In addition to several cytochromes three iron sulfur proteins were detected in mixotrophically grown cells of Ectothiorhodospira shaposhnikovii, a member of the Chromatiaceae. They were identified as a bacterial ferredoxin and two high potential iron sulfur proteins (HIPIPs). The two HIPIPs were purified and characterized. They were named according to their differing retention times on a DEAE-cellulose column using a continuous NaCl gradient: early and late HIPIP. The HIPIPs contain 4 mol of non-heme iron and 4 mol of acid labile sulfur per mol protein. Under the conditions of purification the early HIPIP (E
m, 7+270 mV) was present in a semi-reduced state. Using ion-exchange chromatography the early HIPIP could be split into a reduced green-brown (pI=3.7) and an oxidized red-brown (pI=3.9) fraction. Lipopolysaccharides were isolated from the moderate halophilic Ectothiorhodospira shaposhnikovii slight to and Ectothiorhodospira mobilis and from the extremely halophilic Ectothiorhodospira halophila by the hot phenol-water and purified by the phenol-chloroform-petroleum ether methods. The isolated lipopolysaccharides of all three species contained 3-deoxy-d-manno-octulosonic acid and d-glycero-d-mannoheptose indicating the existence of a core. They contained additionally glucose and uronic acids (E. shaposhnikovii and E. mobilis) or glucose, uronic acids and threonine (E. halophila). Sodium deoxycholate gel-electrophoresis of the three lipopolysaccharides, each showing only one major band, indicated R-type character of the lipopolysaccharides of the three Ectothiorhodospira species.The lipid A fractions of the lipopolysaccharides from E. shaposhnikovii and E. mobilis represented phosphorylated mixed lipid A types with both 2,3-diamino-2,3-dideoxy-d-glucose and d-glucosamine. The lipid A from E. halophila contained also phosphate and 2,3-diamino-2,3-dideoxy-d-glucose but only traces of d-glucosamine, which would indicated lipid ADAG. The fatty acid spectra were characterized by amide-bound 3-OH-10:0 and 3-OH-12:0 (E. shaposhnikovii), 3-OH-10:0 (E. mobilis), or 3-OH-10:0,3-OH-14:0, and 3-oxo-14-0 (E. halophila). The localization of hydrogenase in the phototrophic bacterium Thiocapsa roseopersicina was investigated by subcellular fractionations, and transmission electron microscopic immunocytochemistry. By using sonicated cells and measuring in vitro hydrogenase activities in soluble and membrane fractions, respectively, a weak hydrophobic interaction between the hydrogenase enzyme and the T. roseopersicina membranes was observed. Polyclonal antisera directed against the purified hydrogenase were raised in rabbits and exhibited one band in native-PAGE/Western immunoblot analysis. Native-PAGE/activity stain confirmed the identity of this band as being hydrogenase. Immunocytolocalization experiments using ultrathin sections showed an internal localization of the hydrogenase enzyme. A higher specific labeling was associated with chromatophores, indicating a possible coupling of hydrogenase with the photosynthetic membranes in the T. roseopersicina cells. 相似文献

The lipopolysaccharide of Ectothiorhodospira vacuolata was obtained by the phenol-water procedure. It contained a 3-O-methyl-hexose, glucose, galacturonic and glucuronic acids. The finding of d-glycero-d-mannoheptose and 2-keto-3-deoxyoctonate (tentatively identified) suggested a core-structure. The lipid fraction of the lipopolysaccharide contained phosphate and both, 2,3-diamino-2,3-dideoxy-d-glucose and d-glucosamine. Cooperative interaction of the high-potential hemes (Ch) in the cytochrome subunit of the photosynthesizing bacterium Ectothiorhodospira shaposhnikovii was studied by comparing redox titration curves of the hemes under the conditions of pulse photoactivation inducing single
turnover of electron-transport chain and steady-state photoactivation, as well as by analysis of the kinetics of laser-induced
oxidation of cytochromes by reaction center (RC). A mathematical model of the processes of electron transfer in cytochrome-containing
RC was considered. Theoretical analysis revealed that the reduction of one heme Ch facilitated the reduction of the other heme, which was equivalent to a 60 mV positive shift of the midpoint potential. In
addition, reduction of the second heme Ch caused a three-to four-fold acceleration of the electron transfer from the cytochrome subunit to RC.

A new bacteriochlorophyll b containing phototrophic bacterium was isolated from extremely saline and alkaline soda lakes in Egypt. Enrichment and isolation were performed using a synthetic medium with high contents of sodium carbonate, sodium sulfate and sodium chloride. Photoautotrophic growth occurred with hydrogen sulfide as photosynthetic electron donor. During oxidation of sulfide to sulfate extracellular elemental sulfur globules appeared in the medium. Cells were also capable to grow under photoheterotrophic conditions with acetate, propionate, pyruvate, succinate, fumarate or malate as carbon sources and electron donors. Under these conditions sulfate was assimilated. Optimal growth under the applied experimental conditions occurred at a total salinity of 14–27%, a pH-range between 8.1 and 9.1 and a temperature between 47°C and 50°C. The cells were 0.5–0.6 m wide and, depending on cultural conditions, 2.5–8.0 m long; they were spiral shaped, multiplied by binary fission and were motile by means of bipolar flagella. Intercytoplasmic photosynthetic membranes were present as stacks. Bacteriochlorophyll b was the main photosynthetic pigment; small amounts of carotenoids were mainly present as glucosides of rhodopin and its methoxy derivative. A new phototrophic bacterium was isolated from Jordanian and Kenyan alkaline salt lakes. Cells are rod shaped, 1.5 m wide and 2–4 m long, and motile by polar flagella. They divide by binary fission, and possess photosynthetic membranes as lamellar stacks similar to those in the other species of the genus Ectothiorhodospira and the brown colored Rhodospirillum species. The presence of bacteriochlorophyll a and carotenoids of the normal spirilloxanthin series is indicated by the absorption spectra of living cells. Under certain growth conditions the cells form gas vacuoles, may become immotile and float to the top of the culture medium. Sulfide and thiosulfate are used as photosynthetic electron donors. During the oxidation of sulfide to sulfate, elemental sulfur is formed, which is accumulated outside the cells. The organisms are strictly anaerobic, do not require vitamins, are moderately halophilic and need alkaline pH-values for growth. The new species Ectothiorhodospira vacuolata is proposed. 相似文献

A novel type of purple sulfur bacterium was isolated from a hypersaline sulfur spring on the shore of the Dead Sea. The cells of the isolate are irregularly rod-shaped or curved, and motile by means of a tuft of polar flagella. The photosynthetic system, containing bacteriochlorophyll a and carotenoids of the spirilloxanthin series, is located on stacks of lamellar membranes in the cell cytoplasm. The organism can grow either photoautotrophically with sulfide as electron donor, which is oxidized via extracellular sulfur to sulfate, or photoheterotrophically, using acetate, succinate, fumarate, malate or pyruvate as carbon sources. The bacterium is obligately anaerobic, and requires a source of reduced sulfur for growth. The isolate is moderately halophilic, and grows optimally at NaCl concentrations between 3 and 8%, temperatures between 30 and 45°C, and neutral pH. 16S ribosomal RNA oligonucleotide cataloging suggests a close relationship to purple sulfur bacteria of the genus Ectothiorhodospira. As the isolate differs greatly from the described members of the genus Ectothiorhodospira, we describe the isolate as a new species, and propose the name Ectothiorhodospira marismortui sp. nov. 相似文献

Ectothiorhodospira halochloris grows photoheterotrophically with a variety of sulfur sources. During sulfide oxidation to elemental sulfur considerable amounts of polysulfides may be accumulated transiently. When grown on elemental sulfur no sulfate was produced by oxidation, but sulfide and polysulfide were formed by reduction. Only one soluble cytochrome c-551 was isolated and purified. It was a small acidic hemeprotein with a molecular weight of 6,300, an isoelectric point of 3.1 and a redox potential of-11 mV at pH 7.0. It showed three absorption maxima in the reduced state (=551 nm; =523 nm; =417 nm). The addition of various c-type cytochromes to a suspension of spheroplasts stimulated the velocity of sulfide oxidation. This stimulation was best with the small acidic cytochromes from E. halochloris or Ectothiorhodospira abdelmalekii. Sulfide oxidation was stopped by several uncoupling agents, ionophores and electron transport inhibitors. This study systematically investigated the different types of LH2 produced by Allochromatium (Alc.) vinosum, a photosynthetic purple sulphur bacterium, in response to variations in growth conditions. Three different spectral forms of LH2 were isolated and purified, the B800-820, B800-840 and B800-850 LH2 types, all of which exhibit an unusual split 800 peak in their low temperature absorption spectra. However, it is likely that more forms are also present. Relatively more B800-820 and B800-840 are produced under low light conditions, while relatively more B800-850 is produced under high light conditions. Polypeptide compositions of the three different LH2 types were determined by a combination of HPLC and TOF/MS. The B800-820, B800-840 and B800-850 LH2 types all have a heterogeneous polypeptide composition, containing multiple types of both α and β polypeptides, and differ in their precise polypeptide composition. They all have a mixed carotenoid composition, containing carotenoids of the spirilloxanthin series. In all cases the most abundant carotenoid is rhodopin; however, there is a shift towards carotenoids with a higher conjugation number in LH2 complexes produced under low light conditions. CD spectroscopy, together with the polypeptide analysis, demonstrates that these Alc. vinosum LH2 complexes are more closely related to the LH2 complex from Phs. molischianum than they are to the LH2 complexes from Rps. acidophila. 相似文献

Abstract The photoreaction center isolated from Ectothiorhodospira sp. is known to contain polypeptides L,M and C (a four heme c cytochrome) in a 1:1:1 molar ratio. As a test for the presence of a H subunit, Western blots of the chromatophore and photoreaction center polypeptides were reacted with a monoclonal antibody directed against the H subunit of Rhodospirillum rubrum . The chromatophores but not the isolated photoreaction center cross-reacted with this antibody. It is concluded that the H polypeptide is lost during the purification of the photoreaction center which, in its native state, therefore contains four polypeptides including a genuine H subunit. 相似文献

Trehalase, which hydrolyzes the disaccharide trehalose to -d-glucose was isolated and partially purified (124-fold) from the phototrophic halo-alkaliphilic bacterium Ectothiorhodospira halochloris. The molecular mass was determined to be 480,000 and the isoelectric point pH 5.6. Temperature optimum was found to be 40°C and the pH-optimum 7.8–8.1. In spite of its high K
m-value of 0.5 M, trehalase of E. halochloris was shown to be specific for trehalose. Trehalase is activated by phosphate which is, however, not involved in the reaction mechanism. The enzyme is activated by the compatible solute betaine and inhibited by salts. In the presence of betaine the K
m-value is lowered from 0.5 M to 0.16 M; moreover, betaine partially protects enzymatic activity from salt inhibition. The findings indicate that betaine might regulate the trehalose level in the cells by affecting trehalase activity. 相似文献

The lamellar membrane stacks of Ectothiorhodospira mobilis were isolated and purified by a combination of lysozyme and osmotic shock treatment, followed by differential and density gradient centrifugation. Preparations of lamellar membranes were enriched at least 2.4-fold in the ratio of bacteriochlorophyll a to protein.Thin-sectioning, negative staining, platinumcarbon shadowing and freeze-etching were used to study the architecture of the membrane units. Both platinum-carbon shadowing and freeze-etching showed the outer surfaces of the isolated lamellar membrane stacks to be relatively smooth. Particles averaging 7 nm in diameter were seen on several faces following freeze-ctching.Non-polar amino acids amounted to 60% of the total amino acid composition. Lipids constituted 32% of the membrane dry weight. Phosphatidyl ethanolamine and diphosphatidyl glycerol were the major phospholipids. Fatty acids of 10–15 carbons represented a small fraction of both membrane and whole cell fatty acids. Monoenes constituted 36% of the total membrane fatty acids and 38.4% of the total whole cell fatty acids. The major fatty acids of both whole cells and purified membranes were C16:0, C18:1 and cyclopropane C19:0. 相似文献

This paper presents diffusion data of N2, O2, H2S and SO2 in MFI and 4A zeolites obtained by molecular dynamics simulations, especially its dependence on temperature and loading. At high loadings and temperatures, the order of self-diffusivity of guests in two zeolites is O2 > N2 > H2S>SO2. The diffusion behaviour is different in different zeolites at lower loadings, reflecting different influences from straight channels (MFI) and α-cages (4A). Furthermore, with increasing loading, the self-diffusivity of guest molecules decreases in MFI but generally increases in 4A. The centre of mass (COM) probability densities and diffusion trajectories of guests give insight into molecular-level diffusion process. The simulation results reveal that with increasing loading, the diffusion mechanism would change from the inter-pore to intra-pore diffusion in MFI. However, in 4A, the intra-pore diffusion is predominant at low and high loadings, but inter-pore diffusion is more important at moderate loadings. 相似文献

Evidence is presented which confirms the existence of genetic transformation in the blue-green bacterium, Anacystis nidulans. This process has been demonstrated for three mutations: streptomycin resistance, a phenylalanine requirement and an ornithine requirement. The optimal conditions under which transformation occurs are also investigated, and the potential of this system for genetic mapping is discussed.This work was submitted by K. G. DNA-DNA hybridization reveals low DNA homologies (about 14%) between the species of Ectothiorhodospira genus and indicate clearly that the degree of divergence within this genus exceeds the interspecific level. The degree of genome similarities of E. mobilis and E. vacuolata (more than 80% homology) is high and characteristic for the strains of one and the same species.The results of rRNA-DNA and secondary DNA-DNA hybridization indicate the following: Ectothiorhodospira and Thiocapsa are far less related than the genera of one and the same family; the genus Ectothiorhodospira is equidistant from both families of purple sulfur and nonsulfur bacteria. Thus Ectothiorhodospira is a taxon of a higher rank than a genus; we agree with Imhoff's proposal of a new family Ectothiorhodospiraceae. 相似文献

A detailed analysis of the periplasmic electron carriers of the photosynthetic bacterium Ectothiorhodospira sp. has been performed. Two low mid-point redox potential electron carriers, cytochrome c′ and cytochrome c, are detected. A high potential iron–sulfur protein is the only high mid-point redox potential electron transfer component
present in the periplasm. Analysis of light-induced absorption changes shows that this high potential iron–sulfur protein
acts in vivo as efficient electron donor to the photo-oxidized high potential heme of the Ectothiorhodospira sp. reaction center.

A cytochrome c-551 was isolated and purified from Ectothiorhodospira abdelmalekii. It is a small acidic haemoprotein with a molecular weight of 9,500, an isoelectric point of 3.5 and a redox potential of-7 mV at pH 7.0. It showed three maxima in the reduced state (=551, =529, =417). The best purity index (A280/A417) obtained was 0.29. During sulfide oxidation to elemental sulfur a considerable amount of polysulfides were transiently accumulated. The digestion experiment can be taken to indicate that cytochrome c-551 is localized on the outside of the cell membrane. The addition of cytochrome c-551 to a suspension of spheroplasts stimulated the velocity of sulfide oxidation. In leaves of birch (Betula pendula Roth), changes in the content of total sulphur and its inorganic and organic forms were determined in relation to the decreasing air-pollution load (SO2) in the air-polluted Krusne hory mountains and the Decin sandstone highlands in 1995, 1998, 2001 and 2004. Results have shown that birch is able to use considerable amounts of sulphur taken through leaves from air-pollution load. Birch responds fast to changes in air-pollution load by fall in the content of total and inorganic forms of sulphur in leaves. 相似文献

The halophilic phototrophic bacterium Ectothiorhodospira halochloris is able to synthesize both nitrogen-containing (betaine, ectoine) and nitrogen-free (trehalose) compatible solutes. In the absence of external ammonium and under nitrogen-limited growth conditions ectoine was metabolized and trehalose partly replaced betaine. The cytoplasmic trehalose concentration did not exceeded 0.5 mol/kg water (approx. 30% of total compatible solutes). A decreasing content of betaine in cells growing under nitrogen limitation is a result of decreased biosynthesis. Apparently, the betaine pool cannot be used as a nitrogen source, not even in a situation of total nitrogen depletion. 相似文献
