Oral challenge with a strain of Salmonella kedougou , resistant to nalidixic acid, gave a time-course for salmonella survival in the the alimentary tract of chicks pre-treated with a caecal culture from an eight-week-old bird to increase colonization resistance. In untreated, control chicks, salmonella colonization of the caeca resulted in counts of > 106/g within 48 h and a mean generation time of 1·6 h. With treated birds, however, the salmonellas failed to multiply in the caeca and decreased to a low level over a 48-h period, thus suggesting a mainly bacteriostatic effect. Pretreatment of chicks with the caecal culture also reduced the proportion of salmonella-positive crop samples. Growth of the salmonella in a feed-slurry system resembling moistened crop contents occurred rapidly at 37°C but was prevented by addition of the treatment culture, a decline in numbers coinciding with Lactobacillus populations of ca 109/g and a pH value of 5·5. Incorporation of a feed decontaminant, 1% formic acid (pH 4·0), rapidly eliminated both salmonellas and organisms added from the treatment culture.  相似文献

Day-of-hatch Leghorn chicks were treated by oral gavage with PREEMPT, a continuous-flow competitive exclusion culture containing broiler caecal bacteria, followed by an oral challenge with Listeria monocytogenes , to determine the effects of PREEMPT on L. monocytogenes caecal colonization. Increased ( P < 0·001) concentrations of caecal propionic acid in control chicks compared with PREEMPT-treated chicks at 3 days of age were indicative of the establishment of the PREEMPT bacteria. Caeca from control chicks at 7 days after the oral challenge with L. monocytogenes contained mean 3·4 ± 1·4 log cfu g−1 of caecal content, while caeca from PREEMPT-treated chicks contained no detectable Listeria. Enrichment for L. monocytogenes resulted in 100 % of caeca from control chicks testing culture-positive for L. monocytogenes , while none of the caeca from PREEMPT-treated chicks were culture-positive. The results indicated that prophylactic treatment of newly hatched chicks with PREEMPT significantly reduced caecal colonization by L. monocytogenes.  相似文献

Different serotypes of salmonellas were compared for selectivity and efficiency of recovery using 11 plating media. No optimal growth was obtained after 24h incubation in any of the media, but after 48h, brilliant green, brilliant green-phenol red-lactose-sucrose, bismuth sulphite, xylose-lysine-deoxycholate and Hektoen enteric agars showed optimal recovery of all the salmonella serotypes.
Xylose-lysine-deoxycholate and brilliant green-phenol red-lactose-sucrose agars were the most selective media for all salmonella serotypes. Addition of 10 μg/ml of sodium novobiocin to the tryptic soy-xylose-lysine and tryptic soy-brilliant green agars significantly improved their selectivity but reduced or inhibited the growth of some salmonella serotypes, including Salmonella typhi. Xylose-lysine-deoxycholate agar gave the highest recovery percentage of stressed salmonellas with a double-agar layer technique. Good recovery was also obtained on brilliant green-phenol red-lactose-sucrose, tryptic soy-brilliant green, tryptic soy-brilliant green-novobiocin, tryptic, soy-xylose-lysine and tryptic soy-xylose-lysine-novobiocin agars. Salmonella-shigella agar was the least efficient medium for the recovery of salmonellas under stress-induced or non-stressed conditions.  相似文献

The role of adherence and hydrophobic properties of native gut microflora in competitive exclusion of salmonellas from chicks is reported. Pure bacterial strains were isolated from washed caeca of 3-d-old chicks which had been treated on the day of hatch with a microflora from salmonella-free adult birds. These strains, when added to known mixtures of pure cultures, improved the efficacy of the mixtures in protecting chicks against a challenge of 105 cfu of Salmonella typhimurium. Strains from washed caeca and from other sources were also screened for hydrophobic properties. Undefined microflora and strains freshly isolated from washed caeca and which were hydrophobic improved the efficacy of protective mixtures, while none of the combination of hydrophobic strains obtained from stored strains had any beneficial effect.  相似文献

Studies have been carried out in which growth patterns of a Salmonella sp. and competing micro-organisms, especially other Enterobacteriaceae, were followed during pre-enrichment in buffered peptone water (BPw) and subsequent selective enrichment in tetrathionate broth (TBB). Pre-enrichment cultures were inoculated with minced meat and three reference samples containing nalidixic acid-resistant salmonellas. Irrespective of their initial numbers in BPw, Enterobacteriaceae increased to 10(8)/ml or more. During incubation in TBB at 43 degrees C, numbers of lactose-positive Enterobacteriaceae decreased in most enrichments which resulted in a positive salmonella isolation, but remained constant in the majority of those that did not. Levels of lactose-negative Enterobacteriaceae did not decrease in most salmonella-positive tests, but did so in half of the salmonella-negative ones. In the salmonella-positive tests the numbers of salmonellas had increased to 10(3)-10(7)/ml in BPw and after transfer to TBB slowly reached 10(4)/ml or more. In all cases the numbers of salmonellas exceeded those of the competing flora on brilliant green agar (BGA). In the salmonella-negative tests the numbers of salmonellas had increased less in BPw and decreased in most of the TBB enrichments. In none of these negative tests did the numbers of salmonellas exceed those of the competing flora on BGA. Escherichia coli dominated in most of the salmonella-negative tests. The results suggest more influence of lactose-positive than lactose-negative Enterobacteriaceae on the detection of salmonellas. The effect of competing microorganisms seems to depend not only upon their initial numbers, but also upon the types that can interact with salmonellas during selective enrichment.  相似文献

Three thousand and forty-two samples of post-mortem tissues, environmental swabs, animal feed ingredients and eggs were examined by conventional methods and by a simple rapid culture method. The rapid method gave reliable presumptive identification of salmonella contamination within 48 h of the start of culture. The rapid method also showed enhanced sensitivity for detection of salmonellas, in most types of sample, particularly those taken in poultry houses and hatcheries where high levels of competitive faecal bacteria are present.  相似文献

A 24 h screen which detects three viable salmonella cells per g of faeces was compared with classical isolation procedures for their ability to identify salmonella-positive samples from a pig rearing unit. The screen involved an overnight enrichment in Muller-Kauffmann tetrathionate (MK) broth, subculture for 4 h in M broth containing 10 μg ml−1 novobiocin, followed by detection of the presence of salmonellas by BacTrace and Salmonella-tek ELISAs. The classical protocols were: (1) an overnight and 48 h incubation in MK or selenite cysteine broth; or (2) overnight incubation in buffered peptone water and 24 h subculture in Rappaport-Vassiliadis broth (BPW-RV). Salmonellas were isolated from the broth cultures on xylose lysine deoxycholate and brilliant green agars. Thirty four of 100 samples were positive for salmonellas but no single isolation protocol identified all of them. The best of the classical isolation protocols, 48 h incubation in MK broth, identified 27 (79%) of the 34 positive samples whilst the screen identified 26 (76%) of the 34 positive samples. False-positive results were obtained from all isolation protocols except BPW-RV.  相似文献

A 24 h screen to detect viable salmonellas in faeces was developed by studying growth dynamics of salmonellas and competing flora in combinations of enrichment media and artificially-inoculated pig faeces. Muller-Kauffmann tetrathionate (MK) broth, incubated overnight at 42°C, maintained the lowest ratio of salmonella: competing flora and identified all inoculated samples. A 4 h postenrichment in M broth plus novobiocin reduced the number of false-positive results in subsequent ELISAs. Adjusting the negative cut-off values and incubation time of the chromogenic substrate from that recommended in the ELISA instructions reduced the rate of false-positive results further and allowed the detection of 103 salmonellas per ml in the presence of up to 107 ml−1 aerobic-competing cells. Suspension of faeces diluted 1 in 2 and 1 in 5, rather than 1 in 10 in MK broth did not necessitate further adjustments to the ELISA baseline values. The proposed screen protocol is an overnight incubation of faeces suspended 1 in 10 in MK broth, a 1 in 100 subculture into M broth plus 10 μg ml−1 novobiocin (MbN) for 4 h, steam inactivation of MbN cultures and testing by ELISA, and can detect three salmonella cells per g faeces.  相似文献

Carbohydrases predominate in the crop and their pH optima agree with pH prevailing in crop contents. Major amounts are also found in caecal contents. Aminopeptidase and trypsin are active mainly in the caeca, where they predominate in cells and contents, respectively. Aminopeptidase is partly membrane-bound. Except for trehalase, salivary glands display negligible amounts of digestive enzymes. The specific activity of digestive enzymes is high in all midgut cells and the enzyme molecules do not differ among gut compartments, as judged by polyacrylamide gel electrophoresis. Thus, it is probable that digestive enzymes are synthesized and secreted by all midgut cells (mainly in caeca) and then passed forward into the crop. Digestive enzymes are found in hindgut in concentrations similar to those in ventricular contents and, since they are stable in gut contents, they are likely excreted at a rate similar to undigested food. The data support the hypothesis that carbohydrate and protein are digested mainly in crop and caecal lumina, respectively, with part of the final digestion of proteins occurring at the surface of caecal cells. The peculiar features of the digestion of A. flavolineata grasshoppers, including the lack of midgut countercurrent fluxes, are thought to be derived from putative Polyneoptera ancestors.  相似文献

A method that uses a cation-exchange resin (Chelex 100) and differential centrifugation for the extraction and detection of salmonellas in soil was developed. The extraction efficiencies of a range of materials were examined and Chelex plus polyethylene glycol was identified as the best combination. Shake speeds, shake times and differential centrifugation speeds were selected to give an optimum salmonella recovery. The Chelex method accurately enumerated 1 cell per 10 g of nonsterile soil within 24 h. Addition of glycerol to soil samples enabled storage at — 70°C for 85 d without significant decreases in salmonella numbers. The Oxoid Salmonella Rapid Test (SRT) could be used to pre-screen large numbers of soil samples for the presence of salmonellas, prior to analysis by the Chelex method. The SRT method detected Salmonella typhimurium at levels as low as 2·5 cells per 10 g of nonsterile soil.  相似文献

Aims: Determining the effects of zinc bacitracin, bird age and access to range on bacterial microbiota in the ileum and caeca of broilers. Methods and Results: 16S rRNA gene-based polymerase chain reaction-based denaturing gradient gel electrophoresis (PCR–DGGE) profiling, DNA sequencing and real-time quantitative PCR techniques were used. The richness of both ileal and caecal microbiota increased with chicken age. The microbiota from those birds of the same age demonstrated relatively similar PCR–DGGE profiles and tended to form closely related clusters in the relatedness analyses. Dietary treatment with bacitracin (50 mg kg−1) and access to range did not change the richness but altered the composition of the microbiota. The impact of bacitracin was particularly obvious in 3-day-old chicks. Lactobacilli were abundant in the caecal microbiota of 3-day-old chicks regardless of the dietary treatment with bacitracin. The access to range enriched Bifidobacterium in both the ileum and caeca. Conclusions: Bacitracin, bird age and access to range all influenced bacterial microbiota in the ileum and caeca of broilers, with bird age having the greatest apparent effect. Significance and Impact of the Study: Providing useful information for the development of antibiotic replacement therapy for poultry production.  相似文献

An enzyme-linked immunosorbent assay (ELISA) and a microwell fluorescent antibody (FA) direct count method have been developed for the monitoring of salmonellas in soil. Both methods have a minimum detection level of ca 106 cells per gram of soil. The FA direct count method gave a linear recovery for the inoculum range 106–109 cells per gram of soil. When monitored by plate counts the survival of salmonellas was greater in a sterile than in a non-sterile soil. Evidence was found for the production of viable but non-culturable salmonellas in non-sterile soil; plate counts dropped rapidly with time, but FA direct counts and ELISA remained level. The salmonella cells became progressively smaller and rounder with time. Dead salmonella cells introduced into soil rapidly disappeared.  相似文献

'Competitive exclusion' treatment protected chicks against an environmental challenge with Salmonella enteritidis phage type 4, whether or not feed containing antimicrobial organic acids was also used. Treatment reduced the incidence of Salmonella -positive caecal samples from 82 to 8%, with a corresponding reduction in liver infection from 52 to 13%. Although acid-treated feed had no influence on an environmental challenge with salmonellas, its compatibility with 'competitive exclusion' suggests an advantage in their combined use to maximize protection of poultry against Salmonella infection.  相似文献

An experiment was performed over a period of 5 weeks with 576 one-day-old cockerels (Cobb 500), which were randomly divided into 6 experimental groups (12 replicate cages with 8 birds/cage). The objective of the experiment was to estimate whether different grain types (wheat, rye or triticale) and exogenous xylanase inclusion influence the performance and the gastrointestinal ecosystems of broiler chickens as regards intestinal viscosity, pH as well as the concentration of short-chain fatty acids (SCFA) and lactic acid (LA). The rye-based diets resulted in the poorest production results followed by wheat and triticale-based diets. Ileal viscosity decreased in the range: rye (140 cps), triticale (6.1 cps) and wheat (2.5 cps). Xylanase supplementation reduced ileal viscosity significantly only in birds fed with rye. The cereal type influenced the fermentation process in the broiler gastrointestinal tract more than the xylanase supplementation. The total concentration of organic acids was highest in the caeca, followed by the crop, ileum and gizzard. Lactic acid concentrations were highest in the content of the crop followed by ileum, gizzard and caeca. Propionate was only detected in caecal contents, whereas butyrate was also found in small amounts in the contents of the crop of birds from all treatments. The concentrations of acetate, butyrate and total organic acid concentration in caecal contents of chickens receiving the rye-based diets were lower than in birds fed the other cereals (P<0.001). Enzyme supplementation reduced the relative weight of the caeca in rye-fed birds (P=0.001).  相似文献

Detection of salmonellas in animal feeds by electrical conductance   总被引:10,自引:0,他引:10  
A comparison was made between standard culture methods and electrical conductance using a Malthus AT Microbiological Analyser for the examination of animal feeds for salmonella. Conductance testing with a selenite cystine/trimethylamine-N-oxide/dulcitol medium resulted in the detection of salmonellas in 49 of 55 known positive animal feeds, 13 of 19 spiked feed samples and 36 of 47 salmonella cultures. Testing with a lysine decarboxylase/glucose medium gave significantly better results (P less than 0.05) than with selenite cystine medium but five lysine decarboxylase negative strains of salmonella were undetected. When both media were used in parallel all salmonella positive samples were detected. No difference was found between preenrichment in buffered peptone water containing trimethylamine/mannitol and that containing lysine/glucose. Positive detection criteria for selenite medium of conductance peak at greater than or equal to 500 microsiemens (microS) with a rate of change of greater than or equal to 60 microS/h or 400-499 microS with a rate of change of greater than or equal to 40 microS/h and for lysine medium with a peak of greater than or equal to 100 microS have been established. The method offers savings in media and operating costs over conventional standard culture methods, provides results within 48 h and is recommended for statutory feed monitoring purposes.  相似文献

The lactoperoxidase system (LPS), a natural bactericidal system in milk, was investigated for its activity against salmonellas in vivo and in vitro. In acidified raw milk, in which the LPS was supplemented with an exogenous supply of H2O2, the numbers of salmonellas decreased rapidly. Different salmonella serotypes were affected to the same extent; rough strains, however, were more susceptible than smooth strains. When calves were fed on fresh milk, containing the LPS, and challenged with Salmonella typhimurium in doses of either 10(9) or 10(10), the clinical findings and salmonella excretion patterns were similar to those of control calves fed on heated milk. It was concluded that further studies, perhaps in the field, are necessary to evaluate LPS as a possible non-antibiotic system to control salmonellosis.  相似文献

The effects of two sources of dietary lipids and supplementation of Clostridium butyricum on performance and intestinal metabolism of broilers were investigated. In a 2 x 2 factorial arrangement, 168 one-day-old broiler chicks were divided into four treatment groups, and fed four diets with two lipid sources (soya bean oil or fish oil, at 25 g/kg and 30 g/kg in starter and grower diets respectively), and without or with supplementation of C. butyricum (1 x 10(9) colony forming units per kg diet). C. butyricum had no effect on broiler performance. At 20 and 40 d of age, the pH of caecal digesta and the relative length of caecum were decreased after supplementation of C. butyricum (p < 0.05). After this treatment, the activity of alkaline phosphatase was increased in jejunal mucosa at 40 d of age (p < 0.01). Furthermore, there were increases in populations of lactic acid bacteria and the concentrations of acetic acid, n-butyric acid, n-valeric acid and total short chain fatty acids in caecal digesta of birds fed C. butyricum (p < 0.05). A significant interaction between lipid source and C. butyricum was found in the pH of caecal digesta at 20 d of age (p < 0.01). The results of the present study indicated that dietary supplementation of C. butyricum maybe a benefit for gut health of broiler chickens.  相似文献

Lactic acid is the inhibitory agent in yoghurt responsible for the inhibition of Salmonella typhimurium. Casein, however, may exert a protective effect toward the survival of the salmonella in acid-milk products. Salmonella typhimurium was found to die-off 21.2% more rapidly in 18-h yoghurt-whey than in 18-h yoghurt at 37 degrees C with a pH of 3.85 and 1.42% lactic acid. When casein was added to yoghurt-whey, the die-off rate of the salmonellas was reduced to that found in yoghurt. The rate remained unchanged when 4.8% sodium caseinate was added to the whey. When 0 to 14% casein was added to the acid-whey the die-off rate changed from 9.7 to 24.0 min/log reduction of cells, respectively. There was a direct correlation between the increase in casein concentration and length of survival of the salmonellas. At a pH of 3.85, 4.2 or 4.5, the die-off rate was 6.5, 13.0 or 40 min/log reduction of cells in milk containing 1.42% lactic acid, and was 4.0, 10.0 or 33.3 min/log reduction, respectively, in whey with 1.42% lactic acid. Thus, the protective effect of casein toward Salm. typhimurium increased as the pH increased. This indicated that casein exerts a protective effect on Salm. typhimurium in acid dairy products and the degree of protection depends on the casein concentration, the form of the casein molecule and the pH.  相似文献
