Background and Aims

The source of nitrogen plays an important role in salt tolerance of plants. In this study, the effects of NaCl on net uptake, accumulation and transport of ions were investigated in Nerium oleander with ammonium or nitrate as the nitrogen source in order to analyse differences in uptake and cycling of ions within plants.

Methods

Plants were grown in a greenhouse in hydroponics under different salt treatments (control vs. 100 mm NaCl) with ammonium or nitrate as the nitrogen source, and changes in ion concentration in plants, xylem sap exuded from roots and stems, and phloem sap were determined.

Key Results

Plant weight, leaf area and photosynthetic rate showed a higher salt tolerance of nitrate-fed plants compared with that of ammonium-fed plants. The total amount of Na+ transported in the xylem in roots, accumulated in the shoot and retranslocated in the phloem of ammonium-fed plants under salt treatment was 1·8, 1·9 and 2·7 times more, respectively, than that of nitrate-treated plants. However, the amount of Na+ accumulated in roots in nitrate-fed plants was about 1·5 times higher than that in ammonium-fed plants. Similarly, Cl transport via the xylem to the shoot and its retranslocation via the phloem (Cl cycling) were far greater with ammonium treatment than with nitrate treatment under conditions of salinity. The uptake and accumulation of K+ in shoots decreased more due to salinity in ammonium-fed plants compared with nitrate-fed plants. In contrast, K+ cycling in shoots increased due to salinity, with higher rates in the ammonium-treated plants.

Conclusions

The faster growth of nitrate-fed plants under conditions of salinity was associated with a lower transport and accumulation of Na+ and Cl in the shoot, whereas in ammonium-fed plants accumulation and cycling of Na+ and Cl in shoots probably caused harmful effects and reduced growth of plants.Key words: Mineral cycling, Nerium oleander, nitrogen source, salinity, xylem and phloem transport  相似文献

Background and aims

Urea is the predominant form of N applied as fertilizer to crops, but it is also a significant N metabolite of plants themselves. As such, an understanding of urea metabolism in plants may contribute significantly to subsequent N fertilizer management. It currently appears that arginase is the only plant enzyme that can generate urea in vivo. The aim of this work was, therefore, to gain a more in-depth understanding of the significance of the inhibition of endogenous urease activity and its role in N metabolism depending on the N source supplied.

Methods

Pea (Pisum sativum cv. Snap-pea) plants were grown with either ammonium or nitrate as the sole N source in the presence or absence of the urease inhibitor NBPT.

Results

When supplied, NBPT is absorbed by plants and translocated from the roots to the leaves, where it reduces endogenous urease activity. Different N metabolic responses in terms of N-assimilatory enzymes and N-containing compounds indicate a different degree of arginine catabolism activation in ammonium- and nitrate-fed plants.

Conclusions

The arginine catabolism is more highly activated in ammonium-fed plants than in nitrate-fed plants, probably due to the higher turnover of substrates by enzymes playing a key role in N recycling and remobilization during catabolism and in early flowering and senescence processes, usually observed under ammonium nutrition.  相似文献

Background and aims

The effects of salt stress on the salt marsh halophyte Spartina alterniflora have been well documented. However, plant responses to combined salinity and ammonium toxicity and the underlying mechanisms are relatively unknown. The aim of the present investigation was to study the effects of both salinity (0, 200 and 500 mM NaCl) and nitrogen form (NO3 ?, NH4 + or NH4NO3) on S. alterniflora.

Methods

Plants were cultivated in sandy soil under greenhouse conditions for 3 months. At harvest, growth parameters were measured and leaf samples were analysed for oxidative stress parameters (malondialdehyde, MDA; electrolyte leakage, EL; and hydrogen peroxide, H2O2 concentration) and the activity of antioxidant enzymes (glutathione reductase, GR; superoxide dismutase, SOD; catalase, CAT; ascorbate peroxidase, APX and Guaiacol peroxidase, GPX).

Results

In the absence of NaCl, plant growth rate was the highest in the medium containing both nitrogen forms, and the lowest in the medium containing only nitrate. Irrespective of the nitrogen form, plant growth was generally higher at 200 mM NaCl than without salinity. Ammonium-fed plants showed better growth than nitrate-fed plants under high salinity. In the absence of salinity, ammonium-fed plants showed higher SOD, APX, GR, CAT, and GPX activities than nitrate-fed ones. The antioxidant enzymes exhibited higher activity in saline-treated plants. The considerable advantage of NH4 + nutrition to S. alterniflora under saline conditions was associated with high antioxidant enzyme activities, together with low MDA content, EL, and H2O2 concentration.

Conclusion

These data clearly demonstrate that NH4 + is more favourable for the growth of S. alterniflora under high salinity than NO3 ?. It is suggested that NH4 + nutrition improves the plant’s capacity to limit oxidative damage by stimulating the activities of the major antioxidant enzymes.  相似文献

In plants of wheat (Triticum aestivum L.) grown in the media with nitrate (NO 3 ? plants), ammonium (NH 4 + plants), and without nitrogen (N-deficient plants), the response to oxidative stress induced by the addition of 300 mM NaCl to the nutrient solution was investigated. Three-day-long salinization induced chlorophyll degradation and accumulation of malondialdehyde (MDA) in the leaves. These signs of oxidative stress were clearly expressed in NO 3 ? and N-deficient plants and weakly manifested in NH 4 + plants. In none of the treatments, salinization induced the accumulation of MDA in the roots. Depending on the conditions of N nutrition, salt stress was accompanied by diverse changes in the activity of antioxidant enzymes in the leaves and roots. Resistance of leaves of NH 4 + plants to oxidative stress correlated with a considerable increase in the activities of ascorbate peroxidase and glutathione reductase. Thus, wheat plants grown on the NH 4 + -containing medium were more resistant to the development of oxidative stress in the leaves than those supplied with nitrate.  相似文献

Background and aims

Nickel (Ni) has a critical role in the urea metabolism of plants. This study investigated the impact of seed Ni content along with external Ni supply on the growth, various nitrogen (N) metabolites and N use efficiency (NUE) of soybean plants.

Methods

Soybean plants raised from Ni-poor or Ni-rich seeds were grown in nutrient solution with or without external Ni supply and fed with either urea or nitrate as the sole N source. The changes in growth, leaf chlorophyll levels, Ni and N concentrations of different plant parts, tissue accumulation of various N metabolites and N uptake of soybean as well as NUE and its components were examined.

Results

Nickel starvation reduced the shoot biomass of urea-fed plants by 25 % and the leaf chlorophyll levels by up to 35 %, but nitrate-fed plants were unaffected. Visual toxicity symptoms were not observed in urea-fed plants. Under urea supply, Ni-deficient plants had lower levels of total N, protein and free amino acids in various organs. Root uptake of urea was severely depressed in Ni-deprived plants. Availability of Ni did not have any effect on the NUE of nitrate-fed plants, whereas its deficiency reduced the NUE of urea-fed plants by 30 %. The growth and N nutritional status of urea-fed soybean were significantly improved by high seed Ni reserves as well as external Ni supply.

Conclusion

Adequate Ni supply is required for maximizing the growth, root uptake of urea and NUE of urea-fed plants. Seed Ni reserves contribute significantly to the Ni and thus N nutritional status of soybean.  相似文献

Background and aims

Urea represents over 50 % of nitrogen fertilizers applied worldwide to crop production, however one-third of nitrogen fertilized could be recovered by crops. Previous studies have provided useful knowledge of urea-related plant nitrogen-nutrition, whereas information about crop growth-response to urea without its external degradation seems limiting. We thus assess the ability of rice seedlings to use urea at a physiological level.

Methods

Rice growth on urea versus other nitrogen regimes was tested under aseptic conditions. Activity of urease and GS was analyzed; urea, ammonium, total nitrogen and expression of a nitrogen limitation-responsive gene OsDUR3 were examined.

Results

Growth phenotyping revealed urea-dose-dependent growth improvement but significant growth reduction associated with nitrogen-deficiency of plants compared to those on other nitrogen-sources, indicating a physiological impediment of effective urea utilization by rice. Enzymatic assay showed that activities of urease and GS were well expressed in plants supplied with urea. Low concentrations of urea and ammonium were detected in rice (particularly in roots) on 1 mM urea or other nitrogen-forms, and a less nitrogen-content was determined in urea-fed plants. Additionally, the strongest OsDUR3-expression occurred in seedlings on no-nitrogen or 1 mM urea.

Conclusions

We suggest that insufficient urea-absorption but not assimilation represents likely a factor contraining rice to use urea as sole nitrogen-source.  相似文献

Background and aims

To improve vegetable crops adapted to low input and variable resource availability, better understanding is needed of root system functioning, including nitrogen and water capture.

Methods

This study quantified shoot and root development and patterns of water and nitrate capture of two lettuce cultivars subjected to temporary drought at two development stages (Trial 1) or to continuous, localized drought and/or nitrate shortage (Trial 2).

Results

In Trial 1, early drought slowed down shoot and root growth, whereas late drought enhanced root proliferation in the top 0.1 m. Nitrate capture during drought was sustained by increased nitrate inflow from deeper layers. Plants did not recover fully from drought after re-watering. In Trial 2, root proliferation was stimulated in the drier soil compartment partially compensating reduced water availability and nitrate mobility. Under nitrate shortage, root proliferation was enhanced in the compartment where nitrate was more abundant, irrespective of water availability.

Conclusions

Changes observed in the root system are ‘feed-forward’ mechanisms to sustain resource capture in a limiting growing environment. The type of stress (drought or nitrate shortage) affects coping strategies; nitrate concentration in the soil solution, combined with the nutritional status of the plant will determine the stress response.  相似文献

Background & Aims

The effects of an alfalfa plant (Medicago sativa L.) hydrolysate-based biostimulant (EM) containing triacontanol (TRIA) and indole-3-acetic acid (IAA) were tested in salt-stressed maize plants.

Methods

Plants were grown for 2 weeks in the absence of NaCl or in the presence (25, 75 and 150 mM). On the 12th day, plants were supplied for 48 h with 1.0 mg L?1 EM or 11.2 μM TRIA.

Results

EM and TRIA stimulated the growth and nitrogen assimilation of control plants to a similar degree, while NaCl reduced plant growth, SPAD index and protein content. EM or TRIA increased plant biomass under salinity conditions. Furthermore, EM induced the activity of enzymes functioning in nitrogen metabolism. The activity of antioxidant enzymes and the synthesis of phenolics were induced by salinity, but decreased after EM treatment. The enhancement of phenylalanine ammonia-lyase (PAL) activity and gene expression by EM was consistent with the increase of flavonoids.

Conclusion

The present study proves that the EM increases plant biomass even when plants are grown under salinity conditions. This was likely because EM stimulated plant nitrogen metabolism and antioxidant systems. Therefore, EM may be proposed as bioactive product in agriculture to help plants overcome stress situations.  相似文献

Background and aim

Because the indigenous burrowing lagomorph plateau pika (Ochotona curzoniae) is considered to have negative ecological impacts on alpine meadow steppe grasslands of the Headwaters Region of the Yellow, Yangtze and Mekong Rivers we investigated its effects on ecosystem productivity and soil properties, and especially net ecosystem carbon flux.

Methods

We measured net ecosystem CO2 exchange (NEE) and its components gross ecosystem productivity (GEP) and ecosystem respiration (ER) at peak aboveground biomass by the chamber method with reference to plant and soil characteristics of areas of alpine meadow steppe with different densities of pika burrows.

Results

Higher burrow density decreased NEE, GEP and ER. Above-ground biomass, species number, plant cover and leaf area index decreased with increasing pika density. Higher burrow density was associated with lower soil moisture and higher soil temperature. Responses of NEE were related to changes of abiotic and biotic factors affecting its two components. NEE was positively related to soil moisture, soil ammonium nitrogen, plant cover, leaf area index and above-ground biomass but was negatively correlated with higher soil nitrate nitrogen.

Conclusion

Decrease of NEE by plateau pika may reduce the carbon sink balance of Qinghai-Tibet plateau grassland. Such effects may be influenced by grazing pressure from domestic livestock, population levels of natural predators, and climate change.  相似文献

Background and aims

Characterisation of genetic variation in nitrate accumulation by lettuce will inform strategies for selecting low-nitrate varieties more capable of meeting EU legislation on harvested produce. This study uses a population of recombinant inbred lines (RILs) of lettuce to determine how genotypic differences influence N uptake, N assimilation and iso-osmotic regulation, and to identify key related traits prior to future genetic analysis.

Methods

Measurements were made on plants grown to maturity in soil fertilised with ammonium nitrate, and in a complete nutrient solution containing only nitrate-N. A simple osmotic balance model was developed to estimate variations in shoot osmotic concentration between RILs.

Results

There were significant genotypic variations in nitrate accumulation when plants were grown either with nitrate alone or in combination with ammonium. Ammonium-N significantly reduced nitrate in the shoot but had no effect on its relative variability, or on the ranking of genotypes. Shoot nitrate-N was correlated positively with total-N and tissue water, and negatively with assimilated-C in both experiments. Corresponding relationships with assimilated-N and shoot weight were weaker. Estimated concentrations of total osmotica in shoot sap were statistically identical in all RILs, despite variations in nitrate concentration across the population.

Conclusions

Approximately 73% of the genotypic variability in nitrate accumulation within the population of RILs arose from differences in nitrate uptake and only 27% from differences in nitrate assimilated, irrespective of whether or not part of the N was recovered as ammonium, or whether the plants were grown in soil or solution culture. Genotypic variability in nitrate accumulation was associated with changes in concentrations of other endogenous solutes (especially carboxylates and soluble carbohydrates) and of tissue water, which minimised differences in osmotic potential of shoot sap between RILs. This offers the opportunity of using the regulation of these solutes as additional traits to manipulate nitrate accumulation.  相似文献

Background

Water and nutritional restrictions are limiting factors for the growth of Eucalyptus trees in tropical climates. In the dry season, boron (B) uptake is severely affected.

Aims

The objectives of this study were to evaluate the phloem mobility of B and whether its deficiency can increase plant sensitivity to osmotic stress. It was also tested to what extent foliar application of B could mitigate the negative effects of drought under low B supply.

Methods

Seedlings of a drought tolerant Eucalyptus urophylla (Blake, S. T.) clone were grown in nutrient solution, subjected to low availability of B for 25 days, and then submitted to a progressive osmotic stress. After imposition of osmotic stress, B was applied to young or mature leaves.

Results

B applications, mainly to mature leaf, stimulated root growth and delayed dehydration under osmotic stress and led to an increased B translocation and carbon isotopic composition. The expression of B transporters and pectin metabolism genes were also increased in water-stressed plants supplied with B by foliar application.

Conclusions

B deficiency led to increased plant dehydration and decreased root growth under osmotic stress. The application of B to mature leaf of water-stressed plants proved effective in mitigating the negative effects of water deficit in root growth.  相似文献

Utilization of nitrogen in the form of either nitrate (NO 3 ? ) or ammonium (NH 4 + ) ions may affect the carbohydrate metabolism and energy budget of plants. Recent studies showed that greater expenses of NO 3 ? to NH 4 + reduction mostly occur in the roots and during darkness. Fertilization of corn with 15N-labeled nitrate and ammonium, combined with pulse labeling of plants in a 14CO2 atmosphere at the V6 and V8 growth stages, allowed us to evaluate the effect of N form on the CO2 efflux from soil. NH 4 + oxidation was inhibited by adding dicyandiamide. In respect to ammonium, nitrate addition increased root-derived CO2 efflux from corn by 2.6 times at stage V6 and by 1.8 times at stage V8. The time of peak 14CO2 efflux from soil also differed between two growing stages: at V6, efflux peaked only on the second day after pulse labeling, while at V8 this occurred within the first 6 h. The strong effect of NO 3 ? and NH 4 + on root respiration requires considering the N form in the soil and the nitrate reduction site location in a plant when modeling soil respiration changes and when separately estimating individual CO2 sources that contribute to the total soil CO2 efflux.  相似文献

Background and aims

Biodegradation of polycyclic aromatic hydrocarbons (PAHs) is accelerated in the presence of plants, due to the stimulation of rhizosphere microbes by plant exudates (nonspecific enhancement). However, plants may also recruit specific microbial groups in response to PAH stress (specific enhancement). In this study, plant effects on the development of rhizosphere microbial communities in heterogeneously contaminated soils were assessed for three grasses (ryegrass, red fescue and Yorkshire fog) and four legumes (white clover, chickpea, subterranean clover and red lentil).

Methods

Plants were cultivated using a split-root model with their roots divided between two independent pots containing either uncontaminated soil or PAH-contaminated soil (pyrene or phenanthrene). Microbial community development in the two halves of the rhizosphere was assessed by T-RFLP (bacterial and fungal community) or DGGE (bacterial community), and by 16S rRNA gene tag-pyrosequencing.

Results

In legume rhizospheres, the microbial community structure in the uncontaminated part of the split-root model was significantly influenced by the presence of PAH-contamination in the other part of the root system (indirect effect), but this effect was not seen for grasses. In the contaminated rhizospheres, Verrucomicrobia and Actinobacteria showed increased populations, and there was a dramatic increase in Denitratisoma numbers, suggesting that this genus may be important in rhizoremediation processes.

Conclusion

Our results show that Trifolium and other legumes respond to PAH-contamination stress in a systemic manner, to influence the microbial diversity in their rhizospheres.  相似文献

Background and Aims

One of the most striking attributes of clonal plants is their capacity for physiological integration, which enables movement of essential resources between connected ramets. This study investigated the capacity of physiological integration to buffer differences in resource availability experienced by ramets of the clonal wild strawberry plant, Fragaria vesca. Specifically, a study was made of the responses of connected and severed offspring ramets growing in environments with different water availability conditions (well watered or water stressed) and nitrogen forms (nitrate or ammonium).

Methods

The experimental design consisted of three factors, ‘integration’ (connected, severed) ‘water status’ (well watered, water stressed) and ‘nitrogen form’ (nitrate, ammonium), applied in a pot experiment. The effects of physiological integration were studied by analysing photochemical efficiency, leaf spectral reflectance, photosynthesis and carbon and nitrogen isotope discrimination, the last of which has been neglected in previous studies.

Key Results

Physiological integration buffered the stress caused by water deprivation. As a consequence, survival was improved in water-stressed offspring ramets that remained connected to their parent plants. The nitrogen isotope composition (δ15N) values in the connected water-stressed ramets were similar to those in ramets in the ammonium treatment; however, δ15N values in connected well-watered ramets were similar to those in the nitrate treatment. The results also demonstrated the benefit of integration for offspring ramets in terms of photochemical activity and photosynthesis.

Conclusions

This is the first study in which carbon and nitrogen isotopic discrimination has been used to detect physiological integration in clonal plants. The results for nitrogen isotope composition represent the first evidence of preferential transport of a specific form of nitrogen to compensate for stressful conditions experienced by a member clone. Water consumption was lower in plants supplied with ammonium than in plants supplied with nitrate, and therefore preferential transport of ammonium from parents to water-stressed offspring could potentially optimize the water use of the whole clone.  相似文献

Background and aims

Soil drying leads to the generation of chemical signals in plants that regulate water use via control of the stomatal aperture. The aim of our work was to identify the presence and identity of potential chemical signals, their dynamics, and their relationship with transpiration rate during soil drying in hop (Humulus lupulus (L.)) plants.

Methods

We used pressure chamber technique for measurement of shoot water potential and collection of shoot xylem sap. We analyzed concentrations of abscisic acid (ABA), nitrate, phosphate, sulphate and malate in sap and also the rate of whole plant transpiration.

Results

Transpiration rate decreased prior to changes in shoot water potential. The concentration of ABA in xylem sap continuously increased from early to later stages of water stress, whereas in leaves it increased only at later stages. Shoot sap pH increased simultaneously with the decrease of transpiration rate. Xylem sap alkalization was in some cases accompanied by a decrease in nitrate concentration and an increase in malate concentration. Concentration of sulphate increased in xylem sap during drying and sulphate in combination with a higher ABA concentration enhanced stomatal closure.

Conclusions

Several early chemical signals appear in sap of hop plants during soil drying and their impact on transpiration may vary according to the stage of soil drying.  相似文献

Aims

To evaluate the external and internal morphological differences of roots that might influence rice root radial oxygen loss (ROL) and the corresponding rhizosphere nitrification activity, growth characteristics and nitrogen nutrition of rice.

Methods

The root ROL and rhizosphere oxygen profile were determined using a miniaturised Clark-type oxygen microelectrode system, and the rhizosphere nitrification activity was studied with a short-term nitrification activity assay.

Results

The rice biomass, nitrogen accumulation and nitrogen use efficiency (NUE) of ZH (high yield) were significantly higher than those of HS (low yield). The root biomass, number, diameter and porosity of ZH were also much greater than those of HS. The inner and surface oxygen concentrations of the root of ZH were significantly higher than those of HS. The order of paddy soil oxygen penetration depth was ZH?>?HS?>?CK, and the order of the oxygen concentrations detected in the water layer and rhizosphere soil was the same. The rhizosphere nitrification activity and nitrate concentration of ZH were significantly higher than those of HS.

Conclusions

More porous and thicker roots improved the individual root ROL, and more adventitious root numbers enhanced the entire plant ROL and correspondingly improved the rhizosphere nitrification activity, which might influence the growth and nitrogen nutrition of rice.  相似文献

Abstract

Chilling stress is a crucial factor that limits the geographical distribution and yield of chilling-sensitive plants. Ascorbate (AsA) protects plants by scavenging reactive oxygen species and reduces photoinhibition by promoting the conversion of violaxanthin to zeaxanthin in the xanthophyll cycle to dissipate excess excitation energy. Possible mechanisms of AsA for plant photoprotection under chilling stress were investigated by isolating the tomato GDP-l-galactose phosphorylase gene (SlGGP) and producing transgenic tobacco plants with overexpression of SlGGP. The transgenic plants subjected to chilling stress accumulated less H2O2, demonstrated lower levels of ion leakage and malondialdehyde, and acquired higher net photosynthetic rate, higher maximum photochemical efficiency of PSII, and higher D1 protein content compared with the wild-type (WT) plants. The transgenic plants subjected to chilling stress also showed higher GDP-l-galactose phosphorylase activity, increased AsA content as well as ascorbate peroxidase and oxidizable P700 activities than WT plants. Thus, SlGGP overexpression is crucial in promoting AsA synthesis and alleviating photoinhibition of two photosystems.  相似文献
