We have sought a method capable of detecting small changes in mitochondrial cristae thickness under normal and experimental conditions. Using conventional electron microscopy and goniometry, we studied changes in mitochondria of rat heart muscle and of choroid plexus ependyma caused by treatment with 2,4-dinitrophenol (DNP). Calculation of real thickness was made using goniometric data (formula shown) and the results checked by the method of Casley-Smith and Davy. DNP produced a thickness increase of low statistical significance in choroid plexus ependymal mitochondrial cristae, but a decrease for heart muscle cristae. Although our findings do not resolve the problem of DNP-induced cristae changes, our modified goniometric method may be useful for other studies.  相似文献

Summary A developmental study at the electron microscopic level was conducted of the fat body cells of Hyalophora cecropia (L.). During the last larval instar the fat body increases in volume and the cells exhibit a well developed rough endoplasmic reticulum and protein bodies of diverse sizes. In the pupal fat body, the protein bodies appear to be enclosed by a double membrane and contain glycogen granules, ribosomes and mitochondrion-like structures. In addition, there are large lipid globules, cytolysomes and rough endoplasmic reticulum. The ultrastructure of the protein bodies suggests the development of large bodies by fusion of smaller protein bodies. Changes in fat body cell ultrastructure were followed during adult development and cytological evidence was obtained for the depletion of protein, glycogen and lipid in the female during this period. The female adult fat body cell contains free ribosomes, protein bodies, many mitochondria, a few lipid globules and glycogen granules. The male moth fat body cells have many mitochondria, a few glycogen granules, essentially no protein bodies, but an abundance of large lipid globules.Studies on the influence of egg maturation on the morphology of the fat body of Hyalophora gloveri (L.) revealed that ovariectomy of pupae yielded adults having more fat body than normal females, and that the fat body cells of the ovariectomized animals contained more glycogen, lipid and protein. Male pupae receiving ovarian implants developed into adults containing eggs and possessed more fat body than normal females but less than normal males. Summary The ovary of the liver fluke has been studied with light and electron microscopy. The organ consisted of germ cells and a layer of peripheral cells suggested to be nurse cells, and was surrounded by a capsule containing muscular tissue. The peripheral cells rested on a thick basement membrane and were irregular in outline. Their nuclei were of irregular shape, the mitochondria were dark with few cristae and the endoplasmic reticulum was tubular or vesicular and partly studded with ribosomes. The germ cells were rounded or polyhedral except in the outer part of the ovary where some of them showed irregular processes. The germ cells of the outer region (oogonia) were relatively small and in close contact with the cells suggested to be nurse cells. The inner germ cells (oocytes) were large and loosely packed. Their nuclei were irregular and contained round distinct nucleoli. The nuclear envelopes showed numerous pores. The endoplasmic reticulum was very sparse, but free ribosomes were abundant in the cytoplasm. This corresponded with a strong basophilia removable with RNase. In addition round basophilic bodies formed by densely packed ribosomes and membraneous material occurred in close spatial relation to mitochondria. The latter contained dense granules and few cristae. Groups of vesicles and membraneous lamellae were found in the cytoplasm, but they were considerably smaller than vertebrate Golgi complexes. Numerous dense spherical granules were found mainly in the periphery of the large germ cells. The granules were strongly osmiophilic except in the terminal part of the ovary. The ultrastructure of the pedal muscle of the Megalobulimus oblongus is described. This muscle consists of transverse, longitudinal and oblique bundles ensheathed in collagenous tissue. Each muscle cell is also ensheathed by collagen. The smooth muscle cells contain thin and thick filaments; the thin filaments are attached to dense bodies. These cells contain a simple system of sarcoplasmic reticulum, subsarcolemmal caveolae and mitochondria with dense granules in the matrix, and glycogen. Three types of muscle cells were identified. Type A cells exhibited densely packed myofilaments, abundant glycogen rosettes, numerous mitochondria and sarcoplasmic reticulum profiles. Type B cells exhibited scanty glycogen and mitochondria, few cisternae of sarcoplasmic reticulum and large intermyofibrillar spaces. Type C cells exhibited intermediate characteristics between type A and type B cells. Neither nexus nor desmosomes were observed between the muscle cell membranes. The muscle contains well developed connective tissue and blood vessels. These structures and the distribution of muscle cells are probably involved in the muscular-hydrostat system. The muscle is richly innervated, having neuromuscular junctions with clear and electron-dense synaptic vesicles. The clear vesicles probably contain acetylcholine because the axons to which they are connected arise from acetylcholinesterase positive neurones of the pedal ganglion. The other vesicles may secrete monoamines such as serotonin and/or neuropeptides such as substance P.  相似文献

Large populations (up to 600/cell) of spherical, electron-opaque granules ~0.3 to 0.4 µ in diameter are characteristically found in muscle fibers of mammalian atria. They are absent in muscle fibers of the ventricles. The granules are concentrated in the sarcoplasmic core and occur in lesser numbers in the sarcoplasmic layers between myofibrils and under the plasma membrane. Their intimate association with a central voluminous Golgi complex and the frequent occurrence of material reminiscent of the granular content within the cisternae of the Golgi complex suggest that the latter is involved in the formation of the atrial granules. Atrial granules are larger and more numerous in smaller species (rat, mouse), and generally smaller and less numerous in larger mammals (dog, cat, human); they are absent from the atrial fibers of very young fetuses (rat) but are present in those of newborn animals. A small population of bodies containing glycogen particles and remnants of the endoplasmic reticulum and mitochondria occurs in the sarcoplasmic cores of atrial as well as ventricular muscle fibers in the rat; they contain acid phosphatase and thus appear to be residual bodies of autolytic foci. Their frequency increases with the age of the animal. Typical lipofuscin pigment granules, which are known to contain acid phosphatase and are found in the sarcoplasmic cores in old animals (cat, dog and human), are presumed to arise by progressive aggregation and fusion of small residual bodies.  相似文献

The ultrastructure of liver cells was studied in rooks (Corvus frugilegus) living in radioactive and chemical contamination areas. The ultrastructure of liver cells from rook as well as jackdaw (Corvus monedula) and hooded crow (Corvus cornix) (Corvidae family) from a conventionally clean area was studied as control. Control hepatocytes proved to contain a great number of mitochondria, many of which were swollen and had clear matrix and disorganized cristae. The cristae nearly lacked glycogen and had abundant lipid droplets, which often tightly contacted mitochondria. The cytoplasm of hepatocytes in birds from both ecologically unfavorable areas had numerous mitochondria with the same ultrastructure. In contrast to control, the hepatocyte cytoplasm: (1) contained a lot of glycogen; (2) there were many lipid droplets, which directly contacted glycogen granules; and (3) had more abundant peroxisomes. In addition to normal erythrocytes, the sinusoids contained erythrocytes with mitochondria, vesicles, and lipid droplets in their cytoplasm. Analysis of many micrographs of lipid droplets contacting glycogen granules, mitochondria, peroxisomes, and cisterns of smooth endoplasmic reticulum allowed us to propose that glycogen is synthesized via gluconeogenesis from glycerol and products of fatty acid oxidation in the liver cell cytoplasm of rooks from ecologically unfavorable areas as distinct from control.  相似文献

In the experiment performed on 25 non-inbred male rats ultrastructural changes in cardiomyocytes of the hypertrophied heart have been studied under conditions of stress caused by immobilization and possibility to correct these changes by means of thyroid hormones. The stress intensifies destructive lesions in a number of organelles++, which develop at a prolonged hypertrophy, decreases essentially the ratio mitochondria/myofibrils in section area. Small doses of the thyroid hormones protect the hypertrophied heart from the damaging effect of the stress: prevent the stress-induced+ decrease in the ratio mitochondria/myofibrils, as well as stimulate development of the regenerative-adaptive processes (increase in size and number of mitochondria and their crists, elements of the sarcoplasmic reticulum, glycogen granules, increase in section areas of their nuclei and chromatin in them). The thyroid hormones restrict essentially decrease in correlation of organelles++, resulted from hypertrophy. Thus, the stress-induced disturbances in ultrastructure of the hypertrophied heart can be prevent by means of the thyroid hormones, administered in small doses.  相似文献

Summary Electron microscopically the adrenal X zone was examined in the fourteen SMA female mice aged 40 and 70 days. At these ages, the X zone showed no signs of degeneration. The X zone cell was somewhat smaller than the permanent cortical cell.The mitochondria in the X zone cell were quite bizarre in shape, provided with tubules or cristae. Many intramitochondrial bodies very similar to the cytoplasmic lipid droplets were found in the X zone. A few lipid droplets and globules were also noticed in this zone. The lipid droplets may possibly be formed within the mitochondria.The light and dark cells were differentiated. For the light cells, scant mitochondria and tubular granular endoplasmic reticulum were characteristic in contrast to the abundant mitochondria and multi-lamellated agranular endoplasmic reticulum in the dark cells. By means of electron microscopic autoradiography with 3H-thymidine a study was made of the differentiation degree of DNA synthesizing muscle cells in the sinoatrial node (SAN) of the heart conductive system of the 18 day old mouse embryos. Clear myocytes (CM), predominating in the SAN at this stage, are irregular in shape, with interdigitating protrusions. Nuclei are clear, spherical or ellipsoidal. One hour following 3H-thymidine injection, about 6% of CM display labeled nuclei; this index is considerably lower than in working ventricular myocardium. Like unlabeled myocytes, CM being in phase S contain sparse, randomly located thin myofibrilles. In some areas of the sarcoplasm, only myofilament bundles and Z-disk material can be seen. The number of CM myofibrilles is always considerably less than in the working ventricular myocytes. Accumulations of intermediate (8--11 nm) filaments are present. Mitochondria with a few cristae are not numerous. The sarcoplasmic reticulum and Golgi apparatus being relatively well developed, multivesicular bodies, centrioles, and occasional cilia are often seen. Near the centrioles (basal bodies), striated filamentous bundles are found sometimes showing periodic dense lines separated by 50--70 nm. Specialized contacts between CM are rare, being presented only by desmosomes and primitive intercalated discs. Besides CM, sparse small dark cells occur filled with myofibrilles and mitochondria. In the peripheral regions of the node "transitional" cells are seen. The SAN of the 18 day old embryo mouse heart grown due to proliferation of CM with a poorly developed myofibrillar apparatus.  相似文献

The fine structure of epithelial cells of the small ventral prostate of Pseudomys australis males was studied. The cells were sometimes binucleated, exhibited relatively little granular endoplasmic reticulum, generally few secretory granules (probably reflecting a low proteinaceous secretory activity), but had abundant agranular endoplasmic reticulum (AER), similar to that in steroidogenic cells. Some of the mitochondria also showed tubular cristae. Furthermore, most cells had some irregular dense bodies in the cytoplasm which may have developed, by a gradual transformation process, from the membranes of AER, mitochondria and other organelles; they could be the product of degenerative changes in these organelles. These findings indicate a significant difference in the structure of these cells from those present in the ventral prostate of the hopping mouse, Notomys alexis. It is speculated that this gland secretes relatively little protein but perhaps more lipid or cholesterol-derived substance.  相似文献

Pathological alterations of the fine structure of the cardiac muscle of hereditary muscular dystrophic hamsters (BIO 14.6), with peculiar changes of mitochondria, which were possible degenerative, suggest a mode of formation of intramitochondrial dense bodies. The initial stage of this process appeared in the outer compartment of a mitochondrion where the outer membrane began to vesiculate or interdigit with another outer membrane of an adjacent mitochondrion. Then this vesiculated region developed into an intramitochondrial dense area, which further invaded the central part of the mitochondrion keeping its continuity with the cristae and its vesicle substructure. Independently, some parts of the membrane of the mitochondrial cristae fused with each other and formed dark spots. They were also included in the vesiculated intramitochondrial dense area. The vesiculated outer membrane may be involved in deterioration of the calcium binding site compatible with increased calcium of the prenecrotic stage of the cardiac muscle of dystrophic hamster.  相似文献

Following hibernation, most salamander liver cells undergo alterations. The liver cells containing the structure of their active period present a normal nucleus, glycogen granules disseminated throughout the entire cytoplasm and granular mitochondria with several peripheral cristae. Most cells, however, possess altered mitochondria, transformed into electron-dense masses, or clear vesicles with the possibility of digesting the ambient organelles. Other cells have clear vacuoles with similar digestive possibilities. The endoplasmic reticulum is poorly represented in the liver cells that have suffered advanced alteration. The nucleus of these cells is homogeneously granular. The sytoplasm is populated by electron-dense bodies and lipid vacuoles. The nucleolemma and plasmalemma are thickened. The pigmented cells, altered concomitantly with the liver cells, contain granular aggregates. Some of the pigmented cells exhibit electron-clear formations similar to the reflecting platelets of the iridocytes of the integument of lower vertebrates.  相似文献

Summary This research was undertaken in order to determine if known biochemical processes involving the conversion of fluoroacetate are reflected morphologically at the electron microscopic level. No changes were observed in the hepatocytes after 2.5 mg/kg of fluoroacetate and sacrifice at 3 hours, but with 5 mg/kg of the drug there was an increase in the amount of agranular endoplasmic reticulum, an aberrant shape of mitochondria, and a disappearance of visible glycogen. One hour after 10, 15, 20, or 30 mg/kg of fluoroacetate the results showed a tremendous increase in the amount of agranular endoplasmic reticulum, an aberration in mitochondrial structure, and a disappearance of the intramitochondrial granules and glycogen. A quantitative electron microscopical examination of specialized conducting myocytes of types II and III in the atrioventricular node and working myocytes in perinodal region was performed. The volume fractions of myofibrils, mitochondria, nuclei, sarcoplasmic reticulum, glycogen, atrial granules and "clear" cytoplasm were measured, in addition to the diameters of conducting and working myocytes. These data were compared with those for conducting myocytes of types I, II, and III from sinoatrial region and internodal tracts of the rat heart, and with evidence for atrioventricular node of other animals. Principles of an universal classification of conducting myocytes of the heart are discussed.  相似文献

The ultrastructure stages of Blastocystis galli were studied in chicken's intestine and in laboratory cultures. There were found morphological structures: surface coat (cell from chickens' intestine showed a very thick surface coat); cell membrane--there were some small electron-opaque deepening "pockets" on the membrane; inner membrane; endoplasmic reticulum with attached ribosomes, which present in the cytoplasm; all cells contained numerous of small vacuoles and large glycogen inclusions in cytoplasm; mitochondria with tubular cristae; nucleus with granules condensed chromatin; central vacuole; Golgi complex was represented by number of plates grouped in a pite; the cyst-like forms were surrounded by multilayered wall.  相似文献

The micro-anatomy of the corpuscles of Stannius of the toadfish, Opsanus tau, an aglomerular marine teleost, has been studied by light and electron microscopy. The corpuscles are composed of extensively anastomosed cords of epithelial cells which maintain intimate contact with blood capillaries. Most of the epithelial cells contain acidophilic granules which also show a positive reaction with the periodic acid-Schiff technique and aldehyde fuchsin. On the basis of fine structural criteria, three cell types can be recognized. The granular cells contain abundant quantities of granular endoplasmic reticulum, ribosomes, Golgi apparatus with prosecretory granules, coated vesicles, polymorphic mitochondria with lamellar cristae, filaments, microtubules, a cilium, a variety of lysosome-like dense bodies, glycogen particles, lipid droplets, secretory granules and intranuclear lipid-like inclusions. One variety of agranular cell (type I) is characterized by the total absence of secretory granules, but it contains large amounts of granular endoplasmic reticulum and ribosomes, conspicuous profiles of Golgi apparatus, coated vesicles and sometimes an abundance of glycogen. Another variety of agranular cell (type II) has poorly developed cytoplasmic organelles. The perivascular space between the capillary and parenchyma contains connective tissue cells and abundant nerve fibers. The different types of epithelial cells observed in the corpuscles of Stannius of this fish may represent functional stages of the secretory cycle in a single cell type.  相似文献

Summary The organization of the flagellum abductor muscle and of a scaphognathite levator muscle of the green crab, Carcinus maenas, has been compared quantitatively using light and electron microscopy. These muscles are rhythmically active at relatively high frequencies and for long durations. Fibers of both muscles are interconnected to form fascicles of 50 or more fibers within which there is cytoplasmic continuity. A single muscle is made up of 8–12 fascicles. Individual fibers consist of a peripheral rind of densely packed mitochondria, a thick region of glycogen granules, and myofibrils arranged into scattered central islands. Less than half the volume-density of these muscles is contractile material, the balance being largely mitochondria and glycogen. The fibers within a muscle are structurally similar. They have short sarcomeres (about 2 m), thin to thick filament ratios of about 3:1, and junctions between the sarcoplasmic reticulum and the transverse tubules at the M line. Sarcoplasmic reticulum occupies about 10% of the myofibrillar volume-density. A well developed sarcoplasmic reticulum appears to underlie the capacities of these two muscles for high frequency contraction; extensive mitochondria and glycogen stores should confer fatigue resistance under both aerobic and anaerobic conditions.  相似文献

The results of an electron microscopic histochemical investigation performed in the current study indicate that in heart cells creatine phosphokinase is localized:(1) inside mitochondria on the cristae membranes, (2) on the membrane of the sarcoplasmic reticulum, (3) on myofbrils (and in cytoplasm), (4) on the plasma membrane of the cells, (5) on the membrane of the cell nuclei.  相似文献

Summary The fine structure of rabbit Spermatogonia and primary spermatocytes in meiotic prophase has been studied with different methods of preparation, including a technique for acid phosphatase activity. The spermatogonial cytoplasm is rich in free ribosomes and containes moderate amounts of vesicular, smooth-surfaced endoplasmic reticulum and mitochondria, a simple Golgi-apparatus, some micropinocytotic vesicles, and occasional multivesicular bodies, vacuoles and dense bodies with acid phosphatase activity. The large type A Spermatogonia have a prominent nucleolus and their mitochondria sometimes form clusters with a dense intermitochondrial substance, similar to that in spermatocytes.The nucleus and cytoplasm of primary spermatocytes increase markedly in volume and density during meiotic prophase. The Golgi apparatus enlarges and becomes more differentiated and finally forms small proacrosome granules. The endoplasmic reticulum produces numerous small, mainly smooth vesicles and might also be the source of a new organelle: numerous piles of narrow cisternae with opaque contents. These piles disintegrate late in prophase. The mitochondria become aggregated in clusters with dense intermitochondrial substance and their internal structure is characterized by highly dilated cristae and small particles, interpreted as mitochondrial ribosomes, in the matrix. The role of these structures in the formation of new mitochondria is discussed. The clusters of mitochondria finally disperse and their cores of dense intermitochondrial substance, possibly containing ribonucleoprotein, coalesce into a large chromatoid body similar to that in spermatids. Micropinocytosis and a few lysosomes occur in most spermatocytes. The pachytene nuclei show prominent nucleoli and a distinct sex vesicle without any synaptinemal complex.The importance for spermatid differentiation of some events taking place in the cytoplasm of primary spermatocytes is emphasized.Financial support for this study was received from the Swedish Medical Research Council.  相似文献

The parotid gland of Dasyuroides byrnei was examined by light microscopy, and transmission and scanning electron microscopy. The acini were composed predominantly of seromucous cells with a few mucous cells. The seromucous cells were light or dark cells containing acidophilic spherical granules of moderate to high electron density and had well-developed cytoplasmic organelles-ordinary mitochondria and large mitochondria with tubular cristae, RER with vesicular or tubular elements, and Golgi apparatus with lamellae, vesicles and vacuoles. The mucous cells had basophilic amorphous granules of low electron density, like those of ordinary mucous cells. The intercalated ducts were composed of simple cuboidal light cells having a few electron-dense granules. The striated ducts consisted of tall columnar light cells containing numerous vesicles and mitochondria with tubular cristae, the same as found in acinar seromucous cells.  相似文献
