Background

Schistosoma japonicum causes major public health problems in China and the Philippines; this parasite, which is transmitted by freshwater snails of the species Oncomelania hupensis, causes the disease intestinal schistosomiasis in humans and cattle. Researchers working on Schistosoma in Africa have described the relationship between the parasites and their snail intermediate hosts as coevolved or even as an evolutionary arms race. In the present study this hypothesis of coevolution is evaluated for S. japonicum and O. hupensis. The origins and radiation of the snails and the parasite across China, and the taxonomic validity of the sub-species of O. hupensis, are also assessed.

Methodology/Principal Findings

The findings provide no evidence for coevolution between S. japonicum and O. hupensis, and the phylogeographical analysis suggests a heterochronous radiation of the parasites and snails in response to different palaeogeographical and climatic triggers. The results are consistent with a hypothesis of East to West colonisation of China by Oncomelania with a re-invasion of Japan by O. hupensis from China. The Taiwan population of S. japonicum appears to be recently established in comparison with mainland Chinese populations.

Conclusions/Significance

The snail and parasite populations of the western mountain region of China (Yunnan and Sichuan) appear to have been isolated from Southeast Asian populations since the Pleistocene; this has implications for road and rail links being constructed in the region, which will breach biogeographical barriers between China and Southeast Asia. The results also have implications for the spread of S. japonicum. In the absence of coevolution, the parasite may more readily colonise new snail populations to which it is not locally adapted, or even new intermediate host species; this can facilitate its dispersal into new areas. Additional work is required to assess further the risk of spread of S. japonicum.  相似文献

A mechanistic model of competition on a single resource wastested experimentally with two freshwater rotifers, Brachionusrubens and B.calyciflorus, both grown on the alga Chlorellapyrenoidosa. Using open culture systems for each species wemeasured: (i) the resource-saturated exponential growth rate,µmax, and (ii) the relationship between specific growthrate, µ, at steady-state and the residual algal concentrationover a range of system turnover rates, or dilution rates, D.The µmax of B.calyciflorus was {small tilde}60% higherthan B. rubens. These results were then used to construct agraphical model for predicting the victor in interspecific competitionbetween the two rotifers. Since the two resource-dependent growthrates crossed, one species, B.calyciflorus, was predicted tobe the victor at a high D while B. rubens was predicted as thevictor at low D. Finally, the outcome of competition was determinedfor two turnover rates. As predicted by the graphical competitionmodels, B.calyciflorus was the dominant species at rapid D (0.029h–1) and B.rubens was dominant at slow D (0.0044 h–1).These studies support recent conclusions that mechanistic competitionmodels may be applied to predict dominant species from a prioriinformation on growth potential and resource levels, which isnot possible with traditional Lotka-Volterra models.  相似文献

Species-specific differences in the assimilation of atmosphericCO2 depends upon differences in the capacities for the biochemicalreactions that regulate the gas-exchange process. Quantifyingthese differences for more than a few species, however, hasproven difficult. Therefore, to understand better how speciesdiffer in their capacity for CO2 assimilation, a widely usedmodel, capable of partitioning limitations to the activity ofribulose-1,5-bisphosphate carboxylase-oxygenase, to the rateof ribulose 1,5-bisphosphate regeneration via electron transport,and to the rate of triose phosphate utilization was used toanalyse 164 previously published A/Ci, curves for 109 C3 plantspecies. Based on this analysis, the maximum rate of carboxylation,Vcmax, ranged from 6µmol m–2 s–1 for the coniferousspecies Picea abies to 194µmol m–2 s–1 forthe agricultural species Beta vulgaris, and averaged 64µmolm–2 s–1 across all species. The maximum rate ofelectron transport, Jmax, ranged from 17µmol m–2s–1 again for Picea abies to 372µmol m–2 s–1for the desert annual Malvastrum rotundifolium, and averaged134µmol m–2 s–1 across all species. A strongpositive correlation between Vcmax and Jmax indicated that theassimilation of CO2 was regulated in a co-ordinated manner bythese two component processes. Of the A/Ci curves analysed,23 showed either an insensitivity or reversed-sensitivity toincreasing CO2 concentration, indicating that CO2 assimilationwas limited by the utilization of triose phosphates. The rateof triose phosphate utilization ranged from 4·9 µmolm–2 s–1 for the tropical perennial Tabebuia roseato 20·1 µmol m–2 s–1 for the weedyannual Xanthium strumarium, and averaged 10·1 µmolm–2 s–1 across all species. Despite what at first glance would appear to be a wide rangeof estimates for the biochemical capacities that regulate CO2assimilation, separating these species-specific results intothose of broad plant categories revealed that Vcmax and Jmaxwere in general higher for herbaceous annuals than they werefor woody perennials. For annuals, Vcmax and Jmax averaged 75and 154 µmol m–2 s–1, while for perennialsthese same two parameters averaged only 44 and 97 µmolm2 s–1, respectively. Although these differencesbetween groups may be coincidental, such an observation pointsto differences between annuals and perennials in either theavailability or allocation of resources to the gas-exchangeprocess. Feeding on natural plankton populations and respiration of thesmall cyclopoid copepod Oithona similis were measured duringthe warm season in Buzzards Bay, Massachusetts, USA. AlthoughO.similis did not significantly ingest small autotrophic andheterotrophic flagellates (2–8 µn), this copepodactively fed on >10 µm particles, including autotrophic/heterotrophic(dino)flagel-lates and ciliates, with clearance rates of 0.03–0.38ml animal–1 h–1. The clearance rates increased withthe prey size. O.similis also fed on copepod nauplii (mainlycomposed of the N1 stage of Acartia tonsa with a clearance rateof 0.16 ml animal–1 h–1. Daily carbon ration fromthe combination of these food items averaged 148 ng C animal–1day–1 (41% of body C day–1), with ciliates and heterotrophicdino-flagellates being the main food source ({small tilde}69%of total carbon ration). Respiration rates were 020–0.23µl O2 animal–1 day–1. Assuming a respiratoryquotient of 0.8 and digestion efficiency of 0.7, the carbonrequirement for respiration was calculated to be 125–143ng C animal–1 day–1, close to the daily carbon rationestimated above. We conclude that predation on ciliates andheterotrophic dinoflagellates was important for O.similis tosustain its population in our study area during the warm season.  相似文献

The photoregulation of carotenogenesis in Rhodotorula minutawas found to consist of tow phases, a temperature-independentphotochemical reaction (light process) and temperature-dependentbut light-independent biochemical reactions (dark process).These processes were separately examined by regulating the temperatureand were characterized as follows: 1) The quantity of carotenoid produced [C (µg g–1)]and the rate of carotenoid production [Vc (µg g–1hr–1)] in the dark process were regulated by the lightdose [D (erg cm–2)] to which cells were exposed in thelight process. These relationships were expressed by the equations:C=9.1 log D–62.0 and Vc=0.81 log D–5.60. This photoresponsefollowed the Roscoe-Bunsen reciprocity law. 2) The induced state toward carotenogenesis, once acquired inthe light process, was very stable, suggesting that the proposedphotochemical product is stable as an inducer of carotenogenesisand decreases only in conjunction with carotenoid biosynthesis. 3) The photochemical reaction was oxygen-independent, but subsequentdark reactions were completely dependent on oxygen. 4) Postulated compounds related to the photochemical reactionwere not metabolized in vivo. Hydrococcus brazieri and Arthritica semen on a sandflat at Coodanup,Peel Inlet, Western Australia, accounted for 89.5% of totalmollusc numbers. The mean density of H. brazieri was 9487 m–2and A. semen averaged 8105 m–2. H. brazieri grew at 0.5mm month–1 and reached maturity in 4 months; A. semengrew at 0.3 mm month–1 and reached maturity in 6 months.Somatic production in the Peel-Harvey estuarine system was estimatedto be 0.5 g m–2yr–1for H. brazieri and 4.1 g m–2yr–1for A. semen. Protoplasts were successfully isolated from internodal callustissues of both Oxalis glaucifolia and O. rhombeo-ovata whenthey were digested in a solution containing 0.1% (w/v) MacerozymeR-10, 0.5% (w/v) cellulase Onozuka R-10 and 0.3 mmol m–3sucrose. Protoplasts proliferated to give cell colonies on Gamborget al.'s B5 medium supplemented with 0.3 mmol m–3 mannitol,0.5 mg dm–32, 4-D, and 2.0 mg dm–3 kinetin. Calluswas produced upon transfer of cell colonies to Murashige andSkoog medium containing 2.0 mg dm–3 l-naphthaleneaceticacid (NAA) and 0.1 mg dm–3 kinetin for O. glaucifolia,or with 5.0 mg dm–3 NAA and 0.5 mg dm–3 6-benzylaminopurine,for O. rhombeo-ovata. Plants were regenerated from O. glaucifoliaprotoplasts on a medium containing 0.1 mg dm–3 NAA, 1.0mg dm–3 kinetin and 1.0 mg dm–3 gibberellic acid,but only vascular nodules were differentiated by O. rhombeo-ovataprotoplast-derived calli. 以我国安徽及菲律宾莱特岛的日本血吸虫、湄公血吸虫、马来西亚类似日本血吸虫以及中华血吸虫交互感染其螺蛳宿主。结果表明日本血吸虫及钉螺与湄公血吸虫、马来西亚类似日本血吸虫及其螺蛳宿主亲缘关系较疏远。而湄公血吸虫与马来西亚类似日本血吸虫及其螺蛳宿主有较近的亲缘关系。交互感染结果为马来西亚类似日本血吸虫及钙河罗氏螺的分类问题提供了新的依据。  相似文献

Post-embryonic durations of Tropodiaptomus spectabilis (Kiefer)and Metadiaptomus colonialis(van Douwe) were determined at 20°Cin laboratory factorial experiments involving four algal foodenrichment levels (0, 100, 500 and 2500 µg l–1 Cof Selenastrum added to 20 µm filtered water from respectivesource-lakes) and three suspended sediment levels (filtered,natural, and 2- to 3-fold sediment-enriched lake water). Foodeffects (30, 75, 225 and 600 (µg –1 C of Scenedesmus)were tested alone at 20°C for Metadiaptomus meridianus (vanDouwe). Total naupliar (Dn) and total copepodid (Dc) developmenttimes [summed to give total post-embryonic duration (Dt)] andmetasome lengths at maturity were measured In all taxa, foodsupply maximally affected Dc values 2- to 3-fold, whereas itsmaximal influence on Dn values was relatively slight (generally25%). The measured effect of food supply on Dt, was as strongas the predicted influence of temperature over an appropriateannual range. Food supply influenced size at maturity, and probablythereby fecundity, thus exerting additional demographic influences.Sediment effects were inconsistent, and quantitatively weakerthan food effects Total development of T.spectabilis was 20%raster, and that of M.colonialis 15% slower in sediment-enrichedthan in natural sediment level treatments; contrasting baselinesediment levels (2–3 times higher for the latter species)and different enrichment procedures confound interpretation.Unexpectedly, and inexplicably, development almost invariablyfailed in sediment-free water, implying an apparent dependencyon inorganic particles in these taxa This contrasts with thegenerally adverse influences of high sediment concentrationsupon zooplankton. Minimal male and female Dt values at 20°Cwere comparable and significantly longer in M.colonialis (15.5and 17 7 days) and M meridianus (16.5 and 21.5 days) than inT.spectabilis (11.7 and 12 2 days). These differences in durationare ecologically incongruous in relation to expected rK life history strategies of genera characteristic of temporaryor semipermanent waters and permanent waters respectively.  相似文献

For the elucidation of the isotope effect on cell functionsof deuterium (D) incorporated into cell constituents, alterationsin the heat response of D-exchanged Chlorella ellipsoidea (D-Chlorella)were investigated. D-Chlorella cells obtained by culture inmedium that contained 60 mol% D2O were assayed for their responseto heat in H2O medium to rule out the solvent isotope effectof D2O. Upon heating at 41–45?C, the heat sensitivityof D-Chlorella was greater than that of ordinary (H-Chlorella)cells; at 43?C, the heat sensitivity of D-Chlorella was 1.5–1.6times higher than that of H-Chlorella. For the induction ofresistance to heating, preheating of the cells at a lower temperaturethan that used for heat treatment was effective in the caseof both D- and H-Chlorella. However, the optimum temperaturefor preheating of D-Chlorella (34?C) was lower than for H-Chlorella(36–37?C). With preheating at 34?C, heat-shock proteins(HSPs), in particular proteins of 62 and 79 kDa, were inducedsimilarly in both types of cell. However, the gel-electrophoreticpatterns of HSPs induced at 37?C were differed somewhat betweenD- and H-Chlorella. These results suggest that the responseof cells to heat, in particular the induction of resistanceto heating and the synthesis of HSPs, was altered by deuterationof cell constituents. The habitat, diet, some reproductive and mortality factors forseveral shallow-water species of the boreal turrid genus Oenopotafrom the Puget Sound region were determined. All fed exclusivelyon tubicolous polychaetes. Oenopota fidicula was a dietary andhabitat generalise Oenopota elegans eats Schistocomus hiltoniand lives in areas of shell fragments. Oenopota excunata eatsTharyx multifilis and is a habitat generalist. Oenopota tabulataappears to be a dietary generalist, but lives only on or nearrocks. The widely-distributed species, O. levidensis, eats spionidpolychaetes, primarily Polydora species, or Owenia fusiformis,and is a habitat generalist. Feeding frequencies are low asare population densities; although O. levidensis sometimes exceeds15 animals m–2. Crabs are major predators upon turridsin some areas, but mortality causes are generally obscure. Spawningwas observed in four species, but only O. elegans and O. levidensishad viable larvae. Development takes 13–15 weeks, 6–7weeks in a capsule and 6–7 weeks as planktonic veligers.There are no nurse eggs. Susceptibility of Oncomelania hupensis formosana recombinants and hybrids with Oncomelania hupensis nosophora to infection with Schistosoma japonicum. International Journal for Parasitology11: 391–397. Three generations of Oncomelania hupensis formosana recombinants were produced by mating the Kaohsiung race with the Ilan and Changhua races of the snails. F2 and F3 recombinants were produced by back-crossing F1 and F2 with Kaohsiung O. h. formosana. Subsequently, susceptibility of recombinants to infection with the original strain of Schistosoma japonicum, Ilan or Changhua strain, was investigated. Results indicated that susceptibility of recombinants declined steadily generation by generation. Marked decline of infectivity was observed for Kaohsiung-Ilan recombinants as compared with Kaohsiung-Changhua recombinants. For example, the overall infection rate of Kaohsiung-Ilan F1 recombinants was 7.3 % with a 51.4 % of control snails. The same figures for F2 were 4.2 and 52.6%, and 1 and 40.3 % for F3. On the other hand, the overall infection rate of Kaohsiung-Changhua F1 recombinants was 21.9% with a 46.9% of control snails; and 11.9 and 50.3 % for F2; and 7.6 and 33.2% for F3. The F3 hybrids of Oncomelania hupensis nosophora from Japan and O. h. formosana from Kaohsiung and Ilan were also produced, and susceptibility with the Japanese strain of S. japonicum was studied. A highly significant decline of susceptibility was observed among hybrids (4.4%) in contrast with control snails (85.6 %).Feasibility of applying O. h. formosana in biological control of S. japonicum was discussed. One must determine in the laboratory, prior to application, which race of O. h. formosana should be used depending on the Oncomelania snails of an endemic area. For S. japonicum prevalent in Yamanashi, Japan, the Ilan race of O. h. formosana was found to be better choice than the Kaohsiung race of the snails.  相似文献

Three cultivars of M. sativa and one cultivar of O. viciifoliawere evaluated for their response to inoculation with A. rhizogenesstrain A4T (containing pRiA4b). A cultivar-dependent responsewas observed in M. sativa with 94%, 25%, and 4% of infectedstem explants producing transformed roots in the cultivars Vertus,Regen-S, and Rangelander, respectively. In O. viciifolia cv.Hampshire Giant, an explant-dependent response was observedwith 78% and 50% of seedling cotyledon and hypocotyl explantsresponding, respectively. Leaf explants failed to produce transformedroots. Transformed roots showed plagiotropic and negativelygeotropic growth on hormone-free agar MS medium. Productionof transgenic shoots from O. viciifolia root cultures occurredspontaneously. Recovery of transgenic plants from M. salivacv. Rangelander was achieved by transfer of callus (inducedon UM medium containing 2·0mg dm–3 2,4-D and 0·25mg dm–3 kinetin) to MS medium containing 0·5 ingdm–3 BAP and 0·05 mg dm–3 NAA. Cultured rootsof both species synthesized opines (agropine and mannopine).Extensive morphological variation was observed in plants ofM. sativa (clone Al) and O. viciifolia (clone A4Tl) establishedin the glasshouse. DNA sequences homologous to TL-DNA and TR-DNAwere present in root clones and regenerated plants. Feeding and metabolism of the siphonophore Sphaeronectes gracilis   总被引:1,自引:0,他引:1  
The in situ predation rate of the siphonophore Sphaeronectesgracilis was estimated from gut content analysis of hand-collectedsiphonophores and from laboratory data on digestion rates ofprey organisms. At daytime prey densities of 0.25 copepods 1–1,S. gracilis was estimated to consume 8.1 – 15.4 prey day–1siphonophore–1. From data on abundances of siphonophoresand copepods, S. gracilis was estimated to consume 2–4%of the copepods daily. In laboratory experiments, ingestionrates averaged 13.8 prey day–1 siphonophore–1 atprey densities of 5 copepods 1–1 and 36.9 at 20 copeods1–1. This was equivalent to a specific ingestion rate(for both carbon and nitrogen) of –17% day–1 and45% day–1, respectively, while specific ingestion in situwas only 2% day–1. Ammonium excretion averaged 0.095 µg-atsiphonophore–1 day–1 at 5 prey 1–1, and 0.162at 20 prey 1–1. The effects of root age, temperature, and soil water statuson root hydraulic conductivity (LP) were investigated for twocactus species, Ferocactus acanthodes and Opuntia ficus-indica.The volumetric flux density of water was measured for excisedroot segments, either using negative hydrostatic pressures appliedto the proximal end or using reverse flow of water from theroot to the soil. For both species, LP at 20 ?C increased withroot age, average values reaching a maximum of 3.9 ? 10–7m s–1 MPa–1 for F. acanthodes and 5.2 ? 10–7m s–1 MPa–1 for O.ficus-indica at 11 to 17 weeksof age; LP subsequently declined with increasing root age forboth species. LP was maximal at a temperature of about 10 ?Cfor the youngest roots (1–3 weeks), this optimum shiftingto 40 ?C for 8-week-old roots of both species. For older roots(up to 1.5-years-old), LP increased with temperature from 0?C to 50 ?C, with a Q10 of 1.3 between 20 ?C and 30 ?C. At asoil water potential (soil) of –0.016 MPa, root LP wasindependent of the direction of water flow for both species.Depending on root age, LP declined 45- to 500-fold for F. acanthodesand 90- to 800-fold for O.ficus-indica as soil was reduced from–0.016 to –1.06 MPa, consistent with a rectifier-likebehaviour with respect to water movement between soil and roots.Incorporation of such responses into water uptake models shouldlead to a better understanding of root function. Mesophyll resistance to photosynthetic carboxylation (r'm) wasused as a criterion for leaf integrity. It was measured, at25 °C, in the light, before and after periods of high temperature(3 h at 38 °C) in the dark. During the high temperatureperiods, respiration (RD) of attached leaves of Xanthium strumariumwas suppressed from 27%-36% by either low [O2] (1.04% or 0.21%v.v.) or high [CO2] (840 µl 1–1) in the ambientair. Neither treatment affected rates of RD or photo-respirationduring the second period at 25 °C. There was no significant increase of r'm when RD was not suppressedduring the high temperature treatment. When RD was suppressedat high temperatures, r'm increased from about 3s cm–1before, to about 26 s cm–1 after the high temperaturetreatment. The increase depended upon the degree of suppression. It is concluded that increased RD at high temperature in Xanthiumleaves is partly the result of an increase of energy demandingmaintenance. The subsequent rate of carbon dioxide fixationis reduced when this increase of maintenance-induced respirationis inhibited.  相似文献

Photosynthetic gas exchange has been surveyed in 22 of the 23species currently placed in the genus Oryza and constitutingthe wild relatives of cultivated rice. Unimproved, wild germplasmof a number of species showed light-saturated assimilation ratesin atmospheric air at least as great as cultivars and elitebreeding material of the cultigen O. satlva. One of these specieswas O. australiensis, different accessions of which were significantly(P=0.001) superior in assimilation rate to the 0. satlva genotypestested, including representatives of the Indica, Japonica andJavanica subgroups. Amongst species, assimilation rate was correlatedpositively with light saturation and with carboxylation efficiency.The wild species fell into two distinct groupings accordingto whether they originated from sun or shade habitats, withthe higher assimilation rates being associated with the sunspecies. Assimilation rates were also higher in diploids thanin tetraploids and this was associated with the fact that allsun species are diploids and all tetraploids are shade species.The carbon dioxide compensation concentrations ranged from 28to 43µmol mol–1 with the two lowest values (28 and32µmol mol–1) coming from accessions of O. rufipogon.The mean value for the absolute quantum yield of photosynthesismeas ured on attached leaves was 0.060. There was a large rangein the activity of phosphoenolpyruvate (PEP) carboxylase witha number of species having rates several times those of C3 species.Some species with the highest assimilation rates were assessedfor photorespiratory losses and these were generally around30% and similar to O. satlva cultivars. However, a range ofO. rufipogon accessions had photorespiration rates significantly(P=0.01) lower than the O. sativa genotypes tested. No speciesin the genus possessed C4 photosynthetic metabolism though somedid overlap with compensation concentrations and phosphoenolpyruvatecarboxylase activities reported for C3–C4 intermediatespecies. The growth rates of Appendicularia sicula, Fritillaria borealissargassi, Fritillaria haplostoma, Oikopleura dioica and Oikopleuralongicauda were determined from microcosms incubated in situat 23C in Jamaican waters. Experiments were conducted fromoligotrophic offshore waters, through mesotrophic Lime Cay andeutrophic Kingston Harbour in both natural and nutrient-enhancedphytoplankton communities. Length-weight relationships werecalculated for two of these species: O.longicauda log W=2.47log TL –6.10 and F.haplostoma log W=2.44 log TL –7.37,where weight (W) is in micrograms and trunk length (TL) is inmicrometres. Instantaneous growth rates averaged 1.7–2.5day–1 for the five species and were observed as high as3.3 day–1 These instantaneous rates are equivalent todaily specific growth rates averaging 4.6–11.4 and rangingup to 28. In larger genera, growth rates were related positivelyto picoplankton and nanoplankton concentration, and negativelyto the biomass of larvaceans, but in the smallest species growthwas unrelated to these factors. The Km(CO2) ancl Vmax of ribulose 1,5-bisphosphate (RuBP) carboxylaseand its protein ratio to total soluble protein from Oryza speciesincluding cultivars (25 varieties) and wild types (11 species,21 strains) were surveyed. Their variabilities among cultivarsof O. sativa were very small. The averages of the Km(CO2) andVmax values and the ratio of carboxylase to soluble protein,and their standard errors were 10.2?1.0µM, 1.72?0.13units.mg–1(pH 8.0 and 25?C) and 52?2%, respectively. However, some differencesseemed to exist based on genome constitution in the Oryza genus.RuBP carboxylases from the species with the AgAg genome, O.graberrima and O. breviligulate, exhibited low Km(CO2) values(8.0?0.8 µM). High Vmax was associated with the CC genome,O. eichingeri and O. officinalis (2.08?0.15 units.mg–1).A higher ratio of RuBP carboxylase protein to soluble proteinwas found for the AA genome, O. sativa and O. perennis. Trichoderma viride can be grown in deuterium oxide (D2O) concentrationsas high as 99.7%. Increasing concentrations of D2O (25–90%)progressively extend the lag phase in growth, but do not greatlyaffect the linear growth rate itself. The minimum age and sizeof colony required for photoinduction of the conidiation processis also increased. In 95% D2O, although growth rates are stillrelatively high, both photomorphogenesis and dark conidiationare completely blocked. This block does not appear to involvethe photoprocess itself, but rather post photoinductive processessuch as differentiation of conidiophores. These observations point to an alternative, or at least additional,hypothesis to the one frequently cited—that D2O acts throughstabilizing or "temperature lowering mechanisms.
