Cannulated brown trout, Salmo trutta , were exposed for 36 h to synthetic water with a low calcium content of pH 5 and similar synthetic water dosed with aluminium to raise the filterable A1 from 5 to 290 μg 1−1 over the 36-h period. There were no significant disturbances of plasma concentrations of glucose, cortisol or catecholamine (adrenaline and noradrenaline) in fish held in water of pH 5. The addition of aluminium to this acidic synthetic water resulted in a generalized endocrine stress response with a four-fold increase in plasma glucose concentration after 18 h and a significant increase in plasma cortisol concentration from 24 h onwards when filterable A1 exceeded 200 μg 1−1. Plasma catecholamine concentration indicated an adrenergic stress response in aluminium-exposed brown trout. A transient doubling in noradrenaline after 6 h in A1 was followed by a larger increase in both plasma adrenaline and noradrenaline concentrations in fish surviving the 36-h exposure to A1.  相似文献

The effect of serial removal of fish from aquaria, anaesthesia and stocking density on plasma cortisol, thyroid hormone, metabolite and ion levels was examined in rainbow trout, Salmo gairdneri , to determine the consequences of normal handling and maintenance procedures on the activity of the pituitary-adrenal and pituitary-thyroid axes in the species.
The capture and handling associated with serial removal of fish from an aquarium resulted in a rapid elevation of plasma cortisol concentrations and a slower rise in plasma glucose, Mg2+ and Na+ concentrations; plasma K.+ levels showed a transient fall shortly after commencing the sampling, followed by a significant increase.
A lethal concentration of 3-aminobenzoic acid ethyl ester (MS 222) of 125 mgl−1 (but not a sublethal concentration of 62.5 ng 1−1) prevented the capture stressor-associated changes in most of the measured parameters, including plasma cortisol levels, although plasma protein and ion concentrations were elevated in fish sampled after MS 222-anaesthesia.
Stocking densities from 1 to 64 fish 80 1−1 aquarium did not appear to effect changes in the measured parameters, except for plasma T3/T4 ratios which decreased with increasing stocking density. The exception was found in fish which were stocked in pairs: one member of each pair had a markedly elevated plasma cortisol level and there was a high incidence of mortality, probably related to aggressive social interaction.  相似文献

There were significant inverse correlations between rearing density of rainbow trout, Salmo gairdneri Richardson, and final body weight, plasma L-thyroxine (T4), trüodo-L-tryronine (T3), cortisol and protein concentrations, plasma T4/T3 ratios and thyroid epithelial cell height. In addition, hepatosomatic indices and plasma free fatty acid concentrations were higher in fish reared at low (134 g 1−1) density compared with groups reared at medium (210g1−1) or high density (299g 1−1), and the post-feeding (3.5-4h) elevation in plasma glucose and triglyceride levels evident in trout maintained at low rearing density was not found in those fish reared at higher densities. There were no significant effects of rearing density on hematocrit, carcass composition, hepatic glycogen and lipid levels and interregnal nucleus size.  相似文献

The present study investigates the effect of cannulation and chronic'black-box' confinement, as well as epinephrine administration (4–0 μg kg−1), on the degree and time-course of alterations in trout ( Oncorhynchus mykiss ) catecholamine and cortisol concentrations. Plasma cortisol concentrations in seawater trout acclimated to 3–6° C reached 104 ng ml−1 1 day after cannulation/confinement and remained elevated above resting levels (8 ng ml−1) until 6 days post-confinement. Although plasma epinephrine and norepinephrine generally declined over the period of confinement (day 1 approx. 12 nM; day 7 approx. 6 nM), norepinephrine titres were usually higher and more variable. Epinephrine injection caused elevations in plasma epinephrine levels but not in norepinephrine levels; epinephrine titres reaching 107 ± 26 nM (range 65–238 nM) at 2 min post-injection and returning to pre-injection levels by 30 min post-injection. Plasma cortisol increased by 20 ng ml−1 following epinephrine administration. Based on the time-course for post-confinement alterations in plasma cortisol, it appears that up to a week may be required before cannulated fish are completely acclimated to 'black-box' confinement. The findings suggest that meaningful results from experiments utilizing epinephrine injection and 'black-box confinement are contingent upon: (1) knowledge of circulating epinephrine levels shortly after injection (i.e. within 2 min post-injection); and (2) an experimental design that takes into account the elevated cortisol titres that are inherent with cannulation/confinement and epinephrine injection.  相似文献

Plasma cortisol levels were measured as an indicator of physiological stress in roach subjected to brief handling, or to a 14-day period of confinement, and in undisturbed control fish, during winter (water temperature 5° C) and summer (16° C), at which time plasma 17 β-oestradiol levels were also determined. Cortisol levels in undisturbed roach were low (mean 8·1 ng ml−1 at 5° C; 1·4 ng ml−1 at 16° C) and both handling and handling+confinement elevated blood cortisol levels significantly to 400 and 140 ng ml−1, respectively (at 5° C) and 700 and 600 ng ml−1, respectively (at 16) C). Blood cortisol levels had almost returned to baseline within 4 h following handling alone but in fish subjected to handling and prolonged confinement cortisol levels remained elevated for up to 168 h. Differences in baseline and poststress levels of cortisol, and in the rate of recovery from acute stress, were observed at the two different temperatures and the possible factors underlying these differences are discussed. Circulating levels of 17 β-oestradiol were reduced significantly within 24 h of exposure to either acute handling or chronic confinement indicating that the reproductive endocrine system in roach is sensitive to disruption by stressors.  相似文献

The survival of Atlantic salmon smolts on exposure to constant concentrations of ammonia has been measured under laboratory conditions. At concentrations of dissolved oxygen close to the air-saturation value, the 24-h LC50 of un-ionised ammonia is 0.15 mg NH31−1 in fresh water (hardness 264 mg 1−1 as CaCO3) and 0.3 mg NH31−1 in 30% sea water; at concentrations of dissolved oxygen of 3.5 mg 1−1 in fresh water and 3.1 mg 1−1 in 30% sea water, the 24-h LC50 is 0.09 mg NH3 1−1 and 0.12 mg NH3 1−1 respectively; for fish acclimated for 1 day to a concentration of ammonia close to the 24-h median for un-acclimated fish, the median is increased between 38 and 79%, depending on test conditions.  相似文献

Plasma levels of cortisone, a steroid hormone of potential physiological significance in fish, have rarely been measured. This study examines the interrelationship between circulating levels of cortisone and the major teleost corticosteroid, cortisol, in the blood of two strains of rainbow trout subject to confinement stress, a condition know to stimulate corticosteroidogenic activity. In unstressed fish from both strains, mean plasma cortisol levels were within the range 0.4–7.5 ng ml−1. Mean plasma cortisone levels were within the range 7.1–15.9 ng ml−1. Plasma cortisol levels were elevated within 5 min of the onset of strees and reached peak values within 45 min, although there was a marked difference betweed the maxima observed in the two strains (strain 1:70 ng ml−1; strain 2:150 ng ml−1). The rate of increase of plasma cortisone levels during strees was more rapid than that of cortisol, maximum values (strain 1:100ng ml−1; strain 2:160 ng ml−1) being reached within 10 to 20 min of the onset of stress. This rapid stress-induced elevation of plasme cortisone has not previously been reported in fish. We suggest that rapid conversion of cortisol to cortisone during the initial response to stress accounts for the appearance of large amounts of cortisone in the blood, indicating that circulating for the appearance of large amounts of cortisone in the blood, indicating that circulating levels of cortisol alone do not fully reflect the secretory activity of the interregnal during the initial of cortisol alone do not fully the secretory activity of the interregnal during the initial phase of the stress response. The results also indicate that the rate of clearance of cortisone from the circulation may be a major factor in determining stress-stimulated levels of plasma cortisol.  相似文献

Cortisol levels of black bream Acanthopagrus butcheri at capture did not change with time of day, gonadal stage or season and were 1·9±0·2 and 2·8±0·4 ng ml−1 for male and female fish, respectively. Confinement resulted in significantly elevated cortisol levels at all time periods; however, levels after 24 h of confinement were significantly lower than peak cortisol levels (15 min for males and 1 h for females). Confinement stress resulted in reduced levels of 17β-oestradiol (E2) and testosterone (T) within 1 h in sexually mature females. In mature males, suppression of T and 11-ketotestosterone (11KT) occurred after 30 min and 6 h of confinement, respectively. The relationship between confinement stress and levels of 17,20β-dihydroxy-4-pregnen-3-one (17,20β P) was more complex, with levels in males being elevated after 15 min and 24 h and suppressed after 6 h of confinement. In contrast, 17, 20β P levels in females were elevated after 1 h of confinement. In regressed females, plasma E2 and T concentrations were low at capture and were not affected by confinement stress whereas plasma 17, 20β P was elevated within 1 h. This study indicates that stress exerts a rapid inhibitory effect on gonadal steroidogenesis.  相似文献

Reproductively active female red gurnard Chelidonichthys kumu were captured on long-lines, and placed in confinement tanks for 24, 48, 72 and 96 h to examine the effect of capture and confinement on reproductive parameters (experiment I). Plasma cortisol at the time of capture was elevated to levels typical of stressed fish in other species (53–125 ng ml−1). Final plasma cortisol levels in red gurnard confined for any length of time were not significantly different from one another (ranging from 17 to 43 ng ml−1), indicating that fish were chronically stressed when held in captivity for up to 96 h after capture. When initial and final plasma cortisol levels were compared within confinement groups, cortisol decreased significantly after 24 and 96 h of confinement indicating that some acclimation to captivity may have occurred. In contrast, plasma 17β-estradiol (E2) and testosterone (T) levels decreased significantly to levels comparable to those in post-spawned fish, after any period of confinement, and remained low throughout the experiment. Another group of fish was captured and confined in the same manner as experiment I but subjected to repeated blood sampling every 24 h, until 96 h post-capture. In these fish, plasma cortisol levels decreased significantly from 127 ng ml−1 after 24 h confinement and thereafter showed no change (25–45 ng ml−1). Plasma E2 decreased significantly after 72 h of confinement while plasma T showed no change from levels at capture. Increased amounts of follicular atresia were found in vitellogenic oocytes of fish confined for longer periods of time in experiment I, indicating that capture and confinement stress affect reproduction negatively in captive wild fish.  相似文献

Yearling brown trout, Salmo trutta , were exposed to low mineral content water (nominal concentrations of 20μmol 1−1 magnesium, 7.7 μmol 1−1 potassium, 44 μmol 1−1 sodium) over a pH range of 4.0–5.2 with ambient calcium concentrations of 2.5–60 μmol 1−1. All fish died at pH 4.0 and 4.2 irrespective of ambient calcium concentration and also at pH 4.4 with only 2–3 μmol 1 −1 calcium (that is calcium-free water except for that leached from the diet or excreted by the fish). Good growth rates were obtained over the remaining treatments which extended down to pH 4.4 with as little as 7 μmol 1−1 calcium. When starved, weight loss was inversely correlated with pH. Effects on plasma chloride, percentage dry weight and calcium, potassium sodium, and phosphorus contents of skin, muscle and bone tissue were also investigated. These demonstrated pH effects on mineral metabolism in starved fish, but no effects were detected in fed fish.  相似文献

The Lahontan cutthroat trout lives under highly alkaline and saline conditions in Pyramid Lake, Nevada (pH 9.4; 0.2 mmol 1−1 Ca++; 7.3 mmol 1−1 Mg++). These experiments were conducted to study the possible roles of water Ca++ and Mg++ concentrations on ammonia excretion in the Lahontan cutthroat trout under highly alkaline conditions. The basic protocol of the experiments was to determine ammonia excretion rates during the following three exposure periods (each of 3-h duration) in sequence: (a) in normal lake water; (b) in soft lake water with the divalent cation concentrations reduced; and (c) in the soft lake water with either Ca++ or Mg++ (or no divalent cations added) added back at the appropriate lake water concentration. The soft-water exposure caused a significant reduction in ammonia excretion to about half of the control (original lake water) levels. When either Ca++ or Mg++ was added to the soft water in the third exposure period, the ammonia excretion rates were increased more than twofold back to lake water levels.  相似文献

The effects of water hardness (9 and 220 mgl−1 as CaCO3) upon zinc exchange in brown trout exposed to 0.77 μmol Zn 1−1 have been investigated using artificial soft water (<49.9 μmol Ca l-1, <40.1 μmol Mg 1−1) and mains hard water (1671.7 μmol Ca 1−1, 493.6 μmol Mg 1−1) of known composition. Both hard and soft water-adapted fish exhibited a bimodal pattern of net zinc influx. Net zinc influxes during both fast and slow uptake phases were significantly greater ( P <0.001) in soft (82.9 and 6.2 μmol Zn 100 g−1 h−1) than in hard water (46.3 and 2.4 μmol Zn 100 g h−1). Zinc efflux (- 0.2 μmol Zn 100 g−1 h−1) was enhanced only in hard water during the slow net influx phase.
Brown trout exposed to zinc in hard water and placed in metal-free media exhibited a greater net efflux (- 25.6 μmol Zn 100 g−1 h−1) of the metal than did fish in soft water (-4.2 μmol Zn 100 g−1 h−1) treated in the same manner. Tissue 65Zn activities reflected both the differences in uptake and excretion rates of the metal between hard and soft water fish. During zinc exposure (0.77 μmol Zn 1−1) high water hardness reduced tissue burdens of the metal by reducing net branchial influx, and enhancing efflux of the metal in hard water fish.  相似文献

In an artificial stream channel, wild 1 year old brown trout Salmo trutta were exposed to fluctuations in flow and water level to simulate hydro-peaking conditions downstream of a hydropower installation. Blood plasma cortisol concentrations reached a maximum of 59·4 ± 35·3 ng ml−1 (mean ± 95% CL) 2h after the end of down-ramping. Return to the pre-exposure cortisol level was achieved within 6 h. When subjected to daily cyclical fluctuations over 7 days, plasma cortisol levels were significantly elevated (61·3 ± 26·8 ng ml−1) on the first day compared to undisturbed fish (4·9 ± 3·7 ng ml−1). On the fourth and seventh day, no elevation in plasma cortisol above control levels was observed. No changes in blood glucose that could be attributed to the stressor were found. There was no correlation between plasma cortisol and blood glucose levels. The short-lived cortisol response to daily fluctuations indicates a rapid habituation to this Stressor.  相似文献

Juvenile bull trout Salvelinus confluentus exposed to continuous- or pulsed-DC electroshock exhibited rapid elevations in plasma cortisol and glucose, but plasma chloride did not change. In a 1-h experiment using 240 V at 1·4 A of 60-Hz pulsed DC (voltage gradient 0·81 V cm−1), which proved lethal, plasma cortisol and glucose rose significantly within 15 min of a 10-s electroshock. Plasma cortisol reached a peak level of 156 ± 18 ng ml−1 at 45 min and then decreased, whereas plasma glucose reached its highest level of 179 ± 7·5mg dl−1 at 1 h. In a 24-h experiment using lower dosages, plasma cortisol increased from 6·1-16 ng ml−1 to peak levels of 155–161 ng ml−1 in 1 h in response to a 10-s electroshock of continuous (130 V, 0·5 A, 1·45 V cm−1) or pulsed (120 V, 0·5 A, 60 Hz, 0·55 V cm−1) DC. Although plasma concentrations declined thereafter, levels remained above control values at 24 h. Plasma glucose was elevated from 60–65 to 120–134 mg dl−1 after 1h by both electroshock treatments and remained near or above those levels for the 24-h duration. Plasma cortisol and glucose levels were much higher in electroshocked bull trout at 1 h compared with those in fish 1 h after receiving a 30-s handling stressor (cortisol, 90 ± 12 ng ml−1; glucose, 82 ± 6·1 mg dl−1). The results indicate that both continuous and pulsed DC were more stressful to juvenile bull trout than handling and that recovery, at least for pulsed DC, may take longer than 24 h.  相似文献

Seedlings of two tree species from the Atlantic lowlands of Costa Rica, Ochroma la-gopus Swartz, a fast-growing pioneer species, and Pentaclethra macroloba (Willd.) Kuntze, a slower-growing climax species, were grown under enriched atmospheric CO2 in controlled environment chambers. Carbon dioxide concentrations were maintained at 350 and 675 μl 1−1 under photosynthetic photon flux densities of 500 μol m−2 s−1 and temperatures of 26°C day and 20°C night. Total biomass of both species increased significantly in the elevated CO2 treatment; the increase in biomass was greatest for the pioneer species, O. lagopus . Both species had greater leaf areas and specific leaf weights with increased atmospheric CO2. However, the ratio of non-pho-tosynthetic tissue to leaf area also increased in both species leading to decreased leaf area ratios. Plants of both species grown at 675 μl 1−1 CO2 had lower chlorophyll contents and photosynthesis on a leaf area basis than those grown at 350 μl 1−1. Reductions in net photosynthesis occurred despite increased internal CO2 concentrations in the CO2-enriched treatment. Stomatal conductances of both species decreased with CO2-enrichment resulting in significant increases in water use efficiency.  相似文献

The objective of the current study was to examine whether sublethal (moderate) levels of dissolved ammonia may be beneficial to growth in juvenile walleye Sander vitreus (recent evidence in juvenile rainbow trout Oncorhynchus mykiss has shown significant increases in protein synthesis in the presence of moderately elevated concentrations of dissolved ammonia). Moderately elevated dissolved ammonia concentrations between 100 and 300 μmol l−1 suppressed routine aerobic metabolic activity by 20% during acute trials (2 h), while promoting specific growth rate (>50%) and elevating whole body soluble protein content by 20% in the early stages (14–42 days) in chronic ammonia exposure experiments. Juvenile S. vitreus held at ammonia concentrations between 107·6 ± 5·5 and 225·5 ± 4·7 μmol l−1 (mean ± s . e .) grew significantly faster than control fish and significantly reduced plasma cortisol levels (<3 μg dl−1). Results from this study suggest that chronic exposure to moderate amounts of dissolved ammonia significantly increase growth rates in juvenile S. vitreus by increasing nitrogen accessible for supplementary protein deposition leading to somatic development.  相似文献

A toxicity test with cadmium concentrations ranging from 0·1 to 100·0 mg Cd 1−1 was used to assess the effect of cadmium pretreatment on rainbow trout ( Salmo gairdneri Richardson) alevins. The median period of survival for fish pretreated at 0·01 mg Cd 1−1 was found to be increased at test concentrations up to 10mg Cd 1−1 compared with alevins pretreated with dilution water. However, at concentrations above 10mg Cd 1−1 pretreatment at 0·01 mg Cd 1−1 reduced the median period of survival.  相似文献

This study was designed to investigate whether the blood of Pagothenia borchgrevinki , exhibits a Haldane effect, and whether activation of a Na+/H+ antiporter increases transport of intracellular protons and Bohr protons out of the erythrocytes resulting in inhibition of CO2 excretion in both P. borchgrevinki , and Dissostichus mawsoni. When carbon dioxide dissociation curves were determined from blood samples pooled from three fish under oxygenated and deoxygenated conditions a Haldane effect was observed. Using an in vitro , CO2 excretion assay, the rate of HCO3 dehydration was determined on blood and plasma equilibrated under an N2atmosphere then rapidly oxygenated with air in the presence of 10−5 M noradrenaline or acetazolamide (1004M). Whole blood and plasma from P. borchgrevinki , and D. mawsoni , were equilibrated with 0·5% CO2 in air and assayed in the presence of 10−5 M noradrenaline. Erythrocyte CO2 excretion rates were depressed significantly by noradrenaline in both species. The whole blood HCO3 dehydration rate was depressed significantly following rapid oxygenation in the presence of acetazolamide indicating that the pathway of CO2 excretion included activation of intracellular carbonic anhydrase and an adrenergic receptor.  相似文献

Confinement of juvenile Atlantic salmon Salmo salar from four different populations (all–female diploids, all–female triploids, mixed sex diploids and mixed sex triploids) either before (FW parr) or after (SW smolts) transfer to sea elevated plasma cortisol and plasma lactate from control levels irrespective of ploidy status. Both before and after confinement, plasma cortisol levels in SW smolts (6–174 ng ml–1) were higher than those in FW parr (4–58 ng ml–1), which possibly reflected the physiological challenge of acclimation to SW. Mixed sex populations of SW smolts had higher cortisol levels than all–female populations. The duration of confinement (1, 3 or 6 h) affected the magnitude of the plasma cortisol and lactate responses in SW smolts. Plasma cortisol levels in diploid and triploid SW smolts subjected to 2 h of confinement decreased to pre–stress levels within 6 h post–confinement. Plasma lactate levels were not significantly different from pre–stress levels 48 h after confinement. As no difference exists in primary and secondary stress responses of Atlantic salmon of differing ploidy status, it is unlikely that differences in mortality rates between diploid and triploid populations under commercial conditions can be attributed to differences in their physiological responses to periods of stress lasting up to 6 h.  相似文献

Highbush blueberry plants ( Vaccinium corymbosum L. cv. Bluecrop) growing in containers were flooded in the laboratory for various durations to determine the effect of flooding on carbon assimilation, photosynthetic response to varying CO2 and O2 concentrations and apparent quantum yield as measured in an open flow gas analysis system. Hydraulic conductivity of the root was also measured using a pressure chamber. Root conductivity was lower and the effect of increasing CO2 levels on carbon assimilation less for flooded than unflooded plants after short-(i-2 days), intermediate-(10–14 days) and long-term (35–40 days) flooding. A reduction in O2 levels surrounding the leaves from 21 to 2% for unflooded plants increased carbon assimilation by 33% and carboxylation efficiency from 0.012 to 0.021 mol CO2 fixed (mol CO2)−1. Carboxylation efficiency of flooded plants, however, was unaffected by a decrease in percentage O2, averaging 0.005 mol CO2 fixed (mol CO2)−1. Apparent quantum yield decreased from 2.2 × 10−1 mol of CO2 fixed (mol light)−1 for unflooded plants to 2.0 × 10−3 and 9.0 × 10−4 for intermediate- and long-term flooding durations, respectively. Shortterm flooding reduced carbon assimilation via a decrease in stomatal conductance, while longer flooding durations also decreased the carboxylation efficiency of the leaf.  相似文献
