Testosterone (T) restores the potency of castrated male rhesus monkeys, and our autoradiographic data have demonstrated that 3H-T or its metabolites concentrate in cell nuclei in the corticomedial amygdala, bed nucleus of stria terminalis, preoptic area, and hypothalamus. In rat, 3H-estradiol (3H-E2) is a major nuclear metabolite of 3H-T in areas of the limbic system, but comparable data are lacking for the primate. We have therefore developed an improved technique using high performance liquid chromatography for investigating metabolites of 3H-T that accumulate in cell nuclei in small amounts of tissue obtained from the brain of the rhesus monkey. Two castrated male rhesus monkeys were injected with 5 mCi of 3H-T and were killed 30 min later. In amygdala, preoptic area-bed nucleus of stria terminalis, and hypothalamus, 48–70% of the nuclear radioactivity was in the form of 3H-E2 (Type I tissues). In six other brain areas and in pituitary, 35–85% of the nuclear radioactivity was in the form of 3H-T (Type II tissues), whereas in genital tract tissues, 86–99% of the nuclear radioactivity was in the form of 3H-dihydrotestosterone (3H-DHT) (Type III tissues). In plasma and in supernatants from both Type I and Type II tissues, the proportions of 3H-T were high, and 3H-E2 did not exceed 10% of the total extractable radioactivity. These data suggest that, as in rodents, some of the central actions of T in primates may be mediated by estrogen target neurons.  相似文献

A study was made of the effect of a highly purified preparation of an unusual estrogen-binding protein (UEBP) of rat liver on 3H-estradiol (3H-E2) and 3H-testosterone (3H-T) metabolism by female rat liver homogenates. The UEBP decreased the rate of metabolic conversions of 3H-E2 and 3H-T in a dose-dependent and specific manner. The effectiveness of the inhibitory action of the UEBP declined with increase of metabolic activities of homogenate enzymes. The effects of the UEBP were reduced fully or partly by the ligands specifically binding to the UEBP, estrone (E1), and estriol (E3) respectively. The latter fact was used to demonstrate the effectiveness of the endogenous male rat liver homogenate UEBP in the control of 3H-E2 metabolism. The UEBP did not exhibit any oxidoreductase activity on the use of 3H-E1, 3H-E2, 3H-E2, 3H-T and 3H-androstenedione with NAD+, NADH, NADP+, and NADPH as cofactors. These data present the first direct experimental evidence of the regulatory function of the UEBP in the time course of changes in sex steroids.  相似文献

A method has been described by which floral buds of hermaphroditemelons isolated at various stages from archesporium to preleptotenein the pollen mother cells (PMC) can be grown through to pollen-grainformation in culture. Floral buds cultured for this period ina medium with thymidine-methyl-3H (3H-T) showed incorporationof label into the nuclei of tapetal cells and PMCs. The labelincorporated into the PMC was probably due to pre-meiotic DNAsyn-thesis. In other tissues of the floral buds, 3H-T incorporationproceeded through meiosis. Metabolic and environmental factorsinvolved in normal differentiation of the microsporogenous tissueof floral buds cultured in vitro are discussed.  相似文献

Chronic studies were initiated in rats to determine the effects of high- and low-dose tranylcypromine (TCP) on [3H]tryptamine (3H-T) binding sites. Male Sprague-Dawley rats were administered TCP (0.5 or 2.5 mg/kg/day) or vehicle (distilled water) for 4, 10 or 28 days via Alzet minipumps. After decapitation, the hippocampus and striatum were used to prepare membrane fragments for single point3H-T binding. Hippocampal3H-T binding was reduced after 10 and 28 days with the low dose and after 4, 10 and 28 days with the high dose. Striatal3H-T binding was reduced by both doses at all time intervals. The high dose resulted in a significantly greater reduction in striatal3H-T binding than did the low-dose after 4, 10, and 28 days. These results suggest that a more rapid reduction of3H-T binding in the hippocampus and/or a greater reduction of3H-T binding in the striatum by high-dose TCP than by low-dose TCP may be contributing factors in the reported efficacy of the former in refractory depression.  相似文献

Using electron microscope autoradiography, a study was made of the ultrastructure of early stages of muscle differentiation and 3H-thymidine (3H-T) labelled cells in the wall of the developing lymph heart of larvae of Rana temporaria L. The mononucleated postmitotic myoblasts with small bundles of thin and thick myofilaments deprived of Z-bodies were found in the lymph heart wall. No thin or intermediate-sized subsarcolemmal filaments were detected in the cytoplasm of these myoblasts. Myosatellites occurred under the basal lamina of muscle cells at stages 41-42. The primitive muscle-nerve junction was found at stages 44-45. Four hours after a single 3H-T administration only mononuclear cells without myofilaments were labelled. If the fixation was made 72 hours after a single 3H-T administration, the label was found, in addition, on the muscle cell nuclei. These data evidence that at the early stages of muscle differentiation in the developing lymph heart wall DNA synthesis and muscle specific protein synthesis are incompatible.  相似文献

An androgen receptor (AR) has been detected in both the hypothalamic and extra-hypothalamic area of the brain of the male of the green frog Rana esculenta. 3H-T binding activity was detected separately in the hypothalamic and extra-hypothalamic areas. Experiments of binding assay showed 3H-T binding activity in the nuclear extract but not in the cytosol of both the hypothalamic and the extra-hypothalamic areas. The androgen binding moiety was not strictly specific for androgens, binding also 17 beta-estradiol, although to a lesser extent. 3H-testosterone binding activity fluctuated in both the hypothalamic and the extra-hypothalamic areas throughout the reproductive cycle, and paralleled androgen plasma levels.  相似文献

作为连接城市绿地与居民需求的重要概念,绿地供需是城市绿地研究和规划决策的重要内容。识别城市绿地供给与需求的区别和关联,有助于城市绿地合理配置、人类福祉提升和城市的可持续发展。本文梳理2016—2020年城市绿地供需研究的主要议题和研究模式,探讨不同议题的差异与联系,并对未来城市绿地供需关系研究提出建议。综述发现,多数研究从供给或需求一方面开展研究,较少将城市供给与需求研究结合一起分析。供给方面主要关注绿地时空分布特征与生态系统服务供给,需求方面侧重于研究居民对绿地的主观偏好与行为。绿地供给、需求的研究对象、研究尺度与研究指标存在较大差异,是二者难以建立联系的主要因素。今后的研究应注重绿地供需匹配的情况。基于主观偏好/客观指标的绿地供需整合研究,是城市绿地供需关系研究的重要方向。  相似文献

Rat everted jejunal sacs were incubated for 15 and 30 min at 37 degrees C in oxygenated Krebs-Henseleit buffer, pH 7.4, containing 0.2 microM [3H]-thiamin (3H-T) or [3H]-thiamin monophosphate (3H-TMP) with and without 10 mM 1-phenylalanine (PAL) or 2.5 mM levamisole (LEV). The concentrations of 3H-T and its phosphoesters in sac wall and serosal fluid were determined by a radiometric method after electrophoretic separation. In separate experiments, thiamin pyrophosphokinase (TPKase) and thiamin pyrophosphatase (TPPase) activities were determined in mucosal scrapings, with and without PAL or LEV, by using a radiometric and a colorimetric method, respectively. 3H-TMP was transported partly unchanged by an active mechanism similarly to 3H-T, but less efficiently. During transport, 3H-TMP was also enzymatically transformed to thiamin (T) and thiamin pyrophosphate, which accumulated in the tissue. In the serosal fluid, the concentration of 3H-TMP exceeded that of 3H-T. Presence of PAL or LEV with 3H-T or 3H-TMP in the incubation medium reduced the serosal transport and the tissue content of T compounds. LEV caused a dose-dependent inhibition of TPKase without affecting TPPase, whereas PAL inhibited both activities to about the same extent. These results indicate that the transport of TMP involves a number of different processes similar to those responsible for T transport. The effects of PAL and LEV underline the importance of phosphorylation-dephosphorylation coupling.  相似文献

The composition of the immune system of 33 allophenic mice of four different types [C57BL/6 in equilibrium DBA/1, C57BL/6 in equilibrium (CBA X CBA/H-T6), C57BL/6 in equilibrium (A X SJL), DBA/1 in equilibrium (CBA X CBA/H-T6)] was studied. It was found that the parental composition of the peripheral white blood cells changed significantly during a two-month interval in 11/33 or 33% of the mice studied. This phenomenon has been termed chimeric drift. The animals were sacrificed between 9 and 16 months of age, and the parental composition of the peripheral white blood cells, spleen white blood cells, and thymocytes was determined on the day of sacrifice. It was foound that the peripheral white blood cell and spleen white blood cell compositions showed a high degree of correlation. However 8/33 or 24% of the mice studied showed discordance of the spleen and thymocyte cell populations. Seven of the 8 mice which showed spleen-thymocyte discordance, had also shown evidence of chimeric drift earlier in their lives. We suggest that this is evidence that chimeric drift may have an immunological basis.  相似文献

To demonstrate the existence of unstable or metabolic DNA in normal mammalian neurons and to study the effect of peripheral nerve injury on this metabolic DNA, adult mice were given repeated injections of high doses of 3H-thymidine (3H-T) on the day before injury to the left hypoglossal nerve. The animals were killed at intervals up to 33 days after the injections of 3H-T. Analyses of grain counts showed a low but significant elevation in the number of radioautographic grains per unit area of hypoglossal neuronal nuclei above background levels for up to 5 days after 3H-T injection. Digestion of the tissue with DNase lowered the nuclear grain counts to background levels, confirming that the DNA was indeed labelled. Although there was a loss of labelled material from the neuronal nuclei with time, there was no difference between injured and uninjured neurons at any of the intervals tested after injection of 3H-T.  相似文献

The establishment of prostatic budding patterns occurs early in prostate development but mechanisms responsible for this event are poorly understood. We investigated the role of WNT5A in patterning prostatic buds as they emerge from the fetal mouse urogenital sinus (UGS). Wnt5a mRNA was expressed in UGS mesenchyme during budding and was focally up-regulated as buds emerged from the anterior, dorsolateral, and ventral UGS regions. We observed abnormal UGS morphology and prostatic bud patterns in Wnt5a null male fetuses, demonstrated that prostatic bud number was decreased by recombinant mouse WNT5A protein during wild type UGS morphogenesis in vitro, and showed that ventral prostate development was selectively impaired when these WNT5A-treated UGSs were grafted under under kidney capsules of immunodeficient mice and grown for 28 d. Moreover, a WNT5A inhibitory antibody, added to UGS organ culture media, rescued prostatic budding from inhibition by a ventral prostatic bud inhibitor, 2,3,8,7-tetrachlorodibenzo-p-dioxin, and restored ventral prostate morphogenesis when these tissues were grafted under immunodeficient mouse kidney capsules and grown for 28 d. These results suggest that WNT5A participates in prostatic bud patterning by restricting mouse ventral prostate development.  相似文献

We have monitored estrous cycle and measured serum estradiol, GH, IGF-1, T4 and T3 levels in adult hypothyroid female rats which were divided into four groups: H group, hypothyroid rats without treatment; H-T4 group, hypothyroid rats injected daily with T4; HT4-PTU group, hypothyroid rats injected daily with T4 plus PTU (propylthiouracil), and H-T4-IOP group, hypothyroid rats injected daily with T4 plus IOP (iopanoic acid); Euthyroid rats (E group) were used as control. Our results indicate that the lack of sexual cycle in H animals was associated with lower values of estradiol, GH and IGF-1 in comparison to E group; the restoration of sexual cycle in H-T4 group was associated with values of estradiol, GH and IGF-1 higher than those of H group, whereas in H-T4-PTU and H-T4IOP groups the restoration was associated with higher values of GH and IGF-1 and values of estradiol similar to those of H group. These data could suggest a potential role of GH/IGF-1 axis, at least in part, in the lack of sexual cycle in H group and in the ovulation induction in H-T4, H-T4-PTU and H-T4-IOP groups.  相似文献

Exponentially growing rat islet cells (RINr) and hamster islet cells (HIT T-15) were incubated in presence of tolbutamide (10-1000 microM), gliclazide (0.1-10 microM) or glibenclamide (0.01-10 microM) for 15 hrs. Accumulation of insulin in culture medium was estimated by RIA. Effects of sulfonylureas (SU) on cell proliferation were assessed by 3H-thymidine (3H-T) incorporation into cellular DNA. All of SUs used stimulated insulin production in RIN and HIT cell cultures (with an exception of tolbutamide, which markedly suppressed insulin secretion in HIT cells at 1000 microM). 3H-T incorporation into RIN cells was elevated only in presence of gliclazide (10 microM), whereas tolbutamide at 1000 M significantly inhibited RIN cell proliferation. Gliclazide (0.1 microM) and glibenclamide (0.01-10 microM) enhanced 3H-T incorporation into HIT cells. Further detailed investigations of mechanisms of SU effects on islet cell reproduction will be of use for designing optimal strategy of hypoglycemizing therapy of diabetes mellitus.  相似文献

Iodobenzamides labelled with radioactive iodine are undergoing clinical evaluation as imaging and potential therapeutic agents in malignant melanomas. However, the uptake mechanism in melanic tissues remains controversial. Using secondary ion mass spectrometry (SIMS), we studied the microscopic distribution of N-(2 diethylaminoethyl)-4 iodobenzamide (I-BZA) in B16 murine melanoma inoculated to C57BL/6J1 Co mice as well as in normal pigmented skin. SIMS provides specific detection of iodine-127 atoms entering 127I-BZA composition. In B16 melanoma, 127I-BZA distribution was found to be heterogeneous, with focal areas of high concentration corresponding to cells rich in melanin pigments. In skin, SIMS analysis showed 127I-BZA distribution appearing as multiple small selective concentration areas within the epidermis. The number of these foci decreased from the stratum basale towards the stratum corneum. In both tissues, the intracellular location appeared specifically intracytoplasmic, with no apparent nuclear uptake. Distribution of this molecule mirrored that of melanin pigments. There was no enhancement of uptake at the membrane site. These results suggest that, in melanic tumors as well as in normal pigmented tissue, specific uptake of 127I-BZA occurs in pigment cells, with a possible link to melanin pigments.  相似文献

The dynamics of testosterone-7alpha-3H (T-3H) uptake by the developing genital system organs, as well as by the suprarenals, brain cortex and muscles was studied in vivo experiments with the 14-20 days old female rat embryos. Besides, the T3H uptake by the reproductive tract tissues and muscles just after the isolation of embryos from the uterus and after 2 days cultivation in the diffusion chambers implanted in the abdominal cavity of the adult castrated rats was studied in in vitro experiments. Moreover, the T3H uptake by the embryonic tissues was determined in the absence of ovaries. The specific uptake was noted in all the tissues at all developmental stages under all experimental conditions. The selective T3H uptake was noted during the periods corresponding to the rudiments' morphogenesis carachteristic for males. In the absence of ovaries, the T3H uptake by the reproductive tract tissues in vitro experiments decreased, but after 2 days cultivation in the diffusion chambers increased. The possible participation of ovaries in the female sex differentiation and the bisexual nature of rudiments determined by the presence of receptors both to estro- and androgens are discussed.  相似文献

Summary Cell dedifferentiation was induced inVicia faba root tissues by removing the whole root meristem (decapitation) and the behaviour of the nuclear DNA in the dedifferentiating cells was studied by means of cytophotometric and autoradiographic analyses. Cytophotometric determination after Feulgen-staining showed that: 1. the vast majority of nuclei in differentiated cells were in the DNA postsynthetic phase, but their Feulgen absorption was lower than that of DNA postsynthetic nuclei (G2, 4 C) in the meristem; 2. such a Feulgen absorption was detected in certain nuclei after root decapitation; 3. all the mitoses in the dedifferentiating tissues were diploid, fully matching the Feulgen absorption of mitoses in the meristem.After3H-thymidine (3H-T) feeding of the decapitated roots and autoradiography, the following results were obtained: 1. two populations of labeled nuclei, characterized by two different levels of scattered labeling occurred in dedifferentiating tissues, slightly labeled nuclei being much more numerous than heavily labeled nuclei; 2. the percentage of labeled nuclei was much greater than that of DNA presynthetic nuclei in the root tissues; 3. almost all the mitoses were labeled after a 16-hour3H-T feeding; 4. the percentage of slightly labeled nuclei paralleled that of dedifferentiating cells; 5. the duration of the DNA synthesis phase and that of the gap between completion of DNA synthesis and mitosis differed in heavily and slightly labeled nuclei; 6. all nuclei which entered DNA synthesis also entered mitosis.These results are interpreted to mean that: 1. after decapitation, two different DNA syntheses occur in the dedifferentiating root tissues ofV. faba: DNA reduplication in cells which dedifferentiate starting from a DNA presynthetic nuclear condition (heavily labeled nuclei) and extra DNA synthesis in cells which dedifferentiate starting from a DNA postsynthetic nuclear condition (slightly labeled nuclei); 2. extra DNA synthesis is required in these dedifferentiating cells for entry into mitosis.  相似文献

The Caco-2 cell model was used to study the efficiency of absorption and endogenous excretion of zinc (Zn) regulated by dietary Zn concentration. Cells were seeded onto high pore-density membranes and maintained in medium supplemented with 10% FBS. After confluence, cells were treated with 5 or 25 μmol Zn/L for 7 d, and Zn uptake and transport were measured in both apical (AP) and basolateral (BL) directions by using 65Zn. Similar cells were labeled with 65Zn and the release of Zn to the AP and BL sides was measured. The AP uptake of Zn in cells exposed to 25 μmol Zn/L was slower (p < 0.05) than that in cells exposed to 5 μmol Zn/L. The AP to BL transport rate in the 25 μmol Zn/L group was only 40% (p < 0.05) of that in the 5 μM group. In contrast, the rate of BL Zn uptake was 4-fold higher in cells treated with 25 μmol Zn/L than in those treated with 5 μmol Zn/L (p < 0.05). The BL to AP transport rate was 2-fold higher in cells treated with 25 μmol Zn/L than in those treated with 5 μmol Zn/L (p < 0.05). Basolateral uptake was 6 to 25 times greater (p < 0.05) than AP uptake for cells treated with 5 and 25 μmol Zn/L, respectively. The rate of Zn release was enhanced about 4-fold (p < 0.05) by 25 μmol Zn/L treatment. Release to the BL side was 10 times greater than to the AP side. Zn-induced metallothionein (MT), thought to down-regulate AP to BL Zn transport, was 4-fold higher (p < 0.001) in the 25 μmol Zn/L group than in the 5 μM group, but the rate of BL Zn release was higher in cells treated with 25 μmol Zn/L than in those treated with 5 μmol Zn/L (p < 0.05). Induced changes in transport rates by media Zn concentrations could involve the up- and/or down-regulation of Zn influx and efflux proteins such as the ZIP and ZnT families of Zn transporters.  相似文献

The mammalian urogenital sinus (UGS) develops in a sex specific manner, giving rise to the prostate in the male and the sinus vagina in the embryonic female. Androgens, produced by the embryonic testis, have been shown to be crucial to this process. In this study we show that retinoic acid signaling is required for the initial stages of bud development from the male UGS. Enzymes involved in retinoic acid synthesis are expressed in the UGS mesenchyme in a sex specific manner and addition of ligand to female tissue is able to induce prostate-like bud formation in the absence of androgens, albeit at reduced potency. Functional studies in mouse organ cultures that faithfully reproduce the initiation of prostate development indicate that one of the roles of retinoic acid signaling in the male is to inhibit the expression of Inhba, which encodes the βA subunit of Activin, in the UGS mesenchyme. Through in vivo genetic analysis and culture studies we show that inhibition of Activin signaling in the female UGS leads to a similar phenotype to that of retinoic acid treatment, namely bud formation in the absence of androgens. Our data also reveals that both androgens and retinoic acid have extra independent roles to that of repressing Activin signaling in the development of the prostate during fetal stages. This study identifies a novel role for retinoic acid as a mesenchymal factor that acts together with androgens to determine the position and initiation of bud development in the male UGS epithelia.  相似文献
