采用CO_2激光(波长10.6μm,功率10 w,光束长74 cm)辐照拟微绿球藻(Nannochloropsis sp.YW0980),辐照时间为30 s、60 s、90 s,通过测定藻色素、多糖、蛋白质及油脂含量,研究CO_2激光对藻的生物学效应。结果表明:30 s、60 s、90 s辐照条件下,对拟微绿球藻细胞生长及代谢产物均有一定的促进作用,其中CO_2激光60 s处理组有利于拟微绿球藻的生长及色素的积累,但30 s剂量下更有利于多糖、蛋白质及油脂含量的积累,分别比对照组提高了51.05%(胞外多糖),289.45%(总多糖)、37.05%、172.16%。  相似文献

本文叙述利用柑桔果皮培养黑曲霉进行蛋白质生产的试验。包括培养时间(积温℃·天),无机氨态氮和菌种接种量对干重率、氨态氮转化率和蛋白质产量的影响。并对真菌蛋白产生过程中干物质和氮代谢的平衡参数进行了综合评价。经过适当条件的培养。柑桔果度可由8.3%的起始蛋白质含量(干重)增加到20%以上的蛋白饲料。  相似文献

采用分级提最方法提取银杏(Ginkgo biloba L.)种子和银杏叶蛋白质,并进行各组分蛋白质含量测定和银杏种子后熟过程中蛋白质含量动态变化的分析。结果表明,银杏种子以水溶性和盐溶性蛋白质为主,银杏叶以醇溶性蛋白质为主。银杏叶中主要蛋白质在HPLC柱中的保留时间为3.457min,相对含量达70%以上。银杏叶蛋白质含有丰富的亮氨酸、缬氨酸、赖氨酸和色氨酸,其含量分别为20.23%、13.35%、4.81%和3.73%。萌发种子胚体中的蛋白质主要是醇溶性蛋白质和谷蛋白类蛋白质。在种子萌发过程中,胚乳蛋白质含量明显增加,播种后第3周和萌发时总蛋白质含量达到高峰。  相似文献

运用近红外法预测了中国绿豆主要营养品质蛋白质、淀粉和直链淀粉含量并比较绿豆粉和籽粒两种不同样品类型的差异。102份来自绿豆核心样品为试验材料,采用近红外分析系统扫描了绿豆籽粒和粉样品。光谱数据经预处理后,构建了最小二乘回归预测和交互验证模型并获得了最优校正统计参数。绿豆粉样品最大R2值和最小SECV值蛋白质含量为0.95和0.329,淀粉含量0.90和0.576,直链淀粉含量0.89和0.307;RPD值3.08至4.61。籽粒样品最大R2值和最小SECV值蛋白质含量为0.90和0.404,淀粉含量0.88和0.643,直链淀粉含量0.85和0.426;RPD值2.51至3.23。模型的稳健性采用外部验证法进行了评价。豆粉样品的平均差异1.0%~1.8%比籽粒样品略低。结果表明绿豆粉的三种组分的近红外预测方法具有快速和简单的特点,可应用于绿豆品质的测定。籽粒样品还具有无损种子结构,保持种子活力的特点,可在育种、种质资源创新等方面应用,但其准确度还有待进一步的改善。  相似文献

本文以新鲜的金桂花(Osmanthus fragrans Lour)为实验材料,采用微波-蒸馏萃取法提取桂花中的精油。通过改变微波-蒸馏萃取法中的各种工艺参数(匀浆时间、料液比、微波时间),得到相应工艺参数下的桂花精油提取率。采用单因素试验和正交试验,分析提取工艺过程中的参数和提取率得到最优化的桂花精油提取工艺条件,即匀浆时间4min、料液比1∶5、微波时间60s,同时桂花精油提取率为0.85%。根据对桂花精油的理化性质分析可知,实验所得桂花精油样品香气较为纯正,液体呈橘黄色,酸值大约为7.50,酯值大约为18.03,达到精油的国标标准且质量较好。对桂花精油采用磷钼络合物法进行抗氧化性的研究,表明桂花精油的抗氧化活性较强,同时桂花精油的浓度与抗氧化性成正比关系。  相似文献

本文采用气相色谱法测定了几种生物制品中苯酚含量。样品无需进一步处理,加入内标液后直接上柱测定,色谱峰形良好,线性范围宽,平均回收率和变异系数分别为101.8% 和1.085% ,最小检测限为10 μg/m l,本法操作简单,样品用量少,可作为测定生物制品中苯酚含量的一种常规方法。  相似文献

以氨基酸自动分析仪测定稻米赖氨酸含量为参照标准。在“茚三酮测定赖氨酸含量”(A法)的基础上,对该法中大米蛋白质的提取温度和时间、色温度和时间及茚三酮用量等条件进行了探讨,提出了稻米赖氨酸含量快速测(B法)。提取条件为90℃5分钟,显色条件为90℃20分钟;茚三酮试剂用1 ml。用B法对10个大米样品进行测定,赖氨酸含量与氨基酸分析仪测得的赖含量基本上一致,无显著差异(P>0.05)。  相似文献

不同温度段核桃壳木醋液的组分分析及生物活性研究   总被引:2,自引:0,他引:2  
以核桃壳为材料,根据木材裂解规律、不同温度段木醋液得率和颜色的不同,收集精制得90℃~190℃(K1)、190℃~300℃(K2)、300℃~480℃(K3)及90℃~480℃(K)温度段木醋液。采用种子萌发法对精制后各木醋液进行生物活性测定,GC-MS测定脱水后的K1、K2和K3中有机组分,并利用核磁共振技术对其中的主要成分进行验证。结果显示:不同温度段木醋液含有的有机组分不同,含量差别大。K1、K2和K3分别检出13种、42种和37种物质,11种共有物质的相对总含量分别为76.38%、59.83%和57.08%,其中乙酸的相对含量随收集温度的升高逐渐减少。种子萌发试验表明,K1稀释1 000倍、K2稀释25 000倍时,对小麦根、苗生长和干物质的积累有显著促进作用,K和K3在供试范围对苗长、根长和干重都呈不同程度的抑制作用。  相似文献

The biomass yield, cellulolytic activity, and protein recovery using Aspergillus terreus GN1 with alkali-treated sugarcane bagasse was studied using different levels (250-600 mg of N/L of broth) of organic and inorganic nitrogen sources. e.g., cattle urine, urea, cornsteep liquor, ammonium sulfate, ammonium nitrate, ammonium iron sulfate, ammonium chloride, and sodium nitrate. Among different levels of alkali-treated bagasse substrate concentrations (0.5-4.0% w/v) tested, 1.0% substrate yielded the highest crude protein content, protein recovery, and cellulolytic activity. The biomass recovery with 1.0% substrate ranged from 290-380 mg/500 mg bagasse substrate in a 50-mL broth with a nitrogen level of 250-600 mg of N/L in all the sources except ammonium iron sulfate, which yielded 402-439 mg/500 mg bagasse substrate. However, crude protein content of biomass obtained with an ammonium iron sulfate nitrogen source was the lowest. Cornsteep liquor nitrogen source at the rate of 600 mg of N/L yielded the maximum crude protein of 32.9%, protein recovery of 22.2 g/100 g of bagasse, and carboxymethyl cellulase and filter paper enzyme activities of 1.1 and 0.09 units/mL, among the organic and inorganic nitrogen sources studied. In general, the organic nitrogen sources and inorganic nonammonium nitrogen sources were utilized preferentially for protein production over the inorganic ammonium nitrogen sources. The fermentation time required under optimum cultural and nutritional conditions for A. terreus GN1 was also evaluated. The crude protein content of the biomass increased gradually up to the seventh day of fermentation, but the protein recovery rate was high up to two or three days. It was observed that the cellulose utilization rate increased after an initial lag of one day up to the third day and gradually increased further, which corresponded positively with protein content, biomass protein recovery, and cellulase enzyme activity. On the seventh day of fermentation, the crude protein content, biomass protein recovery, water-soluble carbohydrate, bagasse cellulose utilization, CMCase, and FPase activities were 32.8%, 20.1 g/100 g of bagasse, 6.2%, 82.7%, 1.0. and 0.08 U/mL, respectively. The final biomass recovered contained 32.8% crude protein content and had an in vitro rumen digestibility (IVRD) coefficient of 68.8%. The biomass contained almost all the essential and nonessential amino acids and was comparable with FAO reference protein. It is concluded that a fermentation time of 72 h gave a faster rate of protein production of 16.9 g/100 g of bagasse with 69.8% bagasse cellulose utilization with 76.0% IVRD. and contained almost all the essential and nonessential amino acids.  相似文献

Three-phase partitioning (TPP), a technique used in protein purification, was used to purify invertase from tomato (Lycopersicon esculentum). The method consists of simultaneous addition of ammonium sulfate and t-butanol to the crude enzyme extract in order to obtain the three phases. Different parameters (ammonium sulfate saturation, crude extract to t-butanol ratio and pH) essential for the extraction and purification of invertase were optimized to get highest purity fold and yield. It was seen that, 50% (w/v) ammonium sulfate saturation with 1:1 (v/v) ratio of crude extract to t-butanol at pH 4.5 gave 8.6-fold purification with 190% activity recovery of invertase in a single step. Finally, the purified enzyme was also characterized and the general biochemical properties were determined. The sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis of enzyme showed considerable purification and its molecular weight was nearly found to be as 20 kDa. This work shows that, TPP is a simple, quick and economical technique for purification of invertases.  相似文献

【背景】蛋白饲料的缺乏,促进了蛋白含量高、安全性能好的酵母类单细胞蛋白的研究与应用。【目的】筛选氨氮利用能力强的菌株,为单细胞蛋白的发酵提供优良菌株。【方法】从土壤、奶制品、水果采集样品分离酵母菌,根据形态学和分子生物学鉴定菌株,然后以硫酸铵为唯一氮源培养基,测定菌落大小、菌体干重、蛋白质含量,复筛氨氮利用率高的酵母菌,并对复筛菌株氨同化相关酶活性进行测定。【结果】经过形态学、分子生物学鉴定和氨氮利用能力评价,获得3株高氨氮利用的酵母菌,分别是胶红酵母(Rhodotorula mucilaginosa)、酿酒酵母(Saccharomyces cerevisiae)和戴尔有孢圆酵母(Torulaspora delbrueckii)。通过比较3株酵母菌的谷氨酸脱氢酶、谷氨酸合成酶、谷氨酰胺合成酶活性,酿酒酵母的3种酶活性最高,其次是胶红酵母。【结论】从奶酪和西瓜中分离的胶红酵母N5和酿酒酵母J1具有较强的氨氮利用能力以及酶活性,可为单细胞蛋白发酵提供优良菌株。  相似文献

The Ammonia Recovery Process (ARP) is an award-winning, low-cost, environmentally responsible method of recovering nitrogen, in the form of ammonia, from various dilute waste streams and converting it into concentrated ammonium sulfate. The ThermoEnergy Biogas System utilizes the new chemisorption-based ARP to recover ammonia from anaerobically digested wastes. The process provides for optimal biogas production and significantly reduced nitrogen levels in the treated water discharge. Process flows for the ammonia recovery and ThermoEnergy biogas processes are presented and discussed. A comparison with other techniques such as biological nitrogen removal is made. The ARP technology uses reversible chemisorption and double salt crystal precipitation to recover and concentrate the ammonia. The ARP technology was successfully proven in a recent large-scale field demonstration at New York City's Oakwood Beach Wastewater Treatment Plant, located on Staten Island. This project was a joint effort with Foster Wheeler Environmental Corporation, the Civil Engineering Research Foundation, and New York City Department of Environmental Protection. Independent validated plant data show that ARP consistently recovers up to 99.9% of the ammonia from the city's centrate waste stream (derived from dewatering of sewage sludge), as ammonium sulfate. ARP technology can reduce the nitrogen (ammonia) discharged daily into local bodies of water by municipalities, concentrated animal farming operations, and industry. Recent advances to ARP enhance its performance and economic competitiveness in comparison to stripping or ammonia destruction technologies.  相似文献

A previous study indicated that the oxidative assimilation capacity of a heterogeneous microbial population for acetic acid did not return to the initial value after a period of endogenous respiration in the presence of ammonium sulfate. In view of this finding it was not possible to recommend fully the use of a continuous oxidative assimilation process for the treatment of nitrogen-deficient waste waters of noncarbohydrate nature. To put the process to a severe test, a laboratory-scale pilot plant study was done. Acetate (1,000 mg/liter) was fed continuously to a completely mixed, aerated reactor vessel, from which the mixed liquor was channeled to a settling basin. The settling basin supernatant fluid was continuously discharged, and a portion of the settled biological sludge was subjected to endogenous respiration in the presence of ammonium sulfate and was recycled to the aeration vessel. Experiments were conducted at three levels of ammonia nitrogen in the endogenous phase. Approximately 90% removal of the carbon source (expressed as chemical oxygen demand, COD) was attained with a low level of supplemental nitrogen (COD/N = 70:1) and a fairly low reactor detention time (4 hr). Based upon these and previous results, it has been concluded that the process shows promise for application to the treatment of a wide variety of nitrogen-deficient industrial wastes.  相似文献

采用微波辅助提取、溶剂浸提 ,以正己烷、乙醇、正己烷 +乙醇 (体积比 1∶1 )为溶剂从艾蒿ArtemisiaargyiL啨vl.etVant茎叶得到了提取物。另外 ,还用水蒸气蒸馏法获得了精油。生物活性试验表明 ,它们对菜青虫PierisrapaeL .具有较强的拒食活性和一定的触杀活性。其中 ,正己烷为溶剂的微波辅助提取物的拒食活性和触杀活性最强。  相似文献

Traditional technology of cell disruption has become one of the bottlenecks restricting the industrialization of genetic engineering products due to its high cost and low efficiency. In this study, a novel bioprocess of phage lysis coupled with salting-out extraction (SOE) was evaluated. The lysis effect of T7 phage on genetically engineered Escherichia coli expressing κ-carrageenase was investigated at different multiplicity of infection (MOI), meanwhile the phage and enzyme released into the lysate were separated by SOE. It was found that T7 phage could lyse 99.9% of host cells at MOI = 1 and release more than 90.0% of enzyme within 90 min. After phage lysis, 87.1% of T7 phage and 71.2% of κ-carrageenase could be distributed at the middle phase and the bottom phase, respectively, in the SOE system composed of 16% ammonium sulfate and 20% ethyl acetate (w/w). Furthermore, κ-carrageenase in the bottom phase could be salted out by ammonium sulfate with a yield of 40.1%. Phage lysis exhibits some advantages, such as mild operation conditions and low cost. While SOE can efficiently separate phage and intracellular products. Therefore, phage lysis coupled with SOE is expected to become a viable alternative to the classical cell disruption and intracellular product recovery.  相似文献

应用水、乙醇和硫酸铵三者形成双水相系统,并利用黄酮在乙醇中的溶解度要大于水中的溶解度,以脱除其中一些易溶于水和在高浓度乙醇中溶解度低的物质。尝试了不同盐、不同体积比的乙醇和水、以及盐加入量对分相的影响并测定了其在不同情况下的纯化效果。发现在硫酸铵中的分相情况最好,在加入量到2.5g时既可以达到分相完全。并测定:(1)在5mL乙醇+5mL水+5g硫酸铵+0.15g粗体物(粗体物中黄酮的百分含量为25%)的双水相中,黄酮类物质的回收率为89.2%,而黄酮百分含量提升为47.6%;(2)在4mL乙醇+5mL水+5g硫酸铵+0.15g粗体物(粗体物中黄酮的百分含量为25%)的双水相中,黄酮类物质的回收率为81.9%,而黄酮含量提升为50.9%。水、乙醇和硫酸铵三者形成双水相系统应用于山楂叶中黄酮类物质的纯化在文献中少有报道,而本文应用此法可以将黄酮含量从25%提升到将近47.6%,并且回收率为89%。  相似文献

A mathematical model was developed to describe a dialysis process for the continuous fermentation of whey lactose to lactic acid, with neutralization to a constant pH by ammonia. In the process, whey of a relatively high concentration is fed into the fermentor circuit at a relatively low rate so that the residual concentration of lactose is low. The fermentor effluent contains ammonium lactate, bacterial cells, and residual whey solids and could be used as a nitrogen-enriched feedstuff for ruminant animals. Only water is fed into the dialysate circuit at a relatively high rate. The dialysate effluent contains purified ammonium lactate and could be converted to lactic acid and ammonium sulfate for industry. The fermentation was specifically modeled as a set of equations representing material balances and rate relationships in the two circuits. Dialysis continuous fermentations, in general, were modeled by combining these equations and by using dimensionless parameters. The generalized model was then solved for the steady state and used to simulate the specific fermentation on a digital computer. The results showed the effects of various material and operational and kinetic parameters on the process and predicted that it could be operated efficiently.  相似文献

The transformed glucocorticoid receptor (GR) from rat liver precipitated at 30% saturation of ammonium sulfate and sedimented at 4.3 S on glycerol gradient centrifugation, whereas the nontransformed GR precipitated at higher concentrations of ammonium sulfate (40-50% saturation) and sedimented at 8.6 S on a gradient. Sodium dodecyl sulfate polyacrylamide gel electrophoresis showed that heat shock protein 90 (hsp 90) precipitated at 40-50% saturation of ammonium sulfate. Moreover, hsp 90 and the nontransformed GR were eluted from DEAE high performance ion-exchange chromatography at similar salt concentrations (0.22-0.23 M NaCl), whereas the transformed GR was eluted at 0.1 M NaCl. Therefore, hsp 90 seems to be responsible for the surface charge characteristics of the nontransformed GR.  相似文献
