Effects of polyethylene glycol (PEG)-induced osmotic stress on the mechanical properties of cell walls and the levels of their components were investigated along intact wheat (Triticum aestivum L.) coleoptiles. Stress-relaxation analysis showed that the cell walls of stressed coleoptiles were loosened as compared with those of unstressed ones not only in the apical but in the basal regions. The amounts of wall-bound ferulic acid (FA) and diferulic acid (DFA) of stressed coleoptiles were substantially lower than those of unstressed ones in all regions. The cellulose and hemicellulose contents increased toward the coleoptile base. Osmotic stress reduced the cellulose content in the basal region but it slightly affected the hemicellulose content. The molecular weight of hemicellulose in the apical region of stressed coleoptiles was higher than that of unstressed ones, while that in the basal region was almost the same in both coleoptiles. FA, DFA and cellulose contents correlated with the cell wall mechanical property. The amount and molecular weight of hemicellulose, however, did not correlate. These results suggest that the reduced levels of FA and DFA in all regions and cellulose in the basal region of wheat coleoptiles are involved in maintaining the cell wall extensibility under osmotic stress.  相似文献

The relationship between the formation of cell wall-bound ferulic acid (FA) and diferulic acid (DFA) and the change in activities of phenylalanine ammonia-lyase (PAL) and cell wall-bound peroxidase (CW-PRX) was studied in rice shoots. The length and the fresh mass of shoots increased during the growth period from day 4 to 6, while coleoptiles ceased elongation growth on day 5. The amounts of FA and DFA isomers as well as cell wall polysaccharides continued to increase during the whole period. The activities of PAL and CW-PRX greatly increased in the same manner during the period. There were close correlations between the PAL activity and ferulate content or between the CW-PRX activity and DFA content. The expression levels of investigated genes for PAL and putative CW-PRX showed good accordance with the activities of these enzymes. These results suggest that increases in PAL and CW-PRX activities are cooperatively involved in the formation of ferulate network in cell walls of rice shoots and that investigated genes may be, at least in part, associated with the enzyme activities. The substantial increase in such network probably causes the maturation of cell walls and thus the cessation of elongation growth of coleoptiles.  相似文献

Application of abscisic acid (ABA) to dark-grown wheat (Triticumaestivum L.) roots interfered the cell wall hardening of coleoptilesduring several days of the treatment. Although the amounts ofwall-bound diferulic (DFA) and ferulic (FA) acids in coleoptilesincreased as the coleoptiles grew, ABA substantially reducedtheir increases. When ABA was removed, however, these contentsincreased and reached levels near those of control coleoptiles.A close correlation was observed between the levels of DFA andFA and the mechanical properties of cell walls. The ratio ofthe amount of DFA to FA was almost constant irrespective ofgrowth conditions. The activities of phenylalanine- (PAL) andtyrosine-ammonia-Iyase (TAL) increased rapidly in the controlcoleoptiles. ABA greatly reduced the increases in these enzymeactivities. In response to ABA removal, the enzyme activitiesincreased rapidly. There was a close correlation between theincrease in FA level and the changes in enzyme activities. Theseresults suggest that ABA suppresses the increases in PAL andTAL activities in wheat coleoptiles, resulting in the reducedlevel of wall-bound FA, which, in turn, may cause the reducedDFA level and thereby maintain cell wall extensibility. To elucidate the mechanism by which white fluorescent light (5 W m-2) stimulates the formation of diferulic acid (DFA) in cell walls, the effect of light on phenylalanine-and tyrosine-ammonia-lyase (PAL, EC 4.3.1.5 and TAL, EC 4.3.1.5) and peroxidase activities was studied using coleoptiles of maize ( Zea mays L. cv. Cross Bantam T51). Growth rate of dark-grown coleoptiles was highest at the basal zone and decreased towards the tip, while continuous irradiation caused an inhibition of growth, especially at the basal zone. Light decreased the cell wall extensibility in all zones of the coleoptile. The amounts of DFA, ferulic acid (FA) and p -coumaric acid ( p -CA) increased by severalfold in cell walls of light-grown maize coleoptiles as compared with those grown in the dark. Strong correlations were observed between the increase in the contents of either DFA, FA or p -CA and the decrease in cell wall extensibility. Light decreased the wall-bound peroxidase activity. No correlation was found between DFA content and peroxidase activity. The activities of PAL and TAL were enhanced upon white light irradiation. The increment in either DFA, FA or p -CA content was correlated with an increase in PAL activity, but not with that in TAL activity. White light may promote DFA formation in the cell walls of maize coleoptiles by enhancing PAL activity.  相似文献

Application of 60 mM polyethylene glycol (PEG) to dark-grownwheat (Triticum aestivum L.) roots substantially reduced growthof coleoptiles. However, when PEG was removed, the growth rateof these coleoptiles greatly increased. Cell walls of stressedcoleoptiles remained loosened as compared with those of unstressedones. The osmotic potential of the stressed coleoptiles decreasedto that of the 60 mM PEG solution. On the other hand, the extentof decrease in the osmotic potential of stressed roots was smallerthan that of stressed coleoptiles. The osmotic potential difference() between the cell sap and the incubation medium of stressedroots was substantially higher than that of unstressed ones.The amount of ink moved from roots, where it was applied, tothe apical region of coleoptiles was significantly reduced underosmotic stress conditions. When water was exogenously appliedto abraded coleoptiles, the growth of these stressed coleoptileswas greatly promoted. These results suggest that inhibitionof coleop-tile growth under osmotic stress conditions is notdirectly related to a decrease in cell wall extensibility orto loss of the capacity to maintain osmotic potential gradients,but is caused by the reduction of the water supply from theroots to the coleoptiles. Network structures created by hydroxycinnamate cross-links within the cell wall architecture of gramineous plants make the cell wall resistant to the gravitational force of the earth. In this study, the effects of microgravity on the formation of cell wall-bound hydroxycinnamates were examined using etiolated rice shoots simultaneously grown under artificial 1 g and microgravity conditions in the Cell Biology Experiment Facility on the International Space Station. Measurement of the mechanical properties of cell walls showed that shoot cell walls became stiff during the growth period and that microgravity suppressed this stiffening. Amounts of cell wall polysaccharides, cell wall-bound phenolic acids, and lignin in rice shoots increased as the shoot grew. Microgravity did not influence changes in the amounts of cell wall polysaccharides or phenolic acid monomers such as ferulic acid (FA) and p-coumaric acid, but it suppressed increases in diferulic acid (DFA) isomers and lignin. Activities of the enzymes phenylalanine ammonia-lyase (PAL) and cell wall-bound peroxidase (CW-PRX) in shoots also increased as the shoot grew. PAL activity in microgravity-grown shoots was almost comparable to that in artificial 1 g-grown shoots, while CW-PRX activity increased less in microgravity-grown shoots than in artificial 1 g-grown shoots. Furthermore, the increases in expression levels of some class III peroxidase genes were reduced under microgravity conditions. These results suggest that a microgravity environment modifies the expression levels of certain class III peroxidase genes in rice shoots, that the resultant reduction of CW-PRX activity may be involved in suppressing DFA formation and lignin polymerization, and that this suppression may cause a decrease in cross-linkages within the cell wall architecture. The reduction in intra-network structures may contribute to keeping the cell wall loose under microgravity conditions.  相似文献

Oxalate oxidase (OXO) utilizes oxalate to generate hydrogen peroxide, and thereby acts as a source of hydrogen peroxide. The present study was carried out to investigate whether apoplastic OXO modifies the metabolism of cell wall-bound ferulates in wheat seedlings. Histochemical staining of OXO showed that cell walls were strongly stained, indicating the presence of OXO activity in shoot walls. When native cell walls prepared from shoots were incubated with oxalate or hydrogen peroxide, the levels of ester-linked diferulic acid (DFA) isomers were significantly increased. On the other hand, the level of ester-linked ferulic acid (FA) was substantially decreased. The decrease in FA level was accounted neither by the increases in DFA levels nor by the release of FA from cell walls during the incubation. After the extraction of ester-linked ferulates, considerable ultraviolet absorption remained in the hemicellulosic and cellulose fractions, which was increased by the treatment with oxalate or hydrogen peroxide. Therefore, a part of FA esters may form tight linkages within cell wall architecture. These results suggest that cell wall OXO is capable of modifying the metabolism of ester-linked ferulates in cell walls of wheat shoots by promoting the peroxidase action via supply of hydrogen peroxide.  相似文献

White fluorescent light (5 W m−2) inhibited Avena coleoptile growth. Light caused in increase in minimum stress relaxation time and a decrease in extensibility (strain/load) of coleoptile cell walls. Light increased the contents of ferulic acid (FA) and diferulic acid (DFA) ester-linked to the hemicellulose I in cell walls. These changes in the phenolic contents correlated with those of the mechanical properties of cell walls, suggesting that light stimulates the formation of DFA in hemicellulose I, making cell walls rigid, and thus results in growth inhibition. The ratio of DFA to FA was almost constant in the dark, but decreased in light, although it was almost constant in Oryza coleoptiles either in the dark or in light (Tan et al. 1992). From this fact, it is speculated that in the light condition, the formation of DFA in cell walls is limited in the step of the peroxidase catalyzed coupling reaction to produce DFA, while in the dark it is limited in the step of the feruloylation of hemicellulose I.  相似文献

Changes in the amount and composition of cell wall constituents in response to continuous hypergravity stimuli were studied in wheat ( Triticum aestivum L.) coleoptiles. The lengths of coleoptiles grown under hypergravity (300  g ) conditions for 2–4 days from germination stage were 60–70% of those of 1  g control. However, the net amounts of hemicellulosic polysaccharides and cellulose in hypergravity-treated coleoptiles increased progressively as much as those in the control coleoptiles. As a result, their contents per unit length of coleoptile largely increased under hypergravity conditions. In the hemicellulose fraction, the amounts of arabinose and xylose, the major components of the fraction, prominently increased in response to hypergravity. When hemicellulosic polysaccharides were separated into neutral and acidic polymers by an anion-exchange column, the amounts of the acidic fraction consisting of (glucurono)arabinoxylans were higher in hypergravity-treated coleoptiles than in control coleoptiles. The amounts of cell wall-bound ferulic acid and diferulic acid (DFA) increased dramatically in both 1  g control and hypergravity-treated coleoptiles. Particularly, the amounts of DFA in hypergravity-treated coleoptiles were significantly higher than those in control coleoptiles during the incubation period. These results suggest that continuous hypergravity increases the rigid network structures via arabinoxylan–hydroxycinnamate cross-links within cell wall architecture in wheat coleoptiles. These structures may have a load-bearing function and contribute to construct the stable cell wall against the gravitational force.  相似文献

Suspension-cultured barley cells responded to treatments with crude yeast extract and purified glucan preparation by rapidly and transiently (4 h postelicitation) inducing L-phenylalanine ammonia-lyase activity. Similarly, treatment of cell cultures with chitosan resulted in increased phenylalanine ammonia-lyase activity 2–4 h after elicitation, whereas a mycelium preparation of a fungal pathogen, Bipolaris sorokiniana, and purified chitin caused a more delayed induction of phenylalanine ammonia-lyase (8 h postelicitation). The most abundant of the plant cell wall degrading enzymes produced by Bipolaris sorokiniana, β-1,4-xylanase, had only a weak elicitor activity in barley cells suggesting that fungal cell wall components rather than the hydrolytic enzymes secreted by the fungus function as recognizable components that cause barley cells to induce defences. Treatment of the elicited cells with a phenylalanine ammonia-lyase inhibitor, α-aminooxy-β-phenylpropionic acid, resulted in the superinduction of the enzyme indicating the blocking of the feedback regulation mechanisms, whereas in the presence of 1 mM trans-cinnamic acid the elicitor-induction of phenylalanine ammonia-lyase was completely inhibited. Elicitor treatments increased the accumulation of wall-bound phenolics as evidenced by phloroglucinol-HCl staining and thioglycolic acid methods. However, α-aminooxy-β-phenylpropionic acid applied in combination with the elicitor did not prevent the accumulation of phenolics in barley cell walls. This suggested that phenylalanine ammonia-lyase might not play an important role in the synthesis wall-bound phenolic compounds in barley. However, cinnamic acid, whether applied alone or together with the elicitor, increased the amount of wall-bound phenolics in suspension-cultured barley cells. The cell walls in the elongating zone of submerged floating rice internodes show high susceptibility to expansins. When internode sections corresponding to such an elongation zone were incubated for 24 h under osmotic stress conditions produced by treatment with 100 mM polyethylene glycol 4000 (PEG), the cell wall susceptibility to expansins remained at its initial level, while the susceptibility of internode sections incubated under unstressed conditions decreased considerably during the same period. The contents of polysaccharides and phenolic acids as ferulic, diferulic and p-coumaric acids in the cell walls of internode sections increased substantially under unstressed conditions, but the increases were almost completely prevented by osmotic stress. Ferulic acid applied to internode sections under osmotic stress reduced the susceptibility of the cell walls to expansins and increased the levels of ferulic and diferulic acids in the cell walls, with little effect on the accumulation of polysaccharides. In contrast, applied p-coumaric acid increased the level of p-coumaric acid in the cell walls without a change in the levels of ferulic and diferulic acids but did not reduce the susceptibility to expansins. These results suggest that the deposition of ferulic and diferulic acids is a primary determinant in regulating the reduction of the susceptibility of cell walls to expansins in floating rice internodes.  相似文献

Effects of silicon on the mechanical and chemical properties of cell walls in the second leaf of oat (Avena sativa L.) seedlings were investigated. The cell wall extensibility in the basal region of the second leaf was considerably higher than that in the middle and subapical regions. Externally applied silicon increased the cell wall extensibility in the basal region, but it did not affect the extensibility in the middle and subapical regions. The amounts of cell wall polysaccharides and phenolic compounds, such as diferulic acid (DFA) and ferulic acid (FA), per unit length were lower in the basal region than in the middle and subapical regions of the leaf, and silicon altered these amounts in the basal region. In this region, silicon decreased the amounts of matrix polymers and cellulose per unit length and of DFA and FA, both per unit length and unit matrix polymer content. Silicon treatment also lowered the activity of phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) in the basal region. In contrast, the amount of silicon in cell walls increased in response to silicon treatment in three regions. These results suggest that in the basal region, silicon reduces the net wall mass and the formation of phenolic acid-mediated cross-linkages between wall polysaccharides. Such modifications of wall architecture may be responsible for the silicon-induced increase in the cell wall extensibility in oat leaves.  相似文献

扩展蛋白是植物细胞壁延伸过程中的关键调节因子,在植物的生长发育以及对逆境的响应过程中起着重要作用。本文选用小麦(HF 9703)胚芽鞘为材料,采用Hepes法和SDS法分别提取小麦胚芽鞘扩展蛋白,通过改良的植物组织伸长测定仪测定其活性,并利用扩展蛋白抗体进行免疫印迹以检测其丰度,主要研究了小麦胚芽鞘扩展蛋白的特性及对水分胁迫的响应。结果表明:Hepes法提取的扩展蛋白活性较高,而SDS法的提取效率高;离体小麦胚芽鞘扩展蛋白的活性具有pH依赖性,且随缓冲液的交替更换(pH 4.5:pH 6.8)而反复逆转;扩展蛋白主要定位于细胞壁中;小麦胚芽鞘扩展蛋白和黄瓜下胚轴扩展蛋白具有交叉重组活性,但这种活性具有种属特异性。水分胁迫诱导小麦胚芽鞘扩展蛋白的活性和丰度提高,扩展蛋白活性的提高在小麦对水分胁迫的抗性方面可能具有重要作用。  相似文献

Changes in soluble and cell wall bound peroxidases activities, phenylalanine ammonia-lyase activity and phenolic compounds and lignin contents in roots of calcium-treated soybean (Glycine max (L.) Merr.) seedlings and their relationships with root growth were investigated. Three-day-old soybean seedlings were cultivated in nutrient solution with or without 0.025–5.0 mM calcium for 24 h. In general, length and fresh and dry weights of roots increased, while activities of enzymes (soluble and cell-wall peroxidases and phenylalanine ammonia-lyase) and phenolic compounds and lignin contents decreased against calcium concentrations. In the absence of calcium, phenylalanine ammonia-lyase and peroxidases activities increased by accumulating phenolic compounds and lignin due to restricted growth of roots. Enhanced calcium supply reduced the production of phenolic compounds and lignification due to low phenylalanine ammonia-lyase and peroxidases activities, reinforcing the essential role of calcium to improve the soybean root growth.  相似文献

Phenylalanine ammonia-lyase from Sporobolomyces pararoseus was purified more than 450-fold. Polyacrylamide disc gel electrophoresis of this purified enzyme gave a single major protein band. Tyrosine ammonia-lyase activity was monitored during the purification of phenylalanine ammonia-lyase. Deaminating activities for phenylalanine and tyrosine were not separated during the purification process. The existence of one ammonia-lyase with bisubstrate activity is postulated.  相似文献

经渗透胁迫后 ,CO2 倍增条件下小麦叶片的SOD、POX和CAT的活性均显著高于对照 ,上升或稳定时期较长 ;在渗透胁迫后期MDA含量和电解质泄露率增加较慢 ,显著低于对照 ;H2 O2 含量一直高于对照但进行PEG胁迫后增长较慢。CO2 倍增条件下 ,小麦细胞出现DNA梯的时间较晚而且持续的时间较长 ,DNA梯出现时抗氧化酶和H2 O2 处于相对稳定状态。结果表明在渗透胁迫下CO2 倍增使小麦的抗氧化能力增强从而减轻了对细胞膜和DNA的损伤 ,并且干旱条件下小麦的细胞程序性死亡可能是由于细胞内氧化过强所致  相似文献

Tomato (Lycopersicon esculentum Mill.) cell suspension cultures accumulated wall-bound phenolic materials in response to inoculation with Verticillium albo-atrum Reinke et Berth. in a fashion analogous to that observed in whole plants. Both monomeric and polymeric materials were recovered. Deposition of phenolics into the cell walls of inoculated tomato cell cultures was inhibited by the phenylalanine ammonia-lyase (PAL) inhibitor, 2-amino-2-indanephosphate. Tomato PAL activity was induced over 12-fold by fungal inoculation, with a concomitant increase in the corresponding mRNA. The enzyme was purified >3400-fold, to apparent homogeneity, by anion-exchange chromatography, chromatofocusing, and gel filtration. The holoenzyme had a molecular mass of 280 to 320 kilodaltons, comprising 74-kilodalton subunits, and displayed an isoelectric point of 5.6 to 5.7. Induced PAL displayed apparent Michaelis-Menten kinetics (Km = 116 micromolar) and was not appreciably inhibited by its product cinnamic acid. Chromatographic analysis did not reveal multiple forms of the enzyme in either inoculated or uninoculated cultures.  相似文献

In order to discriminate between the ionic and osmotic components of salt stress, sugarcane (Saccharum officinarum L. cv. Co 86032) plants were treated with salt-NaCl or polyethylene glycol-PEG 8000 solutions (?0.7 MPa) for 15 days. Both the salt and PEG treatments significantly reduced leaf width, number of green leaves, and chlorophyll stability index. Osmotic adjustment (OA) indicated that both the stresses led to significant accumulation of osmolytes and sugars. Salt stressed plants appeared to use salt as an osmoticum while the PEG stressed plants showed an accumulation of sugars. Oxidative damage to membranes was not severe in plants subjected to salt or PEG stress. The salt stressed plants showed an increase in the activities of superoxide dismutase (SOD) and ascorbate peroxidase (APX), while PEG stress led to an increase in SOD but not APX activity as compared to the control. Thus, results indicate that the iso-osmotic salt or PEG stress led to differential responses in plants especially with respect to growth, OA, and antioxidant enzyme activities.  相似文献

用高压液相色谱法研究了豫麦18(抗旱性较强)和扬麦9号(抗旱性较弱)小麦胚芽鞘中三种不同形态的多胺(polyamine,PA):游离态多胺(PA)、高氯酸可溶性结合态多胺(ps结合态PA)和高氯酸不溶性结合态多胺(PIS结合态PA)与渗透胁迫的关系。结果发现:渗透胁迫2d,豫麦18胚芽鞘中的游离态Spd和游离态Spm的含量明显上升,而扬麦9号的游离态Put的上升明显。S-腺苷蛋氨酸脱羧酶(S—AMDC)的抑制剂——甲基乙二醛-双(鸟嘌呤腙)(MGBG)处理豫麦18,明显抑制了渗透胁迫诱导的游离态Spd和游离态Spm的增加,并且加重了渗透胁迫伤害,外源Spd处理扬麦9号明显促进了渗透胁迫诱导的游离态Spd和游离态Spm的增加,并且减缓了渗透胁迫的伤害。渗透胁迫下,豫麦18胚芽鞘中的PS结合态PA和PIS结合态PA的上升幅度都明显大于扬麦9号。菲咯啉(o—Phen)抑制渗透胁迫下PIS结合态PA的合成并加重了渗透胁迫对胚芽鞘的伤害。这些结果表明:小麦胚芽鞘中的游离态Spd、游离态Spm、PS结合态PA和PIS结合态PA的升高有利于增强渗透胁迫抗性。  相似文献

The influence of the allelochemicals ferulic (FA) and vanillic (VA) acids on peroxidase (POD, EC 1.11.1.7) and phenylalanine ammonia-lyase (PAL, EC 4.3.1.5) activities and their relationships with phenolic acid (PhAs) contents and root growth of soybean (Glycine max (L.) Merr.) were examined. Three-day-old seedlings were cultivated in nutrient solution containing FA or VA (0.1 to 1 mM) for 48 h. Both compounds (at 0.5 and 1 mM) decreased root length (RL), fresh weight (FW) and dry weight (DW) and increased PhAs contents. At 0.5 and 1 mM, FA increased soluble POD activity (18% and 47%, respectively) and cell wall (CW)-bound POD activity (61% and 34%), while VA increased soluble POD activity (33% and 17%) but did not affect CW-bound POD activity. At 1 mM, FA increased (82%) while VA reduced (32%) PAL activities. The results are discussed on the basis of the role of these compounds on phenylpropanoid metabolism and root growth and suggest that the effects caused on POD and PAL activities are some of the many mechanisms by which allelochemicals influence plant growth.  相似文献
