population deriving from benthic and littoral food items (0.7–2.7 mg m?3 during July to August, representing a net addition to the water column) was 5–19 times lower in 1991–96 than the recycled P release deriving from zooplankton (8.9–25.7 mg m?3), and too low to explain the increase in the P concentration of the lake water during the summer. Because the biomass‐specific P release and roach diet composition vary with fish age, it is important to consider the age structure of fish populations to obtain correct estimates of P release and net additions to the water column. 5. The removal of roach by fishing diminished the roach stock greatly, but the fish‐mediated P release to the water column changed little. This effect was because of the high compensation capacity of the roach population, leading to high recruitment of young fish with higher biomass‐specific P release rates. 6. External loading is very low during summer months and therefore it cannot explain the increase in the P concentration of water during that time. Internal loading from the sediment might be as high as 10.2 mg P m?2 day?1, i.e. 50 times higher than the maximum net P addition by the total roach population.  相似文献

Xylem recovery from embolism was studied in Laurus nobilis L. stems that were induced to cavitate by combining negative xylem pressure potentials (PX = ?1.1 MPa) with positive air pressures (PC) applied using a pressure collar. Xylem refilling was measured by recording the percentage loss of hydraulic conductance (PLC) with respect to the maximum 2 min, 20 min and 15 h after pressure release. Sodium orthovanadate (an inhibitor of many ATP‐ases) strongly inhibited xylem refilling while fusicoccin (a stimulator of the plasma membrane H+‐ATPase) promoted complete embolism reversal. So, the refilling process was interpreted to result from energy‐dependent mechanisms. Stem girdling induced progressively larger inhibition to refilling the nearer to the embolized stem segment phloem was removed. The starch content of wood parenchyma was estimated as percentages of ray and vasicentric cells with high starch content with respect to the total, before and after stem embolism was induced. A closely linear positive relationship was found to exist between recovery from PLC and starch hydrolysis. This, was especially evident in vasicentric cells. A mechanism for xylem refilling based upon starch to sugar conversion and transport into embolized conduits, assisted by phloem pressure‐driven radial mass flow is proposed.  相似文献

Abstract. Fagus grandifolia var. mexicana seedling dynamics were studied in a relict forest inside the crater of a volcano where it is the only canopy tree species. The objectives of the study were (1) to determine changes in seedling density before and after a mast year, as well as to monitor seedling recruitment, mortality and growth rate in relation to the micro‐environment, and (2) to discuss the impact of masting on seedling dynamics of the population. Before masting, seedling density was 8.8 seedlings m–2, afterwards seedling density peaked at 51.5 seedlings m–2, then decreased by 50% in 3 months and by 85% after 8 months, down to levels of before the mast year (9.9 seedlings m–2) after 16 months. Seedlings in the forest floor before the mast‐seeding event showed a low relative growth rate – 0.14 mm mm–1 mo–1 against 1.27 mm mm–1 mo–1 for seedlings that emerged immediately after masting. Seed germination was high (83%) and rapid (L50= 7 days) following masting. Seedling growth was positively correlated with soil water content but not with temperatures and relative humidity. Data suggested that seed production in mast years is important in maintaining the seedling bank, and thus the viability of relict Fagus populations.  相似文献

The restored Maltañski Reservoir was studied from its filling with water in 1990 till 2000. Total phosphorus, chlorophyll-a, and Secchi depth, as well as the Carlson's trophic state index (TSI) values based on those three parameters showed characteristic patterns of changes among seasons and years. Within each year, the lowest trophic state was usually observed in winter and the highest in summer. Because of the high loads of phosphorus received by the reservoir, this element did not limit primary production. TSI values calculated on the basis of total phosphorus were always markedly higher than calculated on chlorophyll-a and Secchi depth (similar to each other). The trophic upsurge phase lasted only a few months after the filling of the reservoir in 1990. Similar symptoms were observed after its refilling in the spring of 1993. The trophic depression phase lasted until the end of 1995. After that time a significant correlation between phosphorus concentration in the reservoir and in river waters flowing into the reservoir was observed. The successive phases of reservoir ageing, determined on the basis of phosphorus concentration, were not accompanied by changes in chlorophyll-a content. The influence of the top-down mechanism (biomanipulation effect) resulted in relatively low values of chlorophyll-a after the filling of the reservoir with water in 1990 and in 1993. As early as 1992 chlorophyll-a values reached a very high level and stayed at that level until the end of the study in 2000 (except for the short decline in 1993).

Two sudden and pronounced changes in the growth rate of roach, Rutilus rutilus (L.), in Slapton Ley, Devon, have been observed between 1972 and 1976. At first the growth rate declined to the point at which the population became stunted, then subsequently it improved again. These changes were correlated with population density; the numbers of roach were increasing during the late 1960's/early 1970's, and this increase was substantially reinforced by very strong year classes in 1972, and, to a lesser extent, in 1973. The increase in population density resulting from the strong 1972 year class was apparently adequate to accelerate the onset of the stunting observed. The improvement in growth rate in 1976 followed immediately after extensive mortalities amongst the roach during 1975, caused by an epidemic of the parasite Ligula intestinal is (L.). Some improvement in the growth of O group roach had been evident during 1975, but this was not observed in the population. Ford-Walford plots showed a decline in the value of L after the onset of stunting in 1972.
Differences in the reproductive biology of the fish were found following the alleviation of stunting; the gonads of both sexes achieved a greater relative weight, and correspondingly the fecundity of the females was higher, although this latter effect became progressively less marked with increasing size. It was not apparent amongst the largest individuals. There was also some evidence of a decline in the proportion of fish maturing at minimum size. The changes in the reproductive biology were associated with the improvement in conditions for individual fish following the decline in population density.  相似文献

Changes in zooplankton diversity and density in response to reservoir aging in Pawnee Reservoir were investigated. Zooplankton samples collected from April 1992 through April 1993, were compared to a similar study conducted after initial impoundment by Helzer (1971), in 1968–1970. Since this initial study, increases in turbidity and resulting changes in biotic interactions significantly altered the zooplankton community. A significant increase in total zooplankton density and a decrease in species richness were observed between study periods. Density increased from 24.6 to 95.4 individuals L–1, while the number of taxa declined from fourteen to ten. During this time period, Cyclops vernalis became the dominant zooplankter in the reservoir. The density of this predatory copepod increased significantly, from 0.1 l–1 in 1968–1970, to 44.3 l–1 in 1992–1993, which accounted for most of the increase in total zooplankton density. Though a greater spring maximum of another dominant, Bosmina spp. was found during the 1992–1993 study period, the annual density of this cladoceran was not significantly different between study periods. Similar trends for Daphnia ambigua and D. parvula were also observed, as greater spring maxima levels were attained, however overall annual densities were not significantly different. The dominance of C. vernalis (46% of annual density) and Bosmina spp. (33%), indicate that these two zooplankters were tolerant of changes in physical conditions resulting from reservoir aging and biotic interactions that followed in the reservoir during the 22 years between study periods.  相似文献

In ten 0.1 ha drainable ponds O+ cyprinids were stocked to assess their impact on the aquatic community. Bream and roach were stocked in early June, 1986, and carp in early July, 1986. The ponds were drained mid November, 1986. The relationship between the fish community and its food resource was estimated in terms of production and consumption with use of the data on growth and mortality of the fish. The average gross production of the fish species in the ten ponds (48.4 kg during the experiment) was dominated by the carp (58.5%). A minimum estimate of the food consumption of the O+ cyprinids is 760 kJ.m–2. The average production of bream and roach was much lower than that of carp due to a higher mortality and a more specialized feeding pattern. The gross productions of carp, bream and roach in the ten ponds were not significantly related, although stomach analysis showed diet overlap between the carp and the bream.  相似文献

In randomly grown tissue culture cells (mouse leukemic lymphoblast, L5178Y) the number, volume, and Na+ and K+ content increase as an exponential function with a doubling time of 11.3 hr. In synchronously grown cells the volume increase of the population and of single cells follows the same exponential function as in randomly grown cells. In contrast, the cation content fluctuates during a single cell cycle. About 1½ hr after the cell division burst (at the beginning of the S period), a net loss of K+ occurs for a period of about 1 hr amounting to about 20% of the total K. Over the next 5 to 6 hr, the deficit in K+ is eliminated. The Na+ content shows a double fluctuation. It falls during the cell division burst, rises when the K+ content decreases, falls again when K+ content rises, and then increases again before the next cell division burst. The net fluxes of both Na+ and K+ are very small compared to the unidirectional fluxes (less than 5%), thus small changes in the balance of influx and efflux account for the changes in cation content during the growth cycle. Both unidirectional fluxes increase dramatically (by a factor of two) about 2 hr after the cell division burst, and then remain constant until after the next cell division. The pattern of electrolyte regulation during cell division does not follow a simple function such as cell number, cell surface, or cell volume, but must be related to specific internal events in the cell.  相似文献

Short- and long-term effects of temperature on ion flux and energy turnover were studied in hepatocytes from thermally acclimated trout and roach. In trout hepatocytes K+ efflux was insensitive towards acute exposure to low temperature but was downregulated during cold acclimation of the fish so as to balance the uncompensated decreased K+(Rb+) uptake of the cells. In contrast, both K+(Rb+) uptake and K+ efflux of roach hepatocytes were temperature sensitive in the short term. These acute effects, however, were offset during cold acclimation by a near perfect compensation of both fluxes leading to re-establishment of ion flux homeostasis at the original level. Our findings, based on a new method permitting the simultaneous monitoring of K+ efflux and uptake in the same cell population, provide experimental verification of two of the three possible strategies, recently discussed by Cossins et al. (1995), by which the ionic steady state of fish cells may adjust to acute and chronic temperature change. By comparing hepatocytes from two groups of trout, one kept on a maintenance diet (ration I), the other fed ad libitum (ration II), we discovered striking effects of nutritional state on the absolute levels as well as on the temperature relationships of K+ uptake and protein synthetic activity. Both of these functions in the hepatocytes increased in the ration II fed as compared to the ration I fed trouts, but the increase of protein synthetic activity was greater and more uniform at the three experimental temperatures than that of K+ uptake. Moreover, protein synthetic activity proved to be considerably more temperature sensitive than K+ uptake and, in contrast to the latter, showed a compensatory response after cold acclimation. The population growth pattern and related changes in both the nitrogen and phosphorus contents in the cell of the dinoflagellate Peridinium penardii (Lemm.) Lemm., which formed a freshwater red tide in a reservoir, were studied in situ. An exponential increase with time in population density was found. A specific growth rate of 0.25 d–1 was observed. The cellular content of phosphorus per cell decreased from 6.0 × 10–5 µg to 9.2 × 10–6 µg along with an increase in population density from 8.0 × 102 cells ml–1 to 2.5 × 104 cells ml–1. A prominent change in the cellular nitrogen did not occur. Decreasing cell content and continuous uptake of phosphorus were advantageous for P. penardii to form a freshwater red tide under P-limited conditions.  相似文献

We have addressed the hypothesis that pathogen-associated immunomodulatory molecules may influence anti-tumor immunity through their pro- and anti-inflammatory activities and abilities to induce effector and regulatory T (Treg) cells. We found that CpG oligonucleotides (CpG) and cholera toxin (CT), which promote Th1 or Th2/Treg cell biased responses, respectively, had differential effects on tumor growth. Therapeutic peritumoral administration of CpG significantly reduced subcutaneous tumor growth and prolonged survival, whereas CT enhanced tumor growth and reduced survival. Peritumoral administration of CpG enhanced the frequency of IFN-γ-secreting and reduced IL-10-secreting CD4+ and CD8+ T cells, in the tumor and in the draining lymph nodes, whereas, CT significantly enhanced the frequency of CD4+CD25+Foxp3+ Treg cells, but reduced IFN-γ-secreting T cells infiltrating the tumor. In contrast to the beneficial effect of CpG in mice with subcutaneous tumors, CpG or CT had no protective effect against tumor growth in the lungs when given therapeutically by the nasal route. However, prophylactic intranasal administration of CpG significantly reduced the number of lung metastases and this was associated with an enhanced frequency of IFN-γ-secreting CD8+ T cells in the draining lymph node and enhanced tumor-specific CTL responses. Our findings demonstrate that pathogen-associated molecules can either inhibit or enhance anti-tumor immunity by selectively promoting the induction of effector or regulatory T cells, and that the environment of the growing tumor influences the protective effect. Summary In vitro effects of methotrexate (MTX) on interleukin-2(IL-2)-mediated cytotoxicity of peripheral blood mononuclear cells (PBMC) were studied. PBMC were incubated with human recombinant IL-2 (25 U/ml) for 72 h; during the last 24 h, various concentrations (10 pM–1 µM) of MTX were added to the culture. Cytotoxicity against k562 cells was measured by a 4-h51Cr-release assay. The IL-2-mediated cytotoxicity was paradoxically increased at around a concentration (10 nM) MTX. Such a low concentration of MTX showed no anti-proliferative effect on cell growth. This enhancement with 10 nM MTX was shown only in an E-rosette+ (E+) population, but not in E-rosette (E). In addition, when E+ cells were treated with an anti-CD16 monoclonal antibody plus complement after incubation with IL-2 and MTX, MTX-induced enhancement was lost, suggesting that an E+CD16+ cell population was mainly involved in this augmentation. Positively sorted E+CD16+ cells showed similar enhancement of cytotoxicity after treatment with IL-2 plus MTX. On the other hand, MTX treatment did not show the phenotypical changes including of the E+CD16+ cells, indicating that this treatment did not affect the differentiation and proliferation of the specific cell subset. The (Na+---K+)-stimulated Mg2+-dependent ATPase activities of 3T3 and SV40 transformed 3T3 cells were compared as a function of cell population density. For normal cells the enzyme activity remained relatively constant during exponential growth, but decreased sharply coincident with contact inhibition of growth at confluence. This decrease in activity could be reversed by stimulating contact-inhibited cultures to undertake renewed short-term growth either by adding fetal calf serum or changing the medium completely. Transformed cells did not experience a decrease in (Na+---K+)-ATPase activity upon reaching confluence, but this is consistent with the fact that they were still growing exponentially at this stage. However, non-confluent cultures of both normal and transformed cells incurred a marked decrease in levels of the enzyme when growth was inhibited by serum depletion. The results have been interpreted as indicating that levels of (Na+---K+)-ATPase in both normal and transformed cells are correlated with growth. Hence the different patterns of ATPase activity displayed by malignant cells and their normal counterparts with increase in cell number appear to be a reflection of their dissimilar growth behaviours rather than of any innate difference between them.  相似文献

Objective

To determine if the density of FOXP3+ lymphocytes in primary tumors and lymph nodes in pancreatic cancer correlates with the presence of lymph node metastases.

Methods

FOXP3+ lymphocyte density in primary pancreatic cancer tissue and draining lymph nodes was measured using immunohistochemistry. We analyzed the clinical and pathological aspects associated with the accumulation of FOXP3+ lymphocytes in pancreatic cancer. We also analyzed the correlation of density of FOXP3+ lymphocytes in lymph nodes with the nodal status and distance from the primary tumor.

Results

FOXP3+ lymphocyte density in pancreatic cancer was significantly higher than in paratumoral pancreatic tissue. The density of FOXP3+ lymphocytes in local tumor tissue correlated significantly with the histological grade and overall lymph node status. Furthermore, FOXP3+ lymphocyte density was significantly higher in positive lymph nodes than in negative ones, while it had no correlation with the distance of the lymph node from the primary tumor.

Conclusion

FOXP3+ lymphocyte density in primary tumor tissue in patients with pancreatic cancer correlates with lymph node metastasis. Lymph nodes containing metastases having higher FOXP3+ lymphocyte densities than do negative lymph nodes.  相似文献

Growth measurements of hormone-treated roots from willow cuttings were combined with electrophysiological recordings to study hormone-induced changes in membrane potential and in endogenous ion currents. The mean growth rate of roots was 10 ± 2 μm min?1 in regular nutrient solution. It increased to 13 ± 2 μm min+1 after application of spermine and decreased to 0.07 ± 0.01 μm min?1 after treatment with abscisic acid (ABA). Transient depolarizations were elicited in root cortex cells by spermine, while ABA caused a transient hyperpolarization. All changes in membrane potential were accompanied by transient responses of the endogenous current. These responses suggest that first anions, then cations leave the root during spermine-induced depolarizations. From the changes of the endogenous current an apparent efflux of anions (presumably Cl?) and cations (presumably K+) of 200 to 700 pmol cm?2 per depolarization was calculated. To further investigate a possible relation between endogenous ion currents, growth and the growth regulators ABA and spermine, long-lasting extracellular vibrating-probe measurements were performed. Control roots showed an inward current of about 1.5 μA cm?2 at the apical elongation zone and an outward current with a maximum density of 1.3 μA cm?2 at the central and basal elongation zone. The addition of ABA and spermine (final concentration 0.1 mM) to the bathing medium affected the endogenous current in opposite ways: ABA caused a reduction of inward and outward current, while spermine stimulated both. Since protons are a major component of the endogenous current, and sucrose can be taken up by root cells from the apoplast via symport with H+, a role of the endogenous current in growth regulation is indicated.  相似文献

In appropriate environments containing 2-monochloropropionic acid (2MCPA), mutations in a population of nondehalogenatingPseudomonas putida, strain PP40-040 (parent population), resulted in the formation of 2mcpa+ papillae as a result of the decryptification of adehII gene. Increasing the size of the parent population, for example by increasing the availability of a metabolizable substrate such as succinate or lactate, increased the number of 2mcpa+ papillae formed because there were more parent cells available for mutation to the 2mcpa+ phenotype. The presence of a dehalogenating population, such asP. putida strain PP3, in close proximity to the non-dehalogenating population, also increased the number of 2mcpa+ papillae formed. This was due to the excretion of dehalogenases into the growth medium, which caused localized dehalogenation of the available 2MCPA, yielding a metabolizable substrate. This substrate stimulated the growth of the non-dehalogenating population, in turn increasing the number of 2mcpa+ papillae formed. Barriers, such as dialysis membranes, which prevented the excretion of the dehalogenases into the growth medium, prevented the stimulation of 2mcpa+ papillae formation by preventing release of metabolizable substrates from 2MCPA breakdown. Cell-free extracts (CFE) from dehalogenase-producing populations had a similar effect for the same reason. CFE without dehalogenase activity or in which the dehalogenase activity had been destroyed by heating failed to stimulate parent population growth and 2mcpa+ papillae formation. In the case ofPseudomonas putida strain PP3, which carries an easily transposed dehalogenase-encoding transposon, treatment of CFE with DNAase eliminated an additional factor involved in the formation of 2mcpa+ papillae.The authors are with the School of Pure and Applied Biology, University of Wales-Cardiff, P.O. The adenovirus 7-simian virus 40 hybrid virus population E46+ was subjected to fixed-angle equilibrium density gradient centrifugation in CsCl. A difference in buoyant density between the hybrid virion and its nonhybrid adenovirus 7 counterpart was noted, the hybrid virion possessing the lower buoyant density. This difference in buoyant density appeared to be accentuated in a population of adenovirus 2+t7, a derivative of E46+ in which the adenovirus 7-simian virus 40 genome had been transferred to an adenovirus 2 capsid.  相似文献
